In order to build hippocampal prostheses for restoring memory functions, we build sparse multi-input, multi-output (MIMO) nonlinear dynamical models of the human hippocampus. Spike trains are recorded from hippocampal CA3 and CA1 regions of epileptic patients performing a variety of memory-dependent delayed match-to-sample (DMS) tasks. Using CA3 and CA1 spike trains as inputs and outputs respectively, sparse generalized Laguerre-Volterra models are estimated with group lasso and local coordinate descent methods to capture the nonlinear dynamics underlying the CA3-CA1 spike train transformations. These models can accurately predict the CA1 spike trains based on the ongoing CA3 spike trains during multiple memory events, e.g., sample presentation, sample response, match presentation and match response, of the DMS task, and thus will serve as the computational basis of human hippocampal memory prostheses. Objective: As an extension to our study comparing a putative compartmental and data-based model of linear dynamic cerebral autoregulation (CA) and CO2-vasomotor reactivity (VR), we study the CA–VR process in a nonlinear context. Methods: We use the concept of principal dynamic modes (PDM) in order to obtain a compact and more easily interpretable input–output model. This in silico study permits the use of input data with a dynamic range large enough to simulate the classic homeostatic CA and VR curves using a putative structural model of the regulatory control of the cerebral circulation. The PDM model obtained using theoretical and experimental data are compared. Results: It was found that the PDM model was able to reflect accurately both the simulated static CA and VR curves in the associated nonlinear functions (ANFs). Similar to experimental observations, the PDM model essentially separates the pressure-flow relationship into a linear component with fast dynamics and nonlinear components with slow dynamics. In addition, we found good qualitative agreement between the PDMs representing the dynamic theoretical and experimental CO2-flow relationship. Conclusion: Under the modeling assumption and in light of other experimental findings, we hypothesize that PDMs obtained from experimental data correspond with passive fluid dynamical and active regulatory mechanisms. Significance: Both hypothesis-based and data-based modeling approaches can be combined to offer some insight into the physiological basis of PDM model obtained from human experimental data. The scientific and clinical importance of cerebral hemodynamics has generated considerable interest in their quantitative understanding via computational modeling. In particular, two aspects of cerebral hemodynamics, cerebral flow autoregulation (CFA) and CO2 vasomotor reactivity (CVR), have attracted much attention because they are implicated in many important clinical conditions and pathologies (orthostatic intolerance, syncope, hypertension, stroke, vascular dementia, mild cognitive impairment, Alzheimer's disease, and other neurodegenerative diseases with cerebrovascular components). Both CFA and CVR are dynamic physiological processes by which cerebral blood flow is regulated in response to fluctuations in cerebral perfusion pressure and blood CO2 tension. Several modeling studies to date have analyzed beat-to-beat hemodynamic data in order to advance our quantitative understanding of CFA–CVR dynamics. A confounding factor in these studies is the fact that the dynamics of the CFA–CVR processes appear to vary with time (i.e., changes in cerebrovascular characteristics) due to neural, endocrine, and metabolic effects. This paper seeks to address this issue by tracking the changes in linear time-invariant models obtained from short successive segments of data from ten healthy human subjects. The results suggest that systemic variations exist but have stationary statistics and, therefore, the use of time-invariant modeling yields “time-averaged models” of physiological and clinical utility. A major factor involved in providing closed loop feedback for control of neural function is to understand how neural ensembles encode online information critical to the final behavioral endpoint. This issue was directly assessed in rats performing a short-term delay memory task in which successful encoding of task information is dependent upon specific spatio-temporal firing patterns recorded from ensembles of CA3 and CA1 hippocampal neurons. Such patterns, extracted by a specially designed nonlinear multi-input multi-output (MIMO) nonlinear mathematical model, were used to predict successful performance online via a closed loop paradigm which regulated trial difficulty (time of retention) as a function of the “strength” of stimulus encoding. The significance of the MIMO model as a neural prosthesis has been demonstrated by substituting trains of electrical stimulation pulses to mimic these same ensemble firing patterns. This feature was used repeatedly to vary “normal” encoding as a means of understanding how neural ensembles can be “tuned” to mimic the inherent process of selecting codes of different strength and functional specificity. The capacity to enhance and tune hippocampal encoding via MIMO model detection and insertion of critical ensemble firing patterns shown here provides the basis for possible extension to other disrupted brain circuitry. The successful development of neural prostheses requires an understanding of the neurobiological bases of cognitive processes, i.e., how the collective activity of populations of neurons results in a higher level process not predictable based on knowledge of the individual neurons and/or synapses alone. We have been studying and applying novel methods for representing nonlinear transformations of multiple spike train inputs (multiple time series of pulse train inputs) produced by synaptic and field interactions among multiple subclasses of neurons arrayed in multiple layers of incompletely connected units. We have been applying our methods to study of the hippocampus, a cortical brain structure that has been demonstrated, in humans and in animals, to perform the cognitive function of encoding new long-term (declarative) memories. Without their hippocampi, animals and humans retain a short-term memory (memory lasting approximately 1 min), and long-term memory for information learned prior to loss of hippocampal function. Results of more than 20 years of studies have demonstrated that both individual hippocampal neurons, and populations of hippocampal cells, e.g., the neurons comprising one of the three principal subsystems of the hippocampus, induce strong, higher order, nonlinear transformations of hippocampal inputs into hippocampal outputs. For one synaptic input or for a population of synchronously active synaptic inputs, such a transformation is represented by a sequence of action potential inputs being changed into a different sequence of action potential outputs. In other words, an incoming temporal pattern is transformed into a different, outgoing temporal pattern. For multiple, asynchronous synaptic inputs, such a transformation is represented by a spatiotemporal pattern of action potential inputs being changed into a different spatiotemporal pattern of action potential outputs. Our primary thesis is that the encoding of short-term memories into new, long-term memories represents the collective set of nonlinearities induced by the three or four principal subsystems of the hippocampus, i.e., entorhinal cortex-to-dentate gyrus, dentate gyrus-to-CA3 pyramidal cell region, CA3-to-CA1 pyramidal cell region, and CA1-to-subicular cortex. This hypothesis will be supported by studies using in vivo hippocampal multineuron recordings from animals performing memory tasks that require hippocampal function. The implications for this hypothesis will be discussed in the context of "cognitive prostheses"--neural prostheses for cortical brain regions believed to support cognitive functions, and that often are subject to damage due to stroke, epilepsy, dementia, and closed head trauma. The article presents a study that examines the role of automatic neural control on carbon dioxide reactivity and the dynamic pressure autoregulation in the frequency range between 0.005 and 0.40 hertz. The study analyzed the experimental data from healthy persons with the use of a nonlinear multivariate modeling approach. Results show that mean artevial blood pressure (MABP) and the mean cerebral blood flow velocity (MCBFV) decrease during ganglion blockade.

This paper presents the results of a computational study that compares simulated compartmental (differential equation) and Volterra models of the dynamic effects of insulin on blood glucose concentration in humans. In the first approach, we employ the widely accepted "minimal model" and an augmented form of it, which incorporates the effect of insulin secretion by the pancreas, in order to represent the actual closed-loop operating conditions of the system, and in the second modeling approach, we employ the general class of Volterra-type models that are estimated from input-output data. We demonstrate both the equivalence between the two approaches analytically and the feasibility of obtaining accurate Volterra models from insulin--glucose data generated from the compartmental models. The results corroborate the proposition that it may be preferable to obtain data-driven (i.e., inductive) models in a more general and realistic operating context, without resorting to the restrictive prior assumptions and simplifications regarding model structure and/or experimental protocols (e.g., glucose tolerance tests) that are necessary for the compartmental models proposed previously. These prior assumptions may lead to results that are improperly constrained or biased by preconceived (and possibly erroneous) notions--a risk that is avoided when we let the data guide the inductive selection of the appropriate model within the general class of Volterra-type models, as our simulation results suggest. The increasing availability of multiunit recordings gives new urgency to the need for effective analysis of "multidimensional" time-series data that are derived from the recorded activity of neuronal ensembles in the form of multiple sequences of action potentials-treated mathematically as point-processes and computationally as spike-trains. Whether in conditions of spontaneous activity or under conditions of external stimulation, the objective is the identification and quantification of possible causal links among the neurons generating the observed binary signals. A multiple-input/multiple-output (MIMO) modeling methodology is presented that can be used to quantify the neuronal dynamics of causal interrelationships in neuronal ensembles using spike-train data recorded from individual neurons. These causal interrelationships are modeled as transformations of spike-trains recorded from a set of neurons designated as the "inputs" into spike-trains recorded from another set of neurons designated as the "outputs." The MIMO model is composed of a set of multiinputlsingle-output (MISO) modules, one for each output. Each module is the cascade of a MISO Volterra model and a threshold operator generating the output spikes. The Laguerre expansion approach is used to estimate the Volterra kernels of each MISO module from the respective input-output data using the least-squares method. The predictive performance of the model is evaluated with the use of the receiver operating characteristic (ROC) curve, from which the optimum threshold is also selected. The Mann-Whitney statistic is used to select the significant inputs for each output by examining the statistical significance of improvements in the predictive accuracy of the model when the respective inputs is included. A modeling approach to characterize the nonlinear dynamic transformations of the dentate gyrus of the hippocampus is presented and experimentally validated. The dentate gyrus is the first region of the hippocampus which receives and integrates sensory information via the perforant path. The perforant path is composed of two distinct pathways: 1) the lateral path and 2) the medial perforant path. The proposed approach examines and captures the short-term dynamic characteristics of these two pathways using a nonparametric, third-order Poisson-Volterra model. The nonlinear characteristics of the two pathways are represented by Poisson-Volterra kernels, which are quantitative descriptors of the nonlinear dynamic transformations. The kernels were computed with experimental data from in vitro hippocampal slices. The electrophysiological activity was measured with custom-made multielectrode arrays, which allowed selective stimulation with random impulse trains and simultaneous recordings of extracellular field potential activity. The results demonstrate that this mathematically rigorous approach is suitable for the multipathway complexity of the hippocampus and yields interpretable models that have excellent predictive capabilities. The resulting models not only accurately predict previously reported electrophysiological descriptors, such as paired pulses, but more important, can be used to predict the electrophysiological activity of dentate granule cells to arbitrary stimulation patterns at the perforant path. One of the fundamental principles of cortical brain regions, including the hippocampus, is that information is represented in the ensemble firing of populations of neurons, i.e., spatio-temporal patterns of electrophysiological activity. The hippocampus has long been known to be responsible for the formation of declarative, or fact-based, memories. Damage to the hippocampus disrupts the propagation of spatio-temporal patterns of activity through hippocampal internal circuitry, resulting in a severe anterograde amnesia. Developing a neural prosthesis for the damaged hippocampus requires restoring this multiple-input, multiple-output transformation of spatio-temporal patterns of activity. Because the, mechanisms underlying synaptic transmission and generation of electrical activity in neurons are inherently nonlinear, any such prosthesis must be based on a nonlinear multiple-input, multiple-output model. In this paper, we have formulated the transformational process of multi-site propagation of spike activity between two subregions of the hippocampus (CA3 and CA1) as the identification of a multiple-input, multiple-output (MIMO) system, and proposed that it can be decomposed into a series of multiple-input, single-output (MISO) systems. Each MISO system is modeled as a physiologically plausible structure that consists of 1) linear/nonlinear feedforward Volterra kernels modeling synaptic transmission and dendritic integration, 2) a linear feedback Volterra kernel modeling spike-triggered after-potentials, 3) a threshold for spike generation, 4) a summation process for somatic integration, and 5) a noise term representing intrinsic neuronal noise and the contributions of unobserved inputs. Input and output spike trains were recorded from hippocampal CA3 and CA1 regions of rats performing a spatial delayed-nonmatch-to-sample memory task that requires normal hippocampal function. Kernels were expanded with Laguerre basis functions and estimated using a maximum-likelihood method. Complexity of the feedforward kernel was progressively increased to capture higher-order system nonlinear dynamics. Results showed higher prediction accuracies as kernel complexity increased. Self-kernels describe the nonlinearities within each input. Cross-kernels capture the nonlinear interaction between inputs. Second- and third-order nonlinear models were found to successfully predict the CA1 output spike distribution based on CA3 input spike trains. This paper presents a novel segmentation methodology for automated classification and differentiation of soft tissues using multiband data obtained with the newly developed system of high-resolution ultrasonic transmission tomography (HUTT) for imaging biological organs. This methodology extends and combines two existing approaches: the L-level set active contour (AC) segmentation approach and the agglomerative hierarchical k-means approach for unsupervised Clustering (UC). To prevent the trapping of the current iterative minimization AC algorithm in a local minimum, we introduce a multiresolution approach that applies the level set functions at successively increasing resolutions of the image data. The resulting AC clusters are subsequently rearranged by the UC algorithm that seeks the optimal set of clusters yielding the minimum within-cluster distances in the feature space. The presented results from Monte Carlo simulations and experimental animal-tissue data demonstrate that the proposed methodology outperforms other existing methods without depending on heuristic parameters and provides a reliable means for soft tissue differentiation in HUTT images. Inversion layer ultrasonic transducers have been investigated recently as an interesting approach in wideband transducer design. In this paper we present an analytical model of multilayer ultrasonic transducers with an inversion layer. Our analysis of the wave propagation problem of an inversion layer transducer includes a functional decomposition of the electrical input impedance. It becomes clear from this decomposition that an inversion layer transducer can be modeled as three elements in series connection, i.e., a clamped capacitance, a classical motional impedance, and a coupled motional impedance. The first two elements make up the classical model of a single element transducer. The coupled motional impedance describes the coupled interaction between the regular and the inverted piezoelectric sublayers, and thus reflects the effect of an inversion layer. We present examples which show that inversion layer transducers are advantageous in achieving such useful features as dual-frequency operation mode as used in harmonic imaging or broadband performance desired in most ultrasonic applications.
