Total laboratory automation (TLA) is a valuable component of microbiology laboratories and a growing number of publications suggest the potential impact of automation in terms of analysis standardization, streaking quality, and the turnaround time (TAT). The aim of this project was to perform a detailed investigation of the impact of TLA on the workflow of commonly treated specimens such as urine. This is a retrospective observational study comparing two time periods (pre TLA versus post TLA) for urine specimen culture processing. A total of 35,864 urine specimens were plated during the pre-TLA period and 47,283 were plated during the post-TLA period. The median time from streaking to identification decreased from 22.3 h pre TLA to 21.4 h post TLA (p < 0.001), and the median time from streaking to final validation of the report decreased from 24.3 h pre TLA to 23 h post TLA (p < 0.001). Further analysis revealed that the observed differences in TAT were mainly driven by the contaminated and positive samples. Our findings demonstrate that TLA has the potential to decrease turnaround times of samples in a laboratory. Nevertheless, changes in laboratory workflow (such as extended opening hours for plate reading and antibiotic susceptibility testing or decreased incubation times) might further maximize the efficiency of TLA and optimize TATs. The growing interest in low-impact mobility technologies has elevated the significance of fuel cell electric vehicles (FCEVs) in the automotive sector. Given the complexity of the resulting powertrain, the need for an effective energy management strategy (EMS) becomes essential to optimize efficiency and energy consumption in vehicles with diverse energy sources. Model-based control is the main approach to address the EMS in electrified vehicles. In particular, fuel cell power is commonly represented through a 1D look-up table using the current demand as input to simplify the implementation in a vehicle control unit. Uncertainties that may be implemented in maps due to simplifying hypotheses, dynamics, ageing, etc., can be propagated to powertrain control, motivating the adoption of adaptive look-up tables for FC modelling. In this study, an extended Kalman filter (EKF) is proposed to adapt the look-up table to actual FC behaviour by measuring its power and gradually correcting calibration errors, drift, and ageing. Subsequently, a standard equivalent consumption minimization strategy (ECMS) is employed to control the FCEV. The fuel cell model is calibrated with experimental data from an FCEV. The results demonstrate that the adaptive strategy outperforms the base calibration. Following an extensive simulation campaign, an improvement of 1.1% in fuel consumption was observed. Remarkably, after just one hour of operation, there was a notable 85% reduction in fuel cell power estimation error, even when the EMS was initially fed a biased look-up table. This paper highlights the importance of accurately modeling the operational constraints of Combined-Cycle Gas Turbines (CCGTs) within a unit-commitment framework. In practice, in Colombia, when given an initial dispatch by the Independent System Operator, CCGT plants are operated according to the results of heuristic simulation codes. Such heuristics often omit technical operating constraints, including hot, warm, or cold startup ramps; the minimum operation hours required for a gas turbine to start a steam turbine; the relationship between the dispatched number of steam and gas turbines; the load distribution among gas turbines; and supplementary fires. Most unit-commitment models in the literature represent standard technical constraints like startup, shutdown, up/down ramps, and in some cases, supplementary fires. However, they typically overlook other real-life CCGT operating constraints, which were considered in this work. These constraints are crucial in integrated energy systems to avoid equipment damage, which can potentially put CCGT plants out of service and ultimately lead to lower operating costs. This study investigates a low-temperature three-circuit cooling system for a 55 kW industrial electric motor. The cooling system provides an increase of the power-to-dimension ratio by 63%, together with an improvement in motor performance. The three-circuit cooling system includes water cooling of the housing and stator and air-cooling of the motor's interior. The test results show that the motor efficiency was maintained in the range between 92.5 and 94.5%, with respect to the motor's power. With power increases up to 90 kW, a winding temperature of 67 °C was observed during three hours of operation. This advancement is particularly valuable for vehicles, ships, and aircraft applications, where maximizing power within limited space is crucial. An analysis of the experimental data showed that the cooling system operates at an average efficiency of 79.2%, indicating that roughly 20% of heat was accumulated in the rotor. This leads to a gradual temperature rise, particularly in the rotor, posing a risk of overheating and failure during motor overloads above 90 kW. Enhancing the cooling efficiency within the motor's interior can be achieved by incorporating extra heat exchangers, implementing evaporative heat transfer, and employing water-cooling circuits at lower temperatures. When a tribological system is operated for the first time, the first hours of operation are of the utmost importance as the surfaces of the contacting elements mate to each other, involving significant wear processes until the surfaces reach stable topological characteristics. This initial phase in a combustion engine is known as running-in and is of critical importance in the study of friction and wear phenomena. Despite this, there is little information in the literature dedicated to running-in and, therefore, data improving its understanding is greatly anticipated. In this work, a novel wear debris detection system measuring in real time was employed to investigate the running-in of an inline 4-cylinder gasoline engine; it consists primarily of an optical sensor with the capability of detecting very small particles from 4 μ m. For the tests, the engine was mounted on a test bench and operated under stationary working conditions. The results of the wear debris measurements showed interesting insights into the engine running-in process; although in general, the results followed the expected trend, they also showed an unexpected behavior: it was expected to obtain the highest amounts of wear debris at the beginning of the running-in, but instead the number of debris stagnated and only started to increase after about 20 h to then decrease again. The best operating conditions to run-in the engine were identified at the middle of the running-in period, without the presence of large wear debris that could lead to severe wear. Finally, it was found that the engine running-in was not finished until at least 75 h of operation, although commonly, a running-in time of 10 h is used in the industry. [ABSTRACT FROM AUTHOR]

The purpose of this study is to address two major issues: (1) the spread of epidemics such as COVID-19 due to long waiting times caused by a large number of waiting for customers, and (2) excessive energy consumption resulting from the elevator patterns used by various customers. The first issue is addressed through the development of a mobile application, while the second issue is tackled by implementing two strategies: (1) determining optimal stopping strategies for elevators based on registered passengers and (2) assigning passengers to elevators in a way that minimizes the number of floors the elevators need to stop at. The mobile application serves as an input parameter for the optimization toolbox, which employs the exact method and multi-objective variable neighborhood strategy adaptive search (M-VaNSAS) to find the optimal plan for passenger assignment and elevator scheduling. The proposed method, which adopts an even-odd floor strategy, outperforms the currently practiced procedure and leads to a 42.44% reduction in waiting time and a 29.61% reduction in energy consumption. The UK, as elsewhere, has seen an accelerating trend of bank branch closures and reduced opening hours since the early 2000s. The reasons given by the banks are well rehearsed, but the impact assessments they provide to justify such programs and signpost alternatives have been widely criticized as being inadequate. This is particularly so for vulnerable customers dependent on financial services who may face difficulties in accessing remaining branches. There is a need whilst analyzing spatial patterns of access to also include temporal availability in relation to transport opportunities. Drawing on a case study of potential multi-modal accessibility to banks in Wales, we demonstrate how open-source tools can be used to examine patterns of access whilst considering the business operating hours of branches in relation to public transport schedules. The inclusion of public and private travel modes provides insights into access that are often overlooked by a consideration of service-side measures alone. Furthermore, findings from the types of tools developed in this study are illustrative of the additional information that could be included in holistic impact assessments, allowing the consequences of decisions being taken to close or reduce the operating hours of bank branches to be more clearly communicated to customers. The number of microgrids within a smart distribution grid can be raised in the future. Microgrid-based distribution network reconfiguration is analyzed in this research by taking demand response programs and power-sharing into account to optimize costs and reduce power losses. The suggested method determined the ideal distribution network configuration to fulfil the best scheduling goals. The ideal way of interconnecting switches between microgrids and the main grid was also identified. For each hour of operation, the ideal topology of microgrid-based distribution networks was determined using optimal power flow. The results were produced with and without the use of a demand response program and power-sharing in each microgrid. Different load profiles, such as residential, industrial, commercial, and academic, were taken into account and modified using appropriate demand response programs and power-sharing using the Artificial Bee Colony algorithm. Various scenarios were explored independently to suit the diverse aims considered by the distribution network operator for improved observation. The ABC optimization in this research attempted to reduce the system's total operation costs and power losses through efficient networked microgrid reconfiguration. The results of optimal microgrid topology revealed the effects of power-sharing and demand response (TOU) programs. The results obtained in the proposed idea shows that costs were reduced by 8.3% and power losses were reduced by 4%. Digital location traces are a relevant source of insights into how citizens experience their cities. Previous works using call detail records (CDRs) tend to focus on modeling the spatial and temporal patterns of human mobility, not paying much attention to the semantics of places, thus failing to model and enhance the understanding of the motivations behind people's mobility. In this paper, we applied a methodology for identifying individual users' routine locations and propose an approach for attaching semantic meaning to these locations. Specifically, we used circular sectors that correspond to cellular antennas' signal areas. In those areas, we found that all contained points of interest (POIs), extracted their most important attributes (opening hours, check-ins, category) and incorporated them into the classification. We conducted experiments with real-world data from Coimbra, Portugal, and the initial experimental results demonstrate the effectiveness of the proposed methodology to infer activities in the user's routine areas. The heating load of intermittent heating is not always lower than that of continuous heating for heat storage and release of internal walls. Therefore, the threshold value of daily operation hours exists, and is affected by the configuration of internal walls. A comparative study is performed between continuous and intermittent heating modes to investigate the threshold value of daily operation hours for different internal wall configurations by employing computational fluid dynamic (CFD) models. Meanwhile, field tests on the temperature distribution within a thermal mass was carried out to validate the simulation. The results show that the heating load index of intermittent heating is larger than that of continuous heating with increased amplitude ranging from 31.58% to 152.63%. The threshold value of daily operation hours is, respectively, 18.04 h, 15.80 h, 14.59 h, and 13.46 h for four internal wall configurations. Moreover, with the increase in the insulation level of internal walls, the threshold value of daily operation hours decreases. For thermal power generation, the natural draft hybrid cooling system (NDHCs) with airflows in parallel design gives a multi-objective solution for water saving, performance enhancement and maintenance issues, like corrosion, by switching the loads of wet and dry sections. Performances of dry and wet sections interact with each other in the highly integrated system, increasing the complexity of operation strategies. In this context the present paper examines eight different operation schemes to reveal the relationships of ambient conditions and operation schemes. Comprehensive comparisons in the view of cooling efficiency with a same water inlet temperature are conducted firstly. Results show that there exists energy-saving potentials of the water evaporated rate, cooling performances and the pump power for different schemes. Based on the practical boundary conditions, including those of weather data, operation hours and market factors, optimal operation strategies of hybrid cooling are designed to minimize the operation costs of the energy system. For the 660 MW power generating unit integrated with a natural draft dry cooling system (NDDCs), operation costs based on NDHC after optimization decreased about 0.8% in 2010 and 0.35% in 2018 compared with that of the basic system. Toward further implementation of continuous tablet manufacturing, one key issue is the time needed for start-up operation because it could lead to lower product yield and reduced economic performance. The behavior of the start-up operation is not well understood; moreover, the definition of the start-up time is still unclear. This work investigates the effects of process parameters on the start-up operation in continuous wet granulation, which is a critical unit operation in solid drug manufacturing. The profiles of torque and granule size distribution were monitored and measured for the first hour of operation, including the start-up phase. We analyzed the impact of process parameters based on design of experiments and performed an economic assessment to see the effects of the start-up operation. The torque profiles indicated that liquid-to-solid ratio and screw speed would affect the start-up operation, whereas different start-up behavior resulted in different granule size. Depending on the indicator used to define the start-up operation, the economic optimal point was significantly different. The results of this study stress that the start-up time differs according to the process parameters and used definition, e.g., indicators and criteria. The growth in renewable energy integration over the past few years, primarily fueled by the drop in capital cost, has revealed the requirement for more sustainable methods of integration. This paper presents a collocated hybrid plant consisting of solar photovoltaic (PV) and Ternary pumped-storage hydro (TPSH) and designs controls that integrate the PV plant such that the behavior and the controllability of the hybrid plant are similar to those of a conventional plant within operational constraints. The PV array control and hybrid plant control implement a neural–network-based framework to coordinate the response, de-loading, and curtailment of multiple arrays with the response of the TPSH. With the help of the designed controls, a symbiotic relationship is developed between the two energy resources, where the PV compensates for the TPSH nonlinearities and provides required speed of response, while the TPSH firms the PV system and allows the PV to be integrated using its existing infrastructure. Simulations demonstrate that the designed controls enable the PV system to track references, while the TPSH's firming and shifting transforms the PV system into a base load plant for most of the day and extends its hours of operation. The work presented in this paper aims to show how modern wind turbines can help to control the frequency in a small grid which suffers from large power imbalances. It is shown for an exemplary situation, which occurred in Flensburg's distribution grid in 2019: a major blackout, which occurred after almost two hours in islanding operation, affecting almost the entire distribution grid, which supplies approximately 55,000 households and businesses. For the analysis, a wind turbine model and a grid support controller developed at the Wind Energy Technology Institute are combined with real measurements from the day of the blackout to generate a fictional yet realistic case study for such an islanding situation. For this case study, it is assumed that wind turbines with grid support functionalities are connected to the medium voltage distribution grid of the city. It is shown to what extent wind turbines can help to operate the grid by providing grid frequency support in two ways: By supplying synthetic inertia only, where the wind turbines can help to limit the rate of change of frequency in the islanded grid directly after losing the connection to the central European grid. In combination with the primary frequency control capabilities of the wind turbines (WTs), the disconnection of one gen set in the local power station might have been avoided. Furthermore, wind turbines with primary frequency control capabilities could have restored the grid frequency to 50 Hz shortly after the islanding situation even if the aforementioned gen-set was lost. Wind energy siting to minimize impacts to bats would benefit from impact predictions following pre-construction surveys, but whether pre- or even post-construction activity patterns can predict fatalities remains unknown. We tested whether bat passage rates through rotor-swept airspace differ between groups of wind turbines where bat fatalities were found and not found during next-morning dog searches for fatalities. Passage rates differed significantly and averaged four times higher where freshly killed bats were found in next-morning fatality searches. Rates of near misses and risky flight behaviors also differed significantly between groups of turbines where bats were found and not found, and rate of near misses averaged eight times higher where bat fatalities were found in next-morning searches. Hours of turbine operation averaged significantly higher, winds averaged more westerly, and the moon averaged more visible among turbines where and when bat fatalities were found. Although dogs found only one of four bats seen colliding with turbine blades, they found many more bat fatalities than did human-only searchers at the same wind projects, and our fatality estimates were considerably higher. Our rates of observed bat collisions, adjusted for the rates of unseen collisions, would predict four to seven times the fresh fatalities we found using dogs between two wind projects. Despite markedly improved carcass detection through use of dogs, best estimates of bat fatalities might still be biased low due to crippling bias and search radius bias. [ABSTRACT FROM AUTHOR]

This review focuses on the most suitable form of hydrodynamic modeling for the next generation wave energy converter (WEC) design tools. To design and optimize a WEC, it is estimated that several million hours of operation must be simulated, perhaps one million hours of WEC simulation per year of the R&D program. This level of coverage is possible with linear potential flow (LPF) models, but the fidelity of the physics included is not adequate. Conversely, while Reynolds averaged Navier–Stokes (RANS) type computational fluid dynamics (CFD) solvers provide a high fidelity representation of the physics, the increased computational burden of these models renders the required amount of simulations infeasible. To scope the fast, high fidelity options, the present literature review aims to focus on what CFD theories exist intermediate to LPF and RANS as well as other modeling options that are computationally fast while retaining higher fidelity than LPF. Membrane distillation (MD) is fast evolving as a desalination technology for high-salinity waters where scaling remains a major challenge. This paper reports the scaling reduction in carbon nanotube-immobilized membranes (CNIMs) and by the use of the antiscalant polyacrylic acid. High concentrations of CaSO4, CaCO3, and BaSO4 were deliberately used to initiate scaling on the membranes. It was observed that after ten hours of operation in a highly scaling CaSO4 environment, the CNIM showed 127% higher flux than what was observed on a membrane without the CNTs. The trends were similar with CaCO3 and BaSO4, where the CNIM showed significantly improved antiscaling behavior. The normalized flux declination for CNIM was found to be 45%, 30%, and 53% lower compared to the pristine membrane with CaSO4, CaCO3, and BaSO4 solutions, respectively. The use of antiscalant in the feed solution was also found to be effective in improving antiscaling behavior, which reduced salt deposition up to 28%, and the water vapor flux was 100% and 18% higher for the pristine polypropylene and CNIM, respectively. Results also showed that the presence of CNTs facilitated the removal of deposited salts by washing, and the CNIM regained 97% of its initial water flux, whereas the polypropylene only regained 85% of the original value. Separating the particles from the liquid component of sample solutions is important for several microfluidic-based sample preparations and/or sample handling techniques, such as plasma separation from whole blood, sheath-free flow focusing, particle enrichment etc. This paper presents a microfluidic in-flow decantation technique that provides the separation of particles from particle-free fluid while in-flow. The design involves the expansion of sample fluid channel in lateral and depth directions, thereby producing a particle-free layer towards the walls of the channel, followed by gradual extraction of this particle-free fluid through a series of tiny openings located towards one-end of the depth-direction. The latter part of this design is quite crucial in the functionality of this decantation technique and is based on the principle called wee-extraction. The design, theory, and simulations were presented to explain the principle-of-operation. To demonstrate the proof-of-principle, the experimental characterization was performed on beads, platelets, and blood samples at various hematocrits (2.5%–45%). The experiments revealed clog-free separation of particle-free fluid for at least an hour of operation of the device and demonstrated purities close to 100% and yields as high as 14%.
