Abstract

International audience; We developed a fully automated magnetic field scanner dedicated to uniaxial magnetic field measurements to determine remanent magnetization intensities and their variations in sedimentary U-channels. A fluxgate magnetometer located as close as possible to the sedimentary section is used to perform uniaxial measurements of magnetic fields generated by the isothermal remanent magnetization of the sediment. This artificial magnetization, which is known to be a powerful proxy in environmental magnetism, is produced perpendicular to the U-channel long axis, and parallel to the fluxgate axis, using a Halbach cylinder prior to the measurement. The present magnetic scanner offers a maximal spatial resolution of 5.8 mm for point sources. A spatial resolution of 14 mm is obtained for U-channel samples. The magnetic scanner presents a reliable magnetic field range over about 3 orders of magnitude allowing measurement of magnetizations that saturate the Superconducting Rock Magnetometer in its classical configuration. The estimation of remanent magnetization intensities along the U-channel is based on a modeling approach that uses successive uniformly magnetized prisms. In lacustrine laminated sections, comparison between modeling results based on prisms of a constant thickness, on prisms determined from sedimentary facies, and on prisms determined from X-ray fluorescence data helps to understand the detrital versus diagenetic history of the sedimentary succession.

Abstract

This study aimed at determining the seroprevalence of foot‐and‐mouth disease (FMD) in domestic ruminants and at characterizing the virus strains circulating in four areas of Chad (East Batha, West Batha, Wadi Fira and West Ennedi). The study was carried out between October and November 2016. A total of 1,520 sera samples (928 cattle, 216 goats, 254 sheep and 122 dromedaries) were collected randomly for FMD serological analyses. Nine epithelial tissue samples were also collected from cattle showing clinical signs, for FMDV isolation and characterization. Serological results showed an overall NSP seroprevalence of 40% (375/928) in cattle in our sample (95% CrI [19–63]). However, seroprevalences of 84% (27/32), 78% (35/45) and 84% (21/25) were estimated in cattle over 5 years of age in East Batha, West Batha and Wadi Fira, respectively. In cattle under 1 year of age, 67% (18/27) seroprevalence was estimated in Wadi Fira, 64% (14/22) in East Batha and 59% (13/22) in West Batha. It was found that the high seroprevalences have been obtained in areas where pastures are shared by several different herds but also in farms where two to three species (bovine, caprine and ovine) are raised together. ELISA PrioCHECK®FMDV types O and A and in‐house solid phase competition ELISA serotyping results showed that the four O, A, SAT1 and SAT2 serotypes have circulated in Chad in 2016. However, the type SAT2 dominated with an overall seroprevalence of 43% (29/67) and was present in the four areas investigated. The phylogenetic analyses of the VP1 coding sequence allowed determining the serotype SAT2 topotype VII, close to viral strains found in Cameroon in 2015 with a similarity of 98.60%., FMD virus serotyping results showed that four O, A, SAT1 and SAT2 serotypes circulated in Chad in 2015. Abstract

International audience; Foot-and-mouth disease virus (FMDV) causes a highly contagious vesicular disease in livestock, with serious consequences for international trade. The virus persists in the nasopharynx of cattle and this slows down the process to obtain an FMDV-free status after an outbreak. To study biological mechanisms, or to identify molecules that can be targeted to diagnose or interfere with persistence, we developed a model of persistent FMDV infection in bovine dorsal soft palate (DSP). Primary DSP cells were isolated after commercial slaughter and were cultured in multilayers at the air-liquid interface. After 5 weeks of culture without further passage, the cells were infected with FMDV strain O/FRA/1/2001. Approximately, 20% of cells still had a polygonal morphology and displayed tight junctions as in stratified squamous epithelia. Subsets of cells expressed cytokeratin and most or all cells expressed vimentin. In contrast to monolayers in medium, multilayers in air demonstrated only a limited cytopathic effect. Integrin alpha(V)beta(6) expression was observed in mono- but not in multilayers. FMDV antigen, FMDV RNA and live virus were detected from day 1 to 28, with peaks at day 1 and 2. The proportion of infected cells was highest at 24 hr (3% and 36% of cells at an MOI of 0.01 and 1, respectively). At day 28 after infection, at a time when animals that still harbour FMDV are considered carriers, FMDV antigen was detected in 0.2%-2.1% of cells, in all layers, and live virus was isolated from supernatants of 6/8 cultures. On the consensus level, the viral genome did not change within the first 24 hr after infection. Only a few minor single nucleotide variants were detected, giving no indication of the presence of a viral quasispecies. The air-liquid interface model of DSP brings new possibilities to investigate FMDV persistence in a controlled manner.

Abstract

International audience; Une combinaison multi-méthodes de protocoles de prospection géophysique pour la description des milieux littoraux a été testée et appliquée sur deux sites d’étude : le site des ports antiques de Narbonne (Aude) localisé dans l’ancien delta de l’Aude et le site néolithique du Pontet (Charente-Maritime) situé près de l’estuaire de la Charente. Des cartes de conductivité et des sections de résistivité électrique sont effectuées pour décrire la morphologie des structures sédimentaires associées à des environnements littoraux sur de grande surface (> 100 ha). Des diagraphies électriques sont ensuite réalisées pour identifier des variations sédimentaires. Cette approche paléogéographique permet ainsi de caractériser de potentielles structures sédimentaires telles des paléo-chenaux, des formations deltaïques ou des dépôts d’érosion de paléo-falaise. Le couplage de ces méthodes géophysiques est utilisé pour cibler l’implantation de carottages paléoenvironnementaux afin de comprendre l’évolution géomorphologique des zones étudiées.La confrontation entre les carottes et la géophysique permet d’étalonner cette dernière. Ainsi, les cartes de conductivité, les sections électriques et les diagraphies sont plus aisément interprétable en termes de corps sédimentaires, autorisant une lecture continue de la géomorphologie littorale. Cet étalonnage permet aussi de mieux comprendre l’influence de la salinité sur les mesures de résistivité/conductivité qui rendent l’interprétation des données complexes dans ces types de milieux.

Abstract

International audience; At the end of August 2015, a ram located in central France (department of Allier) showed clinical signs suggestive of BTV (Bluetongue virus) infection. However, none of the other animals located in the herd showed any signs of the Bluetongue disease. Laboratory analyses identified the virus as BTV serotype 8. The viro and sero prevalence intraherd were 2.4% and 8.6% in sheep and 18.3% and 42.9% in cattle, respectively. Phylogenetic studies showed that the sequences of this strain are closely related to another BTV-8 strain that has circulated in France in 2006-2008. The origin of the outbreak is unclear but it may be assumed that the BTV-8 has probably circulated at very low prevalence (possibly in livestock or wildlife) since its first emergence in 2007-2008.

Abstract

International audience; This paper focus on an original magnetic survey with a high spatial resolution used to map the Neolithic site of Le Pontet at Saint-Nazaire-sur-Charente (Charente-Maritime, France). The protocol involves to use a motorized total station to locate each magnetic data measured by a G-858 magnetometer. The data processing enables to enhance the magnetic map and to obtain a final error of location of a few centimetre. The accurate localization of measurements by the total station permits to understand the magnetic intensity variations between two consecutive profiles in slope (variation of the height of the sensors during the uphill and the downhill). The using of the topographic data of the total station will enable to correct the variation of magnetic intensity induced by the relief. The results of the magnetic survey allow to bring out several causewayed enclosures, pits and postholes. The archaeology excavations will be positioned on the basis of magnetic survey.

Abstract

International audience; This paper is based on a multidisciplinary study using both paleoenvironmental methods (biological, sedimentological, magnetic and geochemical) and archaeological data in order to characterize landscape and hydrological network changes as well as human impact on a littoral catchment in the Western Mediterranean area. Sedimentary records obtained around a coastal alluvial plain (Cassis, Southeastern France) reveal local environmental changes and human activities since the Neolithic (ca. 6000 cal. BP). Anthropogenic impact is already noticeable in the earliest record, and we follow its evolution through time. According to biological markers, the Holocene landscape of the Calanques is dominated by a mosaic of open herbaceous formations partly generated by human activity. The prevalence of Cernuella virgata in malacofauna successions and of coprophilous fungal spores in the pollen record testifies to the spread of agro-pastoral activities. Two increases of anthropogenic impact are highlighted during the Roman (ca. 2000 cal. BP) and Modern (from ca. 1450 to today) periods. Lead isotope analyses indicate an anthropogenic release of lead, possibly originating from the Greek Cyclades (ca. 2000 cal. BP). Hydrographic and agricultural work is also enhanced during the Roman period. Magnetic parameters and lead isotopes indicate that the Modern anthropogenic impact may be associated with high temperature activities (e.g. lime kiln, fossil fuel).

Abstract

International audience; Le site néolithique du Pontet, sur la commune de Saint-Nazaire-sur-Charente (Charente-Maritime, France), est situé à proximité de l’embouchure du fleuve Charente. Il a été identifié par des photographies aériennes mettant en évidence la présence de quatre fossés discontinus subparallèles. Une prospection magnétique réalisée en août 2014 sur une surface d’environ 1 ha a permis de confirmer la présence de ces fossés. Leur morphologie et le matériel trouvé en surface semblent indiquer qu’ils correspondent au Néolithique récent.Le site est implanté sur une falaise calcaire qui le sépare d’une zone humide contenue dans une petite vallée débouchant sur La Charente. Une étude géophysique multi-méthodes a été menée afin d’établir le contexte paléogéographique du site et de localiser la zone la plus propice pour implanter un carottage. Une cartographie électromagnétique (EM31) a tout d’abord été réalisée pour d’identifier la position d’anciens chenaux et des structures géologiques de surface. La morphologie interne et les variations lithologiques associées au comblement holocène du vallon ont ensuite été déterminées par une série de tomographies de résistivité électrique. Afin d’appuyer et de compléter ces observations, plusieurs essais au pénétromètre dynamique ainsi qu’un carottage ont été réalisés. Divers proxies paléoenvironnementaux ont été déterminés sur les sédiments prélevés. Ces données ont également été mises en relation avec des résultats de diagraphie magnétique

Abstract

International audience; An original karstic system in a coastal alluvial plain located in a carbonate environment (Cassis, Provence -- SE France) was studied using a multi-method approach (electrical resistivity tomography, gravimetric, passive seismic survey), combined with geomorphologic methods, surface observations and drilling. Limestone dissolution and/or cavity collapse led to the development of a polje, which was filled rapidly by erosion of Aptian marls. The combination of several dating and paleoenvironmental proxies indicates that the polje was filled during a glacial period (MIS 6, 8 or 10, i.e. in the 130-360 ka age range). This discovery has implications for the understanding of karst processes. The connection between the polje and the three km-long Bestouan underwater karstic conduit with submarine outlet is strongly suggested by sedimentological studies and geophysical prospections.

Abstract

This Phd research work focuses on : (1) paleo-environment of the Calanques, (2) karst structure formation (polje of Cassis, SE France) and (3) Holocene environmental changes linked to human activities.The study is based on a multi-geophysical methods approach (electrical resistivity tomography, gravimetric, passive sismic survey) and on several sedimentary records studies. The main sedimentary record is 50 m-long core coming from a pleistocene paleolake located 2 km from the Mediterranean Sea. Limestone dissolution and/or cavity collapse led to the development of a polje, rapidly filled by the erosion of Aptian marls in glacial period. The connection between the polje and the three kilometer long Bestouan underwater karstic conduit with submarine outlet is strongly suggested by sedimentological studies and geophysical prospections.The Holocene environment is dominated by dry herbaceous formations. Human activities impact on the environment is visible through (1) local water network changes, (2) lead metallurgy activity and (3) agro-pastoral practices., Ce travail de recherche porte sur l’étude du paléo-environnement du massif des Calanques, sur la mise en place d’une structure karstique de surface (poljé de Cassis, SE France) et sur l’empreinte de l’Homme sur son milieu. L’étude se base sur la réalisation de mesures géophysiques (électrique, gravimétrie, sismique passive) et sur l’analyse de plusieurs sondages, dont le principal est un enregistrement sédimentaire de 50 m de long carotté dans un paléo-lac situé à 2 km du littoral méditerranéen qui retrace le paléo-environnement du massif des Calanques au Pléistocène supérieur et à l’Holocène.Le paléo-lac de Cassis a été formé par la dissolution du calcaire barrémien et/ou par l’effondrement d’une cavité karstique, et a été rempli par une sédimentation laminée fine issue de l’érosion des marnes aptiennes, sédimentation qui caractérise un milieu de dépôt anoxique profond. Les observations spéléologiques et les analyses sédimentaires supposent l’existence d’une connexion entre le poljé et la rivière souterraine du Bestoaun.L’environnement holocène, stable sur toute la période holocène, est dominé par les formations herbacées sèches. L’impact de l’Homme sur l’environnement du massif des Calanques est visible au travers des modifications du réseau hydrographique local, de la métallurgie du plomb et de la mise en place de pratiques agro-pastorales.

Abstract

The encephalomyocarditis virus (EMCV), a Picornaviridae virus, has a wide host spectrum and can cause various diseases. EMCV virulence factors, however, are as yet ill defined. Here, we demonstrate that the EMCV 2A protein is essential for the pathogenesis of EMCV. Infection of mice with the B279/95 strain of EMCV resulted in acute fatal disease, while the clone C9, derived by serial in vitro passage of the B279/95 strain, was avirulent. C9 harbored a large deletion in the gene encoding the 2A protein. This deletion was incorporated into the cDNA of a pathogenic EMCV1.26 strain. The new virus, EMCV1.26Δ2A, was capable of replicating in vitro , albeit more slowly than EMCV1.26. Only mice inoculated with EMCV1.26 triggered death within a few days. Mice infected with EMCV1.26Δ2A did not exhibit clinical signs, and histopathological analyses showed no damage in the central nervous system, unlike EMCV1.26-infected mice. In vitro , EMCV1.26Δ2A presented a defect in viral particle release correlating with prolonged cell viability. Unlike EMCV1.26, which induced cytopathic cell death, EMCV1.26Δ2A induced apoptosis via caspase 3 activation. This strongly suggests that the 2A protein is required for inhibition of apoptosis during EMCV infection. All together, our data indicate that the EMCV 2A protein is important for the virus in counteracting host defenses, since Δ2A viruses were no longer pathogenic and were unable to inhibit apoptosis in vitro .

Abstract

International audience; A few studies have clearly indicated that oxidative stress suppresses the cystic fibrosis transmembrane conductance receptor (CFTR) function and expression. However, the mechanisms by which this occurs are still poorly understood. To clarify this effect, we investigated the role of NF-E2-related factor 2 (Nrf2) transcription factor, a key cellular sensor of oxidative stress. A conserved antioxidant response element (ARE) in the CFTR minimal promoter, which binds Nrf2, has been identified. Surprisingly, Nrf2 exerts an unexpected repressive role on the CFTR gene promoter activity. To decipher the molecular mechanisms involved, we evaluated the role of YY1 in the Nrf2-mediated transcriptional activity and showed cooperation between these two factors. We demonstrated that Nrf2 promotes YY1 nuclear localization and increases its binding to the CFTR promoter. To our knowledge, this study is the first to report a repressor role of Nrf2 through the cooperation with YY1 and contributes to clarify the cascade events leading to the oxidative stress-suppressed CFTR expression.

Abstract

International audience; Growing evidences show that functionally relevant polymorphisms in various promoters alter both transcriptional activity and affinities of existing protein-DNA interactions, and thus influence disease progression in humans. We previously reported the -94G>T CFTR promoter variant in a female CF patient in whom any known disease-causing mutation has been detected. To investigate whether the -94G>T could be a regulatory variant, we have proceeded to in silico analyses and functional studies including EMSA and reporter gene assays. Our data indicate that the promoter variant decreases basal CFTR transcriptional activity in different epithelial cells and alters binding affinities of both Sp1 and USF nuclear proteins to the CFTR promoter. The present report provides evidence for the first functional polymorphism that negatively affects the CFTR transcriptional activity and demonstrates a cooperative role of Sp1 and USF transcription factors in transactivation of the CFTR gene promoter.

Abstract

This paper is the fifth chapter of a book forthcoming in January 2007 under the title Histoire du marché financier français au 19 ème siècle (a history of the French capital market in the 19th century). He discusses the reasons for the development of securities in the early 19th century in France. He presents first the economic and legal conditions for the rise of a market for private issues (mostly utilities) and then the development of a market for foreign (mostly government) securities in Paris., Ce papier constitue le cinquième chapitre de l'Histoire du marché financier français au 19 ème siècle (à paraître en janvier 2007, publications de la Sorbonne). Dans ce chapitre, nous examinons les raisons du développement d'un marché des titres (actions et obligations) à Paris au début du 19 ème siècle : en premier lieu les conditions économiques et légales de l'apparition d'émissions et de cotations de titres privés ; ensuite l'émergence d'un marché de titres étrangers, principalement publics.

Dans ce chapitre, nous examinons et cherchons à expliquer les transformations majeures de l'offre de titres sur le marché financier français de la Belle Epoque : déclin des émissions publiques, montée d'émissions privées non seulement bancaires et d'infrastructures mais aussi industrielles, énorme développement des émissions étrangères privées et publiques.

Abstract

The TREK-1 channel is a temperature-sensitive, osmosensitive and mechano-gated K+ channel with a regulation by Gs and Gq coupled receptors. This paper demonstrates that TREK-1 qualifies as one of the molecular sensors involved in pain perception. TREK-1 is highly expressed in small sensory neurons, is present in both peptidergic and nonpeptidergic neurons and is extensively colocalized with TRPV1, the capsaicin-activated nonselective ion channel. Mice with a disrupted TREK-1 gene are more sensitive to painful heat sensations near the threshold between anoxious warmth and painful heat. This phenotype is associated with the primary sensory neuron, as polymodal C-fibers were found to be more sensitive to heat in single fiber experiments. Knockout animals are more sensitive to low threshold mechanical stimuli and display an increased thermal and mechanical hyperalgesia in conditions of inflammation. They display a largely decreased pain response induced by osmotic changes particularly in prostaglandin E2-sensitized animals. TREK-1 appears as an important ion channel for polymodal pain perception and as an attractive target for the development of new analgesics.

Abstract

Background Chemical and radiological toxicities related to uranium acute exposure have been widely studied in nuclear fuel workers and military personnel. It is well known that uranyl nitrate induces acute renal failure (ARF). However, the mechanisms of this metal-induced injury are not well defined at the molecular level. Results Renal function and histology were assessed in mice receiving uranyl nitrate (UN(+)) and controls (UN(-)). To identify the genomic response to uranium exposure, serial analysis gene expression (SAGE) of the kidney was performed in both groups. Over 43,000 mRNA SAGE tags were sequenced. A selection of the differentially expressed transcripts was confirmed by real-time quantitative PCR and Western blotting. UN(+) animals developed renal failure and displayed the characteristic histological lesions of UN nephropathy. Of the >14,500 unique tags identified in both libraries, 224 had a modified expression level; they are known to participate in inflammation, ion transport, signal transduction, oxidative stress, apoptosis, metabolism, and catabolism. Several genes that were identified had not previously been evaluated within the context of toxic ARF such as translationally controlled tumor protein, insulin like growth factor binding protein 7 and ribosomal protein S29, all apoptosis related genes. Conclusion We report a comprehensive description of the UN induced modifications in gene expression levels, including the identification of genes previously unrelated to ARF. The study of these genes and the metabolisms they control should improve our understanding of toxic ARF and enlighten on the molecular targets for potential therapeutic interventions.

Abstract

The spread of diseases transmitted by Anopheles and Culex mosquitoes, such as malaria and West Nile fever, is a growing concern for human health. Bacillus sphaericus binary toxin (Bin) is one of the few available bioinsecticides able to control populations of these mosquitoes efficiently. We previously showed that Bin binds to Cpm1, an alpha-glucosidase located on the apical side of Culex larval midgut epithelium. We analysed the effects of Bin by expressing a construct encoding Cpm1 in the mammalian epithelial MDCK cell line. Cpm1 is targeted to the apical side of polarized MDCK, where it is anchored by glycosylphosphatidylinositol (GPI) and displays alpha-glucosidase activity. Bin bound to transfected cells and induced a non-specific current presumably related to the opening of pores. The formation of these pores may be related to the location of the toxin/receptor complex in lipid raft microdomains. Finally, Bin promoted the time-dependent appearance of intracytoplasmic vacuoles but did not drive cell lysis. Thus, the dual functionality (enzyme/toxin receptor) of Cpm1 is fully conserved in MDCK cells and Cpm1 is an essential target protein for Bin cytotoxicity in Culex mosquitoes.

Abstract

International audience; Although the nephrotoxkity of uranium has been established through numerous animal studies, relatively little is known about the effects of long-term environmental uranium exposure. Using a combination of conventional biochemical studies and serial analysis of gene expression (SAGE), we examined the renal responses to uranyl nitrate (UN) chronic exposure. Renal uranium levels were significantly increased 4 months after ingestion of uranium in drinking water. Creatinine levels in serum were slightly but significantly increased compared with those in controls. Although no further significant differences in other parameters were noted, substantial molecular changes were observed in toxicogenomic profiles. UN induced dramatic alterations in expression levels of more than 200 genes, mainly up-regulated, including oxidative-response-related genes, genes encoding for cellular metabolism, ribosomal proteins, signal transduction, and solute transporters. Seven differentially expressed transcripts were confirmed by real-time quantitative polymerase chain reaction. In addition, significantly increased peroxide levels support the implication of oxidative stress in UN toxicant response. This report highlights the potential of SAGE for the discovery of novel toxicant-induced gene expression alterations. Here, we present, for the first time, a comprehensive view of renal molecular events after uranium long-term exposure.

Abstract

Molecular events of mechanotransduction in osteoblasts are poorly defined. We show that the mechanosen- sitive BK channels open and recruit the focal adhesion kinase FAK in osteoblasts on hypotonic shock. This could convert mechanical signals in biochemical events, leading to osteoblast activation. Introduction: Mechanical strains applied to the skeleton influence bone remodeling and architecture mainly through the osteoblast lineage. The molecular mechanisms involved in osteoblastic mechanotransduction include opening of mechanosensitive cation channels and the activation of protein tyrosine kinases, notably FAK, but their interplay remains poorly characterized. The large conductance K channel (BK) seems likely as a bone mechanoreceptor candidate because of its high expression in osteoblasts and its ability to open in response to membrane stretch or hypotonic shock. Propagation of the signals issued from the mechanosensitivity of BK channels inside the cell likely implies complex interactions with molecular partners involved in mechanotransduction, notably FAK. Methods: Interaction of FAK with the C terminus of the hSlo -subunit of BK was investigated using the yeast two-hybrid system as well as immunofluorescence microscopy and coimmunoprecipitation experiments with a rabbit anti-hslo antibody on MG63 and CAL72 human osteosarcoma cell lines and on normal human osteoblasts. Mapping of the FAK region interacting with hSlo was approached by testing the ability of hSlo to recruit mutated ot truncated FAK proteins. Results: To the best of our knowledge, we provide the first evidence of the physical association of FAK with the intracellular part of hslo. We show that FAK/hSlo interaction likely takes place through the Pro-1-rich domain situated in the C-terminal region of the kinase. FAK/hSlo association occurs constitutively at a low, but appreciable, level in human osteosarcoma cells and normal human osteoblasts that express endogenous FAK and hSlo. In addition, we found that application of an hypo-osmotic shock to these cells induced a sustained activation of BK channels associated to a marked increase in the recruitment of FAK on hSlo. Conclusions: Based on these data, we propose that BK channels might play a triggering role in the signaling cascade induced by mechanical strains in osteoblasts. J Bone Miner Res 2003;18:1863-1871

Abstract

Objectives: We report the functional expression of four KCNQ1 mutations affecting arginine residues and resulting in Romano–Ward (RW) and the Jervell and Lange–Nielsen (JLN) congenital long QT syndromes. Results: The R539W and R190Q mutations were found in typical RW families with an autosomal dominant transmission. The R243H mutation was found in a compound heterozygous JLN patient who presents with deafness and cardiac symptoms. The fourth mutation, R533W, was a new case of recessive form of the RW syndrome since homozygous carriers experienced syncopes but showed no deafness, whereas the heterozygous carriers were asymptomatic. The R190Q mutation failed to produce functional homomeric channels. The R243H, R533W and R539W mutations induced a positive voltage shift of the channel activation but only when co-expressed with IsK, pointing out the critical role of these positively charged residues in the modulation of the gating properties of KvLQT1 by IsK. The positive shift induced by R533W was merely 15%. This small effect was compatible with the recessive character of the RW phenotype transmission. The average QTc was significantly longer ( P

Abstract

International audience; Mutations in the delayed rectifier K+ channel subunit KvLQT1 have been identified as responsible for both Romano-Ward (RW) and Jervell and Lange-Nielsen (JLN) inherited long QT syndromes. We report the molecular cloning of a human KvLQT1 isoform that is expressed in several human tissues including heart. Expression studies revealed that the association of KvLQT1 with another subunit, IsK, reconstitutes a channel responsible for the IKs current involved in ventricular myocyte repolarization. Six RW and two JLN mutated KvLQT1 subunits were produced and co-expressed with IsK in COS cells. All the mutants, except R555C, fail to produce functional homomeric channels and reduce the K+ current when co-expressed with the wild-type subunit. Thus, in both syndromes, the main effect of the mutations is a dominant-negative suppression of KvLQT1 function. The JLN mutations have a smaller dominant-negative effect, in agreement with the fact that the disease is recessive. The R555C subunit forms a functional channel when expressed with IsK, but with altered gating properties. The voltage dependence of the activation is strongly shifted to more positive values, and deactivation kinetics are accelerated. This finding indicates the functional importance of a small positively charged cytoplasmic region of the KvLQT structure where two RW and one JLN mutations have been found to take place.

Abstract

International audience; The incidence of developing cancer should increase with the body mass, yet is not the case, a conundrum referred to as Peto's paradox. Elephants have a lower incidence of cancer suggesting that these animals have probably evolved different ways to protect themselves against the disease. The paradox is worth revisiting with the realization that most mammals encode an endogenous APOBEC3 cytidine deaminase capable of mutating single stranded DNA. Indeed, the mutagenic activity of some APOBEC3 enzymes has been shown to introduce somatic mutations into genomic DNA. These enzymes are now recognized as causal agent responsible for the accumulation of CG-> TA transitions and DNA breaks leading to chromosomal rearrangements in human cancer genomes. Here, we identified an elephant A3Z1 gene, related to human APOBEC3A and showed that it could efficiently deaminate cytidine, 5-methylcytidine and produce DNA breaks leading to massive apoptosis, similar to other mammalian APOBEC3A enzymes where body mass varies by up to four orders of magnitude. Consequently, it could be considered that eAZ1 might contribute to cancer in elephants in a manner similar to their proposed role in humans. If so, eAZ1 might be particularly well regulated to counter Peto's paradox. The APOBEC3 (A3) locus is bounded by two conserved genes, chromobox 6 and 7 (CBX6 and CBX7) in most pla-cental mammals and encodes a family of cytidine deaminases capable of converting cytidine residues to uridine in single strand DNA (ssDNA). The mutagenic activity of these enzymes is involved with the restriction of retro-viruses and DNA viruses, as well as endogenous retroelements and retrotransposons through hypermutation of viral DNA in a process called editing 1. The A3 repertoire is extremely variable among mammals, the locus being shaped through extensive gene duplications and functionalization in the context of a virus-host arms race. A3 enzymes are made up of three related, but distinct zinc-finger domains referred to as Z1, Z2 and Z3 2-4 presumably already present in the genome of the ancestor of placental mammals 5. The last few years has seen the identification of two human endogenous A3 cytidine deaminases, APOBEC3A (A3A) and APOBEC3B (A3B) capable of introducing multiple mutations in chromosomal DNA 6-9. These findings are grounded by the analysis of many cancer genomes, revealing far more mutations and rearrangements than hitherto imagined, where CG-> TA transitions appears to be the dominant mutations 10-13. Human A3A is composed of a single Z1 domain, while A3B is composed of a double Z2Z1 domain, although only the C terminal Z1 domain being catalytically functional 6. A3A and A3B enzymes are both localized in the nucleus and can edit cytidine residues to uridine in ssDNA during transcription and replication, following DNA repair, and leave TpC to TpT signature mutations that show up in cancer genomes 6,8,9. Both enzymes can mutate 5-methylcytidine (5MeC) to thymidine leaving another distinct signature in cancer genomes 6,14-16. Although A3A and A3B are accepted as intrinsic mutators of cellular chromosomal DNA, analyzed in several cancer types 8,11,17 , debate still persists regarding the contribution of each enzyme in the accumulation of mutations paving the way for oncogenesis. While, it has been described that A3A and A3B could be enzymat-ically active in different cancers 18 , A3A is the more active of the two enzymes and as a consequence, only A3A can produce double stranded breaks (DSBs), at least in an experimental setting 6,7,19. Editing frequencies of >0.5 can be found which is why the phenomenon is referred to as hyperediting or hypermutation 9. Accumulation of substitutions localized in the A3B C-terminal domain attenuated the activity of the enzyme compared to A3A 6 .
