Early diagnosis of paroxysmal atrial fibrillation (PAF) could prompt patients to receive timely interventions in clinical practice. Various PAF onset prediction algorithms might benefit from accurate heart rate variability (HRV) analysis and machine learning classification but are challenged by real-time monitoring scenarios. The aim of this study is to present an end-to-end deep learning-based PAFNet model that integrates a sliding window technique on raw R-R intervals of electrocardiogram (ECG) segments to achieve a real-time prediction of PAF onset. This integration enables the deep convolutional neural network (CNN) to be customized as a light-weight architecture that accommodates the size of sliding windows simply by altering the input layer, and specifically its effectiveness in making a new prediction with each new heartbeat. Catering to the potential influence of input sizes, three CNN models were trained using 50, 100, and 200 R-R intervals, respectively. For each model, the performance of the automated algorithms was evaluated for PAF prediction using a ten-fold cross-validation. As a results, a total of 56,381 PAFN-type and 56,900 N-type R-R interval segments were collected from publicly accessible ECG databases, and a promising prediction performance of the automated algorithm with 100 R-R intervals was achieved, with a sensitivity of 97.12%, a specificity of 97.77%, and an accuracy of 97.45%, respectively. Importantly, the automated algorithm with a sliding window step of 1 could process one sample in only 23.1 milliseconds and identify the onset of PAF at least 45 min in advance. The present results suggest that the sliding window technique on raw R-R interval sequences, along with deep learning-based algorithms, may offer the possibility of providing an accurate, real-time, end-to-end clinical tool for mass monitoring of PAF. To gain deeper insights into the genomic characteristics of Limosilactobacillus reuteri (L. reuteri) YLR001 and uncover its probiotic properties, in the current study, a comprehensive analysis of its whole genome was conducted, explicitly exploring the genetic variations associated with different host organisms. The genome of YLR001 consisted of a circular 2,242,943 bp chromosome with a GC content of 38.84%, along with three circular plasmids (24,864, 38, 926, and 132,625 bp). Among the 2183 protein-coding sequences (CDSs), the specific genes associated with genetic adaptation and stress resistance were identified. We predicted the function of COG protein genes and analyzed the KEGG pathways. Comparative genome analysis revealed that the pan-genome contained 5207 gene families, including 475 core gene families and 941 strain-specific genes. Phylogenetic analysis revealed distinct host specificity among 20 strains of L. reuteri, highlighting substantial genetic diversity across different hosts. This study enhanced our comprehension of the genetic diversity of L. reuteri YLR001, demonstrated its potential probiotic characteristics, and established more solid groundwork for future applications. This study presents a series of Cu-Mn/Al2O3 catalysts prepared by the polyol method to improve the toluene combustion process. The catalytic activity evaluation results showed that the different polyols have a great influence on catalyst activity, in which the catalyst prepared with glycerol through a hydrothermal reaction at 90 °C displayed the highest catalytic activity. The lowest T90 and T50 values could be achieved by CMA-GL-90 with 260 and 237 °C, respectively. Moreover, the XRD and BET results showed that the hydrothermal treatment was more favorable with Cu-Mn crystal formation, and an abundance of mesopores remained in all catalysts with a high specific surface area from 94.37 to 123.03 m2·g−1. The morphology analysis results by SEM and TEM indicated that employing glycerol coupled with hydrothermal treatment at 90 °C could enhance the formation of CuMn2O4 spinel. The toluene catalytic combustion mechanism of Cu-Mn/Al2O3 catalysts was discussed based on XPS and H2-TPR, and a high atomic ratio of Mn3+ could be obtained with 51.03%, and the ratio of Oads/Olatt also increased to 2.85 in CMA-GL-90. The increase in Mn3+ species and oxygen vacancies on the surface of catalysts exhibited excellent activity and stability for toluene combustion. These findings offer valuable insights for optimizing the design and application of Cu-Mn/Al2O3 catalysts in addressing the catalytic oxidation reactions of organic volatile compounds. Biogenetic resources are the foundation of biodiversity and are of great significance to the sustainability of human society. The effective promotion of biogenetic resource conservation depends on the scientific formulation and implementation of relevant policies, so the quantitative evaluation of biogenetic resource conservation policies can provide decision support for the next step of policy formulation. Based on text analysis, social network analysis, and the construction of the PMC index model, this study selected 132 policy samples issued by the Chinese government in the field of biogenetic resources, established an evaluation system for China's biogenetic resources policies, which contains 10 first-level indicators and 55 s-level indicators, and drew the PMC curve diagram accordingly to quantitatively evaluate China's biogenetic resources policies. The results show that China's biogenetic resources policies are generally at a good level, which can meet the current practical needs of biogenetic resources conservation, but there are problems such as the lack of policy forecasts in the relevant policy texts, the lack of flexible planning in the short and medium term, the lack of co-operation among the policy issuers, and the insufficient guidance of innovation. Carbon dots (CDs) are luminescent carbon nanoparticles with significant potential in analytical sensing, biomedicine, and energy regeneration due to their remarkable optical, physical, biological, and catalytic properties. In light of the enduring ecological impact of non-biomass waste that persists in the environment, efforts have been made toward converting non-biomass waste, such as ash, waste plastics, textiles, and papers into CDs. This review introduces non-biomass waste carbon sources and classifies them in accordance with the 2022 Australian National Waste Report. The synthesis approaches, including pre-treatment methods, and the properties of the CDs derived from non-biomass waste are comprehensively discussed. Subsequently, we summarize the diverse applications of CDs from non-biomass waste in sensing, information encryption, LEDs, solar cells, and plant growth promotion. In the final section, we delve into the future challenges and perspectives of CDs derived from non-biomass waste, shedding light on the exciting possibilities in this emerging area of research. The biological degradation of plant residues in the soil or on the soil surface is an integral part of the natural life cycle of annual plants and does not have adverse effects on the environment. Crop straw is characterized by a complex structure and exhibits stability and resistance to rapid microbial decomposition. In this study, we conducted a microcosm experiment to investigate the dynamic succession of the soil microbial community and the functional characteristics associated with lignocellulose-degrading pathways. Additionally, we aimed to identify lignocellulose-degrading microorganisms from the straw of three crop species prevalent in Northeast China: soybean (Glycine max Merr.), rice (Oryza sativa L.), and maize (Zea mays L.). Our findings revealed that both the type of straw and the degradation time influenced the bacterial and fungal community structure and composition. Metagenome sequencing results demonstrated that during degradation, different straw types assembled carbohydrate-active enzymes (CAZymes) and KEGG pathways in distinct manners, contributing to lignocellulose and hemicellulose degradation. Furthermore, isolation of lignocellulose-degrading microbes yielded 59 bacterial and 14 fungal strains contributing to straw degradation, with fungi generally exhibiting superior lignocellulose-degrading enzyme production compared to bacteria. Experiments were conducted to assess the potential synergistic effects of synthetic microbial communities (SynComs) comprising both fungi and bacteria. These SynComs resulted in a straw weight loss of 42% at 15 days post-inoculation, representing a 22% increase compared to conditions without any SynComs. Microbial-Induced Calcium Carbonate Precipitation (MICP) is an environmentally friendly, efficient, and sustainable new soil reinforcement technology. For this study, Bacillus pasteurii were domesticated and cultured in a natural seawater environment with multiple gradients and used for coral reef calcareous sand reinforcement, comparing the mineral composition of the generated precipitates and the reinforcement strength under different domestication gradient conditions. The results revealed that, while the natural seawater environment inhibits the growth of Bacillus pasteurii, the gradient domestication method allows the bacteria to gradually adapt to the natural seawater environment. Notably, their shape becomes thin and long under the seawater environment. Furthermore, the MICP mineralisation reaction rate is faster in the natural seawater environment and, with an increase in the domestication gradient, the mineralisation reaction precipitates increased. At the same time, in the seawater environment, a small amount of mineral components were generated in addition to C a C O 3 , such as M g x C a y (C O 3 ) z , and the M g 2 + mineral content increased with an increase in the domestication gradient. When comparing the curing effect under different gradients in the natural seawater environment, it was found that the Bacillus pasteurii can effectively enhance the curing effect of the calcareous sand after multi-gradient domestication in the seawater environment, with the curing effect increasing with an increase in the domestication gradient. The results of this study provide new ideas for the application of MICP technology in seawater environments for the reinforcement of calcareous sand in the construction of South China Sea islands and reefs. Farming Chinese perch in aquaculture systems with artificial diets is a new method and developing trend. This method of raising Chinese perch has led to outbreaks of new diseases. In 2022, a disease outbreak occurred among farmed Chinese perch fed an artificial diet in Jiangxi Province, China, during which 50% of the fish died. The clinical signs exhibited by the diseased Chinese perch included decreased physical vitality, anorexia, emaciation, and dorsal fin ulceration with white cysts and flocs. Coinfection with ciliate parasites and an oomycete was found. No pathogenic bacteria were isolated from the microbial test, and the viral detection results were negative. After morphological and 18S or 28S rDNA sequence clustering analysis, the parasites were identified as Epistylis wuhanensis and Zoothamnium sp., while the oomycete was identified as Achlya klebsiana. This article discusses the interaction between parasitic and oomycete coinfections in Chinese perch from the perspective of the characteristics of Chinese perch raised with an artificial diet. The relationships between parasites, oomycete, and fish were also briefly discussed. Photoacoustic microscopy (PAM) is a medical-imaging technique with the merits of high contrast and resolution. Nevertheless, conventional PAM scans specimens in a diameter-by-diameter fashion, resulting in a time-consuming process. Furthermore, deep-learning-based PAM image enhancement necessitates acquiring ground-truth data for training purposes. In this paper, we built an optical-resolution photoacoustic microscopy system and introduced an innovative unsupervised-learning algorithm. First, we enhanced the rotational-scanning method, transitioning from a diameter-by-diameter approach to a sector-by-sector one, significantly reducing imaging time (from 280 s to 109 s). Second, by establishing a metric for unsupervised learning, we eliminated the need for collecting reliable and high-quality ground truth, which is a challenging task in photoacoustic microscopy. A total of 324 pairs of datasets (mouse ears) were collected for unsupervised learning, with 274 for training and 50 for testing. Additionally, carbon-fiber data were sampled for lateral resolution and contrast evaluation, as well as the effective rate evaluation of the algorithm. The enhanced images demonstrated superior performance compared with that of maximum projection, both subjectively and objectively. A 76% improvement in the lateral resolution was observed. The effective rate of the algorithm was measured to be 100%, which was tested on 50 random samples. Vegetation classification, biomass assessment, and wildfire dynamics are interconnected wildfire-ecosystem components. The Chongli District, located in Zhangjiakou City, was the venue for skiing at the 2022 Winter Olympics. Its high mountains and dense forests create a unique environment. The establishment of alpine ski resorts highlighted the importance of comprehensive forest surveys. Understanding vegetation types and their biomass is critical to assessing the distribution of local forest resources and predicting the likelihood of forest fires. This study used satellite multispectral data from the Sentinel-2B satellite to classify the surface vegetation in the Chongli District through K-means clustering. By combining this classification with a biomass inversion model, the total biomass of the survey area can be calculated. The biomass inversion equation established based on multispectral remote sensing data and terrain information in the Chongli area have a strong correlation (shrub forest R2 = 0.811, broadleaf forest R2 = 0.356, coniferous forest R2 = 0.223). These correlation coefficients are key indicators for our understanding of the relationship between remote sensing data and actual vegetation biomass, reflecting the performance of the biomass inversion model. Taking shrubland as an example, a correlation coefficient as high as 0.811 shows the model's ability to accurately predict the biomass of this type of vegetation. In addition, through multiple linear regression, the optimal shrub, broadleaf, and coniferous forest biomass models were obtained, with the overall accuracy reaching 93.58%, 89.56%, and 97.53%, respectively, meeting the strict requirements for survey accuracy. This study successfully conducted vegetation classification and biomass inversion in the Chongli District using remote sensing data. Cardiovascular disease (CVD) remains the first cause of mortality globally. Diet plays a fundamental role in cardiovascular health and is closely linked to the development of CVD. Numerous human studies have provided evidence on the relationship between diet and CVD. By discussing the available findings on the dietary components that potentially influence CVD progression and prevention, this review attempted to provide the current state of evidence on healthy dietary choices for CVD. We focus on the effects of individual macronutrients, whole food products, and dietary patterns on the risks of CVD, and the data from population-based trials, observational studies, and meta-analyses are summarized. Unhealthy dietary habits, such as high intake of saturated fatty acids, sugar-sweetened beverages, red meat, and processed meat as well as high salt intake are associated with the increased risk of CVD. Conversely, increased consumption of plant-based components such as dietary fiber, nuts, fruits, and vegetables is shown to be effective in reducing CVD risk factors. The Mediterranean diet appears to be one of the most evidence-based dietary patterns beneficial for CVD prevention. However, there is still great debate regarding whether the supplementation of vitamins and minerals confers cardioprotective benefits. BRAF is one of the most frequently mutated oncogenes, with an overall frequency of about 50%. Targeting BRAF and its effector mitogen-activated protein kinase kinase 1/2 (MEK1/2) is now a key therapeutic strategy for BRAF-mutant tumors, and therapies based on dual BRAF/MEK inhibition showed significant efficacy in a broad spectrum of BRAF tumors. Nonetheless, BRAF/MEK inhibition therapy is not always effective for BRAF tumor suppression, and significant challenges remain to improve its clinical outcomes. First, certain BRAF tumors have an intrinsic ability to rapidly adapt to the presence of BRAF and MEK1/2 inhibitors by bypassing drug effects via rewired signaling, metabolic, and regulatory networks. Second, almost all tumors initially responsive to BRAF and MEK1/2 inhibitors eventually acquire therapy resistance via an additional genetic or epigenetic alteration(s). Overcoming these challenges requires identifying the molecular mechanism underlying tumor cell resistance to BRAF and MEK inhibitors and analyzing their specificity in different BRAF tumors. In recent years, the utilization of deep learning for forest fire detection has yielded favorable outcomes. Nevertheless, the accurate segmentation of forest fires in foggy surroundings with limited visibility remains a formidable obstacle. To overcome this challenge, a collaborative defogging learning framework, known as Defog DeepLabV3+, predicated on an enhanced DeepLabV3+ model is presented. Improved learning and precise flame segmentation are accomplished by merging the defogging features produced by the defogging branch in the input image. Furthermore, dual fusion attention residual feature attention (DARA) is proposed to enhance the extraction of flame-related features. The FFLAD dataset was developed given the scarcity of specifically tailored datasets for flame recognition in foggy environments. The experimental findings attest to the efficacy of our model, with a Mean Precision Accuracy (mPA) of 94.26%, a mean recall (mRecall) of 94.04%, and a mean intersection over union (mIoU) of 89.51%. These results demonstrate improvements of 2.99%, 3.89%, and 5.22% respectively. Coal and gas outbursts are some of the most serious coal mine disasters, and effective prediction of coal and gas outbursts can reduce the likelihood of accidents and fatalities. Previously conducted studies have established that machine learning has achieved results in the prediction of coal and gas outbursts, but there is a problem that the available accident data of coal and gas outbursts are diminished or missing. This paper proposes a prediction model based on multiple filling of chained equations for random forests (miceforest) and the Harris Hawk optimization algorithm with Piecewise chaos mapping (PHHO) to optimize the kernel extreme learning machine (KELM) to solve the problem of missing data in coal and gas outburst prediction and to improve prediction accuracy in the case of missing data. Firstly, the miceforest algorithm was adopted to fill missing values in the salient samples, and then the PHHO algorithm was used to optimize the parameters of KELM. Finally, the datasets before and after filling were input into the PHHO–KELM model for experimentation and comparison with other models. The results show that miceforest filling is effective in improving the salient sample accuracy and overall accuracy of predictions, but the improvement is not significant for non-salient samples. The use of the PHHO–KELM model can effectively avoid falling into a local optimum and further improve the prediction accuracy of the KELM algorithm. The salient sample accuracy and overall accuracy of the miceforest–PHHO–KELM model prediction are 96.77% and 98.50%. Concerning the ever-changing wetland environment, the efficient extraction of wetland information holds great significance for the research and management of wetland ecosystems. China's vast coastal wetlands possess rich and diverse geographical features. This study employs the SegFormer model and Sentinel-2 data to conduct a wetland classification study for coastal wetlands in Yancheng, Jiangsu, China. After preprocessing the Sentinel data, nine classification objects (construction land, Spartina alterniflora (S. alterniflora), Suaeda salsa (S. salsa), Phragmites australis (P. australis), farmland, river system, aquaculture and tidal falt) were identified based on the previous literature and remote sensing images. Moreover, mAcc, mIoU, aAcc, Precision, Recall and F-1 score were chosen as evaluation indicators. This study explores the potential and effectiveness of multiple methods, including data image processing, machine learning and deep learning. The results indicate that SegFormer is the best model for wetland classification, efficiently and accurately extracting small-scale features. With mIoU (0.81), mAcc (0.87), aAcc (0.94), mPrecision (0.901), mRecall (0.876) and mFscore (0.887) higher than other models. In the face of unbalanced wetland categories, combining CrossEntropyLoss and FocalLoss in the loss function can improve several indicators of difficult cases to be segmented, enhancing the classification accuracy and generalization ability of the model. Finally, the category scale pie chart of Yancheng Binhai wetlands was plotted. In conclusion, this study achieves an effective segmentation of Yancheng coastal wetlands based on the semantic segmentation method of deep learning, providing technical support and reference value for subsequent research on wetland values. This study provides a novel method of preparing lotus seed starch nanocrystals (LS-SNCs) using acid hydrolysis combined with ultrasonic-assisted acid hydrolysis (U-LS-SNCs) and evaluates the structural characteristics of starch nanocrystals using scanning electron microscopy; analysis of particle size, molecular weight, and X-ray diffraction patterns; and FT-IR spectroscopy. The results showed that the preparation time of U-LS-SNCs could be reduced to 2 days less than that for LS-SNCs. The smallest particle size and molecular weight were obtained after a 30 min treatment with 200 W of ultrasonic power and 5 days of acid hydrolysis. The particle size was 147 nm, the weight-average molecular weight was 3.42 × 104 Da, and the number-average molecular weight was 1.59 × 104 Da. When the applied ultrasonic power was 150 W for 30 min and acid hydrolysis was applied for 3 days, the highest relative crystallinity of the starch nanocrystals was 52.8%. Metal–organic frameworks (MOFs) are new porous materials composed of metal centers and organic ligand bridges, which received great attention in the field of photocatalysis. In this work, Ag2CrO4@MIL–125(Ti)–NH2 (denoted as AgCr@M125) Z–scheme heterojunctions were synthesized via a simple microemulsion method, by which highly dispersed nano–sized Ag2CrO4 can be anchored uniformly on the surfaces of porous MIL–125(Ti)–NH2 (denoted as M125). Compared with pure M125 and Ag2CrO4, the as–prepared AgCr@M125 hybrids show significant photocatalytic efficiency against inactivated Staphylococcus aureus (S. aureus), reaching over 97% inactivation of the bacteria after 15 min of visible light irradiation. Notably, the photocatalytic activity of the obtained 20%AgCr@M125 is about 1.75 times higher than that of AgCr–M125, which was prepared via a traditional precipitation method. The enhanced photocatalytic antibacterial activity of the AgCr@M125 photocatalytic system is strongly ascribed to a direct Z–scheme mechanism, which can be carefully discussed based on energy band positions and time–dependent electron spin response (ESR) experiments. Our work highlights a simple way to enhance the antibacterial effect by coupling with Ag2CrO4 and M125 via a microemulsion–assisted strategy and affords an ideal example for developing MOFs–based Z–scheme photocatalysts with excellent photoactivity. Mulberry (Morus alba), a widely distributed economic plant, can withstand long-term flooding stress. However, the regulatory gene network underlying this tolerance is unknown. In the present study, mulberry plants were subjected to submergence stress. Subsequently, mulberry leaves were collected to perform quantitative reverse-transcription PCR (qRT-PCR) and transcriptome analysis. Genes encoding ascorbate peroxidase and glutathione S-transferase were significantly upregulated after submergence stress, indicating that they could protect the mulberry plant from flood damage by mediating ROS homeostasis. Genes that regulate starch and sucrose metabolism; genes encoding pyruvate kinase, alcohol dehydrogenase, and pyruvate decarboxylase (enzymes involved in glycolysis and ethanol fermentation); and genes encoding malate dehydrogenase and ATPase (enzymes involved in the TCA cycle) were also obviously upregulated. Hence, these genes likely played a key role in mitigating energy shortage during flooding stress. In addition, genes associated with ethylene, cytokinin, abscisic acid, and MAPK signaling; genes involved in phenylpropanoid biosynthesis; and transcription factor genes also showed upregulation under flooding stress in mulberry plants. These results provide further insights into the adaptation mechanisms and genetics of submergence tolerance in mulberry plants and could aid in the molecular breeding of these plants. Coastal erosion is increasing worldwide due to the increasing frequency of extreme natural phenomena and excessive human exploitation. In this study, a small model experiment was conducted to investigate the solidification effects of three enzyme sources—soybean urease, freshwater Bacillus pasteurella, and seawater domesticated Bacillus pasteurella—on coastal sediments and their impacts in a seawater environment. The solidifying effect of different enzyme sources was determined by measuring the mechanical properties and corrosion resistance of the cured specimen model. The influence of solidified seawater in a seawater environment was obtained by measuring the changes in the pH value, calcium ion concentration, and ammonia nitrogen content of solidified seawater. The results show that different enzyme sources have a certain strengthening effect on coastal sediments. The mechanical properties of coastal sediments can be enhanced by increasing the amount of enzyme solution or level of solidification and can effectively resist simulated flow erosion. Comparing the reinforcement effects of different enzyme sources, it can be seen. It was observed that Bacillus pasteurella acclimated in seawater had better reinforcement effects than Bacillus pasteurella fresh water, and Bacillus pasteurella fresh water had better reinforcement effects than soybean urease. In the seawater measurement tests, the solidification of coastal sediments using different enzyme sources led to a decrease in the seawater pH value, and the acidification of seawater dissolved the generated calcium carbonate, increased the concentration of calcium ions in seawater, and produced ammonia nitrogen as a byproduct in the seawater. It was observed that, compared with the other two enzyme source solutions, the seawater-domesticated Bacillus pasteurella can better adapt to the high-salt environment of seawater, microbial metabolism is not inhibited, urea decomposition ability is improved, and calcium carbonate production is higher, which can effectively improve the engineering characteristics of coastal sediments and play a positive role in coastal protection and development. In order to maximize the reuse of used electronic component resources, while reducing environmental pollution, Cu metal was recycled from wasted CPU sockets by the reformative slurry electrolysis method. However, the influences on the regulation of the Cu recovery rate and purity from waste CPU slots, by slurry electrolysis, has not been systematically elucidated in previous studies. In this work, the effects of H2SO4 concentration, slurry density, NH4Cl concentration, current density, and reaction time, on the recovery rate and purity of Cu in slurry electrolysis, were researched by systematic experimental methods. The results showed that the recycled metal elements were mainly present as powders from the cathode, rather than in the electrolyte. Moreover, the metallic elements in the cathode powder consisted of mostly Cu and small amounts of Sn and Ni. The recovery rate and purity of Cu were up to 96.19% and 93.72%, respectively, with the optimum conditions being: an H2SO4 concentration of 2 mol/L, slurry density of 30 g/L, NH4Cl concentration of 90 g/L, current density of 80 mA/cm2, and reaction time of 7 h. Compared with previous studies, the Cu recovered in this experiment was present in the cathode powder, which was more convenient for the subsequent processing. Streptococcus pneumoniae is one of the most common bacterial pathogens of a wide range of community-acquired infections. It has been more and more recognized that this bacterium could also play a role as a cause of nosocomial infections. In this study, by retrospective analysis of the phenotypic resistance characteristics and genomic characteristics of 52 S. pneumoniae isolates in a hospital in Beijing, China, from 2018 to 2022, we explored the carriage of resistance genes and mutations in penicillin-binding proteins corresponding to the resistances, and identified the population diversity based on the prediction of serotypes and identification of sequence types (STs). The isolates displayed resistances to erythromycin (98%), tetracycline (96%), sulfonamide (72%) and penicillin G (42%). Among the 52 isolates, 41 displayed multiple-drug resistance. In total, 37 STs and 21 serotypes were identified, and the clonal complex 271 serogroup 19 was the most prevalent subtype. Only 24 isolates (46.2%) of 7 serotypes were covered by the 13-valent pneumococcal conjugate vaccination. The isolates showed high carriages of resistance genes, including tet(M) (100%) and erm(B) (98.1%); additionally, 32 isolates (61.5%) had mutations in penicillin-binding proteins. We also observed 11 healthcare-associated infections and 3 cases infected by different subtypes of isolates. We did not find nosocomial transmissions between the patients, and these cases might be associated with the asymptomatic colonization of S. pneumoniae in the human population. Cuproptosis, a new cell death pattern, is promising as an intervention target to treat tumors. Abnormal long non-coding RNA (lncRNA) expression is closely associated with the occurrence and development of papillary renal cell carcinoma (pRCC). However, cuproptosis-related lncRNAs (CRLs) remain largely unknown as prognostic markers for pRCC. We aimed to forecast the prognosis of pRCC patients by constructing models according to CRLs and to examine the correlation between the signatures and the inflammatory microenvironment. From the Cancer Genome Atlas (TCGA), RNA sequencing, genomic mutations and clinical data of TCGA-KIRP (Kidney renal papillary cell carcinoma) were analyzed. Randomly selected pRCC patients were allotted to the training and testing sets. To determine the independent prognostic impact of the training characteristic, the least absolute shrinkage and selection operator (LASSO) algorithm was utilized, together with univariate and multivariate Cox regression models. Further validation was performed in the testing and whole cohorts. External datasets were utilized to verify the prognostic value of CRLs as well. The CRLs prognostic features in pRCC were established based on the five CRLs (AC244033.2, LINC00886, AP000866.1, MRPS9-AS1 and CKMT2-AS1). The utility of CRLs was evaluated and validated in training, testing and all sets on the basis of the Kaplan–Meier (KM) survival analysis. The risk score could be a robust prognostic factor to forecast clinical outcomes for pRCC patients by the LASSO algorithm and univariate and multivariate Cox regression. Analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) data demonstrated that differentially expressed genes (DEGs) are primarily important for immune responses and the PI3K-Akt pathway. Arachidonic acid metabolism was enriched in the high-risk set by Gene Set Enrichment Analysis (GSEA). In addition, Tumor Immune Dysfunction and Exclusion (TIDE) analysis suggested that there was a high risk of immune escape in the high-risk cohort. The immune functions of the low- and high-risk sets differed significantly based on immune microenvironment analysis. Finally, four drugs were screened with a higher sensitivity to the high-risk set. Taken together, a novel model according to five CRLs was set up to forecast the prognosis of pRCC patients, which provides a potential strategy to treat pRCC by a combination of cuproptosis and immunotherapy. A7N01P-T4 aluminum alloy plates for high-speed trains will experience a certain amount of fatigue damage under alternating loads. Three groups of samples, P0 (no fatigue damage), P1 (loading stress 30 MPa), and P2 (loading stress 70 MPa), were created, and corrosion fatigue crack growth (CFCG) tests were conducted in 3.5 wt.% NaC1 solution. The crack growth rate was found to increase after fatigue damage as the damage degree increased. In addition, the A7N01P-T4 aluminum alloy base metal exhibited obvious secondary cracks and crack bifurcations after fatigue damage. It is believed that fatigue damage causes stress concentration in the material, while a certain degree of stress corrosion cracking occurs during the CFCG growth process. This is because hydrogen (H) easily accumulates and diffuses along the grain boundary, which reduces the strength of the grain boundary, thereby becoming the preferred orientation for crack growth. Molybdenum has a broad application and good prospect in the field of nuclear energy, aerospace, electronics, etc., due to its high melting point, high hardness, corrosion resistance and other excellent performances. In this paper, an isothermal and isochronous annealing heat treatment, at the temperature of 800–1300 °C for 0.5–2 h, was applied to pure molybdenum (PM) sheets with deformation of 70%, 80%, 90%, and 95%. The initial deformation of the PM sheet was increased from 70% to 95%. After annealing at 900–1200 °C for 1 h, the recrystallized grain size gradually decreased. The Goss texture ({110}<001>) was always present in the pure molybdenum sheet with 95% deformation during heat treatment, but its strength decreased with the increase of the temperature. The copper texture ({112}<110>) deflected to a cubic texture, and its orientation changed from {001}<110> to that of cube texture {110}<100>. With the increase of the temperature, the cubic texture was obtained more easily in the pure molybdenum sheet. The recrystallization nucleation mechanism of the pure molybdenum sheet with 95% deformation was mainly in situ nucleation and orientation nucleation. The present study aimed to evaluate the feasibility of a formulated diet to replace live baits as feed for Siniperca chuatsi. A 2 × 2 factorial design with three replicates was used to investigate the effects of conventional live baits (LB) and a formulated diet (FD) on the growth, muscle nutrition composition, and digestive enzyme activities of S. chuatsi at two growth stages. The results showed that the feed conversion ratio (FCR) in the FD group was significantly lower than that in the LB group. The final body weight (FBW) and weight gain (WG) were not significantly different between the two feeding modes. However, at the juvenile stage, the specific growth rate (SGR) in the FD group was significantly lower than that in the LB group. Muscle moisture, crude protein, and crude lipid contents were not influenced by dietary treatments. There were no significant differences in ∑EAA, ∑NEAA, and ∑AA compositions of the juvenile and adult stages fed the two kinds of diets. At the juvenile stage, histidine (His) content in the FD group was significantly higher than that in the LB group; at the adult stage, cysteine (Cys) content in the FD group was significantly higher than that in the LB group; at both growth stages, Met content in the FD group was significantly lower than that in the LB group. The FD group showed higher levels of ∑SFA, ∑HUFA, ∑n-3PUFA, n-3/n-6, EPA, and DHA than the LB group did, whilst the opposite occurred in the MUFA levels. In addition, fish fed with LB had significantly higher values of pepsin and intestinal trypsin activities in the juvenile fish compared to the FD group, but the activities were not significantly different between the two feeding modes at the adult stage. The activities of stomach and intestine amylase in the FD group were significantly higher than those in the LB group. Overall, these results showed that under long-term feeding conditions, S. chuatsi fed with the artificial diet had no significant difference in muscle crude protein, crude lipid, amino acid composition, and digestive capacity, but showed superiority in the composition of fatty acids such as EPA and DHA levels compared to the live baits group. Circular fringe projection profilometry (CFPP), as a branch of carrier fringe projection profilometry, has attracted research interest in recent years. Circular fringe Fourier transform profilometry (CFFTP) has been used to measure out-of-plane objects quickly because the absolute phase can be obtained by employing fewer fringes. However, the existing CFFTP method needs to solve a quadratic equation to calculate the pixel displacement amount related to the height of the object, in which the root-seeking process may get into trouble due to the phase error and the non-uniform period of reference fringe. In this paper, an improved CFFTP method based on a non-telecentric model is presented. The calculation of displacement amount is performed by solving a linear equation instead of a quadratic equation after introducing an extra projection of circular fringe with circular center translation. In addition, Gerchberg iteration is employed to eliminate phase error of the region close to the circular center, and the plane calibration technique is used to eliminate system error by establishing a displacement-to-height look-up table. EA4T steel has been widely used in the manufacture of railway axles. Laser cladding remanufacturing technology has been used to repair railway axles, and the failure behaviors of railway axles has been studied. In this work, a class of original FeCrNiMo alloy layers that contain rare-earth elements was successfully fabricated through a laser cladding technique on EA4T steel substrates. The microstructure and elemental distribution of the cladding layers were analyzed through scanning electron microscopy (SEM), transmission electron microscopy (TEM) and electron probe microanalysis (EPMA). Simultaneously, the microhardness, bond strength and fatigue properties of different samples were determined. The results indicated that the phase composition of the cladding layer was mainly martensite. The grains in the cladding layer were gradually refined with an increase in the distance from the interface, and the chemical composition distribution was more uniform. The alignment problem of a rotating optical measurement system composed of a charge-coupled device (CCD) camera and a turntable is discussed. The motion trajectory model of the optical center (or projection center in the computer vision) of a camera rotating with the rotating device is established. A method based on camera calibration with a two-dimensional target is proposed to calculate the positions of the optical center when the camera is rotated by the turntable. An auxiliary coordinate system is introduced to adjust the external parameter matrix of the camera to map the optical centers on a special fictitious plane. The center of the turntable and the distance between the optical center and the rotation center can be accurately calculated by the least square planar circle fitting method. Lastly, the coordinates of the rotation center and the optical centers are used to provide guidance for the installation of a camera in a rotation measurement system. Inflammatory responses have been shown to induce hyperglycemia, yet the underlying mechanism is still largely unclear. GLP-1 is an important intestinal hormone for regulating glucose homeostasis; however, few studies have investigated the influence of digestive tract Salmonella infection on enteroendocrine L cell secretions. In this study, we established a model of Salmonella-infected piglets by oral gavage in order to analyze the effects of Salmonella infection on enteroendocrine L cell function. Furthermore, in vitro lipopolysaccharide (LPS) was administered to STC-1 cells to clarify its direct effect on GLP-1 secretion. The results showed that significantly increased blood glucose in the group of Salmonella-infected piglets was observed, and Salmonella infection decreased blood GLP-1 content. Then, ileal epithelium damage was observed by histological detection, and this was further verified by TUNEL staining. We identified activation of TLR signaling demonstrating up-regulated expressions of TLR4 and nuclear factor-kappa B (NF-ΚB). Furthermore, it was shown that Salmonella induced pyroptosis of enteroendocrine L cells and enhanced the secretion of IL-1β through augmenting gene and protein expressions of NOD-like receptor protein 3 (NLRP3), apoptosis-associated speck-like protein containing a carboxyl-terminal CARD (ASC), Caspase 1, and gasdermin D (GSDMD). Meanwhile, in vitro LPS treatment induced the pyroptosis of STC-1 cells and reduced the secretion of GLP-1. Altogether, the results demonstrated that Salmonella infection can reduce secretion of GLP-1 by inducing pyroptosis of intestinal L cells, which may eventually result in hyperglycemia. Leaf trait is an important target trait in crop breeding programs. Moderate leaf curling may be a help for improving crop yield by minimizing the shadowing by leaves. Mining locus for leaf curling trait is of significance for plant genetics and breeding researches. The present study identified a novel rapeseed accession with up-curling leaf, analyzed the up-curling leaf trait inheritance, and fine mapped the locus for up-curling leaf property (Bnuc3) in Brassica napus. Genetic analysis revealed that the up-curling leaf trait is controlled by a single dominant locus, named BnUC3. We performed an association study of BnUC3 with single nucleotide polymorphism (SNP) markers using a backcross population derived from the homozygous up-curling leaf line NJAU-M1295 and the canola variety 'zhongshuang11' with typical flat leaves, and mapped the BnUC3 locus in a 1.92 Mb interval of chromosome A02 of B. napus. To further map BnUC3, 232 simple sequence repeat (SSR) primers and four pairs of Insertion/Deletion (InDel) primers were developed for the mapping interval. Among them, five SSR markers and two InDel markers were polymorphic. By these markers, the mapping interval was narrowed to 92.0 kb using another F2 population. This fine mapping interval has 11 annotated genes among which BnaA02T0157000ZS were inferred to be candidate casual genes for up-curling leaf based on the cloned sequence analysis, gene functionality, and gene expression analysis. Artificial grasslands have been regarded as an effective method to improve grass production and quality, especially on the Qinghai–Tibet Plateau. Soil ecosystem multifunctionality (EMF) plays an important role in sustainable regional development. However, few studies have investigated the impacts of artificial grasslands on soil EMF. Here, we constructed single-species artificial grasslands in a natural temperate steppe and investigated soil microbial communities, abiotic factors (soil moisture and pH), and functions related to biogeochemical cycles to explore (1) how the transformation from temperate steppe to artificial grasslands affected soil EMF and (2) the roles of species and phylogenetic microbial diversities, microbial community composition, and abiotic factors in driving differences in soil EMF. Our results showed that artificial grasslands decreased soil EMF regardless of planting species; that the bacterial and fungal community composition contributed more to soil EMF prediction than species and phylogenetic diversities; and that microbial phylogenetic diversities were negatively associated with soil EMF. Soil pH played an important role in the effects of artificial grasslands on soil EMF—artificial grasslands increased soil pH, which was negatively associated with soil EMF. Overall, the benefits of establishing artificial grasslands, for example, higher grass production and quality, might be at the expense of soil EMF. Crossed-grating phase-shifting profilometry (CGPSP) has great utility in three-dimensional shape measurement due to its ability to acquire horizontal and vertical phase maps in a single measurement. However, CGPSP is extremely sensitive to the non-linearity effect of a digital fringe projection system, which is not studied in depth yet. In this paper, a mathematical model is established to analyze the phase error caused by the non-linearity effect. Subsequently, two methods used to eliminate the non-linearity error are discussed in detail. To be specific, a double five-step algorithm based on the mathematical model is proposed to passively suppress the second non-linearity. Furthermore, a precoding gamma correction method based on probability distribution function is introduced to actively attenuate the non-linearity of the captured crossed fringe. The comparison results show that the active gamma correction method requires less fringe patterns and can more effectively reduce the non-linearity error compared with the passive method. Finally, employing CGPSP with gamma correction, a faster and reliable inverse pattern projection is realized with less fringe patterns. [ABSTRACT FROM AUTHOR]

In managed orchards, fertilization brings out not only high productivity expectations but also severe environmental pollution. Because economic profit takes priority over environmental cost, increasing amounts of fertilizer have been used in mature subtropical Torreya grandis orchards. However, given the magnitude of global nitrogen deposition, it's worth considering whether heavy fertilizer treatment is necessary. To elucidate the balance between T. grandis nutrient demands and fertilizer supply, we determined the C, N, and P concentrations of foliar and soil ([C], [N], [P]) at 9 orchards undergoing long-term fertilizer treatments in two scenarios of N and N + P addition with different intensity. After documenting the dynamic variation of plant growth, nutrients characteristic, and the corresponding resorption efficiency, we found that excessive N addition interfered T. grandis' sensibility to P availability in this N-enrichment area, leading to an increasing foliar [P] and resorption efficiency (PRE) and decoupling plant C:N:P ratios. As a result, enhanced fertilizer supply failed to improve carbon accumulation, plant growth, and yield effectively. These results demonstrate that extra fertilization in the N-saturated study area highly reduced the economic and ecological efficiency of fertilizers. Thus, our research suggests that N addition in the studied orchards should be rejected, and we recommend organic management as a more conducive method to achieve sustainable development. Simple Summary: Protein ubiquitination is catalyzed by many enzymes, whose functions and substrate specificity are not fully understood. This study reports the expression patterns of the membrane-associated RING-CH (MARCH) family members in breast cancer and their association with patient outcomes. Specifically, MARCH8 is a newly identified tumor suppressor with a role in inhibiting breast cancer metastasis and enhancing cancer cell death. MARCH8 not only promotes the degradation of membrane proteins such as the breast cancer stem-cell marker CD44 through the lysosomal degradation pathway, but also recruits a previously unknown nonmembrane target protein, signal transducer and transcription activator 3 (STAT3), for proteosome-dependent degradation. Protein stability is largely regulated by post-translational modifications, such as ubiquitination, which is mediated by ubiquitin-activating enzyme E1, ubiquitin-conjugating enzyme E2, and ubiquitin ligase E3 with substrate specificity. Membrane-associated RING-CH (MARCH) proteins represent one novel family of transmembrane E3 ligases which target glycoproteins for lysosomal destruction. While most of the MARCH family members are known to degrade membrane proteins in immune cells, their tumor-intrinsic role is largely unknown. In this study, we found that the expression of one MARCH family member, MARCH8, is specifically downregulated in breast cancer tissues and positively correlated with breast cancer survival rate according to bioinformatic analysis of The Cancer Genomic Atlas (TCGA) dataset. MARCH8 protein expression was also lower in a variety of human breast cancer cell lines in comparison to immortalized human mammary epithelial MCF-12A cells. Restoration of MARCH8 expression induced apoptosis in human breast cancer cell lines MDA-MB-231 and BT549. Stable expression of MARCH8 inhibited tumorigenesis and lung metastases of MDA-MB-231 cells in mice. Moreover, we discovered that the breast cancer stem-cell marker and metastasis driver CD44, a membrane protein, interacts with MARCH8 and is one of the glycoprotein targets subject to MARCH8-dependent lysosomal degradation. Unexpectedly, we identified a nonmembrane protein, signal transducer and transcription activator 3 (STAT3), as another essential ubiquitination target of MARCH8, whose degradation through the proteasome pathway is responsible for the proapoptotic changes mediated by MARCH8. These findings highlight a novel tumor-suppressing function of MARCH8 in targeting both membrane and nonmembrane protein targets required for the survival and metastasis of breast cancer cells. Recently, many convolutional networks have been built to fuse a low spatial resolution (LR) hyperspectral image (HSI) and a high spatial resolution (HR) multispectral image (MSI) to obtain HR HSIs. However, most deep learning-based methods are supervised methods, which require sufficient HR HSIs for supervised training. Collecting plenty of HR HSIs is laborious and time-consuming. In this paper, a self-supervised spectral-spatial residual network (SSRN) is proposed to alleviate dependence on a mass of HR HSIs. In SSRN, the fusion of HR MSIs and LR HSIs is considered a pixel-wise spectral mapping problem. Firstly, this paper assumes that the spectral mapping between HR MSIs and HR HSIs can be approximated by the spectral mapping between LR MSIs (derived from HR MSIs) and LR HSIs. Secondly, the spectral mapping between LR MSIs and LR HSIs is explored by SSRN. Finally, a self-supervised fine-tuning strategy is proposed to transfer the learned spectral mapping to generate HR HSIs. SSRN does not require HR HSIs as the supervised information in training. Detecting a variety of biological samples accurately and swiftly in an integrated way is of great practical significance. Currently, biofluorescent spectrum detection still largely relies on microscopic spectrometers. In this study, we propose an integrable method to detect biofluorescent spectrums with designed liquid crystal tunable filter (LCTF), in order to identify typical biological samples such as cells and bacteria. Hela cells labeled with red and green fluorescent proteins and Pseudomonas with fluorescence wavelengths of 610 nm, 509 nm and 450 nm, respectively, are inspected. High-resolution (6 μm) biofluorescent results have been achieved, together with clear images of the Hela cell clusters and the Pseudomonas bacteria colonies. Biofluorescence signals can be detected at a high transmittance (above 80%), and the response time of the device can reach 20 ms or below. The proposed method has the potential to be integrated into a microfluidic system to detect and identify the biofluorescent signals as a high throughput, low-cost option, for both high resolution and large field observation applications. The high-speed three-dimensional (3-D) shape measurement technique has become more and more popular recently, because of the strong demand for dynamic scene measurement. The single-shot nature of Fourier Transform Profilometry (FTP) makes it highly suitable for the 3-D shape measurement of dynamic scenes. However, due to the band-pass filter, FTP method has limitations for measuring objects with sharp edges, abrupt change or non-uniform reflectivity. In this paper, an improved Temporal Fourier Transform Profilometry (TFTP) algorithm combined with the 3-D phase unwrapping algorithm based on a reference plane is presented, and the measurement of one deformed fringe pattern producing a new 3-D shape of an isolated abrupt objects has been achieved. Improved TFTP method avoids band-pass filter in spatial domain and unwraps 3-D phase distribution along the temporal axis based on the reference plane. The high-frequency information of the measured object can be well preserved, and each pixel is processed separately. Experiments verify that our method can be well applied to a dynamic 3-D shape measurement with isolated, sharp edges or abrupt change. A high-speed and low-cost structured light pattern sequence projection has also been presented, it is capable of projection frequencies in the kHz level. Using the proposed 3-D shape measurement algorithm with the self-made mechanical projector, we demonstrated dynamic 3-D reconstruction with a rate of 297 Hz, which is mainly limited by the speed of the camera. Research Highlights: Soil enzymes have a significant impact on the production of glomalin-related soil protein (GRSP), directly and indirectly affecting the nutrient metabolism balance, but there is little available information on ecological stoichiometry in soil aggregates. Background and Objectives: Vegetation restoration changes community structure and species composition in ecosystems, thus changing the physicochemical properties of soil. Soil aggregate is the most basic physical structure of the soil. Therefore, in order to understand dynamic changes in soil aggregate nutrients as vegetation restoration progresses, we set out to investigate the nutrient distribution and utilization in aggregates, and how enzymes respond to the nutrient changes in achieving a nutritional balance along successive stages of vegetation restoration. Materials and Methods: We collected and analyzed soil from plots representing six different stages of a vegetation restoration chronosequence (0, 30, 60, 100, 130, and 160 years) after farmland abandonment on the Loess Plateau, China. We investigated soil nutrient stoichiometry, GRSP, and enzyme stoichiometry in the different successional stages. Results: The results revealed that soil organic carbon, total nitrogen, enzyme activity, and GRSP increased with vegetation recovery age, but not total phosphorus, and not all enzymes reached their maximum in the climax forest community. The easily extractable GRSP/total GRSP ratio was the largest at the shrub community stage, indicating that glomalin degradation was the lowest at this stage. Ecological stoichiometry revealed N-limitation decreased and P-limitation increased with increasing vegetation restoration age. Soil enzymes had a significant impact on the GRSP production, directly and indirectly affecting nutrient metabolism balance. Conclusions: Further study of arbuscular mycorrhizal fungi to identify changes in their category and composition is needed for a better understanding of how soil enzymes affect their release of GRSP, in order to maintain a nutrient balance along successive stages of vegetation restoration. Although TiO2 or MnO2-based materials have been widely used for the degradation of phenolic compounds, complete mineralization is still a challenge, especially for TiO2-based materials. Here, we devise a hierarchically-structured TiO2/MnO2 (HTM) hollow sphere, in which hollow TiO2 acts as a skeleton for the deposition of MnO2 in order to prevent the aggregation of MnO2 nanoparticles and to maintain its hollow structure. During the oxidation reaction, the as-synthesized HTM can fully exert their respective advantages of the TiO2 and MnO2 species to realize the first stage of the rapid oxidation degradation of phenol and the second stage of the complete photo-mineralization of residual phenol and its intermediates, which efficiently overcomes the incomplete mineralization of phenolic compounds. The degradation mechanism and pathway of phenol are also proposed according to the analysis of Mass Spectrometry (MS). Human platelet derived growth factor (PDGF) is a major therapeutic protein with great demand in the clinical setting; however, its rate of supply is far from meeting needs. Here, we provide an effective strategy to produce PDGF-BB in large quantities using a transgenic silkworm. The codon-optimized PDGF-B gene regulated by the highly efficient sericin-1 expression system was integrated into the genome of a silkworm. The high transcriptional expression of the PDGF-BB gene in the transgenic silkworm competitively inhibited the transcription expression of the endogenous sericin-1 gene which caused a significant 37.5% decline. The PDGF-BB synthesized in the middle silk gland (MSG) of transgenic silkworms could form a homodimer through intermolecular disulfide bonds, which is then secreted into sericin lumen and finally, distributed in the sericin layer of the cocoon. In this study, a protein quantity of approximately 0.33 mg/g was found in the cocoon. Following a purification process, approximately 150.7 μg of recombinant PDGF-BB with a purity of 82% was purified from 1 g of cocoons. Furthermore, the bioactivity assays showed that the purified recombinant PDGF-BB was able to promote the growth, proliferation and migration of NIH/3T3 cells significantly. These results suggest that the silk gland bioreactor can produce active recombinant PDGF-BB as an efficient mitogen and wound healing agent. [ABSTRACT FROM AUTHOR]

Ganoderma triterpenes (GTs) are the major secondary metabolites of Ganoderma lucidum , which is a popularly used traditional Chinese medicine for complementary cancer therapy. The present study was to establish a fingerprint evaluation system based on Similarity Analysis (SA), Cluster Analysis (CA) and Principal Component Analysis (PCA) for the identification and quality control of G. lucidum . Fifteen samples from the Chinese provinces of Hainan, Neimeng, Shangdong, Jilin, Anhui, Henan, Yunnan, Guangxi and Fujian were analyzed by HPLC-PAD and HPLC-MS n . Forty-seven compounds were detected by HPLC, of which forty-two compounds were tentatively identified by comparing their retention times and mass spectrometry data with that of reference compounds and reviewing the literature. Ganoderic acid B, 3,7,15-trihydroxy-11,23-dioxolanost-8,16-dien-26-oic acid, lucidenic acid A, ganoderic acid G, and 3,7-oxo-12-acetylganoderic acid DM were deemed to be the marker compounds to distinguish the samples with different quality according to both CA and PCA. This study provides helpful chemical information for further research on the anti-tumor activity and mechanism of action of G. lucidum . The results proved that fingerprints combined with chemometrics are a simple, rapid and effective method for the quality control of G. lucidum .

Danshen, the dried root of Salvia miltiorrhiza Bge., is a widely used commercially available herbal drug, and unstable quality of different samples is a current issue. This study focused on a comprehensive and systematic method combining fingerprints and chemical identification with chemometrics for discrimination and quality assessment of Danshen samples. Twenty-five samples were analyzed by HPLC-PAD and HPLC-MS n . Forty-nine components were identified and characteristic fragmentation regularities were summarized for further interpretation of bioactive components. Chemometric analysis was employed to differentiate samples and clarify the quality differences of Danshen including hierarchical cluster analysis, principal component analysis, and partial least squares discriminant analysis. Consistent results were that the samples were divided into three categories which reflected the difference in quality of Danshen samples. By analyzing the reasons for sample classification, it was revealed that the processing method had a more obvious impact on sample classification than the geographical origin, it induced the different content of bioactive compounds and finally lead to different qualities. Cryptotanshinone, trijuganone B, and 15,16-dihydrotanshinone I were screened out as markers to distinguish samples by different processing methods. The developed strategy could provide a reference for evaluation and discrimination of other traditional herbal medicines.

Ganoderma triterpenes (GTs) are the major secondary metabolites of Ganoderma lucidum, which is a popularly used traditional Chinese medicine for complementary cancer therapy. In the present study, systematic isolation, and in silico pharmacological prediction are implemented to discover potential anti-cancer active GTs from G. lucidum. Nineteen GTs, three steroids, one cerebroside, and one thymidine were isolated from G. lucidum. Six GTs were first isolated from the fruiting bodies of G. lucidum, including 3β,7β,15β-trihydroxy-11,23-dioxo-lanost-8,16-dien-26-oic acid methyl ester (1), 3β,7β,15β-trihydroxy-11,23-dioxo-lanost-8,16-dien-26-oic acid (2), 3β,7β,15α,28-tetrahydroxy-11,23-dioxo-lanost-8,16-dien-26-oic acid (3), ganotropic acid (4), 26-nor-11,23-dioxo-5α-lanost-8-en-3β,7β,15α,25-tetrol (5) and (3β,7α)-dihydroxy-lanosta-8,24-dien- 11-one (6). (4E,8E)-N-d-2'-hydroxypalmitoyl-l-O-β-d-glucopyranosyl-9-methyl-4,8-spingodienine (7), and stigmasta-7,22-dien-3β,5α,6α-triol (8) were first reported from the genus Ganodema. By using reverse pharmacophoric profiling of the six GTs, thirty potential anti-cancer therapeutic targets were identified and utilized to construct their ingredient-target interaction network. Then nineteen high frequency targets of GTs were selected from thirty potential targets to construct a protein interaction network (PIN). In order to cluster the pharmacological activity of GTs, twelve function modules were identified by molecular complex detection (MCODE) and gene ontology (GO) enrichment analysis. The results indicated that anti-cancer effect of GTs might be related to histone acetylation and interphase of mitotic cell cycle by regulating general control non-derepressible 5 (GCN5) and cyclin-dependent kinase-2 (CDK2), respectively. This research mode of extraction, isolation, pharmacological prediction, and PIN analysis might be beneficial to rapidly predict and discover pharmacological activities of novel compounds.
