Background: In the pursuit of sustainable energy sources, residual biomass has emerged as a promising renewable resource. However, efficiently managing residual biomass poses significant challenges, particularly in optimizing supply chain operations. Advanced modeling approaches are necessary to address these complexities. This study aims to develop a comprehensive methodological framework for creating simulation models tailored to agroforestry residual biomass supply chains. Methods: The study employs a hybrid simulation approach, integrating geographic information system mapping with a case study analysis. The simulation was conducted over a 365-day period, using the anyLogistix software (version 2.15.3.202209061204) to model various supply chain dynamics. The framework also accounts for financial, operational, customer satisfaction, and environmental metrics. Results: The simulation results showed a total expenditure of EUR 5,219,411.3, with transportation being the primary cost driver, involving 5678 trips and a peak capacity of 67.16 m3. CO2 emissions were measured at 487.7 kg/m3. The model performed as expected, highlighting the need for sustainable logistics strategies to reduce costs, lower losses, and improve productivity. Conclusions: This study presents one of the first detailed methodological frameworks for simulating agroforestry residual biomass supply chains. It provides valuable managerial insights into the financial, operational, and environmental aspects of supply chain management. As environmental sustainability gains importance, enhancing supply chains to minimize environmental hazards is essential, particularly in industries using residual biomass. This study tackles this by investigating the integration of sustainability criteria into supply chain optimization for a biomass energy company in Portugal, using a combination of simulation modeling through anyLogistix software (version: 2.15.3.202209061204) and multi-criteria decision-making. Four supply chain scenarios were designed and simulated, differing in their number of distribution centers, the adoption of green logistics, and split-by-ratio distribution strategies over a 305-day period. Through the weighted sum model, Scenario C emerged as the optimal configuration, achieving a balance between operational efficiency and sustainability by reducing CO2 emissions by up to 90% and lowering transportation costs without compromising revenue. Sensitivity analysis further highlighted the trade-offs between cost efficiency, lead times, and environmental impact, showing that the strategic placement of distribution centers and the use of eco-friendly vehicles significantly improve the sustainability of the biomass supply chain. These findings provide practical insights for decision-makers, demonstrating how digital modeling tools can enhance supply chain management by optimizing environmental and operational goals simultaneously. The ceramic pastes of ca. 31 samples recovered from the Almaraz archaeological site, located in the south bench of Tagus River, were studied in detail using XRF, micro-Raman and GSDR spectroscopies, as well as the XRD technique. The ceramic sherds could be grouped into six categories, red slip tableware, decorated tableware, yellow slip tableware, grey tableware, common tableware, and handmade pottery. Our studies of the mineralogic composition of the sherds' body indicate all ceramics were produced locally, using siliceous clays in most cases and calcareous clays in a few ones. Micro-Raman and ground state diffuse reflectance absorption spectroscopy provided useful information regarding the materials used to produce the coloured ceramics: hematite and brookite for the red slip and decorated ceramics, jacobsite or carbon black for the black decoration or grey ceramics. For the yellow slip tableware, a simple engobe rich in yellow clay was used. XRF spectroscopic studies provided the elemental composition of all samples, and biplots of the potassium (K) versus calcium (Ca) contents, normalized to the silicon content of each ceramic paste, clearly show Pliocene and Miocene local clays sources were used to produce most ceramics. Logistics and the supply chain are areas of great importance within organizations. Due to planning gaps, an increase in extra and unnecessary transport costs is usually observed in several companies due to their commercial commitments and need to comply with the delivery time and the batch quantity of products, leading to a negative economic impact. Thus, the objective of this work was to adjust an optimization model to maximize the shipments usually carried out by the companies. To validate the model, an automotive components manufacturer was selected, allowing us to apply the model to a real case study and evaluate the advantages and drawbacks of this tool. It was found that the company to validate the model exports most of its products, and most pallets sent are not fully optimized, generating excessive expense for the company in terms of urgent transport. To solve this problem, two mathematical optimization models were used for the company's current reality, optimizing the placement of boxes per pallet and customer. With the use of the new tool, it was possible to determine that five pallets should be sent to the customer weekly, which correspond to their needs, and that have the appropriate configurations so that the pallet is sent completely. In order to select for further development novel photosensitizers for photodynamic therapy in cutaneous disorders, three unsymmetrical porphyrins, namely 5-(4-hydroxy-3-methoxyphenyl)-10,15,20-tris-(4-acetoxy-3-methoxyphenyl) porphyrin (P2.2), 5-(2-hydroxy-5-methoxyphenyl)-10,15,20-tris-(4-carboxymethylphenyl) porphyrin (P3.2), and 5-(2,4-dihydroxyphenyl)-10,15,20-tris-(4-acetoxy-3-methoxyphenyl) porphyrin (P4.2), along with their fully symmetrical counterparts 5,10,15,20-tetrakis-(4-acetoxy-3-methoxyphenyl) porphyrin (P2.1) and 5,10,15,20-tetrakis-(4-carboxymethylphenyl) porphyrin (P3.1) were comparatively evaluated. The absorption and fluorescence properties, as well as atomic force microscopy measurements were performed to evaluate the photophysical characteristics as well as morphological and textural properties of the mentioned porphyrins. The cellular uptake of compounds and the effect of photodynamic therapy on the viability, proliferation, and necrosis of human HaCaT keratinocytes, human Hs27 skin fibroblasts, human skin SCL II squamous cell carcinoma, and B16F10 melanoma cells were assessed in vitro, in correlation with the structural and photophysical properties of the investigated porphyrins, and with the predictions regarding diffusion through cell membranes and ADMET properties. All samples were found to be isotropic and self-similar, with slightly different degrees of aggregability, had a relatively low predicted toxicity (class V), and a predicted long half-life after systemic administration. The in vitro study performed on non-malignant and malignant skin-relevant cells highlighted that the asymmetric P2.2 porphyrin qualified among the five investigated porphyrins to be a promising photosensitizer candidate for PDT in skin disorders. P2.2 was shown to accumulate well within cells, and induced by PDT a massive decrease in the number of metabolically active skin cells, partly due to cell death by necrosis. P2.2 had in this respect a better behavior than the symmetric P.2.1 compound and the related asymmetric compound P4.2. The strong action of P2.2-mediated PDT on normal skin cells might be an important drawback for further development of this compound. Meanwhile, the P3.1 and P3.2 compounds were not able to accumulate well in skin cells, and did not elicit significant PDT in vitro. Lentinula edodes, commonly known as shiitake mushroom, is renowned for its potential health advantages. This research delves into the often-overlooked by-product of shiitake cultivation, namely spent mushroom substrate (SMS), to explore its nutraceutical properties. The SMS samples were collected and subjected to different extraction methods, namely short or long agitation, and ultrasound-assisted extractions using different temperatures and distilled water or a 50% (v/v) ethanol as solvents. The extracts were tested for phenolic content (total phenols, ortho-diphenols, and flavonoids), antioxidant capacity (DPPH, 2,2-diphenyl-1 picrylhydrazyl; ABTS, 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid; and FRAP, ferric reducing antioxidant power), and antibacterial activity. The different extraction methods revealed substantial variations (p < 0.05) in phenolic composition and antioxidant capacity. The highest phenolic content and antioxidant capacity were achieved using 24 h extraction, agitation, 50 °C, and ethanol as the solvent. Furthermore, the extracted compounds displayed antibacterial activity in specific tested bacterial strains. This study highlights the nutraceutical potential of L. edodes' SMS, positioning it as a valuable dietary supplement for animal nutrition, with emphasis on its prebiotic properties. This study delves into the effect of repeated low-velocity impacts on the residual tensile strength of composite laminates reinforced with E-glass/epoxy woven fabrics. The specimens underwent a series of low-velocity impacts, each delivering a constant energy of 4 J. Various parameters, such as maximum impact load, displacement, contact time, and absorbed energy, were examined. The residual tensile strength was subsequently analysed for each impact and compared to control specimens that experienced no impact. The results highlighted a significant decrease in residual tensile strength after the initial impact, whereas subsequent impacts exhibited a diminished effect until the puncture of the specimens occurred. The literature is not abundant with mechanical characterizations of cylindrical shells for civil engineering applications, especially in terms of impact response. In this context, this study intends to evaluate the impact response of cylindrical sandwich shells produced by various types of fibers. Analysis was performed on three alternative configurations: carbon fibers only, carbon fibers and glass, and carbon fibers and basalt. All configurations were tested for static and impact strength. It was concluded that the constituents of the cylindrical sandwich shells are determinants of both static and impact strength. In terms of compressive properties, the lowest displacement (4.4 mm) and highest compressive strength (873 N) and stiffness (354 N/mm) are attributed to configuration 6C. However, the incorporation of basalt fibers decreased these properties to the lowest values, and reductions of 22% and 44% were found for the compressive strength and stiffness, respectively, while the displacement increased by around 66%. On the other hand, in terms of impact, significant benefits were achieved with the introduction of glass fibers. Compared with configurations 6C and 2C+2B+2C, for instance, the elastic recuperation was 25% and 64.6% higher, respectively. [ABSTRACT FROM AUTHOR]

The fast development and adoption of IoT technologies has been enabling their application into increasingly sensitive domains, such as Medical and Industrial IoT, in which safety and cyber-security are paramount. While the number of deployed IoT devices increases annually, they still present severe cyber-security vulnerabilities, becoming potential targets and entry points for further attacks. As these nodes become compromised, attackers aim to set up stealthy communication behaviours, to exfiltrate data or to orchestrate nodes in a cloaked fashion, and network timing covert channels are increasingly being used with such malicious intents. The IEEE 802.15.4 is one of the most pervasive protocols in IoT and a fundamental part of many communication infrastructures. Despite this fact, the possibility of setting up such covert communication techniques on this medium has received very little attention. We aim to analyse the performance and feasibility of such covert-channel implementations upon the IEEE 802.15.4 protocol, particularly upon the DSME behaviour, one of the most promising for large-scale time critical communications. This enables us to better understand the involved risk of such threats and help support the development of active cyber-security mechanisms to mitigate these threats, which, for now, we provide in the form of practical network setup recommendations. The effect of the interface stiffness and interface strength on the low-velocity impact response of woven-fabric semicylindrical composite shells is studied using finite element (FE) models generated with continuum shell elements and cohesive surfaces. The intralaminar damage is accounted for using the constitutive model provided within the ABAQUS software, while the interlaminar is addressed utilising cohesive surfaces. The results show that the interface stiffness has a negligible effect on the force and energy histories for values between 10 1 N/mm 3 and 2.43 × 10 6 N/mm 3 . However, it has a significant impact on the delamination predictions. It is observed that only the normal interface strength affects the maximum impact force and the delamination predictions. Increasing its value from 15 MPa to 30 MPa resulted in an 8% growth in the maximum force, and a substantial reduction in the delaminated area. The obtained results serve as guidelines for the accurate and efficient computation of delamination. The successful validation of the FE models establishes a solid foundation for further numerical investigations and offers the potential to significantly reduce the time and expenses associated with experimental testing. This paper presents an efficient and reliable approach to study the low-velocity impact response of woven composite shells using 3D finite element models that account for the physical intralaminar and interlaminar progressive damage. The authors' previous work on the experimental assessment of the effect of thickness on the impact response of semicylindrical composite laminated shells served as the basis for this paper. Therefore, the finite element models were put to the test in comparison to the experimental findings. A good agreement was obtained between the numerical predictions and experimental data for the load and energy histories as well as for the maximum impact load, maximum displacement, and contact time. The use of the mass-scaling technique was successfully implemented, reducing considerably the computing cost of the solutions. The maximum load, maximum displacement, and contact time are negligibly affected by the choice of finite element mesh discretization. However, it has an impact on the initiation and progression of interlaminar damage. Therefore, to accurately compute delamination, its correct definition is of upmost importance. The validation of these finite element models opens the possibility for further numerical studies on of woven composite shells and enables shortening the time and expenses associated with the experimental testing. Despite specialists' efforts to find the best solutions for cancer diagnosis and therapy, this pathology remains the biggest health threat in the world. Global statistics concerning deaths associated with cancer are alarming; therefore, it is necessary to intensify interdisciplinary research in order to identify efficient strategies for cancer diagnosis and therapy, by using new molecules with optimal therapeutic potential and minimal adverse effects. This review focuses on studies of porphyrin macrocycles with regard to their structural and spectral profiles relevant to their applicability in efficient cancer diagnosis and therapy. Furthermore, we present a critical overview of the main commercial formulations, followed by short descriptions of some strategies approached in the development of third-generation photosensitizers. Seed hydropriming or nutripriming has been used for wheat biofortification. Previously, the untreated S1 offspring of bread wheat S0 seeds hydro- and nutriprimed with FeSO4.7H2O and/or ZnSO4.7H2O showed improved yield relative to the offspring of untreated S0 seeds. We hypothesize that such improvement would have its origin in the higher quality of S1 seeds resulting from plants whose seeds were primed. In this work, we characterised biochemically the whole-wheat flour of unprimed S1 offspring whose S0 seeds were hydro- and nutriprimed with Fe and/or Zn and compared it to the offspring of untreated S0 seeds (control). We identified and quantified 16 free amino acids and five soluble sugars per offspring using high-performance liquid chromatography and the Association of Official Analytical Chemists (AOAC) methods. The most abundant amino acids were glutamic acid and glutamine, proline, and glycine, presenting their highest contents in the offspring of seeds nutriprimed with 8 ppm Zn (0.351 mmol∙g−1), 8 ppm Fe + 8 ppm Zn (0.199 mmol∙g−1), and (0.135 mmol∙g−1), respectively. The highest contents of glucose (1.91 mg∙g−1 sample), ash (24.90 g∙kg−1 dry matter, DM), and crude protein (209.70 g∙kg−1 DM) were presented by the offspring resulting from 4 ppm Fe + 4 ppm Zn, 8 ppm Zn, and 8 ppm Fe + 8 ppm Zn, respectively. The highest total starch content (630.10 g∙kg−1 DM) was detected in the offspring of seeds soaked in 8 ppm Fe. The nutritional value of the flour of the S1 offspring resulting from nutripriming was significantly higher than the control. Overall, the novelty of our research is that seed priming can improve the quality of the wheat grain and flour, at least till the first offspring, without the need to repeat the presowing treatment. Renewable Energy Communities (RECs) are emerging as an effective concept and model to empower the active participation of citizens in the energy transition, not only as energy consumers but also as promoters of environmentally friendly energy generation solutions, particularly through the use of photovoltaic panels. This paper aims to contribute to the management and optimization of individual and community Distributed Energy Resources (DER). The solution follows a price and source-based REC management program, in which consumers' day-ahead flexible loads (Flex Offers) are shifted according to electricity generation availability, prices, and personal preferences, to balance the grid and incentivize user participation. The heuristic approach used in the proposed algorithms allows for the optimization of energy resources in a distributed edge-and-fog approach with a low computational overhead. The simulations performed using real-world energy consumption and flexibility data of a REC with 50 dwellings show an average cost reduction, taking into consideration all the seasons of the year, of 6.5%, with a peak of 12.2% reduction in the summer, and an average increase of 32.6% in individual self-consumption. The effects of acute caffeine supplementation on muscular strength remain unclear. We examined the effects of two different doses of caffeine on muscle strength and calcium in plasma compared to placebo using a crossover, randomized, double-blind, placebo-controlled design. Twenty-one (n = 21) recreationally resistance-trained participants were randomly assigned into three experimental conditions: 6 mg·kg bw−1 of caffeine (CF6); 8 mg·kg bw−1 of caffeine (CF8); or placebo (PLA), with a 7-day washout period between conditions. Muscular strength assessments were made for both upper (bench press) and lower body muscles (squat and deadlift). Calcium release in plasma was measured on five different occasions. Bench press (CF8: 100.1 ± 1.9 kg; PLA: 94.2 ± 2.5 kg), deadlift (CF8: 132.8 ± 3.5 kg; PLA: 120.7 ± 5.7 kg), and squat (CF8: 130.1 ± 4.9 kg; PLA 119.4 ± 5.4 kg) strength were all significantly (p < 0.001) improved in CF8 compared to PLA. Calcium release in plasma was significantly increased in CF8, whereas no changes were observed in CF6 or PLA. Overall, 8 mg·kg bw−1 of caffeine appears to be an effective dose to optimize upper and lower body muscular strength and calcium release in recreationally trained participants. Simple Summary: Animal production in mountain regions is changing and improvements to maintain autochthonous breeds and traditions should be defined. One of the strategies to add value and improve productivity is to produce meat with a PDO (protected denomination of origin) label. One example in Portugal is the Arouquesa PDO beef. This work aimed to compare different production systems and understand if they affect growth performance, carcass, and meat parameters for the Arouquesa PDO beef. Systems using supplementation had better results regarding live weight and average daily gain. The finishing period increased subcutaneous fat. The meat quality parameters differ in the improved production system with early weaning leading to lesser exudative and cooking losses. In conclusion, the traditional systems improved with practices, such as supplementation, can in turn improve meat production without affecting beef quality or PDO certification. This study demonstrates that improved production systems can advance the Arouquesa autochthonous breed production, while in turn maintaining the valued characteristics of a PDO product. Arouquesa is an autochthonous bovine breed known for its Arouquesa PDO beef labeling. There are several production systems under the definition of PDO labeling. This study aimed to compare the effect of different production systems on carcass and meat traits for the Arouquesa breed. Two trials differing in diet and weaning age were conducted. The first trial included a TF group fed the traditional way and weaned at 9 months; a TF + S1 group, equal to TF, but with a starter supplement; and finally, a S1 + S2 group that was fed with a starter and a growth supplement and weaned at 5 months. The second trial was composed of a TF + S3 group fed like the TF + S1 group but reared until 12 months with a finishing supplement, and finally, the S3 group fed like the S1 + S2 group but reared until 12 months. In the first trial, the TF + S1 and S1 + S2 groups showed higher final live weight and average daily gain. In the second trial, we observed differences in the subcutaneous fat that was higher in the S3 group. Regarding meat traits, we observed differences in exudative and cooking losses in the first trial. In general, supplementation improved meat production without affecting meat quality parameters. Companies are continuously looking to improve their production systems using excellence models, with lean thinking, the Shingo model, six sigma and lean six sigma being the most comprehensive and applied. It is expected that the initial focus for the survival of companies is their economic profitability, but when economic needs are met, the next step is to achieve operational excellence. For this, in addition to economic objectives, it is necessary to include social and environmental objectives, i.e., the other two pillars of sustainability. This study aims to propose a conceptual model identifying the tools that can help achieve the desired results in the three pillars of sustainability aligned with operational excellence. The design of the conceptual model was based on a bibliometric analysis of the literature that relates the concepts of lean thinking, six sigma, lean six sigma and the Shingo model. The Web of Science was the platform selected for the collection of data, and the timeframe considered was 2010 to 2021. A total of 125 articles were analyzed using the VosViewer software, through which it was possible to analyze different topics of study related to the literature. The bibliometric analysis allowed for the identification of the temporal distribution of publications, the categorization of topics, different areas of application and the importance of the tools used in different practical cases. This study points out that companies have at their disposal several tools to achieve economic objectives. On the other hand, there is a set of more restricted tools that are used to meet the objectives of the social and environmental pillars. Future research should focus on identifying tools that meet social and environmental goals in order to strengthen these pillars that are essential for operational excellence and for the sustainability of companies. This paper discusses the integration of emergent ICTs, such as the Internet of Things (IoT), the Arrowhead Framework, and the best practices from the area of condition monitoring and maintenance. These technologies are applied, for instance, for roller element bearing fault diagnostics and analysis by simulating faults. The authors first undertook the leading industry standards for condition-based maintenance (CBM), i.e., open system architecture–condition-based maintenance (OSA–CBM) and Machinery Information Management Open System Alliance (MIMOSA), which has been working towards standardizing the integration and interchangeability between systems. In addition, this paper highlights the predictive health monitoring methods that are needed for an effective CBM approach. The monitoring of industrial machines is discussed as well as the necessary details are provided regarding a demonstrator built on a metal sheet bending machine of the Greenbender family. Lastly, the authors discuss the benefits of the integration of the developed prototypes into a service-oriented platform, namely the Arrowhead Framework, which can be instrumental for the remotization of maintenance activities, such as the analysis of various equipment that are geographically distributed, to push forward the grand vision of the servitization of predictive health monitoring methods for large-scale interoperability. Featured Application: Potential application of unsymmetrical squaraine cyanine dyes as cancer photodynamic therapy photosensitizers. The search to replace conventional cancer treatment therapies, such as chemotherapy, radiotherapy and surgery has led over the last ten years, to a substantial effort in the development of several classes of photodynamic therapy photosensitizers with desired photophysicochemical and photobiological properties. Herein we report the synthesis of 6-iodoquinoline- and benzothiazole-based unsymmetrical squaraine cyanine dyes functionalized with amine groups located in the four-membered central ring. Their photodegradation and singlet oxygen production ability, as well as their in vitro photocytotoxicity against Caco-2 and HepG2 cell lines using a 630.8 ± 0.8 nm centered light-emitting diode system, were also investigated. All photosensitizer candidates displayed strong absorption within the tissue transparency spectral region (650–850 nm). The synthesized dyes were found to have moderate light stability. The potential of these compounds is evidenced by their cytotoxic activity against both tumor cell lines, highlighting the zwitterionic unsubstituted dye, which showed more intense photodynamic activity. Although the singlet oxygen quantum yields of these iodinated derivatives are considered low, it could be concluded that their introduction into the quinoline heterocycle was highly advantageous as it played a role in increasing selective cytotoxicity in the presence of light. Thus, the novel synthesized dyes present photophysicochemical and in vitro photobiological properties that make them excellent photosensitizer candidates for photodynamic therapy. A water-soluble indocarbocyanine dye was synthesized and its photophysics were studied for the first time on two solid hosts, microcrystalline cellulose and β-cyclodextrin, as well as in homogeneous media. The inclusion of the indocarbocyanine moiety onto microcrystalline cellulose increased the dye aggregation with both H and J aggregates being formed. Adsorption on β-cyclodextrin enhanced aggregation in a similar way. The fluorescence quantum yields were determined for the powdered samples of the cyanine dye on the two hosts and a significant increase was observed relative to homogeneous solution. A remarkable concentration dependence was also detected in both cases. A lifetime distribution analysis has shown that the indocarbocyanine dye mainly occupies the amorphous part of cellulose and is not entrapped in the crystalline part of this host. In the β-CD case, the adsorption occurs outside the host cavity. In both hosts a strong concentration quenching effect is observed and only monomers emit. Both adsorptions may be explained by stereochemical constraints imposed by the two long sulphoethyl tails linked to nitrogen atoms of the indocarbocyanine dye. [ABSTRACT FROM AUTHOR]

The photocatalytic degradation of two commercial textile azo dyes, namely C.I Reactive Black 5 and C.I Reactive Red 239, has been studied. TiO2 P25 Degussa was used as catalyst and photodegradation was carried out in aqueous solution under artificial irradiation with a 125 W mercury vapor lamp. The effects of the amount of TiO2 used, UV-light irradiation time, pH of the solution under treatment, initial concentration of the azo dye and addition of different concentrations of hydrogen peroxide were investigated. The effect of the simultaneous photodegradation of the two azo dyes was also investigated and we observed that the degradation rates achieved in mono and bi-component systems were identical. The repeatability of photocatalytic activity of the photocatalyst was also tested. After five cycles of TiO2 reuse the rate of colour lost was still 77% of the initial rate. The degradation was followed monitoring the change of azo dye concentration by UV-Vis spectroscopy. Results show that the use of an efficient photocatalyst and the adequate selection of optimal operational parameters may easily lead to a complete decolorization of the aqueous solutions of both azo dyes. Cu(II) complexes with asymmetrical and symmetrical porphyrinic ligands were synthesized with superior yields using microwave irradiation. The paper presents the synthesis of 5-(3-hydroxyphenyl)-10,15,20-tris-(4-carboxymethylphenyl)-21,23-Cu(II)-porphine in comparison to its symmetrical complex 5,10,15,20-meso-tetrakis-(4-carboxymethylphenyl)-21,23-Cu(II) porphine. The two compounds were characterized by FT-IR, UV-Vis and EPR spectroscopy, which fully confirmed the structures. The spectral molecular absorption properties of the porphyrinic complexes were studied in organic solvents (methanol, ethanol, iso-propanol, dimethyl sulfoxide, dimethylformamide and methylene chloride), and the influence of the solvent polarity on the absorbance maxima is described. In order to establish their future potential in biomedical applications preliminary toxicological studies consisting of viability and proliferation of standard tumor cell lines (MCF7 and B16) testing was performed. In the northwest of the Iberian Peninsula, characterized by its humid climate, large rural areas are being abandoned, mostly in less-favoured areas covered by heathlands, which present a low nutritive quality for livestock production. The high combustibility of these shrublands is driving a high wildfire incidence with negative environmental and economic effects. In this review, some aspects on wildfire occurrence and the potential of grazing livestock to reduce woody phytomass and fire risk in heathland-dominated areas whilst maintaining quality production and preserving biodiversity are summarized. Heathlands may be partially improved—converted to grassland—to better meet animals' nutritional requirements while acting as 'natural' firebreaks. The specific grazing behaviour offers the opportunity to combine different domestic herbivores (mixed grazing) to achieve sustainable systems utilizing heterogeneous resources. Cattle, sheep, goats, and horses may have a role in the provision of different ecosystem services such as food production and biodiversity conservation. Genotype x environment interactions shape the ability of animals to cope with poor vegetation conditions, with smaller species and breeds performing better than larger animals. Goats and horses are indicated to arrest woody encroachment. Sustainable grazing systems are affordable in heathland–grassland mosaics by selecting appropriate livestock species and breeds for quality production, thus favouring rural economies and lowering fire risk. Sarcopenia is one of the main issues associated with the process of aging. Characterized by muscle mass loss, it is triggered by several conditions, including sedentary habits and negative net protein balance. According to World Health Organization, it is expected a 38% increase in older individuals by 2025. Therefore, it is noteworthy to establish recommendations to prevent sarcopenia and several events and comorbidities associated with this health issue condition. Beef cattle production is an important agricultural activity in Brazil, which influences environmental and resource consumption. This study analyzed greenhouse gas (GHG) emission impacts from 17 farms, representing the Brazil's productive system and determined possible improvements in the production chain. Methane, nitrous oxide, and carbon dioxide emissions were evaluated using the updated Intergovernmental Panel on Climate Change (IPCC) guidelines for national inventories. The GHG inventory included emissions from animals, feeds, and "cradle-to-farm-gate" operations for animal management. Regression analyses of carbon dioxide equivalent (CO2eq) emissions and productive indices were performed to identify possible GHG emission hotspots. The results varied considerably among the farms. The GHG yield ranged from 8.63 to 50.88 CO2eq kg carcass−1. The productive indices of average daily gain (p < 0.0001), area productivity (p = 0.058), and slaughtering age (p < 0.0001) were positively correlated with GHG yield. However, no correlation was found with the stocking rate (p = 0.21). The production chain could be improved through accurate animal management strategies that reduce the slaughtering age and daily weight gain individually or per area using pasture management and strategic animal supplementation, which could subsequently reduce GHG emissions in beef cattle production. This article aims to review the present status of anti-flavivirus subunit vaccines, both those at the experimental stage and those already available for clinical use. Aspects regarding development of vaccines to Yellow Fever virus, (YFV), Dengue virus (DENV), West Nile virus (WNV), Zika virus (ZIKV), and Japanese encephalitis virus (JEV) are highlighted, with particular emphasis on purified recombinant proteins generated in bacterial cells. Currently licensed anti-flavivirus vaccines are based on inactivated, attenuated, or virus-vector vaccines. However, technological advances in the generation of recombinant antigens with preserved structural and immunological determinants reveal new possibilities for the development of recombinant protein-based vaccine formulations for clinical testing. Furthermore, novel proposals for multi-epitope vaccines and the discovery of new adjuvants and delivery systems that enhance and/or modulate immune responses can pave the way for the development of successful subunit vaccines. Nonetheless, advances in this field require high investments that will probably not raise interest from private pharmaceutical companies and, therefore, will require support by international philanthropic organizations and governments of the countries more severely stricken by these viruses. Photodynamic therapy is an innovative treatment approach broadly directed towards oncological diseases. Its applicability and efficiency are closely related to the interaction of three main components, namely a photosensitizer, light and molecular triplet oxygen, which should drive cell death. Recently, several studies have demonstrated that squaraine cyanine dyes have a set of photophysical and photochemical properties that have made of these compounds' potential photosensitizers for this therapeutic modality. In the present research work, we describe the synthesis and characterization of four quinoline- and benzoselenazole-derived unsymmetrical squaraine cyanine dyes. Except for the precursor of aminosquaraine dyes, i.e., O-methylated derivative, all dyes were evaluated for their behavior and absorption capacity in different organic and aqueous solvents, their ability to form singlet oxygen, their light-stability, and in vitro phototherapeutic effects against two human breast cancer cell cultures (BT-474 and MCF-7). Regardless of the nature of the used solvents, the synthesized dyes showed intense absorption in the red and near-infrared spectral regions, despite the formation of aggregates in aqueous media. Dyes showed high light-stability against light exposure. Despite the low ability to produce singlet oxygen, aminosquaraine dyes demonstrated worthy in vitro phototherapeutic activity. Photodynamic therapy is a medical modality developed for the treatment of several diseases of oncological and non-oncological etiology that requires the presence of a photosensitizer, light and molecular oxygen, which combined will trigger physicochemical reactions responsible for reactive oxygen species production. Given the scarcity of photosensitizers that exhibit desirable characteristics for its potential application in this therapeutic strategy, the main aims of this work were the study of the photophysical and photochemical properties and the photobiological activity of several dicyanomethylene squaraine cyanine dyes. Thus, herein, the study of their aggregation character, photobleaching and singlet oxygen production ability, and the further application of the previously synthesized dyes in Caco-2 and HepG2 cancer cell lines, to evaluate their phototherapeutic effects, are described. Dicyanomethylene squaraine dyes exhibited moderate light-stability and, despite the low singlet oxygen quantum yields, were a core of dyes that exhibited relevant in vitro photodynamic activity, as there was an evident increase in the toxicity of some of the tested dyes exclusive to radiation treatments. The use of ionizing radiation processing technologies has proven to be one of the most versatile ways to prepare a wide range of membranes with specific tailored functionalities, thus enabling them to be used in a variety of industrial, environmental, and biological applications. The general principle of this clean and environmental friendly technique is the use of various types of commercially available high-energy radiation sources, like 60Co, X-ray, and electron beam to initiate energy-controlled processes of free-radical polymerization or copolymerization, leading to the production of functionalized, flexible, structured membranes or to the incorporation of functional groups within a matrix composed by a low-cost polymer film. This study aimed to optimize bean flours fermentation through the use of appropriate technological procedure and, thereby, to obtain a high quality and safe product. In this line, cowpea bean flours with different moisture conditions (10, 20 and 30%) were incubated with (1) a single culture of Lactobacillus plantarum, or (2) a consortium of lactic acid and acetic acid bacteria, together with one strain of Saccharomyces cerevisiae. Effects of inoculation of cowpea beans flours on stability (i.e., evaluated by the decrease in pH), and variations in nutritional characteristics (i.e., protein, starch, water soluble carbohydrates, total dietary fibre) were investigated. In both fermented flours, the effect of fermentation was more noticeable in total water-soluble carbohydrate (WSC) concentration during the fermentation process (P < 0.001), accounted for by metabolic activity of the microorganisms. The pH values progressively decreased (P < 0.001) through the fermentation process, particularly in flours fermented with a single culture of L. plantarum. By contrast, titratable acidity increased (P < 0.001) throughout the fermentation process in F2 and F3, although more noticeable in F3. Total dietary fibre (TDF) was not variable over the time. In relation to the protein content, the fermentations behaved very similarly; F2 had a variation over the time, but the effect was not significant (P = 0.0690). Results revealed small changes in chemical composition except in the case of pH and sugar contents with the values lower in the fermented products (i.e., single- or mixed-culture fermentation), leading to a more stable and safety product. Transforming growth factor beta (TGF-β) is a determinant for inflammation and fibrosis in cardiac and skeletal muscle in Chagas disease. To determine its regulatory mechanisms, we investigated the response of Trypanosoma cruzi-infected cardiomyocytes (CM), cardiac fibroblasts (CF), and L6E9 skeletal myoblasts to TGF-β. Cultures of CM, CF, and L6E9 were infected with T. cruzi (Y strain) and treated with TGF-β (1–10 ng/mL, 1 h or 48 h). Fibronectin (FN) distribution was analyzed by immunofluorescence and Western blot (WB). Phosphorylated SMAD2 (PS2), phospho-p38 (p-p38), and phospho-c-Jun (p-c-Jun) signaling were evaluated by WB. CF and L6E9 showed an increase in FN from 1 ng/mL of TGF-β, while CM displayed FN modulation only after 10 ng/mL treatment. CF and L6E9 showed higher PS2 levels than CM, while p38 was less stimulated in CF than CM and L6E9. T. cruzi infection resulted in localized FN disorganization in CF and L6E9. T. cruzi induced an increase in FN in CF cultures, mainly in uninfected cells. Infected CF cultures treated with TGF-β showed a reduction in PS2 and an increase in p-p38 and p-c-Jun levels. Our data suggest that p38 and c-Jun pathways may be participating in the fibrosis regulatory process mediated by TGF-β after T. cruzi infection. [ABSTRACT FROM AUTHOR]

In this work, several benzothiazole-based aminosquaraine dyes, displaying strong absorption within the so-called phototherapeutic window (650–800 nm), were synthesized. The ability, of all the new dyes, to generate singlet oxygen was assessed by determining the correspondent phosphorescence emission and through the comparison with a standard. The quantum yields of singlet oxygen generation were determined and exhibited to be strongly dependent on the nature of the amino substituents introduced in the squaric ring. The photodynamic activity of the synthesized dyes was tested against four human tumor cell lines: breast (MCF-7), lung (NCI-H460), cervical (HeLa) and hepatocellular (HepG2) carcinomas; and a non-tumor porcine liver primary cell culture (PLP2). All the compounds synthesized were found to be able to inhibit tumor cells growth upon irradiation more than in the dark, in most of the cases, very significantly. Considering the photodynamic activity exhibited and the low toxicity displayed for the non-tumor cells, some of the synthetized dyes can be regarded as potential candidates as photosensitizers for PDT.
