Magnesium-based alloy WE43 is a state-of-the-art bioresorbable metallic implant material. There is a need for implants with both complex geometries to match the mechanical properties of bone and refined microstructure for controlled resorption. Additive manufacturing (AM) using laser powder bed fusion (LPBF) presents a viable fabrication method for implant applications, as it offers near-net-shape geometrical control, allows for geometry customization based on an individual patient, and fast cooling rates to achieve a refined microstructure. In this study, the laser–alloy interaction is investigated over a range of LPBF-relevant processing conditions to reveal melt-pool dynamics, pore formation, and the microstructure of laser-melted WE43. In situ X-ray imaging reveals distinct laser-induced vapor depression morphology regimes, with minimal pore formation at laser-scan speeds greater than 500 mm/s. Optical and electron microscopy of cross-sectioned laser tracks reveal three distinct microstructural regimes that can be controlled by adjusting laser-scan parameters: columnar, dendritic, and banded microstructures. These regimes are consistent with those predicted by the analytic solidification theory for conduction-mode welding, but not for keyhole-mode tracks. The results provide insight into the fundamental laser–material interactions of the WE43 alloy under AM-processing conditions and are critical for the successful implementation of LPBF-produced WE43 parts in biomedical applications. The purpose of this study was to investigate the role of sphingosine kinase 1 (SphK1), which generates sphingosine-1-phosphate (S1P), in corneal neovascularization (NV). Wild-type (WT) and Sphk1 knockout (Sphk1−/−) mice received corneal alkali-burn treatment to induce corneal NV by placing a 2 mm round piece of Whatman No. 1 filter paper soaked in 1N NaOH on the center of the cornea for 20 s. Corneal sphingolipid species were extracted and identified using liquid chromatography/mass spectrometry (LC/MS). The total number of tip cells and those positive for ethynyl deoxy uridine (EdU) were quantified. Immunocytochemistry was done to examine whether pericytes were present on newly forming blood vessels. Cytokine signaling and angiogenic markers were compared between the two groups using multiplex assays. Data were analyzed using appropriate statistical tests. Here, we show that ablation of SphK1 can significantly reduce NV invasion in the cornea following injury. Corneal sphingolipid analysis showed that total levels of ceramides, monohexosyl ceramides (HexCer), and sphingomyelin were significantly elevated in Sphk−/− corneas compared to WT corneas, with a comparable level of sphingosine among the two genotypes. The numbers of total and proliferating endothelial tip cells were also lower in the Sphk1−/− corneas following injury. This study underscores the role of S1P in post-injury corneal NV and raises further questions about the roles played by ceramide, HexCer, and sphingomyelin in regulating corneal NV. Earthquakes threaten humanity globally in complex ways that mainly include various socioeconomic consequences of life and property losses. Resilience against seismic risks is of high importance in the modern world and needs to be sustainable. Sustainable earthquake resilience (SER) from the perspective of structural engineering means equipping the built environment with appropriate aseismic systems. Shape memory alloys (SMAs) are a class of advanced materials well suited for fulfilling the SER demand of the built environment. This article explores how this capability can be realized by the innovative SMA-based superelasticity-assisted slider (SSS), recently proposed for next-generation seismic protection of structures. The versatility of SSS is first discussed as a critical advantage for an effective SER. Alternative configurations and implementation styles of the system are presented, and other advantageous features of this high-tech isolation system (IS) are studied. Results of shaking table experiments, focused on investigating the expected usefulness of SSS for seismic protection in hospitals and conducted at the structural earthquake engineering laboratory of the University of Bonab, are then reported. SSS is compared with currently used ISs, and it is shown that SSS provides the required SER for the built environments and outperforms other ISs by benefitting from the pioneered utilization of SMAs in a novel approach. Electrical energy plays a key role in the development of the social as well as the economic front. The power sector has historically been owned and operated by state agencies due to its tremendous importance. It has been restructured over time, and the power market is being deregulated. In terms of consumer prices, efficiency, and environmental implications, both regulated and deregulated electricity markets offer advantages and disadvantages. Policy-based techniques are typically used in regulated markets to address the costs of fossil-fuel resources and boost the viability of renewable energy sources. Renewables can be integrated into deregulated markets through a combination of regulatory and market-based measures to extend the system's economic stability which has been deployed in this paper. As the need for energy has expanded dramatically over the last few decades, particularly in developing countries, the amount of greenhouse gas emissions has climbed rapidly, as have fuel prices, which are the key driving forces behind initiatives to use renewable energy sources more effectively. Despite the apparent benefits of renewable energy, it has significant downsides, such as generation of optimization methods applied to renewable consistency, because most renewable energy supplies are climate-dependent, necessitating complicated design, planning, and control optimization methods. There have been numerous optimization strategies applied to the renewable integrated deregulated electricity system. With the increased use of renewable energy, energy storage technology has grown in importance, as these devices can capture electricity generated by renewables during off-peak demand hours and put it back into the grid during peak demand periods. Using stored renewable energy instead of adding generation based on fossil fuel can help to minimize greenhouse gas emissions. There is an interest in better utilizing available power system capacity by implementing FACTS to maximize the social benefit in a deregulated system. As a result, effective FACTS device placement provides novel control capabilities in both steady-state power flow regulation and dynamic stability control. This study reviews several aspects of renewable integrated deregulated power systems and provides a clear picture of the most recent research developments on this subject. The Western corn rootworm (WCR; Diabrotica virgifera virgifera) is an economically important belowground pest of maize. Belowground feeding by WCR is damaging because it weakens the roots system, diminishes nutrient uptake, and creates entry points for fungal and bacterial pathogens and increases lodging, all of which can significantly suppress maize yields. Previously, it was demonstrated that belowground herbivory can trigger plant defense responses in the roots and the shoots, thereby impacting intraplant communication. Although several aspects of maize-WCR interactions have been reported, co-transcriptomic remodeling in the plant and insect are yet to be explored. We used a maize genotype, Mp708, that is resistant to a large guild of herbivore pests to study the underlying plant defense signaling network between below and aboveground tissues. We also evaluated WCR compensatory transcriptome responses. Using RNA-seq, we profiled the transcriptome of roots and leaves that interacted with WCR infestation up to 5 days post infestation (dpi). Our results suggest that Mp708 shoots and roots had elevated constitutive and WCR-feeding induced expression of genes related to jasmonic acid and ethylene pathways, respectively, before and after WCR feeding for 1 and 5 days. Similarly, extended feeding by WCR for 5 days in Mp708 roots suppressed many genes involved in the benzoxazinoid pathway, which is a major group of indole-derived secondary metabolites that provides resistance to several insect pests in maize. Furthermore, extended feeding by WCR on Mp708 roots revealed several genes that were downregulated in WCR, which include genes related to proteolysis, neuropeptide signaling pathway, defense response, drug catabolic process, and hormone metabolic process. Green mold (caused by Trichoderma harzianum) is a destructive disease in mushrooms which limits commercial production. The present investigation was carried out to verify the in vitro and in vivo effect of locally available botanicals and bacterial biocontrol agents against this disease. The in vitro evaluation of ethanol extract of botanicals against mycelial growth of T. harzianum at 1, 2, and 3% concentrations showed that Juglans regia and Allium sativum exhibited maximum mycelial growth inhibition of 84.9 and 79.8%, respectively. When the same botanicals were tested against the mycelial growth of A. bisporus, it was observed that J. regia, Curcuma longa, and Azadirachta mellea were least inhibitory (4.66–7.4%). From the in vivo evaluation of plant botanicals at 2% concentration, J. regia and C. longa had the highest average weight (11.8–11.9 g) of a single fruit body and a combined button yield of 11.3–11.9 kg/quintal compost. Among the bacterial bioagents evaluated in vitro, Pseudomonas flourescens, Azotobacter sp., and Bacillus subtilis displayed stimulatory effects of varying degrees on the mycelial growth of A. bisporus but exhibited antagonistic effects on T. harzianum. B. subtilis-38, and P. flourescens-104. Azotobacter-108 caused the highest mycelial growth inhibition of 97.6, 97.4, and 90.3% of T. harzianum, respectively. The current study reveals that the integration of botanical and bacterial antagonists in pathogen-infested white button mushroom casing reduces green mold infection with corresponding yield gains. Various solitary wave excitations are found for a Bose-Einstein condensate in presence of two hybrid potentials in the form of triple mixtures of optical lattices. One of these potentials comprises of a combination of two important lattice profiles, such as frustrated optical lattice and double-well super-lattice, within one. Another represents a composite lattice combination, resulting in a wider and deeper frustrated optical lattice. The dynamical equation for such a system is solved by the exact analytical method to obtain a bright solitary wave, periodic wave and cnoidal wave excitations. We also report Anderson localization, bifurcation of condensate at the center and a competition between two different types of localizations upon trap engineering. Dynamical and structural stability analyses are also carried out, which reveal the obtained solutions as extremely stable for structural noise incorporation and sufficiently stable for dynamical stability. Proteins are vital for the significant cellular activities of living organisms. However, not all of them are essential. Identifying essential proteins through different biological experiments is relatively more laborious and time-consuming than the computational approaches used in recent times. However, practical implementation of conventional scientific methods sometimes becomes challenging due to poor performance impact in specific scenarios. Thus, more developed and efficient computational prediction models are required for essential protein identification. An effective methodology is proposed in this research, capable of predicting essential proteins in a refined yeast protein–protein interaction network (PPIN). The rule-based refinement is done using protein complex and local interaction density information derived from the neighborhood properties of proteins in the network. Identification and pruning of non-essential proteins are equally crucial here. In the initial phase, careful assessment is performed by applying node and edge weights to identify and discard the non-essential proteins from the interaction network. Three cut-off levels are considered for each node and edge weight for pruning the non-essential proteins. Once the PPIN has been filtered out, the second phase starts with two centralities-based approaches: (1) local interaction density (LID) and (2) local interaction density with protein complex (LIDC), which are successively implemented to identify the essential proteins in the yeast PPIN. Key-based circuit obfuscation or logic-locking is a technique that can be used to hide the full design of an integrated circuit from an untrusted foundry or end-user. The technique is based on creating ambiguity in the original circuit by inserting "key" input bits into the circuit such that the circuit is unintelligible absent a correct secret key. Clock signals have traditionally been avoided in locking in order to not corrupt the timing behavior of the locked circuit. In this paper, we explore the case where the clock signal itself may be obfuscated by ambiguating its frequency or pattern. Along with discussing formal notions of security in this context, we present practical ways to deobfuscate such designs using techniques from multi-rate model-checking. We present experimental data on deobfuscation runtime on a set of sequential benchmark circuits. Our results show that naive random clock obfuscation may not provide more security per overhead than traditional random keyed-gate insertion. We discuss how clock obfuscation may be a more attractive choice for special circuit designs that are inherently multi-clock/asynchronous. [ABSTRACT FROM AUTHOR]

Trees greater than 150 years old growing in the current treelines were most likely isolated tree outposts above previous treelines of the Little Ice Age (LIA). An intuitive question is, how did these isolated trees grow at such a high elevation in the cold environment? Here, we tackle this question using tree-ring width data of the Northern Hemisphere's highest treelines at 4900 m a.s.l. (Basu) and 4680 m a.s.l. (Langkazi) on the Tibetan Plateau. The results showed that an age-related exponential growth trend did not exist in most of the ring-width sequences of the sampled trees. The values of ring widths in the isolated trees had a similar pattern of probability distribution during and after the LIA. The coefficients of variation in ring widths of the isolated trees were significantly greater than those of the non-isolated trees in their common growth period. Synchronicity of annual change in radial growth among trees varied in time. These results indicated that the isolated trees in the LIA developed an adaptive ability to slow down radial growth rate and modulate growth synchronicity among individuals in cold stressful environments. Several studies have identified improvements in the risks of cardiovascular disease in adults following a Mediterranean dietary pattern. However, data are scarce on its association with coronary artery calcification (CAC) and pericardial adiposity (PAT) in US adults with and without diabetes. To address this gap, we conducted a case-control study using baseline data from the Coronary Artery Calcification in Type 1 Diabetes (CACTI) study [n = 1255; Type 1 Diabetes (T1D): n = 563; non-Diabetes Mellitus (non-DM): n = 692]. Participants completed a validated food frequency questionnaire, fasting (12 h overnight fast) biochemical analyses, and a physical examination including anthropometric measures. CAC and PAT were measured using electron beam-computed tomography. Logistic regression models were used to examine the associations of the Mediterranean-Style Dietary Pattern Score (MSDPS) with CAC (presence or absence), and linear regression models were applied to PAT analyses. In all of the adjusted models, no significant associations with CAC were found. For PAT, an increasing MSDPS was consistently associated with its lower volume in models adjusted for age, sex, diabetes status, total calories, and body mass index (all p < 0.05). The association between MSDPS and PAT was attenuated after adjusting for serum lipids and physical activity. In conclusion, the baseline data from the CACTI study show that a greater adherence to MSDPS is associated with a lower PAT volume and provide evidence that the Mediterranean dietary pattern is associated with lower cardiovascular risk markers. Annually, more than a million individuals are diagnosed with gastrointestinal (GI) cancers worldwide. With the advancements in radio- and chemotherapy and surgery, the survival rates for GI cancer patients have improved in recent years. However, the prognosis for advanced-stage GI cancers remains poor. Site-specific GI cancers share a few common risk factors; however, they are largely distinct in their etiologies and descriptive epidemiologic profiles. A large number of mutations or copy number changes associated with carcinogenesis are commonly found in noncoding DNA regions, which transcribe several noncoding RNAs (ncRNAs) that are implicated to regulate cancer initiation, metastasis, and drug resistance. In this review, we summarize the regulatory functions of ncRNAs in GI cancer development, progression, chemoresistance, and health disparities. We also highlight the potential roles of ncRNAs as therapeutic targets and biomarkers, mainly focusing on their ethnicity-/race-specific prognostic value, and discuss the prospects of genome-wide association studies (GWAS) to investigate the contribution of ncRNAs in GI tumorigenesis. Glial cell transplantation using olfactory ensheathing cells (OECs) holds a promising approach for treating spinal cord injury (SCI). However, integration of OECs into the hostile acute secondary injury site requires interaction and response to macrophages. Immunomodulation of macrophages to reduce their impact on OECs may improve the functionality of OECs. Vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF), known for their immunomodulatory and neuroprotective functions, have provided improved outcomes in SCI animal models. Thus, VEGF and PDGF modulation of the SCI microenvironment may be beneficial for OEC transplantation. In this in vitro study, the effect of VEGF and PDGF on macrophages in an inflammatory condition was tested. Combined VEGF + PDGF reduced translocation nuclear factor kappa B p65 in macrophages without altering pro-inflammatory cytokines. Further, the ability of OECs to phagocytose myelin debris was assessed using macrophage-conditioned medium. Conditioned medium from macrophages incubated with PDGF and combined VEGF + PDGF in inflammatory conditions promoted phagocytosis by OECs. The growth factor treated conditioned media also modulated the expression of genes associated with nerve repair and myelin expression in OECs. Enterococcus faecium has become an important drug-resistant nosocomial pathogen because of widespread antibiotic abuse. We developed short and chemically simple antimicrobial peptides (AMPs) with a selective amino acid composition, fixed charge, and hydrophobicity ratio based on the core antimicrobial motif of bovine lactoferrin (LfcinB6). Among these peptides, 5L and 6L (both 12 residues long) demonstrated a narrow spectrum and high antibacterial activity against drug-resistant E. faecium isolates with a minimal inhibitory concentration (MIC) that ranged from 4–16 µg/mL. At 32 µg/mL, peptides 5L and 6L inhibited E. faecium strain C68 biofilm formation by 90% and disrupted established biofilms by 75%. At 40 µg/mL, 5L reduced 1 × 107E. faecium persister cells by 3 logs within 120 min of exposure, whereas 6L eliminated all persister cells within 60 min. At 0.5× MIC, 5L and 6L significantly downregulated the expression of a crucial biofilm gene ace by 8 folds (p = 0.02) and 4 folds (p = 0.01), respectively. At 32 µg/mL, peptides 5L and 6L both depolarized the E. faecium membrane, increased fluidity, and eventually ruptured the membrane. Physiologically, 5L (at 8 µg/mL) altered the tricarboxylic acid cycle, glutathione, and purine metabolism. Interestingly, in an ex vivo model of porcine skin infection, compared to no treatment, 5L (at 10× MIC) effectively eliminated all 1 × 106 exponential (p = 0.0045) and persister E. faecium cells (p = 0.0002). In conclusion, the study outlines a roadmap for developing narrow-spectrum selective AMPs and presents peptide 5L as a potential therapeutic candidate to be explored against E. faecium. Saskatchewan Torus-Modified (STOR-M) is a small tokamak, well known for various fusion-related basic experimental studies such as edge turbulent heating, different instabilities, Alternating Current (AC) tokamak operation, Ohmic H-mode triggering by the electrode biasing, fueling and momentum injection by Compact Torus (CT) injection, and the effects of Resonance Magnetic Perturbations (RMPs), among others. Some of those experiments require real-time visualization of magnetic surface reconstructions either through EFIT or quick analyses and visualization of experimental data during experiments. Recently, experimental studies of Geodesic Acoustic Mode (GAM) and zonal flows were performed in the STOR-M tokamak. The GAM experiments strongly require the collection of fluctuation data from different Langmuir probes installed at different poloidal locations, but on the same magnetic surfaces. This is requires the adjustment of radial locations between discharges. It is therefore important to analyze and visualize the features of all probe data quickly during discharges. Because the non-chronic wounds are just about healed at 20 days, we compared the log fold change of the genes in these modules at full chronicity (wounds are fully chronic at 20 days post-wounding) with those at early chronicity to identify differences in wound progression to chronicity (Tables S6-S8). Results For this study, RNASeq was performed during early chronicity (first 10 days post-wounding) and at full chronicity (20 days post-wounding) to provide an overview of the progression of the wound to chronicity, so that we can compare progression to healing in acute wounds to failure to healing in chronic wounds. Marine picocyanobacteria belonging to the genus Synechococcus are one of the most abundant photosynthetic organisms on Earth. They are often exposed to large fluctuations in temperature and CO2 concentrations in the ocean, which are expected to further change in the coming decades due to ocean acidification and warming resulting from rising atmospheric CO2 levels. To decipher the effect of changing temperature and CO2 levels on Synechococcus, six Synechococcus strains previously isolated from various coastal and open ocean sites were exposed to a matrix of three different temperatures (22 °C, 24 °C and 26 °C) and CO2 levels (400 ppm, 600 ppm and 800 ppm). Thereafter, the specific growth rates, photophysiological parameters (σPSII and Fv/Fm), C/N (mol/mol) ratios and the nitrogen stable isotopic composition (δ15N (‰)) of the strains were measured. Temperature was found to be a stronger driver of the changes in specific growth rates and photophysiology in the Synechococcus strains. Carbon-concentrating mechanisms (CCM) operational in these strains that shield the photosynthetic machinery from directly sensing ambient changes in CO2 possibly played a major role in causing minimal changes in the specific growth rates under the varying CO2 levels. The efficient use of renewable energy sources requires development of new electrocatalytic materials for electrochemical energy storage systems, particularly fuel cells. To increase durability of high temperature polymer electrolyte fuel cell (HT-PEMFC), Pt/carbon black based catalysts should be replaced by more durable ones, for example Pt/carbon nanofibers (CNF). Here, we report for the first time the quantitative ascorbic acid assisted deposition of Pt onto electrospun polyacrylonitrile-based CNF composite materials. The effect of their subsequent post-treatment at various temperatures (250 and 500 °C) and media (vacuum or argon-hydrogen mixture) on the Pt/C catalyst morphology is investigated. Limbus-derived stromal/mesenchymal stem cells (LMSCs) are vital for corneal homeostasis and wound healing. However, despite multiple pre-clinical and clinical studies reporting the potency of LMSCs in avoiding inflammation and scarring during corneal wound healing, the molecular basis for the ability of LMSCs remains unknown. This study aimed to uncover the factors and pathways involved in LMSC-mediated corneal wound healing by employing RNA-Sequencing (RNA-Seq) in human LMSCs for the first time. We characterized the cultured LMSCs at the stages of initiation (LMSC−P0) and pure population (LMSC−P3) and subjected them to RNA-Seq to identify the differentially expressed genes (DEGs) in comparison to native limbus and cornea, and scleral tissues. Of the 28,000 genes detected, 7800 DEGs were subjected to pathway-specific enrichment Gene Ontology (GO) analysis. Since 2021, the National Evaluation exam in Romania (the exam aimed to assess 14- to 15-year-old students' knowledge at the end of lower secondary education and just before high school) has presented a novel examination structure that resembles PISA tests. The current investigation analyses the 2021 National Evaluation exam results compared to the results obtained in the previous two years (2019–2020) as an evaluation of upper education institutions' effectiveness in Romania. The results put forward the same conclusions as proposed by extant literature on Bucharest high schools. Even though the educational institutions show apparent progress and great adaptability to change, a more in-depth analysis reveals great inequality between educational institutions. As in the case of Bucharest, nationally there are only a small number of top-performing high schools in Romania, with the majority of high schools ranking in the lowest category as conceptualised in the study. The current investigation puts together a novel methodology for classification based on the main instruments proposed in literature: a letter grade classification and Turner's f-index. The results and the methodological proposal are especially relevant considering the latest PISA (2018) conclusions on Romania characterising the national educational system as underperforming. The present systematic research on cultural, morphological, and pathogenic variability was carried out on eighty isolates of Sclerotinia sclerotiorum collected from major common bean production belts of North Kashmir. The isolates were found to vary in both cultural and morphological characteristics such as colony color and type, colony diameter, number of days for sclerotia initiation, sclerotia number per plate, sclerotial weight, and size. The colony color ranged between white and off-white with the majority. The colony was of three types, in majority smooth, some fluffy, and a few fluffy-at-center-only. Colony diameter ranged between 15.33 mm and 29 mm after 24 h of incubation. The isolates took 4 to 7 days for initiation of sclerotia and varied in size, weight, and number per plate ranging between 14 and 51.3. The sclerotial arrangement pattern on plates was peripheral, sub peripheral, peripheral, and subperipheral, arranged at the rim and scattered. A total of 22 Mycelial compatibility groups (MCGs) were formed with seven groups constituted by a single isolate. The isolates within MCGs were mostly at par with each other. The six isolates representing six MCGs showed variability in pathogenicity with isolate G04 as the most and B01 as the least virulent. The colony diameter and disease scores were positively correlated. Sclerotia were observed to germinate both myceliogenically and carpogenically under natural temperate conditions of Kashmir. Coronavirus disease 2019 (COVID-19) is caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The first COVID-19 case was reported in Wuhan, China, in December 2019. In March 2020, the World Health Organization (WHO) declared COVID-19 a global pandemic. The first COVID-19 case in Nepal was reported in January 2020 in a Nepalese man who had returned from Wuhan to Nepal. This study aims to evaluate the government of Nepal's (GoN) response to the COVID-19 pandemic and explore ways to prevent COVID-19 and other pandemic diseases in the future. As of May 2022, a total of 979,140 cases and 11,951 deaths associated with COVID-19 have been reported in Nepal. To prevent the spread of the virus, the GoN initiated various preventive and control measures, including lockdown strategies. The effects of COVID-19 are expected to persist for many years; the best strategies a resource-limited country such as Nepal can implement to control pandemic diseases such as COVID-19 in the pre-vaccine stage are to increase testing, tracing, and isolation capacity. A DNA Holliday junction (HJ) has been used as a versatile scaffold to create a variety of covalently templated molecular dye aggregates exhibiting strong excitonic coupling. In these dye-DNA constructs, one way to attach dyes to DNA is to tether them via single long linkers to thymine modifiers incorporated in the core of the HJ. Here, using photoinduced [2 + 2] cycloaddition (photocrosslinking) between thymines, we investigated the relative positions of squaraine-labeled thymine modifiers in the core of the HJ, and whether the proximity of thymine modifiers correlated with the excitonic coupling strength in squaraine dimers. Photocrosslinking between squaraine-labeled thymine modifiers was carried out in two distinct types of configurations: adjacent dimer and transverse dimer. The outcomes of the reactions in terms of relative photocrosslinking yields were evaluated by denaturing polyacrylamide electrophoresis. We found that for photocrosslinking to occur at a high yield, a synergetic combination of three parameters was necessary: adjacent dimer configuration, strong attractive dye–dye interactions that led to excitonic coupling, and an A-T neighboring base pair. The insight into the proximity of dye-labeled thymines in adjacent and transverse configurations correlated with the strength of excitonic coupling in the corresponding dimers. To demonstrate a utility of photocrosslinking, we created a squaraine tetramer templated by a doubly crosslinked HJ with increased thermal stability. These findings provide guidance for the design of HJ-templated dye aggregates exhibiting strong excitonic coupling for exciton-based applications such as organic optoelectronics and quantum computing. With the aim of maintaining a decent level of accessibility, the presence of intersections, often in high numbers, is one of the typical features of rural roads. However, evidence from literature shows that increasing intersection density increases the risk of accidents. Accident analysis literature regarding intersection density primarily consists of accident prediction models which are a useful tool for measuring safety performance of roads, but the literature is lacking in terms of evaluation of driver behavior using direct measurements of driver performance. This study focuses on the influence of intersection density on the risk perception of drivers through experiments carried out with a driving simulator. A virtual driving environment of a rural roadway was constructed. The road consisted of segments featuring extra-urban and village driving environments with varying intersection density level. Participants were recruited to drive through this virtual driving environment. Various driver performance measures such as vehicle speed and brake and gas pedal usage were collected from the experiment and then processed for further analysis. Results indicate an increase in driver's perceived risk when the intersection density increased, according with the findings from the accident prediction modeling literature. However, at the same time, this driving simulator study revealed some interesting insights about oscillating perceived risk among drivers in the case of mid-level intersection separation distances. Beyond the accident research domain, findings from this study can also be useful for engineers and transportation agencies associated with access management to make more informed decisions. The viscoelastic properties of a cell cytoskeleton contain abundant information about the state of a cell. Cells show a response to a specific environment or an administered drug through changes in their viscoelastic properties. Studies of single cells have shown that chemical agents that interact with the cytoskeleton can alter mechanical cell properties and suppress mitosis. This envisions using rheological measurements as a non-specific tool for drug development, the pharmacological screening of new drug agents, and to optimize dosage. Although there exists a number of sophisticated methods for studying mechanical properties of single cells, studying concentration dependencies is difficult and cumbersome with these methods: large cell-to-cell variations demand high repetition rates to obtain statistically significant data. Furthermore, method-induced changes in the cell mechanics cannot be excluded when working in a nonlinear viscoelastic range. To address these issues, we not only compared narrow-gap rheometry with commonly used single cell techniques, such as atomic force microscopy and microfluidic-based approaches, but we also compared existing cell monolayer studies used to estimate cell mechanical properties. This review provides insight for whether and how narrow-gap rheometer could be used as an efficient drug screening tool, which could further improve our current understanding of the mechanical issues present in the treatment of human diseases. Lactic acid is the meekest hydroxyl carboxylic acid (2-hydroxy propionic acid) which is a colorless, odorless, hygroscopic, organic compound with no toxic effect, a very inevitable and versatile chemical used in the Food, cosmetics, textile, and pharmaceutical industries for very long years. Lactic acid was produced as non-racemic when specific microbial strains were used; therefore, microbial fermentation gained more attention. Sugarcane molasses is the best source of components for microbial growth and cheap raw material for Lactic acid fermentation. This study produced sustainable lactic acid from sugarcane molasses by the Bacillus amyloliquefaciens J2V2AA strain with a higher production of 178 gm/L/24 h. Mycoparasites cause serious losses in profitable mushroom farms worldwide. The negative impact of green mold (Trichoderma harzianum) reduces cropping surface and damages basidiomes, limiting production and harvest quality. The goal of the current study was to evaluate new generation fungicides, to devise suitable management strategies against the green mold disease under prevailing agro-climatic conditions. Six non-systemic and five systemic fungitoxicants were evaluated for their efficacy against pathogen, T. harzianum, and host, Agaricus bisporus, under in vitro conditions. Among non-systemic fungicides, chlorothalonil and prochloraz manganese with mean mycelium inhibition of 76.87 and 93.40 percent, respectively, were highly inhibitory against the pathogen. The least inhibition percentage of 7.16 of A. bisporus was exhibited by chlorothalonil. Under in vivo conditions, use of captan 50 WP resulted in a maximum yield of button mushroom of 14.96 kg/qt. So far, systemic fungicides were concerned, carbendazim proved extremely inhibitory to the pathogen (89.22%), with least inhibitory effect on host mycelium (1.56%). However, application of non-systemic fungitoxicants further revealed that fungicide prochloraz manganese 50 WP at 0.1–0.2 percent or chlorothalonil 50 WP at 0.2 percent, exhibited maximum disease control of 89.06–96.30 percent. Moreover, the results of systemic fungitoxicants showed that carbendazim 50 WP or thiophanate methyl 70 WP at 0.1 percent reduced disease to 2.29–3.69 percent, hence exhibiting the disease control of 80.11–87.66 percent. The field of immunotherapy has undergone radical conceptual changes over the last decade. There are various examples of immunotherapy, including the use of monoclonal antibodies, cancer vaccines, tumor-infecting viruses, cytokines, adjuvants, and autologous T cells carrying chimeric antigen receptors (CARs) that can bind cancer-specific antigens known as adoptive immunotherapy. While a lot has been achieved in the field of T-cell immunotherapy, only a fraction of patients (20%) see lasting benefits from this mode of treatment, which is why there is a critical need to turn our attention to other immune cells. B cells have been shown to play both anti- and pro-tumorigenic roles in tumor tissue. In this review, we shed light on the dual nature of B cells in the tumor microenvironment. Furthermore, we discussed the different factors affecting the biology and function of B cells in tumors. In the third section, we described B-cell-based immunotherapies and their clinical applications and challenges. Biosorption has great potential in removing toxic effluents from wastewater, especially heavy metal ions such as cobalt, lead, copper, mercury, cadmium, nickel and other ions. Mathematically modeling of biosorption process is essential for the economical and robust design of equipment employing the bioadsorption process. However, biosorption is a complex physicochemical process involving various transport and equilibrium processes, such as absorption, adsorption, ion exchange and surface and interfacial phenomena. The biosorption process becomes even more complex in cases of multicomponent systems and needs an extensive parametric analysis to develop a mathematical model in order to quantify metal ion recovery and the performance of the process. The biosorption process involves various process parameters, such as concentration, contact time, pH, charge, porosity, pore size, available sites, velocity and coefficients, related to activity, diffusion and dispersion. In this review paper, we describe the fundamental physical and chemical processes involved in the biosorption of heavy metals on various types of commonly employed biosorbents. The most common steady state and dynamic mathematical models to describe biosorption in batch and fixed-bed columns are summarized. Mathematical modeling of dynamic process models results in highly coupled partial differential equations. Approximate methods to study the sensitivity analysis of important parameters are suggested. Alphaviruses are positive-strand RNA viruses, mostly being mosquito-transmitted. Cells infected by an alphavirus become resistant to superinfection due to a block that occurs at the level of RNA replication. Alphavirus replication proteins, called nsP1-4, are produced from nonstructural polyprotein precursors, processed by the protease activity of nsP2. Trans-replicase systems and replicon vectors were used to study effects of nsP2 of chikungunya virus and Sindbis virus on alphavirus RNA replication in mosquito cells. Co-expressed wild-type nsP2 reduced RNA replicase activity of homologous virus; this effect was reduced but typically not abolished by mutation in the protease active site of nsP2. Mutations in the replicase polyprotein that blocked its cleavage by nsP2 reduced the negative effect of nsP2 co-expression, confirming that nsP2-mediated inhibition of RNA replicase activity is largely due to nsP2-mediated processing of the nonstructural polyprotein. Co-expression of nsP2 also suppressed the activity of replicases of heterologous alphaviruses. Thus, the presence of nsP2 inhibits formation and activity of alphavirus RNA replicase in protease activity-dependent and -independent manners. Pterins are bicyclic heterocycles that are found widely across Nature and are involved in a variety of biological functions. Notably, pterins are found at the core of molybdenum cofactor (Moco) containing enzymes in the molybdopterin (MPT) ligand that coordinates molybdenum and facilitates cofactor activity. Pterins are diverse and can be widely functionalized to tune their properties. Exosomes are cell-secreted vesicles secreted by a majority of cells and, hence, populating most of the biological fluids, namely blood, tears, sweat, swab, urine, breast milk, etc. They vary vastly in size and density and are influenced by age, gender and diseases. The composition of exosomes includes lipids, DNA, proteins, and coding and noncoding RNA. There is a significant interest in selectively isolating small exosomes (≤50 nm) from human serum to investigate their role in different diseases and regeneration. However, current techniques for small exosome isolation/purification are time-consuming and highly instrument-dependent, with limited specificity and recovery. Thus, rapid and efficient methods to isolate them from bio fluids are strongly needed for both basic research and clinical applications. In the present work, we explored the application of a bench-top centrifuge for isolating mostly the small exosomes (≤50 nm). This can be achieved at low g-force by adding additional weight to the exosomes by conjugating them with citrate-capped gold nanoparticles (CGNP). CGNPs were functionalized with polyethylene glycol (PEG) to form PEGylated GNP (PGNP). EDC/SNHS chemistry is used to activate the –COOH group of the PEG to make it suitable for conjugation with antibodies corresponding to exosomal surface proteins. These antibody-conjugated PGNPs were incubated with the serum to form PGNP-exosome complexes which were separated directly by centrifugation at a low g-force of 7000× g. This makes this technique efficient compared to that of standard ultracentrifugation exosome isolation (which uses approximately 100,000× g). Using the technique, the exosome isolation from serum was achieved successfully in less than two hours. The purification of small exosomes, characterized by the presence of CD63, CD9 and CD81, and sized between 20 nm to 50 nm, was confirmed by western blot, dynamic light scattering (DLS), transmission electron microscopy (TEM) and nanoparticle tracking analyser (NTA). Beetroot juice is a food high in nitrate and is associated with cardiometabolic health benefits and enhanced exercise performance through the production of nitric oxide in the nitrate–nitrite–nitric oxide pathway. Since various food components influence this pathway, the aim of this trial was to study the effect of beetroot juice alone and in conjunction with vitamin C or protein on the acute response to plasma nitrate and nitrite levels in healthy middle- to older-aged adults. In this cross-over trial, each participant received, in a randomized order, a single dose of Beet It Sport® alone; Beet It Sport®, plus a 200 mg vitamin C supplement; and Beet It Sport® plus 15 g of whey protein. Plasma levels of nitrate and nitrite were determined prior to and at 1 and 3 h after intervention. Log plasma nitrate and nitrite was calculated to obtain data that were normally distributed, and these data were analyzed using two-way within-factors ANOVA, with time and treatment as the independent factors. There were no statistically significant differences for log plasma nitrate (p = 0.308) or log plasma nitrite (p = 0.391) values across treatments. Log plasma nitrate increased significantly from pre-consumption levels after 1 h (p < 0.001) and 3 h (p < 0.001), but plasma nitrate was lower at 3 h than 1 h (p < 0.001). Log plasma nitrite increased from pre to 1 h (p < 0.001) and 3 h (p < 0.001) with log values at 3 h higher than at 1 h (p = 0.003). In this cohort, we observed no differences in log plasma nitrate and nitrite at 1 h and 3 h after co-ingesting beetroot juice with vitamin C or a whey protein supplement compared to beetroot juice alone. Further research needs to be undertaken to expand the blood-sampling time-frame and to examine factors that may influence the kinetics of the plasma nitrate to nitrite efficacy, such as differences in fluid volume and osmolarity between treatments employed. The western Himalayan region is susceptible to minor climate changes because of its fragile ecology, which might threaten the valley's prestigious ecosystems and socio-economic components. The Himalayas's local climate and weather are vulnerable to and interlinked with world-scale climatic changes since the region's hydrology is predominantly dominated by snow and glaciers. The Himalayas, notably the Jammu and Kashmir region in the western Himalayas, has clearly shown distinct and robust evidence of climate change. This study used observed data to examine the climatic variability and trends of change in precipitation and temperature for the Kashmir valley between 1980 and 2020. Gulmarg, Pahalgam, Kokernag, Qazigund, Kupwara, and Srinagar (Shalimar) meteorological stations in the Kashmir valley were studied in detail for long- and short-term as well as localized fluctuations in temperature and precipitation. The annual temperature and precipitation fluctuations were calculated using Sen's slope approach, and the sloping trend was determined using linear regression. The research showed statistically insignificant growing trends in maximum and minimum temperatures throughout the Kashmir valley. The average annual temperature in the Kashmir valley increased by 1.55 °C during the last 41 years (from 1980 to 2020), with a higher rise in maximum and minimum temperature by 2.00 and 1.10 °C, respectively. However, precipitation showed a non-significant decreasing trend concerning time series analysis over 1980 to 2020 in Kashmir valley. Results of annual average maximum temperature at all the stations revealed that Pahalgam (2.2 °C), Kokernag (1.8 °C), and Kupwara (1.8 °C) displayed a steep upsurge and statistically significant trends; however, annual average minimum temperature followed an increasing trend from 1980 to 2020 at all the stations except Shalimar. However, non-significant declining trends in precipitation were recorded at all the locations in Kashmir valley. This changing pattern of temperature and precipitation could have significant environmental consequences, affecting the western Himalayan region's food security and ecological sustainability. Short-time, abrupt events—such as earthquakes and other shock loadings—often lead to damage that is difficult to detect in structures using output-only vibration measurements. The time-variant transmissibility is proposed to tackle this problem. The approach is based on two-dimensional wavelet power spectra. The time-frequency transmissibility and relevant coherence function are used for structural damage detection in structural elements in buildings. Numerical simulations and experimental tests are used in these investigations. The results are compared with the classical transmissibility and time-variant input-output wavelet approach. Optical coherence tomography (OCT) is analogous to ultrasound biometry in the cross sectional imaging of ocular tissues. Development of current devices with deeper penetration and higher resolution has made it popular tool in clinics for visualization of anterior segment structures. In this review, the authors discussed the application of AS-OCT for diagnosis and management of various corneal and ocular surface disorders. Further, recent developments in the application of the device for pediatric corneal disorders and extending the application of OCT angiography for anterior segment are introduced. The fashion industry is the second most polluting industry, contributing 8% of all carbon emissions and 20% of all global wastewater, with an anticipated 50% increase in greenhouse gas emissions by 2030. To gain a better understanding of the state of the academic literature on the environmental impacts of the fast fashion industry, we systematically identified 65 publications from 1996 to November 2021 that were subjected to (i) bibliometric, (ii) text, and (iii) content analysis. We found that there is a growing research interest surrounding fast fashion and water quality, with 74% of the articles published in the last 5 years, and the majority of publications and citations are from China and European countries. We summarise the evaluation of production processes, such as carbon and water footprints, along with recycling practices aimed to increase the sustainability of the fashion industry. While Polo-like kinase 1 (PLK1) inhibitors have shown promise in clinical settings for treating triple-negative breast cancer tumors and other solid tumors, they are limited by their ability to bind non-selectively to the ATP kinase domain. Therefore, we sought to develop a PLK1 allosteric inhibitor targeting the PLK1 T-loop (a switch responsible for activation) and evaluate its effects in triple-negative breast cancer cells. A novel compound, RK-10, was developed based on an in silico model, and its effects on specificity, viability, migration, and cell cycle regulation in MCF-10A and MDA-MB 231 cells were evaluated. When MDA-MB 231 cells were treated with 0–50 µg/mL RK-10, phospho-PLK1 (Thr-210) was decreased in cells cultured adherently and cells cultured as mammospheres. RK-10 significantly inhibited viability after 24 h; however, by 48 h, 25–50 µM RK-10 caused >50% reduction. RK-10 attenuated wound healing by up to 99.7% and caused S and G2/M cell cycle arrest, which was associated with increased p21 expression. We developed a novel allosteric inhibitor which mediates anti-proliferative and anti-migratory properties through targeting phospho-PLK1 (Thr-210) in mammospheres and causing S phase and G2/M cell cycle arrest. A research trial was conducted at Agronomy Farm (SKUAST-K, Wadura, Jammu & Kashmir), during kharif 2017 and 2018 to evaluate nutrient removal in rice under various rice establishment methods and weed control measures. The study comprised of two factors: rice establishment techniques {Transplanting (TPR); Direct seeding (DSR) and System of rice intensification (SRI)} as main plot treatments and weed control measures {Butachlor @ 1500 g a.i ha−1 (B); Penoxsulam @ 22.5 g a.i ha−1 (P); Pyrazosulfuron ethyl + Pretilachlor @ 15 and 600 g a.i ha−1 (PP); Bensulfuron methyl + Pretilachlor @ 60 and 600 g a.i ha−1 (BP); 2 Conoweeding/Hand Weeding (CW/HW); Weed free (WF) and weedy check (WC)} as sub-plot treatments meant to evaluate the best establishment method and weed management practice for rice. Over DSR and transplanted rice, the SRI technique yielded a significant increase in dry biomass accumulation (17.04 and 17.20 t ha−1) and grain (7.92 and 8.17 t ha−1) and straw (9.60 and 10.17 t ha−1) yields. Penoxsulam herbicide significantly showed higher grain and straw yield of 8.19 and 8.28 t ha−1 and 10.13 and 10.44 t ha−1, respectively, than other weed management measures by comparing the means using critical difference. TPR excelled in reducing dry weed biomass more than other established methods. All herbicides considerably reduced dry weed biomass, but Penoxsulam herbicide showed the greatest reduction in dry weed biomass and proved superior against complex weed flora. Weeds showed maximum contribution towards total Biomass under DSR, among rice establishment techniques. In contrast, among different weed control measures, it was maximum in weedy check treatment (Untreated Control) and minimum in penoxsulam treatment. SRI significantly excelled in crop (grain and straw) nutrient uptake compared to the DSR and TPR method, although different crop establishment techniques non-significantly influenced nutrient concentrations. Furthermore, penoxsulam treatment demonstrated higher crop (grain and straw) nutrient uptake among the various weed management measures. However, available soil nutrients were observed among establishment techniques, highest in DSR and lowest in SRI. Moreover, direct-seeded rice excelled SRI and transplanted rice in weed nutrient uptake, and among the different herbicidal treatments, penoxsulam recorded the lowest uptake in weeds. Nutrient budgeting demonstrated that DSR showed the maximum percentage of nutrient removal by weeds, and the minimum ratio was in TPR. Acute ocular burns have varied manifestations which require prompt diagnosis and management to prevent chronic sequelae. Of these, the detection of limbal ischemia poses a challenge because of the subjective nature of its clinical signs. Anterior segment optical coherence tomography angiography (AS-OCTA) offers an objective method of assessing ischemia in these eyes. This review provides an overview of the technology of AS-OCTA and its applications in acute burns. AS-OCTA generates images by isolating the movement of erythrocytes within blood vessels from sequentially obtained b-scans. Limbal ischemia manifests in these scans as absent vasculature and the extent of ischemia can be quantified using different vessel-related parameters. Of these, the density of vessels is most commonly used and correlates with the severity of the injury. Incorporation of the degree of ischemia in the classification of acute burns has been attempted in animal studies and its extension to human trials may provide an added dimension in determining the final prognosis of these eyes. Thus, AS-OCTA is a promising device that can objectively evaluate limbal ischemia. This will facilitate the identification of patients who will benefit from revascularization therapies and stem cell transplants in acute and chronic ocular burns, respectively. This post-authorization study was conducted to evaluate the safety of insulin degludec/insulin aspart (IDegAsp) in adult patients with diabetes mellitus (DM) during routine clinical care under a real-world setting in India. Eligible patients received IDegAsp for a minimum of 12 months during routine clinical management. Data were collected at 0, 3, 6, and 12 months. In total, 1029 adult patients with DM were included; 65.2% (n = 671) were men; mean age was 55.0 ± 12.2 years, and the mean duration of diabetes mellitus was 10.8 ± 7.4 years. Thirty adverse events were reported in 23 patients (2.2%) during the follow-up: two adverse events in two patients were serious with fatal outcomes, which were unrelated to IDegAsp use. At baseline, there were 176 confirmed hypoglycemic events in 67 (6.7%) patients while they were on their previous treatment options. At 12 months of treatment with IDegAsp, 11 confirmed hypoglycemic events were reported in 11 (1.1%) patients since the previous visit; there were no reported episodes of severe hypoglycemia. Mean glycosylated hemoglobin value reduced from 9.5% ± 1.8% at baseline to 7.7% ± 1.1% at 12 months. The finishing of additive manufactured (AM) components is crucial for endowing them with fatigue resistance. Unfortunately, current AM processes naturally promote anisotropic surface characteristics that make it challenging to optimize finishing processes. In this study, bead-blasting is explored as a process for finishing Electron Beam Melted (EBM) Ti-6Al-4V. The effects of anisotropic roughness characteristics on the mechanics of bead-blasting are delineated using surface texture measurements via optical profilometry and residual stress measurements via X-ray diffraction. As-received surfaces resulting from AM, as well as those that have been Electrical Discharge Machined (EDM), are studied. It is seen that pre-processed roughness textures heavily influence the final textures and residual stresses. These linkages are quantified using a plasticity index as the governing metric—a rougher surface features a larger plastic index, which results in comparatively greater evolution of its texture characteristics than a smoother surface after equivalent bead-blasting treatments. The mechanics of this evolution are delineated using energy-controlled indentation as a model representing a single impact in bead-blasting. It is seen that rougher surfaces featuring complex textures in as-received states also produce complex stress states featuring a greater level of locally tensile stresses during indentation compared with smoother surfaces. Approaches to address these complications are proposed that can potentially transform a printed, non-functional surface into one that is optimized for fatigue resistance. [ABSTRACT FROM AUTHOR]
