Protease biocatalysis in a high-salt environment is very attractive for applications in the detergent industry, the production of diagnostic kits, and traditional food fermentation. However, high-salt conditions can reduce protease activity or even inactivate enzymes. Herein, in order to explore new protease sources, we expressed a salt-tolerant pseudolysin of Pseudomonas aeruginosa SWJSS3 isolated from deep-sea mud in Saccharomyces cerevisiae. After optimizing the concentration of ion cofactors in yeast peptone dextrose (YPD) medium, the proteolytic activity in the supernatant was 2.41 times more than that in the control group when supplemented with 5 mM CaCl2 and 0.4 mM ZnCl2. The extracellular proteolytic activity of pseudolysin reached 258.95 U/mL with optimized expression cassettes. In addition, the S. cerevisiae expression system increased the salt tolerance of pseudolysin to sodium chloride (NaCl)and sodium dodecyl sulfate (SDS) and the recombinant pseudolysin retained 15.19% activity when stored in 3 M NaCl for 7 days. The recombinant pseudolysin was able to efficiently degrade the β-conglycinin from low-denatured soy protein isolates and glycinin from high-denatured soy protein isolates under high temperatures (60 °C) and high-salt (3 M NaCl) conditions. Binomial autoregressive models are frequently used for modeling bounded time series counts. However, they are not well developed for more complex bounded time series counts of the occurrence of n exchangeable and dependent units, which are becoming increasingly common in practice. To fill this gap, this paper first constructs an exchangeable Conway–Maxwell–Poisson-binomial (CMPB) thinning operator and then establishes the Conway–Maxwell–Poisson-binomial AR (CMPBAR) model. We establish its stationarity and ergodicity, discuss the conditional maximum likelihood (CML) estimate of the model's parameters, and establish the asymptotic normality of the CML estimator. In a simulation study, the boxplots illustrate that the CML estimator is consistent and the qqplots show the asymptotic normality of the CML estimator. The rapid growth in demand for portable and intelligent hardware has caused tremendous pressure on signal sampling, transfer, and storage resources. As an emerging signal acquisition technology, compressed sensing (CS) has promising application prospects in low-cost wireless sensor networks. To achieve reduced energy consumption and maintain a longer acquisition duration for high sample rate electromyogram (EMG) signals, this paper comprehensively analyzes the compressed sensing method using EMG. A fair comparison is carried out on the performances of 52 ordinary wavelet sparse bases and five widely applied reconstruction algorithms at different compression levels. The experimental results show that the db2 wavelet basis can sparse EMG signals so that the compressed EMG signals are reconstructed properly, thanks to its low percentage root mean square distortion (PRD) values at most compression ratios. In addition, the basis pursuit (BP) reconstruction algorithm can provide a more efficient reconstruction process and better reconstruction performance by comparison. The experiment records and comparative analysis screen out the suitable sparse bases and reconstruction algorithms for EMG signals, acting as prior experiments for further practical applications and also a benchmark for future academic research. Recently, there has been a growing interest in integer-valued time series models, especially in multivariate models. Motivated by the diversity of the infinite-patch metapopulation models, we propose an extension to the popular bivariate INAR(1) model, whose innovation vector is assumed to be time-dependent in the sense that the mean of the innovation vector is linearly increased by the previous population size. We discuss the stationarity and ergodicity of the observed process and its subprocesses. We consider the conditional maximum likelihood estimate of the parameters of interest, and establish their large-sample properties. Sonoporation employs ultrasound accompanied by microbubble (MB) cavitation to induce the reversible disruption of cell membranes and has been exploited as a promising intracellular macromolecular delivery strategy. Due to the damage to cells resulting from strong cavitation, it is difficult to balance efficient delivery and high survival rates. In this paper, a traveling surface acoustic wave (TSAW) device, consisting of a TSAW chip and a polydimethylsiloxane (PDMS) channel, was designed to explore single-cell sonoporation using targeted microbubbles (TMBs) in a non-cavitation regime. A TSAW was applied to precisely manipulate the movement of the TMBs attached to MDA-MB-231 cells, leading to sonoporation at a single-cell level. The impact of input voltage and the number of TMBs on cell sonoporation was investigated. In addition, the physical mechanisms of bubble cavitation or the acoustic radiation force (ARF) for cell sonoporation were analyzed. The TMBs excited by an ARF directly propelled cell membrane deformation, leading to reversible perforation in the cell membrane. When two TMBs adhered to the cell surface and the input voltage was 350 mVpp, the cell sonoporation efficiency went up to 83%. [ABSTRACT FROM AUTHOR]

Excess zeros is a common phenomenon in time series of counts, but it is not well studied in asymmetrically structured bivariate cases. To fill this gap, we first considered a new first-order, bivariate, random coefficient, integer-valued autoregressive model with a bivariate innovation, which follows the asymmetric Hermite distuibution with five parameters. An attractive advantage of the new model is that the dependence between series is achieved by innovative parts and the cross-dependence of the series. In addition, the time series of counts are modeled with excess zeros, low counts and low over-dispersion. Next, we established the stationarity and ergodicity of the new model and found its stochastic properties. We discuss the conditional maximum likelihood (CML) estimate and its asymptotic property. We assessed finite sample performances of estimators through a simulation study. Finally, we demonstrate the superiority of the proposed model by analyzing an artificial dataset and a real dataset. The Li-ion battery is of paramount importance to electric vehicles (EVs). Propelled by the rapid growth of the EV industry, the performance of the battery is continuously improving. However, Li-ion batteries are susceptible to the working temperature and only obtain the optimal performance within an acceptable temperature range. Therefore, a battery thermal management system (BTMS) is required to ensure EVs' safe operation. There are various basic methods for BTMS, including forced-air cooling, liquid cooling, phase change material (PCM), heat pipe (HP), thermoelectric cooling (TEC), etc. Every method has its unique application condition and characteristic. Furthermore, based on basic BTMS, more hybrid cooling methods adopting different basic methods are being designed to meet EVs' requirements. In this work, the hybrid BTMS, as a more reliable and environmentally friendly method for the EVs, will be compared with basic BTMS to reveal its advantages and potential. The characteristics of the transcritical CO2 heat pump water heater (HPWH) system are; a lower inlet hot water temperature (Ti-hw) (sometimes this is lower than the water source temperature), and an outlet gas cooler temperature (To-gc) which is affected by the Ti-hw and often lower than the critical temperature. In order to study the effects of the internal heat exchanger (IHX) on the operational performance of the transcritical CO2 HPWH when To-gc is low, a transcritical CO2 water source HPWH experiment platform is established to conduct experimental research and comparative analysis on the operational performance of the transcritical CO2 water source HPWH, with or without IHX. It is found that, if only the coefficient of performance (COP) and heating at the optimal exhaust pressure of the transcritical CO2 water source HPWH were considered, COP and the heating of the non-IHX system would be slightly higher than those of the IHX system at the lower hot water flow and water source temperature, and this increase was not obvious. At the higher hot water flow rate and water source temperature, COP and the heating of the non-IHX system were also higher than those of the IHX system, and the increase was obvious. The experiment results showed that, near the optimal exhaust pressure, the variation range of COP and heating of the IHX system is relatively small, and the system has a relatively high stability. Cell lysis is a process of breaking cell membranes to release intracellular substances such as DNA, RNA, protein, or organelles from a cell. The detection of DNA, RNA, or protein from the lysed cells is of importance for cancer diagnostics and drug screening. In this study, we develop a microbubble array that enables the realization of multiple cell lysis induced by the shear stress resulting from the individual oscillating microbubbles. The oscillating microbubbles in the channel have similar vibration amplitudes, and the intracellular substances can be released from the individual cells efficiently. Moreover, the efficiency of cell lysis increases with increments of input voltage and sonication time. By means of DNA agarose-gel electrophoresis, a sufficient extraction amount of DNA released from the lysed cells can be detected, and there is no significant difference in lysis efficiency when compared to cell lysis achieved using commercial kits. With the advantages of the simple manufacturing process, low cost, high efficiency, and high speed, this device can serve as an efficient and versatile tool for the single-cell sequencing of cell biology research, disease diagnosis, and stem cell therapy. Spray cooling with liquid nitrogen is increasingly utilized as an efficient approach to achieve cryogenic cooling. Effects of injection mass flow rate fluctuations on the evaporation, temperature distribution, and droplet distribution of a spray field were examined by employing a validated Computational Fluid Dynamics (CFD) numerical model. The numerical results indicated that injection fluctuations enhanced the volume-averaging turbulent kinetic energy and promoted the evaporation of the whole spray field. The strengthened mass and heat transfer between the liquid nitrogen droplets and the surrounding vapor created by the fluctuating injection led to a lower temperature of the whole volume. A relatively smaller droplet size and a more inhomogeneous droplet distribution were obtained under the unsteady inlet. The changes of the frequency and the amplitude of the fluctuations had little effects on the overall spray development. The results could enrich the knowledge of the relation between the inevitable fluctuations and the overall spray development and the cooling performance in a practical spray cooling system with cryogenic fluids. In order to implement the omnidirectional wireless power transfer (WPT), a novel three-phase-shifted drive omnidirectional WPT system is proposed. This system is comprised of three independent phase-adjusted excitation sources, three orthogonal transmitting coils, and one planar receiving coil. Based on the mutual coupling theory, the power transfer efficiency is derived and the corresponding control mechanism for maximizing this efficiency is presented. This control mechanism only depends on the currents’ root-mean-square (RMS) values of the three transmitting coils and simple calculations after each location and/or posture change of the receiving coil, which provides the real-time possibility to design an omnidirectional WPT system comparing with the other omnidirectional systems. In aid of computer emulation technique, the efficiency characteristic versus the omnidirectional location and posture of the receiving coil is analyzed, and the analytical results verify the validity of the control mechanism. Lastly, a hardware prototype has been set up, and its omnidirectional power transmission capacity has been successfully verified. The experimental results show that the wireless power is omnidirectional and it can be effectively transmitted to a load even though its receiving coil moves and/or rotates in a 3-D energy region.
