4-methyl-5-vinylthiazole (MVT) is a significant volatile of caladium (Caladium bicolor) which produces a very high level of thiamin thiazole synthase (THI4) in male flowers. We explored transcriptomes upregulating MVT using RNA-seq during the six developmental stages of the male flower (Day−10 to Day0) in C. bicolor 'Tapestry'. THI4 was the highest transcript throughout the male flower development. Additionally, the genes showing the high expression associated with floral volatiles of caladium on Day0 were trans-resveratrol di-O-methyltransferase (ROMT), chalcone synthase (CHS), 3-ketoacyl-CoA thiolase 2 (KAT2), and linalool synthase (TPS). These four genes correspond to the following elevated volatiles of caladium: 1,3,5-trimethoxybenzene, MVT, indole, methyl salicylate, and linalool on Day0 compared to Day−10. The upstream THI4 gene was cloned to drive a fluorescent gene (ZsGreen1) in transient and stable transgenic petunia and tobacco plants, showing the gene expression only in the male tissue. The tissue-specific expression of the caladium THI4 promoter could benefit crop production with minimal modification of plants. The study of gender markers is essential in forensic genetic analysis. Mutations in the X or Y homologs of the amelogenin gene can be misleading, resulting in serious mistakes in forensic genetic analysis. We recently discovered two male cases of the X homolog of the amelogenin (AMELX) allelic dropout while analyzing short tandem repeat genotypes obtained from crime scene evidence. Subsequently, we evaluated the molecular characteristics of AMELX allelic dropout in this study. We used two previously reported amelogenin primers to verify a half level of amelogenin gene amplification intensity in the two male cases, which we confirmed was caused by AMELX allelic dropout. We then characterized the point mutation using Sanger sequencing and designed mutation-specific primers that could overcome AMELX allelic dropout. Short tandem repeat genotyping analysis confirmed that the AMELX allelic dropout was recovered by the mutation-specific primer designed specifically for this case. The sequencing of the AMELX allele revealed a single-point variant from A→G at base position 7 downstream from the 3′ end in the amelogenin forward primer-binding region. This point mutation was identically found in two different male cases, resulting in AMELX allelic dropout. To our knowledge, these mutations and the X homolog amplification failure of amelogenin have not been reported in the Korean population. Patients with obstructive sleep apnea (OSA) exhibit a high prevalence of pulmonary hypertension and right ventricular (RV) hypertrophy. However, the exact molecule responsible for the pathogenesis remains unknown. Given the resistance to RV dilation observed in transient receptor potential canonical 3(Trpc3)−/− mice during a pulmonary hypertension model induced by phenylephrine (PE), we hypothesized that TRPC3 also plays a role in chronic intermittent hypoxia (CIH) conditions, which lead to RV dilation and dysfunction. To test this, we established an OSA mouse model using 8- to 12-week-old 129/SvEv wild-type and Trpc3−/− mice in a customized breeding chamber that simulated sleep and oxygen cycles. Functional parameters of the RV were evaluated through analysis of cardiac cine magnetic resonance images, while histopathological examinations were conducted on cardiomyocytes and pulmonary vessels. Following exposure to 4 weeks of CIH, Trpc3−/− mice exhibited significant RV dysfunction, characterized by decreased ejection fraction, increased end-diastole RV wall thickness, and elevated expression of pathological cardiac markers. In addition, reactive oxygen species (ROS) signaling and the endothelin system were markedly increased solely in the hearts of CIH-exposed Trpc3−/− mice. Notably, no significant differences in pulmonary vessel thickness or the endothelin system were observed in the lungs of wild-type (WT) and Trpc3−/− mice subjected to 4 weeks of CIH. In conclusion, our findings suggest that TRPC3 serves as a regulator of RV resistance in response to pressure from the pulmonary vasculature, as evidenced by the high susceptibility to RV dilation in Trpc3−/− mice without notable changes in pulmonary vasculature under CIH conditions. Simple Summary: This study developed a prediction model for late bladder toxicity in patients with uterine cervical cancer undergoing radiation therapy. A deep learning (DL) model was trained on data from 281 patients and compared its performance with a multivariable logistic regression model. The DL model outperformed the regression model, achieving higher accuracy, recall, F1-score, and area under the receiver operating characteristic curve. Specifically, based on the feature importance analysis, the DL model identified the doses for the most exposed 2 cc volume of the bladder (BD2cc), BD5cc, and ICRU bladder point as high-priority features. Finally, the lightweight DL model, which was designed to focus on the top five important features, demonstrated superior predictive capabilities, highlighting its potential in improving patient outcomes and minimizing treatment-related complications with secured reliability. (2) We collected data obtained from 281 uterine cervical cancer patients who underwent definitive radiation therapy. It achieved an F1-score of 0.76 and an AUROC of 0.81, while the corresponding values for the multivariable logistic regression were 0.14 and 0.43, respectively. The DL model identified the doses for the most exposed 2 cc volume of the bladder (BD2cc) as the most important feature, followed by BD5cc and the ICRU bladder point. In the case of the lightweight DL model, the F-score and AUROC were 0.90 and 0.91, respectively. Numerous pedestrians interact with the subway station space by finding entrances into this closed area to use the subway system; further, they may use transfer transportation facilities or the complex functions nearby, such as commercial. Many studies examine pedestrian behaviors in subway stations, but most focus on special situations such as disasters and evacuation. Because it is important to analyze gait patterns in everyday situations, this study aims to verify the explanatory power of actual gait behavior by using space syntax theory in constructing an optimal agent-based model. To this end, first, pedestrian characteristics and space types are classified using pedestrian data from Gangnam Station. Second, the depthmapX program is used to develop an appropriate agent-based model for stations. Third, a simulation is run to calculate the frequency of the agent movement at each gate, which is matched with the observed pedestrian volume. Fourth, the relationship between the frequency of the agent movement and pedestrian volume is analyzed using Statistical Package for the Social Sciences. The results show that although agent-based models have limitations in explaining pedestrian patterns in the entire subway station, they are capable of explaining these patterns along the shortest paths between ticket gates and station entrances. Urban regeneration by participatory methods is being discussed in many parts of the world, but conflicts between stakeholders emerge as a major challenge. In order to address this problem, a new approach to urban regeneration has been attempted in Korea. By targeting towns with university campuses, this project encourages active participation from university students as well as local residents. As a result of COVID-19 restrictions, the project adopted an online-based communication strategy. First, the online data was collected; second, the data for each participant was classified through data refinement; and third, the data analysis and data visualization were carried out at each stage using program R. The results revealed that the stakeholders exhibited different perceptions about the process, indicating a potential benefit of distinct role division for the success of the multiparty project. The significance of this study lies in the fact that it analyzes participants' perceptions of urban regeneration using a text-mining process. We explore the spatial layouts of mega-shelters and suggest better spatial planning strategies. A mega-shelter for refugees contains multiple functions, such as dormitory, dining, medical, kitchen, storage, and community areas. Post-disaster refugees often suffer from PTSD that affects their mental health and spatial cognitive ability. The spatial configuration of a mega-shelter can accelerate their recovery by providing an environment that not only satisfies the basic needs, but one that can improve their spatial cognitive ability and promote a sense of community in this new, albeit temporary, small society. Four mega-shelters in the U.S., Australia, and Japan were analyzed using space syntax methods, specifically axial line analysis and visibility graph analysis (VGA), as well as justified graph analysis. The comparative analysis shows that while specific spatial layouts are different, all shelters were designed from a manager's perspective. The movements of the refugees were sometimes unnecessarily exposed to supervision and control, and community areas were often found in locations with low accessibility. By incorporating strategies such as siting community space in areas with high global integration values and adopting transition areas, mega-shelters can create an environment that can enhance the refugees' will to recover and rebuild by promoting communications with neighbors and various community activities. Simple Summary: Proton minibeam radiation therapy (pMBRT) using multislit collimator (MSC) and scatterers has been proposed to spare healthy tissues and organs on the beam path and beyond the Bragg peak. An MSC that was much thicker than the maximum range of the proton beam could provide a sufficiently high peak-to-valley dose ratio at the patient's skin, and the scatterers could actively convert the spatially fractionated proton beam to a uniform and broad beam in tumors by changing their thickness. The combination of the MSC and the scatterers can be a good solution for implementing pMBRT in clinical proton therapy facilities. The feasibility of proton minibeam radiation therapy (pMBRT) using a multislit collimator (MSC) and a scattering device was evaluated for clinical use at a clinical proton therapy facility. We fabricated, through Monte Carlo (MC) simulations, not only an MSC with a high peak-to-valley dose ratio (PVDR) at the entrance of the proton beam, to prevent radiation toxicity, but also a scattering device to modulate the PVDR in depth. The slit width and center-to-center distance of the diverging MSC were 2.5 mm and 5.0 mm at the large end, respectively, and its thickness and available field size were 100 mm and 76 × 77.5 mm2, respectively. Spatially fractionated dose distributions were measured at various depths using radiochromic EBT3 films and also tested on bacterial cells. MC simulation showed that the thicker the MSC, the higher the PVDR at the phantom surface. Dosimetric evaluations showed that lateral dose profiles varied according to the scatterer's thickness, and the depths satisfying PVDR = 1.1 moved toward the surface as their thickness increased. The response of the bacterial cells to the proton minibeams' depth was also established, in a manner similar to the dosimetric pattern. Featured Application: This paper's application of the proposed novel method is reducing the ski-slope effect in direct PCR, thus aiding more efficient amplification of DNA obtained from crime scenes. Analytical techniques such as DNA profiling are widely used in various fields, including forensic science, and novel technologies such as direct polymerase chain reaction (PCR) amplification are continuously being developed in order to acquire DNA profiles efficiently. However, non-specific amplification may occur depending on the quality of the crime scene evidence and amplification methods employed. In particular, the ski-slope effect observed in direct PCR amplification has led to inaccurate interpretations of the DNA profile results. In this study, we aimed to reduce the ski-slope effect by using dimethyl sulfoxide (DMSO) in direct PCR. We confirmed that DMSO (3.75%, v/v) increased the amplification yield of large-sized DNA sequences more than that of small-sized ones. Using 50 Korean buccal samples, we further demonstrated that DMSO reduced the ski-slope effect in direct PCR. These results suggest that the experimental method developed in this study is suitable for direct PCR and may help to successfully obtain DNA profiles from various types of evidence at crime scenes. This study presents an experimental seismic investigation of a 1000 kVA cast resin-type hybrid mold transformer through tri-axial shaking table tests. The input acceleration time histories were generated in accordance with the specifications recommended by the International Code Council Evaluation Services Acceptance Criteria ICC-ES AC156 code, with scaling factors in the range of 25–300%. The damage and the dynamic characteristics of the mold transformer were evaluated in terms of the fundamental frequency, damping ratio, acceleration time history responses, dynamic amplification factors, and relative displacement. The shaking table test results showed that the damage of the mold transformer was mainly governed by the severe slippage of the spacers and the loosening of the linked bolts between the bottom beam and the bed beam. In addition, the maximum relative displacement at the top beam in Y and Z-directions exceeded the boundary limit recommended by the Korean National Radio Research Agency. Moreover, the operational test of the specimen was performed based on the IEC 60076-11 Standard before and after the shaking table test series to ensure the operational capacity of the transformer. In the present study, the seismic vulnerability of a hybrid mold transformer was investigated using a dynamic analytical approach incorporating the experimental results of shaking table tests. The analytical model consisted of linear springs and plastic beam elements, and it has six degrees of freedom simulating the hybrid mold transformer. The dynamic characteristics of the analytical model were determined based on the shaking table tests. The reliability of the analytical model was verified by comparing the test results and analytical results. In order to assess the seismic vulnerability, three critical damage states observed during the shaking table tests were investigated by incorporating the three performance levels specified in ASCE 41-17. Comprehensive dynamic analyses were performed with a set of twenty earthquakes in consideration of the variation of the uncertain parameters (such as the effective stiffness and coil mass) of the mold transformer. A proposed countermeasure to COVID-19 is a robust healthcare system that can respond and identify transmission paths using information technology. This involves the use of smart city services for tracking an infected person. However, during the COVID-19 pandemic, the healthcare system could only provide data on the number of infected people. Additionally, smart city services could respond neither timely nor sequentially. This study proposed a method for timely and sequential responses, through a flexible combination of the healthcare system and smart city services by envisioning a scenario that sequentially grafts the current status of COVID-19 in Korea. The results are the following. First, the COVID-19 outbreak was summarized in the context of the healthcare system and current smart city services. A method by which the latter could respond to the various needs of the former was suggested. Second, recommendations on combining or dismissing certain smart city services, as per the needs of coping with COVID-19, were summarized. Third, smart city services must be utilized only for addressing pandemics, as data from the healthcare system consists of personal information. It has been claimed that high-rise apartments, unlike perimeter block housing, cause social pathology; however, no studies have quantitatively proven this. Therefore, this study aimed to analyze the difference between space-use behavior and social interactions with a focus on high-rise apartments, the main residential mode in South Korea, and the newly created perimeter block housing. This was done by first comparing and analyzing the spatial configurations of high-rise apartments and perimeter block housing using the space syntax methodology. Second, the space-use behaviors that affect interaction were explained by regression analysis after analyzing correlations among the spatial configurations of individual residence locations, the frequency of use of community facilities, and interaction. Third, differences in interaction for people living in complexes with different spatial configurations were analyzed using t-tests. The main finding was that people whose living arrangements include a good spatial configuration or that often use children's playgrounds interact more. Additionally, when the spatial configuration of a complex is systematic and the complex is closely connected to the city, the interaction between neighbors appears better. Simple Summary: Pelvic nodal involvement is frequently present in early-stage cervical cancer patients on pretreatment imaging studies. However, it is unclear whether radical chemoradiotherapy (CRT) or radical hysterectomy RH followed by tailored adjuvant radiotherapy is more appropriate in these patients. We compared oncological outcomes of up-front surgery followed by tailored adjuvant radiotherapy and definitive CRT in these patients. We found no differences in outcomes existed between definitive CRT and hysterectomy with tailored adjuvant radiotherapy. However, after surgery, 88.7% of patients required adjuvant radiotherapy. These findings suggest that definitive CRT can avoid unplanned tri-modality therapy without compromising oncologic outcomes. To compare the oncologic outcomes between chemoradiotherapy (CRT) and radical hysterectomy followed by tailored adjuvant therapy in patients with early cervical cancer presenting with pelvic lymph node metastasis. We retrospectively analyzed the medical records of women with early cervical cancer presenting with positive pelvic nodes identified on pretreatment imaging assessment. Propensity score matching was employed to control for the heterogeneity between two groups according to confounding factors. Overall survival, disease-free survival, and pattern of failure were compared between the two groups. A total of 262 patients were identified; among them, 67 received definitive CRT (group A), and 195 received hysterectomy (group B). Adjuvant therapy was administered to 88.7% of group B. There were no significant differences between group A and group B regarding the 5-year overall survival rates (89.2% vs. 89.0%) as well as disease-free survival rates (80.6% vs. 82.7%), and patterns of failure. Distant metastasis was the major failure pattern identified in both groups. In multivariate analysis, non-squamous histology was significantly associated with poorer overall survival. As there are no significant differences in 5-year OS, DFS, and patterns of failure, definitive CRT could avoid the combined modality therapy without compromising oncologic outcomes. The COVID-19 outbreak in 2020 has changed the way people travel due to its highly contagious nature. In this study, changes in the travel behavior of passengers due to COVID-19 in the first half of 2020 were examined. To determine whether COVID-19 has affected the use of transportation by passengers, paired t-tests were conducted between the passenger volume of private vehicles in Seoul prior to and after the pandemic. Additionally, the passenger occupancy rate of different modes of transportation during the similar time periods were compared and analyzed to identify the changes in monthly usage rate for each mode. In the case of private vehicles and public bicycles, the usage rates have recovered or increased when compared to those of before the pandemic. Conversely, bus and rail passenger service rates have decreased from the previous year before the pandemic. Furthermore, it is found that existing bus and rail users have switched to the private auto mode due to COVID-19. Background: In this study, we analyzed the association between antibiotic use and obesity and metabolic syndrome (MS) in a Korean adult population. Methods: Subjects using the Korean National Health Insurance Service sample cohort were retrospectively analyzed in 2015. The differences in obesity and metabolic syndrome (MS) status were compared and analyzed according to duration of systemic antibiotic treatment in the previous 10 years (non-users, 1st, 2nd, and 3rd tertile). Results: Subjects who used systemic antibiotics for longer periods were older, satisfied more criteria for MS, and had more comorbidities than non-users (non-users vs. 3rd tertile, p < 0.0001 for all). After adjusting for confounding factors, the risk of obesity was higher in subjects who used systemic antibiotics for longer periods than in non-users (non-users vs. 3rd tertile, OR (odds ratio) (95% CI (confidence interval)); 1.20 (1.12–1.38)). The criteria for MS were more satisfied in the 3rd tertile than in non-users. A higher obesity risk was also found in subjects treated with antibiotics targeting Gram-negative organisms than in those targeting Gram-positive organisms. Conclusion: The risk of obesity was higher in subjects who took systemic antibiotics more frequently. The risk was more prominent when they took antibiotics targeting Gram-negative bacteria. A decision tree is a well-known machine learning technique. Recently their popularity has increased due to the powerful Gradient Boosting ensemble method that allows to gradually increasing accuracy at the cost of executing a large number of decision trees. In this paper we present an accelerator designed to optimize the execution of these trees while reducing the energy consumption. We have implemented it in an FPGA for embedded systems, and we have tested it with a relevant case-study: pixel classification of hyperspectral images. In our experiments with different images our accelerator can process the hyperspectral images at the same speed at which they are generated by the hyperspectral sensors. Compared to a high-performance processor running optimized software, on average our design is twice as fast and consumes 72 times less energy. MicroRNAs (miRNAs) can be used to target a variety of human malignancies by targeting their oncogenes or tumor suppressor genes. Recent evidence has shown that miRNA-1208 (miR-1208) was rarely expressed in a variety of cancer cells, suggesting the possibility that miR-1208 functions as a tumor suppressor gene. Herein, ectopic expression of miR-1208 induced the accumulation of sub-G1 populations and the cleavage of procaspase-3 and PARP, which could be prevented by pre-treatment with the pan-caspase inhibitor, Z-VAD. In addition, miR-1208 increased the susceptibility to cisplatin and TRAIL in Caki-1 cells. Luciferase reporter assay results showed that miR-1208 negatively regulates TBC1 domain containing kinase (TBCK) expression by binding to the miR-1208 binding sites in the 3′-untranslated region of TBCK. In addition, miR-1208 specifically repressed TBCK expression at the transcriptional level. In contrast, inhibition of endogenous miR-1208 by anti-miRs resulted in an increase in TBCK expression. Downregulation of TBCK induced by TBCK-specific siRNAs increased susceptibility to cisplatin and TRAIL. These findings suggest that miR-1208 acts as a tumor suppressor and targets TBCK directly, thus possessing great potential for use in renal cancer therapy. [ABSTRACT FROM AUTHOR]

Precise and timely evaluation of an individual's hearing loss plays an important role in determining appropriate treatment strategies, including medication and aural rehabilitation. However, currently available hearing assessment systems do not satisfy the need for an objective assessment tool with a simple and non-invasive procedure. In this paper, we propose a new method for pure-tone audiometry, which may potentially be used to assess an individual's hearing ability objectively and quantitatively, without need for the user's active response. The proposed method is based on the auditory oculogyric reflex, where the eyes involuntary rotate towards the source of a sound, in response to spatially moving pure-tone audio stimuli modulated at specific frequencies and intensities. We quantitatively analyzed horizontal electrooculograms (EOG) recorded with a pair of electrodes under two conditions—when pure-tone stimuli were (1) "inaudible" or (2) "audible" to a participant. Preliminary experimental results showed significantly increased EOG amplitude in the audible condition compared to the inaudible condition for all ten healthy participants. Zinc finger with KRAB and SCAN domain 3 (ZKSCAN3) upregulates genes encoding proteins involved in cell differentiation, proliferation and apoptosis. ZKSCAN3 has been reported to be overexpressed in several human cancers such as colorectal cancer and prostate cancer and is proposed as a candidate oncoprotein. However, the molecular mechanism by which ZKSCAN3 participates in carcinogenesis is largely unknown. Here, we evaluated ZKSCAN3 expression in uterine cervical cancers (CC) by immunohistochemistry using formalin-fixed, paraffin-embedded tissues from 126 biopsy samples from 126 patients. The clinicopathological findings were analyzed and compared with ZKSCAN3 expression levels. ZKSCAN3 was strongly overexpressed in CCs compared to adjacent non-neoplastic cervical mucosa tissues. Moreover, a gene copy number assay showed amplified ZKSCAN3 in CC samples. ZKSCAN3 overexpression was also significantly associated with poor overall survival of the patients. Overall, our findings indicate that ZKSCAN3 overexpression is a frequent event in uterine CC and is correlated with a poor clinical outcome.
