Tiliroside is a natural polyphenolic compound with a wide range of biological activity, and defatted strawberry seeds are its rich source. The goal of this study was to optimize accelerated solvent extraction (ASE) conditions, including temperature, solvent composition, and the number of extraction cycles, using Box–Behnken design to maximize the yield of tiliroside. UPLC-DAD-MS was applied to investigate the polyphenolic composition of the extracts, and preparative liquid chromatography (pLC) was used for isolation. All obtained mathematical models generally showed an increase in the efficiency of isolating polyphenolic compounds with an increase in temperature, ethanol content, and the number of extraction cycles. The optimal established ASE conditions for tiliroside were as follows: a temperature of 65 ◦C, 63% ethanol in water, and four extraction cycles. This allowed for the obtainment of a tiliroside-rich fraction, and the recovery of isolated tiliroside from plant material reached 243.2 mg from 100 g. Our study showed that ASE ensures the isolation of a tiliroside-rich fraction with high effectiveness. Furthermore, defatted strawberry seeds proved to be a convenient source of tiliroside because the matrix of accompanying components is relatively poor, which facilitates separation. Isoflavones, belonging to polyphenolic compounds, show structural similarity to natural estrogens, and in this context, they have been extensively studied. Some of them are also applied as cosmetic additives; however, little is known regarding their effects on skin cells. In this investigation, common isoflavones, including genistein, daidzein, glycitein, formononetin, and biochanin A, as well as coumestrol, were evaluated for antioxidant activity and their impact on human skin fibroblasts and keratinocytes. Antioxidant effects were assessed using DPPH, ABTS, and FRAP tests, and the ability to scavenge reactive oxygen species (ROS) was tested in cells with H2O2-provoked oxidative stress. The impact on the activity of antioxidant enzymes (SOD, CAT, GSH) and lipid peroxidation (MDA) was also explored. As shown by Alamar Blue and neutral red uptake assays, the compounds were not toxic within the tested concentration range, and formononetin and coumestrol even demonstrated a stimulatory effect on cells. Coumestrol and biochanin A demonstrated significant antioxidative potential, leading to a significant decrease in ROS in the cells stimulated by H2O2. Furthermore, they influenced enzyme activity, preventing depletion during induced oxidative stress, and also reduced MDA levels, demonstrating protection against lipid peroxidation. Carlina vulgaris is a little-understood plant with unexplored biological potential, and the papers regarding its chemical composition are scarce. In our study, for the first time, the phytochemical profile of the plant, focusing on polar metabolites, was established using modern chromatographic techniques including LC-HRMS-QTOF-CAD, UHPLC-PDA-MS. Phytochemical analysis revealed that the species is a rich source of polyphenolic components, with the most abundant being chlorogenic acid and C-glycosides of luteolin, including carlinoside, orientin, isoorientin, and C-glycosides of apigenin, schaftoside, isoschaftoside, and vitexin. Furthermore, we assessed the impact of the polyphenolic-rich fraction of C. vulgaris extracts on human skin fibroblasts using the MTT and NR assays. It was found that the extract was non-toxic and exhibited potent antioxidant activity in the cells subjected to induced oxidative stress. Additionally, it effectively protected the cells against H2O2-induced cytotoxicity. Our study contributes to the general trend of searching for new phytotherapeutics with potential applications in pharmacy and medicine. The period of middle adulthood presents people with self-actualization challenges related, among others, to their religious beliefs and sexual satisfaction. The aim of this study was to explore the associations between religiosity, nonconformity and sexual satisfaction, as well as to examine whether nonconformity moderates the relationship between religiosity and sexual satisfaction in middle adulthood. The following questionnaires were used: the Religious Meaning System Questionnaire, the Sexual Satisfaction Questionnaire, and the Creative Behavior Questionnaire CBQIII. A series of regression analyses using Hayes PROCESS macro was run to test the models in a sample of 667 middle adults aged between 35 and 55 (M = 42.62; SD = 5.37). The analyses revealed significant relationships between the variables. The religious meaning system showed negative correlations with both nonconformity and sexual satisfaction, whereas nonconformity was positively correlated with sexual satisfaction. An interaction effect of religiosity and nonconformity on sexual satisfaction, when controlling for gender and age, was also demonstrated. More specifically, religiosity was negatively related to sexual satisfaction in individuals with a high level of nonconformity. By contrast, for low and average levels of nonconformity, the relationship between religiosity and sexual satisfaction was insignificant. Phlebotropic flavonoids, including diosmin and its aglycone diosmetin, are natural polyphenols widely used in the prevention and treatment of chronic venous insufficiency (CVI). As oxidative stress plays an important role in the development of pathophysiology of the cardiovascular system, the study aimed to investigate the protective effects of diosmin and diosmetin on hydrogen peroxide (H2O2)-induced oxidative stress in endothelial cells. The cells were pretreated with different concentrations of the flavonoid prior to the H2O2 exposure. The cell viability, the level of intracellular reactive oxygen species (ROS), the activity of cellular antioxidant enzymes—including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase GPx—and the malondialdehyde (MDA) level were assessed. It was found that the H2O2-induced oxidative stress was ameliorated by diosmin/diosmetin in a concentration-dependent manner. The flavonoids restored the activity of cellular antioxidant enzymes and lowered the MDA level upregulated by the H2O2 exposure. These results indicate that diosmin and diosmetin may prevent oxidative stress in endothelial cells; therefore, they may protect against the development and progression of oxidative-stress-related disorders. In this article, we have presented the development and validation of a rapid and sensitive reversed-phase liquid chromatography with tandem mass spectrometry (LC-MS/MS) method for the determination of vincristine (VCR) in patient serum samples. Chromatographic separation was achieved on a Kinetex® (Singapore) column using a mobile phase consisting of 25 mM acetic acid and 0.3% formic acid (A) and methanol (B) in a gradient elution mode at a flow rate of 0.3 mL/min. The VCR and internal standard (vinblastine) were monitored using the multiple reaction monitoring mode under positive electrospray ionization. The lower limit of quantification (LLOQ) was 0.67 ng/mL, and the upper limit of quantification (ULOQ) was 250 ng/mL for VCR. The calculated values of LOD and LOQ for VCR were 0.075 and 0.228 ng/mL, respectively. The calibration curve was linear over the VCR concentration range of 1.0–250 ng/mL in serum. The intra- and inter-day precision and precision were within the generally accepted criteria for the bioanalytical method (<15%). Omission of frequency-dependent hydraulic resistance (skin friction) during modelling of the water hammer phenomenon is unacceptable. This resistance plays a major role when the transient liquid flow occurs in rigid-walled pipes (steel, copper, etc.). In the literature, there are at least two different modelling approaches to skin friction. The first group consists of models based on instantaneous changes in local and convective velocity derivatives, and the second group are models based on the convolution integral and full history of the flow. To date, more popular models are those from the first group, but their use requires empirical coefficients. The second group is still undervalued, even if based on good theoretical foundations and does not require any empirical coefficients. This is undoubtedly related to the calculation complexity of the convolution integral. In this work, a new improved effective solution of this integral is further validated, which is characterised with the use of a simplified weighting function consisting of just two exponential terms. This approach speeds the numerical calculations of the basic flow parameters (pressure and velocity) significantly. Presented comparisons of calculations using the new procedure with experimental pressure runs show the usefulness of the proposed solution and prove that it maintains sufficient accuracy. This paper experimentally and numerically investigates the water hammer phenomenon in serially connected steel and HDPE pipes with different diameters. The aim of the laboratory tests was to obtain the time history of the pressure head at the downstream end of the pipeline system. Transient tests were conducted on seven different pipeline system configurations. The experimental results show that despite the significantly smaller diameter of the HDPE pipe compared to the steel pipe, introducing an HDPE section makes it possible to suppress the valve-induced pressure surge. By referring to the results of the experimental tests conducted, the comparative numerical calculations were performed using the fixed-grid method of characteristics. To reproduce pressure wave attenuation in a steel pipe, Brunone-Vitkovský instant acceleration-based model of unsteady friction was used. To include the viscoelastic behavior of the HDPE pipe wall, the one-element Kelvin–Voigt model was applied. By calibrating the unsteady friction coefficient and creep parameters, satisfactory agreement between the calculated and observed data was obtained. The calibrated values of parameters for a single experimental test were introduced in a numerical model to simulate the remaining water hammer runs. It was demonstrated that using the same unsteady friction coefficient and creep parameters in slightly different configurations of pipe lengths can be effective. Rapid transients are particularly dangerous in industrial hydro-transport systems, where solid-liquid mixtures are transported via long pressure pipelines. A mathematical description of such flow is difficult due to the complexity of phenomena and difficulties in determining parameters. The main aim of the study was to examine the influence of the simplified mixture density and wave celerity description on satisfactory reproduction of pressure characteristics during the transient flow of slurry at low concentrations. The paper reports and discusses the selected aspects of experimental and numerical analyses of transient slurry flow in a polyethylene pipe. The experiments were conducted by using the physical model of a slurry's transportation pressure. The aim of the experiments was to determine the wave celerity during a transient flow in slurries. A low concertation of slurries, which was used during experiments, is typical for one of the biggest slurry networks in Poland. A comparison of the effects of different wave celerity descriptions was performed. The research reported that the theoretical formulas for slurry wave celerity and mixture density were not sufficiently accurate to obtain satisfactory compliance between calculations and observations. To improve the model, the experimental values of wave celerity and the concept of equivalent mixture density have been applied to indirectly consider the influence of variable mixture parameters. With such modifications, the calculated pressure characteristics in all analyzed episodes demonstrated satisfactory compliance with observations. The case study presents the results of numeric simulations of deformations of Vistula River bed downstream the Włocławek Dam, with and without development of the downstream dam cascade. Calculations were performed using a one-dimensional flow model MIKE11 with a river transport module. Using synthetic inflow hydrographs, predictions were performed for 39 year period (2016–2055). This study analyzes the possibilities of using an irrigation sluice gate in submerged conditions to measure water flow rate. Hydraulic experiments on sluice gate discharge capacity were performed on a model made on a 1:2 scale. Measurements were taken for the submerged flow of the sluice gate. Nomograms and relationships for discharge coefficients of the analyzed sluice gate were developed. The possibility of using the existing nomogram for discharge capacity of the submerged sluice gate to determine the discharge capacity of the modeled gate was also investigated. The effect of narrowing of the sluice gate cross-section resulting from different mounting techniques on its capacity was explained. This paper concerns the start-up process of a hydrostatic transmission with a fixed displacement pump, with particular emphasis on dynamic surplus pressure. A numerically controlled transmission using a proportional directional valve was analysed by simulation and experimental verification. The transmission is controlled by the throttle method, and the variable resistance is the throttling gap of the proportional spool valve. A mathematical description of the gear start-up process was obtained using a lumped-parameters model based on ordinary differential equations. The proportional spool valve was described using a modified model, which significantly improved the performance of the model in the closed-loop control process. After assuming the initial conditions and parameterization of the equation coefficients, a simulation of the transition start-up was performed in the MATLAB–Simulink environment. Simulations and experimental studies were carried out for control signals of various shapes and for various feedback from the hydraulic system. The pressure at the pump discharge port and the inlet port of the hydraulic motor, as well as the rotational speed of the hydraulic motor, were analysed in detail as functions of time. Despite significant advances in early diagnosis and treatment, cancer is one of the leading causes of death. Photodynamic therapy (PDT) is a therapy for the treatment of many diseases, including cancer. This therapy uses a combination of a photosensitizer (PS), light irradiation of appropriate length and molecular oxygen. The photodynamic effect kills cancer cells through apoptosis, necrosis, or autophagy of tumor cells. PDT is a promising approach for eliminating various cancers but is not yet as widely applied in therapy as conventional chemotherapy. Currently, natural compounds with photosensitizing properties are being discovered and identified. A reduced toxicity to healthy tissues and a lower incidence of side effects inspires scientists to seek natural PS for PDT. In this review, several groups of compounds with photoactive properties are presented. The use of natural products has been shown to be a fruitful approach in the discovery of novel pharmaceuticals. This review focused on the anticancer activity of furanocoumarins, polyacetylenes, thiophenes, tolyporphins, curcumins, alkaloid and anthraquinones in relation to the light-absorbing properties. Photodynamic therapy (PDT) is a non-invasive and modern form of therapy. It is used in the treatment of non-oncological diseases and more and more often in the treatment of various types of neoplasms in various locations including bladder cancer. The PDT method consists of local or systemic application of a photosensitizer, i.e., a photosensitive compound that accumulates in pathological tissue. Light of appropriate wavelength is absorbed by the photosensitizer molecules, which in turn transfers energy to oxygen or initiates radical processes that leads to selective destruction of diseased cells. The technique enables the selective destruction of malignant cells, as the photocytotoxicity reactions induced by the photosensitizer take place strictly within the pathological tissue. PDT is known to be well tolerated in a clinical setting in patients. In cited papers herein no new safety issues were identified. The development of anti-cancer PDT therapies has greatly accelerated over the last decade. There was no evidence of increased or cumulative toxic effects with each PDT treatment. Many modifications have been made to enhance the effects. Clinically, bladder cancer remains one of the deadliest urological diseases of the urinary system. The subject of this review is the anti-cancer use of PDT, its benefits and possible modifications that may lead to more effective treatments for bladder cancer. Bladder cancer, if localized, would seem to be a good candidate for PDT therapy since this does not involve the toxicity of systemic chemotherapy and can spare normal tissues from damage if properly carried out. It is clear that PDT deserves more investment in clinical research, especially for plant-based photosensitizers. Natural PS isolated from plants and other biological sources can be considered a green approach to PDT in cancer therapy. Currently, PDT is widely used in the treatment of skin cancer, but numerous studies show the advantages of related therapeutic strategies that can help eliminate various types of cancer, including bladder cancer. PDT for bladder cancer in which photosensitizer is locally activated and generates cytotoxic reactive oxygen species and causing cell death, is a modern treatment. In pressurised pipeline systems, various water hammer events commonly occur. This phenomenon can cause extensive damage or even lead to a failure of the pumping system. The aim of this work is to experimentally re-examine the possibility of using an additional polymeric pipe, installed at the downstream end of the main pipeline, to control water hammer. A previous study on this topic investigated additional polymeric pipes connected to the hydraulic system with a short joint section of the same diameter as the main pipeline. In the current research, a different method of including an additional pipe was considered which involved connecting it with a pipe of a smaller diameter than the main pipeline. Three additional HDPE pipes, with different volumes, were investigated. The performance of the devices was studied for hydraulic transients induced by both rapid and slow, manual valve closures. Experimental results show that the additional polymeric pipe can provide significant pressure surge damping during rapid water hammer events. As the valve closing time lengthens, the influence of the additional pipe on the maximum pressure increase is reduced. The additional HDPE pipe does not provide notable protection against hydraulic transients induced by slow valve closure in terms of reducing the first pressure peak. No relationship between the volume of the additional pipe and the damping properties was noticed. The observed pressure oscillations were used to evaluate a one-dimensional numerical model, in which an additional pipe is described as a lumped parameter of the system. The viscoelastic properties of the device were included using the one element Kelvin–Voigt model. Transient flow equations were solved with the implicit method of characteristics. Calculation results demonstrate that this approach allows one to reasonably reproduce unsteady flow oscillations registered during experiments in terms of the maximum pressure increase and pressure wave oscillation period. Most of today's water supply systems are based on plastic pipes. They are characterized by the retarded strain (RS) that takes place in the walls of these pipes. The occurrence of RS increases energy losses and leads to a different form of the basic equations describing the transient pipe flow. In this paper, the RS is calculated with the use of convolution integral of the local derivative of pressure and creep function that describes the viscoelastic behavior of the pipe-wall material. The main equations of a discrete bubble cavity model (DBCM) are based on a momentum equation of two-phase vaporous cavitating flow and continuity equations written initially separately for the gas and liquid phase. In transient flows, another important source of pressure damping is skin friction. Accordingly, the wall shear stress model also required necessary modifications. The final partial derivative set of equations was solved with the use of the method of characteristics (MOC), which transforms the original set of partial differential equations (PDE) into a set of ordinary differential equations (ODE). The developed numerical solutions along with the appropriate boundary conditions formed a basis to write a computer program that was used in comparison analysis. The comparisons between computed and measured results showed that the novel modified DBCM predicts pressure and velocity waveforms including cavitation and retarded strain effects with an acceptable accuracy. This article introduces a flow controller for an upstream water head designed for pipe culverts used in drainage ditches or wells. The regulator is applicable to water flow rates in the range of Qmin < Q < Qmax and the water depth H0, exceeding which causes the gate to open. Qmin flow denotes the minimum flow rate that allows water to accumulate upstream of the controller. Above the maximum flow rate Qmax, the gate remains in the open position. In the present study, the position of the regulator's gate axis was related to the water depth H0 in front of the device. Derived dependencies were verified in hydraulic experiments. Water flow in channels with a compound cross-section involves an exchange of water mass and momentum between the slower flowing water in the floodplains and the faster water in the main channel. This process is called the streams interaction. As a result, the water velocity in the main channel decreases, and at the same time the velocity and depth of flow increase in the part of the floodplains adjacent to the main channel. Diversification of the surface roughness of the main channel and floodplains significantly affects the form of interactions. The results of laboratory experiments were used to characterize the influence of interactions on the discharge capacity of the channel with diversified roughness. The reduction in velocity of the main channel caused by the stream interactions is described with the apparent friction coefficients introduced at the boundary between the main channel and the floodplain. The obtained values of resistance coefficients, supplemented with the values from experiments reported in the literature, were used to establish a relationship useful in assessing the discharge capacity of such channels. It is well known that the water hammer phenomenon can lead to pipeline system failures. For this reason, there is an increased need for simulation of hydraulic transients. High-density polyethylene (HDPE) pipes are commonly used in various pressurised pipeline systems. Most studies have only focused on water hammer events in a single pipe. However, typical fluid distribution networks are composed of serially connected pipes with various inner diameters. The present paper aims to investigate the influence of sudden cross-section changes in an HDPE pipeline system on pressure oscillations during the water hammer phenomenon. Numerical and experimental studies have been conducted. In order to include the viscoelastic behaviour of the HDPE pipe wall, the generalised Kelvin–Voigt model was introduced into the continuity equation. Transient equations were numerically solved using the explicit MacCormack method. A numerical model that involves assigning two values of flow velocity to the connection node was used. The aim of the conducted experiments was to record pressure changes downstream of the pipeline system during valve-induced water hammer. In order to validate the numerical model, the simulation results were compared with experimental data. There is an urgent need to seek new molecular biomarkers helpful in diagnosing and treating breast cancer. In this elaboration, we performed a molecular analysis of mutations and expression of genes within the PI3K/Akt/mTOR pathway in patients with ductal breast cancer of various malignancy levels. We recognized significant correlations between the expression levels of the studied genes. We also performed a bioinformatics analysis of the data available on the international database TCGA and compared them with our own research. Studies on mutations and expression of genes were conducted using High-Resolution Melt PCR (HRM-PCR), Allele-Specific-quantitative PCR (ASP-qPCR), Real-Time PCR molecular methods in a group of women with ductal breast cancer. Bioinformatics analysis was carried out using web source Ualcan and bc-GenExMiner. In the studied group of women, it was observed that the prevalence of mutations in the studied PIK3CA and AKT1 genes was 29.63%. It was stated that the average expression level of the PIK3CA, PIK3R1, PTEN genes in the group of breast cancer patients is lower in comparison to the control group, while the average expression level of the AKT1 and mTOR genes in the studied group was higher in comparison to the control group. It was also indicated that in the group of patients with mutations in the area of the PIK3CA and AKT1 genes, the PIK3CA gene expression level is statistically significantly lower than in the group without mutations. According to our knowledge, we demonstrate, for the first time, that there is a very strong positive correlation between the levels of AKT1 and mTOR gene expression in the case of patients with mutations and without mutations. Nowadays films occupy a significant portion of the media products consumed by people. In Russia, cinema is being considered as a means of individual and social transformation, which makes a contribution to the formation of the Russian audience's outlook, including their attitudes towards topical social issues. At the same time, the question of the effectiveness of films' impact remains an open question in psychological science. According to the empirical orientation of our approach to the study of mass media influence, our goal was to obtain new data on the positive impact of films based on specific experimental research. The task was to identify changes in the attitudes of young people, as the most active viewers, towards topical social issues after watching a specifically selected film. Using a psychosemantic technique that included 25 scales designed to identify attitudes towards elderly people, respondents evaluated their various characteristics before and after watching the film. Using a number of characteristics related to the motivational, emotional and cognitive spheres, significant changes were revealed. At the same time, significant differences were found in assessments of the elderly between undergraduate students and postgraduate students. After watching the film, postgraduate students' attitudes towards elderly people changed in a positive way, while undergraduate students' negative assessments only worsened. The revealed opposite trends can be explained by individual differences of respondents, which include age, educational status as an indicator of individual psychological characteristics, the experience of interaction with elderly people and, as a result, attitudes towards elderly people at the time before watching the movie. The finding that previous attitudes mediate the impact of the film complements the ideas of the contribution of individual differences to media effects. Most of the changes detected immediately after watching the movie did not remain over time. A single movie viewing did not have a lasting effect on viewers' attitudes, and it suggests the further task of identifying mechanisms of the sustainability of changes. [ABSTRACT FROM AUTHOR]

The article is devoted to the assessment of the network community as a collective subject, as a group of interconnected and interdependent persons performing joint activities. According to the main research hypothesis, various forms of group subjectness, which determine its readiness for joint activities, are manifested in the discourse of the network community. Discourse constitutes a network community, mediates the interaction of its participants, represents ideas about the world, values, relationships, attitudes, sets patterns of behavior. A procedure is proposed for identifying discernible traces of the subjectness of a network community at various levels (lexical, semantic, content-analytical scales, etc.). The subjective structure of the network community is described based on experts' implicit representations. The revealed components of the subjectness of network communities are compared with the characteristics of the subjectness of offline social groups. It is shown that the structure of the subjectness of network communities for some components is similar to the structure of the characteristics of the subjectness of offline social groups: the discourse of the network community represents a discussion of joint activities, group norms, and values, problems of civic identity. The specificity of network communities' subjectness is revealed, which is manifested in the positive support of communication within the community, the identification and support of distinction between "us" and "them". Two models of the relationship between discursive features and the construct "subjectness" are compared: additive-cumulative and additive. The equivalence of models is established based on the discriminativeness and the level of consistency with expert evaluation by external criteria. [ABSTRACT FROM AUTHOR]

The relationship between dietary intake and body composition changes during cancer treatment has not been well characterized. The aim of this study was to compare dietary intake at diagnosis and end of treatment in relation to changes in muscle mass and adiposity in head and neck cancer (HNC) patients. Dietary intakes (three-day food record) and body composition using computed tomography (CT) were assessed at diagnosis (baseline) and after treatment completion (post-treatment). Skeletal muscle (SM) loss was explored as a consequence of energy and protein intake in relation to the minimum and maximum European Society of Parenteral and Enteral Nutrition (ESPEN) guidelines. Higher energy intakes (kcal/kg/day) and increases in energy intake (%) from baseline to post-treatment were correlated with attenuated muscle loss (r = 0.62, p < 0.01; r = 0.47, p = 0.04, respectively). Post-treatment protein intake demonstrated a weak positive correlation (r = 0.44, p = 0.05) with muscle loss, which did not persist when controlling for covariates. Meeting minimum ESPEN energy guidelines (25 kcal/kg/day) did not attenuate SM loss, whereas intakes >30 kcal/kg/day resulted in fewer participants losing muscle. Greater baseline adiposity correlated with greater SM loss (p < 0.001). Energy intakes of 30 kcal/kg/day may be required to protect against SM loss during treatment in HNC patients. Background: The natural compounds have been researched extensively as an alternative to the conventional chemotherapy and radiation. Stilbene derivatives appear as a group of therapeutics which deserves special attention. The present study was designed to analyze the effects of stilbene derivatives on drug resistant human leukemic cells. The aim of this work was to evaluate the apoptotic effect of stilbene derivatives in various concentrations on leukemic cells (LC) with and without resistant phenotype. Methods: Human acute promyelocytic leukemia (APL) cell lines (HL60, HL60/MX1, HL60/MX2) and acute lymphoblastic leukemia (ALL) cell lines (CEM/C1, CCRF-CEM) were studied. T-resveratrol, piceatannol, rhaponticin, deoxyrhaponticin, pterostilbene were used to stimulate apoptosis. Mitoxantrone (MIT) was applied to induce drug resistance. Results:t-Resveratrol (RES), deoxyrhaponticin (D-RHAP), rhaponticin (RHAP), pterostilbene (PTER), and piceatannol (PIC) influenced viability and induced apoptosis in all investigated cell lines. Conclusions: Our results confirmed that RES, PIC, RHAP, D-RHAP, and PTER are essential therapeutic compounds with anticancer activity exhibited by induction of apoptosis in leukemic cells with and without resistant phenotype. Stilbene-induced apoptosis in HL60/MX1, HL60/MX2, CEM/C1, and CCRF-CEM leukemia cell lines have been presented in very few studies so far and our research is an important contribution to the investigation of these substances. Background: The insensitivity of cancer cells to therapeutic agents is considered to be the main cause of failure of therapy and mortality of patients with cancer. A particularly important problem in these patients is the phenomenon of multidrug resistance, consisting of abnormal, elevated expression of transport proteins (ABC family). The aim of this research included determination of IC50 values of selected furanocoumarins in the presence and absence of mitoxantrone in leukemia cells and analysis of changes in apoptosis using anexinV/IP and Casp3/IP after 24 h exposure of cell lines to selected coumarins in the presence and absence of mitoxantrone in IC50 concentrations. Methods: Research was conducted on 3 cell lines derived from the human hematopoietic system: HL-60, HL-60/MX1 and HL-60/MX2. After exposure to coumarin compounds, cells were subjected to cytometric analysis to determine the induction of apoptosis by two methods: the Annexin V test with propidium iodide and the PhiPhiLux-G1D2 reagent containing caspase 3 antibodies. Results: All of the furanocoumarin derivatives studied were found to induce apoptosis in leukemia cell lines. Conclusions: Our results clearly show that the furanocoumarin derivatives are therapeutic substances with antitumor activity inducing apoptosis in human leukemia cells with phenotypes of resistance.
