The paper expands the finite difference method to the complex plane, and thus obtains an improvement in the resolution of differential equations with an increase in numerical precision and a generalization in the mathematical modeling of problems. The article begins with a selection of the best techniques for obtaining finite difference coefficients for approximating derivatives in the real domain. Then, the calculation is expanded to the complex domain. The research expands forward, backward, and central difference approximations of the real case by a quadrant approximation in the complex plane, which facilitates the use in boundary conditions of differential equations. The article shows many real and complex finite difference equations with their respective order of error, intended to serve as a basis and reference, which have been tested in practical examples of solving differential equations used in engineering. Finally, a comparison is made between the real and complex techniques of finite difference methods applied in the Theory of Elasticity. As a surprising result, the article shows that the finite difference method has great advantages in numerical precision, diversity of formulas, and modeling generalities in the complex domain when compared to the real domain. The aim of this study was to verify the accuracy of predicting oxygen consumption (O2) in predominantly aerobic activities based on net heart rate (netHR), sex, and body mass index (BMI) in active adults. NetHR is the value of the difference between the resting HR (HRrest) and the average HR value obtained during a given session or period of physical activity. These activities must be continuous, submaximal, and of a stabilized intensity. The magnitude of the netHR depends mainly on the intensity of the exercise. The HR is measured in beats per minute (bpm). A total of 156 participants, 52 women and 104 men, between the ages of 18 and 81, had their netHR and net oxygen intake (netVO2) assessed. There were 79 participants in group 1 (prediction sample) (52 males and 27 females). There were 77 people in group 2 (validation sample) (52 males and 25 females). The results of the multiple linear regression showed that netVO2 (R2 = 85.2%, SEE = 3.38) could be significantly predicted by sex (p < 0.001), netHR (p < 0.001), and BMI (p < 0.001). The Bland–Altman plots satisfied the agreement requirements, and the comparison of the measured and estimated netVO2 revealed non-significant differences with a trivial effect size. We calculated the formula NetVO2 (mL/(kg·min)) = 16 + 3.67 (sex) + 0.27 (netHR) − 0.57 (BMI) to predict netVO2, where netVO2 is the amount of oxygen uptake (mL/(kg·min)) above the resting value, netHR is the heart rate (beats per minute) above the resting value measured during exercise, sex is equal to zero for women and one for men, and BMI is the body mass index. In addition, based on the knowledge of VO2, it was possible to estimate the energy expenditure from a particular training session, and to determine or prescribe the exercise intensity in MET (metabolic equivalent of task). Different training intensity distributions (TIDs) have been proposed to improve cycling performance, especially for high-competition athletes. The objectives of this study were to analyze the effect of a 16-week pyramidal training intensity distribution on somatic and power variables in recreational cyclists and to explore the training zone with the greatest impact on performance improvement. The sample consisted of 14 male recreational cyclists aged 41.00 ± 7.29 years of age. A number of somatic variables were measured. During an incremental protocol, power at a 4 mMol·L−1 blood lactate concentration (P4), corresponding power to body mass ratio (P/W P4), and heart rate (HR P4) were also measured. Among the somatic variables, the percentage of fat mass showed the greatest improvement between moments (p < 0.001, d = 0.52). Both P4 (p < 0.001, d = 1.21) and P/W P4 (p < 0.001, d = 1.54) presented a significant increase between moments. The relative improvement in P4 (% P4) showed a significant correlation (Rs = 0.661, p = 0.038) and relationship (R2 = 0.61, p = 0.008) mainly with training zone Z2 (blood lactate levels ≥ 2 and <4 mMol·L−1). Enhancing recovery is a fundamental component of high-performance sports training since it enables practitioners to potentiate physical performance and minimise the risk of injuries. Using a new sports legging embedded with an intelligent system for electrostimulation, localised heating and compression (completely embodied into the textile structures), we aimed to analyse acute recovery following a fatigue protocol. Surface electromyography- and torque-related variables were recorded on eight recreational athletes. A fatigue protocol conducted in an isokinetic dynamometer allowed us to examine isometric torque and consequent post-exercise acute recovery after using the sports legging. Regarding peak torque, no differences were found between post-fatigue and post-recovery assessments in any variable; however, pre-fatigue registered a 16% greater peak torque when compared with post-fatigue for localised heating and compression recovery methods. Our data are supported by recent meta-analyses indicating that individual recovery methods, such as localised heating, electrostimulation and compression, are not effective to recover from a fatiguing exercise. The aims of this study were (1) to evaluate the prevalence of type 2 diabetes (T2D) in a middle-aged north-eastern Portuguese population, (2) to analyze the prevalence of impaired fasting glucose (IFG), and (3) to assess the risk of T2D in this community-based sample. An exploratory, retrospective, and cross-sectional study was conducted from a total of 6570 individuals aged 18–102 years, among which 3865 were women (57.4 ± 18.1 years) and 2705 were men (60.0 ± 16.8 years). T2D diagnosis, IFG, and the diabetes risk score (low to very high risk) were assessed. The prevalence of T2D in this adult and an older north-eastern Portuguese population was 17.4%. A higher prevalence of T2D was reported in men (22.2%) than in women (14.0%); however, this was without significant differences (p = 0.086). Otherwise, the prevalence of T2D was significantly different among the age groups and increased with age (p < 0.001). Regarding IFG, a higher percentage of cases was observed in men (14.1%) than in women (8.4%) (p < 0.001). The risk of developing T2D in the next 10 years showed an association with sex and age group (p < 0.001) with a small-to-moderate effect (V = 0.1–0.3). Men and the elderly had the highest percentage of cases in the moderate-to-very high-risk bands. The current research confirmed a higher prevalence of T2D, IFG, and diabetes risk than previous Portuguese epidemiological reports. The results also suggest potential prediabetes cases, which should be carefully monitored. Arthur de Carvalho Drops® (ACD) is a traditional Brazilian herbal medicine used to treat functional gastrointestinal disorders (FGIDs). ACD is a formulation of herbal extracts from Matricaria recutita (chamomile), Foeniculum vulgare (fennel) and Gentiana lutea L. (gentian). Considering the popular use for FGIDs, the aim of this work was to investigate the ACD effect on gastric and intestinal parameters with emphasis in a mechanistic approach using isolated duodenal preparations of rodents. Analytical method was developed and validated for quantify three actives principles/markers (Apigenin-7-glucoside, gentiopicroside and anethole) in ACD. The treatment with ACD significantly reduced the emetogenic stimuli induced by cisplatin in rats, showed a laxative effect, reduced the bethanechol-enhanced gastrointestinal transit and completely reversed the contraction induced by carbachol in rat duodenum. However, ACD did not alter the secretory gastric volume or total gastric acidity. The ACD affect the contractions of duodenal smooth muscle mediated by Ca2+ channels and it is also able to inhibit the contractile response mediated by the release from its intracellular store. Furthermore, the relaxant effects of ACD appear independent of the nitric oxide pathway in rat duodenum. G-quadruplex (G4)-interactive small molecules have a wide range of potential applications, not only as drugs, but also as sensors of quadruplex structures. The purpose of this work is the synthesis of analogues of the bis-methylquinolinium-pyridine-2,6-dicarboxamide G4 ligand 360A, to identify relevant structure–activity relationships to apply to the design of other G4-interactive small molecules bearing bis-quinoline or bis-isoquinoline moieties. Thermal denaturation experiments revealed that non-methylated derivatives with a relative 1,4 position between the amide linker and the nitrogen of the quinoline ring are moderate G4 stabilizers, with a preference for the hybrid h-Telo G4, a 21-nt sequence present in human telomeres. Insertion of a positive charge upon methylation of quinoline/isoquinoline nitrogen increases compounds' ability to selectively stabilize G4s compared to duplex DNA, with a preference for parallel structures. Among these, compounds having a relative 1,3-position between the charged methylquinolinium/isoquinolinium nitrogen and the amide linker are the best G4 stabilizers. More interestingly, these ligands showed different capacities to selectively block DNA polymerization in a PCR-stop assay and to induce G4 conformation switches of hybrid h-Telo G4. Molecular dynamic simulations with the parallel G4 formed by a 21-nt sequence present in k-RAS gene promoter, showed that the relative spatial orientation of the two methylated quinoline/isoquinoline rings determines the ligands mode and strength of binding to G4s. Anadenanthera colubrina var. cebil (Griseb.) Altschul (Fabaceae family), commonly known as the red angico tree, is a medicinal plant found throughout Brazil's semi-arid area. In this study, a chemical analysis was performed to investigate the antidiarrheal activity and safety profile of red angico gum (RAG), a biopolymer extracted from the trunk exudate of A. colubrina. Upon FT-IR spectroscopy, RAG showed bands in the regions of 1608 cm−1, 1368 cm−1, and 1029 cm−1, which relate to the vibration of O–H water molecules, deformation vibration of C-O bands, and vibration of the polysaccharide C-O band, respectively, all of which are relevant to glycosidic bonds. The peak molar mass of RAG was 1.89 × 105 g/mol, with the zeta potential indicating electronegativity. RAG demonstrated high yield and solubility with a low degree of impurity. Pre-treatment with RAG reduced the total diarrheal stool and enteropooling. RAG also enhanced Na+/K+-ATPase activity and reduced gastrointestinal transit, and thereby inhibited intestinal smooth muscle contractions. Enzyme-Linked Immunosorbent Assay (ELISA) demonstrated that RAG can interact with GM1 receptors and can also reduce E. coli-induced diarrhea in vivo. Moreover, RAG did not induce any signs of toxicity in mice. These results suggest that RAG is a possible candidate for the treatment of diarrheal diseases. [ABSTRACT FROM AUTHOR]

Human epidermal growth factor 2 (HER2) is a ligand-free tyrosine kinase receptor of the HER family that is overexpressed in some of the most aggressive tumours. Although it is known that HER2 dimerization involves a specific region of its extracellular domain, the so-called "dimerization arm", the mechanism of dimerization inhibition remains uncertain. However, uncovering how antibody interactions lead to inhibition of HER2 dimerization is of key importance in understanding its role in tumour progression and therapy. Herein, we employed several computational modelling techniques for a molecular-level understanding of the interactions between HER and specific anti-HER2 antibodies, namely an antigen-binding (Fab) fragment (F0178) and a single-chain variable fragment from Trastuzumab (scFv). Specifically, we investigated the effects of antibody-HER2 interactions on the key residues of "dimerization arm" from molecular dynamics (MD) simulations of unbound HER (in a total of 1 µs), as well as ScFv:HER2 and F0178:HER2 complexes (for a total of 2.5 µs). A deep surface analysis of HER receptor revealed that the binding of specific anti-HER2 antibodies induced conformational changes both in the interfacial residues, which was expected, and in the ECDII (extracellular domain), in particular at the "dimerization arm", which is critical in establishing protein–protein interface (PPI) interactions. Our results support and advance the knowledge on the already described trastuzumab effect on blocking HER2 dimerization through synergistic inhibition and/or steric hindrance. Short-term load forecasting (STLF) plays a vital role in ensuring the safe, efficient, and economical operation of power systems. Accurate load forecasting provides numerous benefits for power suppliers, such as cost reduction, increased reliability, and informed decision-making. However, STLF is a complex task due to various factors, including non-linear trends, multiple seasonality, variable variance, and significant random interruptions in electricity demand time series. To address these challenges, advanced techniques and models are required. This study focuses on the development of an efficient short-term power load forecasting model using the random forest (RF) algorithm. RF combines regression trees through bagging and random subspace techniques to improve prediction accuracy and reduce model variability. The algorithm constructs a forest of trees using bootstrap samples and selects random feature subsets at each node to enhance diversity. Hyperparameters such as the number of trees, minimum sample leaf size, and maximum features for each split are tuned to optimize forecasting results. The proposed model was tested using historical hourly load data from four transformer substations supplying different campus areas of the University of Beira Interior, Portugal. The training data were from January 2018 to December 2021, while the data from 2022 were used for testing. The results demonstrate the effectiveness of the RF model in forecasting short-term hourly and one day ahead load and its potential to enhance decision-making processes in smart grid operations. Leveling the curve of Spee is a commonly-used strategy to correct deep bites. Although several techniques have been proposed to intrude mandibular incisors (MI), flaring of these teeth is often observed and in many instances undesired. A three-dimensional (3D) finite element model (FEM) was used to locate the ideal point of force application (PFA) to achieve pure MI intrusion with the three-piece arches' technique. It comprised (1) a 0.021 × 0.025 in. stainless steel (SS) wire that passively filled the slots of the canine and premolar brackets and the first and second molar tubes, bilaterally; (2) a 0.0215 × 0.0275 in. SS intrusion base arch (IBA) inserted into the MI brackets, that presented a step down distal to the lateral incisors brackets and a posterior extension arm; (3) titanium-molybdenum tip-back springs designed to apply the intrusion force, fitted inside the first molar gingival tube. Four PFA on the IBA were simulated (FEM 1, 2, 3, and 4). FEM 3 resulted in pure MI and was considered the ideal PFA. FEM1 and 2 showed intrusion and buccal crown flaring of the MI, whereas FEM4 resulted in intrusion and lingual crown flaring of those teeth. Clinicians may consider three-piece arch mechanics to achieve pure MI intrusion. However, they must be aware that when force was applied anteriorly or posteriorly to the ideal PFA, the incisors would incline labially or lingually, respectively. [ABSTRACT FROM AUTHOR]

Stocking densities were evaluated for the rearing tambaqui in an RAS system. Experiment 1 used juveniles weighing 0.54 g at the following densities for 15 days: D0.3—0.3; D0.6—0.6; and D0.9—0.9 kg m−3. Experiment 2 used juveniles weighing 8.22 g at the following densities for 75 days: D0.8—0.8; D1.2—1.2; and D1.6—1.6 kg m−3. Experiment 3 used juveniles weighing 142.18 g at the following densities for 75 days: D2—2; D4—4 and D6—6 kg m−3. In Experiment 1, density did not influence performance (p > 0.05), with the exception of biomass, which was greatest in D0.9 (p < 0.05). In Experiment 2, final weight, weight gain and daily weight gain were highest for D0.8 (p < 0.05), as was triglycerides (p < 0.05), whereas biomass increased with increasing density (p < 0.05). In Experiment 3, weight, weight gain and daily weight gain were greater for D2 and D4 (p < 0.05), while final biomass was lowest for D2 (p < 0.05). Hemoglobin was lower for D4 and D6 (p < 0.05), while cholesterol and glucose levels were higher for D2 and D6 (p < 0.05). This study examines the notion of governance while corruption and polity act in a negotiated approach. It adopts a theory synthesis approach to design the research paradigm and brings renewed attention to governance from a national perspective. This study argues that corruption and polity collectively define the state of governance in a particular country, which might offer some new insights to the remaining parts of the world. The principal aim of the study is to bring relevant evidence from the literature to develop a solid foundation on governance from a macro perspective. Deploying a qualitative approach, this study highlights available literature on corruption, polity, and their connections to define the state of governance. From this specific target, we have initiated this study deploying a conceptual fashion in exploring governance which is shaped by the interplay between two loosely connected themes: polity and corruption. The outcome of this synthesis is to renew our understanding on governance to strengthen the governance mechanism whereby corruption could be checked through sound polity in action. The arguments presented in the paper are expected to be useful for regulators and policymakers as they prepare governance-related rules, acts, or directives in their respective countries. This article investigates the extent to which the COVID-19 pandemic fostered significant shifts in election campaigning. The argument is that COVID-19 might have had an impact on both digitalization and professionalization, which might have been regarded as necessary strategies to curb the difficulties brought about by the pandemic. We apply a most similar systems design with a threefold comparative scheme in order to capture and isolate such effects in the campaigns preceding the 2021 Portuguese presidential elections, using data from campaign spending, campaign activities, and social media activity and impact. Results show that the pandemic crisis has not, generally speaking, brought about a higher level of digitalization of electoral campaigns, in spite of online events having become more common. On the contrary, while there were signs of feebler patterns of normalization of online competition in 2021 vis-à-vis 2016, namely in terms of engagement, normalization was stronger after the lockdown than before. Lastly, relative investment in professionalization was similar in 2016 and 2021, and the difference between the budgeted and the actual investment in 2021 cannot be attributed to the worsening of the pandemic situation or to the lockdown. In sum, we depict a scenario of remarkable stability of the electoral campaigns put forward by presidential candidates in terms of digitalization and professionalization. Food waste occurs from harvesting to consumption. Applying procedures and technologies, changing attitudes, and promoting awareness have positive social, economic, and environmental impacts that can contribute to reducing food waste. The paper presents a decision support system (DSS) to predict the quality evolution of fruits and vegetables, particularly of peaches, and estimate its commercialization period at the highest overall perceived quality by consumers, thus contributing to reducing food waste. The Fuzzy Logic DSS predicts the evolution of the physical-chemical parameters of peaches (hardness, soluble solids content, and acidity) depending on the cultivar (Royal Summer and Royal Time), storage time, and temperature. As the range of the values of these physical-chemical parameters of peaches that consumers perceive to be at their highest quality are known, the DSS predicts the marketable period in days. Case studies were developed to analyze the influence of each physical-chemical parameter on the commercialization days (number and time to start). It is concluded that temperature is the most important parameter for fruit conservation. A low value of conservation temperature allows for the significant extension of the time that peaches can be sold at the highest quality. Hardness is used to determine the harvest date since it is an index of fruit ripeness. The same conclusion is obtained for the influence of the soluble solids content. The influence of acidity on marketable days is less than the other physical-chemical parameters. This DSS helps retailers to sell their peaches at the highest quality with benefits for all parties. It also helps in the decision-making concerning the actions to take when fruits are reaching the end of their highest quality by predicting the range of the commercialization days. This formulation can be extended to other fruits and vegetables and in the last instance contribute to the reduction of food loss and waste, consequently promoting social, economic, and environmental aspects of our daily life. The present paper intends to provide an overview and a critical assessment of the administrative simplification policies implemented in Portugal over the past 20+ years. Throughout these decades, the major instruments for administrative simplification have been: (i) Decree-Law n. 135/99, of 22 April; (ii) the Simplex programme; (iii) the 2015 new Code of Administrative Procedure and its 2021 amendment; and (iv) the COVID-19 legislation. Although Decree-Law n. 135/99 (i) was the first attempt to specifically address simplification, it was a very thin one. The Simplex programme (ii), created in 2006, was the first cross-sectoral robust policy of simplification; its motto is "the simpler the better" and it encompassed reforms in all government areas aiming at cutting red tape, promoting administrative efficiency, and making the citizens' and the corporations' lives easier when dealing with State. The new Code of Administrative Procedure (iii), approved in 2015, brought about specific normative solutions to simplify procedures, namely, prior communications, administrative assistance, procedural conferences, and the electronic one-stop shop. Finally, the COVID-19 legislation (iv) was made necessary by the COVID-19 pandemic and by the fact that public health restrictions made it impossible for citizens to establish personal contact with the administration in many cases. The already enormous burden caused by Mycobacterium tuberculosis and Human Immunodeficiency Virus type 1 (HIV-1) alone is aggravated by co-infection. Despite obvious differences in the rate of evolution comparing these two human pathogens, genetic diversity plays an important role in the success of both. The extreme evolutionary dynamics of HIV-1 is in the basis of a robust capacity to evade immune responses, to generate drug-resistance and to diversify the population-level reservoir of M group viral subtypes. Compared to HIV-1 and other retroviruses, M. tuberculosis generates minute levels of genetic diversity within the host. However, emerging whole-genome sequencing data show that the M. tuberculosis complex contains at least nine human-adapted phylogenetic lineages. This level of genetic diversity results in differences in M. tuberculosis interactions with the host immune system, virulence and drug resistance propensity. In co-infected individuals, HIV-1 and M. tuberculosis are likely to co-colonize host cells. However, the evolutionary impact of the interaction between the host, the slowly evolving M. tuberculosis bacteria and the HIV-1 viral "mutant cloud" is poorly understood. These evolutionary dynamics, at the cellular niche of monocytes/macrophages, are also discussed and proposed as a relevant future research topic in the context of single-cell sequencing. Dams provide water supply, flood protection, and hydropower generation benefits, but also harm native species by altering the natural flow regime, and degrading the aquatic and riparian habitats. In the present study, which comprised the Douro River basin located in the North of Portugal, the cost-benefit assessment of dams was based upon a balance between the touristic benefits of a dammed Douro, and the ecological benefits of less fragmented Douro sub-catchments. Focused on four sub-catchments (Sabor, Tâmega, Côa and Corgo), a probabilistic stream connectivity model was developed and implemented to recommend priorities for dam removal, where this action could significantly improve the movement of potadromous fish species along the local streams. The proposed model accounts for fish movement across the dam or weir (permeability), which is a novel issue in connectivity models. However, before any final recommendation on the fate of a dam or weir, the connectivity results will be balanced with other important socio-economic interests. While implementing the connectivity model, an inventory of barriers (dams and weirs) was accomplished through an observation of satellite images. Besides identification and location of any obstacles, the inventory comprised the compilation of data on surrounding land use, reservoir water use, characteristics of the riparian gallery, and permeability conditions for fish, among others. All this information was stored in a geospatial dataset that also included geographical information on the sub-catchment drainage network. The linear (drainage network) and point (barriers) source data were processed in a computer program that provided or returned numbers for inter-barrier stream lengths (habitat), and the barrier permeability. These numbers were finally used in the same computer program to calculate a habitat connector index, and a link improvement index, used to prioritize dam removal based upon structural connectivity criteria. The results showed that habitat patch connectivity in the Sabor, Tâmega and Côa sub-catchments is not dramatically affected by the installed obstacles, because most link improvement values were generally low. For the opposite reason, in the Corgo sub-catchment, obstacles may constitute a relatively higher limitation to connectivity, and in this case the removal of eight obstacles could significantly improve this connectivity. Using the probabilistic model of structural connectivity, it was possible to elaborate a preliminary selection of dams/weirs that critically limit stream connectivity, and that will be the focus of field hydraulic characterization to precisely determine fish movement along the associated river stretches. Future work will also include the implementation of a multi-criteria decision support system for dam removal or mitigation of the critical structures, as well to define exclusion areas for additional obstacles.
