Time-of-Flight Diffraction (TOFD) is a method of ultrasonic testing (UT) that is widely established as a non-destructive technique (NDT) mainly used for the inspection of welds. In contrast to other established UT techniques, TOFD is capable of identifying discontinuities regardless of their orientation. This paper proposes a redesign of the typical TOFD transducers, featuring an innovative curved sole aimed at enhancing their defect detection capabilities. This design is particularly beneficial for thick-walled samples, as it allows for deeper inspections without compromising the resolution near the surface area. During this research, an evaluation consisting in simulations of the ultrasonic beam distribution and experimental tests on a component with artificially manufactured defects at varying depths has been performed to validate the new design. The results demonstrate a 30 to 50% higher beam distribution area as well as an improvement in the signal-to-noise ratio (SNR) resulting in a 24% enhancement in the capability of defect detection compared to the traditional approach.

In Cameroon, Aedes mosquitoes transmit various arboviruses, posing significant health risks. We aimed to characterize the Aedes virome in southwestern Cameroon and identify potential core viruses which might be associated with vector competence. A total of 398 Aedes mosquitoes were collected from four locations (Bafoussam, Buea, Edea, and Yaounde). Aedes albopictus dominated all sites except for Bafoussam, where Aedes africanus prevailed. Metagenomic analyses of the mosquitoes grouped per species into 54 pools revealed notable differences in the eukaryotic viromes between Ae. africanus and Ae. albopictus , with the former exhibiting greater richness and diversity. Thirty-seven eukaryotic virus species from 16 families were identified, including six novel viruses with near complete genome sequences. Seven viruses were further quantified in individual mosquitoes via qRT-PCR. Although none of them could be identified as core viruses, Guangzhou sobemo-like virus and Bafoussam mosquito solemovirus, were highly prevalent regionally in Ae. albopictus and Ae. africanus , respectively. This study highlights the diverse eukaryotic virome of Aedes species in southwestern Cameroon. Despite their shared genus, Aedes species exhibit limited viral sharing, with varying viral abundance and prevalence across locations. Ae. africanus , an understudied vector, harbors a rich and diverse virome, suggesting potential implications for arbovirus vector competence.

The effect of birthplace (the place where a player is born and grows up) is one of the key variables associated with soccer player development and achievement. However, recent studies have questioned the influence of contextual variables on selection and promotion processes related to birthplace. The main purposes of this study were: (1) establish whether there is a difference between the birthplace and sporting growth according to the stages of entry into the academy, (2) to evaluate the influence of geographical and contextual variables on sporting development, and (3) assess the chances of making a professional team debut. Applied logistic regression was used in order to analyse the birthplace and growth of 1411 male soccer players, of which 40.1% are incorporated in the U-12 category from a Spanish First division club, and the results showed a statistically significant association between a change from one's birthplace to the place of sporting growth and later success (79%). Key contextual variables such as number of inhabitants, population density, proximity to sports facilities, average household income and presence of sports clubs have been considered. The analysis of the contextual variables revealed that there was a positive relationship between certain variables, like a higher population, and being selected for the academy ( p < 0.01; r = 0.28). Finally, the findings showed that players who experienced a geographical transition were 38% more likely to make a professional debut. The importance of considering the place of sporting growth when analysing the "birthplace effect" has therefore been demonstrated.

The burrowing nematode Radopholus similis is considered a major problem of intensive banana cultivation. It can cause extensive root damage resulting in the toppling disease of banana, which means that plants fall to the ground. Soaking R. similis in double-stranded (ds) RNA of the nematode genes Rps13 , chitin synthase ( Chs-2 ), Unc-87 , Pat-10 or beta-1,4-endoglucanase ( Eng1a ) suppressed reproduction on carrot discs, from 2.8-fold ( Chs-2 ) to 7-fold ( Rps13 ). The East African Highland Banana cultivar Nakitembe was then transformed with constructs for expression of dsRNA against the same genes, and for each construct, 30 independent transformants were tested with nematode infection. Four months after transfer from in vitro culture to the greenhouse, the banana plants were transferred to a screenhouse and inoculated with 2000 nematodes per plant, and thirteen weeks later, they were analyzed for several parameters including plant growth, root necrosis and final nematode population. Plants with dsRNA constructs against the nematode genes were on average showing lower nematode multiplication and root damage than the nontransformed controls or the banana plants expressing dsRNA against the nonendogenous gene. In conclusion, RNAi seems to efficiently protect banana against damage caused by R. similis , opening perspectives to control this pest.

Individuals with Down syndrome (DS) are more prone to develop severe respiratory tract infections. Although a RSV infection has a high clinical impact and severe outcome in individuals with DS, no vaccine nor effective therapeutics are available. Any research into infection pathophysiology or prophylactic and therapeutic antiviral strategies in the specific context of DS would greatly benefit this patient population, but currently such relevant animal models are lacking. This study aimed to develop and characterize the first mouse model of RSV infection in a DS-specific context. Ts65Dn mice and wild type littermates were inoculated with a bioluminescence imaging-enabled recombinant human RSV to longitudinally track viral replication in host cells throughout infection progression. This resulted in an active infection in the upper airways and lungs with similar viral load in Ts65Dn mice and euploid mice. Flow cytometric analysis of leukocytes in lungs and spleen demonstrated immune alterations with lower CD8+ T cells and B-cells in Ts65Dn mice. Overall, our study presents a novel DS-specific mouse model of hRSV infection and shows that potential in using the Ts65Dn preclinical model to study immune-specific responses of RSV in the context of DS and supports the need for models representing the pathological development.

The final quality of sheet and tube metal-formed components strongly depends on the tribology and friction conditions between the tools and the material to be formed. Furthermore, it has been recently demonstrated that friction is the numerical input parameter that has the biggest effect in the numerical models used for feasibility studies and process design. For these reasons, industrial dedicated software packages have introduced friction laws which are dependent on sliding velocity, contact pressure and sometimes strain suffered by the sheet, and currently, temperature dependency is being implemented as it has also a major effect on friction. In this work, three lubricants having different viscosity have been characterized using the tube-sliding test. The final aim of the study is to fit friction laws that are contact pressure and sliding velocity dependent for their use in tube hydroforming modeling. The tests performed at various contact pressures and velocities have demonstrated that viscosity has a major effect on friction. Experimental hydroforming tests using the three different lubricants have corroborated the importance of the lubricant in the final forming of a triangular shape. The measurement of the axial forces and the final principal strains of the formed tubes have shown the importance of using advanced friction laws to properly model the hydroforming process using the finite element modeling.

As end-users, employees appropriate technologies. Technology appropriation is generally conceived as a covert phenomenon. In particular, alternative ways and new purposes for which employees deploy technologies tend to remain hidden. Therefore, the potential of technologies as a source of organizational improvements may remain undisclosed. Continuous improvement (CI) programs, in contrast, are explicitly oriented at disclosing organizational improvements. In essence, CI programs encourage employees to openly discuss how to improve their work practices. Such continuous movements towards novel, often better, ways of working may be perfectly suited to bring the covert nature of technology appropriation into the open. Based on a case study on a personal digital assistant (PDA) in a Belgian nursing home with such a CI program in place, we document and analyze to what extent and why functionalities of the PDA were discussed and further developed. We distinguish between the functionalities that, upon implementation, intended to improve particular work practices, and those that surfaced after the technology had been introduced. To conclude, we point at employees' perceived usefulness of their work practices and their willingness to improve these, rather than only the technology itself, to further the debate on technology appropriation.

Today's stamping simulations are realized by ignoring the elastic deformation of the press and tooling system through the assumption of a rigid behavior and a perfect press stroke. However, in reality, the press and tool components deform elastically and are one of the major error sources for the final adjustment and blue-spotting of the dies. In order to tackle this issue, a new approach is proposed in this study that substitutes the press stiffness by means of a substitutive model composed of cost-effective shell and beam elements. The substitute model was calibrated using full-scale measurements, in which a 20,000 kN trial press was experimentally characterized by measuring its deformation under static loads. To examine the robustness of the substitute model, a medium-size tool and a large-size tool were simulated together with the substitutive model. To this end, a B-pillar tool was re-machined based on the substitute-model results and a new cambering procedure was proposed and validated throughout the blue-painting procedure. The newly developed substitute model was able to replicate the global stiffness of the press with a high accuracy and affordable calculation time. The implementation of the findings can aid toolmakers in eliminating most of the reworking and home-line trials.

The use of alternative fuels derived from residues in energy-intensive industries that rely on fossil fuels can cause considerable energy cost savings, but also significant environmental benefits by conserving non-renewable resources and reducing waste disposal. However, the switching from conventional to alternative fuels is challenging for industries, which require a sound understanding of the properties and combustion characteristics of the alternative fuel, in order to adequately adapt their industrial processes and equipment for its utilization. In this work, a solid recovered fuel (SRF) obtained from the polymeric fraction of an automotive shredder residue is tested for use as an alternative fuel for scrap preheating in an aluminium refinery. The material and chemical composition of the SRF has been extensively characterized using proximate and ultimate analyses, calorific values and thermal degradation studies. Considering the calorific value and the chlorine and mercury contents measured, the SRF can be designated as class code NCV 1; Cl 2; Hg 2 (EN ISO 21640:2021). The combustion of the SRF was studied in a laboratory-scale pilot plant, where the effects of temperature, flow, and an oxidizer were determined. The ash remaining after combustion, the collected liquid, and the generated gas phase were analysed in each test. It was observed that increasing the residence time of the gas at a high temperature allowed for a better combustion of the SRF. The oxidizer type was important for increasing the total combustion of the vapour compounds generated during the oxidation of the SRF and for avoiding uncontrolled combustion.

Reactive oxygen species present a challenge for marine organisms releasing gametes into the water. Thiol-containing molecules protect cells against oxidative stress, and ovothiol (OSH), an antioxidant-reducing mercaptohistidine, has been described as especially relevant in the oocytes of marine invertebrates. Ovothiol synthase ( ovoA ), in charge of the first step in OSH synthesis, was sequenced in mussels, Mytilus galloprovincialis . Transcription levels of ovoA in mantle did not significantly change along the reproductive cycle. No alterations of ovoA transcription were observed after a laboratory copper (10 µg/L) exposure or in mussels captured in a highly polluted site. Conversely, the metabolomic analysis of the hydrophilic metabolite content in mantle clearly classified mussels according to their site of origin, especially at the most advanced stages of oogenesis. Quantification of OSH-A and -B and glutathione (GSH), revealed stable levels in mantle at early gametogenesis in the unpolluted sampling site, but a strong increase in female mantle previous to spawning in the polluted site. These increased concentrations under pollution suggest that OSH-A accumulates along oogenesis, independent of gene transcription regulation. The concerted accumulation of OSH-A and GSH suggests the building of a balanced cellular redox-system to scavenge ROS produced in the oocyte before and during fertilization., Competing Interests: The authors declare no conflict of interest. Workpiece rejection originated by thermal damage is of great concern in high added-value industries, such as automotive or aerospace. Surface temperature control is vital to avoid this kind of damage. Difficulties in empirical measurement of surface temperatures in-process imply the measurement in points other than the ground surface. Indirect estimation of temperatures demands the use of thermal models. Among the numerous temperature measuring techniques, infra-red measurement devices excel for their speed and accurate measurements. With all of this in mind, the current work presents a novel temperature estimation system, capable of accurate measurements below the surface as well as correct interpretation and estimation of temperatures. The estimation system was validated by using a series of tests in different grinding conditions that confirm the hypotheses of the error made when measuring temperatures in the workpiece below the surface in grinding. This method provides a flexible and precise way of estimating surface temperatures in grinding processes and has shown to reduce measurement error by up to 60%.

Nematodes are a very diverse phylum that has adapted to nearly every ecosystem. They have developed specialized lifestyles, dividing the phylum into free-living, animal, and plant parasitic species. Their sheer abundance in numbers and presence in nearly every ecosystem make them the most prevalent animals on earth. In this research nematode-specific profiles were designed to retrieve predicted lectin-like domains from the sequence data of nematode genomes and transcriptomes. Lectins are carbohydrate-binding proteins that play numerous roles inside and outside the cell depending on their sugar specificity and associated protein domains. The sugar-binding properties of the retrieved lectin-like proteins were predicted in silico. Although most research has focused on C-type lectin-like, galectin-like, and calreticulin-like proteins in nematodes, we show that the lectin-like repertoire in nematodes is far more diverse. We focused on C-type lectins, which are abundantly present in all investigated nematode species, but seem to be far more abundant in free-living species. Although C-type lectin-like proteins are omnipresent in nematodes, we have shown that only a small part possesses the residues that are thought to be essential for carbohydrate binding. Diatoms are known to produce a variety of halogenated compounds, which were recently shown to have a role in allelopathic interactions between competing species. The production of these compounds is linked to haloperoxidase activity. This research, has shown that this system may also be involved in diatom-bacteria interactions via the H2O2 dependent inactivation of a type of quorum sensing (QS) molecule, i.e., N-β-ketoacylated homoserine lactones (AHLs), by a natural haloperoxidase system from the benthic diatom Nitzschia cf pellucida. The AHL degradation pathway towards corresponding halogenated derivatives was elucidated via HPLC-MS analysis and the synthesis of a broad series of novel halogenated AHL analogues as reference compounds. Furthermore, their biological activity as quorum sensing modulators was directly compared and evaluated against a series of naturally occurring β-keto-AHLs. It has been demonstrated that the loss of the QS activity results from the final cleavage of the halogenated N-acyl chain of the signal molecules.
