Abstract

Background. Hepatocellular carcinoma (HCC) ranks as the third leading cause of cancer-related mortality in China with increasing incidence. This study is designed to explore early genetic changes implicated in HCC tumorigenesis and progression by whole-exome sequencing. Methods. We firstly sequenced the whole exomes of 5 paired hepatitis B virus-related early-stage HCC and peripheral blood samples, followed by gene ontological analysis and pathway analysis of the single-nucleotide variants discovered. Then, the mutations of high frequency were further confirmed by Sanger sequencing. Results. We identified a mutational signature of dominant T:A>A:T transversion in early HCC and significantly enriched pathways including ECM-receptor interaction, axon guidance, and focal adhesion and enriched biological processes containing cell adhesion, axon guidance, and regulation of pH. Eight genes, including MUC16, UNC79, USH2A, DNAH17, PTPN13, TENM4, PCLO, and PDE1C, were frequently mutated. Conclusions. This study reveals a mutational profile and a distinct mutation signature of T:A>A:T transversion in early-stage HCC with HBV infection, which will enrich our understanding of genetic characteristics of the early-stage HCC.

Abstract

The purpose of this study was to prepare a novel paclitaxel (PTX) microemulsion containing a reduced amount of Cremophor EL (CrEL) which had similar pharmacokinetics and antitumor efficacy as the commercially available PTX injection, but a significantly reduced allergic effect due to the CrEL. The pharmacokinetics, biodistribution, in vivo antitumor activity and safety of PTX microemulsion was evaluated. The results of pharmacokinetic and distribution properties of PTX in the microemulsion were similar to those of the PTX injection. The antitumor efficacy of the PTX microemulsion in OVCRA-3 and A 549 tumor- bearing animals was similar to that of PTX injection. The PTX microemulsion did not cause haemolysis, erythrocyte agglutination or simulative reaction. Abstract

A novel hybrid network composed of benzoxazines (BZ) and novolac epoxy resin (F-51) was prepared successfully. Thermal properties, curing kinetics, and decomposition process were studied using isothermal differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA) in this paper. The reactive mechanism of F-51/BZ mixture system is different from the BZ homopolymers at low temperatures; two resin systems follow the autocatalytic model mainly at high temperatures. Thermogravimetric analysis indicates that F-51 can have no significant effect on thermal degradation temperatures and on increasing char yield.

Abstract

A pedestrian counting method based on Haar-like detection and template-matching algorithm is presented. The aim of the method is to count pedestrians that are in a metro station automatically using video surveillance camera. The most challenging problem is to count pedestrians accurately in the case of not changing the position of the surveillance camera, because the view that surveillance camera uses in a metro station is always short-shot and nondirect downward view. In this view, traditional methods find it difficult to count pedestrians accurately. Hence, we propose this novel method. In addition, in order to improve counting accuracy more, we present a method to set the parameter value with a threshold-curve instead of a fixed threshold. The results of experiments show the high accuracy of our method.

Abstract

Curcuminoids, and mainly curcumin, are potential therapeutic agents for the prevention of various diseases; however, little is known about the factors that influence their accumulation in Curcuma species. In this study, the effects of factors such as sucrose concentration, different ratios of 6-benzylaminopurine (6-BA) and α-naphthalene acetic acid (NAA), and light quality on the accumulation of curcumin and other curcuminoids in Curcuma aromatica were investigated. Microrhizomes grown on media containing 3% sucrose produced more curcumin and other curcuminoids than those grown on higher concentrations. Moreover, when compared to other ratios of 6-BA and NAA, microrhizomes induced on 3% sucrose media supplemented with 3.0 mg/L 6-BA and 0.5 mg/L NAA produced more curcumin and other curcuminoids; however, the amount was less than in microrhizomes grown on 3% sucrose alone. We determined that a 5% sucrose medium supplemented with 3.0 mg/L of 6-BA and 0.5 mg/L of NAA enhanced the levels of curcumin and curcuminoids and that exposure to red light further increased production. Abstract

In this article, we present an efficient technique for the accurate design of wideband substrate integrate waveguide directional couplers. By tapering the coupling section, the bandwidth of substrate integrated waveguide (SIW) directional couplers can be enlarged. Two design aspects are involved in this approach. First, the even-mode propagation constant in the tapered coupling section is accurately extracted with the help of a numerical thru-reflect-line calibration technique. Then, it is fitted into the model of a uniform dielectric-filled rectangular waveguide and thereafter extrapolated to the operation range of the odd mode. Second, equivalent circuit models of the waveguide bifurcation effects are also presented together with parametric values. Based on the results of extraction, a 90° 3-dB directional coupler is developed to validate the proposed design approach. To achieve the reverse phasing at two output ports, the prototyped 90° 3-dB directional coupler is subsequently integrated with a novel broadband fixed phase shifter developed with the SIW technology, of which a systematic synthesis procedure has been proposed in this article. Measured performance of both 90° and 180° 3-dB couplers confirms the accuracy of our proposed design approach.
