2. Vehicle–Bridge Interaction Dynamic Analysis of Continuous Rigid Frame Composite Box Girder Bridge with Corrugated Steel Webs under Seismic Excitation.

Abstract

To study the vehicle–bridge interaction (VBI) of highway bridges under seismic excitation, a vehicle–bridge couple analysis method based on Ansys is proposed. The 1/2 vehicle model and space beam element model were established to analyze the VBI response of Lanzhou Xiaoshagou bridge. The self-excited excitation of the system is represented by road surface irregularity randomness, while the external excitation is represented by an earthquake. The impact of seismic types, seismic direction, seismic intensity, vehicle speed, and road surface irregularity on the bridge vibration under the vehicle–bridge coupling during an earthquake is thoroughly analyzed. The results reveal that the type of earthquake significantly influences the dynamic response of the bridge, showing a minimum difference of 31.4%. The intensity of the earthquake is positively correlated with the dynamic response of the bridge. Longitudinal and vertical earthquakes have a more noticeable effect on the bridge's vertical vibration compared to lateral earthquakes. The ratio of the bridge response under vertical or longitudinal seismic excitation to the response of lateral earthquakes ranges from 1.50 to 26.61. Vehicle speed, road irregularity grade, and randomness have a negligible impact on the dynamic response of vehicle–bridge interaction under an earthquake, accounting for less than 3%. These findings indicate that the analysis of earthquake-bridge vibration can simplify the VBI analysis for continuous rigid frame composite box girder bridges with corrugated steel webs under seismic conditions. 3. The Pyroptosis-Related Risk Genes APOBEC3D, TNFRSF14, and RAC2 Were Used to Evaluate Prognosis and as Tumor Suppressor Genes in Breast Cancer

Abstract

Background. Pyroptosis is a type of cell death that plays an important role in predicting prognosis and immunoregulation in cancers. However, the pyroptosis-related gene signature for prognosis and immune infiltration prediction has not been studied in breast cancer (BC). Methods. The Gene Expression Omnibus (GEO) and Cancer Genome Atlas (TCGA) databases were used to obtain the expression and clinical data of genes. 52 pyroptosis-related genes were obtained from TCGA-BC and estimated differentially expressed genes by the limma program. To categorize the molecular subtypes of pyroptosis-related genes, the ConsensusClusterPlus tool was utilized. Cox and Lasso regression analyses were used to create a signature. TCGA-BC dataset as the training set and the GSE37751 test set for risk research. Gene set enrichment analysis (GSEA) was used to conduct KEGG and GO studies of subtype groups. We also used the ssGSEA approach in the GSVA package to calculate the risk score of immune cells. Finally, pyroptosis-related genes in BC were validated using qPCR and immunohistochemical assays. Clone formation and EDU assays were used to explore the ability of signature genes to regulate the proliferation of BC cells. Results. Based on pyroptosis-related genes, the C1 and C2 subtypes were obtained. Survival analysis results showed that the C2 group had a better prognosis. Then, a three-gene signature (APOBEC3D, TNFRSF14, and RAC2) were created by Lasso regression analysis, which had a good prediction effect in the TCGA-BC and GSE37751 datasets. Our nomogram has a fair degree of accuracy in predicting the survival rates of BC patients. The pyroptosis-related signature has a good predictive effect in evaluating the tumour microenvironment score, 28 types of immune cells and response to immune checkpoint therapy. Finally, qPCR and immunohistochemistry staining results indicated that APOBEC3D, TNFRSF14, and RAC2 expression in BC tissues was low. The results of clone formation and EdU assays showed that high expression of signature genes inhibited the proliferation ability of BC cells. Conclusions. Based on pyroptosis-related genes (APOBEC3D, TNFRSF14, and RAC2), we built a novel prognostic molecular model for BC that might be used to assess prognostic risk and immune infiltration in BC patients. These signature genes are also tumor suppressor genes and may serve as potential targets for BC.

4. Simultaneous Separation and Determination of Nine Active Ingredients in Sanyetangzhiqing by Cyclodextrin-Modified Micellar Electrokinetic Capillary Electrophoresis-Diode Array Detector.

Abstract

A simple and sensitive strategy using cyclodextrin-modified micellar electrokinetic chromatography with diode array detector was developed and applied for the simultaneous separation and determination of nine components in Sanyetangzhiqing (SYTZQ), a hypoglycemic and hypolipidemic agent. Several important parameters affecting separation performance were evaluated and optimized using single variable methods. Under the optimal conditions, baseline separation of the nine components, including four flavonoids (hyperoside, isoquercitrin, quercetin-3-O-glucuronoside, and astragalin), four phenolic acids (chlorogenic acid, rosmarinic acid, salvianolic acid B, and lithospermic acid), and a monoterpenoids (paeoniflorin), were achieved in less than 16 min. The correlation coefficients of the calibration curves were over 0.9996 for all the analytes. Intraday and interday precisions ranged from 0.4% to 4.8% and 1.7% to 5.0%, respectively. Recoveries of analytes varied from 95.3% to 105%. Validation results as well as the application to analyse SYTZQ samples demonstrated the applicability of the proposed method and thus provided an effective tool for the quality control of SYTZQ. Moreover, with the advantages of short time consuming, low energy consumption, high efficiency, and low cost, this method has laid a foundation for the determination and quality evaluation of multicomponents in Chinese herbal compounds. Abstract

This paper deals with fault estimation problem for a class of nonlinear system with parameter uncertainties subjecting to Bernoulli-distributed white sequences with known conditional probabilities. In order to reflect the reality more closely, parameter uncertainties are considered in both the state parameter matrix and the output parameter matrix. Compared with existing observer-based fault estimation approaches, the proposed iterative learning observer considers the state error information and fault estimating information from the previous iteration to improve the fault estimation performance in the current iteration. Simultaneously, the stability and convergence of the designed observer are achieved by employing the Lyapunov stability theory. On the other hand, a novel optimal function using expectation is presented to ensure the uniform convergence of the fault estimation scheme, thus reducing the impact of randomly occurring parameter uncertainties. Finally, linear matrix inequality (LMI) is employed to obtain the solutions of sufficient condition for further improvement of iterative learning law performance. The results are suitable for the systems with time-varying uncertainties as well as constant uncertainties. Additionally, a numerical example is given to demonstrate the effectiveness of the proposed design scheme.
