Motivated by studies of speech deficits in humans, several studies over the past two decades have investigated the potential role of a forkhead domain transcription factor, FoxP2, in the central control of acoustic signaling/vocalization among vertebrates. Comparative neuroanatomical studies that mainly include mammalian and avian species have mapped the distribution of FoxP2 expression in multiple brain regions that imply a greater functional significance beyond vocalization that might be shared broadly across vertebrate lineages. To date, reports for teleost fish have been limited in number and scope to nonvocal species. Here, we map the neuroanatomical distribution of FoxP2 mRNA expression in a highly vocal teleost, the plainfin midshipman (Porichthys notatus). We report an extensive overlap between FoxP2 expression and vocal, auditory, and steroid-signaling systems with robust expression at multiple sites in the telencephalon, the preoptic area, the diencephalon, and the midbrain. Label was far more restricted in the hindbrain though robust in one region of the reticular formation. A comparison with other teleosts and tetrapods suggests an evolutionarily conserved FoxP2 phenotype important to vocal-acoustic and, more broadly, sensorimotor function among vertebrates. Among fishes, acoustic communication is best studied in toadfishes, a single order and family that includes species commonly known as toadfish and midshipman. However, there is a lack of comparative anatomical and physiological studies, making it difficult to identify both shared and derived mechanisms of vocalization among toadfishes. Here, vocal nerve labeling and intracellular in vivo recording and staining delineated the hindbrain vocal network of the Gulf toadfish Opsanus beta. Dextran-biotin labeling of the vocal nerve or intracellular neurobiotin fills of motoneurons delineated a midline vocal motor nucleus (VMN). Motoneurons showed bilaterally extensive dendritic arbors both within and lateral to the paired motor nuclei. The motoneuron activity matched that of the spike-like vocal nerve motor volley that determines the natural call duration and frequency. Ipsilateral vocal nerve labeling with biocytin or neurobiotin yielded dense bilateral transneuronal filling of motoneurons and coextensive columns of premotor neurons. These premotor neurons generated pacemaker-like action potentials matched 1:1 with vocal nerve and motoneuron firing. Transneuronal transport further revealed connectivity within and between the pacemaker-motor circuit and a rostral prepacemaker nucleus. Unlike the pacemaker-motor circuit, prepacemaker firing did not match the frequency of vocal nerve activity but instead was predictive of the duration of the vocal nerve volley that codes for call duration. Transneuronally labeled terminal-like boutons also occurred in auditory-recipient hindbrain nuclei, including neurons innervating the inner ear and lateral line organs. Together with studies of midshipman, we propose that separate premotor populations coding vocal frequency and duration with direct premotor coupling to auditory-lateral line nuclei are plesiomorphic characters for toadfishes. Unlike in midshipman, transneuronal labeling in toadfishes reveals an expansive column of pacemaker neurons that is weakly coupled to prepacemaker neurons, a character that likely depends on the extent of gap junction coupling. We propose that these and other anatomical characters contribute to neurophysiological properties that, in turn, sculpt the species-typical patterning of frequency and amplitude-modulated vocalizations. © 2014 S. The neuropeptide arginine vasotocin (AVT) is well known to modulate both aggression and affiliation, yet few studies relate individual behavioral state to a quantitative assessment of AVT distribution in the brain. Here, using a wild population of beaugregory damselfish, Stegastes leucostictus, we assess: (1) the effect of AVT on courtship, and (2) with reference to our previous study on AVT modulation of aggression in this species, the relationship between AVT-like immunoreactive (ir) fiber distribution in the forebrain’s preoptic area and individual courtship and aggression levels. Exogenous AVT did not affect courtship, yet Manning compound, an arginine vasopressin (AVP) V1a receptor antagonist, significantly lowered but did not eradicate courtship. Consistent with AVT’s known facilitation of aggression in this species, the density of AVT-ir fibers in the preoptic area was significantly negatively correlated to aggression. Our findings match similar behavioral and immunoreactive patterns of neuropeptide secretion in other taxa. Unlike aggression, preoptic AVT-ir fiber density was not significantly correlated to individual courtship levels. Sound production has evolved independently a number of times among teleost fishes. In most cases, sound is generated by fast contracting muscles that vibrate the swim bladder by way of their direct attachment (intrinsic muscles) or indirectly by way of other skeletal elements (extrinsic muscles). This study focuses on the red and black piranha, Pygocentrus nattereri and Serrasalmus rhombeus (superorder Ostariophysi, Order Characiformes), that have extrinsic swim bladder sonic muscles innervated by the third and fourth spinal nerves. This innervation pattern diverges from that found in most teleosts, including the closely related catfishes (Ostariophysi, Siluriformes), where sonic muscles are innervated by ventral occipital nerve roots that arise just caudal to the vagus nerve. Here, we tested the hypothesis that piranhas would also differ from most other teleosts in the location of their sonic motor neurons. Following biotin labeling of branches of the third and fourth spinal nerves that innervate the sonic muscles in the red and black piranha, sonic motor neurons were identified amongst other non-sonic motor neurons in the central part of the spinal cord, slightly ventrolateral to the central canal. To our knowledge, this is the first example of sonic motor neurons positioned entirely within the spinal cord. In the other species so far studied, sonic motor neurons form well-defined nuclei that extend from far caudal levels of the medulla into the rostral spinal cord and are located either within the ventral motor column or near the midline, close to or just ventrolateral to the fourth ventricle and central canal. A piranha-like pattern may be more widespread among characiforms and is likely present in other teleost orders, e.g., Sciaenidae (drumfishes), that also have sonic muscles innervated by spinal nerves. Teleost fishes are the ‘champions’ of sexual plasticity among vertebrates. Several species have two male reproductive morphs with distinct suites of behavioral, somatic, neuronal, endocrinological, and life history traits. Here, we consider recent studies of the social and neural modulation of sexual plasticity for such species with a focus on two neuropeptides, gonadotropin releasing hormone (GnRH) and arginine vasotocin (AVT, teleostean analogue of mammalian arginine vasopressin). The major premise of this review is that phenotypic changes in GnRH and AVT expression in the brain can orchestrate events leading to changes in either sexual status or the expression of morph specific display behaviors important in reproduction.Copyright © 2001 S. The organization of the descending and secondary octaval nuclei in the hindbrain of the Gulf toadfish, Opsanus beta, was revealed following the injection of biotin compounds into a physiologically identified auditory region of the torus semicircularis. The results show retrogradely-filled neurons mainly in a dorsomedial division of the descending octaval nucleus, and dorsal and ventral divisions of a secondary octaval nucleus; minor labeling also appeared in dorsolateral and rostromedial intermediate divisions of the descending nucleus. The pattern identified is consistent with that reported in other teleosts, including both vocal and non-vocal species, and clarifies earlier reports of the organization of hindbrain octaval nuclei in toadfish and the closely related midshipman fish.Copyright © 2001 S. Similar to many teleost fish, squirrelfish (family Holocentridae) produce vocalizations by the contraction of muscles that lead to vibration of the swimbladder. We used biotinylated compounds to identify the position and extent of vocal motor neurons in comparison to additional motor neuron groups, namely those of red and white dorsal epaxial muscle and opercular muscle that are located adjacent to or near the sonic muscle. The sonic motor nucleus (SMN) was located in the caudal medulla and rostral spinal cord in a ventrolateral position with dendrites extending dorsally in a dense bundle along the lateral edge of the medulla and axons exiting via ventral occipital nerve roots. Transneuronal transport of biocytin identified premotor neurons within the SMN and in the medially adjacent reticular formation that projected to the contralateral SMN and more rostrally to the octavolateralis efferent nucleus and nucleus praeeminentialis, suggesting interactions between vocal and octavolateralis systems as seen in other teleosts. Motor neurons innervating the red and white dorsal muscle formed a loose aggregate in the dorsal motor column, adjacent to the medial longitudinal fasciculus, sending fibers bilaterally throughout the spinal cord with axons exiting via ventral spinal nerve roots. Opercular motor neurons were located within the facial motor nucleus. The anatomical characteristics of the SMN of squirrelfish, a representative member of the order Beryciformes, are similar to those of representative members of the closely related order Scorpaeniformes, but diverge from the SMN of more distantly related orders of paracanthopterygiian and ostariophysan teleosts. Among teleost fishes, representatives of several distantly related groups have sound-producing (sonic/vocal) muscles associated with the swimbladder or pectoral girdle/fin. Here, the diversity of vocal organs and central motor pathways in four families of catfish, order Siluriformes, is compared to that in families from two distantly related orders, the Scorpaeniformes and Batrachoidiformes. Several catfish families have two sonic mechanisms – a swimbladder vibration established by ‘drumming muscles’ that differ in origin and insertion between families, and a pectoral spine stridulatory apparatus. In ariids, mochokids and doradids, sonic swimbladder muscles originate at various cranial or postcranial elements and insert onto an ‘elastic spring’ that vibrates the swimbladder, while in pimelodids the muscles insert ventrally at the swimbladder. Sonic motoneurons are located along the midline, ventral to the fourth ventricle/central canal in doradids and mochokids but lateral to the medial longitudinal fasciculus in ariids; pimelodids have motoneurons in both locations. The axonal trajectory for the lateral motoneurons in pimelodids and ariids implies that they are a migrated, midline population of sonic motoneurons. Pectoral spine-associated motoneurons are located in the ventral motor column. Unlike catfishes, a diversity of sonic mechanisms in Scorpaeniformes is not associated with different positions for sonic motoneurons. Cottids (sculpin) lack a swimbladder but have sonic muscles that originate at the occipital cranium and insert at the pectoral girdle; sonic motoneurons are located within the ventral motor column. Some triglids have intrinsic swimbladder muscles, although ontogenetic data indicate a transient association with the pectoral girdle; sonic motoneurons are in the same location as in cottids. Among Batrachoidiformes, all known representatives have intrinsic swimbladder muscles that are never associated with the pectoral girdle and are innervated by midline sonic motoneurons. The results suggest two patterns of organization for sound-producing systems in teleost fishes: pectoral fin/girdle-associated muscles are innervated by sonic motoneurons positioned within the ventral motor column, adjacent to the ventral fasciculus; non-pectoral associated muscles are innervated by sonic motoneurons located on or close to the midline, adjacent to the medial longitudinal fasciculus.
