This paper examines the socio-economic significance of forest visits and the collection of forest berries and mushrooms (FBMs) in the Czech Republic, emphasising their role in enhancing human well-being and contributing to regional economies. Over a 30-year period, data were collected on the quantities and economic values of FBMs, alongside the intensity of forest visits by the Czech population. This study incorporates a detailed analysis of time series data on FBM collection, exploring trends and fluctuations in the harvested quantities and their economic value. A Lorenz curve analysis reveals significant disparities in the distribution of economic benefits, with a small segment of the population accounting for the majority of the FBM-derived value. Additionally, the research investigates the impact of forest visitation on well-being at the regional level, highlighting the relationship between forest access, visitation intensity, and public health benefits. This study also examines visitors' expectations, motivations, and perceptions regarding an ideal forest for visitation, providing recommendations for effective marketing strategies. Furthermore, the study explores the contribution of FBMs to net income across different regions, demonstrating substantial regional variation in their economic importance. Notably, the analysis shows that the value of FBMs represents approximately 37% of the net income generated by traditional forestry activities, underscoring its significant economic potential. The findings emphasise the potential of territorial marketing strategies to enhance well-being, particularly in economically disadvantaged regions, and advocate for sustainable forest management practices to protect these valuable resources and ensure equitable access to the benefits provided by forest ecosystems. The article presents a methodology for the management of repairs to property damaged by a disaster. The article describes the procedure for the development of an organizational layout, along with planning and managing activities leading to the restoration of damaged property. Last but not least, the article deals with financial planning. The methodology presented was applied to a case study that was conducted in the village of Hrušky, South Moravian region, the Czech Republic, which was hit by a tornado in 2021. More than three years have passed since this tragedy, and repairs to the village's property are still planned to continue until the middle of 2025. Qualitative approaches to data collection and evaluation were used for data collection, especially in-depth interviews related to the given issue. The mayor of the village, the village management, and representatives of construction companies were involved in the in-depth interviews. Another important source of data was the project documentation and the municipality's accounting records. The main outputs include a list of construction works, which served as a basis for the financial damages; the municipality's participation in state subsidy programs for the financing of the restoration; and, last but not least, a list of works that was used to create the project documentation for the reconstruction of individual buildings. Other outputs of the work include a structural plan, which has been developed for the purposes of planning and managing activities leading to the restoration of the damaged area, and a financial plan, which contains information on expenses and funding sources. The key contribution of the paper is a methodical procedure for the identification, the design, and the execution of crucial activities connected with the restoration of the territory after a disaster. The methodological procedure is designed considering the experience associated with a specific disaster, namely, a tornado in the southeastern part of the Czech Republic, which served as the basis for this case study. Background: The SARS-CoV-2 pandemic has become the greatest public health challenge worldwide. Soon after the appearance of the virus in 2019, intensive efforts to develop vaccines were initiated, and by late 2020, delivery of vaccines for the targeted population as a campaign had started. Aim: Collect information from European Union countries regarding how and to what extent were family physicians (FPs)/general practitioners (GPs) involved in the vaccination campaigns in 2021 and how these were organized at the national level. Method: A short questionnaire was distributed through the secretariats of WONCA (World Organization of Family Doctors) Europe and the European Forum for Primary Care (EFPC). Results: In most of the countries, participation of FPs/GPs was compulsory. The vaccination was usually centrally organized by governmental authorities. In the beginning, registration (web-based) of patients was required, mainly at the national level. By the middle of 2021, vaccination on a walk-in basis became available in almost every country for the first immunization as well as for the booster injections. The remunerations of GPs/FPs differed; in some countries, no extra payments were offered. The Pfizer vaccine was used in all countries, while in nine countries, non-European Medicines Agency (EMA)-approved vaccines were also given in primary care settings and at vaccination centers. In some countries, professional homepages helped the GPs. The involvement of primary health care (PHC) providers did not correlate to the vaccination coverage of the entire population of the respective countries. It was the highest in the more developed countries with higher living standards, where participation of GPs was voluntary and appropriate financial incentives were offered to them. Conclusions: The vaccination campaign was a professional and logistic challenge and an excellent performance of PCH providers. The cabbage whitefly has become an important pest on brassica vegetables in Central Europe. It does not destroy the affected plants, but the product becomes unmarketable, causing considerable economic losses. The pest is also difficult to control due to its way of life and because it develops resistance to some of the active components of insecticides. In organic farming systems, insecticides are strictly restricted, but neither predators nor whitefly parasitoids are able to keep the pest at a tolerable level. It is, therefore, necessary to become familiar with the whitefly's life cycle and habits, including mass migration from winter hosts to vegetables. We inspected 44 rapeseed fields across the republic in the period 2014–2021 in order to find the connection between the presence of oilseed rape fields near vegetable growing areas (VGAs) and the abundance of the overwintering cabbage whiteflies. We also conducted regular weekly monitoring of whitefly occurrence in the main cultivation area of the Czech Republic (Polabí) with the aim of specifying critical data important for the successful control of this pest. We found that the cabbage whitefly incidences were many times higher in rapeseed fields close to VGAs compared to areas where the crops are not adjacent. The average number of whiteflies was 0.59 individuals per plant in VGA-1 (oilseed rape grown inside this area or up to 1 km far), 0.052 in VGA-2 (distance 3–10 km from vegetable fields) and 0.014 in VGA-3 (more than 20 km). In the extremely warm year 2016, the difference was up to sixty times. The first CW eggs laid on cruciferous vegetables were usually found around 20 May. The period of mass migration of CW adults to cruciferous vegetables was between 6 June and 2 August. At this time, vegetables are most vulnerable to damage. Successful control of the cabbage whitefly requires the use of fabric netting, combined with an insecticide as needed and trap plants as needed; the latter have to be destroyed before adult whiteflies hatch—typically in early July. The aim of this study is to provide a comprehensive view of matters of cross-border cooperation, with the main emphasis on the typology and development of structures supporting cross-border cooperation in a selected region of the Czech Republic (South Bohemian Region). The study focuses on the analysis of changes in the methods and ways of cooperation of municipalities in the programming periods 2007–2013 and 2014–2020 (Euroregions, cross-border impulse centers, European territorial cooperation groups, partner and friendly cities, etc.). The research not only analyzes the actual development of cooperation between municipalities in the region studied, but also identifies the prerequisites of and barriers to cross-border cooperation. The data obtained show a significant decrease in the interest of municipalities in cross-border cooperation within the conditions of the Czech Republic in recent years. The current situation with regard to the existence and development of cross-border cooperation in 2024 is not very satisfactory, although it is paradoxically an area strongly supported by the EU Cohesion Policy. This study employs a mixed-methods approach, combining both qualitative and quantitative research methods to provide a comprehensive analysis of cross-border cooperation in the South Bohemian Region. A thorough review of existing literature on cross-border cooperation and related policies was conducted to establish a theoretical framework. The practical part of the study is based on field research carried out by the author in 2007–2013 and 2014–2020. This research examined the level of cooperation between the 623 municipalities in the South Bohemian Region (in 2016, there were 624 municipalities in the region). In the case of cross-border impulse centers, the input data were represented by e-mail and telephone inquiries to representatives of individual municipalities and the websites of Euroregions. To conduct a comprehensive study on cross-border cooperation between municipalities in the South Bohemian Region, logical and scientific methods were used. Universal methods of logic ensured the correctness and orderliness of the thought process, with abstraction identifying key elements. Synthesis was used to divide the study into parts, analyze them in detail, and integrate them into a comprehensive analysis. Scientific methods included analysis to examine the current status and frameworks of cross-border cooperation and comparison to identify similarities or differences between the Czech Republic and neighboring countries. Honey contains a wide range of inorganic substances. Their content can be influenced, i.e., by the type of soil on which the bee pasture is located. As part of this study, the mineral profile of 32 samples of honey from hobby beekeepers from the Czech Republic wasevaluated and then compared with soil types in the vicinity of the beehive location. Pearson's correlation coefficient was used to express the relationship between mineral substances and soil type. There was a high correlation between antroposol and Zn (R = 0.98), Pb (R = 0.96), then between ranker and Mn (0.95), then regosol and Al (R = 0.97) (p < 0.05). A high negative correlation was found between regosol and Mg (R = −0.97), Cr (R = −0.98) and between redzinas and Al (R = −0.97) (p < 0.05). Both positive and negative high correlations were confirmed for phaeozem. The CART method subsequently proved that the characteristic elements for individual soil types are B, Ca, Mg, Ni, and Mn. The soil types of cambisol, fluvisol, gleysol, anthrosol, and kastanozem had the closest relationship with the elements mentioned, and it can therefore be assumed that their occurrence indicates the presence of these soil types within the range of beehive location. Gyrophoric acid (GA), a lichen secondary metabolite, has attracted more attention during the last years because of its potential biological effects. Until now, its effect in vivo has not yet been demonstrated. The aim of our study was to evaluate the basic physicochemical and pharmacokinetic properties of GA, which are directly associated with its biological activities. The stability of the GA in various pH was assessed by conducting repeated UV-VIS spectral measurements. Microsomal stability in rat liver microsomes was performed using Ultra-Performance LC/MS. Binding to human serum albumin (HSA) was assessed using synchronous fluorescence spectra, and molecular docking analysis was used to reveal the binding site of GA to HSA. In the in vivo experiment, 24 Sprague-Dawley rats (Velaz, Únetice, Czech Republic) were used. The animals were divided as follows. The first group (n = 6) included healthy males as control intact rats (♂INT), and the second group (n = 6) included healthy females as controls (♀INT). Groups three and four (♂GA/n = 6 and ♀GA/n = 6) consisted of animals with daily administered GA (10 mg/kg body weight) in an ethanol-water solution per os for a one-month period. We found that GA remained stable under various pH and temperature conditions. It bonded to human serum albumin with the binding constant 1.788 × 106 dm3mol−1 to reach the target tissue via this mechanism. In vivo, GA did not influence body mass gain, food, or fluid intake during the experiment. No liver toxicity was observed. However, GA increased the rearing frequency in behavioral tests (p < 0.01) and center crossings in the elevated plus-maze (p < 0.01 and p < 0.001, respectively). In addition, the time spent in the open arm was prolonged (p < 0.01 and p < 0.001, respectively). Notably, GA was able to pass through the blood–brain barrier, indicating its ability to permeate into the brain and to stimulate neurogenesis in the hilus and subgranular zone of the hippocampus. Carbon dioxide capture and storage (CCS) is a necessary requirement for high-emitting CO2 industries to significantly reduce volumes of greenhouse gases released into the atmosphere and mitigate climate change. Geological CO2 storage into depleted oil and gas fields is the fastest and most accessible option for CCS deployment allowing for re-purposing existing infrastructures and utilizing significant knowledge about the subsurface acquired during field production operations. The location of such depleted fields in the neighborhoods of high-emitting CO2 industries is an additional advantage of matured on-shore European fields. Considering these advantages, oil and gas operators are now evaluating different possibilities for CO2 sequestration projects for the fields approaching end of production. This article describes an integrated approach to reservoir simulations focused on evaluating a CO2 injection pilot at one of these matured fields operated by MND and located in the Czech Republic. The CO2 injection site in focus is a naturally fractured carbonate reservoir. This oil-bearing formation has a gas cap and connection to a limited aquifer and was produced mainly by pressure depletion with limited pressure support from water injection. The article summarizes the results of the efforts made by the multi-disciplinary team. An integrated approach was developed starting from geological modeling of a naturally fractured reservoir, integrating the results of laboratory studies and their interpretations (geomechanics and geochemistry), dynamic field data analysis (pressure transient analysis, including time-lapse) and history matching reservoir model enabling simulation of the pilot CO2 injection. The laboratory studies and field data analysis provided descriptions of stress-sensitive fracture properties and safe injection envelope preventing induced fracturing. The impact of potential salt precipitation in the near wellbore area was also included. These effects are considered in the context of a pilot CO2 injection and addressed in the reservoir simulations of injection scenarios. Single-porosity and permeability reservoir simulations with a dominating fracture flow and black-oil formulation with CO2 simulated as a solvent were performed in this study. The arguments for the choice of the simulation approach for the site in focus are shortly discussed. The reservoir simulations indicated a larger site injection capacity than that required for the pilot injection, and gravity-driven CO2 migration pathway towards the gas cap in the reservoir. The application of the approach to the site in focus also revealed large uncertainties, related to fracture description and geomechanical evaluations, resulting in an uncertain safe injection envelope. These uncertainties should be addressed in further studies in preparation for the pilot. The article concludes with an overview of the outcomes of the integrated approach and its application to the field in focus, including a discussion of the issues and uncertainties revealed. Regions densely populated with archaeological monuments pose significant challenges for construction investors and archaeologists during the planning stages of major construction projects. Recognising the archaeological potential of these areas is crucial for planning effective rescue excavations, which have become a standard procedure in construction. This study explores the utility of non-invasive prospection techniques, including artefact field surveys, multispectral imaging, and magnetic surveys, in assessing the area chosen for the I/16 Slaný–Velvary road construction. We specifically focused on the contributions these methods make towards understanding the archaeological context of the proposed construction site. The findings from the magnetic survey were compared with the results of actual archaeological excavations. Through manual visual analysis and statistical spatial correlation, we assessed the effectiveness of the magnetic survey. The ability of magnetic survey to locate different types of archaeological objects proved to be dependent on various factors. The variability of the environmental setting, particularly pedological and geological conditions, is essential. However, the modern anthropogenic impact and the very nature of individual archaeological objects, especially their dimensions, also play an essential role. [ABSTRACT FROM AUTHOR]

This paper describes the in situ spores of the Calamospora type, macerated from sixty-one specimens of calamitean cones belonging to sixteen species of genera, such as the Palaeostachya, Macrostachya, Calamostachys, and Huttonia from the Pennsylvanian Czech Republic period, specifically from the Moscovian/Kasimovian ages (i.e., Duckmantian-Stephanian). The in situ spores were compared to twenty dispersed species of Calamospora. The majority of spores were microspores; however, some cones yielded both micro- and megaspores. Morphological variations of the in situ spores, including the diameter, labrum, contact area, ontogenetic stages, and secondary folds of the exine, are described, including their importance for the classification of calamospores. The relationships of Elaterites, Pteroretis, Vestispora, and some monopseudosaccate spores are discussed. All Paleozoic Calamospora-producing parent plants are summarized. [ABSTRACT FROM AUTHOR]

Diabetes mellitus is a complex, multifactorial, progressive condition with a variety of approved therapeutic options. The purpose of this study was to offer an overview of the authorized antidiabetic medicines (excluding insulin) compared with marketed products in seven European countries. Data were obtained from primary sources, including the websites of national authorities and directly from specialists in the countries of interest. The range of marketed medicines compared with the authorized group was assessed in terms of active pharmaceutical ingredients (>60% in Bulgaria, France, Serbia), brand names (>70% in Bulgaria, the Czech Republic, Romania, Serbia, Spain), pharmaceutical forms (>60% in all countries), strengths (>60% in Bulgaria, the Czech Republic, Romania, Serbia, Spain), marketing authorization holder (≥50% in all countries) and the status of medicine. Spain was found to have the highest number of products based on most of these attributes. Over 90% of authorized medicines had a pharmacy price in Serbia. Regarding the newer class of GLP-1 receptor agonists, a retail price for all approved substances was available in Bulgaria, Romania, Serbia, and Spain. Only one brand name with one concentration was found available for some agents, being susceptible to drug shortages: glibenclamide (Romania, Serbia, Spain), glipizide (the Czech Republic, Poland, Romania, Spain), glisentide (Spain), acarbose (the Czech Republic), sitagliptin (Bulgaria, Poland), vildagliptin (the Czech Republic, Poland) and saxagliptin (the Czech Republic, France, Romania, Serbia). An overview of the national and international therapeutic options may allow competent authorities and health professionals to take rapid measures in case of supply problems or health crises. Mineralogy and formation conditions were studied in a newly found vein wolframite mineralization, cutting migmatitized paragneisses in the exocontact of a small Carboniferous granite body in the Pohled quarry, Moldanubian Zone of the Bohemian Massif, Czech Republic. The early stage of the rich mineral assemblage (36 mineral species) involves wolframite, columbite-group minerals, molybdenite, and scheelite hosted by quartz–muscovite–chlorite gangue, which was followed by base-metal sulfides in a quartz gangue, whereas the last stage included calcite gangue with fluorite and minor sulfides. The mineral assemblage points to the mobility of usually hardly soluble elements, including W, Sn, Zr, Nb, Th, Ti, Sc, Y, and REEs. A fluid inclusion study indicates a significant decrease in homogenization temperatures from 350–370 °C to less than 100 °C during vein formation. Fluids were aqueous, with a low salinity (0–12 wt. % NaCl eq.) and traces of CO2, N2, CH4, H2, and C2H6. The δ18O values of the fluids giving rise to quartz and scheelite are positive (min. 4‰–6‰ V-SMOW). The Eh and pH of the fluid also changed during evolution of the vein. Both wolframite and columbite-group minerals are anomalously enriched in Mg. We suggest that the origin of this distinct mineralization was related to the mixing of Mo,W-bearing granite-derived magmatic fluids with external basinal waters derived from contemporaneous freshwater (but episodically evaporated) piedmont basins. The basinal waters infiltrated into the subsurface along fractures formed in the extensional tectonic regime, and their circulation continued even after the ending of the activity of magmatic fluids. The studied wolframite mineralization represents the most complete record of the 'hydrothermal' history of a site adjacent to a cooling granite body in the study area. Moreover, there are broad similarities in the mineral assemblages, textures, and chemical compositions of individual minerals from other occurrences of wolframite mineralization around the Central Moldanubian Plutonic Complex, pointing to the genetic similarities of the Variscan wolframite-bearing veins in this area. The aim of this study was to characterize the influence of CO2 in geological structures on mineralogical changes in rocks and assess the sequestration capacity in mineral form and solution of a potential pilot storage site in the Czech Republic. Rock samples from a dolomite reservoir and the overburden level, as well as the corresponding pore water, were used. The most important chemical process occurring in the reservoir rock is the dissolution of carbonate minerals and feldspars during the injection of CO2 into the structure, which increases the porosity of the structure by approximately 0.25 percentage points and affects the sequestration capacity of the reservoir rock. According to the results of geochemical modeling, the secondary carbonate minerals (dolomite, siderite, and ephemeral dawsonite) were present only during the first 50 years of storage, and the porosity at this stage decreased by 1.20 pp. In the caprocks, the decomposition of K-feldspar and calcite resulted in an increase in porosity by 0.15 percentage points at the injection stage only, while no changes in porosity were noted during storage. This suggests that their insulation efficiency can be maintained during the injection and post-injection periods. However, further experimental research is needed to support this observation. The results of this study indicate that the analyzed formation has a low potential for CO2 sequestration in mineral form and solution over 10,000 years of storage, amounting to 5.50 kg CO2/m3 for reservoir rocks (4.37 kg CO2/m3 in mineral form and 1.13 kg CO2/m3 in dissolved form) and 3.22 kg CO2/m3 for caprock rocks (3.01 kg CO2/m3 in mineral form and 0.21 kg CO2/m3 in dissolved form). These values are lower than in the case of the depleted Brodské oil field, which is a porous reservoir located in the Moravian part of the Vienna Basin. [ABSTRACT FROM AUTHOR]

This article deals with the liability of the supervisory authority of the financial market. It could be questioned whether the supervisory authority, as the public authority, is liable for the supervisory performance. If the answer is yes, then the question is what kind of liability could be found and if any special conditions (prerequisites) are needed. In general, there could be two lines of public liability found. One is systemic liability for the safe financial market—e.g., financial market stability. The other perspective is individual liability for damages caused by unlawful administrative procedure or maladministration, where unlimited strict liability is granted. This kind of liability might be widely questioned, especially when the central bank is the supervisory authority, like in the Czech Republic, Slovakia, Hungary, and other EU member states. The use of new technologies in tourism is bringing about a genuine revolution in the sector, where automated services, such as chatbots, are increasingly being used to perform some of the tasks involved in service delivery. However, the emergence of new technologies in a sector as globalized as tourism can mean that not all users are able to perceive the benefits of these innovations. Therefore, the aim of this study is to determine whether the digital literacy (DL) of a tourist may lead to different evaluations of the quality of the electronic service they received, both overall and as per the different dimensions of quality. This study compares a more innovative and interactive service-provision technology (a chatbot) with a more traditional one (email). To this end, an experiment was conducted in which 124 participants from Spain (higher DL) and the Czech Republic (lower DL) were asked to interact with these two technologies in a simulated hotel customer-service scenario. The results show that individuals with higher DL rated the quality of service received via chatbot higher than individuals with lower DL. The latter prefer email because they perceived it to provide greater security, empathy, reliability, and information quality. However, participants with higher DL preferred the chatbot due to its greater competence in completing the task. Finally, the participants rated the responsiveness of the chatbot higher than that of email. These results can help the introduction of chatbot-based customer service in the tourism sector. Recently, an outbreak of Ips sexdentatus (Börner, 1776) has caused considerable damage in the pine forests of the Czech Republic. As historical data on the biology of this pest are scarce due to its rare occurrence in recent decades, our work focused on monitoring flight activity and voltinism and investigating methods for monitoring its activity during the growing season. Observations were conducted from March to September 2021 and 2022 at three sites using 12 Theysohn traps with four types of pheromone lures (ACUMIPROTECT, ACUWIT, SEXOWIT and Pheagr IAC) together with data loggers to record weather conditions. The first beetles occurred in early May (daily mean temperatures above 13 °C). After the first egg laying stage, females re-emerged to establish a sister brood. The maximum flight activity appeared between late June and mid-July (daily mean temperatures about 20 °C), and the offspring occurred at the turn of June/July and in the first half of August. Since then, flight activity had a downward trend and quietened in September. According to the data, monitoring of I. sexdentatus should be conducted between May and September using the ACUMIPROTECT pheromone bait exhibiting the highest capturing efficacy. Using the liverwort Hygrobiella laxiflora as an example, this study aims to investigate the impact of climate change on montane species and their ecosystems. The study area is the Elbe Sandstone Mountains in Germany and the Czech Republic, where Hygrobiella laxifolia has an isolated occurrence in deeply carved gorges characterized by a specific microclimate (cellar climate). The focus is on determining the rate at which populations are affected. After little change in the population over a long period, a sudden sharp decline has been observed in recent years. The decline correlates with the mass proliferation of the European spruce bark beetle (Ips typographus) observed since 2017, which has led to a strong thinning of the forests in the area. In many areas of occurrence, the forests now have a much sparser tree layer or none at all. Competition from other, more competitive species also plays an important role as a cause of decline, as these have spread more widely on the sites due to the now higher light levels. The severe bark beetle infestation and the large-scale forest fire of 2022 both represent events that led to the almost complete deforestation of large parts of the study area, which was previously unknown on this scale. Since both the bark beetle infestation and the forest fire are strongly intensified by climate change, we assume that climate change is the main reason for the decline of Hygrobiella laxifolia in the area. The populations of Hygrobiella laxifolia of the Elbe Sandstone Mountains show some morphological peculiarities. As two new species have recently been described within Hygrobiella, we have integrated a sample from the area into a molecular phylogeny of Hygrobiella based on the plastid sequences of trnL-trnF intergenic spacer and matK to clarify the relationships. The planned pilot CO2 storage Zar-3 is an oil field with a gas cap in the final production stage in the SE Czech Republic. It is composed of a dolomite Jurassic reservoir sealed by three different formations that differ significantly in lithology. Previous studies left open questions on the nature of pore space and connectivity and the quality of the seal in the future CO2 storage complex. Microscopic petrography of the reservoir suggests dolomitisation in shallow water followed by karstification and brecciation with fracture-correct-dominated porosity. The seal horizons have porosity limited to the micro- and nanoscales. The oil consists of significantly biodegraded black oil of Jurassic origin mixed with less biodegraded gasoline-range hydrocarbons. Biomarkers in the caprock bitumens trapped in nanopores show a genetic relationship to the reservoir oil. Gas in the not yet fully depleted gas cap of the field is of thermogenic origin with no contribution of microbial methane. The formation water has total dissolved solids typical of isolated brines not diluted by infiltrated fresh water. The geochemical characteristics of the storage system together with the fact that the initial oil column is about 105 m tall with another 150 m of gas cap suggest that the seals are efficient and the Zar-3 future storage complex is tight and safe. The climate challenges posed by the European Union are particularly difficult for emerging and developing economies. This causes reluctance to undertake them and even denial of their validity. For this reason, this article seeks to answer the question of whether it is possible to carry out an effective transformation of energy resources in the developing economies of the Visegrad Group. Analyses in this area are conducted in a long-term, 58-year research perspective (1965–2022) and concern the Czech Republic, Hungary, Poland, and Slovakia. The effectiveness of the examined transformation is assessed in two dimensions as follows: (1) resource and (2) emission, which are then synthesized using a multi-criteria analysis. This research shows that an effective resource transformation aimed at a zero-emission economy is possible even in emerging and developing economies, although it undoubtedly requires time and consistency in the implementation of the once-chosen energy policy. Hungary—a leader in assessed climate effectiveness—has systematically decarbonized while reducing the use of total non-renewable resources. The Hungarian resource transition strategy assumed the use of nuclear energy and an increase in the share of renewable resources. The result of these activities is a quite diversified energy mix, which is greater resource self-sufficiency and low carbon dioxide emissions. The Czech Republic also achieved a similar strategy and results. Poland and Slovakia coped much worse with the climate challenge: although they gave up coal, they replaced it largely with other non-renewable resources. It is worth emphasizing that Poland has managed to significantly increase the use of renewable resources, and nuclear energy and hydropower have also appeared in the Slovak mix. This study examines the strategic use of life cycle cost analyses (LCCAs) in the management and conservation of heritage sites, emphasizing the need for comprehensive financial planning. With an increasing number of heritage sites showing signs of deterioration, our aim was to improve the sustainability and effectiveness of restoration practices. We used dynamic life cycle costing methods and developed the MONUREV software V2 to simulate different restoration scenarios, providing accurate, data-driven projections for maintaining structural, functional and aesthetic integrity. The field research involved testing these methods through case studies of heritage buildings in the Czech Republic, focusing on holistic cost management from initial analysis to practical application. The results showed that LCC analysis can significantly assist in making informed decisions, balancing economic and cultural values, and ensuring long-term conservation outcomes. This study concludes that the integration of a detailed LCC analysis into heritage conservation strategies represents a methodological advance that can significantly improve the economic and operational planning of the maintenance of heritage buildings, thereby ensuring their preservation for future generations. The profile of secondary metabolites present in the apple cuticular layer is not only characteristic of a particular apple cultivar; it also dynamically reflects various external factors in the growing environment. In this study, the possibility of authenticating apple samples by analyzing their cuticular layer extracts was investigated. Ultra-high-performance liquid chromatography coupled with high-resolution tandem mass spectrometry (UHPLC-HRMS/MS) was employed for obtaining metabolomic fingerprints. A total of 274 authentic apple samples from four cultivars harvested in the Czech Republic and Poland between 2020 and 2022 were analyzed. The complex data generated, processed using univariate and multivariate statistical methods, enabled the building of classification models to distinguish apple cultivars as well as their geographical origin. The models showed very good performance in discriminating Czech and Polish samples for three out of four cultivars: "Gala", "Golden Delicious" and "Idared". Moreover, the validity of the models was tested over several harvest seasons. In addition to metabolites of the triterpene biosynthetic pathway, the diagnostic markers were mainly wax esters. Bark beetle outbreaks damage forests and kill trees worldwide, but many aspects of their dynamics remain unexplained. Our aim was to identify predictors for individual tree deaths within the small (0.7 km2) Plešné Lake catchment in the Šumava National Park in southwestern Czechia. Within this area, >60,000 trees were geo-referenced and categorized from ten aerial images (20 cm spatial resolution) between 2000 and 2015. For each year for which aerial images were available, we calculated tree densities of different categories and diameters. Tree height was evaluated by means of LiDAR in two terrestrial campaigns (2010 and 2011). A machine learning technique was then used to evaluate the most important variables. The resulting relationships were largely nonlinear and differed among years; however, individual trait tree height proved to be the most influential variable in each year. Higher trees were more likely to have died during either the undisturbed phase (2000 and 2003), the disturbed phase (2005–2011), or the recovery phase (2013). Norway spruce (Picea abies) forests in temperate zones are already reacting to short-term extreme summer heatwaves, threatening the vitality of trees and forest productivity, and can even lead to local and regional dieback events. Examining quantitative wood anatomy can provide helpful information in terms of understanding the physiology mechanisms and related responses of conifer trees to local environmental interactions in relation to tracheid adaptive capacity. This study analysed the tracheid functional anatomical traits (FATs) plasticity of six young Norway spruce trees growing in two mesic research plots with high annual precipitation (~43%) and air temperature differences during 2010–2017. The research plots are located in the sub-mountainous (Rájec Němčice) and mountainous (Bílý Kříž) belts of the Moravia region, Czech Republic. Vapour pressure deficit and cell wall reinforcement index (CWRI) were shown to be the most representative environmental parameters as proxies of dry conditions. Tracheid FATs indicated latewood phenological plasticity sensitivity, with more pronounced variability in the warmer and drier plots. Latewood tracheids of Norway spruce trees grown in the RAJ formed significantly thicker cell walls than BK during the studied period. The observed differences between the two research plots indicate additional support for tracheid cells' hydraulic safety against cavitation and potential traces of adaptive acclimatization response. Caseous lymphadenitis (CLA) is a worldwide disease of small ruminants caused by Corynebacterium pseudotuberculosis, a facultative intracellular pathogen that is able to survive and multiply in certain white blood cells of the host. In this study, 33 strains of C. pseudotuberculosis were isolated from sheep and goats suffering from CLA on nine farms in the Czech Republic. All these strains were tested for their antibiotic susceptibility, ability to form a biofilm and resistance to the effects of commonly used disinfectant agents. To better understand the virulence of C. pseudotuberculosis, the genomes of strains were sequenced and comparative genomic analysis was performed with another 123 genomes of the same species, including ovis and equi biovars, downloaded from the NCBI. The genetic determinants for the virulence factors responsible for adherence and virulence factors specialized for iron uptake and exotoxin phospholipase D were revealed in every analyzed genome. Carbohydrate-Active Enzymes were compared, revealing the presence of genetic determinants encoding exo-α-sialidase (GH33) and the CP40 protein in most of the analyzed genomes. Thirty-three Czech strains of C. pseudotuberculosis were identified as the biovar ovis on the basis of comparative genome analysis. All the compared genomes of the biovar ovis strains were highly similar regardless of their country of origin or host, reflecting their clonal behavior. [ABSTRACT FROM AUTHOR]

This article compares different production, economic, and selected environmental aspects of agroforestry systems in a standard (alley cropping) and a newly proposed design with fast-growing trees grown in short-rotation coppice. Our models of agroforestry systems (AFSs) are as follows: (i) alley cropping AFS with cherry and walnut trees in single rows (tree strips) with 28 m-wide arable fields between them (crop strips), and (ii) coppiced tree belt AFS with poplars and willows and 25 m-wide arable fields between them (crop strips). To evaluate the production characteristics of trees, we used yield curves from experimental plantations in conditions of the Czech Republic from previous research projects. Cost data were collected from long-term experimental plantations and combined with current operation and energy prices. The article presents an economic methodology for assessing the competitiveness of biomass production in AFSs under the current identified market conditions. Our results show that AFSs with short-rotation coppice can have similar economic and production results as annual crops if grown on suitable sites and with appropriate quality of agronomy. In comparison, alley cropping AFSs with fruit trees would not be economically viable for farmers without a significant subsidy for establishment and maintenance in the first years after establishment. Concerning the latest economic and political developments, the product from SRC (energy woodchips) can be evaluated as strategic, increasing the producer's independence from purchased energy fuels. Background/Objectives: Molnupiravir (MOV), an oral antiviral COVID-19 treatment, was introduced in the Czech Republic in December 2021 for COVID-19 patients at a high risk of progression to severe disease requiring hospitalization. In this observational, retrospective study, we aimed to describe the characteristics and healthcare resource utilization in non-hospitalized, adult COVID-19 patients prescribed MOV in the Czech Republic between 1 January and 30 April 2022. Methods: A total of 621 patients were included and followed up with for 28 days. Results: The median age was 68.0 (20–99) years, 77.8% were overweight or obese, 14.1% smoked, and 85.7% were vaccinated. The overall cumulative incidence (95% CI) of all-cause hospitalization was 0.71 (0.37; 1.24) per 1000 person years or 1.9%, with similar rates across sexes, age groups, BMI category, multimorbidity category, polypharmacy category, and COVID-19 vaccination status. Among patients reported hospitalized, oxygen-based resources were not observed, and no deaths occurred. Conclusions: These data describe the characteristics and healthcare resource utilization in Czech MOV-treated patients whose clinical characteristics may put them at increased risk of severe disease. Featured Application: The used method was introduced by the Railway Administration's methodological regulations for the determination of points of primary and secondary system points and is currently used in connection with the determination of the spatial position of the center of the track. This article deals with the analysis of the accuracy of the geodetic real-time GNSS measurement procedure used in railway operating conditions in the Czech Republic. The purpose was to determine to what extent the operating conditions affect the accuracy of the measurement result and whether an accuracy of standard deviation σx,y = 5 mm in the horizontal plane could be achieved. The use of geodetic GNSS equipment with an IMU unit was also tested. The accuracy obtained in operational conditions is compared with the accuracy obtained on a calibration base using the same measurement procedure. The consistency between the accuracy of the primary system (satellite-based) and the secondary system (terrestrially measured by the traverse method) is also discussed. The analysis includes the issue of residual inhomogeneities of the uniform transformation key when converted to the Czech national coordinate system S-JTSK. It is shown that a homogeneous accuracy in coordinate standard deviation better than σx,y = 5 mm can be achieved. The results indicate that the accuracy under operational conditions is two–three times worse than the accuracy achieved by the same procedure under ideal conditions on a calibration base. This is due to the non-ideal observing conditions, i.e., horizon occlusion by overlays, surrounding vegetation and multipath effects. It has been shown that the effect of multipath can be reduced by repeating short observations 3–4 h apart. Older GNSS instruments using an IMU unit in combination with an electronic compass (eCompass) are at risk of a systematic bias of up to several tens of millimeters, which can be detected by rotating the antenna by 180°. The current uniform transformation key used in the Czech Republic for the conversion of GNSS coordinates into the national system has residual geometric inhomogeneities (p = 0.90 to 10 mm/km, sporadically up to 20 mm/km), which metrologically deteriorate the results of the calculation of the terrestrially measured secondary system inserted into the GNSS measured primary system. Achieving homogeneous accuracy in coordinate standard deviation in a horizontal plane better than σx,y = 5 mm has been demonstrated in non-ideal railway operating conditions with increased risk of multipath. The innovative aspect of the approach used is that it simplifies and thus increases the efficiency of the measurement with respect to the availability of GPS, GLONASS, Galileo and BeiDou satellites, as well as reducing the effect of multipath on the noise by repeating the measurement procedure. Central Europe is not a typical wildfire region; however, an increasingly warm and dry climate and model-based projections indicate that the number of forest fires are increasing. This study provides new insights into the drivers of forest fire occurrence in the Czech Republic, during the period 2006 to 2015, by focusing on climate, land cover, and human activity factors. The average annual number of forest fires during the study period was 728, with a median burned area of 0.01 ha. Forest fire incidence showed distinct spring (April) and summer (July to August) peaks, with median burned areas of 0.04 ha and 0.005 ha, respectively. Relationships between the predictors (climate data, forest-related data, socioeconomic data, and landscape-context data) and the number of forest fires in individual municipality districts were analyzed using Generalized Additive Models (GAM) on three time scales (annually, monthly, and during the summer season). The constructed GAMs explained 48.7 and 53.8% of forest fire variability when fire occurrence was analyzed on a monthly scale and during the summer season, respectively. On an annual scale, the models explained 71.4% of the observed forest fire variability. The number of forest fires was related to the number of residents and overnight tourists in the area. The effect of climate was manifested on monthly and summer season scales only, with warmer and drier conditions associated with higher forest fire frequency. A higher proportion of conifers and the length of the wildland–urban interface were also positively associated with forest fire occurrence. Forest fire occurrence was influenced by a combination of climatic, forest-related, and social activity factors. The effect of climate was most pronounced on a monthly scale, corresponding with the presence of two distinct seasonal peaks of forest fire occurrence. The significant effect of factors related to human activity suggests that measures to increase public awareness about fire risk and targeted activity regulation are essential in controlling the risk of fire occurrence in Central Europe. An increasing frequency of fire-conducive weather, forest structure transformations due to excessive tree mortality, and changing patterns of human activity on the landscape require permanent monitoring and assessment of possible shifts in forest fire risk. Members of the genus Armillaria are widespread forest pathogens against which effective protection has not yet been developed. Due to their longevity and the creation of large-scale cloning of Armillaria individuals, the use of mycoviruses as biocontrol agents (BCAs) against these pathogens could be an effective alternative. This work describes the detection and characterization of viruses in Armillaria spp. collected in the Czech Republic through the application of stranded total RNA sequencing. A total of five single-stranded RNA viruses were detected in Armillaria ostoyae and A. cepistipes, including viruses of the family Tymoviridae and four viruses belonging to the recently described "ambivirus" group with a circular ambisense genome arrangement. Both hammerhead (HHRz) and hairpin (HpRz) ribozymes were detected in all the ambiviricot sequences. Armillaria viruses were compared through phylogenetic analysis and confirmed their specific host by direct RT-PCR. A multi-residue UHPLC–MS/MS analytical method, previously developed for monitoring 52 pharmaceuticals in drinking water, was used to analyse these pharmaceuticals in wastewater originating from healthcare facilities in the Czech Republic. Furthermore, the methodology was expanded to include the evaluation of the effectiveness of drug removal in Czech wastewater treatment plants (WWTPs). Of the 18 wastewater samples analysed by the validated UHPLC-MS/MS, each sample contained at least one quantifiable analyte. This study reveals the prevalence of several different drugs; mean concentrations of 702 μg L−1 of iomeprol, 48.8 μg L−1 of iopromide, 29.9 μg L−1 of gabapentin, 42.0 μg L−1 of caffeine and 82.5 μg L−1 of paracetamol were present. An analysis of 20 samples from ten WWTPs revealed different removal efficiencies for different analytes. Paracetamol was present in the inflow samples of all ten WWTPs and its removal efficiency was 100%. Analytes such as caffeine, ketoprofen, naproxen or atenolol showed high removal efficiencies exceeding 80%. On the other hand, pharmaceuticals like furosemide, metoprolol, iomeprol, zolpidem and tramadol showed lower removal efficiencies. Four pharmaceuticals exhibited higher concentrations in WWTP effluents than in the influents, resulting in negative removal efficiencies: warfarin at −9.5%, indomethacin at −53%, trimethoprim at −54% and metronidazole at −110%. These comprehensive findings contribute valuable insights to the pharmaceutical landscape of wastewater from healthcare facilities and the varied removal efficiencies of Czech WWTPs, which together with the already published literature, gives a more complete picture of the burden on the aquatic environment. Our study wants to clarify the structure of spirituality by applying existing multidimensional theories of religiosity and spirituality to in-depth interviews conducted among a sample of students. The current research analyzes 15 qualitative semi-structured interviews conducted among a sample of students at the Adventist Theological Institute in the Czech Republic and was conducted in May 2022. Since religiosity is a multidimensional phenomenon and we wished to investigate development in each dimension, we based our analyses on Glock and Stark's model with four of their dimensions of religiosity: "belief", "practice", "experience", and "knowledge". Our study reflects on existing multidimensional religiosity/spirituality; six dimensions by Huber overlap with the religiosity/spirituality model we chose as the applied multidimensional model. As a result, based on the interviewees' statements, we distinguished these dimensions in the context of specific conditions in the respondents' individual personal experiences in the absence of personal group interaction. In our research, we ask the question: "What impact has the move of the entire formal and informal environment of an educational institution to an online environment had on the spirituality of each student?" An important finding is that the COVID-19 pandemic period brought about an exciting stimulus for spiritual support in theological education. To achieve the elimination of the negative impacts of transport on road safety, the European Union is taking various measures resulting from its commitment to improve road safety. The main objective of this paper is to assess the impact of social legislation on road transport safety using real research and to identify the factors influencing the violation of the legislation depending on the type of transport carried out in domestic or international road freight transport. Answers from the respondents are used in the segmentation of truck drivers based on input data describing the factors influencing the violation of traffic rules. Two-step cluster analysis is a suitable method for the segmentation of truck drivers based on input data. These data are obtained through a questionnaire from a total of 472 Czech and Slovak truck drivers. The results show that the optimal number of types of truck drivers with different characteristics is four. All of the clusters are described and compared. The majority of truck drivers do not violate social legislation for several reasons, such as traffic accidents (almost 67%), complications in meeting the loading or unloading deadline (less than 88%), poorly planned transport routes (more than 90%) and driving home (almost 80%). Currently, the recovery of resources from urban wastewater (WW) represents a priority. On this topic, the potential recovery of cellulose for its subsequent reuse in different sectors is gaining interest. In this work, a large-size conventional wastewater treatment plant (WWTP) was selected as a case study. A preliminary mechanical treatment was used, with the aim of separating, quantifying, and characterizing cellulose in WW. The results suggest that the per-capita production of dry primary cellulosic sludge (D-PCS) is equal to 1.46 ± 0.13 kgD-PCS PE−1 y−1, with an average calorific value of 21.04 MJ kg−1DM. Cellulosic fibres have an average length of >100 µm and a thickness of 2–5 µm. The D-PCS was subsequently treated via medium-temperature pyrolysis; a total of 29.5% of the initial D-PCS was converted into pyrolyzed primary cellulosic sludge (P-PCS) and only 26% into pyrolytic gas. More than 44.5% of the dried cellulose can be converted into pyrolytic oil. Moreover, three different scenarios of recovery have been considered, and the impact of cellulose separation in terms of COD fluxes entering the WWTP and potential energy recovery has been studied. The results suggested that, in this case study, the potential separation of the primary cellulosic sludge from the influent water flux would have no significant impact on COD load entering the biological treatments and biogas production in the anaerobic digestion of the secondary sludge. In this work, we develop a trade model that explains the pattern of trade between countries based on differences in total factor productivity (TFP) while also accounting for differences in relative factor endowments. The novelty stems from the introduction of production functions derived by combining the Ricardian and Heckscher–Ohlin–Samuelson (H-O-S) theories, with TFP differences serving as the basis of comparative advantage. To this end, a testable hypothesis is derived. For the empirical measurement of the TFP in each industry and country, a constant elasticity of substitution (CES)-type production function was employed, and the TFP was calculated as the Solow residual from the production function's fixed term. To offer a better understanding, the model was tested for the bilateral trade between Germany and Russia, and Germany and the Czech Republic. It was found that TFP differences can be used as a basis for explaining comparative advantages and, consequently, the bilateral pattern of trade between two countries. [ABSTRACT FROM AUTHOR]

Forestry reclamation practices have been very popular in the second half of the last century, and many spoil heaps have been converted into forests since. In our experiment, three forest reclamations of different ages (~90, ~50, and ~30 years) and three soil vegetation covers (I—maple and cherry, II—maple, and III—alder) from Sokolov, Czech Republic, were investigated. In each of the three stands, two soil profiles have been dug, and both disturbed and undisturbed soil samples were taken from all recognized horizons. Samples were tested for bulk and particle density, porosity, water retention capacity, pH (H2O, KCl), cation exchange capacity, oxidizable carbon content, organic matter quality, plant available nutrients, and risk elements. A comparison of these properties throughout the profile, as well as between the stands, was presented. A significant role of stand age in soil profile development and soil quality was observed, as well as the tendency of the anthropogenic mine Technosol to evolve into a forest Cambisol in this climate region and parent material. Influence of forest vegetation cover was observed to ameliorate soil properties by accumulating organic matter, thus reducing compaction and increasing CEC and nutrient availability. The Developmental Coordination Disorder Questionnaire (DCDQ) is a widely used parent questionnaire for screening motor coordination disorders in children aged 5–15 years. Despite increasing motor difficulties in children, a validated version is lacking in Central Europe. In addition, previous studies pointed out that several DCDQ items were shown to be problematic in different cultural environments. We found that the majority of these studies did not assess the item's content validity approach for keeping the semantic form and linguistic intelligibility of the original items. Therefore, this study aimed to translate the DCDQ, determine the content validity of items, and adapt the DCDQ for Czech children aged 6–10 years, where the identification of motor difficulties is crucial. Back-translation was employed, and face validity was consulted with linguistic experts and occupational therapists. A sample of 25 bilingual parents and practitioners evaluated the translated version, with content validity assessed using the Content Validity Ratio coefficient (CVR). Initial CVR scores ranged from 0.6 to 1.0. Lower scores were found for items 14 and 15, which were shown to be problematic in previous studies. The reason for the lower content validity in these items was due to double negation. Following linguistic modifications, the CVR values improved (range: 0.87–1.0), indicating content and semantic stability. Our findings underscore the importance of considering content validity and language specificity, including issues like double negation, during cross-cultural questionnaire validation to mitigate potential psychometric concerns in the future. Introduction: Dual diagnosis is used in addiction medicine to refer to the co-occurrence of an addiction-related disorder and another psychiatric disorder in the same individual. Adolescence is a key period for the development of both mental disorders and addictions. Objective: The aim of this study is to describe the relationships between psychiatric and addiction-related disorders in patients of the Outpatient Addiction Treatment Clinic for Children and Adolescents at the 1st Faculty of Medicine, Charles University in Prague in 2015–2022. Methods: Data were retrospectively analyzed from the hospital's medical system, which collects basic diagnostic data on patients. Descriptive statistics and cluster analysis were performed to identify relationships between psychiatric and addiction-related disorders. Results: Of the 450 patients, 153 patients (34%) met the criteria for dual diagnosis. The most common addiction-related disorders were mental and behavioural disorders due to the use of cannabinoids (35%) and internet gaming disorder (35%). The most common psychiatric diagnoses were behavioural and emotional disorders with usual onset in childhood and adolescence (64%), with a lower prevalence in girls than in boys. Conclusions: These findings may be important for the diagnosis and treatment of risky behaviours and addictions in children and adolescents. [ABSTRACT FROM AUTHOR]

Bovine respiratory syncytial virus (BRSV) is one of the most important respiratory pathogens of cattle. In this study, frequency of infection, analysis of variants, and the immune status of vaccinated and non-vaccinated cattle were studied. Blood (n = 162) and nasal/oropharyngeal (n = 277) swabs were collected from 62 cattle herds in Turkey. Lung samples (n = 37) were also taken from dead animals and abattoirs. Antibodies to BRSV were detected in 76 (46%) out of 162 sera. The antibody levels in the vaccinated and non-vaccinated groups were statistically significant. Among 277 nasal/oropharyngeal swabs and 37 lungs, ten nasal/oropharyngeal and four lung samples were positive for BRSV-RNA. BRSV-G gene sequences of 5 out of 14 RT-PCR positive samples showed that all viruses clustered as Group-III in phylogenetic analysis with 88–100% homology. Similarity with previous Turkish BRSVs was 89–98%, and that with BRSVs detected in the USA and Czechia was 89.47–93.12%. BRSV continues to circulate in Turkish cattle, and vaccination seems beneficial in preventing BRSV. The diversity of the BRSVs found in this study needs be considered in vaccination strategies. This review aims to explore the role of professional diagnostic rapid testing of acute respiratory infections (ARIs), especially COVID-19 and influenza, ensuring proper disease management and treatment in Europe, and particularly in Czech Republic, Poland, and Romania. The paper was constructed based on a review of scientific evidence and national and international policies and recommendations, as well as a process of validation by four experts. The development of new testing technologies, treatment options, and increased awareness of the negative multidimensional impact of ARI profiles transformed differential diagnosis into a tangible and desirable reality. This review covers the following topics: (1) the multidimensional impact of ARIs, (2) ARI rapid diagnostic testing platforms and their value, (3) the policy landscape, (4) challenges and barriers to implementation, and (5) a set of recommendations illustrating a path forward. The findings indicate that rapid diagnostic testing, including at the point of care (POC), can have a positive impact on case management, antimicrobial and antibiotic stewardship, epidemiological surveillance, and decision making. Integrating this strategy will require the commitment of governments and the international and academic communities, especially as we identified room for improvement in the access and expansion of POC rapid testing in the focus countries and the inclusion of rapid testing in relevant policies. The Czech Republic is one of the countries along the Belt and Road Initiative, and classifying land cover in the Czech Republic helps to understand the distribution of its forest resources, laying the foundation for forestry cooperation between China and the Czech Republic. This study aims to develop a practical approach for land cover classification in the Czech Republic, with the goal of efficiently acquiring spatial distribution information regarding its forest resources. This approach is based on multi-level feature extraction and selection, integrated with advanced machine learning or deep learning models. To accomplish this goal, the study concentrated on two typical experimental regions in the Czech Republic and conducted a series of classification experiments, using Sentinel-2 and DEM data in 2018 as the main data sources. Initially, this study extracted various features, including spectral, vegetation, and terrain features, from the study area, then assessed and selected key features based on their importance. Additionally, this study also explored multi-level spatial contextual features to improve classification performance. The extracted features include texture and morphological features, as well as deep semantic information learned by utilizing a deep learning model, 3D CNN. Finally, an AdaBoost ensemble learning model with the random forest as the base classifier is designed to produce land cover classification maps, thus obtaining the spatial distribution of forest resources. The experimental results demonstrate that feature optimization significantly enhances the extraction of high-quality features of surface objects, thereby improving classification performance. Specifically, morphological and texture features can effectively enhance the discriminability between different features of surface objects, thereby improving classification accuracy. Utilizing deep learning networks enables more efficient extraction of deep feature information, further enhancing classification accuracy. Moreover, employing an ensemble learning model effectively boosts the accuracy of the original classification results from different individual classifiers. Ultimately, the classification accuracy of the two experimental areas reaches 92.84% and 93.83%, respectively. The user accuracies for forests are 92.24% and 93.14%, while the producer accuracies are 97.71% and 97.02%. This study applies the proposed approach for nationwide classification in the Czech Republic, resulting in an overall classification accuracy of 90.98%, with forest user accuracy at 91.97% and producer accuracy at 96.2%. The results in this study demonstrate the feasibility of combining feature optimization with the 3D Convolutional Neural Network (3DCNN) model for land cover classification. The energy sector faces a pressing need for significant transformation to curb CO2 emissions. For instance, Czechia and Germany have taken steps to phase out fossil thermal power plants by 2038, opting instead for a greater reliance on variable renewable energy sources like wind and solar power. Nonetheless, thermal power plants will still have roles, too. While the conventional multistage axial turbine design has been predominant in large-scale power plants for the past century, it is unsuitable for small-scale decentralized projects due to complexity and cost. To address this, the study investigates less common turbine types, which were discarded as they demonstrated lower efficiency. One design is the Elektra turbine, characterized by its velocity compounded radial re-entry configuration. The Elektra turbine combines the advantages of volumetric expanders (the low rotational speed requirement) with the advantages of a turbine (no rubbing seals, no lubrication in the working fluid, wear is almost completely avoided). Thus, the research goal of the authors is the implementation of a 10 kW-class ORC turbine driving a cost-effective off-the-shelf 3000 rpm generator. The paper introduces the concept of the Elektra turbine in comparison to other turbines and proposes this approach for an ORC working fluid. In the second part, the 1D design and 3D–CFD optimization of the 7 kW Elektra turbine working with Hexamethyldisiloxane (MM) is performed. Finally, CFD efficiency characteristics of various versions of the Elektra are presented and critically discussed regarding the originally defined design approach. This study focused on the viruses of the Tymoviridae family that infect grapevines in the Czech Republic. Complete sequences of GFkV (grapevine fleck virus) and GRGV (grapevine red globe virus) from the genus Maculavirus and GRVFV (grapevine rupestris vein feathering virus) and GSyV-1 (grapevine Syrah virus 1) from the genus Marafivirus were obtained using high-throughput sequencing of small RNAs and total RNAs. Mixed infections with these viruses were observed, as well as several variants of these viruses in the same plant. Phylogenetic analysis showed the position of the newly obtained virus isolates within the Tymoviridae family. Recombinant analysis provided evidence of single and multiple intraspecific recombinations in GRGV, GSyV-1, and GRVFV. Additionally, GAMaV, a grapevine virus from the genus Marafivirus, was reported for the first time in the Czech Republic. [ABSTRACT FROM AUTHOR]

Cercariae of Plagiorchis spp. are frequently reported in European freshwater snails, but their true diversity is difficult to estimate due to subtle differences in morphology. We molecularly characterized 67 isolates of Plagiorchis cercariae collected from four lymnaeid snail hosts, Ampullaceana balthica, Ampullaceana lagotis, Radix auricularia and Lymnaea stagnalis in freshwater ecosystems in the Czech Republic and Poland. Based on mitochondrial cox1 and nuclear 28S sequences, ten species or species-level lineages were identified, including the first molecular evidence of P. vespertilionis from snail hosts and two species-level lineages reported for the first time. Previously undescribed species and species-level lineages are characterized morphometrically. We confirm the overlapping spatial distribution of Plagiorchis spp. in their snail hosts from Central Europe with those from Western and sub-Arctic Europe. Our results increase the known diversity of Plagiorchis spp. in Europe to 25 species/lineages in snails, but further research is needed to establish links between life cycle stages and to assess the host specificity of these parasites. Gallibacterium anatis, recognized as a resident and opportunistic pathogen primarily in poultry, underwent investigation in unwell domestic mammals and birds. The study encompassed the mapping and comparison of G. anatis isolates, evaluation of their genetic diversity, and determination of their susceptibility to antimicrobials. A total of 11,908 clinical samples were analyzed using cultivation methods and MALDI-TOF. Whole-genome sequencing was performed on seven calf isolates and six hen isolates. Among mammals, G. anatis was exclusively detected in 22 young dairy calves, while among domestic birds, it was found in 35 individuals belonging to four species. Pathological observations in calves were predominantly localized in the digestive tract, whereas in birds, multi-organ infections and respiratory system infections were most prevalent. Distinct groups of genes were identified solely in calf isolates, and conversely, those unique to hen isolates were also recognized. Novel alleles in the multilocus sequence typing scheme genes and previously unidentified sequence types were observed in both calf and hen isolates. Antimicrobial susceptibility exhibited variation between bird and calf isolates. Notably, G. anatis isolates from calves exhibited disparities in genotype and phenotype compared to those from hens. Despite these distinctions, G. anatis isolates demonstrated the capability to induce septicemia in both species. [ABSTRACT FROM AUTHOR]

Pectinolytic bacteria of the family Enterobacteriaceae, specifically Dickeya solani, are known to cause potato blackleg. This study aimed to evaluate the effectiveness of a mixture of two bacteriophages from the genus Limestonevirus in controlling Dickeya solani in both greenhouse and field trials. The potential of bacteriophages for ecological potato control was also assessed. The phages φDs3CZ and φDs20CZ were isolated from soil in the Czech Republic between 2019 and 2021. They were applied preventively and curatively as a solution on artificially wounded and inoculated seed tubers immediately prior to planting. The phage-treated variant showed a highly significant reduction in the extent of D. solani infection compared to the untreated control in both the greenhouse and field trial. The effect of the phages depended on the concentration of the solution, the rate of tuber injury, and the sequence of application. When applied preventively, the phages caused a significantly higher reduction in the rate of blackleg symptoms (86.7% and/or 87.1%) compared to the curative application (54.6 and/or 36.6%). A reliable supply of quality drinking water is a fundamental prerequisite for a healthy society and its economic development. Public ownership of water infrastructure is prevalent in most European countries. In the Czech Republic, however, water infrastructure is highly fragmented, which entails multiple risks. The fragmentation of the sector leads to a low economic efficiency of operations, the unstable quality of service provision, and significant price differences. The aim of the paper is to use the IFE matrix to analyze the strengths and weaknesses of different ways of operating water supply systems in the Czech Republic. Furthermore, through the FMEA method (failure mode and effects analysis), this paper tries to identify the risks and threats to drinking water supplies for selected operators (representing the most frequently used operating models) and, subsequently, it proposes measures to mitigate the identified risks. The topic was addressed in the form of a case study of selected water system operators in the Czech Republic, and the findings indicate the compartmentalized model to be the most appropriate operating model. In 2022/2023, Europe experienced its third consecutive season of high-pathogenicity avian influenza. During this period, the Czech Republic was again severely affected. For the first time, the number of culled birds approached one million, which was three times higher than in previous seasons. In parallel to the outbreaks in poultry, mass die-offs of gulls were also observed. In the present study, we performed whole-genome sequencing and phylogenetic analysis of 137 H5N1 strains collected in the Czech Republic in 2022/2023 (94.6% of all outbreaks or locations). The analysis revealed four distinct genotypes: AB, CH, BB and AF. Phylogenetic analysis suggested that the AF genotype persisted from the previous H5N1 season without reassortment. In addition, the genotype BB, which was detected mainly in gulls, showed a noticeable strain diversity at the local level. This virus was also responsible for a single outbreak in commercially bred turkeys. Finally, an interesting spatio-temporal cluster with three co-circulating H5N1 genotypes, AB, CH and AF, was identified with no evidence of intrasubtype reassortment. Highly sensitive molecular surveillance and the timely sharing of genomic sequences and associated metadata could greatly assist in tracking the spread and detecting molecular changes associated with the increased virulence of this potentially zoonotic pathogen. This contribution deals with the assessment of air pollution caused by atmospheric aerosol particulate matter fraction PM10 and benzo[a]pyrene (BaP) in the urban agglomeration of Ostrava, located in the Czech Republic in Central Europe. The motivation for this research was the need to identify the sources of air pollution in the area, particularly in locations where the contribution of different sources to concentrations of pollutants of concern has not been elucidated yet. In this study, source apportionment in the vicinity of the industrial hot spot was performed by statistically evaluating measured pollutant concentrations as a function of meteorological variables and using the Positive Matrix Factorization (PMF) receptor model. A significant methodological innovation and improvement over previous assessments was the higher monitoring resolution of benzo[a]pyrene, with samples collected at three-hour intervals instead of the standard 24 h collection period. The key findings indicate that in the cold part of the year, secondary particles—specifically sulfates and ammonium nitrates—were responsible for the most significant portion of PM10 air pollution throughout the area of interest. The contribution of these particles ranged from one-third to two-fifths of the total concentration, except at the industrial site of Ostrava–Radvanice (TORE), where they accounted for approximately one-fifth of the measured pollution concentration level. Emissions from individual household heating were identified as the main source of this type of pollution. With regards to benzo[a]pyrene air pollution, this study found that in the whole area of interest, except for the Ostrava–Radvanice site, it mainly originated from individual heating with coal (90%). In contrast, at the Ostrava–Radvanice site, two-thirds of the benzo[a]pyrene pollution came from the premises of Liberty Ostrava a.s., primarily from coke production, and less than one-third came from local domestic heating. This study also determined the spatial extent of the occurrence of extremely high benzo[a]pyrene concentrations (above 5 ng/m3), which are estimated to affect nearly 10,000 inhabitants. The results confirm that the data from the TORE station are only representative of its immediate surroundings and are not applicable to the assessment of air quality and causes of air pollution in the whole city of Ostrava or the urban area of Ostrava–Radvanice as a whole. After years of research, these findings provide the Ministry of the Environment of the Czech Republic and the government with an accurate basis for implementing measures to address the identified pollution sources in the area of interest. The success of this study was made possible by the adoption of a more detailed sampling approach, which involved a resolution of 3 h instead of 24 h. SARS-CoV-2 respiratory infection is associated with significant morbidity and mortality in hospitalized patients. We aimed to assess the risk factors for hospital mortality in non-vaccinated patients during the 2021 spring wave in the Czech Republic. A total of 991 patients hospitalized between January 2021 and March 2021 with a PCR-confirmed SARS-CoV-2 acute respiratory infection in two university hospitals and five rural hospitals were included in this analysis. After excluding patients with unknown outcomes, 790 patients entered the final analyses. Out of 790 patients included in the analysis, 282/790 (35.7%) patients died in the hospital; 162/790 (20.5) were male and 120/790 (15.2%) were female. There were 141/790 (18%) patients with mild, 461/790 (58.3%) with moderate, and 187/790 (23.7%) with severe courses of the disease based mainly on the oxygenation status. The best-performing multivariate regression model contains only two predictors—age and the patient's state; both predictors were rendered significant (p < 0.0001). Both age and disease state are very significant predictors of hospital mortality. An increase in age by 10 years raises the risk of hospital mortality by a factor of 2.5, and a unit increase in the oxygenation status raises the risk of hospital mortality by a factor of 20. The aim of this contribution is to present the R-WIM (Rainwater Information Management) tool, which was created based on an extensive database of territory parameters, weather, surface runoff, etc., and in accordance with the requirements of municipalities. This tool was created especially for the purpose of identifying places where it is appropriate to implement elements of blue-green infrastructure. This tool was created on the basis of the smart urbido s.r.o. software 2.0, which allows working with a wide range of graphic and non-graphic information so that it is possible to link them together functionally and computationally, and to model the necessary spatial phenomena within the environment of selected cities of the Moravian-Silesian Region of the Czech Republic.
