Today's adolescents represent an elective target in addressing environmental challenges. Education is a key factor in achieving a sustainable future for them. However, formal education can represent a challenge when youths are its target, as they are considered the "interactive generation". Game-based learning, and, in particular, Digital Educational Escape Rooms (DERs), have emerged as innovative methods in education, with promising applications in environmental sustainability studies. In this study, we developed 14 Digital Escape Rooms using the Social Cognitive Theory of Bandura as a theoretical framework. These were focused on environmental education and we tested them on a sample of 411 students (aged 12–18 years; 158 female, 38.4%). A one-group quasi-experimental research design was adopted, carrying out a pre-test post-test analysis. Each participant completed assessments at two time points: before engaging in the escape rooms (T0) and after (T1). The assessment tools included the Goal Assessment Scale (GAS) and the Perceived Climate Self-Efficacy Scale. Our findings revealed no significant gender differences in goal achievement. However, at T0, females exhibited higher levels of perceived climate self-efficacy in both individual and collective dimensions. Instead, the Digital Escape Rooms appeared particularly effective in increasing self-efficacy among male participants. These results suggest that Digital Escape Rooms hold potential for enhancing environmental self-efficacy, although gender differences in baseline efficacy levels warrant further exploration. Climate change (CC) has a significant impact on human health, resulting in both physical and mental illnesses. Eco-anxiety—the excessive and pervasive fear about the consequences of CC—is the most studied psychoterratic state. This study presents the validation of Italian versions of Hogg's Eco-Anxiety Scale (HEAS) and the Eco-Paralysis Scale. It also investigates the effects of worry on eco-anxiety and eco-paralysis. The study was conducted on 150 Italian individuals who responded to the two scales and to other questionnaires to make comparisons with the two above. Internal consistency and factorial structure were assessed through Cronbach's alpha, Confirmatory Factor Analysis and Exploratory Factor Analysis. A median regression was used to assess the association between the EPS and the HEAS and Climate Change Worry Scale (CCWS) and their interaction. HEAS and EPS showed good psychometric properties: HEAS resulted in good internal consistency (Cronbach's α = 0.986), and the Eco-Paralysis scale had good test-retest reliability (r = 0.988). In both cases, a one-factor structure was suggested to be retained. The interaction terms between HEAS and CCWS (β = −0.02; 95% CI: −0.03, −0.01; p < 0.001) and between HEAS and education (β = −0.05; 95% CI: −0.08, −0.02; p < 0.001) were significant. Therefore, the feeling of worry seems to act as a moderator between climate change anxiety and eco-paralysis since it may appear to influence individuals and their ability to transform anxiety into action. Education plays a role in reducing the risk of Eco-Paralysis in subjects affected by climate change anxiety. Thus, data suggest that working on reinforcing a more cognitive concern might result in more problem-solving-focused strategies to face climate change anxiety and eco-paralysis. Tomato (Solanum lycopersicum L.) is one of the most widely grown vegetables in the world and is impacted by many diseases which cause yield reduction or even crop failure. Breeding for disease resistance is thus a key objective in tomato improvement. Since disease arises from a compatible interaction between a plant and a pathogen, a mutation which alters a plant susceptibility (S) gene facilitating compatibility may induce broad-spectrum and durable plant resistance. Here, we report on a genome-wide analysis of a set of 360 tomato genotypes, with the goal of identifying defective S-gene alleles as a potential source for the breeding of resistance. A set of 125 gene homologs of 10 S-genes (PMR 4, PMR5, PMR6, MLO, BIK1, DMR1, DMR6, DND1, CPR5, and SR1) were analyzed. Their genomic sequences were examined and SNPs/indels were annotated using the SNPeff pipeline. A total of 54,000 SNPs/indels were identified, among which 1300 were estimated to have a moderate impact (non-synonymous variants), while 120 were estimated to have a high impact (e.g., missense/nonsense/frameshift variants). The latter were then analyzed for their effect on gene functionality. A total of 103 genotypes showed one high-impact mutation in at least one of the scouted genes, while in 10 genotypes, more than 4 high-impact mutations in as many genes were detected. A set of 10 SNPs were validated through Sanger sequencing. Three genotypes carrying high-impact homozygous SNPs in S-genes were infected with Oidium neolycopersici, and two highlighted a significantly reduced susceptibility to the fungus. The existing mutations fall within the scope of a history of safe use and can be useful to guide risk assessment in evaluating the effect of new genomic techniques. [ABSTRACT FROM AUTHOR]

The aim of this study was to examine the association between different coping strategies and physical and emotional consequences, depressive symptoms, state anxiety, and trait anxiety, distinguishing between victims with previous offline victimization experiences (e.g., bullying, domestic violence) and those without such experiences. A self-administered questionnaire was distributed in a snowball system to more than 700 young adults in Italy. A total of 689 individuals completed the instrument. Of these, 305 participants (44%) reported having been victims of at least one form of cyberstalking. A total of 201 participants (66% of victims) reported having experienced both cyberstalking and other forms of victimization in their lifetime, while 89 (29% of victims) reported having experienced only cyberstalking. Overall, the results of this study show that victims with previous victimization had significantly higher scores on physical, emotional, depressive, and anxiety symptoms than victims who had never been victimized. In addition, results showed that victims who have been victimized before are more likely to use all three strategies (proactive, avoidant, passive) to stop cyberstalking than victims who have never been victimized. Phytophthora infestans, the causal agent of late blight (LB) in tomato (Solanum lycopersicum L.), is a devastating disease and a serious concern for plant productivity. The presence of susceptibility (S) genes in plants facilitates pathogen proliferation; thus, disabling these genes may help provide a broad-spectrum and durable type of tolerance/resistance. Previous studies on Arabidopsis and tomato have highlighted that knock-out mutants of the PMR4 susceptibility gene are tolerant to powdery mildew. Moreover, PMR4 knock-down in potato has been shown to confer tolerance to LB. To verify the same effect in tomato in the present study, a CRISPR–Cas9 vector containing four single guide RNAs (sgRNAs: sgRNA1, sgRNA6, sgRNA7, and sgRNA8), targeting as many SlPMR4 regions, was introduced via Agrobacterium-tumefaciens-mediated transformation into two widely grown Italian tomato cultivars: 'San Marzano' (SM) and 'Oxheart' (OX). Thirty-five plants (twenty-six SM and nine OX) were selected and screened to identify the CRISPR/Cas9-induced mutations. The different sgRNAs caused mutation frequencies ranging from 22.1 to 100% and alternatively precise insertions (sgRNA6) or deletions (sgRNA7, sgRNA1, and sgRNA8). Notably, sgRNA7 induced in seven SM genotypes a −7 bp deletion in the homozygous status, whereas sgRNA8 led to the production of fifteen SM genotypes with a biallelic mutation (−7 bp and −2 bp). Selected edited lines were inoculated with P. infestans, and four of them, fully knocked out at the PMR4 locus, showed reduced disease symptoms (reduction in susceptibility from 55 to 80%) compared to control plants. The four SM lines were sequenced using Illumina whole-genome sequencing for deeper characterization without exhibiting any evidence of mutations in the candidate off-target regions. Our results showed, for the first time, a reduced susceptibility to Phytophtora infestans in pmr4 tomato mutants confirming the role of KO PMR4 in providing broad-spectrum protection against pathogens. Despite the numerous advances made in Italy over the years in the study of sexual harassment in the workplace (SHW), research has focused exclusively on victims, perpetrators, and their relationships, and not on the consequences that the experience of sexual harassment can produce in witnesses. The present study aims to address this gap by examining how the indirect experience of SHW, in conjunction with variables such as gender, age, self-efficacy, and coping strategies, affects the mental health status of witnesses of SHW. A sample of 724 employees completed a questionnaire that included a modified version of the Sexual Experience Questionnaire (SEQ), the Oldenburg Burnout Inventory (OLBI), the General Health Questionnaire (GHQ), the Satisfaction With Life Scale (SWLS), and the Emotional Self-Efficacy Scale (RESE). Of the group, 321 participants reported witnessing sexual harassment in the workplace (28.2% of women and 16.2% of men). Results show that witnesses were younger than participants who described themselves as non-witnesses. Results also show that women and men who were witnesses were more likely to suffer the emotional and psychological consequences of the experience than non-witnesses. In addition, female witnesses expressed more positive emotions than men, which enabled them to manage their anxiety and emotional states when triggered in response to sexual harassment in the workplace. Finally, a significant association was found between perceptions of mental health and age, gender, experience with SHW, and self-efficacy strategies. The findings underscore the importance of sexual harassment intervention in the workplace, women and men who witness sexual harassment suffer vicarious experiences, psychological impact, exhaustion, disengagement, and negative feelings. Robberies and other violent acts by strangers and intruders are common in banks, which can lead to post-traumatic stress disorder in employees. However, the literature indicates that more subtle and less measurable forms of violence, such as psychological violence, can also cause discomfort among employees. The aim of the present work is to investigate the prevalence of verbal aggression and its consequences among 311 Italian bank employees. A self-administered questionnaire was completed by 197 employees. The results showed a high prevalence of verbal aggression in the sample. It is important to investigate and find the causes and effects of verbal aggression in order to understand the phenomenon in this workplace. Cases of verbal aggression need to be prevented and responded to appropriately. In addition, it should be noted that the effects of verbal aggression do not only affect the victim, but also the workplace, as productivity decreases, and the feeling of a safe environment is missing. Anemone coronaria L. (2n = 2x = 16) is a perennial, allogamous, highly heterozygous plant marketed as a cut flower or in gardens. Due to its large genome size, limited efforts have been made in order to develop species-specific molecular markers. We obtained the first draft genome of the species by Illumina sequencing an androgenetic haploid plant of the commercial line "MISTRAL® Magenta". The genome assembly was obtained by applying the MEGAHIT pipeline and consisted of 2 × 106 scaffolds. The SciRoKo SSR (Simple Sequence Repeats)-search module identified 401.822 perfect and 188.987 imperfect microsatellites motifs. Following, we developed a user-friendly "Anemone coronaria Microsatellite DataBase" (AnCorDB), which incorporates the Primer3 script, making it possible to design couples of primers for downstream application of the identified SSR markers. Eight genotypes belonging to eight cultivars were used to validate 62 SSRs and a subset of markers was applied for fingerprinting each cultivar, as well as to assess their intra-cultivar variability. The newly developed microsatellite markers will find application in Breeding Rights disputes, developing genetic maps, marker assisted breeding (MAS) strategies, as well as phylogenetic studies. Workplace violence (WV) is defined as an intentional misuse of power, including threats of physical force against another person or group, which can cause physical, mental, spiritual, moral or social harm. The aim of this study was to describe the prevalence, characteristics and consequences of WV in a sample of Italian teachers. Our hypothesis was that WV impacted workplace satisfaction, self-efficacy and burnout. A self-administered questionnaire was answered by 331 teachers. A total of 192 (58%) subjects reported experiencing a physical or psychological form of WV. Overall, findings confirmed our hypothesis: teacher victims of WV showed high levels of burnout—both in terms of exhaustion (F = 3.96; p = 0.04) and disengagement (F = 5.85; p = 0.016), lower levels of workplace satisfaction (F = 13.24; p < 0.001) and regulatory emotional self-efficacy—especially for negative emotions (F = 5.45; p = 0.02) compared with teachers who have never experienced WV. This investigation suggests the importance of preventing WV and offering support to victims. Doing so will increase teachers' ability to manage and cope with violent behavior. Prevention and intervention may also decrease serious consequences in relation to victims' wellbeing, and improve the general stability of the classroom, as well as motivation and academic commitment. Simple Summary: In health care, animal-assisted intervention has been used primarily to enhance the positive effects of therapy. For example, it has been used with patients suffering from autism spectrum symptoms, medical difficulties, behavioral problems, and emotional disorders. More recently, this type of intervention has been increasingly used in the workplace to mitigate the effects of stress in employees (including healthcare workers). The aim of this systematic review was to analyze the potential benefits of animal-assisted intervention in healthcare workers. Healthcare settings have recently increased the use of companion animals in the workplace to provide emotional support to people with disabilities, but there is limited empirical research on the effects of these programs on healthcare workers. However, it is reasonable to speculate that Animal-Assisted Interventions (AAIs) may have positive effects on health care workers (HCWs) by buffering the negative effects of work-related stress and other occupational psychosocial risk factors. The aim of this review was to examine the beneficial effects of AAIs on the psychological well-being of HCWs. A systematic review was conducted in December 2021 to gain insight into the positive effects of pets on HCWs in the workplace. Searches were conducted in the following databases: Scopus, PubMed/Medline, Web of Science, and Google Scholar, including studies between 2001 and December 2021, and 12 articles were included in the review. The results indicate that implementing the AAI program in a busy clinic is feasible and that the program is accepted by medical professionals because of the immense psychological benefits it provides. In a search for natural compounds with anti-HIV-1 activity, we studied the effect of the ethanolic extract obtained from leaves, bark, and peels of Punica granatum L. for the inhibition of the HIV-1 reverse transcriptase (RT)-associated ribonuclease H (RNase H) and integrase (IN) LEDGF-dependent activities. The chemical analyses led to the detection of compounds belonging mainly to the phenolic and flavonoid chemical classes. Ellagic acid, flavones, and triterpenoid molecules were identified in leaves. The bark and peels were characterized by the presence of hydrolyzable tannins, such as punicalins and punicalagins, together with ellagic acid. Among the isolated compounds, the hydrolyzable tannins and ellagic acid showed a very high inhibition (IC50 values ranging from 0.12 to 1.4 µM and 0.065 to 0.09 µM of the RNase H and IN activities, respectively). Of the flavonoids, luteolin and apigenin were found to be able to inhibit RNase H and IN functions (IC50 values in the 3.7–22 μM range), whereas luteolin 7-O-glucoside showed selective activity for HIV-1 IN. In contrast, betulinic acid, ursolic acid, and oleanolic acid were selective for the HIV-1 RNase H activity. The strategies that non-profit organizations choose for volunteer engagement and financial sustainability are of the utmost importance for successful work. The main purpose of this study was to investigate feasible strategies for volunteer engagement and financial sustainability. Interviews were conducted to obtain data on the experience of volunteer managers in dealing with volunteers and financial matters together. Qualitative data analysis methods were used to code and analyze the data. Significant themes emerged from the data gathered through interviews that highlighted the strategies for volunteer engagement and financial sustainability chosen by the managers of non-profit organizations. From the manager's perspective, the following strategies were considered important for engaging volunteers at non-profit organizations (NPOs): 'building the skill sets of volunteers', 'fulfilling ulterior motives', and 'administering a culture of administrative support'. Moreover, 'local fundraising preference', 'transparency', and 'building trust' were regarded as successful strategies for maintaining financial sustainability. The findings of this study showed that, to function smoothly, non-profit organizations need to follow certain strategies to be cautious about volunteers as well as finance. The findings provide fruitful implications for practitioners and policy makers, and these are discussed in the paper. Soybean (Glycine max Merr.) is a worldwide important legume crop, whose growth and yield are negatively affected by heat stress at germination time. Here, we tested the role of a biostimulant based on lignin derivatives, plant-derived amino acids, and molybdenum in enhancing soybean heat stress tolerance when applied on seeds. After treatment with the biostimulant at 35 °C, the seed biometric parameters were positively influenced after 24 h, meanwhile, germination percentage was increased after 72 h (+10%). RNA-Seq analyses revealed a modulation of 879 genes (51 upregulated and 828 downregulated) in biostimulant-treated seeds as compared with the control, at 24 h after incubation at 35 °C. Surprisingly, more than 33% of upregulated genes encoded for ribosomal RNA (rRNA) methyltransferases and proteins involved in the ribosome assembly, acting in a specific protein network. Conversely, the downregulated genes were involved in stress response, hormone signaling, and primary metabolism. Finally, from a biochemical point of view, the dramatic H2O2 reduction 40%) correlated to a strong increase in non-protein thiols (+150%), suggested a lower oxidative stress level in biostimulant-treated seeds, at 24 h after incubation at 35 °C. Aims: In the last decade, the use of smartphones, computers and devices has progressively increased, and prolonged use of technology and the internet has generated new arenas (and tools) for victimization. The first aim of this study was to analyze the use of coping strategies in young adult self-declared victims of cyberstalking. The coping strategies were categorized as proactive behavior, avoidance tactics and passivity. To better understand these strategies, they were analyzed in light of the experience of victimization in terms of incurred misconduct. The second aim was to analyze the coping strategies and the consequences (in terms of depression and anxiety) that occurred in victims; a comparison was made between males and females. Methods: A self-administered questionnaire was distributed to over 433 young adults living in Italy. The questionnaires were filled out by 398 (92%) subjects, 41% males and 59% females. Their ages ranged from 18 to 30 years (M = 23.5, SD = 2.76). Respondents took part on a voluntary basis and did not receive any compensation (or extra credit) for their participation. Results: Findings from this investigation confirmed that among victims, females were more prone than males to experience cyberstalking (respectively, 65% and 35%), with females experiencing a higher percentage of more than one form of cyberstalking behavior than males. Young adult male victims used the internet principally for online gaming, and for this activity, they experienced more cyberstalking behavior than females. In most cases, the perpetrator was a male, and the victim–cyberstalker relationship was a friendship or an acquaintance. For the coping strategies adopted, the findings indicated that the victims were more prone to use avoidance tactics than proactivity behavior and passivity strategies. Young adults involved in this investigation mainly used avoidance tactics to cope with the stressful situation, which implies that they preferred to decrease the use of the internet or stop online contact than collect evidence and try to contact and reason with the cyberstalker or increase the misuse of alcohol of psychotropic substances. Moreover, females were less prone to use proactive behavior than expected. Our findings suggested that males were more prone than females to adopt passivity strategies, while females were more prone to adopt avoidance tactics. Moreover, the data showed that proactivity behavior was adopted more in the case of online contacts and online identity fraud, while passivity strategies were adopted in the case of online threats. Conclusion: Findings from this investigation show the importance of improving the knowledge about the coping strategies that could be suggested to victims and the impact on their psychological health. Castanea sativa is an important multipurpose species in Europe for nut and timber production as well as for its role in the landscape and in the forest ecosystem. This species has low tolerance to chestnut gall wasp (Dryocosmus kuriphilus Yasumatsu), which is a pest that was accidentally introduced into Europe in early 2000 and devastated forest and orchard trees. Resistance to the gall wasp was found in the hybrid cultivar 'Bouche de Bétizac' (C. sativa × C. crenata) and studied by developing genetic linkage maps using a population derived from a cross between 'Bouche de Bétizac' and the susceptible cultivar 'Madonna' (C. sativa). The high-density genetic maps were constructed using double-digest restriction site-associated DNA-seq and simple sequence repeat markers. The map of 'Bouche de Bétizac' consisted of 1459 loci and spanned 809.6 cM; the map of 'Madonna' consisted of 1089 loci and spanned 753.3 cM. In both maps, 12 linkage groups were identified. A single major QTL was recognized on the 'Bouche de Bétizac' map, explaining up to 67–69% of the phenotypic variance of the resistance trait (Rdk1). The Rdk1 quantitative trait loci (QTL) region included 11 scaffolds and two candidate genes putatively involved in the resistance response were identified. Green pruning residues (GPRs) and leaves from 16 red and white Vitis vinifera L. cultivars from Piedmont (Italy) were studied. The investigated samples were extracted by ultrasound-assisted extraction optimized by an experimental design, and quali- and quantitatively analyzed by HPLC-PDA-MS/MS. GPRs and leaves show a similar polyphenolic pattern, with quercetin 3-O-glucuronide, caftaric acid, and quercetin 3-O-glucoside as the main components, although in variable proportions. The HPLC results were related to the antioxidant activity, measured as total phenolic content and through DPPH and ABTS assays with similar results. Colorimetric in vitro assays, offline combined with HPLC-PDA analysis, determine which compounds contribute to the antioxidant activity in terms of radical scavenging abilities. Valorization of GPRs is a potential source of natural compounds that could be of interest in the health field, increasing their economic value together with a positive effect on the environment. Aims: The aim of this study was to compare victims of one type of cyberstalking (OneType) with victims of more than one type of cyberstalking (MoreType) regarding (1) the impact of cyberstalking and (2) attitudes related to telling someone about the experience of cyberstalking and the coping strategies used by victims. Methods: A self-administered questionnaire was distributed to over 250 students at the University of Torino. Results: About half of the participants experienced at least one incident of cyberstalking. Among them, more than half experienced more than one type of cyberstalking. Victims suffered from depression more than those who had never experienced cyberstalking. No statistically significant difference emerged for anxiety. The coping strategies used by MoreType were more varied than those used by OneType victims of cyberstalking. Moreover, MoreType victims told someone about their victimization more than OneType victims. Conclusion: The work presented suggests implications for health care professionals, police officers, and government. For example, our suggestion is to pay attention to cyberstalking victims and provide flyers in schools, universities, and cafeterias that explain the risk of certain online behaviors and their consequences in physical and emotional spheres. Globe artichoke represents one of the main horticultural species of the Mediterranean basin, and 'Spinoso sardo' is the most widespread and economically relevant varietal type in Sardinia, Italy. In the last decades, in vitro culture of meristematic apices has increased the frequency of aberrant plants in open-field production. These off-type phenotypes showed highly pinnate-parted leaves and late inflorescence budding, and emerged from some branches of the true-to-type 'Spinoso sardo' plants. This phenomenon cannot be foreseen and is reversible through generations, suggesting the occurrence of epigenetic alterations. Here, we report an exploratory study on DNA methylation patterns in off-type/true-to-type globe artichoke plants, using a modified EpiRADseq technology, which allowed the identification of 2897 differentially methylated loci (DML): 1998 in CG, 458 in CHH, and 441 in CHG methylation contexts of which 720, 88, and 152, respectively, were in coding regions. Most of them appeared involved in primary metabolic processes, mostly linked to photosynthesis, regulation of flower development, and regulation of reproductive processes, coherently with the observed phenotype. Differences in the methylation status of some candidate genes were integrated with transcriptional analysis to test whether these two regulation levels might interplay in the emergence and spread of the 'Spinoso sardo' non-conventional phenotype. Cyberstalking has been defined as the use of electronic communication devices (including the Internet and email) to stalk another person. The aim of this study was to compare the effects of cyberstalking between victims of cyberstalking and victims of cyberstalking and previous offline victimization in their lifetimes. Our hypothesis was that cyberstalking had an impact on victims' wellbeing and contributed to increases in physical and emotional symptoms, anxiety and depression, and that those symptoms increased in victims who had suffered previous offline victimization. In an effort to investigate the effects of cyberstalking, a questionnaire was self-administered to 229 Italian students. A total of 107 participants (46.7%) indicated that they had been victims of cyberstalking. Seventy-two of them (67.3%) were victims of both cyberstalking and other forms of offline victimization in their lifetimes. Overall, our findings showed that the prevalence of cyberstalking in our sample was higher than in previous investigations. With regard to consequences, victims indicated higher scores for depression and anxiety than non-victims. In particular, victims of cyberstalking and previous offline victimization in their lifetimes experienced more depression and symptoms of trait anxiety than victims of cyberstalking only and non-victims.
