Stress on the Endoplasmic reticulum (ER) can severely disrupt cellular function by impairing protein folding and post-translational modifications, thereby leading to the accumulation of poor-quality proteins. However, research on its impact on photosynthesis remains limited. In this study, we investigated the impact of ER stress on the photosynthetic efficiency of Chlamydomonas reinhardtii using pharmacological inducers, tunicamycin (TM) and brefeldin A (BFA), which specifically target the ER. Our measurements of photosynthetic parameters showed that these ER stress-inducing compounds caused a significant decline in photosynthetic efficiency. A proteomic analysis confirmed that TM and BFA effectively induce ER stress, as evidenced by the upregulation of ER stress-related proteins. Furthermore, we observed a widespread downregulation of photosynthesis-related proteins, which is consistent with the results obtained from our measurements of photosynthetic parameters. These findings suggest that the stress on ER has a profound impact on chloroplast function, disrupting photosynthetic processes. This study highlights the critical interdependence between the ER and chloroplasts, and it underscores the broader implications of ER stress on the cellular metabolism and energy efficiency of photosynthetic organisms.

(1) T-2 toxin synthesized by Fusarium oxysporum ( F. oxysporum ) can cause deterioration of dried fish and endanger human health. (2) The molecular mechanism by which antibacterial lipopeptides surfactin inhibited F. oxysporum growth and toxin production was elucidated by investigating the intracellular ROS production pathway and the subcellular distribution and transcriptional activity of the transcription factor Yap1 and its regulation of Tri5 gene in F. oxysporum . (3) Surfactin caused hyphal damage and nucleic acid and protein leakage; thus, the growth of F. oxysporum was disrupted. Surfactin's excessive accumulation of intracellular ROS triggered the translocation of transcription factor Yap1 into the nucleus, resulting in toxin cluster gene Tri5 expression inhibition, thereby blocking T-2 toxin synthesis. (4) This is a novel mechanism by which surfactin inhibits the growth and T-2 toxin synthesis of F. oxysporum from multiple aspects, including cell structural integrity and the ROS- Yap1 signaling pathway. (5) This study provides a theoretical basis for the application of surfactin in the antifungal control of aquatic dry products.

Ti 4+ -doped V 2 O 5 films with nanowires on top and a dense, long nanorod layer on the bottom were successfully fabricated using the spin-coating route. During the electrochromic cycling, charge transfer resistance ( R ct ) decreases while ion-diffusion ability ( K Ω ) rapidly drops in the first ten cycles and then levels off. Low R ct and morphology of nanowires collaboratively improved the electrochromic behavior of Ti 4+ -doped V 2 O 5 films by enhancing the charge transfer speed and minimizing polarization and dissolution. The obtained Ti 4+ -doped V 2 O 5 film shows better electrochromic properties than the undoped V 2 O 5 film, with a coloration efficiency (CE) of 34.15 cm 2 /C, coloration time of 9.00 s, and cyclic retention of 82.6% at cycle 100. In contrast, the corresponding values for the undoped V 2 O 5 film were 23.57 cm 2 /C, 13.16 s, and 43.6%.

There is a growing trend in incorporating biomass-based engineered nanomaterials into food products to enhance their quality and functionality. The zeta potential, droplet size, microstructure, and content of free fatty acid (FFA) release were determined to investigate the influence of a plant-derived particle stabilizer, i.e., lignin-containing cellulose nanofibrils (LCNFs). Remarkable differences were observed during digestion stages, which were found to be correlated with the concentrations of LCNFs. The gradual FFA release in the small intestine stage from LCNF-coated lipid droplets was monitored over time, with a final lowest release of FFAs amounting to 26.3% in the emulsion containing 20.0% ( v / v ) of the dispersed phase stabilized by 3 mg/mL of LCNFs. This release can be attributed to the physical barrier at lipid droplet surfaces and the network effect created by the free LCNFs in the continuous phase. This work provides a foundation for the potential application of nature-derived LCNF materials in reducing fat absorbance.

Perovskite is an advanced optoelectronic semiconductor material that has garnered significant attention in recent years. However, its drawback lies in its environmental instability, limiting its practical applications. To tackle this issue, this research delved into the idea of creating a space-confined structure and used electrospinning to produce a film of perovskite nanocomposite fibers. By effectively encapsulating perovskite nanocrystals into a polymer matrix, the perovskite could be shielded from water and oxygen in the environment, thereby reducing the likelihood of perovskite decomposition and enhancing the stability of its structure and properties. This study examined the influence of material composition and the spinning process on the nanofiber structure to create good spatial confinement. This strategy resulted in a high photoluminescence quantum yield of over 80% and a long-term environmental stability of as long as 1000 h over 90% of the original PLQY. By harnessing the flexibility of the composite fibers, this study demonstrated the potential applications and performance of this nanocomposite film in flexible quantum fluorescence conversion for LED applications.

Optical Coherence Tomography (OCT)-based angiography (OCTA) is a high-resolution, high-speed, and non-invasive imaging method that can provide vascular mapping of subcutaneous tissue up to approximately 2 mm. In dermatology applications of OCTA, handheld probes are always designed with a piece of transparent but solid contact window placed at the end of the probe to directly contact the skin for achieving better focusing between the light source and the tissue, reducing noise caused by minor movements. The pressure between the contact window and the skin is usually uncontrollable, and high external pressure affects the quality of microvascular imaging by compressing the vessels and obstructing the underlying blood flow. Therefore, it is necessary to determine a pressure range to ensure that the vessels can be fully imaged in high-quality images. In this paper, two pressure sensors were added to the existing handheld OCT probe, and the imaging probe was fixed to a metal stand and adjusted vertically to change the pressure between the probe and the tested skin site, a gradient of roughly 4 kPa (with 1-2 kPa error) increase was applied in each experiment, and the impact of pressure to the vessel was calculated. The experiment involved a total of five subjects, three areas of which were scanned (palm, back of the hand, and forearm). The vessel density was calculated to evaluate the impact of external pressure on angiography. In addition, PSNR was calculated to ensure that the quality of different tests was at a similar level. The angiography showed the highest density (about 10%) when the pressure between the contact window on the probe and the test area was between 3 and 5 kPa. As the pressure increased, the vascular density decreased, and the rate of decrease varied in different test areas. After fitting all the data points according to the different sites, the slope of the fitted line, i.e., the rate of decrease in density per unit value of pressure, was found to be 4.05% at the palm site, 6.93% at the back of the hand, and 4.55% at the forearm site. This experiment demonstrates that the pressure between the skin and contact window is a significant parameter that cannot be ignored. It is recommended that in future OCTA data collection processes and probe designs, the impact of pressure on the experiment be considered.

Concrete coatings show significant promise in shielding concrete substrates from corrosion by effectively resisting harmful ions and moisture. Thanks to their practicality, high efficiency, and cost-effectiveness, coatings are considered a potent technique for enhancing the chloride resistance of reinforced concrete structures. Over recent decades, extensive research has concentrated on employing coatings to bolster concrete's ability to withstand chloride penetration. This paper provides a holistic review of the current studies on chloride infiltration in concrete surfaces treated with coating materials, primarily focused on chloride resistance improvement efficiency and chloride transport modeling. Firstly, by comparing the functions of assorted coatings, four inherent protection mechanisms are summarized and elaborated thoroughly. Afterwards, the chloride resistance improvement efficiency of assorted coatings reported in current studies are reviewed and compared in great detail, with a specific focus on inorganic, organic, and organic-inorganic composite coatings. Furthermore, the theoretical research about methodologies for chloride transport behavior prediction is summarized. Finally, this paper outlines the potential research directions in this field and the theoretical, technical, and practical application challenges. This review not only identifies critical areas necessitating further investigation and problem-solving in this domain but also aids in selecting appropriate coating materials and refining corrosion management strategies.

Alpinae oxyphyllae fructus (AOF), the dried mature fruit of Alpinia oxyphylla Miquel of the Zingiberaceae family, shows many special pharmacological effects. In recent years, there has been an abundance of research results on AOF. In this paper, the new compounds isolated from AOF since 2018 are reviewed, including terpenes, flavonoids, diarylheptanoids, phenolic acid, sterols, alkanes, fats, etc. The isolation methods that were applied include the microwave-assisted method, response surface method, chiral high-performance liquid chromatography-multiple reaction monitoring-mass spectrometry (HPLC-MRM-MS) analytical method, ultra-high-performance liquid chromatography-quadrupole-electrostatic field orbitrap high-resolution mass spectrometry (UPLC-Orbitrap-HRMS) method, ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method, hot water leaching method, ethanol leaching method, and so on. Additionally, the pharmacological effects of AOF found from 2018 to 2024 are also summarized, including neuroprotection, regulation of metabolic disorders, antioxidant activity, antiapoptosis, antiinflammatory activity, antidiabetic activity, antihyperuricemia, antiaging, antidiuresis, immune regulation, anti-tumor activity, renal protection, hepatoprotection, and anti-asthma. This paper provides a reference for further research on AOF.

The key components of a polarization-independent electro-optic (EO) interferometer, including the beam splitter, mode converter, and directional coupler, are designed based on a lithium niobate (LN) platform on an integrated insulator to ensure high extinction ratios. By elaborately designing the geometric structure of the multimode interference (MMI) coupler, beam splitting of equal proportions and directional coupling of higher-order modes are realized. The most prominent characteristic of the proposed interferometer is polarization insensitivity, which is realized by converting TM polarization into TE polarization using a mode converter, providing conditions for the study of light with different polarizations. At 1550 nm, the visibility of the interferometer is 97.59% and 98.16% for TE and TM, respectively, demonstrating the high performance of the proposed LN polarization-independent interferometer.

Transition-metal-catalyzed directed C-H functionalization with various carbene precursors has been widely employed for constructing a wide range of complex and diverse active molecules through metal carbene migratory insertion processes. Among various carbene precursors, iodonium ylides serve as a novel and emerging carbene precursor with features including easy accessibility, thermal stability and high activity, which have attracted great attention from organic chemists and have achieved tremendous success in organic transformation. In this review, recent progress on the application of iodonium ylides with multifunctional coupling characteristics in C-H bond activation reactions is summarized, and the potential of iodonium ylides is discussed.

Panama boasts an expansive mangrove area and stands as one of the most biodiverse countries in America. While mangrove plants have long been utilized in traditional medicine, there are still unstudied species whose potential medicinal applications remain unknown. This study aimed to extract bioactive compounds from Mora oleifera (Triana ex Hemsl.) Ducke, an understudied mangrove species. Through bioassay-guided fractionation of the crude extract, we isolated seven active compounds identified as lupenone ( 1 ), lupeol ( 2 ), α-amyrin ( 3 ), β-amyrin ( 4 ), palmitic acid ( 5 ), sitosterol ( 6 ), and stigmasterol ( 7 ). Compound structures were determined using spectroscopic analyses, including APCI-HR-MS and NMR. Compounds 1-7 displayed concentration-dependent inhibition of the alpha-glucosidase enzyme, with IC 50 values of 0.72, 1.05, 2.13, 1.22, 240.20, 18.70, and 163.10 µM, respectively. Their inhibitory activity surpassed acarbose, the positive control (IC 50 241.6 µM). Kinetic analysis revealed that all compounds acted as competitive inhibitors. Docking analysis predicted that all triterpenes bonded to the same site as acarbose in human intestinal alpha-glucosidase (PDB: 3TOP). A complementary metabolomic analysis of M. oleifera active fractions revealed the presence of 64 compounds, shedding new light on the plant's chemical composition. These findings suggest that M. oleifera holds promise as a valuable botanical source for developing compounds for managing blood sugar levels in individuals with diabetes.

Iron-chalcogenide superconductors continue to captivate researchers due to their diverse crystalline structures and intriguing superconducting properties, positioning them as both a valuable platform for theoretical investigations and promising candidates for practical applications. This review begins with a comprehensive overview of the fabrication techniques employed for various iron-chalcogenide superconductors, accompanied by a summary of their phase diagrams. Subsequently, it delves into the upper critical field, anisotropy, and critical current density. Furthermore, it discusses the successful fabrication of meters-long coated conductors and explores their applications in superconducting radio-frequency cavities and coils. Finally, several prospective avenues for future research are proposed.

Pleural mesothelioma (PM) is a highly aggressive tumor that is caused by asbestos exposure and lacks effective therapeutic regimens. Current procedures for PM diagnosis are invasive and can take a long time to reach a definitive result. Small extracellular vesicles (sEVs) have been identified as important communicators between tumor cells and their microenvironment via their cargo including circular RNAs (circRNAs). CircRNAs are thermodynamically stable, highly conserved, and have been found to be dysregulated in cancer. This study aimed to identify potential biomarkers for PM diagnosis by investigating the expression of specific circRNA gene pattern (hsa&#95;circ&#95;0007386) in cells and sEVs using digital polymerase chain reaction (dPCR). For this reason, 5 PM, 14 non-PM, and one normal mesothelial cell line were cultured. The sEV was isolated from the cells using the gold standard ultracentrifuge method. The RNA was extracted from both cells and sEVs, cDNA was synthesized, and dPCR was run. Results showed that hsa&#95;circ&#95;0007386 was significantly overexpressed in PM cell lines and sEVs compared to non-PM and normal mesothelial cell lines ( p < 0.0001). The upregulation of hsa&#95;circ&#95;0007386 in PM highlights its potential as a diagnostic biomarker. This study underscores the importance and potential of circRNAs and sEVs as cancer diagnostic tools.

The grooming behavior of honeybees serves as a crucial auto-protective mechanism against Varroa mite infestations. Compared to Apis mellifera , Apis cerana demonstrates more effective grooming behavior in removing Varroa mites from the bodies of infested bees. However, the underlying mechanisms regulating grooming behavior remain elusive. In this study, we evaluated the efficacy of the auto-grooming behavior between A. cerana and A. mellifera and employed RNA-sequencing technology to identify differentially expressed genes (DEGs) in bee brains with varying degrees of grooming behavior intensity. We observed that A. cerana exhibited a higher frequency of mite removal between day 5 and day 15 compared to A. mellifera , with day-9 bees showing the highest frequency of mite removal in A. cerana . RNA-sequencing results revealed the differential expression of the HTR2A and SLC17A8 genes in A. cerana and the CCKAR and TpnC47D genes in A. mellifera . Subsequent homology analysis identified the HTR2A gene and SLC17A8 gene of A. cerana as homologous to the HTR2A gene and SLC17A7 gene of A. mellifera . These DEGs are annotated in the neuroactive ligand-receptor interaction pathway, the glutamatergic synaptic pathway, and the calcium signaling pathway. Moreover, CCKAR , TpnC47D , HTR2A , and SLC17A7 may be closely related to the auto-grooming behavior of A. mellifera , conferring resistance against Varroa infestation. Our results further explain the relationship between honeybee grooming behavior and brain function at the molecular level and provide a reference basis for further studies of the mechanism of honeybee grooming behavior.

N -aryl-substituted pyrrolidines are important moieties widely found in bioactive substances and drugs. Herein, we present a practical reductive amination of diketones with anilines for the synthesis of N -aryl-substituted pyrrolidines in good to excellent yields. In this process, the N -aryl-substituted pyrrolidines were furnished via successive reductive amination of diketones via iridium-catalyzed transfer hydrogenation. The scale-up performance, water as a solvent, simple operation, as well as derivation of drug molecules showcased the potential application in organic synthesis.

Stretchable conductive composites play a pivotal role in the development of personalized electronic devices, electronic skins, and artificial implant devices. This article explores the fabrication and characterization of stretchable composites based on natural rubber (NR) filled with molybdenum disilicide (MoSi 2 ) nanoparticles and multi-walled carbon nanotubes (MWCNTs). Experimental characterization and molecular dynamics (MD) simulations are employed to investigate the static and dynamic properties of the composites, including morphology, glass transition temperature ( T g ), electrical conductivity, and mechanical behavior. Results show that the addition of MoSi 2 nanoparticles enhances the dispersion of MWCNTs within the NR matrix, optimizing the formation of a conductive network. Dynamic mechanical analysis (DMA) confirms the T g reduction with the addition of MWCNTs and the influence of MoSi 2 content on T g . Mechanical testing reveals that the tensile strength increases with MoSi 2 content, with an optimal ratio of 4:1 MoSi 2 :MWCNTs. Electrical conductivity measurements demonstrate that the MoSi 2 /MWCNTs/NR composites exhibit enhanced conductivity, reaching optimal values at specific filler ratios. MD simulations further support experimental findings, highlighting the role of MoSi 2 in improving dispersion and mechanical properties. Overall, the study elucidates the synergistic effects of nanoparticles and nanotubes in enhancing the properties of stretchable conductive composites.

The development of an advanced dye adsorbent that possesses a range of beneficial characteristics, such as high adsorption capacity, swift adsorption kinetics, selective adsorption capability, and robust reusability, remains a challenge. This study introduces a facile method for fabricating an amine-rich porous adsorbent (ARPA), which is specifically engineered for the adsorptive removal of anionic dyes from aqueous solutions. Through a comprehensive assessment, we have evaluated the adsorption performance of ARPA using two benchmark dyes: amaranth (ART) and tartrazine (TTZ). Our findings indicate that the adsorption process reaches equilibrium in a remarkably short timeframe of just 20 min, and it exhibits an excellent correlation with both the Langmuir isotherm model and the pseudo-second-order kinetic model. Furthermore, ARPA has demonstrated an exceptional maximum adsorption capacity, with values of 675.68 mg g -1 for ART and 534.76 mg g -1 for TTZ. In addition to its high adsorption capacity, ARPA has also shown remarkable selectivity, as evidenced by its ability to selectively adsorb TTZ from a mixed dye solution, a feature that is highly desirable for practical applications. Beyond its impressive adsorption capabilities, ARPA can be efficiently regenerated and recycled. It maintains a high level of original removal efficiency for both ART (76.8%) and TTZ (78.9%) even after five consecutive cycles of adsorption and desorption. Considering the simplicity of its synthesis and its outstanding adsorption performance, ARPA emerges as a highly promising material for use in dye removal applications. Consequently, this paper presents a straightforward and feasible method for the production of an effective dye adsorbent for environmental remediation.

One previously undescribed alkaloid, named penifuranone A ( 1 ), and three known compounds ( 2 - 4 ) were isolated from the mangrove endophytic fungus Penicillium crustosum SCNU-F0006. The structure of the new alkaloid ( 1 ) was elucidated based on extensive spectroscopic data analysis and single-crystal X-ray diffraction analysis. Four natural isolates and one new synthetic derivative of penifuranone A, compound 1a , were screened for their antimicrobial, antioxidant, and anti-inflammatory activities. Bioassays revealed that penifuranone A ( 1 ) exhibited strong anti-inflammatory activity in vitro by inhibiting nitric oxide (NO) production in lipopolysaccharide-activated RAW264.7 cells with an IC 50 value of 42.2 μM. The docking study revealed that compound 1 exhibited an ideal fit within the active site of the murine inducible nitric oxide synthase (iNOS), establishing characteristic hydrogen bonds.

To monitor the position and profile of therapeutic carbon beams in real-time, in this paper, we proposed a system called HiBeam-T. The HiBeam-T is a time projection chamber (TPC) with forty Topmetal-II- CMOS pixel sensors as its readout. Each Topmetal-II- has 72 × 72 pixels with the size of 83 μm × 83 μm. The detector consists of the charge drift region and the charge collection area. The readout electronics comprise three Readout Control Modules and one Clock Synchronization Module. This Hibeam-T has a sensitive area of 20 × 20 cm and can acquire the center of the incident beams. The test with a continuous 80.55 MeV/u 12 C 6+ beam shows that the measurement resolution to the beam center could reach 6.45 μm for unsaturated beam projections.

The application of recycled coarse aggregate (RA) in structural concrete can save non-renewable resources and reduce land occupation. Developing comprehensive knowledge of chloride penetration and service life modeling of recycled coarse aggregate concrete (RAC) is a prerequisite for practice. However, compared with the natural aggregate concrete (NAC), the inferior durability performance, especially chloride penetration resistance, of RAC hinders its application in structural concrete. Therefore, many RAC performance enhancement methods have been proposed. This paper presents a holistic review focused on the chloride penetration of RAC with/without enhancement methods and service life prediction. The current RAC performance enhancement methods are introduced. The improvement effect of the corresponding enhancement methods on the chloride penetration resistance of RAC are discussed and analyzed in turn. Based on the reviewed data on the chloride diffusion coefficient, the modification efficiencies of assorted enhancement methods are summarized. With the hope of promoting RAC application in structural concrete, the current literature on chloride-ingress-based service life prediction for RAC is also overviewed. In addition, the typical influencing factors on chloride transport properties are also discussed, i.e., RA quality. It can be concluded that enhancement techniques can effectively improve the chloride penetration resistance of RAC. The old mortar enhancement or removal methods can improve the chloride penetration resistance by 15-30%, depending on the specific treatment measures. The modification efficiency of the modifier material depends on the specific type and content of the incorporated substance, which ranges from approximately 5% to 95%. The estimated service life of RAC structures decreases with the increasing RA replacement ratio. Finally, concluding remarks are provided concerning future research on the chloride transport behavior of RAC.

Using sodium lignosulfonate as feedstock, ZnCl 2 and NaHCO 3 co-activated the hierarchical porous carbons (HPCs) were prepared by one-pot pyrolysis with different NaHCO 3 dosages (0-4 g) and carbonization temperatures (400-600 °C). Subsequently, phosphotungstate (HPW) was supported with the resulting biochar for the α-pinene hydration reaction to produce α-terpineol. The optimum preparation conditions were determined according to the yield of α-terpineol. The formation mechanism and physicochemical properties of HPCs were analyzed through TG, SEM, XPS, XRD, FT-IR, and N 2 adsorption-desorption isotherms. The results demonstrated that NaHCO 3 underwent a two-step reaction which liberated a substantial quantity of CO 2 , thereby enhancing activated carbon's macroporous and mesoporous structures. Simultaneously, NaHCO 3 mitigated strong acid gas (HCl) emissions during ZnCl 2 activation. Compared with AC450-4:8:0 prepared by ZnCl 2 activation alone, the total pore volume of AC450-4:8:2 prepared by co-activation is increased from 0.595 mL/g to 0.754 mL/g and the mesopore rate from 47.7% to 77.8%, which is conducive to reducing the steric hindrance of the hydration reaction and improving the selectivity. Hydration experiments show that the selectivity of α-terpineol is 55.7% under HPW/AC450-4:8:2 catalysis, higher than 31.0% for HPW and 47.4% for HPW/AC450-4:8:0.

When backfilling narrow spaces, controlled low-strength materials (CLSM) can be used to achieve an effective backfilling effect. The pipeline engineering in Yahnghe Avenue of Suqian, China, provides a favorable on-site condition for the use of CLSM. However, no guidance exists for the determination of the material mixture ratio of CLSM for this geological condition. Laboratory tests were performed to investigate the basic physical parameters of excavated soil and the optimal mixture ratio of CLSM. Results indicate that the sand and silt account for 29.76% and 57.23% of the weight of excavated soil, respectively. As the water content increases (from 40% to 50%), the flowability of the CLSM approximately shows a linear increase (slumps values from 154.3 mm to 269.75 mm for 9% cement content), while its compressive strength shows a linear decreasing trend (from 875.3 KPa to 468.3 KPa after curing for 28 days); as the cement content increases (from 6% to 12%), the flowability approximately shows a linear decreasing trend (from 238.8 mm to 178.5 mm for 45% water content), while the compressive strength shows a linear increasing trend (from 391.6 KPa to 987.6 KPa after curing for 28 days). By establishing the relationship between compressive strength/flowability and the water-cement ratio, the optimal material ratio is determined to be 9% cement content and 40-43% water content. The engineering application results indicate that the use of CLSM can achieve efficient and high-quality backfilling effects for pipeline trenches. The findings of this research may provide a reference for the application of CLSM in fields with similar geological conditions.

Capillary-gradient wicks can achieve fast or directional liquid transport, but they face fabrication challenges by traditional methods in terms of precise patterns. Laser processing is a potential solution due to its high pattern accuracy, but there are a few studies on laser-processed capillary-gradient wicks. In this paper, capillary step-gradient micro-grooved wicks (CSMWs) were fabricated by an ultraviolet nanosecond pulsed laser, and their capillary performance was studied experimentally. The CSMWs could be divided into three regions with a decreasing capillary radius. The equilibrium rising height of the CSMWs was enhanced by 124% compared to the non-gradient parallel wick. Different from the classical Lucas-Washburn model describing a uniform non-gradient wick, secondary capillary acceleration was observed in the negative gradient direction of the CSMWs. With the increase in laser power and the decrease in scanning speed, the capillary performance was promoted, and the optimal laser processing parameters were 4 W-10 mm/s. The laser-enhanced capillary performance was attributed to the improved hydrophilicity and reduced capillary radius, which resulted from the increased surface roughness, protrusion morphology, and deep-narrow V-shaped grooves induced by the high energy density of the laser. Our study demonstrates that ultraviolet pulsed laser processing is a highly efficient and low-cost method for fabricating high-performance capillary gradient wicks.

Titanium alloy components often experience damage from impact loads during usage, which makes improving the mechanical properties of TC4 titanium alloys crucial. This paper investigates the influence of laser scanning irradiation on the tensile properties of thin titanium alloy sheets. Results indicate that the tensile strength of thin titanium alloy sheets exhibits a trend of initial increase followed by a decrease. Different levels of enhancement are observed in the elongation at break of a cross-section. Optimal improvement in the elongation at break is achieved when the laser fluence is around 8 J/cm 2 , while the maximum increase in tensile strength occurs at approximately 10 J/cm 2 . Using femtosecond laser surface irradiation, this study compares the maximum enhancement in the tensile strength of titanium alloy base materials, which is approximately 8.54%, and the maximum increase in elongation at break, which reaches 25.61%. In addition, the results verify that cracks in tensile fractures of TC4 start from the middle, while laser-induced fracture cracks occur from both ends.

R2R3-MYB represents a substantial gene family that plays diverse roles in plant development. In this study, 102 SmR2R3-MYB genes were identified from eggplant fruit and classified into 31 subfamilies. Analysis indicated that segmental duplication events played a pivotal role in the expansion of the SmR2R3-MYB gene family. Furthermore, the prediction of miRNAs targeting SmR2R3-MYB genes revealed that 60 SmR2R3-MYBs are targeted by 57 miRNAs, with specific miRNAs displaying varying numbers of target genes, providing valuable insights into the regulatory functions of miRNAs in plant growth, development, and responses to stress conditions. Through expression profile analysis under various treatment conditions, including low temperature (4 °C), plant hormone (ABA, Abscisic acid), and drought stress (PEG, Polyethylene glycol), diverse and complex regulatory mechanisms governing SmR2R3-MYB gene expression were elucidated. Notably, EGP21875.1 and EGP21874.1 exhibited upregulation in expression under all treatment conditions. Transcriptome and metabolome analyses demonstrated that, apart from anthocyanins (delphinidin-3-O-glucoside, cyanidin-3-O-(6-O-p-coumaroyl)-glucoside, and malvidin-3-O-(6-O-p-coumaroyl)-glucoside), overexpression of SmMYB75 could also elevate the content of various beneficial compounds, such as flavonoids, phenolic acids, and terpenes, in eggplant pulp. This comprehensive study enhances our understanding of SmR2R3-MYB gene functions and provides a strong basis for further research on their roles in regulating anthocyanin synthesis and improving eggplant fruit quality.

Peanuts play a pivotal role as an economic crop on a global scale, serving as a primary source of both edible oil and protein. Peanut rust ( Puccinia arachidis Speg.) disease constitutes a significant global biotic stress, representing a substantial economic threat to the peanut industry by inducing noteworthy reductions in seed yields and compromising oil quality. This comprehensive review delves into the distinctive characteristics and detrimental symptoms associated with peanut rust, scrutinizing its epidemiology and the control strategies that are currently implemented. Notably, host resistance emerges as the most favored strategy due to its potential to surmount the limitations inherent in other approaches. The review further considers the recent advancements in peanut rust resistance breeding, integrating the use of molecular marker technology and the identification of rust resistance genes. Our findings indicate that the ongoing refinement of control strategies, especially through the development and application of immune or highly resistant peanut varieties, will have a profound impact on the global peanut industry.

Triple combination conversion therapy, involving transcatheter arterial chemoembolization (TACE) or hepatic arterial infusion chemotherapy (HAIC) combined with tyrosine kinase inhibitors (TKIs) and immune checkpoint inhibitors (ICIs), has shown an encouraging objective response rate (ORR) and successful conversion surgery rate in initially unresectable hepatocellular carcinoma (HCC). However, the safety and long-term survival outcomes of subsequent liver resection after successful conversion still remain to be validated. From February 2019 to February 2023, 726 patients were enrolled in this retrospective study (75 patients received hepatectomy after conversion therapy [CLR group], and 651 patients underwent pure hepatectomy [LR group]). Propensity score matching (PSM) was used to balance the preoperative baseline characteristics. After PSM, 68 patients in the CLR group and 124 patients in the LR group were analyzed, and all the matching variables were well-balanced. Compared with the LR group, the CLR group experienced longer Pringle maneuver time, longer operation time, and longer hospital stays. In addition, the CLR group had significantly higher incidence rates of intra-abdominal bleeding, biliary leakage, post-hepatectomy liver failure (PHLF), and Clavien-Dindo grade IIIa complications than the LR group. There were no significant statistical differences in overall survival (OS) (hazard ratio [HR] 0.724; 95% confidence interval [CI] 0.356-1.474; p = 0.374) and recurrence-free survival (RFS) (HR 1.249; 95% CI 0.807-1.934; p = 0.374) between the two groups. Liver resection following triple combination conversion therapy in initially unresectable HCC may achieve favorable survival outcomes with manageable safety profiles; presenting as a promising treatment option for initially unresectable HCC.

Acute liver injury (ALI) has the potential to compromise hepatic function rapidly, with severe cases posing a considerable threat to human health and wellbeing. Conventional treatments, such as the oral administration of antioxidants, can inadvertently lead to liver toxicity and other unwanted side effects. Mesenchymal stromal cells (MSCs) can target therapeutic agents directly to inflammatory sites owing to their homing effect, and they offer a promising avenue for the treatment of ALI. However, the efficacy and feasibility of these live cell products are hampered by challenges associated with delivery pathways and safety concerns. Therefore, in this work, MSC membranes were ingeniously harnessed as protective shells to encapsulate synthesized PLGA nanoparticle cores (PLGA/MSCs). This strategic approach enabled nanoparticles to simulate endogenous substances and yielded a core-shell nano-biomimetic structure. The biomimetic nanocarrier remarkably maintained the homing ability of MSCs to inflammatory sites. In this study, cryptotanshinone (CPT)-loaded PLGA/MSCs (CPT@PLGA/MSC) were prepared. These nanoparticles can be effectively internalized by LO2 cells. They reduced cellular oxidative stress and elevated inflammatory levels. In vivo results suggested that, after intravenous administration, CPT@PLGA/MSCs significantly reduced uptake by the reticuloendothelial system and immune recognition compared to PLGA nanoparticles without MSC membrane coatings, subsequently resulting in their targeted and enhanced accumulation in the liver. The effectiveness of CPT@PLGA/MSCs in alleviating carbon tetrachloride-induced oxidative stress and inflammation in a mouse model was unequivocally demonstrated through comprehensive histological examination and liver function tests. This study introduces a pioneering strategy with substantial potential for ALI treatment.

Chlorophyll is the main photosynthetic pigment and is crucial for plant photosynthesis. Leaf color mutants are widely used to identify genes involved in the synthesis or metabolism of chlorophyll. In this study, a spontaneous mutant, yellow-green leaf 19 ( ygl19 ), was isolated from rice ( Oryza sativa ). This ygl19 mutant showed yellow-green leaves and decreased chlorophyll level and net photosynthetic rate. Brown necrotic spots appeared on the surface of ygl19 leaves at the tillering stage. And the agronomic traits of the ygl19 mutant, including the plant height, tiller number per plant, and total number of grains per plant, were significantly reduced. Map-based cloning revealed that the candidate YGL19 gene was LOC&#95;Os03g21370. Complementation of the ygl19 mutant with the wild-type CDS of LOC&#95;Os03g21370 led to the restoration of the mutant to the normal phenotype. Evolutionary analysis revealed that YGL19 protein and its homologues were unique for photoautotrophs, containing a conserved Ycf54 functional domain. A conserved amino acid substitution from proline to serine on the Ycf54 domain led to the ygl19 mutation. Sequence analysis of the YGL19 gene in 4726 rice accessions found that the YGL19 gene was conserved in natural rice variants with no resulting amino acid variation. The YGL19 gene was mainly expressed in green tissues, especially in leaf organs. And the YGL19 protein was localized in the chloroplast for function. Gene expression analysis via qRT-PCR showed that the expression levels of tetrapyrrole synthesis-related genes and photosynthesis-related genes were regulated in the ygl19 mutant. Reactive oxygen species (ROS) such as superoxide anions and hydrogen peroxide accumulated in spotted leaves of the ygl19 mutant at the tillering stage, accompanied by the regulation of ROS scavenging enzyme-encoding genes and ROS-responsive defense signaling genes. This study demonstrates that a novel yellow-green leaf gene YGL19 affects tetrapyrrole biosynthesis, photosynthesis, and ROS metabolism in rice.

Nd-based polydiene elastomers, including NdIR and NdBR, are regarded as indispensable key raw materials in preparing green tires with excellent performance capabilities, but their wide application is still limited by the relative higher cost of Nd precatalysts. Nd-mediated coordinative chain transfer polymerization (CCTP) of diene provides an effective strategy to reduce the precatalyst cost, because this method involves very high atom economy, i.e., each Nd molecule can generate multiple polymer chains. Nevertheless, all possible factors that could influence such CCTP behaviors are still mostly unexplored to date. In this report, the basic chemistry on the influence of external donors on the overall CCTP behaviors of isoprene was established for the first time. It was found that increasing the amount of external donors had a negative influence on the chain transfer efficiencies, resulting in gradually decreasing atom economies. Catalyst addition order studies revealed that the coordination of donors with cationic Nd active species, rather than alkylaluminium CTAs, contributed mostly to such decreased efficiencies. Moreover, it was found that when the ratio of donors and Nd compounds was higher than 1.0, the CCTP behaviors were corrupted, resulting in polymers with broad distributions, as well as resulting in low atom economies; nevertheless, when the ratio was lower than 0.5, the system still displayed CCTP characteristics, implying that the critical ratio for maintaining the CCTP was 0.5. Additionally, when such a ratio was 0.01, the high atom economy was almost the same as donor-free CCTP systems. Detailed kinetic studies at such a ratio demonstrated that the donor-contained system proceeded in a well-controlled manner, as concluded from the good linear relationship between the M n of the PIps against the polymer yields, as well as the good linearity between the M n against the (IP)/(Nd) ratios. Such maintained CCTP properties also allowed for seeding two-step polymerizations to prepare diblock copolymers with precisely controlled molecular weights. Expanding the types of donors to more phosphine, oxygen, and nitrogen containing compounds showed that they also affected the CCTP behaviors depending on their steric and electronic properties.

Cryptography is very essential in our daily life, not only for confidentiality of information, but also for information integrity verification, non-repudiation, authentication, and other aspects. In modern society, cryptography is widely used; everything from personal life to national security is inseparable from it. With the emergence of quantum computing, traditional encryption methods are at risk of being cracked. People are beginning to explore methods for defending against quantum computer attacks. Among the methods currently developed, quantum key distribution is a technology that uses the principles of quantum mechanics to distribute keys. Post-quantum encryption algorithms are encryption methods that rely on mathematical challenges that quantum computers cannot solve quickly to ensure security. In this study, an integrated review of post-quantum encryption algorithms is conducted from the perspective of traditional cryptography. First, the concept and development background of post-quantum encryption are introduced. Then, the post-quantum encryption algorithm Kyber is studied. Finally, the achievements, difficulties and outstanding problems in this emerging field are summarized, and some predictions for the future are made.

Crocins are a group of highly valuable water-soluble carotenoids that are reported to have many pharmacological activities, such as anticancer properties, and the potential for treating neurodegenerative diseases including Alzheimer's disease. Crocins are mainly biosynthesized in the stigmas of food-medicine herbs Crocus sativus L. and Gardenia jasminoides fruits. The distribution is narrow in nature and deficient in resources, which are scarce and expensive. Recently, the synthesis of metabolites in the heterologous host has opened up the potential for large-scale and sustainable production of crocins, especially for the main active compounds crocin I and crocin II. In this study, GjCCD4a , GjALDH2C3 , GjUGT74F8 , and GjUGT94E13 from G. jasminoides fruits were expressed in Nicotiana benthamiana . The highest total content of crocins in T1 generation tobacco can reach 78,362 ng/g FW (fresh weight) and the dry weight is expected to reach 1,058,945 ng/g DW (dry weight). Surprisingly, the primary effective constituents crocin I and crocin II can account for 99% of the total crocins in transgenic plants. The strategy mentioned here provides an alternative platform for the scale-up production of crocin I and crocin II in tobacco.

Zinc finger protein (ZFP) transcription factors play a pivotal role in regulating plant growth, development, and response to biotic and abiotic stresses. Although extensively characterized in model organisms, these genes have yet to be reported in bamboo plants, and their expression information is lacking. Therefore, we identified 21 B-box ( BBX ) genes from a transcriptome analysis of Bambusa pervariabilis × Dendrocalamopsis grandis . Consequently, multiple sequence alignments and an analysis of conserved motifs showed that they all had highly similar structures. The BBX genes were divided into four subgroups according to their phylogenetic relationships and conserved domains. A GO analysis predicted multiple functions of the BBX genes in photomorphogenesis, metabolic processes, and biological regulation. We assessed the expression profiles of 21 BBX genes via qRT-PCR under different adversity conditions. Among them, eight genes were significantly up-regulated under water deficit stress ( BBX4 , BBX10 , BBX11 , BBX14 , BBX15 , BBX16 , BBX17 , and BBX21 ), nine under salt stress ( BBX2 , BBX3 , BBX7 , BBX9 , BBX10 , BBX12 , BBX15 , BBX16 , and BBX21 ), twelve under cold stress ( BBX1 , BBX2 , BBX4 , BBX7 , BBX10 , BBX12 , BBX14 , BBX15 , BBX17 , BBX18 , BBX19 , and BBX21 ), and twelve under pathogen infestation stress ( BBX1 , BBX2 , BBX4 , BBX7 , BBX10 , BBX12 , BBX14 , BBX15 , BBX17 , BBX18 , BBX19 , and BBX21 ). Three genes ( BBX10 , BBX15 , and BBX21 ) were significantly up-regulated under both biotic and abiotic stresses. These results suggest that the BBX gene family is integral to plant growth, development, and response to multivariate stresses. In conclusion, we have comprehensively analyzed the BDBBX genes under various adversity stress conditions, thus providing valuable information for further functional studies of this gene family.

Doxorubicin, a widely used chemotherapeutic drug in clinical oncology, causes a series of cardiac side effects referred to as doxorubicin-induced cardiotoxicity. Hyperhomocysteinaemia is an independent risk factor for multiple cardiovascular diseases. However, whether hyperhomocysteinaemia contributes to doxorubicin-induced cardiotoxicity is currently unknown. In this study, we explored the pathogenic effects of hyperhomocysteinaemia induced by dietary methionine supplementation (2% wt/wt in rodent chow) in a mouse model of doxorubicin-induced cardiotoxicity. Our data showed that methionine supplementation doubled serum homocysteine levels, inducing mild hyperhomocysteinaemia. Doxorubicin at a cumulative dosage of 25 mg/kg body weight led to significant weight loss and severe cardiac dysfunction, which were further exacerbated by methionine-induced mild hyperhomocysteinaemia. Doxorubicin-induced cardiac atrophy, cytoplasmic vacuolisation, myofibrillar disarray and loss, as well as cardiac fibrosis, were also exacerbated by methionine-induced mild hyperhomocysteinaemia. Additional folic acid supplementation (0.006% wt/wt) prevented methionine-induced hyperhomocysteinaemia and inhibited hyperhomocysteinaemia-aggravated cardiac dysfunction and cardiomyopathy. In particular, hyperhomocysteinaemia increased both serum and cardiac oxidative stress, which could all be inhibited by folic acid supplementation. Therefore, we demonstrated for the first time that hyperhomocysteinaemia could exacerbate doxorubicin-induced cardiotoxicity in mice, and the pathogenic effects of hyperhomocysteinaemia might at least partially correlate with increased oxidative stress and could be prevented by folic acid supplementation. Our study provides preliminary experimental evidence for the assessment of hyperhomocysteinaemia as a potential risk factor for chemotherapy-induced cardiotoxicity in cancer patients.

Constitutional deletions of chromosome 1q42 region are rare. The phenotype spectrum associated with this copy number change is variable, including developmental delay, intellectual disability, seizures, and dysmorphology. This study describes a patient with developmental delays and brain abnormalities. G-banded karyotype, FISH, SNP oligonucleotide microarray analysis (SOMA), and whole exome sequencing analysis were performed. Postnatal reanalysis of prenatal SOMA and follow-up parental testing revealed a paternally inherited 63 kb deletion at 1q42.11 in the patient. We characterized the clinical features of this patient, providing insight into the clinical phenotype associated with deletions of the 1q42.11 sub-band. Our study provides new evidence supporting the potential functional importance of the FBXO28 3' UTR region and the hypothesis that FBXO28 is a critical gene in the pathogenesis of chromosome 1q41q42 microdeletion syndrome. It also highlights the different goals and reporting criteria between prenatal and postnatal microarray tests.

To study the evolution of stress on the ring and segment interfaces during the construction process of the concrete encapsulation of the main arch ring in a rigid-frame arch bridge, alongside its impact on the ultimate load-bearing capacity of the main arch ring, a 1:10 scale model experiment was conducted by taking the 600 m Tian'e Longtan Bridge as the prototype. The key cross-section concrete strain data were collected during the entire construction process of the main arch ring via fiber-optic strain sensors, which were used to investigate the stress evolution at ring and segment interfaces. ANSYS APDL was employed to simulate the ultimate bearing capacity under various loading conditions of two different finite element models, which were, respectively, formed segmentally and by single pouring. The results revealed that (1) after the closure of the concrete encapsulation of the main arch ring, the concrete stress in the cross-section exhibits significant stress disparities. At the same cross-section, the level of the web concrete stress can reach 76% of the floor concrete stress, while the roof concrete stress level is less than 20% of the floor concrete stress. (2) At the junction of two adjacent work planes, there are considerable differences in the stress levels of the concrete on both sides. After the closure of the main arch ring, the intersegment stress ratios of the floor, web, and roof concrete are 60~70%, 40~60%, and 0~5%, respectively. (3) Loading conditions remarkably affected the ultimate bearing capacity of the main arch ring. Under mid-span loading and 1/4 span symmetrical loading conditions, compared to single-pour concrete encapsulation, the ultimate bearing capacity of the main arch ring with concrete encapsulated by segmented and ring-divided pouring decreased by 19.16% and 5.23%, respectively, compared to single-pour concrete encapsulation. This suggests that the non-uniformity of stress distribution in the concrete sheath can lead to reductions in the ultimate bearing capacity of the arch ring.

Piwi proteins play a significant role in germ cell development and the silencing of transposons in animals by associating with small non-coding RNAs known as Piwi-interacting RNAs (piRNAs). While the Piwi gene has been well characterized in various insect species, the role of the Piwi ( PxPiwi ) gene in the diamondback moth ( Plutella xylostella ), a globally distributed pest of cruciferous crops, remains unclear. Expression analysis demonstrated the upregulation of PxPiwi in pupae and testes. Furthermore, we generated a PxPiwi -knockout mutant using CRISPR/Cas9 technology, which resulted in a significantly prolonged pupal stage and the failure of pupae to develop into adults. Additionally, the knockdown of PxPiwi , through RNA interference (RNAi), led to a substantial decrease in the oviposition and hatchability of P. xylostella . These findings indicate that PxPiwi is specifically expressed and essential for the development and reproduction of P. xylostella . This is the first report indicating the involvement of the Piwi gene in the development of lepidopteran insects, except for reproduction and germ cell development, which provides a foundation for future investigations into the functions of PxPiwi .

Porcine reproductive and respiratory syndrome (PRRS) is a highly contagious disease in the pig industry, but its pathogenesis is not yet fully understood. The disease is caused by the PRRS virus (PRRSV), which primarily infects porcine alveolar macrophages and disrupts the immune system. Unfortunately, there is no specific drug to cure PRRS, so vaccination is crucial for controlling the disease. There are various types of single and combined vaccines available, including live, inactivated, subunit, DNA, and vector vaccines. Among them, live vaccines provide better protection, but cross-protection is weak. Inactivated vaccines are safe but have poor immune efficacy. Subunit vaccines can be used in the third trimester of pregnancy, and DNA vaccines can enhance the protective effect of live vaccines. However, vector vaccines only confer partial protection and have not been widely used in practice. A PRRS vaccine that meets new-generation international standards is still needed. This manuscript provides a comprehensive review of the advantages, disadvantages, and applicability of live-attenuated, inactivated, subunit, live vector, DNA, gene-deletion, synthetic peptide, virus-like particle, and other types of vaccines for the prevention and control of PRRS. The aim is to provide a theoretical basis for vaccine research and development.

The identification of the growth and development period of rice is of great significance to achieve high-yield and high-quality rice. However, the acquisition of rice growth period information mainly relies on manual observation, which has problems such as low efficiency and strong subjectivity. In order to solve these problems, a lightweight recognition method is proposed to automatically identify the growth period of rice: Small-YOLOv5, which is based on improved YOLOv5s. Firstly, the new backbone feature extraction network MobileNetV3 was used to replace the YOLOv5s backbone network to reduce the model size and the number of model parameters, thus improving the detection speed of the model. Secondly, in the feature fusion stage of YOLOv5s, we introduced a more lightweight convolution method, GsConv, to replace the standard convolution. The computational cost of GsConv is about 60-70% of the standard convolution, but its contribution to the model learning ability is no less than that of the standard convolution. Based on GsConv, we built a lightweight neck network to reduce the complexity of the network model while maintaining accuracy. To verify the performance of Small-YOLOv5s, we tested it on a self-built dataset of rice growth period. The results show that compared with YOLOv5s (5.0) on the self-built dataset, the number of the model parameter was reduced by 82.4%, GFLOPS decreased by 85.9%, and the volume reduced by 86.0%. The mAP (0.5) value of the improved model was 98.7%, only 0.8% lower than that of the original YOLOv5s model. Compared with the mainstream lightweight model YOLOV5s- MobileNetV3-Small, the number of the model parameter was decreased by 10.0%, the volume reduced by 9.6%, and the mAP (0.5:0.95) improved by 5.0%-reaching 94.7%-and the recall rate improved by 1.5%-reaching 98.9%. Viruses, such as Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), infect hosts and take advantage of host cellular machinery for genome replication and new virion production. Identifying and elucidating host pathways for viral infection is critical for understanding the development of the viral life cycle and novel therapeutics. The SARS-CoV-2 N protein is critical for viral RNA (vRNA) genome packaging in new virion formation. Using our quantitative Förster energy transfer/Mass spectrometry (qFRET/MS) coupled method and immunofluorescence imaging, we identified three SUMOylation sites of the SARS-CoV-2 N protein. We found that (1) Small Ubiquitin-like modifier (SUMO) modification in Nucleocapsid (N) protein interaction affinity increased, leading to enhanced oligomerization of the N protein; (2) one of the identified SUMOylation sites, K65, is critical for its nuclear translocation. These results suggest that the host human SUMOylation pathway may be critical for N protein functions in viral replication and pathology in vivo. Thus, blocking essential host pathways could provide a novel strategy for future anti-viral therapeutics development, such as for SARS-CoV-2 and other viruses.

MADS-box genes constitute a large family of transcription factors that play important roles in plant growth and development. However, our understanding of MADS-box genes involved in anther development and male sterility in Salvia miltiorrhiza is still limited. In this study, 63 MADS-box genes were identified from the genome of the male sterility ecotype Sichuan S. miltiorrhiza ( S. miltiorrhiza &#95;SC) unevenly distributed among eight chromosomes. Phylogenetic analysis classified them into two types and 17 subfamilies. They contained 1 to 12 exons and 10 conserved motifs. Evolution analysis showed that segmental duplication was the main force for the expansion of the SmMADS gene family, and duplication gene pairs were under purifying selection. Cis-acting elements analysis demonstrated that the promoter of SmMADS genes contain numerous elements associated with plant growth and development, plant hormones, and stress response. RNA-seq showed that the expression levels of B-class and C-class SmMADS genes were highly expressed during anther development, with SmMADS11 likely playing an important role in regulating anther development and male fertility in S. miltiorrhiza &#95;SC. Overall, this study provides a comprehensive analysis of the MADS-box gene family in S. miltiorrhiza , shedding light on their potential role in anther development and male sterility.

Salvia miltiorrhiza Bunge is an important traditional herb. Salvia miltiorrhiza is distributed in the Sichuan province of China (here called SC). Under natural conditions, it does not bear seeds and its sterility mechanism is still unclear. Through artificial cross, there was defective pistil and partial pollen abortion in these plants. Electron microscopy results showed that the defective pollen wall was caused by delayed degradation of the tapetum. Due to the lack of starch and organelle, the abortive pollen grains showed shrinkage. RNA-seq was performed to explore the molecular mechanisms of pollen abortion. KEGG enrichment analysis suggested that the pathways of phytohormone, starch, lipid, pectin, and phenylpropanoid affected the fertility of S. miltiorrhiza . Moreover, some differentially expressed genes involved in starch synthesis and plant hormone signaling were identified. These results contribute to the molecular mechanism of pollen sterility and provide a more theoretical foundation for molecular-assisted breeding.

Mulberry leaves are a well-known traditional Chinese medicine herb, and it has been observed since ancient times that leaves collected after frost have superior medicinal properties. Therefore, understanding the changes in critical metabolic components of mulberry leaves, specifically Morus nigra L., is essential. In this study, we conducted widely targeted metabolic profiling analyses on two types of mulberry leaves, including Morus nigra L. and Morus alba L., harvested at different times. In total, we detected over 100 compounds. After frost, 51 and 58 significantly different metabolites were identified in the leaves of Morus nigra L. and Morus alba L., respectively. Further analysis revealed a significant difference in the effect of defrosting on the accumulation of metabolites in the two mulberries. Specifically, in Morus nigra L., the content of 1-deoxynojirimycin (1-DNJ) in leaves decreased after frost, while flavonoids peaked after the second frost. In Morus alba L., the content of DNJ increased after frost, reaching its peak one day after the second frost, whereas flavonoids primarily peaked one week before frost. In addition, an analysis of the influence of picking time on metabolite accumulation in two types of mulberry leaves demonstrated that leaves collected in the morning contained higher levels of DNJ alkaloids and flavonoids. These findings provide scientific guidance for determining the optimal harvesting time for mulberry leaves.

Soilbags are expandable three-dimensional geosynthetic bags made from high-density polyethylene or polypropylene. This study conducted a series of plate load tests to explore the bearing capacity of soft foundations reinforced by soilbags filled with solid wastes based on an onshore wind farm project in China. The effect of contained material on the bearing capacity of the soilbag-reinforced foundation was investigated during the field tests. The experimental studies indicated that soilbag reinforcement with reused solid wastes could substantially improve the bearing capacity of soft foundations under vertical loading conditions. Solid wastes like excavated soil or brick slag residues were found to be suitable as contained material, and the soilbags with plain soil mixed with brick slag had higher bearing capacity than those with pure plain soil. The earth pressure analysis indicated that stress diffusion occurred through the soilbag layers to reduce the load transferred to the underlying soft soil. The stress diffusion angle of soilbag reinforcement obtained from the tests was approximately 38°. In addition, combining soilbag reinforcement with bottom sludge permeable treatment was an effective foundation reinforcement method, which required fewer soilbag layers due to its relatively high permeability. Furthermore, soilbags are considered sustainable construction materials with advantages such as high construction efficiency, low cost, easy reclamation and environmental friendliness while making full use of local solid wastes.

Insulator defect detection is of great significance to compromise the stability of the power transmission line. The state-of-the-art object detection network, YOLOv5, has been widely used in insulator and defect detection. However, the YOLOv5 network has limitations such as poor detection rate and high computational loads in detecting small insulator defects. To solve these problems, we proposed a light-weight network for insulator and defect detection. In this network, we introduced the Ghost module into the YOLOv5 backbone and neck to reduce the parameters and model size to enhance the performance of unmanned aerial vehicles (UAVs). Besides, we added small object detection anchors and layers for small defect detection. In addition, we optimized the backbone of YOLOv5 by applying convolutional block attention modules (CBAM) to focus on critical information for insulator and defect detection and suppress uncritical information. The experiment result shows the mean average precision (mAP) is set to 0.5, and the mAP is set from 0.5 to 0.95 of our model and can reach 99.4% and 91.7%; the parameters and model size were reduced to 3,807,372 and 8.79 M, which can be easily deployed to embedded devices such as UAVs. Moreover, the speed of detection can reach 10.9 ms/image, which can meet the real-time detection requirement.

One of the main obstacles in biocatalysis is the substrate inhibition (SI) of enzymes that play important roles in biosynthesis and metabolic regulation in organisms. The promiscuous glycosyltransferase UGT72AY1 from Nicotiana benthamiana is strongly substrate-inhibited by hydroxycoumarins (inhibitory constant Ki < 20 µM), but only weakly inhibited when monolignols are glucosylated ( Ki > 1000 µM). Apocarotenoid effectors reduce the inherent UDP-glucose glucohydrolase activity of the enzyme and attenuate the SI by scopoletin derivatives, which could also be achieved by mutations. Here, we studied the kinetic profiles of different phenols and used the substrate analog vanillin, which has shown atypical Michaelis-Menten kinetics in previous studies, to examine the effects of different ligands and mutations on the SI of NbUGT72AY1. Coumarins had no effect on enzymatic activity, whereas apocarotenoids and fatty acids strongly affected SI kinetics by increasing the inhibition constant Ki . Only the F87I mutant and a chimeric version of the enzyme showed weak SI with the substrate vanillin, but all mutants exhibited mild SI when sinapaldehyde was used as an acceptor. In contrast, stearic acid reduced the transferase activity of the mutants to varying degrees. The results not only confirm the multi-substrate functionality of NbUGT72AY1, but also reveal that the enzymatic activity of this protein can be fine-tuned by external metabolites such as apocarotenoids and fatty acids that affect SI. Since these signals are generated during plant cell destruction, NbUGT72AY1 likely plays an important role in plant defense by participating in the production of lignin in the cell wall and providing direct protection through the formation of toxic phytoalexins.

Calcineurin, a key regulator of the calcium signaling pathway, is involved in calcium signal transduction and calcium ion homeostasis. Magnaporthe oryzae is a devastating filamentous phytopathogenic fungus in rice, yet little is known about the function of the calcium signaling system. Here, we identified a novel calcineurin regulatory-subunit-binding protein, MoCbp7, which is highly conserved in filamentous fungi and was found to localize in the cytoplasm. Phenotypic analysis of the MoCBP7 gene deletion mutant (Δ Mocbp7 ) showed that MoCbp7 influenced the growth, conidiation, appressorium formation, invasive growth, and virulence of M. oryzae . Some calcium-signaling-related genes, such as YVC1 , VCX1 , and RCN1 , are expressed in a calcineurin/MoCbp7-dependent manner. Furthermore, MoCbp7 synergizes with calcineurin to regulate endoplasmic reticulum homeostasis. Our research indicated that M. oryzae may have evolved a new calcium signaling regulatory network to adapt to its environment compared to the fungal model organism Saccharomyces cerevisiae .

As a common harmful pollutant, cadmium (Cd) can easily enter the human body through the food chain, posing a major threat to human health. Gut microbiota play a key role in Cd absorption. Docosahexaenoic acid (DHA) is thought to have a potential role in the treatment of Cd poisoning. This study investigated the therapeutic effect and mechanism of DHA in Cd-exposed mice from the perspective of the gut microbiota. The results showed that DHA significantly increased the Cd content in feces and decreased the Cd accumulation in the organs of mice. The gut microbiota results showed that DHA significantly restored the abundance of Parabacteroides in the gut microbiota of Cd-exposed mice. Parabacteroides distasonis ( P. distasonis ), a representative strain of the Parabacteroides , also showed Cd- and toxicity-reduction capabilities. P. distasonis significantly restored the gut damage caused by Cd exposure. At the same time, P. distasonis reduced the Cd content in the liver, spleen, lung, kidneys, gut, and blood to varying degrees and significantly increased the Cd content in feces. The succinic acid produced by P. distasonis plays an important role in promoting Cd excretion in Cd-exposed mice. Therefore, these results suggest that P. distasonis may have a potential role in DHA-mediated Cd excretion in Cd-exposed mice.

Although one of the poster children of high-performance MEMS (Micro Electro Mechanical Systems) gyroscopes, the MEMS hemispherical resonator gyroscope (HRG) is faced with the barrier of technical and process limits, which makes it unable to form a resonator with the best structure. How to obtain the best resonator under specific technical and process limits is a significant topic for us. In this paper, the optimization of a MEMS polysilicon hemispherical resonator, designed by patterns based on PSO-BP and NSGA-II, was introduced. Firstly, the geometric parameters that significantly contribute to the performance of the resonator were determined via a thermoelastic model and process characteristics. Variety regulation between its performance parameters and geometric characteristics was discovered preliminarily using finite element simulation under a specified range. Then, the mapping between performance parameters and structure parameters was determined and stored in the BP neural network, which was optimized via PSO. Finally, the structure parameters in a specific numerical range corresponding to the best performance were obtained via the selection, heredity, and variation of NSGAII. Additionally, it was demonstrated using commercial finite element soft analysis that the output of the NSGAII, which corresponded to the Q factor of 42,454 and frequency difference of 8539, was a better structure for the resonator (generated by polysilicon under this process within a selected range) than the original. Instead of experimental processing, this study provides an effective and economical alternative for the design and optimization of high-performance HRGs under specific technical and process limits.

The high vascularization of glioma highlights the potential value of anti-angiogenic therapeutics for glioma treatment. Previously, we designed a novel vascular-targeting and blood-brain barrier (BBB)-penetrating peptide, TAT-AT7, by attaching the cell-penetrating peptide TAT to a vascular-targeting peptide AT7, and we demonstrated that TAT-AT7 could target binding to the vascular endothelial growth factor receptor 2 (VEGFR-2) and Neuropilin-1 (NRP-1), which are both highly expressed in endothelial cells. TAT-AT7 has been proven to be a good targeting peptide which could effectively deliver the secretory endostatin gene to treat glioma via the TAT-AT7-modified polyethyleneimine (PEI) nanocomplex. In the current study, we further explored the molecular binding mechanisms of TAT-AT7 to VEGFR-2 and NRP-1 and its anti-glioma effects. Accordingly, TAT-AT7 was proven to competitively bind to VEGFR-2 and NRP-1 and prevent VEGF-A165 binding to the receptors by the surface plasmon resonance (SPR) assay. TAT-AT7 inhibited endothelial cells' proliferation, migration, invasion, and tubule formation, as well as promoted endothelial cells' apoptosis in vitro. Further research revealed that TAT-AT7 inhibited the phosphorylation of VEGFR-2 and its downstream PLC-γ, ERK1/2, SRC, AKT, and FAK kinases. Additionally, TAT-AT7 significantly inhibited angiogenesis of zebrafish embryo. Moreover, TAT-AT7 had a better penetrating ability and could penetrate the BBB into glioma tissue and target glioma neovascularization in an orthotopic U87-glioma-bearing nude mice model, and exhibited the effect of inhibiting glioma growth and angiogenesis. Taken together, the binding and function mechanisms of TAT-AT7 were firstly revealed, and TAT-AT7 was proven to be an effective and promising peptide for the development of anti-angiogenic drugs for targeted treatment of glioma.
