The green alga Chlamydomonas reinhardtii can use the ureides allantoin and allantoate as sole nitrogen sources. Once the uptake systems for allantoin and allantoate were induced, the uptake and growth rates were identical for the two ureides. However, the enzymatic activities involved in the degradation of the two ureides (allantoinase and allantoicase) were regulated differently. Allantoinase seems to be constitutive, since it was detected in all the nitrogen sources studied, while allantoicase behaved as an inducible enzyme, since it was present only in cells cultured in ureides or any metabolic precursor of these compounds. Neither allantoinase nor allantoicase activities were repressed by ammonium in the presence of ureides. Allantoicase activity was not induced under nitrogen starvation conditions, while it was induced in cells that had been cultured with allantoin or allantoate in the dark. This report describes the isolation of the genes encoding allantoicase (DAL2) and ureidoglycolate hydrolase (DAL3), which are components of the large DAL gene cluster on the right arm of chromosome IX of Saccharomyces cerevisiae. During this work a new gene (DAL7) was identified and found to be regulated in the manner expected for an allantoin pathway gene. Its expression was (i) induced by allophanate, (ii) sensitive to nitrogen catabolite repression, and (iii) responsive to mutation of the DAL80 and DAL81 loci, which have previously been shown to regulate the allantoin degradation system. Hybridization probes generated from these cloned genes were used to analyze expression of the allantoin pathway genes in wild-type and mutant cells grown under a variety of physiological conditions. When comparison was possible, the patterns of mRNA and enzyme levels observed in various strains and physiological conditions were very similar, suggesting that the system is predominantly regulated at the level of gene expression. Although all of the genes seem to be controlled by a common mechanism, their detailed patterns of expression were, at the same time, highly individual and diverse.

A remarkable accumulation and utilization of allantoin are observed in soybean plant bearing modules (Glycine max variety A62-1). To study ita physiological role, changes in activities and distribution of the enzymes concerning the purine catabolism (i.e., xanthine oxidase, uricase, allantoinase and allantoicase) were measured during development. Uricase activity was found in the radicles in the early stage of seedling development, and no difference of the enzyme activity was detected between the radicles of the nodulating variety (A62-) and non-nodulating variety (A62-2). On maturation the activity disappeared rapidly in radicles and appeared in leaves, pods and nodules. The activity in nodules was much higher than that in the leaves and the pods throughout the culture period. Xanthine oxidase activity was found in the radicles and nodules.
