Tomato (Solanum lycopersicon L.) is one of the most widely consumed and produced vegetable crops worldwide. It offers numerous health benefits due to its rich content of many therapeutic elements such as vitamins, carotenoids, and phenolic compounds. Biotic stressors such as bacteria, viruses, fungi, nematodes, and insects cause severe yield losses as well as decreasing fruit quality. Conventional breeding strategies have succeeded in developing resistant genotypes, but these approaches require significant time and effort. The advent of state-of-the-art genome editing technologies, particularly CRISPR/Cas9, provides a rapid and straightforward method for developing high-quality biotic stress-resistant tomato lines. The advantage of genome editing over other approaches is the ability to make precise, minute adjustments without leaving foreign DNA inside the transformed plant. The tomato genome has been precisely modified via CRISPR/Cas9 to induce resistance genes or knock out susceptibility genes, resulting in lines resistant to common bacterial, fungal, and viral diseases. A highly efficient low-cost adsorbent was prepared using raw and chemically modified cellulose isolated from sugarcane bagasse for decontamination of Cr(VI) from wastewater. First, cellulose pulp was isolated from sugarcane bagasse by subjecting it to acid hydrolysis, alkaline hydrolysis and bleaching with sodium chlorate (NaClO3). Then, the bleached cellulose pulp was chemically modified with acrylonitrile monomer in the presence Fenton's reagent (Fe+2/H2O2) to carry out grafting of acrylonitrile onto cellulose by atom transfer radical polymerization. The developed adsorbent (acrylonitrile grafted cellulose) was analyzed by X-ray diffraction analysis (XRD), scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FT-IR). Both raw cellulose and acrylonitrile grafted cellulose were used for chromium removal from wastewater. The effects of metal ion concentration, pH, adsorbent dose and time were studied, and their values were optimized. The optimum conditions for the adsorption of Cr(VI) onto raw and chemically modified cellulose were: metal ion concentration: 50 ppm, adsorbent dose: 1 g, pH: 6, and time: 60 min. The maximum efficiencies of 73% and 94% and adsorption capacities of 125.95 mg/g and 267.93 mg/g were achieved for raw and acrylonitrile grafted cellulose, respectively. High removal efficiency was achieved, owing to high surface area of 79.92 m2/g and functional active binding cites on grafted cellulose. Isotherm and kinetics studies show that the experimental data were fully fitted by the Freundlich isotherm model and pseudo first-order model. The adsorbent (acrylonitrile grafted cellulose) was regenerated using three different types of regenerating reagents and reused thirty times, and there was negligible decrease (19%) in removal efficiency after using it for 30 times. Stem rust, caused by Puccinia graminis f.sp. tritici, is one of the most dangerous rust diseases on wheat. Through physiological, biochemical, and molecular analysis, the relationship between the change in resistance of 15 wheat cultivars to stem rust disease and the response of 41 stem rust resistance genes (Sr,s) as well as TTKSK, TTKST, and TTTSK races was explained. Some cultivars and Sr genes, such as Gemmeiza-9, Gemmeiza-11, Sids-13, Sakha-94, Misr-1, Misr-2, Sr31, and Sr38, became susceptible to infection. Other new cultivars include Mir-3 and Sakha-95, and Sr genes 13, 37, 40, GT, and FR*2/SRTT3-SRTT3-SR10 remain resistant. Some resistance genes have been identified in these resistant cultivars: Sr2, Sr13, Sr24, Sr36, and Sr40. Sr31 was not detected in any cultivars. Reactive oxygen species such as hydrogen peroxide and superoxide, enzymes activities (catalase, peroxidase, and polyphenoloxidase), and electrolyte leakage were increased in the highly susceptible cultivars, while they decreased in the resistant ones. Anatomical characteristics such as the thickness of the epidermis, ground tissue, phloem tissue and vascular bundle diameter in the midrib were decreased in susceptible cultivars compared with resistant cultivars. Our results indicated that some races (TTKSK, TTKST, and TTTSK) appeared for the first time in Egypt and many other countries, which broke the resistant cultivars. Epigenetic modifications, mainly aberrant DNA methylation, have been shown to silence the expression of genes involved in epigenetic diseases, including cancer suppression genes. Almost all conventional cancer therapeutic agents, such as the DNA hypomethylation drug 5-aza-2-deoxycytidine, have insurmountable side effects. To investigate the role of the well-known DNA protectant (ectoine) in skin cell DNA methylation and cancer cell proliferation, comprehensive methylome sequence analysis, 5-methyl cytosine (5mC) analysis, proliferation and tumorigenicity assays, and DNA epigenetic modifications-related gene analysis were performed. The results showed that extended ectoine treatment globally hypomethylated DNA in skin cells, especially in the CpG island (CGIs) element, and 5mC percentage was significantly reduced. Moreover, ectoine mildly inhibited skin cell proliferation and did not induce tumorigenicity in HaCaT cells injected into athymic nude mice. HaCaT cells treated with ectoine for 24 weeks modulated the mRNA expression levels of Dnmt1, Dnmt3a, Dnmt3b, Dnmt3l, Hdac1, Hdac2, Kdm3a, Mettl3, Mettl14, Snrpn, and Mest. Overall, ectoine mildly demethylates DNA in skin cells, modulates the expression of epigenetic modification-related genes, and reduces cell proliferation. Sustainable agriculture is a prerequisite for food and environmental security. Chemical fertilization, especially nitrogenous fertilization, is considered the most consumed for field crops. In rice crops, plants consume much less than half of the applied N-fertilizer. In the current investigation, multiple N environments were generated by applying different N doses of urea fertilizer to a permanent transplanted field for two successive summer growing seasons at the rice research and training center, Kafrelsheikh, Egypt. A set of 55 genotypes consisting of 25 Jabonica, 4 Tropical Japonica, 20 Indica, and 6 Indica/Japonica were transplanted under no N (0N), Low N (LN), medium N (MN), and High N (HN) (i.e., 0, 48, 96, and 165 Kg N ha−1, respectively). Highly significant differences were detected among the tested genotypes. AMMI analysis of variance revealed the existence of the genotype via nitrogen interaction (GNI) on yield performance. The GNI principal components (IPCA); IPCA1 and IPCA2 scores were significant and contributed values of 71.1 and 21.7%, respectively. The highest-ranked genotypes were MTU1010, IR22, SK2046, SK2058, IR66, and Yabani LuLu based on their grain yield means (30.7, 29.9, 29.5, 29.3, 28.8, and 28.5 g plant−1). These genotypes were grouped into the same subcluster (SCL) according to the stability analysis ranking matrix. Based on AMMI analysis and biplots, MTU1010 and Yabani LuLu showed yield stability across environments. Meanwhile, the which-won-where biplot showed that IR22 was superior under unfavorable N-levels and MTU1010 was stable across the different environments. The current study was conducted in a pot experiment with sand bed soil for two winter seasons (2019/20, 2020/21) to illuminate the impact of calcium lignosulfonate (Ca-LIGN) (100 mg/L) in alleviating various levels of NaCl (0, 100, 200, and 300 mM) on two barley cultivars, Giza132 and Giza133. Giza133 outgrew Giza132 under salinity stress by accumulating less Na+ content and retaining more K+ content. Surprisingly, Ca-LIGN was shown to be involved in both cultivars' capacity to efflux Na+ in return for greater K+ influx under 100 and 200 mM NaCl, resulting in an increased dry weight of shoots and roots as well as leaf area compared with the untreated salinity levels. Physiological parameters were measured as relative water content (RWC), electrolyte leakage rate (ELR), peroxidase activity (POD) in leaf and root and grain yield, and grain protein content were evaluated. Adding Ca-LIGN ameliorated both cultivars' growth in all the recorded characteristics. Under salinity stress, Ca-LIGN induced a higher RWC in both cultivars compared to those without Ca-LIGN. Although the ELR increased significantly in Giza132 leaves under the different NaCl concentrations compared to in Giza133 leaves, applying Ca-LIGN for both cultivars reduced the deterioration in their leaf and root by significantly lowering the ELR. As a result, applying Ca-LIGN to the salinity-affected plants (Giza133 and Giza132) under (100 and 200 mM NaCl), respectively, inhibited POD activity by about (10-fold, 6-fold, and 3-fold, 5-fold). The impact of Ca-LIGN on grain yield was more effective in Giza133 than in Giza132, with (61.46, 35.04, 29.21% and 46.02, 24.16, 21.96%) at various salinity levels. Moreover, while both cultivars recorded similar protein content under normal conditions, adding Ca-LIGN increased protein accumulation by raising salinity concentration until it reached 3% and 2% increases in both cultivars, Giza133 and Giza132, respectively, under 300 mM NaCl. A pot experiment was conducted during the 2019/2020 and 2020/2021 seasons to evaluate the effect of silver nanoparticles (SNPs), iron nanoparticles (FeNPs), zinc nanoparticles (ZnNPs), and nitrogen, phosphorus, and potassium nanoparticles (NPK NPs) and humic acid (HA) in improving the growth of Philodendron plants. Our findings indicated that the highest increase in plant height and leaf width was recorded with 60 mg/L SNPs. Additionally, the highest values in the number of leaves/plant were recorded with 60 mg/L SNPs compared to the control. FeNPs at 150 mg/L treatment gave the best result of total chlorophyll and carotenoid content, followed by SNPs at 60 mg/L and then NPK NPs at 2 mL/L in the two seasons. Furthermore, ZnNPs at 200 mg/L, SNPs at 20 mg/L, SNPs at 40 mg/L, and SNPs at 60 mg/L gave the best results of enzyme activity (catalase, peroxidase, and polyphenol oxidase). However, the treatments with 40 and 60 mg/L SNPs led to improve the anatomical characters of leaves and stem such as thickness of the blade, mesophyll tissue, xylem vessel diameter, vascular bundle dimension, stem diameter, and epidermis cell dimension compared with other treatments and the control. This experiment was conducted to investigate the effects of different nitrogen fertilizers (potassium nitrate and/or urea) on shoot parameters, relative growth rate, net assimilation rate, and nitrogen fractions, as well as to conduct transmission electron microscopy, of Vigna sinensis L. (cowpea) and Helianthus annuus L. (sunflower) leaves. A general improvement was recorded in the shoot parameters of the two plants, except for a decrease in the net assimilation rate by treatment of the two plants with 100% potassium nitrate plus 100% urea. The total nitrogen, insoluble protein, and total soluble nitrogen generally decreased in cowpea shoots from the treatments but increased in case of cowpea roots and sunflower shoots and roots. The examination of the ultrastructure changes in cowpea leaves confirmed the presence of two starch granules (in response to 100% potassium nitrate, 100% potassium nitrate plus 100% urea, and the control) and three granules (in response to 50% potassium nitrate plus 50% urea) and the disappearance of the starch granules (in response to 100% urea). Despite the starch granules not being detected in the leaves of the untreated sunflower, the treated plant showed the appearance of the highest number after treatment with 50% potassium nitrate plus 50% urea (2) and the most cell size with the 100% potassium nitrate treatment. Generally, our findings demonstrated that fertilization with 50% potassium nitrate plus 50% urea has the best influence on the growth parameters and nitrogen content in the two plants, but the magnitude of response was more pronounced in case of cowpea plants. Rice (Oryza sativa) is an important staple food crop worldwide, especially in east and southeast Asia. About one-third of rice cultivated area is under saline soil, either natural saline soils or irrigation with brackish water. Salinity stress is among the devastating abiotic stresses that not only affect rice growth and crop productivity but also limit its cultivation area globally. Plants adopt multiple tolerance mechanisms at the morphological, physiological, and biochemical levels to tackle salinity stress. To identify these tolerance mechanisms, this study was carried out under both a controlled glass house as well as natural saline field conditions using 22 green super rice (GSR) lines along with two local varieties ("IRRI 6 and Kissan Basmati"). Several morpho-physiological and biochemical parameters along with stress-responsive genes were used as evaluation criteria under normal and salinity stress conditions. Correlation and Principal Component Analysis (PCA) suggested that shoot-related parameters and the salt susceptible index (SSI) can be used for the identification of salt-tolerant genotypes. Based on Agglomerative Hierarchical Cluster (AHC) analysis, two saline-tolerant ("S19 and S20") and saline-susceptible ("S3 and S24") lines were selected for further molecular evaluation. Quantitative RT-PCR was performed, and results showed that expression of 1-5-phosphoribosyl -5-5-phosphoribosyl amino methylidene amino imidazole-4-carboxamide isomerase, DNA repair protein recA, and peptide transporter PTR2 related genes were upregulated in salt-tolerant genotypes, suggesting their potential role in salinity tolerance. However, additional validation using reverse genetics approaches will further confirm their specific role in salt tolerance. This study aimed to identify new rice lines and hybrids that are tolerant to water deficit and produce high yields under water stress conditions. A line × tester mating design was used to study the lines and testers' general combining ability (GCA) effects. The specific combining ability (SCA) of the hybrid rice combinations was measured under three different irrigation regimes; 6, 9, and 12 days. The study was carried out at the experimental farm of Sakha Agricultural Research Station, Sakha, Kafr El-Sheikh, Egypt, during the 2018 and 2019 rice growing seasons. Due to the genotypes and their partitions to the parents and the crosses, the mean squares were highly significant for all studied traits under the three irrigation regimes. The additive gene effects play an important role in expressing most of the studied traits. Therefore, the selection procedures based on the accumulation of the additive effect would be successful at improving these traits and the grain yield. The cytoplasmic male sterile (CMS) line G46A (L1) was the best combiner for most yield component traits in the three irrigation regimes. The newly devolved restorer lines T11, T1, T2, T5, T4, and T3, as well as the new hybrids L2 × T10, L2 × T6, L1 × T7, L1 × T5, L1 × T3, L2 × T7, L2 × T9, L2 × T8, L2 × T4, L1 × T4, L2 × T2, L1 × T8, L1 × T9, and L2 × NRL 10, showed good, desirable values of the studied traits such as earliness of flowering, short plant height, number of panicles/plant, panicle length, number of spikelets/panicle, number of filled grains/panicle, panicle weight, 1000-grain weight, hulling percentage, milling percentage, head rice percentage, and grain yield under the irrigation regimes of 6, 9, and 12 days. This research was carried out at the Experimental Farm of Sakha Agricultural Research Station, Sakha, Kafr El-Sheikh, Egypt, during the 2018–2020 rice-growing seasons to develop and evaluate four iso-cytoplasmic rice-restorer lines: NRL79, NRL80, NRL81, and NRL82, as well as Giza 178, with ten new hybrids in order to estimate genotypic coefficient, phenotypic coefficient, heritability in a broad sense, and advantage over Giza 178 as a check variety (control) of new restorer lines. This study also estimated combining ability, gene action, better-parent heterosis (BP), mid-parents heterosis (MP), and standard heterosis (SH) over Egyptian Hybrid one (IR69A × Giza 178) as a check hybrid (control) for grain yield, agronomic traits, and some grain quality characters in restorer lines and hybrids. The percentage of advantage over commercial-variety Giza 178 (check) was significant, and highly significant among the newly developed restorer fertility lines for all the studied traits. This indicates that the selection is a highly effective factor in improving these traits. New restorer fertility lines showed highly significant positive values over commercial restorer for grain yield; the values ranged from 51% for NRL80 to 100.4% for NRL82, respectively. Meanwhile, in regard to the grain shape of paddy rice, three lines of the promising lines showed highly significant negative desirable values compared with Giza 178; the values ranged from −7.7% for the NRL80 to −15.2% for NRL79, respectively. Based on the superiority of the new lines, the new lines can be used as new restorer fertility lines to breed promising new hybrids and new inbred rice lines or varieties. From the results of the testcross experiment, the four promising lines were identified as effective restorer fertility lines for two cytoplasmic male sterile (CMS) lines. Moreover, the six rice hybrids showed values for SH heterosis of grain yield/plant of more than 15% over the check hybrid variety, with high values of 1000-grain weight and desirable grain shape; these hybrids were G46A × NRL81 (125.1%), G46A × NRL80 (66.9%), IR69A × NRL79 (47.2%), G46A × NRL79 (24.6%), IR69A × NRL81 (23.4%), and IR69A × NRL82 (16.2%). The capture and utilization of rainwater by crops under various mulching conditions have great importance in agriculture production systems, especially in dry-prone regions. Understanding the effect of mulching on rainwater use efficiency growth and yield of a crop is very important. For this purpose, field experiments were conducted in 2017 and 2018 to evaluate the potential of ridge-furrow mulching on maize growth and development under rain-fed conditions. The field study compared four treatments, i.e., ridge-furrow without mulch (WM), black plastic mulch (BM), transparent plastic mulch (TM) and grass mulch (GM). The BM treatment consistently increased the soil moisture and temperature, resulting in earlier emergence, as well as increased plant height and plant biomass, compared to the WM treatment. Compared to WM, the two-years mean yield of maize with BM, TM and GM were recorded to be increased by 33.6%, 28.1% and 10.8%, respectively. The BM produced a maximal crop growth rate at 90 days after sowing (DAS) as specified by a greater leaf area index. Transpiration rate and leaf stomatal conductance were significantly higher with BM and TM than with WM, however, the BM treatment showed the highest net photosynthetic rate in both years. Net income for the BM treatment was the highest (USD 1226 ha−1) of all the treatments and USD 335 ha−1 greater than WM. As growth, yield and net income of maize were improved with BM, therefore this treatment was found to be the most effective for maize production in rain-fed conditions. Garlic is an important vegetable in terms of its economic value and also as a medicinal plant. In this study, chitosan (300 mM) and yeast extract (8 g/L) were used individually or in combination to improve the yields of garlic plants under drought conditions (i.e., 75% and 50% of the water they would normally receive from irrigation) for two seasons. Significant decreases in numbers of leaves per plant and plant height, plant dry weight, relative water content, and chlorophyll a and b concentrations were found in stressed garlic plants in both seasons. The greatest reductions in these characters were recorded in plants that received only 50% of the normal irrigation in both seasons. Levels of hydrogen peroxide, products of lipid peroxidation such as malondialdehyde, and superoxide, as well as percentages of electrolyte leakage, were elevated considerably and were signals of oxidative damage. The application of the yeast extract (8 g/L) or chitosan (300 mM) individually or in combination led to a remarkable increase in the most studied characters of the stressed garlic plants. The combination of yeast extract (8 g/L) plus chitosan (300 mM) led to increase plant height (44%), ascorbic acid levels (30.2%), and relative water content (36.8%), as well as the chlorophyll a (50.7%) and b concentrations (79%), regulated the proline content and levels of antioxidant enzymes in stressed garlic plants that received 75% of the normal irrigation, and this decreased the signs of oxidative stress (i.e., percentage of electrolyte leakage and levels of malondialdehyde, hydrogen peroxide, and superoxide). The technology of hybrid rice utilizing heterosis is an essential requirement for achieving food security. The current study was aimed at assessing the genetic parameters and the gene actions of 15 yield-component traits associated with heterosis, in 9 new parental lines of hybrid rice and their generated hybrids. Five cytoplasmic male sterile (CMS) lines were crossed with four restorer (R) lines using twenty generated line × tester designation hybrid combinations. The results revealed that all the traits were controlled by additive and non-additive gene actions. However, the additive variance was the main component of the total genotypic variance. Assessment of the general combining ability (GCA) detected the best combiners among the genotypes. The hybrid combinations that expressed the highest-positive specific combining ability (SCA) for grain-yield were detected. The correlation between the GCA and SCA was evaluated. The hybrid crosses with high-positive heterosis, due to having a better parent for grain yield, were detected. The principal component analysis (PCA) recorded the first four principal axis displayed Eigenvalues >1 and existing variation cumulative of 83.92% in the genotypes for yield component characteristics. Three-dimensional plots corresponding to the studied traits illustrated that the genotypes Guang8A × Giza181, Quan-9311A × Giza179, II-32A × Giza181, and II-32A × Giza179 are classified as possessing superior grain yield. (1) Background: Anaplasmosis is an infectious disease in camels caused by an obligate intracellular bacterium that is transmitted by ticks. (2) Methods: A cross-sectional study was conducted during 2020 to study the seroprevalence of Anaplasma spp. among Camelus dromedarius in three governorates in Egypt and assess the associated risk factors. Serum samples from 365 camels were examined by a competitive enzyme-linked immunosorbent assay (cELISA) test. (3) Results: Overall, the seroprevalence of anaplasmosis among camels was 18.6%. Multivariable logistic regression was performed, and it was discovered that tick infestation, application of acaricides, grooming practice and body condition were potential risk factors for Anaplasma spp. infection (odds ratio > 1) in dromedary camels. In contrast, the locality in which the camels lived and their age were not significant effects with regard to the occurrence of anaplasmosis. (4) Conclusions: The current findings suggest that improvement of protective measures to limit the effects of the identified risk factors can help to reduce the spread of anaplasmosis among camels in Egypt. Morphological and simple sequence repeat (SSR) approaches were used to determine the genetic diversity of 29 ryegrass (Lolium rigidum) genotypes belonging to eight populations collected from several regions in Saudi Arabia. In this study, 50 in Silico-developed SSR markers derived from genomic and expressed sequence tag (EST) microsatellites were examined. Analysis of variance showed highly significant differences in all studied traits. Cluster analysis based on the morphological data of the 29 Lolium genotypes and using PAST (paleontological statistics) software was performed. According to the results, clustering was based mostly on genotype location. The sensitive genotypes for herbicide were clustered in one group. In addition, using EST-SSR markers, we observed the existence of a considerable number of genetic variations among Lolium genotypes. From these markers, only 31 produced reasonable amplification products. The results showed that 23 SSR markers revealed that 74.19% were polymorphic. The number of alleles detected per primer ranged from one to five in the primer LTC SSR1. The tested primers amplified 1434 bands across eight populations, with an average of 46.26 bands per primer. The polymorphism information content (PIC) values ranged from 0.11 to 0.76 for the primers LT EST-SSR5 and LTC SSR1. The unweighted pair group method with arithmetic average (UPGMA) clustering of the 29 genotypes representing eight populations was based essentially on their locations and herbicide-tolerance levels. Most of the populations formed into four clusters, together representing genotypes. Moreover, the tolerant populations were distinguished from the sensitive ones. The relationship between the genetic diversity and geographical source of Lolium rigidum populations of Saudi Arabia was revealed through this study. The results showed that the efficiency of developed SSR markers are transferable across species. They have been helpful to assess the genetic diversity of the ryegrass population as this could be applied to differentiate between tolerant and sensitive populations of ryegrass. Simple Summary: The small ubiquitin-related modifier genes regulate the function of the cellular proteins, which are associated with cell stress-tolerance. Identification and understanding the functional localization of these genes are very important to mitigate the stresses. In this study, we identified a novel small ubiquitin-related modifier gene and studied its functional localization in the cell. This new finding will be very valuable in increasing our understanding of the mechanism of stress-tolerance. Small ubiquitin-related modifier (SUMO) regulates the cellular function of diverse proteins through post-translational modifications. The current study defined a new homolog of SUMO genes in the rice genome and named it OsSUMO7. Putative protein analysis of OsSUMO7 detected SUMOylation features, including di-glycine (GG) and consensus motifs (ΨKXE/D) for the SUMOylation site. Phylogenetic analysis demonstrated the high homology of OsSUMO7 with identified rice SUMO genes, which indicates that the OsSUMO7 gene is an evolutionarily conserved SUMO member. RT-PCR analysis revealed that OsSUMO7 was constitutively expressed in all plant organs. Bioinformatic analysis defined the physicochemical properties and structural model prediction of OsSUMO7 proteins. A red fluorescent protein (DsRed), fused with the OsSUMO7 protein, was expressed and localized mainly in the nucleus and formed nuclear subdomain structures. In the present investigation, we study the effect of Bacillus thuringiensis MH161336 (106–8 CFU/cm3), silicon (25 mL L−1), and carrot extract (75 mL L−1) as seed primers, individually or in combination, on morphological, physio-biochemical and yield components of drought-stressed pea plants (Master B) during 2019/2020 and 2020/2021 seasons. Our results indicated that drought causes a remarkable reduction in plant height, leaf area, number of leaves per plant, and number of flowers per plant in stressed pea plants during two seasons. Likewise, number of pods, pod length, seeds weight of 10 dried plants, and dry weight of 100 seeds were decreased significantly in drought-stressed pea plants. Nevertheless, seed priming with the individual treatments or in combination boosted the morphological, physio-biochemical, and yield characters of pea plants. The best results were obtained with the Bacillus thuringiensis + carrot extract treatment, which led to a remarkable increase in the number of leaves per plant, leaf area, plant height, and number of flowers per plant in stressed pea plants in both seasons. Moreover, pod length, number of seeds per pod, seeds weight of 10 dried plants, and dry weight of 100 seeds were significantly increased as well. Rice is considered a strategic crop for many countries around the world, being the main cash crop for farmers. Water shortage stress occurrence as a result of climate change is among the main threats challenging rice breeders in the last few decades. In the current study, 19 Fn-lines were developed from four populations by crossing a reverse thermo-responsive genic male sterile (rTGMS) line, M.J.5460S, with the three high-quality Egyptian commercial cultivars Giza177, Sakha105, Sakha106 and the promising line GZ7768 as male parents. These newly developed lines, along with their parents, and two water shortage stress-tolerant international genotypes (Azucena and IRAT170), were cultivated under water-shortage stress conditions and compared with their performance under well-watered conditions. Results indicated that the yielding ability of the 19 newly developed lines exceeded those for the two Egyptian parents (Giza177 and Sakha105) under well-watered conditions. The lines M.J5460S/GIZA177-3 and M.J5460S/GIZA177-12 were the best performing genotypes under water shortage stress conditions. The genetic and heritability in broad sense estimates indicated that direct selection for grain yield (GY) under water-shortage stress is highly effective in the current study. Molecular marker analysis revealed that M.J5460S/GIZA177-3 had accumulated the quantitative trait loci (QTL)s, on the chromosomes 2, 3, and 9, which contribute to GY under water-shortage stress from their high yielding tolerant ancestor, M.J5460S. It could be concluded that those lines are high yielding under both well-watered and water-stress conditions harboring several QTLs for yield enhancement under both conditions and that the markers RM555, RM14551, RM3199, RM257, RM242, and RM410 are among the markers that could be used in marker-assisted selection (MAS) breeding programs for such stress condition. Simple Summary: Among abiotic stress factors, drought is one of the most detrimental factors in arid and semiarid regions, causing a significant decrease in plant growth and yield in most species, including crops. Under drought conditions, morphological, physiological and biochemical characteristics such as plant height and enzymatic activities are negatively affected. This negative effect may be alleviated with the aid of plant growth-promoting bacteria. Application of plant growth-promoting bacteria such as Paenibacillus, Azospirillum, Rhizobium, Bacillus, Azotobacter, Klebsiella, Pseudomonas and Serratia can enhance hormonal balance, maintain nutrient status and improve plant growth characters as well as increase yield. This review discusses the pivotal role of plant growth-promoting bacteria in mitigating drought stress by improving plant growth characters and yield. Plant growth-promoting bacteria play an essential role in enhancing the physical, chemical and biological characters of soils by facilitating nutrient uptake and water flow, especially under abiotic stress conditions, which are major constrains to agricultural development and production. Drought is one of the most harmful abiotic stress and perhaps the most severe problem facing agricultural sustainability, leading to a severe shortage in crop productivity. Drought affects plant growth by causing hormonal and membrane stability perturbations, nutrient imbalance and physiological disorders. Furthermore, drought causes a remarkable decrease in leaf numbers, relative water content, sugar yield, root yield, chlorophyll a and b and ascorbic acid concentrations. However, the concentrations of total phenolic compounds, electrolyte leakage, lipid peroxidation, amounts of proline, and reactive oxygen species are considerably increased because of drought stress. This negative impact of drought can be eliminated by using plant growth-promoting bacteria (PGPB). Under drought conditions, application of PGPB can improve plant growth by adjusting hormonal balance, maintaining nutrient status and producing plant growth regulators. This role of PGPB positively affects physiological and biochemical characteristics, resulting in increased leaf numbers, sugar yield, relative water content, amounts of photosynthetic pigments and ascorbic acid. Conversely, lipid peroxidation, electrolyte leakage and amounts of proline, total phenolic compounds and reactive oxygen species are decreased under drought in the presence of PGPB. The current review gives an overview on the impact of drought on plants and the pivotal role of PGPB in mitigating the negative effects of drought by enhancing antioxidant defense systems and increasing plant growth and yield to improve sustainable agriculture. We investigated the impact of Bacillus thuringiensis as seed treatment and application with silicon on lettuce plants exposed to salinity levels (4 dS m−1 and 8 dS m−1). Results revealed that leaves number, head weight, total yield, relative water content (RWC), and chlorophyll a and b declined considerably due to two salinity levels. Oxidative stress markers, i.e., hydrogen peroxide (H2O2), superoxide (O2−), and lipid peroxidation (MDA) dramatically augmented in stressed plants. On the other hand, leaves number, total yield, RWC, and chlorophyll a, b in stressed lettuce plants were considerably enhanced because of the application of Si or B. thuringiensis. In contrast, EL%, MDA, and H2O2 were considerably reduced in treated lettuce plants with Si and B. thuringiensis. In addition, the treatment with Si and B. thuringiensis increased head weight (g) and total yield (ton hectare-1), and caused up-regulation of proline and catalase, superoxide dismutase, peroxidase, and polyphenol oxidase activity in lettuce leaves under salinity conditions. Drought stress deleteriously affects growth, development and productivity in plants. So, we examined the silicon effect (2 mmol) and proline (10 mmol) individually or the combination (Si + proline) in alleviating the harmful effect of drought on total phenolic compounds, reactive oxygen species (ROS), chlorophyll concentration and antioxidant enzymes as well as yield parameters of drought-stressed sugar beet plants during 2018/2019 and 2019/2020 seasons. Our findings indicated that the root diameter and length (cm), root and shoot fresh weights (g plant−1) as well as root and sugar yield significantly decreased in sugar beet plants under drought. Relative water content (RWC), nitrogen (N), phosphorus (P) and potassium (K) contents and chlorophyll (Chl) concentration considerably reduced in stressed sugar beet plants that compared with control in both seasons. Nonetheless, lipid peroxidation (MDA), electrolyte leakage (EL), hydrogen peroxide (H2O2) and superoxide (O2●−) considerably elevated as signals of drought. Drought-stressed sugar beet plants showed an increase in proline accumulation, total phenolic compounds and up-regulation of antioxidant enzymes catalase (CAT) and superoxide dismutase (SOD) activity to mitigate drought effects. Si and proline individually or the combination Si + proline considerably increased root and sugar yield, sucrose%, Chl concentration and RWC, MDA and EL were remarkably reduced. The treatments led to adjust proline and total phenolic compounds as well as CAT and SOD activity in stressed sugar beet plants. Hybrid rice parental lines with better combining abilities provide an efficient tool to increase rice production. In the current study, twenty hybrid combinations were generated from five aromatic cytoplasmic male sterile (CMS) lines and four restorer lines (three of them aromatic) using a line × tester mating design. The hybrids and their parental lines were evaluated under two water regimes: normal irrigation and water-stress. Ten yield-component traits were studied over a period of 2 years, and the significant differences between the parents and hybrids are reported in this investigation. Overall, all yield component traits were significantly affected by the water deficit and were governed by both additive and non-additive gene actions. More specifically, the grain yield (GY) was mainly controlled by non-additive gene action under both normal and water-stress conditions. The contribution of the additive variance (σ2 A) was more prominent in the genetic components of traits as compared to the dominance variance (σ2 D). The aromatic parental line CMS IR58025A and the restorer line PR2 were recorded as the best combiners for the GY and good combiners for many other characteristics under both growth conditions. The cross combinations Pusa12A/IR25571-31R and Pusa12A/Giza-Basmati-201 revealed significantly positive specific combining ability (SCA) effects for the GY under both normal and water-stress conditions. The inconsistent correlation between the general combining ability (GCA) and SCA manifested complex interactions among the positive and negative alleles of the genes controlling the yield traits. Generally, the findings of this investigation demonstrated the importance of the GCA and SCA for understanding the genetic components and gene actions of the yield characteristics in new aromatic hybrid rice parental lines. Concluding, the number of spikelets, and/or number of kernels were identified to be unbiased traits for assessing wheat DHLs under salinity conditions, given their contribution and direct impact on the grain yield. Moreover, the two most salt-tolerant genotypes DHL2 and DHL21 can be useful as genetic resources for future breeding programs. However, the assessment of salt tolerance potential in bread wheat doubled haploid lines (DHL) through agro-physiological traits and stress-related gene expression analysis could potentially minimize the cost of breeding programs and be a powerful way for the selection of the most salt-tolerant genotype. We used an extensive set of agro-physiologic parameters and salt-stress-related gene expressions. Multivariate analysis was used to detect phenotypic and genetic variations of wheat genotypes more closely under salinity stress, and we analyzed how these strategies effectively balance each other. Four doubled haploid lines (DHLs) and the check cultivar (Sakha93) were evaluated in two salinity levels (without and 150 mM NaCl) until harvest. The five genotypes showed reduced growth under 150 mM NaCl; however, the check cultivar (Sakha93) died at the beginning of the flowering stage. Salt stress induced reduction traits, except the canopy temperature and initial electrical conductivity, which was found in each of the five genotypes, with the greatest decline occurring in the check cultivar (Sakha-93) and the least decline in DHL2. The genotypes DHL21 and DHL5 exhibited increased expression rate of salt-stress-related genes (TaNHX1, TaHKT1, and TaCAT1) compared with DHL2 and Sakha93 under salt stress conditions. Principle component analysis detection of the first two components explains 70.78% of the overall variation of all traits (28 out of 32 traits). A multiple linear regression model and path coefficient analysis showed a coefficient of determination (R2) of 0.93. Broomrape parasitism on faba bean (Vicia faba L.) is the most destructive factor for this crop in Egypt. Pot experiments were conducted during the two successive seasons 2017/2018 and 2018/2019 to study the mitigation of broomrape stress on faba bean using a ten-fold dilution of 10% (w/v) spent mushroom substrate extract (SMSE) of Pleurotus ostreatus and the same dilution of culture filtrate of mushroom (MCF) grown in potato dextrose broth (PDB) at a rate of 48 l hectare−1 compared with the commercial herbicide Roundup (Glyphosate 48% emulsifiable concentrate) at a rate of 144 cm3 ha−1 on the two varieties (Misr3 and Sakha3) cultivated in broomrape-infested soil. The treatments include the use of mushroom products as foliar spray and/or soil amendment in addition to Roundup spraying as a recommended treatment. Using Gas Chromatography-Mass Spectrometry (GC-MS) spectroscopy, our results indicate that the major components of the two mushroom products were bioactive compounds such as polyphenol and high molecular weight aliphatic and aromatic hydrocarbons that may interfere with parasite and host metabolism. These results indicated that SMSE of P. ostreatus and MCF of the same mushroom grown in potato dextrose broth (PDB) gave the best control of broomrape, and increased plant height, root length, leaf area, chlorophyll concentration, relative water content and seed yield (g plant−1), as well as anatomical characters of leaves in the two faba bean varieties (Misr3 and Sakha3), such as upper and lower epidermis, palisade tissue, spongy tissue and vascular bundles. Additionally, electrolyte leakage was decreased in the treated plants compared to control plants and the plants treated with Roundup (glyphosate) because of the important role of SMSE and MCF in the improvement of faba bean water status. Desert plants are able to survive under harsh environmental stresses inherent to arid and semiarid regions due to their association with bacterial endophytes. However, the identity, functions, and the factors that influence the association of bacterial endophytes with desert plants are poorly known. These bacterial endophytes can be used as an untapped resource to favor plant growth and development in agro-ecosystems of arid regions. The present study is therefore focused on the isolation and identification of bacterial endophytes from two native medicinal plants (Fagonia mollis Delile and Achillea fragrantissima (Forssk) Sch. Bip.) growing spontaneously in the arid region of the South Sinai (Egypt), and characterization of their plant growth promoting (PGP) traits. Thirteen putative bacterial endophytes were isolated from the leaves of both plant species and characterized for their plant growth promoting abilities using molecular and biochemical approaches, as well as greenhouse trials. Selected endophytic bacterial strains were applied to maize plants (Zea mays L. var. Single cross Pioneer 30K08) to further evaluate their PGP abilities under greenhouse conditions. Isolated bacterial strains have variable plant growth promoting activities. Among these activities, isolated bacterial endophytes have the efficacy of phosphate solubilizing with clear zones ranging from 7.6 ± 0.3 to 9.6 ± 0.3 mm. Additionally, the obtained bacterial endophytes increased the productivity of indole acetic acid (IAA) in broth media from 10 to 60 µg·mL−1 with increasing tryptophan concentration from 1 to 5 mg·mL−1. Bacillus and Brevibacillus strains were frequently isolated from the leaves of both plant species, and had significant positive effects on plant growth and shoot phosphorus (P) and nitrogen (N) contents. Results suggest that these endophytes are good candidates as plant growth promoting inoculants to help reduce chemical input in conventional agricultural practices and increase nutrient uptake and stress resilience in plant species. Salinity stress deleteriously affects the growth and yield of many plants. Plant growth promoting rhizobacteria (PGPR) and chitosan both play an important role in combating salinity stress and improving plant growth under adverse environmental conditions. The present study aimed to evaluate the impacts of PGPR and chitosan on the growth of sweet pepper plant grown under different salinity regimes. For this purpose, two pot experiments were conducted in 2019 and 2020 to evaluate the role of PGPR (Bacillus thuringiensis MH161336 106–8 CFU/cm3) applied as seed treatment and foliar application of chitosan (30 mg dm−3) on sweet pepper plants (cv. Yolo Wonder) under two salinity concentrations (34 and 68 mM). Our findings revealed that, the chlorophyll fluorescence parameter (Fv/Fm ratio), chlorophyll a and b concentrations, relative water content (RWC), and fruit yield characters were negatively affected and significantly reduced under salinity conditions. The higher concentration was more harmful. Nevertheless, electrolyte leakage, lipid peroxidation, hydrogen peroxide (H2O2), and superoxide (O2−) significantly increased in stressed plants. However, the application of B. thuringiensis and chitosan led to improved plant growth and resulted in a significant increase in RWC, chlorophyll content, chlorophyll fluorescence parameter (Fv/Fm ratio), and fruit yield. Conversely, lipid peroxidation, electrolyte leakage, O2−, and H2O2 were significantly reduced in stressed plants. The impact of biochar and chitosan on barley plants under drought stress conditions was investigated during two field experiments. Our results confirmed that drought stress negatively affected morphological and physiological growth traits of barley plants such as plant height, number of leaves, chlorophyll concentrations, and relative water content. However, electrolyte leakage (EL%), lipid peroxidation (MDA), soluble sugars, sucrose and starch contents significantly increased as a response to drought stress. Additionally, 1000 grain weight, grains yield ha−1 and biological yield significantly decreased in stressed barley plants, also anatomical traits such as upper epidermis, lower epidermis, lamina, and mesophyll tissue thickness as well as vascular bundle diameter of flag leaves significantly decreased compared with control. The use of biochar and chitosan led to significant increases in plant height, number of leaves, and chlorophyll concentrations as well as relative water content; nevertheless these treatments led to significant decreases in electrolyte leakage (EL%) and lipid peroxidation (MDA) in the stressed plants. Moreover, anatomical and yield characters of stressed barley plants were improved with application of biochar and chitosan. Barley is a very important crop worldwide and has good impact in preserving food security. The impacts of 10 mM proline and 0.5 mM salicylic acid were evaluated on water stressed barley plants (Hordeum vulgare L. Giza126). Salicylic acid and proline treatments led to increased stem length, plant dry weights, chlorophyll concentration, relative water content, activity of antioxidant enzymes, and grain yield under drought stress. Chitin is the second most prevalent polysaccharide found in nature, following cellulose. Amino-oligosaccharides, the byproducts of chitin degradation, exhibit favorable biological properties and potential for various uses. Chitinases play a crucial function in the breakdown of chitin, and their exceptionally effective production has garnered significant interest. Here, in this study, the exochitinase PbChiA, obtained from Paenibacillus barengoltzii , was recombinantly produced and immobilized using the CotG surface protein of Bacillus subtilis WB800N. The resulting strain Bacillus subtilis WB800N pHS-CotG-Chi exhibited exceptional heat stability and efficacy across various pH levels. The chitinolytic activity of the enzyme, which had been isolated and immobilized on the spore surface, was measured to be approximately 16.06 U/mL. Including Ni 2+ , Zn +2 , and K + , and EDTA at various concentration levels in the reaction system, has significantly enhanced the activity of the immobilized enzyme. The immobilized exochitinase demonstrated a notable rate of recycling, as the recombinant spores sustained a relative enzyme activity of more than 70% after three cycles and 62.7% after four cycles. These findings established a basis for additional investigation into the role and practical use of the immobilized bacterial exochitinase in industry.

OsARF1 is the first full-length member of auxin response factor (ARF) gene family to be cloned from monocot plant. Using quantitative RT-PCR this study found that, the transcript abundance of OsARF1 was significantly higher in embryonic tissues than in vegetative tissues. To investigate the effect of OsARF1 on the phenotype of rice, a cDNA fragment of OsARF1 was inserted in inverse orientation to the 35S promoter in vector pBin438 to produce an antisense (AS) construction. The AS-OsARF1 construct was transferred into rice (Oryza sativa L. japonica) calli via Agrobacterium tumefaciens-mediated transformation. Molecular analysis of transgenic plants showed that the functional expression of OsARF1 was inhibited at mRNA level efficiently. The AS-OsARF1 plants showed extremely low growth, poor vigor, short curled leaves and tillered but were sterile. Therefore, the OsARF1 was shown to be essential for growth in vegetative organs and seed development.
