Cutin, one of the main structural components of tomato peels, is a waxy biopolymer rich in hydroxylated fatty acids. In this study, 10,16-dihydroxyhexadecanoic acid (10,16-diHHDA) was extracted and isolated from tomato peels and exploited to develop fully crosslinked polyesters as potential candidates for replacing fossil-based metal protective coatings. A preliminary screening was conducted to select the base formulation, and then a design of experiments (DoE) was used as a methodology to identify the optimal composition to develop a suitable coating material. Different formulations containing 10,16-diHHDA and other biorefinery monomers, including 2,5-furandicarboxylic acid, were considered. To this end, all polyesters were characterized through differential scanning calorimetry (DSC) and gel content measurements to determine their T g value and crosslinking efficiency. Compositions exhibiting the best trade-off between T g value, chemical resistance, and sufficiently high 10,16-diHHDA content between 39 and 48 wt.% were used to prepare model coatings that were characterized for assessing their wettability, scratch hardness, chemical resistance, and adhesion to metal substrates. These polyester coatings showed a T g in the range of 45-55 °C, a hydrophobic behavior with a water contact angle of around 100°, a good solvent resistance (>100 MEK double rubs), and an adhesion strength to steel higher than 2 MPa. The results obtained confirmed the potential of cutin-based resins as coatings for metal protection, meeting the requirements for ensuring physicochemical properties of the final product, as well as for optimizing the valorization of such an abundant agri-food waste as tomato peels.

The purpose of this study was to evaluate the evolution of technical performance indicators over the course of football matches in the UEFA Champions League. Three elite football teams were the sample of the present study and were analyzed throughout four consecutive seasons within the previously mentioned competition. Data from 15 min periods were collected from Wyscout and elaborated. The effects of match location and competition stage were analyzed on nine technical indicators, including ball possession and variables related to offense and goal scoring. The effects of independent variables were assessed both independently and combined. The results showed a significant increase in the frequency of occurrence and accuracy of most of the parameters towards the end of the match. The effect of match location was generally significant with higher rates for teams playing at home. Differences were noted between the two stages of the competition with higher values in the technical indicators registered during the group stage. The existence of significant differences between the initial and final periods of football games was suggested by the results. The so-called home advantage was confirmed. Different team approaches between longer stages (e.g., group stage) and elimination games (i.e., knockout phase) were suggested by the results.

Quartz Crystal Microbalances (QCM) are widely used instruments thanks to their stability, low mass, and low cost. Nevertheless, the sensitivity to temperature is their main drawback and is often a driver for their design. Though the crystal average temperature is mostly considered as the only disturbance, temperature affects the QCM measurements also through the in-plane temperature gradients, an effect identified in the past but mostly neglected. Recently, it has been shown that this effect can prevail over that of the average temperature in implementations where the heat for thermal control is released directly on the crystal through deposited film heaters. In this study, the effect of temperature gradients for this kind of crystal is analyzed, the sensitivity of frequency to the average temperature gradient on the electrode border is determined, and a correction is proposed and verified. A numerical thermal model of the QCM has been created to determine the temperature gradients on the electrode borders. The frequency versus temperature-gradient function has been experimentally determined in different thermal conditions. The correction function has been eventually applied to a QCM implementing a crystal of the same manufacturing lot as the one used for the characterization. The residual errors after the implementation of the correction of both average temperature and temperature gradients were always lower than 5% of the initial temperature disturbance. Moreover, using the correlation between the heater power dissipation and the generated temperature gradients, it has been shown that an effective correction strategy can be based on the measurement of the power delivered to the crystal without the determination of the temperature gradient.

This work aims to improve the corrosion protection features of poly(phenylene methylene) (PPM) by sidechain engineering inserting methoxy units along the polymer backbone. The influence of side methoxy groups at different concentrations (4.6% mol/mol and 9% mol/mol) on the final polymer properties was investigated by structural and thermal characterization of the resulting copolymers: co-PPM 4.6% and co-PPM 9%, respectively. Then, coatings were processed by hot pressing the polymers powder on aluminum alloy AA2024 and corrosion protection properties were evaluated exposing samples to a 3.5% w/v NaCl aqueous solution. Anodic polarization tests evidenced the enhanced corrosion protection ability (i.e., lower current density) by increasing the percentage of the co-monomer. Coatings made with co-PPM 9% showed the best protection performance with respect to both PPM blend and PPM co-polymers reported so far. Electrochemical response of aluminum alloy coated with co-PPM 9% was monitored over time under two "artificially-aged" conditions, that are: (i) a pristine coating subjected to potentiostatic anodic polarization cycles, and (ii) an artificially damaged coating at resting condition. The first scenario points to accelerating the corrosion process, the second one models damage of the coating potentially occurring either due to natural deterioration or due to any accidental scratching of the polymer layer. In both cases, an intrinsic self-healing phenomenon was indirectly argued by the time evolution of the impedance and of the current density of the coated systems. In vitro models of corticogenesis from pluripotent stem cells (PSCs) have greatly improved our understanding of human brain development and disease. Among these, 3D cortical organoid systems are able to recapitulate some aspects of in vivo cytoarchitecture of the developing cortex. Here, we tested three cortical organoid protocols for brain regional identity, cell type specificity and neuronal maturation. Overall, all protocols gave rise to organoids that displayed a time-dependent expression of neuronal maturation genes such as those involved in the establishment of synapses and neuronal function. Comparatively, guided differentiation methods without WNT activation generated the highest degree of cortical regional identity, whereas default conditions produced the broadest range of cell types such as neurons, astrocytes and hematopoietic-lineage-derived microglia cells. These results suggest that cortical organoid models produce diverse outcomes of brain regional identity and cell type specificity and emphasize the importance of selecting the correct model for the right application.

Quartz crystal microbalances are widely used sensors with applications for the detection of very-low-mass deposition in many different fields, from contamination monitoring in the high vacuum of deep space missions to the monitoring of biological activity or pollution using specifically designed active substrates. These sensors are very stable over time; nevertheless, their sensitivity to the temperature is well known, and different implementations have been devised to correct it, e.g., through compensation with a dual crystal. This paper deals with the effects of temperature on QCM but separates the case of uniform crystal temperature from the case of in-plane temperature gradients considering a QCM based on quartz crystals with deposited film resistors used as both RTDs and heaters. This configuration allows both an accurate temperature measurement and efficient thermal control, allowing the achievement of crystals temperatures in the order of 400 °C higher than the environment with a low power dissipation of the order of 1 W. The film resistors deposited around the electrodes allow directly measuring the average crystal temperature and directly delivering power to the crystal for thermal control. The localized delivery of the heat nevertheless also determines uncommon temperature fields on the crystal, and thus, an analysis of both the effects of temperature on the new microbalance was performed. The temperature gradient has strong effects on the frequency; therefore, along with the temperature, the thermal gradients have tobe compensated. The calibration of the QCM thermometers and the assessment of the achievable measurement accuracy were performed, as well as the determination of the frequency-temperature relationship. The comparison between frequency changes in the case of uniform temperature and those observed while using crystal heaters proved that temperature gradients have a strong effect on the crystal frequency. To identify the temperature field on the crystal surface of a QCM crystal, the gold coating of the deposited films was removed to achieve an emissivity acceptable for thermal imaging with an IR camera. Moreover, image processing for emissivity correction was developed. In order to correlate the temperature gradient with the frequency variation, a test campaign was performed to measure the frequency changes derived from different power levels delivered to the crystal heaters. From this test campaign and thermal analysis, the effect of the thermal gradient was assessed.

Poly(phenylene methylene) (PPM) is a multifunctional polymer featuring hydrophobicity, high thermal stability, fluorescence and thermoplastic processability. Accordingly, smart corrosion resistant PPM-based coatings (blend and copolymer) were prepared and applied by hot pressing on aluminum alloy AA2024. The corrosion protection properties of the coatings and their dependence on coating thickness were evaluated for both strategies employed. The accelerated cyclic electrochemical technique (ACET), based on a combination of electrochemical impedance spectroscopy (EIS), cathodic polarizations and relaxation steps, was used as the main investigating technique. At the coating thickness of about 50 µm, both blend and copolymer PPM showed effective corrosion protection, as reflected by |Z| 0.01Hz of about 10 8 Ω cm 2 over all the ACET cycles. In contrast, when the coating thickness was reduced to 30 µm, PPM copolymer showed neatly better corrosion resistance than blended PPM, maintaining |Z| 0.01Hz above 10 8 Ω cm 2 with respect to values below 10 6 Ω cm 2 of the latter. Furthermore, the analysis of many electrochemical key features, in combination with the optical investigation of the coating surface under 254 nm UV light, confirms the intrinsic self-healing ability of the coatings made by PPM copolymer, contrary to the reference specimen (i.e., blend PPM).

β-glucocerebrosidase is a lysosomal hydrolase involved in the catabolism of the sphingolipid glucosylceramide. Biallelic loss of function mutations in this enzyme are responsible for the onset of Gaucher disease, while monoallelic β-glucocerebrosidase mutations represent the first genetic risk factor for Parkinson's disease. Despite this evidence, the molecular mechanism linking the impairment in β-glucocerebrosidase activity with the onset of neurodegeneration in still unknown. In this frame, we developed two in vitro neuronal models of β-glucocerebrosidase deficiency, represented by mouse cerebellar granule neurons and human-induced pluripotent stem cells-derived dopaminergic neurons treated with the specific β-glucocerebrosidase inhibitor conduritol B epoxide. Neurons deficient for β-glucocerebrosidase activity showed a lysosomal accumulation of glucosylceramide and the onset of neuronal damage. Moreover, we found that neurons react to the lysosomal impairment by the induction of their biogenesis and exocytosis. This latter event was responsible for glucosylceramide accumulation also at the plasma membrane level, with an alteration in lipid and protein composition of specific signaling microdomains. Collectively, our data suggest that β-glucocerebrosidase loss of function impairs the lysosomal compartment, establishing a lysosome-plasma membrane axis responsible for modifications in the plasma membrane architecture and possible alterations of intracellular signaling pathways, leading to neuronal damage.

Mature T-cell lymphomas (MTCLs) represent a heterogeneous group of aggressive non-Hodgkin lymphomas comprising different entities. Anthracycline-based regimens are considered the standard of care in the front-line treatment. However, responses to these approaches have been neither adequate nor durable, and new treatment strategies are urgently needed to improve survival. Genomic instability is a common feature of cancer cells and can be caused by aberrations in the DNA damage response (DDR) and DNA repair mechanisms. Consistently, molecules involved in DDR are being targeted to successfully sensitize cancer cells to chemotherapy. Recent studies showed that some hematological malignancies display constitutive DNA damage and intrinsic DDR activation, but these features have not been investigated yet in MTCLs. In this study, we employed a panel of malignant T cell lines, and we report for the first time the characterization of intrinsic DNA damage and basal DDR activation in preclinical models in T-cell lymphoma. Moreover, we report the efficacy of targeting the apical kinase ATM using the inhibitor AZD0156, in combination with standard chemotherapy to promote apoptotic cell death. These findings suggest that DDR is an attractive pathway to be pharmacologically targeted when developing novel therapies and improving MTCL patients' outcomes.

MicroMED (Micro Martian Environmental Dust Systematic Analyzer (MEDUSA)) instrument was selected for the ExoMars 2020 mission to study the airborne dust on the red planet through in situ measurements of the size distribution and concentration. This characterization has never been done before and would have a strong impact on the understanding of Martian climate and Aeolian processes on Mars. The MicroMED is an optical particle counter that exploits the measured intensity of light scattered by dust particles when crossing a collimated laser beam. The measurement technique is well established for laboratory and ground applications but in order to be mounted on the Dust Suite payload within the framework of ExoMars 2020 mission, the instrument must be compatible with harsh mechanical and thermal environments and the tight mass budget of the mission payload. This work summarizes the thermo-mechanical design of the instrument, the manufacturing of the flight model and its successful qualification in expected thermal and mechanical environments., Competing Interests: The authors declare no conflicts of interest.
