Although β-glucan has diverse benefits for fish health, the potential adverse impacts of excessive supplementation are poorly understood. This study investigated the optimal dosage of β-glucan for coral trout and explored the adverse effects of excessive supplementation. The results revealed that moderate β-glucan supplementation (1) significantly enhanced the weight gain rate and specific growth rate (SGR); (2) significantly improved the intestinal villus length (0.10%), muscle thickness (0.05–0.15%), and α-amylase and chymotrypsin activities (0.05–0.15%); (3) significantly increased liver catalase (CAT), glutathione reductase (GR), alkaline phosphatase, complement 3, immunoglobulin M (IgM), lysozyme, serum superoxide dismutase, CAT, glutathione peroxidase, GR, total antioxidant capacity, acid phosphatase, complement 4, and IgM activities and significantly reduced malondialdehyde contents; (4) upregulated genes in the liver associated with copper-zinc superoxide dismutase (SOD-1), manganese superoxide dismutase (SOD-2), CAT, GSH-Px1a, ACP6, AKP, LZ-c, IgM, C3, and C4-b. However, 0.20% β-glucan significantly inhibited the growth performance of coral trout compared with 0.10% β-glucan. Thus, 0.10% β-glucan represents the optimal dosage for promoting growth, antioxidant activity, and immune responses in coral trout, while higher β-glucan levels weakened these beneficial effects. With an SGR established by a cubic regression analysis, the optimal level of β-glucan for maximal growth of coral trout is 0.082%. Simple Summary: In aquaculture, probiotics are widely used to improve growth, digestion, immunity, and so on. Employing a combination of multiple probiotic strains to enhance host health promotion is commonly acknowledged. However, studies assessing the synergistic effects of mixed probiotic additives on hosts and comparing the properties of each strain in their composition remain unclear. Therefore, this study aims to elucidate the mechanisms underlying the effects of individual and combined endogenous probiotics on hybrid groupers. Our findings revealed that the addition of mixed probiotics (Bacillus cereus and Exiguobacterium acetylicum) effectively enhances the growth performance, immune response, and intestinal health of hybrid groupers. Therefore, we recommend selecting combination strains of endogenous probiotics for optimal growth and immunity in hybrid groupers. This study aims to examine the effects of the mixture of Bacillus cereus G1–11 and Exiguobacterium acetylicum G1–33, isolated from the gut of hybrid groupers (Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂), on the host. The hybrid groupers were divided into a control (C, without any probiotics), B. cereus (BC, 1010 cfu/g), E. acetylicum (EA, 108 cfu/g), compound (mix, a 1:1 mixture of B. cereus and E. acetylicum), and positive reference group (P, Lactobacillus acidophilus, 5 × 108 cfu/L). Each group had four replicates, with 30 fish per replicate (53.30 ± 0.50 g), and were fed for 60 days. The results showed that adding probiotics to the feed significantly improved the weight gain, weight growth rate, specific growth rate, and digestive enzyme activities of hybrid groupers compared to the C group. The compound group was the most significant. In addition, composite probiotics added to feed significantly upregulated the expression levels of several growth-related genes in the liver and muscles. The activities of alkaline phosphatase, catalase, glutathione peroxidase, glutathione transferase, lysozyme, and total antioxidant capacity in the serum and liver were significantly influenced through mixed probiotic feeding. Moreover, the expression levels of several immune-related genes in the liver, spleen, and head kidney were significantly enhanced by adding single and mixed probiotics to feed, with the synergy of mixed probiotics being the best. An analysis of the gut microbiota showed that adding composite bacteria enhanced the richness and diversity of the gut microbiota, significantly increasing the relative abundance of potential probiotics (Cetobacterium and Microbacterium) while decreasing the presence of potential pathogens (Mycoplasma). Overall, our findings highlighted the efficacy of mixed probiotics (B. cereus and E. acetylicum) in enhancing growth performance, nutritional value of hybrid grouper feed, antioxidant capacity, immune response, and intestinal health, in finding the best combination of functional feed additives. Simple Summary: Siganus oramin, as a commercial species, is a well-received table fish in southeastern China. However, valuable reproduction-related data are still scarce. In the present study, gonad transcriptome analysis was first performed using Illumina Novaseq technology. Comparative transcriptome analysis of adult male and female gonads identified a large number of differentially expressed genes. Among them, many reproduction-related genes that play important roles in gonad differentiation and development were found. These data may contribute greatly to the regulation of rabbitfish reproduction. The rabbitfish, Siganus oramin, is a commercially important table fish in southeastern China. However, there have been few studies on its gonad development and reproduction regulation. Comparative transcriptome analysis was first performed on adult male and female gonads of S. oramin. In total, 47,070 unigenes were successfully assembled and 22,737 unigenes were successfully annotated. Through comparative transcriptome analysis of male and female gonads, a total of 6722 differentially expressed genes were successfully identified, with 3528 upregulated genes and 3154 downregulated genes in the testes. In addition, 39 differentially expressed reproduction-related genes were identified. Finally, quantitative real-time PCR was used to validate the expression levels of several differentially expressed genes. These results provide important data for further studying the function of reproduction-related genes and the molecular mechanism regulating gonad development and reproduction in S. oramin. The rabbitfish Siganus oramin is an important naturally caught and aquaculture fish species. Intestinal microbiota can affect the metabolism and immunity of fish, which is closely related to the habitat of the host. In this study, we collected the wild fry S. oramin from a natural sea area, and cultured them in outdoor and indoor ponds, respectively, and investigated the changes in serum biochemical indexes and intestinal microbial community in three different habitats. The results showed that compared with the wild population, the serum total protein content of the outdoor culture population increased significantly. The indoor culture population had significantly higher triglyceride and total cholesterol contents than that of the outdoor culture population. Additionally, the intestinal microbial richness indexes ACE and Chao1 of the cultured population were higher than those of the wild population, especially the indoor culture, but Shannon and Simpson had no obvious changes. The relative abundances of Firmicutes, Spirochaetae and Bacteroidetes increased in the outdoor culture population, but decreased in the indoor culture population; those of Proteobacteria and Cyanobacteria were completely the opposite. Some putative beneficial bacteria (Lactobacillus, Clostridium sensu stricto 1, and Intestinibacter) and harmful bacteria (Vibrio, Photobacterium, Desulfovibrio, Streptococcus) all decreased in the intestines of the cultured population when compared with the wild population, while Akkermansia and Faecalibacterium were enriched in the outdoor culture population. These results reveal that a change in habitat environment, whether an outdoor or an indoor pond, positively influenced the intestinal microbiota of the rabbitfish, which is beneficial to the healthy culture of the fish from the perspective of microbial community. The present study was conducted to investigate the effects of dietary fermented cottonseed meal (FCSM) substitution for fish meal on intestinal enzymatic activity, inflammatory and physical-barrier-related gene expression, and intestinal microflora of juvenile golden pompano. The 375 golden pompanos were divided into 15 groups of 25 fish each, with three replicates for each experimental group. The fish were fed five experimental diets (0 (FM), 12.5% (CSM12.5), 25% (CSM25), 50% (CSM50), and 100% (CSM100) substitution levels) for 8 weeks. The fish were reared and fed the experimental diets under a natural-day light cycle. Compared with the control group, the activities of AMY (amylase) enzymes in the CSM12.5 group and all other groups were elevated (p < 0.05). The CSM25 group exhibited a considerable up-regulation of IL-10 (Interleukin-10) expression relative to the FM group (p < 0.05). With an increase in dietary FM substitution with FCSM from 0 to 25%, the relative expressions of NF-κB (Nuclear factor kappa-B), IL-1β (Interleukin-1 beta), and IL-8 (Interleukin-8) were down-regulated. In this study, the relative expressions of ZO-1 (zonula occluden-1) and Occludin were up-regulated, and those of Claudin-3 and Claudin-15 significantly up-regulated, when the FCSM substitution ratio was 25%. The results of high-throughput sequencing of the intestinal microflora showed that ACE indices the lowest in the CSM25 group, which was significantly different from those in the CSM100 group (p < 0.05). The CSM50 group had the highest Shannon and Simpson indices and the highest community diversity. In addition, replacing a high percentage of fish meal with FCSM can negatively affect the intestinal flora of fish. In this study, the 25% substitution ratio improved nutrient absorption, reduced intestinal inflammation, improved intestinal physical barrier damage, did not affect intestinal microecology, and had no adverse effects on fish. The present study was conducted to investigate the effects of high dietary carbohydrate levels on growth performance, enzyme activities, and gene expressions related to liver glucose metabolism and the intestinal microbiota of Lateolabrax maculatus juveniles. Two experimental diets with levels of carbohydrates (20% and 30%, named the NCD group and the HCD group, respectively) were designed to feed L. maculatus (initial weight 9.45 ± 0.03 g) for 56 days. The results showed that, compared with the NCD group, the condition factor (CF) was significantly elevated in the HCD group (p < 0.05). The plasma advanced glycosylation end products (AGEs), glycated serum protein (GSP), total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL), low-density lipoprotein (LDL), and glutamate aminotransferase (AST) were significantly higher in the HCD group than those in the NCD group (p < 0.05). The intestinal lipase, chymotrypsin, and α-amylase in the HCD group were significantly higher than those in the NCD group (p < 0.05). The liver superoxide dismutase (SOD), total antioxidant capacity (T-AOC), and catalase (CAT) were significantly lower in the HCD group than in the NCD group (p < 0.05). The liver malondialdehyde (MDA) and hexokinase (HK) levels were significantly higher than those in the NCD group (p < 0.05). In the histopathological findings, liver cells in the HCD group appeared to have many vacuoles, and the number of lipid droplets increased. Compared with the NCD group, the relative expression of liver glucokinase (GK) and glycogen synthetase kinase-3 (GSK3β) genes in the HCD group was significantly increased (p < 0.05), while the relative expression of phosphoenolpyruvate carboxykinase (PEPCK) and glycogen phosphorylase (GP) genes in the HCD group was significantly reduced (p < 0.05). High-throughput 16S rRNA gene sequencing showed that high dietary carbohydrate intake changed the composition and structure of the intestinal microbiota. At the phylum level of the intestinal microbiota, high dietary carbohydrates decreased the relative abundance of Firmicutes and increased the relative abundance of Proteobacteria and Bacteroidetes. At the genus level of the intestinal microbiota, high carbohydrates decreased the relative abundance of Bacillus and increased the relative abundance of Photobacterium and Paraclostridium. From the results of this experiment on L. maculatus, high carbohydrates led to increased condition factor and liver glycogen, lipid deposition, decreased antioxidant capacity of the liver, increased relative abundance of harmful intestinal microorganisms, and disrupted glucose metabolism. In this study, we investigated the effect of dietary curcumin on non-specific immune responses and antioxidative ability in Seriola dumerili under ammonia stress and post-recovery. Three diets were prepared to contain 0, 75, and 150 mg/kg of curcumin. A total of 225 greater amberjack (initial weight: 100.90 ± 0.03 g) were distributed into nine cylindrical tanks, constituting an experimental design with three treatments and three replicates. After 56 days of feeding, plasma, intestinal, and hepatic enzyme activities were evaluated. Then, an acute ammonia challenge experiment was conducted. Ten fish per tank were subjected to acute ammonia stress (total ammonia-N: 1000 mg/L) for eight minutes followed by six minutes of recovery. The results indicated that dietary curcumin significantly promoted intestinal and hepatic alkaline phosphatase (ALP) and acid phosphatase (ACP) levels as well as hepatic antioxidative enzymes such as superoxide dismutase (SOD), total antioxidant capacity (T-AOC), reduced glutathione (GSH), and glutathione peroxidase (GSH-Px) of greater amberjack. In addition, curcumin addition improved the activities of antioxidant enzymes, such as SOD, T-AOC, GSH, GSH-Px, and catalase (CAT), and reduced malondialdehyde (MDA) content in liver, spleen, head kidney, and brain tissues after post-recovery. The indexes related to immunity and antioxidant enzymes in the liver, gill, and spleen rose again to some extent, but they showed the worst recovery ability in the head kidney and brain tissue samples. These results indicate that dietary curcumin supplementation could increase non-specific immune responses, antioxidant ability, and enhance resistance to high ammonia stress in juvenile S. dumerili.
