转化生长因子 β （TGF-β） 是一种多功能生长因子, 既参与调控细胞增殖分化和基因表 达, 也 是 组 织 修 复 和 纤 维 化 反 应 中 的 中 枢 效 应 因 子, 在 纤 维 化 疾 病 的 发 生 发 展 中 发 挥 重 要 作 用。 TGF-β 通常以潜伏复合物的形式分泌并储存于细胞外基质 （ECM） 中, 被激活后通过经典的 Smad 通 路和丝裂原活化蛋白激酶 （MAPK） 等非 Smad 通路发挥作用。在遗传性牙龈纤维瘤、药物性牙龈增 生和口腔黏膜下纤维化等口腔软组织纤维化疾病中, TGF-β 被激活且高表达, 其在通过刺激Ⅰ型胶原 增生、诱导肌成纤维细胞分化和促进细胞外基质 （ECM） 合成的同时抑制 ECM 降解, 使 ECM 过度 沉积进而导致纤维化。现结合近年来国内外相关研究, 分析 TGF-β 活化及信号传导功能, 并对其在遗 传性牙龈纤维瘤、药物性牙龈增生和口腔黏膜下纤维化发生发展中的影响进行阐述, 旨在为口腔软组 织纤维化疾病的机制研究和精准治疗提供新的思路和方向。 [ABSTRACT FROM AUTHOR]
