The variety and widespread of coronavirus in natural reservoir animals is likely to cause epidemics via interspecific transmission, which has attracted much attention due to frequent coronavirus epidemics in recent decades. Birds are natural reservoir of various viruses, but the existence of coronaviruses in wild birds in central China has been barely studied. Some bird coronaviruses belong to the genus of Deltacoronavirus. To explore the diversity of bird deltacoronaviruses in central China, we tested faecal samples from 415 wild birds in Hunan Province, China. By RT‐PCR detection, we identified eight samples positive for deltacoronaviruses which were all from common magpies, and in four of them, we successfully amplified complete deltacoronavirus genomes distinct from currently known deltacoronavirus, indicating four novel deltacoronavirus stains (HNU1‐1, HNU1‐2, HNU2 and HNU3). Comparative analysis on the four genomic sequences showed that these novel magpie deltacoronaviruses shared three different S genes among which the S genes of HNU1‐1 and HNU1‐2 showed 93.8% amino acid (aa) identity to that of thrush coronavirus HKU12, HNU2 S showed 71.9% aa identity to that of White‐eye coronavirus HKU16, and HNU3 S showed 72.4% aa identity to that of sparrow coronavirus HKU17. Recombination analysis showed that frequent recombination events of the S genes occurred among these deltacoronavirus strains. Two novel putative cleavage sites separating the non‐structural proteins in the HNU coronaviruses were found. Bayesian phylogeographic analysis showed that the south coast of China might be a potential origin of bird deltacoronaviruses existing in inland China. In summary, these results suggest that common magpie in China carries diverse deltacoronaviruses with novel genomic features, indicating an important source of environmental coronaviruses closed to human communities, which may provide key information for prevention and control of future coronavirus epidemics. Our previous study of coxsackievirus B3 (CVB3)‐induced unfolded protein responses (UPR) found that overexpression of ATF6a enhances CVB3 VP1 capsid protein production and increases viral particle formation. These findings implicate that ATF6a signalling benefits CVB3 replication. However, the mechanism by which ATF6a signalling is transduced to promote virus replication is unclear. In this study, using a Tet‐On inducible ATF6a HeLa cell line, we found that ATF6a signalling downregulated the protein expression of the endoplasmic reticulum (ER) degradation‐enhancing α‐mannosidase‐like protein 1 (EDEM1), resulting in accumulation of CVB3 VP1 protein; in contrast, expression of a dominant negative ATF6a had the opposite effect. Furthermore, we found that EDEM1 was cleaved by both CVB3 protease 3C and virus‐activated caspase and subsequently degraded via the ubiquitin‐proteasome pathway. However, overexpression of EDEM1 caused VP1 degradation, likely via a glycosylation‐independent and ubiquitin‐lysosome pathway. Finally, we demonstrated that CRISPR/Cas9‐mediated knockout of EDEM1 increased VP1 accumulation and thus CVB3 replication. This is the first study to report the ER protein quality control of non‐enveloped RNA virus and reveals a novel mechanism by which CVB3 evades host ER quality control pathways through cleavage and degradation of the UPR target gene EDEM1, to ultimately benefit its own replication. Coronavirus (CoV) pandemics have become a huge threat to the public health worldwide in the recent decades. Typically, severe acute respiratory syndrome CoV (SARS‐CoV) caused SARS pandemic in 2003 and SARS‐CoV‐2 caused the ongoing COVID‐19 pandemic. Both viruses are most likely originated from bats. Thus, direct or indirect inter‐species transmission from bats to humans is required for the viruses to cause pandemics. Receptor utilization is a key factor determining the host range of viruses which is critical to the inter‐species transmission. Angiotensin‐converting enzyme 2 (ACE2) is the receptor of both SARS‐CoV and SARS‐CoV‐2, but only ACE2s of certain animals can be utilized by the viruses. Here, we employed pseudovirus cell‐entry assay to evaluate the receptor‐utilizing capability of ACE2s of 20 animals by the two viruses and found that SARS‐CoV‐2 utilized less ACE2s than SARS‐CoV, indicating a narrower host range of SARS‐CoV‐2. Especially, SARS‐CoV‐2 tended not to use murine or non‐mammal ACE2s. Meanwhile, pangolin‐CoV, another SARS‐related coronavirus highly homologous to SARS‐CoV‐2 in its genome, yet showed similar ACE2 utilization profile with SARS‐CoV rather than SARS‐CoV‐2. Nevertheless, the actual susceptibility of these animals to the coronaviruses should be further verified by in vivo studies. To clarify the mechanism underlying the receptor utilization, we compared the amino acid sequences of the 20 ACE2s and found 5 amino acid residues potentially critical for ACE2 utilization, including the N‐terminal 20th and 42nd amino acid residues that might determine the different receptor utilization of SARS‐CoV, SARS‐CoV‐2 and pangolin‐CoV. Our studies enhance the understanding of receptor utilization of pandemic coronaviruses, potentially contributing to the virus tracing, intermediate host screening and epidemic prevention for pathogenic coronaviruses. In situ Raman spectra of aqueous K2WO4-HCl, K2WO4-HCl-NaCl, and K2WO4-CO2-NaCl solutions were collected at elevated temperature (T , 100–400°C) and constant pressure (P) of 30 MPa. The stretching vibration band of the W=O bond ( v 1 ) was analyzed to reveal the species of tungsten (W) responsible for the hydrothermal transport of W during mineralization. Results showed that monomeric tungstates with v 1 (W=O) bands at ~930 and 950 cm-1 are the dominant W species in weakly alkaline (room temperature, pH = 7.7) to near-neutral (room temperature, pH = 7.2) solutions under the investigated T ‐ P conditions. Overall, the stability of polymeric tungstate species with v 1 (W=O) bands at ~965–995 cm-1 decreases with rising temperature and was not detected at ≥300°C in moderately acidic solution (room temperature, pH = 4.9). However, increased fluid acidity and salinity obviously enlarged the temperature stability field of polymeric tungstate species. In highly acidic solution (room temperature, pH = 1.4), polymeric tungstates are the only stable W species even at 400°C. In the presence of 1.9 mol/kg NaCl, polymeric tungstate(s) can persist to at least 350°C in moderately acidic solution. Considering that 300–350°C is the major W-mineralizing T range and W-mineralizing fluid is generally characterized by a moderately acidic nature, we propose that, in addition to monomeric tungstates, polymeric tungstates can also be important W species in some natural geological fluids that are responsible for the mineralization of W. Previously we found that prolonged endoplasmic reticulum ( ER) stress caused by coxsackievirus B3 ( CVB3) infection led to p58IPK downregulation and subsequent cell apoptosis. This finding implies that p58IPK expression benefits cell survival and counteracts CVB3-induced apoptosis. In testing this hypothesis, we first found that PI3 K/ Akt survival pathway is more sensitive than ERK1/2 in response to p58IPK expression. This finding was further verified by silencing p58IPK with specific siRNAs, which led to the significant suppression of phosphorylation of Akt (p- Akt) but not ERK1/2. Further, using CVB3-infected cell line expressing dominant negative ATF6a ( DN- ATF6a), we found that expression of p58IPK and p- Akt was significantly reduced, which led to the decreased cell viability. However, when the DN- ATF6a cells were transiently transfected with p58IPK, an opposite result was obtained. Finally, by CVB3 infection of cells stably expressing p58IPK, we found that CVB3-induced mitochondria-mediated apoptosis was suppressed, which was evidenced by the reduced cytochrome c release and upregulation of the mitochondrial membrane protein mitofusin 2. However, silencing p58IPK with either specific siRNAs or DN- ATF6a sensitized cells to CVB3-induced apoptosis. These results suggest that p58IPK suppresses CVB3-induced apoptosis through selective activation of PI3 K/ Akt pathway that requires activation of ATF6a and subsequently upregulates mitofusin 2. Anatase TiO2 nanosheet porous films were prepared by calcination of the orthorhombic titanic acid films at 400°C. They showed an excellent photocatalytic activity for CO2 photoreduction to methane, which should be related to their special porous structure and large Brunauer-Emmett-Teller (BET) surface area. In order to further improve the photocatalytic activity, Pt nanoparticles were loaded uniformly with the average size of 3-4nm on TiO2 porous films by the photoreduction method. It was found that the loading of Pt expanded the light absorption ability of the porous film and improved the transformation efficiency of CO2 to methane. The conversion yield of CO2 to methane on Pt/TiO2 film reached 20.51 ppm/h·cm2.The Pt/TiO2 nanosheet porous film was characterized by means of X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscope (TEM), and ultraviolet-visible light diffuse reflectance spectra (UV-vis DRS). Moreover, the transient photocurrent-time curves showed that the Pt/TiO2 nanosheet porous film exhibited higher photocurrent, indicating that the higher separation efficiency of the photogenerated charge carriers was achieved. Bank customer validation is carried out with the aim of providing a series of services to users of a bank and financial institutions. It is necessary to perform various analytical methods for user's accounts due to the high volume of banking data. This research works in the field of money laundering detection from real bank data. Banking data analysis is a complex process that involves information gathered from various sources, mainly in terms of personality, such as bills or bank account transactions which have qualitative characteristics such as the testimony of eyewitnesses. Operational or research activities can be greatly improved if supported by proprietary techniques and tools, due to the vast nature of this information. The application of data mining operations with the aim of discovering new knowledge of banking data with an intelligent approach is considered in this research. The approach of this research is to use the spiking neural network (SNN) with a group of sparks to detect money laundering, but due to the weakness in accurately identifying the characteristics of money laundering, the maximum entropy principle (MEP) method is also used. This approach will have a mapping from clustering and feature extraction to classification for accurate detection. Based on the analysis and simulation, it is observed that the proposed approach SNN-MFP has 87% accuracy and is 84.71% more functional than the classical method of using only the SNN. In this analysis, it is observed that in real banking data from Mellat Bank, Iran, in its third and fourth data, with a comprehensive analysis and reaching different outputs, there have been two money laundering cases. The intestinal microbiota play a vital role in the growth and health of the host. Probiotics could influence the homeostasis of intestinal microbiota, but the relationship between species and ecosystem stability is poorly understood. Here, four diets with different doses of Bacillus cereus (control, 0 cfu/kg; BL7, 107 cfu/kg; BL9, 109 cfu/kg; and BL11, 1011 cfu/kg) were designed to investigate the intestinal microbiota in Paramisgurnus dabryanus. The growth experiment demonstrated that the growth performance and the indices of Chao1, phylogenetic diversity, Simpson and Shannon improved significantly after loaches were fed a BL11 diet. The dominant composition of intestinal microbiota at phylum, class and genus levels did not change in loach after feeding different diets. Categories of bacterial taxa revealed that rare taxa and conditionally rare taxa were the dominant bacterial taxa in intestinal microbiota of loach. B. cereus supplemented with 1011 cfu/kg promoted the stability of the ecological network of intestinal microbiota by reinforcing the cooperation of interspecific interactions, promoting operational taxonomic units to shape larger submodules and increasing module hubs species and connectors species. Results of functional prediction revealed that a total of 5692 functions were shared among the four groups, which accounted for 91.7% of overall functions. To sum up, dietary supplementation with B. cereus had no significant influences on the composition and bacterial taxa of intestinal microbiota and the composition of bacterial functions. A dose of B. cereus appropriate to the diet could improve the growth performance and promote the homeostasis of intestinal microbiota in loach. [ABSTRACT FROM AUTHOR]
