Old trees are irreplaceable conservation resources with numerous ecological and socio-cultural values. While many forests have experienced significant declines under recent climate warming, the risk of growth declines in old trees remains unknown. Here, we tackle this problem by dendrochronological studies of 30 old trees in a Platycladus orientalis forest at the northern boundary of the Taihang Mountain of China. We examined annual growth trajectories of trees at individual level and discovered four severe growth decline events over the last 150 years, including the periods of 1894–1899, 1913–1919, 1964–1967 and 2004–2018. The most recent growth decline event lasted for 15-year and involced 50% to 75% of the old trees. This decline was unprecedented in both its extent and duration. Furthermore, the growth–climate relationship of these old trees has changed since 1990. Before 1990, tree growth was significantly correlated with minimum winter; after 1990, tree growth became significantly correlated with the self-calibrating Palmer Drought Index. These results suggest that warming-induced droughts after 1990 could be the primary driver of the recent growth decline. If climate warming continues and drought stresses intensify, the old trees may face an increased risk of growth decline and even mortality. [ABSTRACT FROM AUTHOR]

Climate change plays a pivotal role in shaping the shifting patterns of plant distribution, and gaining insights into how medicinal plants in the plateau region adapt to climate change will be instrumental in safeguarding the rich biodiversity of the highlands. Gymnosia orchidis Lindl. (G. orchidis) is a valuable Tibetan medicinal resource with significant medicinal, ecological, and economic value. However, the growth of G. orchidis is severely constrained by stringent natural conditions, leading to a drastic decline in its resources. Therefore, it is crucial to study the suitable habitat areas of G. orchidis to facilitate future artificial cultivation and maintain ecological balance. In this study, we investigated the suitable zones of G. orchidis based on 79 occurrence points in the Qinghai–Tibet Plateau (QTP) and 23 major environmental variables, including climate, topography, and soil type. We employed the Maximum Entropy model (MaxEnt) to simulate and predict the spatial distribution and configuration changes in G. orchidis during different time periods, including the last interglacial (LIG), the Last Glacial Maximum (LGM), the Mid-Holocene (MH), the present, and future scenarios (2041–2060 and 2061–2080) under three different climate scenarios (SSP126, SSP370, and SSP585). Our results indicated that annual precipitation (Bio12, 613–2466 mm) and mean temperature of the coldest quarter (Bio11, −5.8–8.5 °C) were the primary factors influencing the suitable habitat of G. orchidis, with a cumulative contribution of 78.5%. The precipitation and temperature during the driest season had the most significant overall impact. Under current climate conditions, the suitable areas of G. orchidis covered approximately 63.72 × 104/km2, encompassing Yunnan, Gansu, Sichuan, and parts of Xizang provinces, with the highest suitability observed in the Hengduan, Yunlin, and Himalayan mountain regions. In the past, the suitable area of G. orchidis experienced significant changes during the Mid-Holocene, including variations in the total area and centroid migration direction. In future scenarios, the suitable habitat of G. orchidis is projected to expand significantly under SSP370 (30.33–46.19%), followed by SSP585 (1.41–22.3%), while contraction is expected under SSP126. Moreover, the centroids of suitable areas exhibited multidirectional movement, with the most extensive displacement observed under SSP585 (100.38 km2). The number of people in a crowd is crucial information in public safety, intelligent monitoring, traffic management, architectural design, and other fields. At present, the counting accuracy in public spaces remains compromised by some unavoidable situations, such as the uneven distribution of a crowd and the difference in head scale caused by people's differing distances from the camera. To solve these problems, we propose a deep learning crowd counting model, multi-scale dilated convolution networks (MSDCNet), based on crowd density map estimation. MSDCNet consists of three parts. The front-end network uses the truncated VGG16 to obtain preliminary features of the input image, with a proposed spatial pyramid pooling (SPP) module replacing the max-pooling layer to extract features with scale invariance. The core network is our proposed multi-scale feature extraction network (MFENet) for extracting features in three different scales. The back-end network consists of consecutive dilation convolution layers instead of traditional alternate convolution and pooling to expand the receptive field, extract high-level semantic information and avoid the spatial feature loss of small-scale heads. The experimental results on three public datasets show that the proposed model solved the above problems satisfactorily and obtained better counting accuracy than representative models in terms of mean absolute error (MAE) and mean square error (MSE). The conversion of skeletal muscle fiber from fast-twitch to slow-twitch is crucial for sustained contractile and stretchable events, energy homeostasis, and anti-fatigue ability. The purpose of our study was to explore the mechanism and effects of garcinol on the regulation of skeletal muscle fiber type transformation. Forty 21-day-old male C57/BL6J mice (n = 10/diet) were fed a control diet or a control diet plus garcinol at 100 mg/kg (Low Gar), 300 mg/kg (Mid Gar), or 500 mg/kg (High Gar) for 12 weeks. The tibialis anterior (TA) and soleus muscles were collected for protein and immunoprecipitation analyses. Dietary garcinol significantly downregulated (p < 0.05) fast myosin heavy chain (MyHC) expression and upregulated (p < 0.05) slow MyHC expression in the TA and soleus muscles. Garcinol significantly increased (p < 0.05) the activity of peroxisome proliferator-activated receptor gamma co-activator 1α (PGC-1α) and markedly decreased (p < 0.05) the acetylation of PGC-1α. In vitro and in vivo experiments showed that garcinol decreased (p < 0.05) lactate dehydrogenase activity and increased (p < 0.05) the activities of malate dehydrogenase and succinic dehydrogenase. In addition, the results of C2C12 myotubes showed that garcinol treatment increased (p < 0.05) the transformation of glycolytic muscle fiber to oxidative muscle fiber by 45.9%. Garcinol treatment and p300 interference reduced (p < 0.05) the expression of fast MyHC but increased (p < 0.05) the expression of slow MyHC in vitro. Moreover, the acetylation of PGC-1α was significantly decreased (p < 0.05). Garcinol promotes the transformation of skeletal muscle fibers from the fast-glycolytic type to the slow-oxidative type through the p300/PGC-1α signaling pathway in C2C12 myotubes. In this paper, the transverse vibration of a fractional viscoelastic beam is studied based on the fractional calculus, and the corresponding scheme of a viscoelastic beam is established by using the mixed finite volume element method. The stability and convergence of the algorithm are analyzed. Numerical examples demonstrate the effectiveness of the algorithm. Finally, the values of different parameter sets are tested, and the test results show that both the damping coefficient and the fractional derivative have significant effects on the model. Pansy (Viola × wittrockiana Gams.) is an important and attractive ornamental plant with a wide variety of flower colors. To date, the molecular genetic understanding of its colorful petal pigment patterns remains largely unknown. In this study, we analyzed the bicolor petals of "Mengdie" in cytological, physiological, and transcriptomic aspects. Results showed that the difference of flower colors was mainly determined by the pigment distribution in the epidermal cells. Pigment analysis indicated that anthocyanins had strong correlations with color parameters, which acted as the main factor in flower coloration. Comparative transcriptome analysis found a total of 43,908 unigenes with the mean length of 682 bp. There were 24,323, 16,668, 8507, and 7680 unigenes annotated in the Nr, Swiss-Prot, KOG, and KEGG databases, respectively. Differential expression genes (DEGs) showed that the expression of anthocyanin late biosynthesis genes (LBGs), VwF3′H, VwF3′5′H, and VwUFGT, would be likely to play a major role in the color formation of pansy. The expression patterns of selected DEGs were verified by qRT-PCR. In the early morning of 21 April 2021 local time, the Indonesian Navy submarine KRI nanggala-402 crashed in the Bali Sea (BS). As internal solitary waves (ISWs) are a great threat to submarine navigation, this paper analyzes the characteristics of ISWs in the BS by surveying satellite remote sensing images collected from 12–21 April 2021. The satellite images revealed active ISWs in the BS near the submarine wreck site with crest lengths approaching 200 km. Originating from the Lombok Strait (LS), the waves travelled northwestward across the BS deep basin, passed through the submarine wreck site, and shoaled onto the continental shelf west of the Kangean Islands, during which process, the propagation speed reached 2.69 m/s in the deep basin and 0.71 m/s in the shallow water. Based on the satellite images, the wave amplitude near the wreck site was reconstructed to be 41 m, and the reconstructed underwater wave structure showed a maximum vertical velocity of 10 cm/s. Satellite images also demonstrated the near-source evidence of ISWs near the Nusa Penida sill of the LS, and their generation were estimated to be related to the southward tidal current troughs. Weaning significantly alters hepatic aromatic amino acid (AAA) metabolism and physiological functions. However, less is known about the regulating mechanism of hepatic AAA metabolism after weaning. A total of 200 21-day-old piglets (Duroc × Landrace) were assigned randomly to the control group and the weaning group. In this study, weaning significantly decreased the concentration of phenylalanine, tryptophan, and tyrosine in piglet livers (p < 0.05). Additionally, through the detection of liver AAA metabolites and metabolic enzyme activity, it was observed that hepatic tryptophan catabolism was enhanced, while that of phenylalanine was weakened (p < 0.05). Intriguingly, acetyl-proteome profiling of liver from weaned piglets showed that weaning exacerbated the acetylation of phenylalanine hydroxylase (PAH) and the deacetylation of tryptophan 2,3-dioxygenase (TDO). Analysis of PAH and TDO acetylation in Chang liver cells showed that acetylation decreased the PAH activity, while deacetylation increased the TDO activity (p < 0.05). Furthermore, metabolites of AAAs and the acetylation statuses of PAH and TDO in primary hepatocytes from weaned piglets were consistent with the results in vivo. These findings indicated that weaning altered the PAH and TDO activity by affecting the acetylation state of the enzyme in piglets" livers. The liver is the central organ of glycolipid metabolism, which regulates the metabolism of lipids and glucose to maintain energy homeostasis upon alterations of physiological conditions. Researchers formerly focused on the phosphorylation of glucagon in controlling liver metabolism. Noteworthily, emerging evidence has shown glucagon could additionally induce acetylation to control hepatic metabolism in response to different physiological states. Through inducing acetylation of complex metabolic networks, glucagon interacts extensively with various energy-sensing factors in shifting from glucose metabolism to lipid metabolism during prolonged fasting. In addition, glucagon-induced acetylation of different energy-sensing factors is involved in the advancement of nonalcoholic fatty liver disease (NAFLD) to liver cancer. Here, we summarize the latest findings on glucagon to control hepatic metabolism by inducing acetylation of energy-sensing factors.
