4. Preparation and Separation of Angiotensin I Converting Enzyme Inhibitory Peptides

Abstract

イワシすり身のプロテアーゼ分解物からACE阻害活性を有するペプチドを調製する際の分解条件,精製法について検討し,さらに得られたペプチドのin vivoにおける活性を測定して次の結果を得た. (1) 分解に用いるプロテアーゼとしてペプシン,トリプシン,キモトリプシン,デナチームAPについて検討した結果,最も高い活性はペプシン分解物に認められた. (2) プロテアーゼによる加水分解前に,イワシ蛋白質を加熱処理することにより,未処理のものよりも高いACE阻害活性を有する分解液を得ることができた. (3) 予備加熱を行ったイワシ蛋白質のペプシンによる加水分解液をODS充填カラムに通し, 25%エタノールで溶出することにより,高いACE阻害活性(IC50=0.064mg protein/ml)を有するペプチドを一段階の精製操作で得ることができた. (4) ODSカラムによる一段階精製で得られたペプチドはラットへの静脈内投与により,アンジオテンシンIに対する血圧上昇を明らかに抑制する効果が認められた.
