The present study was conducted to evaluate the analytical precision of finite element analysis models of wind turbine bearings. In the finite element analysis models, balls were modeled as finite element meshes as a solid model or replaced by nonlinear springs as two kinds of spring models. In addition, test bench modeling was performed to calculate the displacement following the application of a turnover moment by means of a global model analysis and to calculate the contact stress by means of a sub-model analysis. The comparison of the results of the finite element analyses with the results of the bearing bench test showed that the analytical precision was 17% in the single-spring model, 9% in the Daidie spring model, and 3% in the finite element mesh ball model, indicating that the finite element mesh ball model exhibited the highest precision. HMGB1 is a prototypical danger-associated molecular pattern (DAMP) molecule that co-localizes with amyloid beta (Aβ) in the brains of patients with Alzheimer's disease. HMGB1 levels are significantly higher in the cerebrospinal fluid of patients. However, the cellular and subcellular distribution of HMGB1 in relation to the pathology of Alzheimer's disease has not yet been studied in detail. Here, we investigated whether HMGB1 protein levels in brain tissue homogenates (frontal cortex and striatum) and sera from Tg-APP/PS1 mice, along with its cellular and subcellular localization in those regions, differed. Total HMGB1 levels were increased in the frontal cortices of aged wildtype (7.5 M) mice compared to young (3.5 M) mice, whereas total HMGB1 levels in the frontal cortices of Tg-APP/PS1 mice (7.5 M) were significantly lower than those in age-matched wildtype mice. In contrast, total serum HMGB1 levels were enhanced in aged wildtype (7.5 M) mice and Tg-APP/PS1 mice (7.5 M). Further analysis indicated that nuclear HMGB1 levels in the frontal cortices of Tg-APP/PS1 mice were significantly reduced compared to those in age-matched wildtype controls, and cytosolic HMGB1 levels were also significantly decreased. Triple-fluorescence immunohistochemical analysis indicated that HMGB1 appeared as a ring shape in the cytoplasm of most neurons and microglia in the frontal cortices of 9.5 M Tg-APP/PS1 mice, indicating that nuclear HMGB1 is reduced by aging and in Tg-APP/PS1 mice. Consistent with these observations, Aβ treatment of both primary cortical neuron and primary microglial cultures increased HMGB1 secretion in the media, in an Aβ-dose-dependent manner. Our results indicate that nuclear HMGB1 might be translocated from the nucleus to the cytoplasm in both neurons and microglia in the brains of Tg-APP/PS1 mice, and that it may subsequently be secreted extracellularly. We configured a long-distance ranging apparatus to test the principle of dual-comb time-of-flight measurement using ultrashort lasers. Emphasis was given to the evaluation of open-air performance quantitatively in terms of the measurement resolution and stability. The test results revealed that our dual-comb asynchronous optical pulse sampling permits micrometer-resolved ranging with a repeatability of 2.05 μm over a 648 m distance in dry weather conditions. Further atmospheric effects were evaluated in three different weather conditions with corresponding Allan deviations. Finally, the capability of simultaneous determination of multiple targets was verified with the potential of advanced industrial applications, such as manufacturing, surveying, metrology, and geodesy. We present a phase-locked synthetic wavelength interferometer that enables a complete elimination of cyclic errors in absolute distance measurements. With this method, the phase difference between the reference and measurement paths is fed back into a phase lock-in system, which is then used to control the synthetic wavelength and set the phase difference to zero using an external cavity acousto-optic modulator. We validated the cyclic error removal of the proposed phase-locked method by comparing it with the conventional phase-measuring method of the synthetic wavelength interferometer. By analyzing the locked error signal, we achieved a precision of 0.6 mrad in phase without any observed cyclic errors. [ABSTRACT FROM AUTHOR]

Inconel 718 is a well-known nickel-based superalloy used for high-temperature applications. The aim of the present study was to formulate a constitutive equation (CE) that can be used to account for the deformation behavior of Inconel 718. Compression tests were performed using Gleeble 3800, a thermomechanical simulator, at temperatures ranging from 900 to 1200 °C, at strain rates varying from 0.1 to 10/s. Before compression tests, each specimen was rapidly heated to the desired test temperature while the initial contact pressure was kept relatively low. Thus, compression was performed while the temperature of the entire system, including the specimen and the die, was not uniform. Before conducting an upsetting finite element analysis to determine CE parameters, the heating conditions applied in the Gleeble tests were first subjected to a Joule heating analysis, to simulate the temperature distribution in each specimen prior to the compression process. The spatial temperature distribution of the specimen and the die were determined using a Joule heating analysis, and these results were used as input data for the subsequent finite element analysis of the compression process. From this, the parameters in the obtained Hansel–Spittel equation were estimated for each temperature condition, by employing the regression optimization method, which was used to minimize the deviation between experimental and simulated load values. To validate this optimization process, the experimentally measured flow stresses with respect to the strain rate for each temperature condition were compared with the forming load, determined by the finite element analysis of the compression process using the optimized CE obtained in the present study. It was confirmed that when the optimization process was applied, there was a decrease in the root mean square error. The major findings confirmed the validity of the CE optimization method combined with Joule heating analysis for determining the CE's parameters for high-temperature applications. The high mobility group box 1 (HMGB1), a well-known danger-associated molecule pattern (DAMP) molecule, is a non-histone chromosomal protein localized in the nucleus under normal physiological conditions. HMGB1 exhibits diverse functions depending on its subcellular location. In the present study, we investigated the role of HMGB1-induced autophagy in the lipopolysaccharide (LPS)-treated BV2 microglial cell line in mediating the transition between the inflammatory and autophagic function of the nucleotide-binding oligomerization domain-containing 2 (NOD2), a cytoplasmic pattern-recognition receptor. The induction of the microtubule-associated protein 1 light chain 3 (LC3), an autophagy biomarker, was detected slowly in BV2 cells after the LPS treatment, and peak induction was detected at 12 h. Under these conditions, NOD2 level was significantly increased and the binding between HMGB1 and NOD2 and between HMGB1 and ATG16L1 was markedly enhanced and the temporal profiles of the LC3II induction and HMGB1-NOD2 and HMGB1-ATG16L1 complex formation coincided with the cytosolic accumulation of HMGB1. The LPS-mediated autophagy induction was significantly suppressed in BV2 cells after HMGB1 or NOD2 knock-down (KD), indicating that HMGB1 contributes to NOD2-mediated autophagy induction in microglia. Moreover, NOD2-RIP2 interaction-mediated pro-inflammatory cytokine induction and NF-κB activity were significantly enhanced in BV2 cells after HMGB1 KD, indicating that HMGB1 plays a critical role in the modulation of NOD2 function between pro-inflammation and pro-autophagy in microglia. The effects of the cell-autonomous pro-autophagic pathway operated by cytoplasmic HMGB1 may be beneficial, whereas those from the paracrine pro-inflammatory pathway executed by extracellularly secreted HMGB1 can be detrimental. The ligand exchange process on gold nanorods (Au NRs) was explored by using laser desorption/ionization time-of-flight mass spectrometry (LDI-TOF-MS). Cetyltrimethylammonium bromide (CTAB) adsorbed on Au NRs was replaced with alkanethiol derivatives presenting different functional groups. The ligand exchange process was investigated under various conditions, such as in the presence of different functional groups in the ligands and with different concentrations of CTAB. The ligand-exchanged Au NRs were characterized by using a combination of UV–Vis spectroscopy and LDI-TOF-MS. Based on the results, it was revealed that LDI-TOF-MS analysis can provide crucial and distinct information about the degree of ligand exchange on Au NRs. [ABSTRACT FROM AUTHOR]

We report an absolute interferometer configured with a 1 GHz microwave source photonically synthesized from a fiber mode-locked laser of a 100 MHz pulse repetition rate. Special attention is paid to the identification of the repeatable systematic error with its subsequent suppression by means of passive compensation as well as active correction. Experimental results show that passive compensation permits the measurement error to be less than 7.8 μm (1 σ) over a 2 m range, which further reduces to 3.5 μm (1 σ) by active correction as it is limited ultimately by the phase-resolving power of the phasemeter employed in this study. With precise absolute distance ranging capability, the proposed scheme of the photonic microwave interferometer is expected to replace conventional incremental-type interferometers in diverse long-distance measurement applications, particularly for large machine axis control, precision geodetic surveying and inter-satellite ranging in space. Laser ablation of metal thin films draws attention as a fast means of clean micropatterning. In this study, we attempt to remove only the metal thin film layer selectively without leaving thermal damage on the underneath substrate. Specifically, our single-pulse ablation experiment followed by two-temperature analysis explains that selective ablation can be achieved for gold (Au) films of 50–100 nm thickness by the lift-off process induced as a result of vaporization of the titanium (Ti) interlayer with a strong electron–phonon coupling. With increasing the film thickness comparable to the mean free path of electrons (100 nm), the pulse duration has to be taken shorter than 10 ps, as high-temperature electrons generated by the ultrashort pulses transfer heat to the Ti interlayer. Matrix-assisted laser desorption/ionization (MALDI) has been considered as one of the most powerful analytical tools for mass spectrometry (MS) analysis of large molecular weight compounds such as proteins, nucleic acids, and synthetic polymers thanks to its high sensitivity, high resolution, and compatibility with high-throughput analysis. Despite these advantages, MALDI cannot be applied to MS analysis of small molecular weight compounds (<500 Da) because of the matrix interference in low mass region. Therefore, numerous efforts have been devoted to solving this issue by using metal, semiconductor, and carbon nanomaterials for MALDI time-of-flight MS (MALDI-TOF-MS) analysis instead of organic matrices. Among those nanomaterials, graphene oxide (GO) is of particular interest considering its unique and highly tunable chemical structures composed of the segregated sp2 carbon domains surrounded by sp3 carbon matrix. Chemical modification of GO can precisely tune its physicochemical properties, and it can be readily incorporated with other functional nanomaterials. In this review, the advances of GO derivatives and their nanohybrid structures as alternatives to organic matrices are summarized to demonstrate their potential and practical aspect for MALDI-TOF-MS analysis of small molecules. Multilateration tracking systems (MLTSs) are used in industrial three-dimensional (3D) coordinate measuring applications. For high-precision measurement, system parameters must be calibrated properly in advance. For an MLTS using absolute distance measurement (ADM), the conventional self-calibration method significantly reduces estimation efficiency because all system parameters are estimated simultaneously using a complicated residual function. This paper presents a novel self-calibration method that optimizes ADM to reduce the number of system parameters via highly precise and separate estimations of dead paths. Therefore, the residual function to estimate the tracking station locations can be simplified. By applying a suitable mathematical procedure and solving the initial guess problem without the aid of an external device, estimation accuracy of the system parameters is significantly improved. In three self-calibration experiments, with ADM repeatability of approximately 3.4 µm, the maximum deviation of the system parameters estimated by the proposed self-calibration method was 68.6 µm, while the maximum deviation estimated by the conventional self-calibration method was 711.9 µm. Validation of 3D coordinate measurements in a 1000 mm × 1000 mm × 1000 mm volume showed good agreement between the proposed ADM-based MLTS and a commercial laser tracker, where the maximum difference based on the standard deviation was 17.7 µm. Conversely, the maximum difference was 98.8 µm using the conventional self-calibration method. These results confirmed the efficiency and feasibility of the proposed self-calibration method. In ischemic stroke, neutrophils infiltrate damaged brain tissue immediately following the ischemic insult and aggravate inflammation via various mechanisms which include neutrophil extracellular traps (NETs) formation. In the present study, we showed that adenosine triphosphate (ATP), a DAMP molecule, accumulates in the brain and induces NETosis in brain parenchyma and in circulating neutrophils (PMNs) isolated from a murine model of stroke induced by middle cerebral artery occlusion (MCAO). Expression of peptidylarginine deiminase-4 (PAD4), which induces citrullination of histones H3 (CitH3) and initiates NETosis, was significantly enhanced in brain parenchyma and blood PMNs following MCAO. ATP or BzATP (a prototypic P2X7R agonist) significantly enhanced the inductions of PAD4 and CitH3 in a P2X7R-dependent manner and intracellular Ca2+ influx, PKCα activation, and NADPH oxidase-dependent reactive oxygen species (ROS) production play critical roles in this ATP-P2X7R-mediated NETosis. In our MCAO animal model, NETosis was markedly suppressed by treatment with apyrase, an enzyme hydrolyzing ATP, but enhanced by co-treatment of BzATP, confirming ATP-P2X7R-mediated NETosis. Since ATP not only induced NETosis but was also extruded after NETosis, our results indicate that ATP accumulated in the ischemic brain induces NETosis, mediating a cross-talk linking NETosis with neuronal damage that might aggravate inflammation and brain damage. Neutrophil extracellular traps (NETs) comprise decondensed chromatin, histones and neutrophil granular proteins and are involved in the response to infectious as well as non-infectious diseases. The prothrombotic activity of NETs has been reported in various thrombus-related diseases; this activity can be attributed to the fact that the NETs serve as a scaffold for cells and numerous coagulation factors and stimulate fibrin deposition. A crosstalk between NETs and thrombosis has been indicated to play a role in numerous thrombosis-related conditions including stroke. In cerebral ischemia, neutrophils are the first group of cells to infiltrate the damaged brain tissue, where they produce NETs in the brain parenchyma and within blood vessels, thereby aggravating inflammation. Increasing evidences suggest the connection between NETosis and thrombosis as a possible cause of "tPA resistance", a problem encountered during the treatment of stroke patients. Several damage-associated molecular pattern molecules have been proven to induce NETosis and thrombosis, with high mobility group box 1 (HMGB1) playing a critical role. This review discusses NETosis and thrombosis and their crosstalk in various thrombosis-related diseases, focusing on the role of HMGB1 as a mediator in stroke.
