Nonionic-zwitterionic diblock copolymers are designed to feature a coil-to-globule collapse transition with an upper critical solution temperature (UCST) in aqueous media, including physiological saline solution. The block copolymers that combine presumably highly biocompatible blocks are synthesized by chain extension of a poly(ethylene glycol) (PEG) macroinitiator via atom transfer radical polymerization (ATRP) of sulfobetaine and sulfabetaine methacrylates. Their thermoresponsive behavior is studied by variable temperature turbidimetry and ¹H NMR spectroscopy. While the polymers with polysulfobetaine blocks exhibit phase transitions in the physiologically interesting window of 30-50 °C only in pure aqueous solution, the polymers bearing polysulfabetaine blocks enabled phase transitions only in physiological saline solution. By copolymerizing a pair of structurally closely related sulfo- and sulfabetaine monomers, thermoresponsive behavior can be implemented in aqueous solutions of both low and high salinity. Surprisingly, the presence of the PEG blocks can affect the UCST-transitions of the polyzwitterions notably. In specific cases, this results in "schizophrenic" thermoresponsive behavior displaying simultaneously an UCST and an LCST (lower critical solution temperature) transition. Exploratory experiments on the UCST-transition triggered the encapsulation and release of various solvatochromic fluorescent dyes as model "cargos" failed, apparently due to the poor affinity even of charged organic compounds to the collapsed state of the polyzwitterions. The study addresses the effect of thermal treatment on the internal structure of polyelectrolyte multilayers (PEMs). In order to get insight into the internal structure of PEMs, Neutron Reflectometry (NR) was used. PEMs with a deuterated inner block towards the substrate and a non-deuterated outer block were prepared and measured in 1% RH and in D2O before and after a thermal treatment. Complementarily, PEMs with the same number of layers but completely non-deuterated were investigated by ellipsometry. The analysis for the overall thickness (d), the average scattering length density (SLD) and the refractive index (n) indicate a degradation of the PEM. The loss in material is independent of the number of layers, i.e., only a constant part of the PEM is affected by degradation. The analysis of the internal structure revealed a more complex influence of thermal treatment on PEM structure. Only the outermost part of the PEM degenerates, while the inner part becomes denser during the thermal treatment. In addition, the swelling behavior of PEMs is influenced by the thermal treatment. The untreated PEM shows a well pronounced odd--even effect, i.e., PDADMAC-terminated PEMs take up more water than PSS-terminated PEMs. After the thermal treatment, the odd-even effect becomes much weaker. A series of new fluorescent dye bearing monomers, including glycomonomers, based on maleamide and maleic esteramide was synthesized. The dye monomers were incorporated by radical copolymerization into thermo-responsive poly(N-vinyl-caprolactam) that displays a lower critical solution temperature (LCST) in aqueous solution. The effects of the local molecular environment on the polymers' luminescence, in particular on the fluorescence intensity and the extent of solvatochromism, were investigated below as well as above the phase transition. By attaching substituents of varying size and polarity in the close vicinity of the fluorophore, and by varying the spacer groups connecting the dyes to the polymer backbone, we explored the underlying structure-property relationships, in order to establish rules for successful sensor designs, e.g., for molecular thermometers. Most importantly, spacer groups of sufficient length separating the fluorophore from the polymer backbone proved to be crucial for obtaining pronounced temperature regulated fluorescence responses. The structures and synthesis of polyzwitterions ("polybetaines") are reviewed, emphasizing the literature of the past decade. Particular attention is given to the general challenges faced, and to successful strategies to obtain polymers with a true balance of permanent cationic and anionic groups, thus resulting in an overall zero charge. Also, the progress due to applying new methodologies from general polymer synthesis, such as controlled polymerization methods or the use of "click" chemical reactions is presented. Furthermore, the emerging topic of responsive ("smart") polyzwitterions is addressed. The considerations and critical discussions are illustrated by typical examples. [ABSTRACT FROM AUTHOR]

Polyelectrolytes in dilute solutions (0.01 mmol/L) adsorb in a two-dimensional lamellar phase to oppositely charged lipid monolayers at the air/water interface. The interchain separation is monitored by Grazing Incidence X-ray Diffraction. On monolayer compression, the interchain separation decreases to a factor of two. To investigate the influence of the electrostatic interaction, either the line charge density of the polymer is reduced (a statistic copolymer with 90% and 50% charged monomers) or mixtures between charged and uncharged lipids are used (dipalmitoylphosphatidylcholine (DPPC)/ dioctadecyldimethylammonium bromide (DODAB)) On decrease of the surface charge density, the interchain separation increases, while on decrease of the linear charge density, the interchain separation decreases. The ratio between charged monomers and charged lipid molecules is fairly constant; it decreases up to 30% when the lipids are in the fluid phase. Thermoresponsive polymer coatings allow the control of adhesion of cells on synthetic substrates. In particular, decreasing the temperature below the lower critical solution temperature (LCST) of the polymer triggers the non-invasive detachment of cells from their cultivation substrate. Widening the range of applications of these coatings in cellular biotechnology requires a better understanding of their interaction with cells. By monitoring the morphological changes of cells during their detachment at various temperatures, we provide evidence that cell detachment is an active process. Analyses of cell residues that are left behind by the cells on the substrate during their detachment, further support this notion. In the second part of this work, we show that the kinetics of adhesion and the efficiency of detachment of cells can be controlled through the coadsorption of molecules bearing the peptide motif RGD (arginine-glycine-aspartic acid) with the polymers. Films of zwitterionic polymers are increasingly explored for conferring fouling resistance to materials. Yet, the structural diversity of polyzwitterions is rather limited so far, and clear structure-property relationships are missing. Therefore, we synthesized a series of new polyzwitterions combining ammonium and sulfate groups in their betaine moieties, so-called poly(sulfabetaine)s. Their chemical structures were varied systematically, the monomers carrying methacrylate, methacrylamide, or styrene moieties as polymerizable groups. High molar mass homopolymers were obtained by free radical polymerization. Although their solubilities in most solvents were very low, brine and lower fluorinated alcohols were effective solvents in most cases. A set of sulfabetaine copolymers containing about 1 mol % (based on the repeat units) of reactive benzophenone methacrylate was prepared, spin-coated onto solid substrates, and photo-cured. The resistance of these films against the nonspecific adsorption by two model proteins (bovine serum albumin—BSA, fibrinogen) was explored, and directly compared with a set of references. The various polyzwitterions reduced protein adsorption strongly compared to films of poly(n-butyl methacrylate) that were used as a negative control. The poly(sulfabetaine)s showed generally even somewhat higher anti-fouling activity than their poly(sulfobetaine) analogues, though detailed efficacies depended on the individual polymer–protein pairs. The hydrolytic stability of polymers to be used for coatings in aqueous environments, for example, to confer anti-fouling properties, is crucial. However, long-term exposure studies on such polymers are virtually missing. In this context, we synthesized a set of nine polymers that are typically used for low-fouling coatings, comprising the well-established poly(oligoethylene glycol methylether methacrylate), poly(3-(N-2-methacryloylethyl-N,N-dimethyl) ammoniopropanesulfonate) (“sulfobetaine methacrylate”), and poly(3-(N-3-methacryamidopropyl-N,N-dimethyl)ammoniopropanesulfonate) (“sulfobetaine methacrylamide”) as well as a series of hitherto rarely studied polysulfabetaines, which had been suggested to be particularly hydrolysis-stable. Hydrolysis resistance upon extended storage in aqueous solution is followed by 1H NMR at ambient temperature in various pH regimes. Whereas the monomers suffered slow (in PBS) to very fast hydrolysis (in 1 M NaOH), the polymers, including the polymethacrylates, proved to be highly stable. No degradation of the carboxyl ester or amide was observed after one year in PBS, 1 M HCl, or in sodium carbonate buffer of pH 10. This demonstrates their basic suitability for anti-fouling applications. Poly(sulfobetaine methacrylamide) proved even to be stable for one year in 1 M NaOH without any signs of degradation. Polysulfobetaines in aqueous solution show upper critical solution temperature (UCST) behavior. We investigate here the representative of this class of materials, poly (N,N-dimethyl-N-(3-methacrylamidopropyl) ammonio propane sulfonate) (PSPP), with respect to: (i) the dynamics in aqueous solution above the cloud point as function of NaBr concentration; and (ii) the swelling behavior of thin films in water vapor as function of the initial film thickness. For PSPP solutions with a concentration of 5 wt.%, the temperature dependence of the intensity autocorrelation functions is measured with dynamic light scattering as function of molar mass and NaBr concentration (0–8 mM). We found a scaling of behavior for the scattered intensity and dynamic correlation length. The resulting spinodal temperatures showed a maximum at a certain (small) NaBr concentration, which is similar to the behavior of the cloud points measured previously by turbidimetry. The critical exponent of susceptibility depends on NaBr concentration, with a minimum value where the spinodal temperature is maximum and a trend towards the mean-field value of unity with increasing NaBr concentration. In contrast, the critical exponent of the correlation length does not depend on NaBr concentration and is lower than the value of 0.5 predicted by mean-field theory. For PSPP thin films, the swelling behavior was found to depend on film thickness. A film thickness of about 100 nm turns out to be the optimum thickness needed to obtain fast hydration with H2O.
