Background: We aimed to elucidate the quantitative relationship between the neuromuscular blockade depth and intraoperative motor-evoked potential amplitudes. Methods: This prospective, single-arm, open-label, observational study was conducted at a single university hospital in Seoul, Korea, and included 100 adult patients aged ≥19 years undergoing brain tumor removal surgery under general anesthesia. We measured the neuromuscular blockade degree and motor-evoked potential amplitude in the deltoid, abductor pollicis brevis, tibialis anterior, and abductor hallucis muscles until dural opening. Results: The pharmacokinetic-pharmacodynamic model revealed the exposure-response relationship between the rocuronium effect-site concentration and motor-evoked potential amplitudes. The mean motor-evoked potential amplitudes decreased proportionally with increasing neuromuscular blockade depth. As the mean amplitude increased, the coefficient of variation decreased bi-exponentially. The critical ratio of the first evoked response to the train-of-four stimulation (T1)/control response (Tc) thresholds beyond which the coefficient of variation exhibited minimal change were found to be 0.63, 0.65, 0.68, and 0.63 for the deltoid, abductor pollicis brevis, tibialis anterior, and abductor hallucis muscles, respectively. Conclusions: Our results reveal that the motor-evoked potential amplitude exhibits deterioration proportional to the degree of neuromuscular blockade. In light of the observed bi-exponential decline of the coefficient of variation with the motor-evoked potential amplitude, we recommend maintaining a T1/Tc ratio higher than 0.6 for partial neuromuscular blockade. The increasing utilization of artificial intelligence algorithms in drug development has proven to be highly efficient and effective. One area where deep learning-based approaches have made significant contributions is in drug repositioning, enabling the identification of new therapeutic applications for existing drugs. In the present study, a trained deep-learning model was employed to screen a library of FDA-approved drugs to discover novel inhibitors targeting JAK2. To accomplish this, reference datasets containing active and decoy compounds specific to JAK2 were obtained from the DUD-E database. RDKit, a cheminformatic toolkit, was utilized to extract molecular features from the compounds. The DeepChem framework's GraphConvMol, based on graph convolutional network models, was applied to build a predictive model using the DUD-E datasets. Subsequently, the trained deep-learning model was used to predict the JAK2 inhibitory potential of FDA-approved drugs. Based on these predictions, ribociclib, topiroxostat, amodiaquine, and gefitinib were identified as potential JAK2 inhibitors. Notably, several known JAK2 inhibitors demonstrated high potential according to the prediction results, validating the reliability of our prediction model. To further validate these findings and confirm their JAK2 inhibitory activity, molecular docking experiments were conducted using tofacitinib—an FDA-approved drug for JAK2 inhibition. Experimental validation successfully confirmed our computational analysis results by demonstrating that these novel drugs exhibited comparable inhibitory activity against JAK2 compared to tofacitinib. In conclusion, our study highlights how deep learning models can significantly enhance virtual screening efforts in drug discovery by efficiently identifying potential candidates for specific targets such as JAK2. γ-Aminobutyric acid aminotransferase (GABA-AT) is a pyridoxal 5′-phosphate (PLP)-dependent enzyme that degrades γ-aminobutyric (GABA) in the brain. GABA is an important inhibitory neurotransmitter that plays important neurological roles in the brain. Therefore, GABA-AT is an important drug target that regulates GABA levels. Novel and potent drug development to inhibit GABA-AT is still a very challenging task. In this study, we aimed to devise novel and potent inhibitors against GABA-AT using computer-aided drug design (CADD) tools. Since the crystal structure of human GABA-AT was not yet available, we utilized a homologous structure derived from our previously published paper. To identify highly potent compounds relative to vigabatrin, an FDA-approved drug against human GABA-AT, we developed a pharmacophore analysis protocol for 530,000 Korea Chemical Bank (KCB) compounds and selected the top 50 compounds for further screening. Preliminary biological analysis was carried out for these 50 compounds and 16 compounds were further assessed. Subsequently, molecular docking, molecular dynamics (MD) simulations, and binding free energy calculations were carried out. In the results, four predicted compounds, A07, B07, D08, and H08, were found to be highly potent and were further evaluated by a biological activity assay to confirm the results of the GABA-AT activity inhibition assay. Despite their clonal origins, tumors eventually develop into complex communities made up of phenotypically different cell subpopulations, according to mounting evidence. Tumor cell-intrinsic programming and signals from geographically and temporally changing microenvironments both contribute to this variability. Furthermore, the mutational load is typically lacking in childhood malignancies of adult cancers, and they still exhibit high cellular heterogeneity levels largely mediated by epigenetic mechanisms. Ewing sarcomas represent highly aggressive malignancies affecting both bone and soft tissue, primarily afflicting adolescents. Unfortunately, the outlook for patients facing relapsed or metastatic disease is grim. These tumors are primarily fueled by a distinctive fusion event involving an FET protein and an ETS family transcription factor, with the most prevalent fusion being EWS/FLI1. Despite originating from a common driver mutation, Ewing sarcoma cells display significant variations in transcriptional activity, both within and among tumors. Recent research has pinpointed distinct fusion protein activities as a principal source of this heterogeneity, resulting in markedly diverse cellular phenotypes. In this review, we aim to characterize the role of the EWS/FLI fusion protein in Ewing sarcoma by exploring its general mechanism of activation and elucidating its implications for tumor heterogeneity. Gamma-aminobutyric acid (GABA) transaminase—also called GABA aminotransferase (GABA-AT)—deficiency is a rare autosomal recessive disorder characterized by a severe neonatal-infantile epileptic encephalopathy with symptoms such as seizures, hypotonia, hyperreflexia, developmental delay, and growth acceleration. GABA transaminase deficiency is caused by mutations in GABA-AT, the enzyme responsible for the catabolism of GABA. Mutations in multiple locations on GABA-AT have been reported and their locations have been shown to influence the onset of the disease and the severity of symptoms. We examined how GABA-AT mutations influence the structural stability of the enzyme and GABA-binding affinity using computational methodologies such as molecular dynamics simulation and binding free energy calculation to understand the underlying mechanism through which GABA-AT mutations cause GABA-AT deficiency. GABA-AT 3D model depiction was carried out together with seven individual mutated models of GABA-AT. The structural stability of all the predicted models was analyzed using several tools and web servers. All models were evaluated based on their phytochemical values. Additionally, 100 ns MD simulation was carried out and the mutated models were evaluated using RMSD, RMSF, Rg, and SASA. gmxMMPBSA free energy calculation was carried out. Moreover, RMSD and free energy calculations were also compared with those obtained using online web servers. Our study demonstrates that P152S, Q296H, and R92Q play a more critical role in the structural instability of GABA-AT compared with the other mutated models: G465R, L211F, L478P, and R220K. Dosage-sensitive sex reversal, adrenal hypoplasia critical region, on chromosome X, gene 1 (DAX1) is an orphan nuclear receptor encoded by the NR0B1 gene. The functional study showed that DAX1 is a physiologically significant target for EWS/FLI1-mediated oncogenesis, particularly Ewing Sarcoma (ES). In this study, a three-dimensional DAX1 structure was modeled by employing a homology modeling approach. Furthermore, the network analysis of genes involved in Ewing Sarcoma was also carried out to evaluate the association of DAX1 and other genes with ES. Moreover, a molecular docking study was carried out to check the binding profile of screened flavonoid compounds against DAX1. Therefore, 132 flavonoids were docked in the predicted active binding pocket of DAX1. Moreover, the pharmacogenomics analysis was performed for the top ten docked compounds to evaluate the ES-related gene clusters. As a result, the five best flavonoid-docked complexes were selected and further evaluated by Molecular Dynamics (MD) simulation studies at 100 ns. The MD simulation trajectories were evaluated by generating RMSD, hydrogen bond plot analysis, and interaction energy graphs. Our results demonstrate that flavonoids showed interactive profiles in the active region of DAX1 and can be used as potential therapeutic agents against DAX1-mediated augmentation of ES after in-vitro and in-vivo evaluations. Plasmodium vivax (P. vivax) is one of the human's most common malaria parasites. P. vivax is exceedingly difficult to control and eliminate due to the existence of extravascular reservoirs and recurring infections from latent liver stages. Traditionally, licorice compounds have been widely investigated against viral and infectious diseases and exhibit some promising results to combat these diseases. In the present study, computational approaches are utilized to study the effect of licorice compounds against P. vivax Duffy binding protein (DBP) to inhibit the malarial invasion to human red blood cells (RBCs). The main focus is to block the DBP binding site to Duffy antigen receptor chemokines (DARC) of RBC to restrict the formation of the DBP–DARC complex. A molecular docking study was performed to analyze the interaction of licorice compounds with the DARC binding site of DBP. Furthermore, the triplicates of molecular dynamic simulation studies for 100 ns were carried out to study the stability of representative docked complexes. The leading compounds such as licochalcone A, echinatin, and licochalcone B manifest competitive results against DBP. The blockage of the active region of DBP resulting from these compounds was maintained throughout the triplicates of 100 ns molecular dynamic (MD) simulation, maintaining stable hydrogen bond formation with the active site residues of DBP. The coronavirus disease (COVID-19) outbreak affected the utilization and management of blood products in hospitals. Blood shortages occurred owing to social distancing policies and reduction in blood donors. However, only a few studies examined whether these changes affected blood usage and transfusion patterns. We retrospectively reviewed blood component usage according to hospital departments and phases of surgery in transfused patients admitted between 1 March 2019 and 28 February 2021, in a single center in Anyang, Korea. We also analyzed the length of hospital stay and mortality to determine prognosis. In 2020, 32,050 blood components were transfused to 2877 patients, corresponding to 15.8% and 11.8% less than the rates in 2019, respectively. Postoperative usage of blood products significantly decreased in 2020 (3.87 ± 6.50) compared to 2019 (7.12 ± 21.71) (p = 0.047). The length of hospital stay of the patients who underwent postoperative transfusion in 2019 (n = 197) was 13.97 ± 11.95 days, which was not significantly different from that in 2020 (n = 167), i.e., 16.44 ± 17.90 days (p = 0.118). Further, 9 of 197 postoperative transfusion patients died in 2019, while 8 of 167 patients died in 2020 (p = 0.920). Surface imprinting used for protein recognition in functional cavities is highly effective in imprinting biomacromolecules to avoid template encapsulation during the formation of a molecularly imprinted polymer (MIP) matrix. Herein, we introduce a facile surface-imprinting method based on two-step spin-coating and photopolymerization to design highly efficient imprinted sites on polymeric films to detect trypsin (TRY). Well-distributed template imprinting is successfully achieved for maximized sensing responses by controlling the composition of functional monomers and crosslinkers in the precursor solution and the concentration of TRY in the imprinting solution. The MIP film exhibits higher sensitivity (−841 ± 65 Hz/(μg/mL)) with a coefficient of determination of 0.970 and a higher imprinting factor of 4.5 in a 0.24 µg/mL TRY solution compared to the nonimprinted polymer (NIP) film. Moreover, the limit of detection and limit of quantification are calculated to be 25.33 and 84.42 ng/mL, respectively. Finally, the selectivity coefficient is within the range of 3.90–6.78 for TRY against other proteins. These sensing properties are superior to those of the corresponding nonimprinted polymer matrix. Ewing sarcoma (ES) is a highly malignant carcinoma prevalent in children and most frequent in the second decade of life. It mostly occurs due to t(11;22) (q24;q12) translocation. This translocation encodes the oncogenic fusion protein EWS/FLI (Friend leukemia integration 1 transcription factor), which acts as an aberrant transcription factor to deregulate target genes essential for cancer. Traditionally, flavonoids from plants have been investigated against viral and cancerous diseases and have shown some promising results to combat these disorders. In the current study, representative flavonoid compounds from various subclasses are selected and used to disrupt the RNA-binding motif of EWS, which is required for EWS/FLI fusion. By blocking the RNA-binding motif of EWS, it might be possible to combat ES. Therefore, molecular docking experiments validated the binding interaction patterns and structural behaviors of screened flavonoid compounds within the active region of the Ewing sarcoma protein (EWS). Furthermore, pharmacogenomics analysis was used to investigate potential drug interactions with Ewing sarcoma-associated genes. Finally, molecular dynamics simulations were used to investigate the stability of the best selected docked complexes. Taken together, daidzein, kaempferol, and genistein exhibited a result comparable to ifosfamide in the proposed in silico study and can be further analyzed as possible candidate compounds in biological in vitro studies against ES. This study investigates the impact of the sea surface temperature (SST) on the forecast of two typhoons, which consecutively hit South Korea in 2020. SST data were obtained from the Daily Optimum Interpolation Sea Surface Temperature (OISST) version 2 and HYbrid Coordinate Ocean Model/Navy Coupled Ocean Data Assimilation (HYCOM/NCODA; GLBy0.08/expt_93.0). When verified using in situ observational data, the OISST data did not accurately estimate the changes in SST during each typhoon's landfall period compared to the HYCOM data since it has a relatively low temporal resolution. To investigate the impact of these two SST data on typhoon forecasts, we conducted sensitivity experiments using the Weather Research and Forecasting (WRF) model. The results showed that simulated typhoon intensities were significantly improved in the simulations with HYCOM data (HY runs), while typhoon track forecast performances were similar in both runs. In addition, the forecast performances of the maximum wind speed at 10 m during the typhoon landfall period were improved in the HY runs. Therefore, this study showed that the overall typhoon intensity and forecast performances during the landfall period could be improved when the higher temporal-resolution SST data were prescribed in the model boundary conditions for a better representation of typhoon-induced SST changes. The copper molten marks at a fire site provide important clues for determining the causes of fire. Four factors have been presented to quantitatively discriminate copper molten marks, namely the fraction of (001) component perpendicular to the demarcation line, the grain aspect ratio, the fraction of Σ3 boundaries, and the fraction of maximum grain size. However, only laboratory-level results of these parameters have been presented, and their applicability in actual fires is yet to be verified. In this study, a fire reproduction experimental system was configured to generate molten marks similar to those in actual fire sites. The molten marks were measured by electron backscatter diffraction and applied to the four discriminant factors. The results obtained similar characteristics to those of the laboratory unit, confirming the applicability of the four discriminant factors. Discriminant equations and processes that can distinguish the primary and secondary arc beads were derived using the molten marks generated in the laboratory and reproduction experiments. Furthermore, a probabilistic discrimination method and classification model developed by machine learning were proposed. This study examined whether information and communication technology (ICT), over the top (OTT) market growth, and economic freedom affect tourism at the global level. Toward this end, the present study tested regression models of inbound tourism and outbound tourism with panel data from 50 countries covering the years 2013 to 2020. The results of the panel-data analysis suggest that high levels of OTT advertising revenue and mobile broadband penetration contribute to high levels of inbound tourism. The results also reveal that high OTT advertising revenue, labor freedom, and income are associated with high levels of outbound tourism. The microstructure of molten marks changes according to ambient temperatures, when a short circuit occurs. Investigation of microstructural changes is important for understanding the properties of copper and examining the cause of a fire. In this study, the boundary characteristics and grain-size distribution of molten marks—primary-arc beads (PABs), which short-circuited at room temperature (25 °C), and secondary-arc beads (SABs), which short-circuited at high temperatures (600 °C, 900 °C)—were compared using electron backscatter diffraction. The distribution of Σ3 boundaries was compared, and it was found that SABs have a higher fraction of Σ3 boundaries than PABs. Moreover, it was confirmed that the ratio of maximum grain size (area) to the total area of the molten mark in SABs is larger than that in PABs. Thus, reliable discriminant factors were suggested, such as the fraction of Σ3 boundaries and normalized maximum grain size, which can distinguish PABs and SABs. The four discriminant factors, such as the (001)//LD, GAR, fraction of Σ3 boundaries, and fraction of maximum grain size to the total molten-mark area, were verified using the machine learning of t-SNE and Pearson correlation analyses. Combination therapies for the treatment of oral squamous cell carcinoma have been studied extensively and represent a synergistic approach with better outcomes than monotherapy. In this study, a novel combination therapy was investigated using gold nanoparticles (GNP) conjugated to programmed cell death protein ligand 1 (PD-L1) antibodies and nonthermal plasma (NTP). The present study describes the effectiveness of NTP using PD-L1 antibody conjugated to GNP in PD-L1 expressing SCC-25 cells, an oral squamous cell carcinoma line. Immunocytochemistry revealed higher levels of PD-L1 expression and an increase in the selective uptake of PD-L1 antibody + GNP on SCC-25 cells compared to HaCaT cells. In addition, cell viability analyses confirmed higher levels of cell death of SCC-25 cells after treatment with PD-L1 antibody, GNP, and NTP compared to HaCaT cells. Among the experimental groups, the highest cell death was observed upon treatment with PD-L1 antibody + GNP + NTP. Following the Western blot analysis and immunofluorescence staining, the expression of apoptosis-related proteins was found to increase after treatment with PD-L1 antibody + GNP + NTP among the other experimental groups. In conclusion, the treatment of SCC-25 cells with PD-L1 antibody + GNP + NTP significantly increased the number of dead cells compared to other experimental groups. The purpose of this study was to investigate the accuracy of the airway volume measurement by a Regression Neural Network-based deep-learning model. A set of manually outlined airway data was set to build the algorithm for fully automatic segmentation of a deep learning process. Manual landmarks of the airway were determined by one examiner using a mid-sagittal plane of cone-beam computed tomography (CBCT) images of 315 patients. Clinical dataset-based training with data augmentation was conducted. Based on the annotated landmarks, the airway passage was measured and segmented. The accuracy of our model was confirmed by measuring the following between the examiner and the program: (1) a difference in volume of nasopharynx, oropharynx, and hypopharynx, and (2) the Euclidean distance. For the agreement analysis, 61 samples were extracted and compared. The correlation test showed a range of good to excellent reliability. A difference between volumes were analyzed using regression analysis. The slope of the two measurements was close to 1 and showed a linear regression correlation (r2 = 0.975, slope = 1.02, p < 0.001). These results indicate that fully automatic segmentation of the airway is possible by training via deep learning of artificial intelligence. Simple Summary: An abnormal expression of microRNA is commonly observed in cancer. Since a single miRNA can target numerous genes, it is important to understand the exact mechanism for the regulation of cancer growth by miRNAs. Here, we show that miR-101-3p, which is downregulated in several cancers, regulates RPL11 ubiquitination by targeting USP47, thereby controlling p53 levels by affecting the localization of RPL11 and its interaction with MDM2. Our results provide a novel mechanism for the inhibition of cancer cell growth by miR-101-3p, and suggest that miR-101-3p could be a potential target as an anticancer agent. MicroRNAs (miRNAs) are a class of small non-coding RNA molecules that regulate a countless number of genes in the cell, and the aberrant expression of miRNA can lead to cancer. Here, we demonstrate that miR-101-3p regulates the RPL11–MDM2–p53 pathway by targeting ubiquitin-specific peptidase 47 (USP47), consequently inhibiting cancer cell proliferation. We confirm that miR-101-3p directly binds to the 3′-UTR region of the USP47 gene and inhibits USP47 expression. In addition, the overexpression of miR-101-3p suppresses cell proliferation in a p53-dependent manner. MiR-101-3p promotes interaction between RPL11 and MDM2 by inducing the translocation of RPL11 from the nucleolus to the nucleoplasm, thus preventing the MDM2-mediated proteasomal degradation of p53. However, these phenomena are restored by the overexpression of USP47, but not by its catalytically inactive form. Indeed, miR-101-3p regulates RPL11 localization and its interaction with MDM2 by inhibiting the USP47-induced deubiquitination of RPL11. Finally, the expression of miR-101-3p is downregulated in lung cancer patients, and the patients with low miR-101-3p expression exhibit a lower survival rate, indicating that miR-101-3p is associated with tumorigenesis. In this work, sensing behaviors and mechanisms of two crosslinked copolymers with dimethylamine and dimethylamide functional groups were compared and investigated for their ability to detect hydrogen chloride (HCl) gas. The crosslinked copolymer films were photopolymerized on quartz crystal electrodes using a micro-contact printing technique. The gas sensing behaviors were analyzed by measuring resonant frequency (Δf) of quartz crystal microbalance (QCM). The HCl binding capacity of photopolymerized films, with a mass between 4.6 and 5.9 μg, was optimized. Under optimized film mass conditions, the poly(2-dimethylaminoethyl methacrylate-co-ethylene glycol dimethacrylate) (DMAEMA-co-EGDMA), poly(DMAEMA-co-EGDMA), film, C2-DMA, showed a 13.9-fold higher binding capacity than the poly(N,N-dimethylacrylamide-co-ethylene glycol dimethacrylate, poly(DMAA-co-EGDMA), film, C0-DMA, during HCl gas adsorption. HCl gas was effectively adsorbed on the C2-DMA film because of the formation of tertiary amine salts through protonation and strong ionic bonding. Furthermore, the C2-DMA film exhibited excellent sensitivity, of 2.51 (ng/μg) (1/ppm), and selectivity coefficient (k* = 12.6 for formaldehyde and 13.5 for hydrogen fluoride) compared to the C0-DMA film. According to the experimental results, and due to its high functionality and stability, the C2-DMA film-coated QC electrode could be used as an HCl gas sensor, with low-cost and simple preparation, in future endeavors. Fatty acid synthase (FASN) plays an important role in cancer development, providing excess lipid sources for cancer growth by participating in de novo lipogenesis. Although several inhibitors of FASN have been developed, there are many limitations to using FASN inhibitors alone as cancer therapeutics. We therefore attempted to effectively inhibit cancer cell growth by using a FASN inhibitor in combination with an inhibitor of a deubiquitinating enzyme USP14, which is known to maintain FASN protein levels in hepatocytes. However, when FASN and USP14 were inhibited together, there were no synergistic effects on cancer cell death compared to inhibition of FASN alone. Surprisingly, USP14 rather reduced the protein levels and activity of FASN in cancer cells, although it slightly inhibited the ubiquitination of FASN. Indeed, treatment of an USP14 inhibitor IU1 did not significantly affect FASN levels in cancer cells. Furthermore, from an analysis of metabolites involved in lipid metabolism, metabolite changes in IU1-treated cells were significantly different from those in cells treated with a FASN inhibitor, Fasnall. These results suggest that FASN may not be a direct substrate of USP14 in the cancer cells. Consequently, we demonstrate that USP14 regulates proliferation of the cancer cells in a fatty acid synthase-independent manner, and targeting USP14 in combination with FASN may not be a viable method for effective cancer treatment. Zinc plays a pivotal role in the function of cells and can induce apoptosis in various cancer cells, including Raji B lymphoma. However, the metabolic mechanism of Zn-induced apoptosis in Raji cells has not been explored. In this study, we performed global metabolic profiling using UPLC−Orbitrap−MS to assess the apoptosis of Raji cells induced by Zn ions released from ZnO nanorods. Multivariate analysis and database searches identified altered metabolites. Furthermore, the differences in the phosphorylation of 1380 proteins were also evaluated by Full Moon kinase array to discover the protein associated Zn−induced apoptosis. From the results, a prominent increase in glycerophosphocholine and fatty acids was observed after Zn ion treatment, but only arachidonic acid was shown to induce apoptosis. The kinase array revealed that the phosphorylation of p53, GTPase activation protein, CaMK2a, PPAR−γ, and PLA−2 was changed. From the pathway analysis, metabolic changes showed earlier onset than protein signaling, which were related to choline metabolism. LC−MS analysis was used to quantify the intracellular choline concentration, which decreased after Zn treatment, which may be related to the choline consumption required to produce choline-containing metabolites. Plants produce numerous secondary metabolites with diverse physicochemical properties. Because different parts of a single plant produce various components, several spectroscopic methods are necessary to inspect their chemical profiles. Mass spectral data are recognized as one of the most useful tools for analyzing components with a wide range of polarities. However, interpreting mass spectral data generated from positive and negative ionization modes is a challenging task because of the diverse chemical profiles of secondary metabolites. Herein, we combine and analyze mass spectral data generated in two ionization modes to detect as many metabolites as possible using the molecular networking approach. We selected different parts of a single plant, Morus alba (Moraceae), which are used in the functional food and medicinal herb industries. The mass spectral data generated from two ionization modes were combined and analyzed using various molecular networking workflows. In this study, the general impact of high-resolution moving nesting domains on tropical cyclone (TC) intensity and track forecasts was verified, for a total of 107 forecast cases of 33 TCs, using the Weather Research and Forecasting (WRF) model. The experiment, with a coarse resolution of 12 km, could not significantly capture the intensification process, especially for maximum intensities (>60 m s−1). The intense TCs were better predicted by experiments using a moving nesting domain with a horizontal resolution of 4 km. The forecast errors for maximum wind speed and minimum sea-level pressure decreased in the experiment with higher resolution; the forecast of lifetime maximum intensity was improved. For the track forecast, the experiment with a coarser resolution tended to simulate TC tracks deviating rightward to the TC motions in the best-track data; this erroneous deflection was reduced in the experiment with a higher resolution. In particular, the track forecast in the experiment with a higher resolution improved more frequently for intense TCs that were generally distributed at relatively lower latitudes among the test cases. The sensitivity of the track forecast to the model resolution was relatively significant for lower-latitude TCs. On the other hand, the track forecasts of TCs moving to the mid-latitudes, which were primarily influenced by large-scale features, were not sensitive to the resolution. [ABSTRACT FROM AUTHOR]

Surface imprinting used for template recognition in nanocavities can be controlled and improved by surface morphological changes. Generally, the lithographic technique is used for surface patterning concerning sensing signal amplification in molecularly imprinted polymer (MIP) thin films. In this paper, we describe the effects of silanized silica molds on sensing the properties of MIP films. Porous imprinted poly(MAA–co–EGDMA) films were lithographically fabricated using silanized or non-treated normal silica replica molds to detect 2,4-dichlorophenoxyacetic acid (2,4-D) herbicide as the standard template. The silanized mold MIP film (st-MIP) (Δf = −1021 Hz) exhibited a better sensing response than the non-treated normal MIP (n-MIP) (Δf = −978 Hz) because the imprinting effects, which occurred via functional groups on the silica surface, could be reduced through silane modification. Particularly, two non-imprinted (NIP) films (st-NIP and n-NIP) exhibited significantly different sensing responses. The st-NIP (Δfst-NIP = −332 Hz) films exhibited lower Δf values than the n-NIP film (Δfn-NIP = −610 Hz) owing to the remarkably reduced functionality against nonspecific adsorption. This phenomenon led to different imprinting factor (IF) values for the two MIP films (IFst-MIP = 3.38 and IFn-MIP = 1.86), which was calculated from the adsorbed 2,4-D mass per poly(MAA–co–EGDMA) unit weight (i.e., QMIP/QNIP). Moreover, it was found that the st-MIP film had better selectivity than the n-MIP film based on the sensing response of analogous herbicide solutions. As a result, it was revealed that the patterned molds' chemical surface modification, which controls the surface functionality of imprinted films during photopolymerization, plays a role in fabricating enhanced sensing properties in patterned MIP films. Rhizofiltration experiments were conducted using uranium-contaminated groundwater and lettuce (Lactuca sativa), Chinese cabbage (Brassica campestris L.), radish (Raphanus sativus L.), and buttercup (Oenanthe javanica), which are commonly grown and consumed in South Korea. The results of the rhizofiltration experiments with artificial solutions with different initial uranium concentrations (18, 32, 84, 116, 173, and 263 μg/L) show that the uranium accumulation and bioconcentration factor (BCF) of plant roots increase with increasing uranium concentration in the groundwater. Among the four plants, the uranium concentration in the roots of Raphanus sativus L. is 1215.8 μg/g dry weight, with a maximum BCF value of 2692.7. The BCF value of the artificial solutions with various pH values (pH 3, 5, 7, and 9) is the highest under acidic conditions (pH 3) for all four plants. The uranium BCF values based on different hydroponic conditions range from 170.5 to 11580.3 and the results are comparable with those of other studies using similar methods; the highest BCF value was determined for Brassica campestris L. at pH 3. The BCF values of Raphanus sativus L. after the rhizofiltration experiments with genuine groundwater contaminated with uranium are the highest among the four species; that is, 1684.7 and 1700.1 in Oesam-dong and Bugokdong groundwater samples with uranium concentrations of 83 and 173 μg/L, respectively. The results of the scanning electron microscope/electron dispersive X-ray spectroscope analyses show that uranium in contaminated groundwater is adsorbed as a solid phase on the root surface. These results demonstrate that Raphanus sativus L. has a high tolerance to high concentrations of uranium and low pH conditions and a remarkable potential for uranium accumulation. Fatty acid synthase (FAS) is a key enzyme involved in de novo lipogenesis that produces lipids that are necessary for cell growth and signal transduction, and it is known to be overexpressed, especially in cancer cells. Although lipid metabolism alteration is an important metabolic phenotype in cancer cells, the development of drugs targeting FAS to block lipid synthesis is hampered by the characteristics of cancer cells with metabolic flexibility leading to rapid adaptation and resistance. Therefore, to confirm the metabolic alterations at the cellular level during FAS inhibition, we treated LNCaP-LN3 prostate cancer cells with FAS inhibitors (Fasnall, GSK2194069, and TVB-3166). With untargeted metabolomics, we observed significant changes in a total of 56 metabolites in the drug-treated groups. Among the altered metabolites, 28 metabolites were significantly changed in all of the drug-treated groups. To our surprise, despite the inhibition of FAS, which is involved in palmitate production, the cells increase their fatty acids and glycerophospholipids contents endogenously. Also, some of the notable changes in the metabolic pathways include polyamine metabolism and energy metabolism. This is the first study to compare and elucidate the effect of FAS inhibition on cellular metabolic flexibility using three different FAS inhibitors through metabolomics. This study successfully fabricated BPA-imprinted poly(4-vinylpyridine-co-ethylene glycol dimethacrylate) (poly(4-VP-co-EGDMA)) quartz crystal microbalance (MIP-QCM) sensors on a silica skeleton surface and gold pinholes of silica inverse opal through surface-initiated atom transfer radical polymerization (SI-ATRP). The sensing features of the two MIP films on the structured silica surface and nano-scale local gold surface were investigated by measuring the resonant frequency change (∆f) in QCM sensors. The ∆f values for the p-MIP (MIP on gold pinholes) and s-MIP films (MIP on silica skeleton surface) were obtained with the ∆f value of −199 ± 4.9 Hz and −376 ± 19.1 Hz, respectively, whereas for p-/s-NIP films, the ∆f values were observed to be −115 ± 19.2 Hz and −174 ± 5.8 Hz by the influence of non-specific adsorption on the surface of the films. Additionally, the imprinting factor (IF) appeared to be 1.72 for p-MIP film and 2.15 for s-MIP film, and the limits of quantitation (LOQ) and detection (LOD) were 54.924 and 18.125 nM (p-MIP film) and 38.419 and 12.678 nM (s-MIP film), respectively. Using the Freundlich isotherm model, the binding affinity of the BPA-imprinted films was evaluated. This was measured in an aqueous solution of BPA whose concentration ranged between 45 and 225 nM. It was found that the p-MIP film (m = 0.39) was relatively more heterogeneous than the s-MIP film (m = 0.33), both of which were obtained from the slope of the linear regressions. Finally, the selectivity of the MIP-QCM sensors for BPA detection was determined by measuring the effect of other analogous chemicals, such as bisphenol F (BPF), bisphenol AP (BPAP), and bisphenol B (BPB), in aqueous solutions. The selectivity coefficients (k*) of the two MIP films had ~1.9 for the p-MIP and ~2.3 for the s-MIP films, respectively. The results reveal that, with respect to signal amplification of the QCM sensors, the s-MIP film has better sensing features and faster detection responses than the p-MIP film. Deubiquitinating enzymes regulate various cellular processes, particularly protein degradation, localization, and protein–protein interactions. The dysregulation of deubiquitinating enzyme (DUB) activity has been linked to several diseases; however, the function of many DUBs has not been identified. Therefore, the development of methods to assess DUB activity is important to identify novel DUBs, characterize DUB selectivity, and profile dynamic DUB substrates. Here, we review various methods of evaluating DUB activity using cell lysates or purified DUBs, as well as the types of probes used in these methods. In addition, we introduce some techniques that can deliver DUB probes into the cells and cell-permeable activity-based probes to directly visualize and quantify DUB activity in live cells. This review could contribute to the development of DUB inhibitors by providing important information on the characteristics and applications of various probes used to evaluate and detect DUB activity in vitro and in vivo. Patterning of surfaces with a simple strategy provides insights into the functional interfaces by suitable modification of the surface by novel techniques. Especially, highly ordered structural topographies and chemical features from the wide range of interfaces have been considered as important characteristics to understand the complex relationship between the surface chemistries and biological systems. Here, we report a simple fabrication method to create patterned surfaces over large areas using evaporative self-assembly that is designed to produce a sacrificial template and lithographic etch masks of polymeric stripe patterns, ranging from micrometer to nanoscale. By facilitating a roll-on-plate geometry, the periodically patterned surface structures formed by repetitive slip-stick motions were thoroughly examined to be used for the deposition of the Au nanoparticles decorated graphene oxide (i.e., AuNPs, ~21 nm) and the formation of conductive graphene channels. The fluorescently labeled thiol-modified DNA was applied on the patterned arrays of graphene oxide (GO)/AuNPs, and biotin-streptavidin sensitive devices built with graphene-based transistors (GFETs, effective mobility of ~320 cm2 V−1 s−1) were demonstrated as examples of the platform for the next-generation biosensors with the high sensing response up to ~1 nM of target analyte (i.e., streptavidin). Our strategy suggests that the stripe patterned arrays of polymer films as sacrificial templates can be a simple route to creating highly sensitive biointerfaces and highlighting the development of new chemically patterned surfaces composed of graphene-based nanomaterials. Ultrasound stimulation (US) is reported to be a safe and useful technology for improving injured nerve regeneration. However, the intracellular mechanisms underlying its stimulatory effects are only partially understood. Mammalian target of rapamycin (mTOR) signaling is involved in neuronal survival and axonal outgrowth. In this study, we investigated the effect of US on regeneration of injured dorsal root ganglion (DRG) neurons and activation of the mTOR pathway. We showed that US significantly increased neurite regeneration and enhanced mTOR activation. Moreover, the expression of growth-associated protein-43 (GAP-43), a crucial factor for axonal outgrowth and regeneration in neurons, was significantly increased by US. Hydrogen chloride (HCl) gas is highly toxic to the human body. Therefore, HCl gas detection sensors should be installed at workplaces where trace HCl gas is continuously generated. Even though various polymer-based HCl-gas-sensing films have been developed, simpler and novel sensing platforms should be developed to ensure the cost effectiveness and reusability of the sensing platforms. Therefore, we present a simple strategy to fabricate reusable HCl-gas-sensing platforms using aminated polystyrene (a-PS) colloids and investigate their sensitivity, reusability, and selectivity using a quartz crystal microbalance (QCM). The reusable a-PS(1.0) colloidal sensor with a high degree of amination (DA) exhibited the highest binding capacity (102 μg/mg) based on the frequency change (Δf) during the HCl gas adsorption process. Further, its sensitivity and limit of detection (LOD) were 3.88 Hz/ppm and 5.002 ppm, respectively, at a low HCl gas concentration (<10 ppm). In addition, the sensitivity coefficient (k*) of the a-PS(1.0) colloid sensor with respect to HCHO was higher than that in the case of HF because of the lower binding affinity of the former with the a-PS(1.0) colloids. Based on these results, highly sensitive and reproducible a-PS colloids could be reused as an HCl-gas-sensing platform and used as an HCl sorbent in a gas column filter. [ABSTRACT FROM AUTHOR]

The carbonation of recycled aggregate was accelerated by sparging with supercritical carbon dioxide (scCO2) to reduce the amount of time needed for carbonation, which is necessary for the pH neutralization of recycled aggregate. To accelerate the carbonation process, pressurized scCO2 was sparged into two different types of recycled aggregates immersed in water for 1 h, followed by standstill for 2 h (in total, a 3 h treatment process). The reduction of the pH of the treated aggregates due to carbonation was investigated using batch extraction experiments. A continuous column extraction experiment for the scCO2-sparged recycled aggregate was also performed to identify the effect of pH reduction under the condition of non-equilibrium reaction. From XRD, SEM/EDS, and TG/DTA analyses, much of the portlandite in the recycled aggregates was consumed. In its place, calcite was created as a secondary mineral during only 3 h of treatment (1 h scCO2 sparging and 2 h stationing), indicating satisfactory carbonation of the aggregate. The results of the batch extraction experiments for both of the two recycled aggregate types also showed that the average pH of scCO2-sparged aggregate decreased from 12.0 to <9.8 (the tolerance limit for recycling). The pH of the eluent from the column packed with the scCO2-sparged aggregate also remained as <9.8, suggesting that a 1 h scCO2 sparging process is sufficient to carbonate waste concrete aggregate and to create an alternative construction material resource. p53 is activated in response to cellular stresses such as DNA damage, oxidative stress, and especially ribosomal stress. Although the regulations of p53 by E3 ligase and deubiquitinating enzymes (DUBs) have been described, the cellular roles of DUB associated with ribosomal stress have not been well studied. In this study, we report that Ubiquitin Specific Protease 47 (USP47) functions as an important regulator of p53. We show that ubiquitinated ribosomal protein S2 (RPS2) by Mouse double minute 2 homolog (MDM2) is deubiquitinated by USP47. USP47 inhibits the interaction between RPS2 and MDM2 thereby alleviating RPS2-mediated suppression of MDM2 under normal conditions. However, dissociation of USP47 leads to RPS2 binding to MDM2, which is required for the suppression of MDM2, consequently inducing up-regulation of the p53 level under ribosomal stress. Finally, we show that depletion of USP47 induces p53 and therefore inhibits cell proliferation, colony formation, and tumor progression in cancer cell lines and a mouse xenograft model. Mitosis is a complex and dynamic process that is tightly regulated by a large number of mitotic proteins. Dysregulation of these proteins can generate daughter cells that exhibit genomic instability and aneuploidy, and such cells can transform into tumorigenic cells. Thus, it is important for faithful mitotic progression to regulate mitotic proteins at specific locations in the cells at a given time in each phase of mitosis. Ubiquitin-dependent modifications play critical roles in this process by regulating the degradation, translocation, or signal transduction of mitotic proteins. Here, we review how ubiquitination and deubiquitination regulate the progression of mitosis. In addition, we summarize the substrates and roles of some deubiquitinating enzymes (DUBs) crucial for mitosis and describe how they contribute error correction during mitosis and control the transition between the mitotic phases. Laboratory experiments were performed to measure the supercritical CO2 (scCO2) storage ratio (%) of conglomerate and sandstone in the Janggi Basin, which are classified as rock in Korea that are available for CO2 storage. The scCO2 storage capacity was evaluated by direct measurement of the amount of scCO2 replacing the pore water in each reservoir rock core. The scCO2 sealing capacity of the cap rock (i.e., tuff and mudstone) was also compared by measuring the scCO2 capillary entry pressure (Δp) into the rock core. The measured average scCO2 storage ratio of the conglomerate and the sandstone were 30.7% and 13.1%, respectively, suggesting that the scCO2 storage capacity was greater than 360,000 metric tons. The scCO2 capillary entry pressure for the tuff ranged from 15 to 20 bar and for the mudstone it was higher than 150 bar, suggesting that the mudstone layers had enough sealing capacity from the aspect of hydromechanics. From XRF analyses, before and after 90 d of the scCO2-water-cap rock reaction, the mudstone and the tuff were investigated to assure their geochemical stability as the cap rock.
