Introduction Solid lipid nanoparticles (SLNs) are considered a promising system in enhancing the oral bioavailability of poorly water-soluble drugs; owing to their intrinsic ability to increase the solubility together with protecting the incorporated drugs from extensive metabolism. Objective Exploiting such properties, SLNs loaded with gliclazide (GLZ) were developed in an attempt to improve the oral bioavailability and the anti-diabetic action of GLZ, together with prolonging its duration of action for better glycemic control. Methods SLNs were prepared by ultra-sonication technique using glyceryl behenate (Compritol®888 ATO) as a lipid matrix and poloxamer 188 (PLX) as a stabilizer. A 2*3 asymmetrical factorial design was adopted to study the effect of different stabilizer concentrations at different sonication times on the shape, and size of the particles, PDI and drug loading. The selected optimum formulation was then freeze dried using trehalose di-hydrate as a cryo-protectant in different ratios with respect to glyceryl behenate concentration. After freeze drying, the formulation was tested for in-vitro drug release, pharmacokinetics, and pharmacodynamics. Safety of the selected formula was established after carrying out a subacute toxicity study. Results The factorial design experiment resulted in an optimum formulation coded 10F2 (150 mg PLX/10 min sonication). Scanning electron micrographs showed spherical particles with smooth surface, whereas a ratio of 2:1 cryo-protectant:lipid was found to be optimum with particle size of 245.9 ± 26.2 nm, polydispersity index of 0.482 ± 0.026, and biphasic in-vitro release with an initial burst effect, followed by a prolonged release phase. On the other hand, the selected SLNs exhibited prolonged drug release when compared with the GLZ commercial immediate release (IR) tablets (Diamicron®). Pharmacokinetics study showed about 5-fold increase in GLZ oral bioavailability loaded in SLNs when compared with raw GLZ powder. Pharmacodynamics study on a diabetic rat model confirmed the better anti-diabetic action of GLZ loaded SLNs when compared to raw GLZ powder. Subacute toxicity study indicated the safety of SLNs upon repetitive oral administration.

Antibiotic resistance is an increasing worldwide crisis. In Egypt, it is prohibited by law to dispense antibiotics with no prescription; however, there are no active and enforced governmental policies and laws to regulate such a process. This has led to increased antibiotic dispensing and consumption.To assess some basic knowledge and attitudes towards antibiotic resistance and the practice of antibiotic dispensing among Egyptian community pharmacists in Greater Cairo.An observational cross-sectional self-administrated survey targeting licensed community pharmacists in Greater Cairo, Egypt. The survey was validated by both face and content validation.Exactly 531 pharmacists were invited to join the survey, and only 461 pharmacists agreed to participate. Approximately half of the participants (51.2%) were considered to have good basic knowledge regarding antibiotic use and antibiotic resistance. The majority (74%) of the respondents strongly agreed that antibiotics are over-prescribed in Egypt, and almost all of them (90%) were hoping the Ministry of Health could publish guidelines for antibiotic dispensing in Egypt. Male pharmacists were more likely to engage and communicate with either patient or prescriber during the antibiotic dispensing process compared to females.The Egyptian Ministry of Health should regulate and monitor the process of dispensing antibiotics without a prescription with law enforcements. Moreover, community pharmacists should have more awareness on the consequences of the problem of antibiotic dispensing without a prescription, regardless of the business profit behind it.

The effect of graphene nanosheets (GNS) on the rheological characteristics and thermal conductivity of calcium grease used in marine applications, and water pumps for lower demanding applications, has been experimentally evaluated in this paper. Various volume fractions of GNS (0.5, 1, 2, 3, and 4% wt.) have been immersed in the grease aiming at finding the better percentages that improve the properties of nanogrease. The rheological characteristics and thermal conductivity were evaluated with a Brookfield Rheometer DV-III ULTRA and KD2 thermal analyzer, respectively. The results of the modified calcium grease indicated that the best concentration of GNS is 3% wt. The rheological characteristics of the grease with a different volume fraction of GNS indicated a non-Newtonian behavior. Thermal conductivity of nanogreas also increases with an increase of GNS volume fraction. The addition of graphene nanosheets (GNSs) in lubricating grease could significantly reduce the interfacial friction and improve the load-bearing capacity of the parts. Therefore, it has been considered as having great potential as lubricant additives. In this study, we synthesized GNSs that are prepared by a modified Hummer method, and investigated the effect of GNS with different concentration (0.5%, 1%, 2%, 3%, and 4 wt%) on the tribological properties of the calcium grease. The friction and wear experiments were performed using a four-ball tribometer. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) were employed to examine the GNS and the friction mechanisms. This study examined the tribological properties of two lubricating oils, mobil gear 627 and paraffinic mineral oils, with multi-walled carbon nanotubes (MWCNTs) nanoparticles used as additives with various concentrations (0.1, 0.5, 1, and 2 wt.%). The friction and wear experiments were performed using a four ball tribotester. The samples were tested for their anti-wear, load carrying capacity, and friction coefficients according to ASTM D-2783, ASTM D-2596, and ASTM D-5183 standards. The experimental results show that the addition of MWCNTs to base oils exhibit good friction reduction and anti-wear properties. The wear test results show a decreased wear by 68% and 39% in the case of MWCNTs-based mineral oil as compared with base mobil gear 627 and paraffinic mineral oils, respectively. The cooling towers are reinforced concrete structures affected by different ageing mechanisms. The material data, the loading actions and the environmental conditions are subject to important fluctuations. It is necessary to have efficient tools allowing to evaluate and to supervise the reliability level of these structures, by taking account for these fluctuations as well as the available measurements on the structure. We propose herewith to show the capability of the reliability methods to evaluate the failure risk of the reinforced concrete shells in function of the amplification of the deformation measured in-situ.

The mechanical-reliability combination represents a link between finite element and reliability procedures, in such a way to obtain the safety evaluation by the most efficient method. In this combination scheme, the reliability analysis is very sensitive to the mechanical modeling accuracy: the mechanical error may be reduced, or, mostly, amplified during the reliability analysis. In practical engineering, the desired mechanical accuracy should not be limited to deterministic requirements but should take account for the filtration effect in the mechanical-reliability combination. In this paper, we develop, for linear elasto-static behavior, the filtration factors, illustrated by numerical example of thick sphere under pressure.
