Purpose This study was designed to expound the underlying mechanism of microtubule-directed chemotherapeutic drugs resistance induced by cancer-associated fibroblasts (CAFs) in breast cancer. Materials and methods We collected 10 microtubule-directed chemotherapeutic drugs resistant breast tumor samples and 10 normal breast tumor samples to analyze the CAFs distribution by immunohistochemistry and flow cytometry. We also detected the collagen expression in CAFs by real-time PCR. We detected the activation of PI3K/AKT signaling pathway in tumor cells by Western blotting and immunofluorescence. The subcutaneous 4T1/MCF-7 bearing mice were used to investigate the anticancer effects of integrin β1 inhibitor combined with microtubule-directed chemotherapeutic drugs. Results In our studies, accumulation of CAFs was observed in tumor samples from microtubule-directed chemotherapeutic drugs resistant patients. Those isolated CAFs could efficiently induce the acquisition of microtubule-directed chemotherapeutic drugs resistance in breast cancer cells. More importantly, we found that CAFs could regulate the microtubule-directed chemotherapeutic drugs resistance through the secretion of collagen to activate the integrin β1/PI3K/AKT signaling pathway. Combination of integrin α2β1 inhibitor and paclitaxel/vincristine sulfate could efficiently overcome the microtubule-directed chemotherapeutic drugs resistance induced by CAFs and enhanced the anticancer effects of chemotherapy in subcutaneous 4T1/MCF-7 bearing mice. Conclusion Our results demonstrated that CAFs constitute a supporting niche for cancer drug resistance acquisition. Thus, traditional microtubule-directed chemotherapeutic drugs combined with integrin β1 inhibitor may present an innovative therapeutic strategy for breast cancer therapy.

Sea urchins (Echinoidea) are key components of marine benthic communities and many are commercially important fishery resources as luxury and healthy seafood. However, despite their high ecological and economic value, the mitochondrial genomes of all sea urchins have yet to be analyzed. In this study, we report the first complete mitochondrial genome of Stomopneustidae from Stomopneustes variolaris. The mitogenome has 15,767 base pairs (59.77% A + T content) and contains 37 genes (13 protein-coding, 22 transfer RNAs and 2 ribosomal RNAs), plus a putative control region. This study provides useful molecular resources for clarifying evolutionary and phylogenetic histories of sea urchins.

In this research, additive friction stir processing (AFSP) was successfully employed to prepare the aluminum–tungsten carbide (Al–WC) surface composite upon 5083Al substrate. The resulting microstructures were detailedly characterized by X-ray diffraction, optical microscopy (OM), scanning electron microscopy, energy dispersive spectroscopy and electron backscattered diffraction. Microhardness tests and wear tests were used to evaluate the surface properties of the AFSP surface composite. Results showed that the prepared Al–WC surface composite by AFSP had a combination of homogeneous distribution of WC particles, excellent WC/Al interfacial integrity and remarkably refined grains. The continuous dynamic recrystallization (DRX) mechanism was mainly responsible for the grain refining of 5083Al during FSP. Friction stir welding (FSW) was performed on 304 stainless steel sheets. The welding speed was varied from 30 to 70 mm/min. Because the FSW of thin plates is sensitive to heat input. The surface of the welds is smooth and without any macroscopic cracks and cavities only when the rotation speed is 50 mm/min. The stir zone had equiaxed grains with austentic structure due to dynamic recrystallization. There was not any evidence for the presence of sigma phases being found in this study. The black sea cucumber, Holothuria leucospilota, is an ecologically and economically important holothuroid in China due to broader environmental adaptation and valuable pharmacological compounds. However, the wild stocks of H. leucospilota, have been declining steadily due to overexploitation in recent years. The stock enhancement programme has become an increasingly important priority. In this study, we report the complete mitochondrial genome of H. leucospilota. The mitogenome has 15,839 base pairs (57.6% A + T content) and made up of total of 37 genes (13 protein-coding, 22 transfer RNAs and 2 ribosomal RNAs), and a putative control region. The complete mitogenomes of H. leucospilota will provide useful genetic information for future conservation and management of this valuable and vulnerable species.

The sea cucumber, Stichopus monotuberculatus, is an economically important holothuroid in China due to its larger body size and valuable nutrition. However, the taxonomic revision studies of Stichopodidae have been one of the most controversial issues in recent years. Moreover, there remain considerable doubts about a complex of cryptic species within S. monotuberculatus. In this study, we report the complete mitochondrial genome of S. monotuberculatus. The mitogenome has 16,274 base pairs (60.4% A + T content) and made up of total of 37 genes (13 protein-coding, 22 transfer RNAs and 2 ribosomal RNAs), and a putative control region. This study adds one more available complete mitogenomes of Stichopus and will provide useful genetic information for future evolutionary and taxonomic classification of Stichopodidae.

Hemifusus tuba is an ecologically and economically important species of Buccinoidea, which comprises an ecologically diverse group of carnivorous marine gastropods. However, the taxonomic classification and phylogenetic studies have so far been limited. In this study, we report the first complete mitochondrial genome of Melongenidae from H. tuba. The mitogenome has 15,483 bp (68.2% A + T content) and made up of total of 37 genes (13 protein-coding, 22 transfer RNAs, and 2 ribosomal RNAs), and a control region. This study was the first to provide complete mitogenome of Melongenidae and will provide useful genetic information for future phylogenetic and taxonomic classification of Buccinoidea.
