Abstract

Glyphosate-based herbicides (GBHs) are massively used in agriculture. However, few studies have investigated the effects of glyphosate-based herbicides on avian species although they are largely exposed via their food. Here, we investigated the potential reversibility of the effects of chronic dietary exposure to glyphosate-based herbicides in broiler hens. For 42 days, we exposed 32-week-old hens to glyphosate-based herbicides via their food (47 mg/kg/day glyphosate equivalent, glyphosate-based herbicides, n = 75) corresponding to half glyphosate’s no-observed-adverse-effect-level in birds. We compared their performance to that of 75 control animals (CT). Both groups (glyphosate-based herbicides and control animals) were then fed for 28 additional days without glyphosate-based herbicides exposure (Ex-glyphosate-based herbicides and Ex-control animals). Glyphosate-based herbicides temporarily increased the plasma glyphosate and AMPA (aminomethylphosphonic acid) concentrations. Glyphosate and aminomethylphosphonic acid mostly accumulated in the liver and to a lesser extent in the leg muscle and abdominal adipose tissue. Glyphosate-based herbicides also temporarily increased the gizzard weight and plasma oxidative stress monitored by TBARS (thiobarbituric acid reactive substances). Glyphosate-based herbicides temporarily decreased the cecal concentrations of propionate, isobutyrate and propionate but acetate and valerate were durably reduced. The cecal microbiome was also durably affected since glyphosate-based herbicides inhibited Barnesiella and favored Alloprevotella. Body weight, fattening, food intake and feeding behavior as well as plasma lipid and uric acid were unaffected by glyphosate-based herbicides. Taken together, our results show possible disturbances of the cecal microbiota associated with plasma oxidative stress and accumulation of glyphosate in metabolic tissues in response to dietary glyphosate-based herbicides exposure in broiler hens. Luckily, glyphosate-based herbicides at this concentration does not hamper growth and most of the effects on the phenotypes are reversible.

Abstract

International audience; Metformin is a drug used for the treatment of type 2 diabetes and disorders associated with insulin resistance. Metformin is also used in the treatment of pregnancy disorders such as gestational diabetes. However, the consequences of foetal exposure to metformin on the fertility of exposed offspring remain poorly documented. In this study, we investigated the effect of in utero metformin exposure on the fertility of female and male offspring. We observed that metformin is detectable in the blood of the mother and in amniotic fluid and blood of the umbilical cord. Metformin was not measurable in any tissues of the embryo, including the gonads. The effect of metformin exposure on offspring was sex specific. The adult females that had been exposed to metformin in utero presented no clear reduction in fertility. However, the adult males that had been exposed to metformin during foetal life exhibited a 30% reduction in litter size compared with controls. The lower fertility was not due to a change in sperm production or the motility of sperm. Rather, the phenotype was due to lower sperm head quality – significantly increased spermatozoa head abnormality with greater DNA damage – and hypermethylation of the genomic DNA in the spermatozoa associated with lower expression of the ten-eleven translocation methylcytosine dioxygenase 1 (TET1) protein. In conclusion, while foetal metformin exposure did not dramatically alter gonad development, these results suggest that metabolic modification by metformin during the foetal period could change the expression of epigenetic regulators such as Tet1 and perturb the genomic DNA in germ cells, changes that might contribute to a reduced fertility.

Abstract

International audience; Reproduction is a complex and essential physiological process required by all species to produce a new generation. This process involves strict hormonal regulation, depending on a connection between the hypothalamus-pituitary-gonadal axis and peripheral organs. Metabolic homeostasis influences the reproductive functions, and its alteration leads to disturbances in the reproductive functions of humans as well as animals. For a long time, adipose tissue has been recognised as an endocrine organ but its ability to secrete and release hormones called adipokines is now emerging. Adipokines have been found to play a major role in the regulation of metabolic and reproductive processes at both central and peripheral levels. Leptin was initially the first adipokine that has been described to be the most involved in the metabolism/reproduction interrelation in mammals. In avian species, the role of leptin is still under debate. Recently, three novel adipokines have been discovered: adiponectin (ADIPOQ, ACRP30), visfatin (NAMPT, PBEF), and chemerin (RARRES2, TIG2). However, their mode of action between mammalian and nonmammalian species is different due to the different reproductive and metabolic systems. Herein, we will provide an overview of the structure and function related to metabolic and reproductive mechanisms of the latter three adipokines with emphasis on avian species.

Abstract

International audience; In order to follow the composition of parental broilers (Cobb500) during growth from hatching to adulthood (32 weeks of age), we evaluated kinetics of fattening, growth rate, reproduction parameters and body composition of the animals by using non-invasive tools such as medical imaging and blood sample analysing. The use of CT scanner allowed us to monitor the development of body composition on the same animals who are sleeping (fatness, bone, muscle, ovary and testis growth). These analyses were associated with biochemical blood analysis. Parental broilers fed the recommended diet by breeders. The analyses were performed with a CT scanner of bio-medical grade (Siemens As Definition ; 100K Volta and 120 mA / s). Five hundred images were performed every 0.6 mm, 0.45 Pitch and reconstruction filter Safire I26 was used to characterize fat tissues, skeletal and muscle. Image processing by thresholding was used to quantify and monitor the tissues from 4 weeks to 32 weeks of age. The acquired data were used to compare males and females. A significant difference in volume of adipose tissue was observed between male and female. From 20 weeks of age, shortly before the onset of lay, the females had 1.6 fold more adipose tissue than males (P < 0,001). In the female group, the increase in adipose tissue is associated to elevated plasma triglycerides levels (average 8 fold more in comparison to male at the beginning of the laying period. In addition, females, from 16 weeks of age, present a slow bone development in comparison to male (P < 0,001). The ratio bone/bodyweight is 30% lower in female compared to male chicken at 32 weeks of age. A section of the pectoral muscle volume was also analysed but no difference has been noted between both sexes. The correlation between the number of yellow follicles of hens and volume of ovarian adipose tissue determined in vivo was 0.80. In conclusion, the use of CT scanner and ultrasound system has permitted to follow the body composition of an actual parental breed without dissection

Abstract

Energy metabolism (in connection with growth, adipose tissue, etc.) is an important modulator of fertility. In order to improve performance in fertility, we have evaluated the feasibility to analyze the adipose tissue composition in 3 poultry species: chicken, turkey and guinea fowl by using a non-invasive method such as medical imaging (computed tomography, CTScan). In these 3 species, the adipose tissue composition varies between 10 and 15% depending the age. In particular, we carried out kinetics of fattening in the hen to associate it with the feeding, growth, and egg quality. In the experiment, meat-type chickens (Cobbgenetics) were bred with 2 diets conditions (ad libitum and restricted diet) from 3 weeks to 35 weeks of age. During this period, analyzes were performed with a biomedical scanner (Siemens, 100K Volta and 120mA / s). 500 images were taken every 0.6 mm. Image processing by thresholding was used to quantify adipose tissue. We calculated a correlation r² of 0.94 between the weight of the abdominal adipose tissue measured and the weight of the adipose tissue determined throughout the organism in vivo by computed tomography. The correlation between the chicken body weight and the volume of adipose tissue determined in vivo is r²= 0.82. The location and development of adipose tissue revealed significant differences as early as 6 weeks between ad libitum and restricted groups. In the ad libitum group, the increase in volume of adipose tissue is also associated with a high plasma triglyceride concentration and early onset of the laying period. Preliminary measurements of muscle, bone and liver volume have been measured, and allow opening new perspectives on the acquisition of new data that could be extrapolated with the feed and fertility parameters. In conclusion, this work has shown the importance of the CTscan to follow the evolution of the body composition in a non-invasive method, and to associate it with the growth parameters. Similar experiments are underway in turkey and guinea fowl., Le métabolisme énergétique (en lien avec la croissance, la masse du tissu adipeux,...) est un modulateur important de la fertilité. Afin d'améliorer les performances de fertilité, nous avons évalué la faisabilité d’analyser la composition en tissu adipeux de 3 espèces avicoles d’importance autour de la période de stimulation lumineuse : la poule, la dinde et la pintade en utilisant des outils non invasifs tels que l'imagerie médicale (tomodensitométrie, CTScan) qui permettent de localiser le tissu. Chez ces 3 espèces, lacomposition en tissu adipeux varie entre 10 et 15 % en fonction de l’âge. Plus particulièrement, nous avons réalisé une cinétique d'engraissement chez la poule afin de la mettre en lien avec l’alimentation, la croissance, et la qualité des oeufs. Pour cela, des poules de type chair (génétique Cobb) ont été nourries selon 2 régimes (ad libitum; régime restreint) de 3 semaines à 35 semaines d'âge. Sur cette période, des analyses ont été effectuées avec un scanner biomédical (Siemens Comme Définition; 100K Volta et 120 mA / s). 500 images ont été prises tous les 0,6 mm. Un traitement de l'image par seuillage a été utilisé pour quantifier le tissuadipeux. Nous avons calculé une corrélation r² de 0,94 entre le poids du tissu adipeux abdominal mesuré et le poids du tissu adipeux déterminé dans tout l’organisme in vivo par tomodensitométrie. La corrélation entre le poids corporel des poules et le volume de tissu adipeux déterminé in vivo est de r²=0,82. La localisation et le développement du tissu adipeux a révélé des différences significatives dès l’âge de 6 semaines entre les lots ad libitum et restreint. Dans le groupe ad libitum, l'augmentation de volume du tissu adipeux est également associée à une concentration élevée en triglycérides plasmatiques et un début de la période de ponte plus précoce. Des mesures préliminaires de masse musculaire, de volume osseux et du foie ont été réalisées, etpermettent d’ouvrir sur l’acquisition de nouvelles données qui pourraient être extrapolées avec les paramètres de fertilité ou de l’alimentation utilisée. En conclusion, ce travail montre l’importance du CTscan pour suivre l'évolution de la composition corporelle de façon non invasive, et de l’associer aux paramètres de croissance. Des expériences similaires sont en cours chez la dinde et la pintade.

Abstract

Effect of food restriction and fatty acid supplementation on metabolic parameters and plasma levels ofseveral adipokines in reproductive hens In livestock, reproductive hens are subjected to restricted diet to limit the decline of fertility associated with body mass changes. Thus, we have established an in vivo protocol of assessing the effect of different levels of restricted diet with or without polyunsaturated fatty acid supplementation (FAS) on metabolic parameters and plasma levels of adipokines (chemerin, visfatin, adiponectin) in reproductive hens. Our analysis indicates that hens fed with ad libitum diet (Ad, receiving an amount of feeding to achieve growth 1,7 times greater than the animals of the restricted diet) have a body weight, a percentage of fat, a surface of pectoral and “sot l’y laisse” muscles significantly (p, En élevage, les poules reproductrices sont soumises à une restriction alimentaire pour limiter la baisse de lafertilité, associée à des changements de masse corporelle. In broiler hens, rapid growth is often associated with a disruption of reproduction. One of the methods used byfarmers to limit the decline of fertility is feed restriction. The aim of this study is to assess the consequences ofthis type of feed intake management and polyunsaturated fatty acid supplementation (FAS) known to increaseadipokines (hormones produced by the adipose tissue) production, on fertility, steroidogenesis and eggs quantity and quality. Hence, we followed 320 hens divided into 4 equal groups fed with a restricted (Rt) or ad libitum (Ad, receiving an amount of feed to reach a 1.7 times heavier weight than the animals of the restricted diet), supplemented or not with FAS (1%).
