This comprehensive review explores the chemical composition, bioactivities, processing techniques, and economic potential of BC milk in China. The distinctive chemical composition of BC milk, including protein, lipid, carbohydrate, vitamin, and mineral content, is discussed, emphasizing its differences from other mammalian milk. The review highlights the various bioactivities of BC milk, such as anti-inflammatory, antidiabetic, lipid-lowering, and anticancer properties, as well as its modulatory effects on intestinal microbiota. The technological properties of BC milk, focusing on its heat stability, coagulation behavior, and potential for product development, are examined. The review also addresses current processing techniques and their impact on milk quality. Finally, the economic potential and future perspectives of BC milk in China are evaluated. Simple Summary: Fourier Transform Infrared Spectroscopy (FT-MIRS) is widely used in milk quality detection, dairy herd improvement (DHI), and other fields. It is an economical, fast, accurate, and nondestructive batch tool for determining production performance phenotypes of dairy cows. Milk is one of the most important ways to provide the human body with the fatty acids it needs. There are a huge number of dairy cows in China. Therefore, it is possible to control fatty acid production in the milk source through targeted husbandry and breeding on large pastures to improve the quality of milk production. However, this work has not yet officially begun in China. In summary, our work uses FT-MIRS for the first time to study the phenotypic properties of milk fatty acid content and the genetic mechanism of its formation and to estimate genetic parameters. At the same time, SNPs significantly related to fatty acid content were discovered and the genes or adjacent genes had critical regulatory effects on milk fat synthesis, milk protein synthesis, adipocyte differentiation, mammary gland development, milk synthesis, and growth and development in dairy cows, thus providing a new perspective for cow genetic selection in China. Fourier Transform Mid-Infrared Spectroscopy (FT-MIRS) can be used for quantitative detection of milk components. Here, milk samples of 458 Chinese Holstein cows from 11 provinces in China were collected and we established a total of 22 quantitative prediction models in milk fatty acids by FT-MIRS. The coefficient of determination of the validation set ranged from 0.59 (C18:0) to 0.76 (C4:0). The models were adopted to predict the milk fatty acids from 2138 cows and a new high-throughput computing software HiBLUP was employed to construct a multi-trait model to estimate and analyze genetic parameters in dairy cows. Finally, genome-wide association analysis was performed and seven novel SNPs significantly associated with fatty acid content were selected, investigated, and verified with the FarmCPU method, which stands for "Fixed and random model Circulating Probability Unification". The findings of this study lay a foundation and offer technical support for the study of fatty acid trait breeding and the screening and grouping of characteristic dairy cows in China with rich, high-quality fatty acids. Simple Summary: This study reports the results of two studies aiming to determine, under commercial farm conditions, the effects of electronic sow feeders on the production and economic yields of lactating sows. The results indicate remarkable technical and economic outputs compared with traditional feeders due to the weaning of heavier piglets with a lower amount of feed per kg of weaned piglet. Adequate nutritional management in maternities is one of the most challenging aspects of swine production. This study reports the results of two studies aiming to determine, under commercial farm conditions, the effects of precision feeding (electronic sow feeders, ESFs) on the production and economic yields of lactating sows and possible nutritional and metabolic differences when compared to a control group fed with traditional feeders. The first trial showed that sows fed with ESFs weaned heavier piglets than sows fed with traditional feeders. Feed intake during the lactation period was similar in the sows of both groups; consequently, the amount of feed per kg of weaned piglet was lower in the sows fed with ESFs, which is a remarkable economic output. The second trial confirmed these findings and showed that, despite similar feed intakes, the sows fed with ESFs had lower bodyweight losses during the lactation period, but there were no major differences in milk composition or metabolic traits of sows and piglets. Simple Summary: This study evaluated the effects of heat stress on the productivity of dairy cows. During the experimental period, THI values ranged from 46.9 to 81.0, with the most severe heat stress occurring in August. A decrease in productivity was observed when the THI exceeded 72. It was found that milk production took about one month to recover after the high-temperature period ended. The THI ratio, calculated based on the daily THI distribution, showed a strong negative correlation with rumen temperature (− 0.744; p < 0.001). During periods of significant productivity decreases due to high-temperature stress, rumen temperatures were higher than 39.15 °C. Based on the rumen temperature and daily THI distribution, it is suggested that these findings could serve as foundational data for developing technologies to manage high-temperature environments for dairy cows. This study was conducted to evaluate the effects of high-temperature stress on dairy cow productivity and the correlation between rumen sensors. The data were collected on the temperature, humidity, milk productivity, milk components, blood components, and rumen sensor data from 125 dairy cows during the experimental period (1 May 2020 to 30 October 2020). High-temperature stress of dairy cows was evaluated based on the temperature–humidity index (THI). The correlations between the high-temperature stress, productivity, and sensor data were analyzed using SAS and R programs. The THI ranged from 46.9 to 81.0 during the experimental period, and a significant decrease was observed in the milk production of dairy cows during August (p < 0.05). Milk production was evaluated to decrease by 1.8% because of high-temperature stress during the experimental period. There was a significantly high negative correlation between the THI ratio of day and rumen temperature (r = 0.744; p < 0.001). The other rumen sensor data did not show a significant correlation with the productivity of dairy cows. Simple Summary: Mastitis in dairy cows is a major problem for livestock production. In this study, we investigated the effects of exogenous melatonin on the milk somatic cell count and yield and the serum anti-inflammatory factors, antioxidant indices, and melatonin in dairy cows. Exogenous melatonin was shown to increase the milk component content, reduce the somatic cell count, and improve the antioxidant capacity and immune responses. Mastitis is an important factor affecting the health of cows that leads to elevated somatic cell counts in milk, which can seriously affect milk quality and result in huge economic losses for the livestock industry. Therefore, the aim of this trial was to investigate the effect of melatonin on performance and mastitis in dairy cows. Forty-eight Holstein cows with a similar body weight (470 ± 10 kg), parity (2.75 ± 1.23), number of lactation days (143 ± 43 days), BCS (3.0–3.5), milk yield (36.80 ± 4.18 kg), and somatic cell count (300,000–500,000 cells/mL) were selected and randomly divided into four groups: control (CON group), trial Ⅰ (T80 group), trial Ⅱ (T120 group), and trial Ⅲ (T160 group). Twelve cows in trial groups I, II, and III were pre-dispensed 80, 120, and 160 mg of melatonin in edible glutinous rice capsules along with the basal ration, respectively, while the control group was fed an empty glutinous rice capsule along with the ration. The trial period was 37 days, which included a 7-day adaptive phase followed by a 30-day experimental period. At the end of the trial period, feeding was ended and the cows were observed for 7 days. Milk samples were collected on days 0, 7, 14, 21, 28, and 37 to determine the somatic cell number and milk composition. Blood samples were collected on days 0, 15, 30, and 37 of the trial to determine the serum biochemical indicators, antioxidant and immune indicators, and the amount of melatonin in the blood. The results showed that the somatic cell counts of lactating cows in the CON group were lower than those in the T120 group on days 14 (p < 0.05) and 28 (p < 0.01) at 1 week after melatonin cessation. The milk protein percentage and milk fat percentage of cows in the T120 group were higher than those in the CON group (p < 0.01). The total protein and globulin content in the T120 group were higher than those in the CON group (p < 0.01). In terms of antioxidant capacity and immunity, the cows 1 week after melatonin cessation showed higher superoxide dismutase activity and interleukin-10 contents (p < 0.01) compared with the CON group and lower malondialdehyde and tumor necrosis factor-alpha contents (p < 0.01) compared with the T120 group. The melatonin content in the T120 group was increased relative to that in the other groups. In conclusion, exogenous melatonin can increase the content of milk components, reduce the somatic cell count, and improve the antioxidant capacity and immune responses to a certain extent. Simple Summary: The increasing use of automated milking systems (AMS) in the dairy industry has led to the necessity for changes in management practices implemented by producers. This study investigated the productive and metabolic impact of three versus six milkings per day (MPD) during early lactation on multiparous Holstein cows managed in an AMS. Mature cows (third and greater parity) with six MPD had greater milk fat production during the experimental phase (4 to 29 days in milk; DIM) and milk production during the carryover phase (30 to 90 DIM) than mature three MPD cows. The composition of milk fatty acids differed between MPD groups with cows, with three MPD having greater short-, medium-, odd- and branched-chain fatty acids during the experimental phase compared to cows with six MPD. Based on greater blood fatty acid and β-hydroxybutyrate concentrations, the mature six MPD cows may have been in a greater postpartum nutrient deficit and at greater risk of metabolic illness. Our findings suggest a need to strategically manage the MPD or nutrition of mature, early lactation dairy cows in an AMS. Automated milking systems (AMS) are increasingly adopted for dairy cow production, promoting individualized cow management dependent on factors like lactation stage, age, and productivity. The study objective was to investigate the effects of early lactation milking frequency on cows milked via AMS. Multiparous Holstein cows blocked by parity and due date were randomly assigned to treatments (n = 8 per treatment): three (3X) or six (6X) milkings per day (MPD). The experimental phase (EXP) was defined as 4 to 29 days in milk (DIM). The AMS settings were programed so 3X cows were limited to three MPD while 6X cows were allowed six MPD. Afterwards was the carry over phase (CO) ranging from 30 to 90 DIM; all cows were allowed up to six MPD. Measurements by the AMS included bodyweight, milk yield (MY), and pellet intake. Weekly composite milk samples were analyzed for macronutrient composition and fatty acid (FA) profile. Coccygeal blood was sampled at 3, 8 ± 1, and 13 ± 1 DIM; concentrations of blood plasma analytes were quantified. Greater MPD was achieved for 6X cows versus 3X cows during EXP, but similar during the CO. Daily MY was non-separable during the EXP while 6X cows in their third or greater lactation group (3 + LG) had greater MY than 3X cows of the same LG during the CO. Milk fat content and 4% fat-corrected MY were both greater for 6X, 3 + LG cows during the EXP compared to 3X, 3 + LG cows. Milk FA methyl esters (FAME) proportions were different between MPD groups, with 6X, 3 + LG cows having the lowest short, even-chain FA from de novo or post-absorptive origin. Differences in analytes indicated that 6X, 3 + LG cows experienced metabolic stress and incorporated greater FA from adipose tissue. Graviera Naxos, a renowned cheese with Protected Designation of Origin status, is crafted from a blend of cow, goat, and sheep milk. This study focused on assessing the Sr isotopic and multi-elemental composition of both the processed cheese and its ingredients, as well as the environmental context of Naxos Island, including samples of milk, water, soil, and feed. The objective was to delineate the geochemical signature of Graviera Naxos cheese and to explore the utility of Sr isotopes as indicators of geographic origin. The 87Sr/86Sr values for Graviera Naxos samples ranged from 0.70891 to 0.70952, indicating a relatively narrow range. However, the Sr isotopic signature of milk, heavily influenced by the feed, which originates from geologically distinct areas, does not always accurately reflect the local breeding environment. Multi-elemental analysis revealed variations in milk composition based on type and season; yet, no notable differences were found between raw and pasteurized milk. A mixing model evaluating the contributions of milk, sea salt, and rennet to the cheese's Sr isotopic signature suggested a significant average contribution of 73.1% from sea salt. Among other bioactive molecules, milk contains high amounts of sialylated milk oligosaccharides (MOs) that influence numerous processes in the offspring. For instance, sialylated MOs inhibit the invasion of pathogens and positively influence the gut microbiome to support the optimal development of the offspring. For these reasons, sialylated MOs are also used in infant formula as well as food supplements and are potential therapeutic substances for humans and animals. Because of the high interest in sialylated bovine MOs (bMOs), we used several analytical approaches, such as gas and liquid chromatography in combination with mass spectrometry, to investigate in detail the profile of sialylated bMOs in the milk of Holstein Friesian cows during early lactation. Most of the 40 MOs identified in this study were sialylated, and a rapid decrease in all detected sialylated bMOs took place during the first day of lactation. Remarkably, we observed a high variance within the sialylation level during the first two days after calving. Therefore, our results suggest that the content of sialylated MOs might be an additional quality marker for the bioactivity of colostrum and transitional milk to ensure its optimized application for the production of milk replacer and food supplements. Simple Summary: Pre-weaning piglet diarrhea is one of the major concerns in the swine industry. Recently, various probiotics have been applied to improve animal gut health and performance. This study investigated the impact of C. butyricum probiotic feed additive on sow and piglet performance, together with alterations in lipidomic and metabolomic profiles of sow milk. Results showed that sows given the probiotics had lower backfat loss and their piglets experienced less diarrhea incidence, although there were no other significant benefits for piglet growth. Changes in certain fatty acids and metabolites present in sow colostrum and milk, which could impact their nutritional profiles and the health of the piglets, were significantly observed. This study provided new insights regarding the impacts of probiotics application that could potentially lead to better outcomes in swine farms. The present study aimed to investigate the impact of Clostridium butyricum probiotic feed additive on sow and piglet performances, together with alterations in the lipidomic and metabolomic profiles of sow milk. Sixty-four Landrace × Yorkshire crossbred sows and 794 piglets were included. Sows were divided into two groups; i.e., (i) conventional gestation diet (control; n = 35) and (ii) conventional diet added with 10 g/sow/day of probiotic C. butyricum spores (treatment; n = 29) from one week before the estimated farrowing day until weaning (29.6 ± 4.8 days). The sow and piglet performances and incidence of piglet diarrhea were recorded. Changes in gross chemical composition, fatty acid and non-volatile polar metabolite profiles of sow colostrum, transient milk and mature milk were evaluated. The results showed that relative backfat loss in the treatment group (−2.3%) was significantly lower than in control group (11.6%), especially in primiparous sows (p = 0.019). The application of C. butyricum probiotics in sows significantly reduced the incidence of diarrhea in piglets (p < 0.001) but no other effect on piglet performance was found. Lipidomic and metabolomic analyses revealed variations in sow colostrum and milk biomolecular profiles, with indicative compounds significantly altered by feeding with the C. butyricum probiotics. In conclusion, the use of C. butyricum probiotics in sows may improve sow body condition and reduce diarrhea incidence in piglets, with underlying changes in milk composition that warrant further investigation. Simple Summary: Summer heat stress adversely affects the productivity and economic viability of lactating Holstein cows. We explored the effects of dietary supplementation with L-tryptophan or L-ascorbic acid on milk production characteristics and plasma metabolites in lactating Holstein cows during the summer. The results showed that the supplementation with L-tryptophan or L-ascorbic acid reduced heat stress in lactating Holstein cows during the summer. In particular, L-tryptophan supplementation increased the concentration of melatonin in milk. However, there were no significant differences in milk yield or composition between the treatments. This study investigated the effects of rumen-protected L-tryptophan or L-ascorbic acid supplementation on the productivity of lactating Holstein cows during a high-temperature period. Thirty cows were assigned to three dietary groups: control (CON), treatment 1 (TRT 1; rumen-protected L-tryptophan, 20 g/cow/d), and treatment 2 (TRT 2; rumen-protected L-ascorbic acid, 20 g/cow/d). As the high-temperature period progressed, the decrease in milk yield and dry matter intake (DMI) in the TRT 1 and TRT 2 groups was lower than that in the CON group. The total protein level in the plasma of the TRT 1 group was higher than that in the CON group (p < 0.05). Milk melatonin concentration was higher in the TRT 1 group than in the CON and TRT 2 groups (p < 0.05). Thus, the present results indicate that rumen-protected L-tryptophan or L-ascorbic acid has positive effects in preventing declines in DMI and milk yield by reducing heat stress in Holstein cows. Background: Breastfeeding is a process for not only nourishing infants but also for building a unique emotional bond between mother and child. Therefore, the ideal contraception during lactation should not affect lactation (milk composition, milk volume) and offspring development. Objectives: This study aims to analyze the literature on the safety of progestogen hormonal contraceptive methods during lactation. Methods: We conducted a thorough search across various databases, including the National Library of Medicine (PubMed), and the Cochrane Database, Drugs and Lactation Database (LactMed). Our search utilized specific phrases such as: "lactation" and "breastfeeding" and "oral contraception" with "drospirenone" or "desogestrel", with "subcutaneous etonogestrel implant" or "etonogestrel implant", with "levonorgestrel-releasing intrauterine system", and "emergency contraception", with "levonorgestrel" or "ulipristal acetate". Conclusions: Based on published scientific reports, progestogen hormonal contraceptives can be considered a relatively safe solution for women desiring to continue feeding their infant with their milk while using hormonal contraception. It is important to seek guidance on selecting the best contraception method based on the latest medical knowledge, tailored to the individual needs and clinical circumstances of each woman and place of residence. Simple Summary: Mules cannot reproduce naturally; however; they can host embryos, gestate normally, and act as surrogate dams, producing milk for the foal. There are only a few studies on the composition of mule milk. We evaluated the chemical quality of the milk obtained from a mule dam that had foaled after receiving a mule embryo transfer. The quality of the mule milk was similar to that of horse and donkey milk. Monounsaturated and polyunsaturated fatty acids made up half of the total fatty acid content. The aim of this investigation was to increase our knowledge of the qualitative composition of mule's milk and its variations throughout the whole lactation period—namely, from 6 h after foaling to 180 days in milk (DIM). Milk was obtained from a mule dam that had foaled after receiving a mule embryo transfer. For each sample, the gross, mineral, and fatty acid composition was evaluated. The average quality of the mule milk was as follows: protein 1.97 g 100 mL−1, fat 0.90 g 100 mL−1, and ash 0.39 g 100 mL−1. Saturated fatty acids made up, on average, 50.00 g 100 g−1 of fat. Monounsaturated and polyunsaturated fatty acids made up half of the total fatty acid content (31.80 g 100 g−1 and 18.2 g 100 g−1 of fat, respectively). Simple Summary: Humanity faced the hottest year in observational history in 2023. If the trend of rising temperatures on Earth continues, irreversible changes in the environment will occur with negative consequences for the lives of us all. In order to limit the further increase in temperatures on Earth, it is necessary to reduce the emissions of greenhouse gases (GHGs) in various sectors. Since ruminant farming is a significant contributor to anthropogenic GHG emissions due to methane emissions, we attempted to reduce methane emissions in dairy cows by using brown seaweed (Ascophyllum nodosum) in cow rations. Indeed, we achieved a reduction in methane emissions, without disturbing the productivity of cows. We also found a more favorable metabolic status and milk composition in supplemented cows. Additional studies are needed to define precisely how brown seaweeds can be included into strategies to reduce methane emissions from the dairy industry. The dairy industry contributes significantly to anthropogenic methane emissions, which have an impact on global warming. This study aimed to investigate the effects of a dietary inclusion of brown seaweed Ascophyllum nodosum on enteric methane emissions (EMEs), hematological and blood biochemical profiles, and milk composition in dairy cows. Eighteen Holstein cows were divided into three groups: CON (non-supplemented cows), BS50 (50 mL of 10% A. nodosum), and BS100 (100 mL of 10% A. nodosum). In each cow, measurements of EME, dry matter intake (DMI), and milk yield (MY), as well as blood and milk sampling with respective analyzes, were performed before supplementation (P1), after 15 (P2) days, and after 30 (P3) days of supplementation. A. nodosum reduced (p < 0.05) methane production, methane yield, and methane intensity in both BS50 and BS100, and raised DMI (p < 0.05) only in BS50. Total bilirubin (p < 0.05) was higher in BS50 compared to CON cows in P2, and triacylglycerols were lower (p < 0.05) in BS50 than in CON cows in P3. Higher milk fat content was found in BS50 than in CON cows in P3. C16:0 proportions were higher (p < 0.05) in BS50 and BS100 than in CON cows, while C18:3n-3 was higher (p < 0.05) in BS100 than in BS50 and CON cows in P3. In this study, our primary aim was to explore the genomic landscape of Barka cattle, a breed recognized for high milk production in a semi-arid environment, by focusing on genes with known roles in milk production traits. We employed genome-wide analysis and three selective sweep detection methods (ZFST, θπ ratio, and ZHp) to identify candidate genes associated with milk production and composition traits. Notably, ACAA1, P4HTM, and SLC4A4 were consistently identified by all methods. Functional annotation highlighted their roles in crucial biological processes such as fatty acid metabolism, mammary gland development, and milk protein synthesis. These findings contribute to understanding the genetic basis of milk production in Barka cattle, presenting opportunities for enhancing dairy cattle production in tropical climates. "Pecorino" is a typical semi-hard cheese obtained with raw or heat-treated sheep milk using procedures to valorize the raw material's chemical and microbiological properties. In the present study, using a high-throughput method of 16S rRNA gene sequencing, we assessed the evolution of the microbiome composition from milk to Pecorino-like cheese in artisanal processes using milk from Comisana and Lacaune sheep breeds. The comparative analysis of the bacterial community composition revealed significant differences in the presence and abundance of specific taxa in the milk microbiomes of the Comisana and Lacaune breeds. Next-Generation Sequencing (NGS) analysis also revealed differences in the curd microbiomes related to dairy farming practices, which have a relevant effect on the final structure of the Pecorino cheese microbiome. Goat milk is considered the optimal substitute for human milk and is characterized by variations in the lipid composition of its fat globules across lactation phases. Therefore, the objective of this study was to thoroughly analyze the differences between goat milk during different lactations and human milk, aiming to offer scientific guidance for the production of functional dairy products. Compared with transitional and mature milk, the findings indicated that the total membrane protein content in goat colostrum exhibited greater similarity to that found in human milk. Additionally, goat milk exhibited higher milk fat globule size, as well as a higher total lipid and protein content than human milk. A total of 1461 lipid molecules across 61 subclasses were identified in goat milk and human milk. The contents of glycerides and glycerophospholipids were higher in goat colostrum, whereas sphingolipids and fatty acids were more abundant in human milk. Meanwhile, the compositions of lipid subclasses were inconsistent. Acquiring real-time feeding information for monitoring lactating sows and their feeding requirements is a challenging task. Real-time data represent an important input for numerous tasks, such as disease monitoring, nutritional regulation, and feeding modeling. However, concurrently monitoring large numbers of sows and processing the real-time information for modeling is challenging using existing platforms. In this paper, we describe the design and development of a system that monitors and processes sows' feed and water consumption in real time. The system was custom-developed using open-source networking technologies. The system consists of three components: an electronic sow feeder connected to a central controller via a CAN network, an MQTT service cluster, and a data processing program. The MQTT service cluster uses Netty to develop a single service node, and it uses Zookeeper and Redis to complete node registration, discovery, and scheduling. The data processing program is based on Spark and Flink. We conducted comparative testing of three common codecs (Java Serializer, Marshalling, and Protostuff) to further speed up data transmission. The results of the experiment show that, with three service nodes, the system can concurrently monitor up to 20,000 sows. This research paper addresses the effects of parity and SCC threshold on the udder morphology, milkability traits, and milk composition during mid-lactation in Canarian goats. Results showed a positive association between SCC and total bacterial count with the parity, indicating an impairment of the udder epithelial tissue, as well as a greater susceptibility to bacterial infections in older goats. The effects of parity and somatic cell count in milk (SCC) threshold on the udder morphology, milkability traits, and milk composition was evaluated in 41 Canarian goats in mid-lactation. The animals were divided according to parity (1st, 2nd, and 3rd), and a SCC threshold of 2000 × 103 cells/mL in milk was set to evaluate the effect of this factor on the different measured parameters. Results showed that primiparous goats had the udder smaller and less distended than multiparous goats, but no differences were detected on milk flow parameters. Furthermore, SCC and total bacterial count (TBC) tended to be higher when the parity increased. On the other hand, goats with SCC ≤ 2000 × 103 had higher cistern-floor distance (CF) and lower TBC values compared with those goats with a count above the predetermined threshold. The results suggest that a reduction in SCC can be achieved by a selection of udder morphological traits. Milk holds a high nutritional value and is associated with diverse health benefits. The understanding of its composition of (poly)phenolic metabolites is limited, which necessitates a comprehensive evaluation of the subject. This study aimed at analyzing the (poly)phenolic profile of commercial milk samples from cows and goats and investigating their sterilization treatments, fat content, and lactose content. Fingerprinting of phenolic metabolites was achieved by using ultra-high-performance liquid chromatography coupled with triple-quadrupole mass spectrometry (UHPLC-QqQ-MS/MS). Two hundred and three potential microbial and phase II metabolites of the main dietary (poly)phenols were targeted. Twenty-five metabolites were identified, revealing a diverse array of phenolic metabolites in milk, including isoflavones and their microbial catabolites equol and O-desmethylangolensin, phenyl-γ-valerolactones (flavan-3-ol microbial catabolites), enterolignans, urolithins (ellagitannin microbial catabolites), benzene diols, and hippuric acid derivates. Goat's milk contained higher concentrations of these metabolites than cow's milk, while the sterilization process and milk composition (fat and lactose content) had minimal impact on the metabolite profiles. Thus, the consumption of goat's milk might serve as a potential means to supplement bioactive phenolic metabolites, especially in individuals with limited production capacity. The aim of this study was to investigate the effect of calcareous marine algae (Lithotamium calcareum)-based rumen content buffer included in total mixed ration, fed to 34 high-producing, peak lactation Holstein dairy cows (group A, n = 17; group B, n = 17). It was hypothesized that through a rumen content buffering effect, buffer would improve feeding behavior, feed intake, rumen function, and performance. Differences between the experimental and control total mixed ration in most parameters under study (feed intake, rumen content acidity, feces composition, milk yield, and milk composition) were statistically insignificant. However, statistically significant differences were noted in the length of eating and chewing epizodes (feeding time; rumination time) between groups. Feed intake was in line with this, although the difference was only numerical and statistically insignificant. This may simply mean that the buffer effects takes some time to show but persists after the buffer withdrawal. One of the reasons for not achieving a significant improvement in other parameters might be that the cows were fed a typical, well-balanced ration that did not induce acidosis. The aim of the study was to investigate the effect of calcareous marine algae (Lithotamium calcareum)-based rumen content buffer (CMA) included in concentrated feed within total mixed ration (TMR), fed to 34 peak lactation (87–144 days in milk) Holstein dairy cows, randomized into two groups (group A, n = 17; group B, n = 17), wearing collars with accelerometers, and housed a in barn with automatic feed-weigh troughs. During the first phase P1, group A received TMR with CMA (TMR-E) and group B was fed TMR without the buffer (TMR-C). For P2, the treatments in the groups were exchanged. Feed intake, feeding time (FT), rumination time (RT), milk yield, milk composition, and rumen pH were measured by barn technologies, and rumen fluid and feces composition were analyzed in the laboratory. Differences between the TMR-E and TMR-C in most parameters under study were statistically insignificant, except overall FT and RT, which differed significantly between the groups. Group A, feeding at P1 by TMR-E, exhibited higher FT and RT than Group B (202 min/cow/day vs. 184 min/cow/day, and 486 min/cow/day vs. 428 min/cow/day, respectively). Each cow's milk lactose, fat, protein, and fat-to-protein ratio were continuously monitored using the BROLIS HerdLine in-line milk analyzer (Brolis Sensor Technology, Vilnius, Lithuania). The RumiWatch noseband sensor (RWS; ITIN + HOCH GmbH, Fütterungstechnik, Liestal, Switzerland) was employed to measure the biomarkers of the rumination, feeding, and locomotion behavior. The measurements were recorded over 5 days at the same time (during morning milking). A total of 502 cows were examined. During these 5 days, 2510 measurements were taken. Based on the lactose content in their milk, the cows were divided into two categories: the first group consisted of cows with milk lactose levels below 4.70%, while the second group included cows with milk lactose levels of 4.70% or higher. Our study showed that cows with higher milk lactose concentrations (≥4.70%) produced significantly more milk (16.14% increase) but had a lower milk protein concentration (5.05% decrease) compared to cows with lower lactose levels. These cows also exhibited changes in rumination and feeding behaviors, as recorded by the RWS: there was an increase in the mastication and rumination behaviors, evidenced by a 14.09% rise in other chews and a 13.84% increase in rumination chews, along with a 16.70% boost in bolus activity. The objective of this study was to evaluate and develop equations to predict forage intake and growth of calves throughout the suckling period of beef calves grazing on forage or dairy calves fed harvested forage. Milk and forage intake and body weight data for individual animals were collected from published theses (one using bottle-fed dairy calves and one using suckling beef calves). A nutrition model was constructed using milk and forage intake equations and growth equations. Additional datasets were compiled from the literature to develop equations to adjust the original nutrition model for forage digestibility, milk composition, and growth. In general, the original nutrition model predicted the forage intake and body weight of dairy calves with moderate-to-high precision (CCC = 0.234 to 0.929) and poor accuracy (MB = −341.16 to −1.58%). Additionally, the original nutrition model predicted forage intake and body weight in beef calves with poor-to-moderate precision (CCC = 0.348 to 0.766) and accuracy (MB = 6.39 to 57.67%). Adjusted nutrition models performed better with the best model precisely (CCC = 0.914) predicting forage intake and precisely (CCC = 0.978) and accurately (MB = 2.83%) predicting body weight in dairy calves. The best adjusted nutrition model predicted forage intake and body weight with high precision (CCC = 0.882 and 0.935) and moderate accuracy (MB = −7.01 and −7.34) in beef calves. This investigation aimed to assess how the somatic cell count (SCC) impacts milk production and the physicochemical quality of milk, including the effects of centrifugation and microfiltration on SCCs and the yield of fresh cheese. Milk production was analysed at different somatic cell counts (SCCs) to observe how centrifugation and microfiltration affected the removal of SCs from the milk and the yield of fresh cheese. Tukey's test was employed at a 5% significance level to compare the chemical composition of the milk, milk production, and fresh cheese yield when using microfiltered and/or centrifuged milk. Milk with an SCC of ≤200,000 somatic cells (SCs) per mL exhibited higher yields. This study looked at the effects of a single 500 mg dose of rbST on the lipid profile, liver and kidney function, and physical, chemical, and cellular constitution of milk in 14 buffalo over 14 days, from the 100th day of lactation, compared with 14 animals in a control group. From the first day after rbST, there was a rise in beta-hydroxybutyrate (β-HBO), possibly due to higher dry matter intake or the biotransformation of NEFA into β-HBO. The treatment did not influence blood glucose, non-esterified fatty acids (NEFAs), triglycerides, cholesterol, total protein, albumin, AST, GGT, bilirubin, urea, or creatinine levels. In 71.3% of the buffalo, there was a gradual increase in milk production, with the maximal response occurring in the first week followed by a gradual decrease, whilst in 21.4%, the increase in production occurred between 7 and 10 days. Only 7.1% of the animals did not respond. On the 3rd, 5th, 7th, and 10th days after treatment, an increase was found in daily milk production between the two groups equal to 1.04, 1.52, 1.42, and 1.06 L, respectively. In relative terms, this means an increase in milk production, respectively, of 15.1%, 21.0%, 19.8%, and 15.1%. The constitution of the milk showed no difference in the amounts of fat, lactose, total solids, or somatic cell count; however, on the third day after rbST administration, there was a decrease in protein. Notably, from the fifth day, the protein values showed no statistical difference. The administration of Bacilli to dairy cows exerts beneficial effects on dry matter intake, lactation performance, and milk composition, but the rationale behind their efficacy is still poorly understood. In this work, we sought to establish whether cellulases and xylanases, among the enzymes secreted by B. subtilis, are involved in the positive effect exerted by Bacilli on ruminal performance. We took advantage of two isogenic B. subtilis strains, only differing in the secretion levels of those two enzymes. A multi-factorial study was conducted in which eight feed ingredients were treated in vitro, using ruminal fluid from cannulated cows, with cultures of the two strains conveniently grown in a growth medium based on inexpensive waste. Feed degradability and gas production were assessed. Fiber degradability was 10% higher (p < 0.001) in feeds treated with the enzyme-overexpressing strain than in the untreated control, while the non-overexpressing strain provided a 5% increase. The benefit of the fibrolytic enzymes was maximal for maize silage, the most recalcitrant feed. Gas production also correlated with the amount of enzymes applied (p < 0.05). Our results revealed that B. subtilis cellulases and xylanases effectively contribute to improving forage quality, justifying the use of Bacilli as direct-fed microbials to increase animal productivity. The aim of our study was to investigate and quantify the impact of heat stress on the milk fat-to-protein ratio (F/P) and the metabolic profile in dairy cows, utilizing in-line registration methods for accurate and real-time data collection. This study was carried out in Lithuania at coordinates 55.819156, 23.773541, from 1 June 2023 until 31 August 2023. Milk composition, including fat and protein, was measured using a BROLIS HerdLine in-line milk analyzer from Brolis Sensor Technology, Vilnius, Lithuania. During the general clinical examinations (twice per week), blood samples were collected and determined for GLU—blood glucose concentration; BHB—blood β-hydroxybutyrate concentration; AST—aspartate transaminase activity; GGT—gamma-glutamyltransferase activity; and NEFAs—non-esterified fatty acids. The parameters based on the Temperature–Humidity Index (THI) were categorized into two groups: group 1, consisting of THI values less than 72, representing the comfort zone, and group 2, with THI values of 72 or higher, indicating a greater risk of thermal stress. Specifically, group 2 exhibited an 8.6% increase in the F/P ratio compared to group 1 (p = 0.049). Additionally, there was a 4.2% decrease in glucose levels in group 2 (p = 0.056) and a notable 5.8% decrease in albumin levels compared to group 1 (p < 0.001). We found a very weak, non-significant correlation between humidity and the milk fat-to-protein ratio (r = 0.043, p = 0.447) and a similarly negligible correlation with Beta-Hydroxybutyrate (BHB; r = 0.046, p = 0.417). We observed significant changes in milk composition, particularly an increase in the milk fat-to-protein ratio, and alterations in metabolic indicators like glucose, albumin, and liver enzymes. These changes, indicative of a negative energy balance and altered metabolic processes such as gluconeogenesis and lipolysis, correspond to previous research. Karachai goats are a breed of local selection, the milk of which is widely used in the regions of the North Caucasus for the production of unique cheeses. While a relatively large number of genome studies have been carried out for transboundary goat breeds, such as Saanen goats, there is not yet enough research on local goat breeds. That is why we conducted a genome-wide association study (GWAS) on the milk component traits of Karachai goats. The use of genomic evaluation in the selection of Karachai goats will increase the rate of breeding progress in terms of enhancing milk productivity traits and thereby increase the profitability of their use. This study is first to perform a genome-wide association study (GWAS) to investigate the milk quality traits in Karachai goats. The objective of the study was to identify candidate genes associated with milk composition traits based on the identification and subsequent analysis of all possible SNPs, both genome-wide (high-confidence) and suggestive (subthreshold significance). To estimate the milk components, 22 traits were determined, including several types of fatty acids. DNA was extracted from ear tissue or blood samples. A total of 167 Karachai goats were genotyped using an Illumina GoatSNP53K BeadChip panel (Illumina Inc., San Diego, CA, USA). Overall, we identified 167 highly significant and subthreshold SNPs associated with the milk components of Karachai goats. A total of 10 SNPs were located within protein-coding genes and 33 SNPs in close proximity to them (±0.2 Mb). The largest number of genome-wide significant SNPs was found on chromosomes 2 and 8 and some of them were associated with several traits. The greatest number of genome-wide significant SNPs was identified for crude protein and lactose (6), and the smallest number—only 1 SNP—for freezing point depression. No SNPs were identified for monounsaturated and polyunsaturated fatty acids. Functional annotation of all 43 SNPs allowed us to identify 66 significant candidate genes on chromosomes 1, 2, 3, 4, 5, 8, 10, 13, 16, 18, 21, 23, 25, 26, and 27. We considered these genes potential DNA markers of the fatty acid composition of Karachai goat milk. Also, we found 12 genes that had a polygenic effect: most of them were simultaneously associated with the dry matter content and fatty acids (METTL, SLC1A 8, PHACTR1, FMO2, ECI1, PGP, ABCA3, AMDHD2). In this study, twenty-four pairs of parental pigeons were randomly divided into four groups, with six replicates, and each pair feeding three squabs. The control group drank normal water. The E. faecium group, B. subtilis group, and mixed group drank water supplemented with 3 × 106 CFU/mL E. faecium, 2 × 107 CFU/mL B. subtilis, and a mixture of these two probiotics, respectively. The experiment lasted 19 days. The results demonstrated that the IgA and IgG levels were significantly higher in the milk of Group D pigeons than in the other groups. At the phylum level, Fimicutes, Actinobacteria, and Bacteroidetes were the three main phyla identified. At the genus level, Lactobacillus, Bifidobacterium, Veillonella, and Enterococcus were the four main genera identified. Simple Summary: Extensive goat breeding follows principles of sustainable agriculture, and it can be achieved with local breeds because their genome has been shaped by centuries of interaction with the environment in which goats were raised. The Sarda goat is well adapted to the arid environment of Sardinia (Italy). The aim of the present work was to study the variability of the casein genes, the main proteins of milk. We measured milk traits such as protein, fat, lactose, total solids, and milk yield, and performed association analyses with casein gene variants. The variability of the four casein genes was associated with milk protein, fat, total solids, and milk energy. The main finding was that intronic variants of the CSN1S2 gene (encoding the αs2-casein protein) were associated with milk yield and protein and fat content. This information might be used in selection schemes, and in future investigations aiming to disclose the direct link between genotype and phenotype. This work aimed to assess the variability of casein genes in a population of 153 bucks and 825 lactating does of the Sarda breed, and to perform association analysis between polymorphic sites and milk yield and composition traits. To genotype the casein genes, we chose an SNP panel including 44 SNPs mapping to the four casein genes CSN1S1, CSN2, CSN1S2, and CSN3. Genotyping (made by KASP™ genotyping assay, based on competitive allele-specific PCR) revealed the high variability of the Sarda goat, and haplotype analysis revealed linkage disequilibrium (LD) between CSN1S1 and CSN2 genes, in addition to two LD blocks within the CSN1S2 and two LD blocks within the CSN3 gene, in bucks and does. Association analysis revealed that variability at all four casein genes was associated with milk protein content, total solids, and milk energy. The three Ca-sensitive casein genes were associated with lipid content, and CSN1S2 showed a unique pattern, with intron variants associated with milk yield, in addition to milk pH, NaCl, and SCS (Somatic Cell Score). Proteins in donkey milk (DM) have special biological activities. However, the bioactive proteins and their expression regulation in donkey milk are still unclear. Thus, the differentially expressed proteins (DEPs) in DM in different lactation stages were first investigated by data-independent acquisition (DIA) proteomics. A total of 805 proteins were characterized in DM. The composition and content of milk proteins varied with the lactation stage. A total of 445 candidate DEPs related to biological processes and molecular functions were identified between mature milk and colostrum. The 219 down-regulated DEPs were mainly related to complement and coagulation cascades, staphylococcus aureus infection, systemic lupus erythematosus, prion diseases, AGE-RAGE signaling pathways in diabetic complications, and pertussis. The 226 up-regulated DEPs were mainly involved in metabolic pathways related to nutrient (fat, carbohydrate, nucleic acid, and vitamin) metabolism. Some other DEPs in milk from the lactation period of 30 to 180 days also had activities such as promoting cell proliferation, promoting antioxidant, immunoregulation, anti-inflammatory, and antibacterial effects, and enhancing skin moisture. DM can be used as a nutritional substitute for infants, as well as for cosmetic and medical purposes. An increasing number of dairy farmers in New Zealand (NZ) have adopted a once-a-day (OAD) milking production system, and little is known about the impact of this production system on milk protein composition. The objective of this study was to evaluate the effect of OAD milking on the protein composition in milk from individual cows. Milk was sampled in early, mid-, and late lactation from cows kept at Massey University farms Dairy No. 1 (OAD milking) and Dairy No. 4 (TAD milking) in Palmerston North, NZ. The yields of total milk and milk solids, the proximate composition, and the protein composition were determined. Results showed that OAD milking yielded less milk and milk solids than TAD milking. However, no significant differences in protein, fat, and lactose contents were found. Over a period of six months, data related to milk quality and udder health and bedding sampling were collected from eight dairy farms. Monthly measurements of the bedding temperature and wind speed inside the CBPB were taken, while temperature and relative humidity data inside the CBPB were recorded using a datalogger. Bedding samples were subjected to analysis of moisture, pH, microbiological count, and carbon/nitrogen ratio. Data on milk composition (fat, protein, milk urea nitrogen, and total solids) and quality (somatic cell count and standard plate count) of bulk tank milk were obtained from DHIA results. Canonical correlation analyses were used to evaluate the association between the analyzed group variables, and linear regression models were used to identify associations between bedding characteristics and SCM occurrence in the studied herds. The bedding characteristics that most influenced milk composition and quality were moisture, temperature at 30 cm depth (T30), and bedding pH. Environmental variables played an important role in bedding composting, as they were closely related to the surface temperature and pH. Overall, 62.71% of the variation in milk quality and composition could be explained by the bedding variables, and 77.50% of the variation in the bedding variables was associated with environmental variables. Nutritional indices, such as the atherogenicity index and thrombogenicity index, remained within the recommended values. Antioxidant properties, including total phenolic content, DPPH, FRAP, and ABTS, showed significant differences, with higher values toward the end of the study. For this reason, health and welfare risks are a great concern to the dairy industry, with mastitis still being among the most significant, as it decreases milk quality and farmer profit. In the present study, 1004 dairy cows from one farm in Greece were included in the study. Data regarding the animals were collected from the farm's database, while for each cow, health and welfare traits were recorded before collecting milk samples. Higher lactation periods and teat hyperkeratosis seemed to significantly lower milk quality traits and increase somatic cell counts. Despite years of research devoted to bovine mastitis, the disease remains a serious problem in dairy cattle, causing economic losses to the dairy industry worldwide due to reduced milk yield, lower milk quality, drug costs and early culling of cows. The aim of this study is to determine the importance of several risk factors affecting milk quality in dairy cows, as well as to highlight proper milking techniques. A cross-sectional study was performed in one Greek dairy farm with the inclusion of a total of 1004 Holstein Friesian cows in the study. The udder and teat traits were recorded for each cow, while individual milk samples were used to estimate the somatic cell count (SCC) and gross milk composition. To address the challenges of a long measurement period, high testing cost, and environmental pollution of traditional milk composition detection methods, a portable detection instrument was developed by combining multi-spectral sensors, machine learning algorithms, and an embedded system to rapidly detect the main components of milk. A broadband near-infrared (NIR) LED constant-current driver circuit and multi-spectral sensor module were designed to obtain six NIR features of milk samples. Based on a comparison of several machine learning algorithms, the XGBoost model was selected for training, and the trained model was ported to a Raspberry Pi unit for sample detection. The validation results showed that the coefficients of determination (R2) for the investigated protein and fat models were 0.9816 and 0.9978, respectively, and the corresponding mean absolute errors (MAE) were 0.0086 and 0.0079. (1) Background: The purpose of study was to compare the safety profile of glatiramer with natalizumab, alemtuzumab and ocrelizumab in pregnant and lactating women affected by multiple sclerosis (MS). (2) Methods: Individual case safety reports (ICSRs) were retrieved from the European spontaneous reporting system database (EudraVigilance). The reporting odds ratios (RORs) were computed to compare the reporting probability of events between natalizumab, alemtuzumab and ocrelizumab vs. glatiramer. (3) Results: A total of 1236 ICSRs reporting at least one DMT as a suspected drug were selected. More adverse drug reactions (ADRs) unrelated to pregnancy and breastfeeding (n = 1171; 32.6%) were reported than ADRs specific to pregnancy and breastfeeding (n = 1093; 30.4%). The most frequently reported unrelated ADR was MS relapse. Alemtuzumab and natalizumab seem to have a lower reporting probability of MS relapse compared to glatiramer (ROR 0.17, 95% CI 0.07–0.45 and ROR 0.34, 95% CI 0.20–0.57). Among pregnancy- and breastfeeding-related ADRs, the first most reported event was spontaneous abortion (n = 321; 8.9%). Natalizumab and ocrelizumab were associated with a higher reporting probability of spontaneous abortion compared to glatiramer (ROR 2.22, 95% CI 1.58–3.12; ROR 2.18, 95% CI 1.34–3.54, respectively), while alemtuzumab had a lower reporting frequency (ROR 0.32, 95% CI 0.17–0.60). Eighteen Nili Ravi buffaloes were assigned to three cooling strategies: CTL (handheld hosepipe cooling twice daily for 5 min), 3CS (sprinkler cooling three times daily), and 5CS (sprinkler cooling five times daily). The 5CS group produced 1.6 and 3.2 kg more milk per day than 3CS and CTL, respectively, with more milk fat. The buffaloes in the 5CS group also had lower core body temperatures and respiration rates than the CTL and 3CS groups. Furthermore, buffaloes in the 5CS group spent significantly more time eating and showed decreased blood cortisol levels. Our study demonstrated that increased cooling sessions had a beneficial effect on milk yield and composition, particularly by increasing milk fat content. Additionally, more cooling sessions positively influenced the welfare of dairy buffaloes by lowering respiration rate, body temperature, and cortisol levels. These findings underscore the critical importance of optimizing cooling practices in dairy buffalo management. Mastitis is one of the most important diseases in dairy cows, leading to substantial economic losses associated with decreased milk production and quality. Early detection of changes in metabolic and milk parameters is crucial for maintaining animal welfare and milk quality. This study aimed to detect patterns in metabolic and milk composition parameters in Serbian dairy cows affected by mastitis. It also examined the relationship between these factors in cows with clinical and subclinical mastitis, as well as in healthy cows. This study included 60 Holstein-Friesian cows with the same body score condition that were in the same lactation phase. They were divided into three groups of 20: clinical and subclinical mastitis and a control group of healthy cows. The categorization was based on clinical udder health and the California mastitis test. Blood serum metabolic profiles were measured using a Rayto spectrophotometer (Shenzhen, China), and milk composition was determined using MilcoScanTM (Foss, Hilleroed, Denmark) and FossomaticTM (Foss, Hilleroed, Denmark) instruments. Significant increases in non-esterified fatty acids (NEFAs), beta-hydroxybutyrate (BHB), total protein, globulin, urea, total bilirubin, magnesium, and enzyme activity were noted in mastitis-affected cows compared to healthy ones. Additionally, mastitis-affected cows had higher total protein and globulin levels and increased somatic cell counts (SCCs), while albumin concentrations were decreased. Furthermore, a negative correlation between total protein and lactose suggested inflammation leading to reduced lactose levels due to cell damage, infection, and lactose use by mastitis pathogens. Hence, indicators of the energy and protein status of the metabolic profile, together with the chemical composition of milk, may be significant diagnostic tools for detecting, monitoring, and predicting the outcome of mastitis in cows. Simple Summary: In this study, our goal was to develop a simple-to-use method for evaluating the health status of dairy cows by analyzing the milk composition data collected via in-line sensors. Cows were divided into the following groups: subclinical ketosis (n = 62), subclinical acidosis (n = 14), and healthy group (H; n = 20). Our focus was on quantifying the levels of fat and protein in cow milk and uncovering intriguing patterns. Specifically, cows struggling with a particular health issue (subclinical ketosis) displayed an elevated fat-to-protein ratio in their milk. We scrutinized their blood samples and unveiled correlations between specific blood constituents and variations in milk composition. Over the study period, farm visits occurred biweekly on consistent days, during which milk composition (specifically fat and protein) was measured using a BROLIS HerdLine in-line milk analyzer (Brolis Sensor Technology, Vilnius, Lithuania). Clinical examinations were performed at the same time as the farm visits. Blood was drawn into anticoagulant-free evacuated tubes to measure the activities of GGT and AST and albumin concentrations. NEFA levels were assessed using a wet chemistry analyzer. Using the MediSense and FreeStyle Optium H systems, blood samples from the ear were used to measure the levels of BHBA and glucose in plasma. Daily blood samples were collected for BHBA concentration assessment. All samples were procured during the clinical evaluations. The cows were categorized into distinct groups: subclinical ketosis (SCK; n = 62), exhibiting elevated milk F/P ratios without concurrent clinical signs of other post-calving diseases; subclinical acidosis (SCA; n = 14), characterized by low F/P ratios (<1.2), severe diarrhea, and nondigestive food remnants in feces, while being free of other post-calving ailments; and a healthy group (H; n = 20), comprising cows with no clinical indications of illness and an average milk F/P ratio of 1.2. The milk fat-to-protein ratios were notably higher in SCK cows, averaging 1.66 (±0.29; p < 0.01), compared to SCA cows (0.93 ± 0.1; p < 0.01) and healthy cows (1.22). A 36% increase in milk fat-to-protein ratio was observed in SCK cows, while SCA cows displayed a 23.77% decrease. Significant differences emerged in AST activity, with SCA cows presenting a 26.66% elevation (p < 0.05) compared to healthy cows. Moreover, SCK cows exhibited a 40.38% higher NEFA concentration (p < 0.001). A positive correlation was identified between blood BHBA and NEFA levels (r = 0.321, p < 0.01), as well as a negative association between BHBA and glucose concentrations (r = −0.330, p < 0.01). Notably, AST displayed a robust positive correlation with GGT (r = 0.623, p < 0.01). In light of these findings, this study posits that milk fat-to-protein ratio comparisons could serve as a non-invasive indicator of metabolic health in cows. Simple Summary: The use of by-products from the olive oil industry in livestock farming is an important resource due to their high content of biomolecules with strong antioxidant properties. In this study, 30 Italian Holstein–Friesian cows were fed with an Olea europaea L. polyphenol-enriched diet (500 mg/cow/day) to evaluate the effects of such supplementation on metabolic parameters and milk quality. Thirty Italian Holstein–Friesian dairy cows in the mid-lactation phase (90 to 210 days) were blocked into experimental groups based on parity class (namely, primiparous (P) (n = 10), secondiparous (S) (n = 10) and pluriparous (PL) (n = 10)) and received, for 60 days, Phenofeed Dry® at 500 mg/cow/day. Milk and blood samples were collected before the start of the treatment (T0), subsequently every 15 days (T1–T4) and at 45 days after the end of treatment (T5). In the serum, glucose and triglycerides, stress, the thyroid, lactation and sex hormones were measured; in the milk, lysozyme content as well as the fatty acid profile were assessed. In the whole animal, the enriched feed helped to maintain hormonal parameters in the physiological range while producing hypoglycemic (T4 vs. T0, for P and PL p < 0.001) and hypolipidemic effects (T4 vs. T0, for P p < 0.001 and for PL p < 0.01). β-casomorphin 7 (BCM7) is a bioactive peptide that is released during the digestion of the β-casein (in particular, the A1 variant) present in cow's milk. BCM7 has been linked to several health concerns such as gastrointestinal disorders. Milk processing alters the composition of milk, which in turn may affect its digestion, thus impacting the amount of BCM7 that is released. This study aimed to understand the impact of microfiltration on BCM7 release after the in vitro digestion (mimicking in vivo digestion) of semi-skimmed filtered milk compared to that of pasteurized milk and pasteurized Jersey milk (which does not contain A1 β-casein, the main source of BCM7). Goat milk is a vital, sustainable dairy product source that contributes significantly to the global dairy market. Different goat breeds may produce milk with varying compositions during different lactation seasons. Therefore, milk composition and quality need to be monitored. As a versatile and non-destructive method, Near Infrared Spectroscopy (NIRS) offers the potential for evaluating various milk parameters. In this study, we aim to establish NIRS models for classifying goat's milk samples between the native Red and Alpine Breed at first and fifth lactation season. Our objective is to assess the efficacy and cost-effectiveness of NIRS as a viable alternative to traditional laboratory techniques. Forty-five milk samples were collected from two breeds, French Alpine goats, and native Red goats. Within each breed, half of the samples originated from goats in their first lactation season and the other half from goats in their fifth lactation season. The NIRS measurements were conducted using homogenized, untreated, and diluted milk samples. The experiment was performed with a benchtop spectrophotometer (740–1700 nm). Qualitative analysis using linear discriminant analysis (LDA) was conducted, demonstrating the feasibility of NIRS in effectively classifying samples. The achieved accuracy ranged between 67.19% to 100%, depending on the breed and lactation period. These findings underscore the potential of NIRS as a rapid, non-destructive analytical tool for quality monitoring and milk analysis. The objective of our study was to evaluate the predictive ability of a multi-trait genomic prediction model that accounts for interactions between marker effects to estimate heritability and genetic correlations of traits including 305-day milk yield, milk fat percentage, milk protein percentage, milk lactose percentage, and milk dry matter percentage in the Polish Holstein Friesian cow population. For this aim, 14,742 SNP genotype records for 586 Polish Holstein Friesian dairy cows from Poland were used. Single-Trait-ssGBLUP (ST) and Multi-Trait-ssGBLUP (MT) methods were used for estimation. We examined 305-day milk yield (MY, kg), milk fat percentage (MF, %), milk protein percentage (MP, %), milk lactose percentage (ML, %), and milk dry matter percentage (MDM, %). The results showed that the highest marker effect rank correlation was found between milk fat percentage and milk dry matter. The weakest marker effect rank correlation was found between ML and all other traits. Obtained accuracies of this study were between 0.770 and 0.882, and 0.773 and 0.876 for MT and ST, respectively, which were acceptable values. All estimated bias values were positive, which is proof of underestimation. The highest heritability value was obtained for MP (0.3029) and the lowest heritability value was calculated for ML (0.2171). Estimated heritability values were low for milk yield and milk composition as expected. The strongest genetic correlation was estimated between MDM and MF (0.4990) and the weakest genetic correlation was estimated between MY and ML (0.001). Simple Summary: Gut microbial profiles have been shown to influence a range of physiological processes contributing to animal performance. In particular, gut microbiota have a close relationship with digestion and metabolism, indicating they may be correlated to weight gain. As previous studies have discussed, gut microbiota ranging from the rumen to the hindgut can influence and potentially indicate differences in important production traits such as milk composition in dairy cattle and feed efficiency in beef cattle. Therefore, the objective of this study was to characterise the faecal microbial profiles of Angus steers with high and low average daily gain (ADG) at both weaning and yearling stages. The results indicate that at yearling stage, there was a distinct difference in the faecal microbial profiles of steers with high or low ADG. Primarily, we observed a reduction in species richness and decrease in between-sample phylogenetic similarity in the high ADG group. Microbial communities inhabiting the gut have the ability to influence physiological processes contributing to livestock production and performance. Livestock enterprises rely on animal production traits such as growth performance for profit. Previous studies have shown that gut microbiota are correlated to growth performance and could even influence it. The aim of this study was to characterise the faecal microbial profiles of Angus steers with high and low ADG at both weaning and yearling stages by profiling 16S rRNA gene sequences from rectal faecal samples. When microbial profiles were compared in terms of relative abundances, LEfSe analysis, alpha diversity metrics, and beta diversity, at the weaning stage, few significant differences were found between the high and low ADG groups. However, at yearling stage, microbial profiles significantly differed between the high and low ADG groups. The relative abundances of eight phyla and six genera significantly differed between the two groups. Alpha diversity metrics showed a significant decrease (p = 0.001) in species richness in the high ADG group. Similarly, beta diversity analysis showed that samples clustered clearly according to high and low ADG groups at yearling stage, indicating that phylogenetic similarity between the two ADG groups was significantly reduced (p = 0.005). Simple Summary: This study evaluated the dietary effects of extruded linseed and oregano on the quality traits and sensory properties of meat obtained from kids of a Lucanian goat population named "Facciuta Lucana". Thirty-six male kids were divided into three homogeneous groups fed a control feed (C), a diet containing 3% extruded linseed (L), or 3% linseed plus 0.6% oregano (L + O). Meat from the Longissimus lumborum muscle obtained from linseed-fed groups showed a lower content of fat and total saturated fatty acids and, in turn, an increase in monounsaturated and polyunsaturated fatty acids, and conjugated linoleic acid, with benefits for human health. Oregano addition to the linseed diet proved to be effective in preserving meat shelf-life, since it lowered the malondialdehyde concentration after 10 days of storage, and improved meat succulence, tenderness, juiciness, and overall acceptance. Bovine milk is a nutrient-dense food and a major component of the human diet. Therefore, understanding the factors that impact its composition is of great importance. Applications of metabolomics provide in-depth analysis of the metabolite composition of milk. The objective of this research was to examine the impact of lactation stage on bovine milk metabolite levels. Metabolomic analysis of bovine milk powder samples across lactation (N = 18) was performed using nuclear magnetic resonance (1H-NMR) spectroscopy and liquid chromatography–tandem mass spectrometry (LC-MS/MS). Forty-one metabolites were identified and quantified in the 1H-NMR spectra. Statistical analysis revealed that 17 metabolites were significantly different across lactation stages (FDR < 0.05), of which the majority had higher levels in early lactation. In total, 491 metabolites were measured using LC-MS/MS, of which 269 had significantly different levels across lactation (FDR < 0.05). Compound classes significantly affected by lactation stage included phosphatidylcholines (59%) and triglycerides (64%), of which 100% of phosphatidylcholines and 61% of triglycerides increased from early lactation onwards. Our study demonstrates significant differences in metabolites across the stages of lactation, with early-lactation milk having a distinct metabolomic profile. Simple Summary: Compared with multiparous sows, primiparous sows have poor reproductive performance and insufficient nutritional reserves, resulting in a decreased growth performance in their offspring, which affects the efficiency of pig production. Yeast culture (YC) is rich in nutrients, and the supplementation of yeast culture in gestation and lactation diets is beneficial to improving the nutritional level of primiparous sows. Therefore, the objective of this experiment was mainly to investigate the effects of supplementing 0.5% and 0.8% yeast cultures into diets during gestation and lactation on the reproductive performance and intestinal health of primiparous sows, and the nutrient content of their colostrum and milk. The results showed that the dietary yeast cultures improved reproductive performance, improved the intestinal environment and energy metabolism, and increased the nutrient content of colostrum from primiparous sows. This study provides a theoretical basis for improving the performance of primiparous sows, and also provides a reference for the application of yeast cultures in sow production. The objective of this study was to evaluate the effects of yeast culture (YC) on reproductive performance, gut microbiota, and milk composition in primiparous sows. A total of 60 primiparous sows were randomly assigned to the control group (CON) and YC group (0.5% YC during gestation and 0.8% YC during lactation) consisting of 30 replicates, with one sow in each. The results showed that dietary YC supplementation increased the piglet birth weight and backfat thickness at 28 d of lactation (p < 0.05). Dietary YC supplementation increased the apparent total tract digestibility (ATTD) of gross energy and calcium during lactation, the content of acetic acid and propionic acid at 110 d of gestation, and the content of acetic acid and butyric acid at 28 d of lactation in feces (p < 0.05). Furthermore, dietary YC supplementation decreased the abundance of Firmicutes, Lachnospiraceae_XPB1014_group, and Terrisporobacter (p < 0.05), and increased the abundance of Prevotellaceae_NK3B31_group and Rikenellaceae_RC9_gut_group (p < 0.05). Compared to the control group, dietary YC supplementation increased the fat and lactose content of the colostrum (p < 0.05). Metabolomics analysis showed that YC increased 26 different metabolites in the colostrum. Among them were mainly pantothenic acid, proline, isoleucine, phenylalanine, acylcarnitine, and other metabolites. In conclusion, these results suggested that dietary YC supplementation improves reproductive performance and gut health and increases the nutrient content in the colostrum of primiparous sows. Simple Summary: Expanding our understanding of the changes induced by chitosan supplementation on the rumen microbiome and its complexity is not only a key factor in finding strategies to decrease CH4 production from ruminants but also in enhancing our knowledge on the mechanisms of action of chitosan in the rumen. Therefore, this study aimed to expand the knowledge about the activity and mode of action of chitosan on methanogenesis and rumen microbial taxonomy. This is the first study, to our knowledge, accounting for the effects of chitosan on ruminal microorganisms on in vivo conditions and using whole-metagenome sequencing. Chitosan did not affect rumen microbial diversity but induced shifts in the relative abundance of some microbial taxa. Chitosan increased relative abundance of Anaeroplasma and tended to reduce fibrolytic fungi and protozoa, resulting in a shift of ruminal fermentation towards a lower acetic to propionic ratio. However, CH4 emissions, microbial protein synthesis, and productive performance were not affected by CHI supplementation. The dose of chitosan used in this study seemed not to be large enough to have a concomitant improvement in animal performance and CH4 reduction. Therefore, future works with higher doses would be necessary to assess the potential use of this additive as methane inhibitor. This study aimed to expand the knowledge about the activity and mode of action of CHI on methanogenesis and rumen microbial populations in vivo. A total of 16 lactating dairy cows were distributed in two groups, one of them receiving 135 mg CHI/kg body weight daily. The effect on productive performance, milk composition, fermentation efficiency, methane emissions, microbial protein synthesis, and ruminal microbial communities was determined. Supplementation with CHI did not affect rumen microbial diversity but increased the relative abundance (RA) of the bacteria Anaeroplasma and decreased those of rumen ciliates and protozoa resulting in a shift towards a lower acetic to propionic ratio. However, no effect on milk yield or methane intensity was observed. In conclusion, supplementing 135 mg CHI/kg body weight increased the RA of Anaeroplasma and decreased those of rumen ciliates and protozoa, both being related to fiber degradation in the rumen in different ways and resulted in a shift of ruminal fermentation towards more propionate proportions, without affecting CH4 emissions, milk yield, or milk composition. A study was undertaken on milk and caciocavallo cheese from Podolica cattle in the Basilicata Region (Southern Italy), with a view of the possible identification of specific traits useful to protect them from imitations. More than 800 individual milk samples and 29 bulk milk samples were taken in spring–early summer from cows registered in the genealogical book of the breed; moreover, 18 samples of caciocavallo cheese were taken in the same geographical area, 9 of which had been manufactured from Podolica milk. The obtained results confirmed the high aptitude of Podolica milk to cheesemaking, even though the exceptional dry weather in the period of sampling decreased the fat content with respect to the literature data. The presence of the variant A of α-lactalbumin, a characteristic trait of Podolica milk, was ascertained in only 14% of the animals considered in the study, indicating that this feature is disappearing in the population under study. The results on caciocavallo gave useful indications, because some possible peculiar characteristics were identified, such as the lower protein to fat ratio and some aroma descriptors. More research is needed to assess if these characteristics can be used for developing a multi-functional protocol, to be extended to all Italian Podolica populations, able to discriminate the cheese from imitations.
