Bumblebees have been the focus of much research, but the taxonomy of many species groups is still unclear, especially for circumpolar species. Delimiting species based on multisource datasets provides a solution to overcome current systematic issues of closely related populations. Here, we use an integrative taxonomic approach based on new genetic and eco-chemical datasets to resolve the taxonomic status of Bombus lapponicus and Bombus sylvicola. Our results support the conspecific status of B. lapponicus and B. sylvicola and that the low gradual divergence around the Arctic Circle between Fennoscandia and Alaska does not imply speciation in this species complex. Therefore, based on our molecular and morphological analyses, we propose to assign them subspecific status: Bombus lapponicus lapponicus from Fennoscandia and West Siberia and Bombus lapponicus sylvicola comb. nov. from Alaska and Yukon. In addition, our analyses reveal a cryptic species in the B. lapponicus complex from Alaska, which we describe here as new: Bombus (Pyrobombus) interacti sp. nov.

International audience; AbstractSpecies taxonomy of bumblebees (Bombus Latreille, 1802) is well known to be problematic due to a potentially high intra-specific variability of morphological traits while different species can converge locally to the same color pattern (cryptic species). Assessing species delimitation remains challenging because it requires to arbitrarily select variable traits whose accuracy continues to be debated. Integrative taxonomic approach seems to be very useful for this group as different independent traits are assessed to propose a rational taxonomic hypothesis. Among operational criteria to assess specific status, the reproductive traits involved in the pre-mating recognition (i.e., the male cephalic labial gland secretions, CLGS) have been premium information. Since these secretions are supposed to be species-specific, these chemical traits can bring essential information where species delimitation is debated. Here, we describe and compare the CLGS of 161 male specimens of nine Alpinobombus taxa: alpinus, balteatus, helleri, hyperboreus, kirbiellus, natvigi, neoboreus, polaris, and pyrrhopygus. We aim also to test the congruence between this new information (reproductive traits) and published genetic dataset. Our results emphasized six distinct groups with diagnostic major compounds: (a) alpinus + helleri with hexadec-9-en-1-ol; (b) polaris + pyrrhopygus with two major compounds hexadec-9-en-1-ol and hexadec-9-enal; (c) balteatus with tetradecyl acetate; (d) kirbiellus with geranyl geranyl acetate; (e) hyperboreus + natvigi with octadec-11-en-1-ol; (f) neoboreus with octadec-9-en-1-ol. Based on this new information, we can confirm the species status of B. alpinus, B. balteatus, B. hyperboreus, B. kirbiellus, B. neoboreus, and B. polaris. We also confirm the synonymy of helleri (Alps) with alpinus (Sweden). However, the specific status of natvigi (Alaska) and pyrrhopygus (Sweden) is questionable and these taxa do not have specific CLGS composition.

Sex-specific chemical secretions have been widely used as diagnostic characters in chemotaxonomy. The taxonomically confused group of bumblebees has reaped the benefit of this approach through the analyses of cephalic labial gland secretions (CLGS). Most of currently available CLGS descriptions concern species from the West-Palearctic region but few from the New World. Here, the CLGS of four East-Palearctic species Bombus deuteronymus, B. filchnerae, B. humilis, and B. exil (subgenus Thoracobombus) are analysed. Our results show high levels of variability in the major compounds in B. exil. In contrast, we describe a low differentiation in CLGS compounds between B. filchnerae and its phylogenetically closely related taxon B. muscorum. Moreover, the chemical profiles of B. filchnerae and B. muscorum are characterized by low concentrations of the C16 component, which is found in higher concentrations in the other Thoracobombus species. This raises the possibility that courtship behavior as well as environmental constraints could affect the role of the bumblebee males' CLGS.

International audience; The species international trade leads to multiple non-native invasions. Besides species invasions, commercial exchanges may also contribute to translocation between closely related taxa or allopatric populations. Consequently, preserving endemic taxa and specificity of local populations require to regulate commercial translocations of species or populations. To be efficient such regulation needs a resolved taxonomy and a thorough analysis of the population structure of native taxa/populations. To provide guidelines for an efficient regulation of the trade of Bombus terrestris within its natural range, we analyzed its taxonomy and its population structure using an integrative taxonomic approach. Our results show that B. terrestris translocations involve two species, three subspecies, and several populations with weak differentiation. These different levels of differentiation imply specific and appropriate regulations of translocations with different levels of prioritization. We ultimately assess the relevance of current policies and propose potentially efficient regulations for policy-makers. Such integrative taxonomic approach should be used in other traded polytypic species.

International audience; In social insects, the high cost of parental investment has promoted the evolution of traits that reduce or avoid care for unrelated offspring. They have evolved a sophisticated recognition system which enables them to reject foreign individuals, and therefore prevent their nest's exploitation by parasites and predators. Nonetheless, many social parasites are able to overcome social insect recognition and parasitize their parental care behaviour by laying their eggs in host nests. Although several studies have already been conducted on the strategies used by social parasites to successfully invade host nests, the strategy used by its offspring to evade host rejection is still poorly understood. In bumblebees, previous investigations indicated that male offspring of Bombus vestalis (Geoffroy) (Hymenoptera: Apidae), the specific obligate social parasite of Bombus terrestris (L.), may use allomones to evade host worker attacks. Following these studies, we performed electrophysiological studies and behavioural experiments to identify the potential repellents. Six compounds elicited electroantennographic responses in host worker antennae. The corresponding mixture of synthetic volatiles as well as pure tetradecyl acetate, and to a lesser extent pure (Z)-11-hexadecenyl acetate, showed a repellent effect on starved host workers in a feeding bioassay. We hypothesize that tetradecyl acetate has a key function in the repellent effect of the secretions of B. vestalis male offspring towards B. terrestris host workers.

International audience; The evolution of signals and reproductive traits involved in the pre-mating recognition has been in focus of abundant research in several model species, such as bumblebees (genus Bombus). However, the most-studied bumblebee reproductive trait, the male cephalic labial gland secretions (CLGS), remains unknown among bumblebee species from South America. In this study, the CLGS of five South American bumblebees of the subgenera Thoracobombus (Bombus excellens and B. atratus) and Cullumanobombus (B. rubicundus, B. hortulanus, and B. melaleucus) were investigated, by comparing the chemical compositions of their secretions to those of closely related European species. The results showed an obvious interspecific differentiation in both subgenera. The interspecific differentiation among the species of the Thoracobombus subgenus involved different compounds present at high contents (main compounds), while those of the Cullumanobombus subgenus shared the same main components. This suggests that among the species of the Cullumanobombus subgenus, the differentiation in minor components could lead to species discrimination.

International audience; Delimitation of closely related species is often hindered by the lack of discrete diagnostic morphological characters. This is exemplified in bumblebees (genus Bombus). There have been many attempts to clarify bumblebee taxonomy by using alternative features to discrete morphological characters such as wing shape, DNA, or eco-chemical traits. Nevertheless each approach has its own limitations. Recent studies have used a multisource approach to gather different lines of speciation evidence in order to draw a strongly supported taxonomic hypothesis in bumblebees. Yet, the resulting taxonomic status is not independent of selected evidence and of consensus methodology (i.e. unanimous procedure, majority, different weighting of evidence). In this article, we compare taxonomic conclusions for a group of taxonomically doubtful species (the Bombus lapidarius-group) obtained from the four commonly used lines of evidence for species delimitation in bumblebees (geometric morphometric of wing shape, genetic differentiation assessment, sequence-based species delimitation methods and differentiation of cephalic labial gland secretions). We ultimately aim to assess the usefulness of these lines of evidence as components of an integrative decision framework to delimit bumblebee species. Our results show that analyses based on wing shape do not delineate any obvious cluster. In contrast, nuclear/mitochondrial, sequence-based species delimitation methods, and analyses based on cephalic labial gland secretions are congruent with each other. This allows setting up an integrative decision framework to establish strongly supported species and subspecies status within bumblebees.

International audience; AbstractMany species display local variations in pre-mating signals and in mating preferences. This may lead to discrimination against potential foreign mates that may ultimately lead to reproductive isolation. However, the extent to which population differentiation in mating cues affects the species recognition has received little empirical support. Here, we investigate the consequence of geographic differentiation in male reproductive traits on female preferences to these traits in Bombus terrestris. We characterise (1) the geographic differentiation in male cephalic labial gland secretions (CLGS), a key trait for mate attraction, and (2) the preference of virgin females to the CLGS of different subspecies. Our results show geographic CLGS differences parallel with divergences in female preferences for these secretions. This geographic CLGS differentiation in males, along with female preference for sympatric males, could lead to or reflect a pre-mating isolation among subspecies.

Populations on islands often exhibit lower levels of genetic variation and ecomorphological divergence compared to their mainland relatives. While phenotypic differentiation in characters, such as size or shape among insular organisms, has been well studied, insular differentiation in quantitative reproductive traits involved in chemical communication has received very little attention to date. Here, we investigated the impact of insularity on two syntopic bumblebee species pairs: one including species that are phylogenetically related (Bombus terrestris and B. lucorum), and the other including species that interact ecologically (B. terrestris and its specific nest inquiline B. vestalis). For each bumblebee species, we characterized the patterns of variation and differentiation of insular (Corsican) vs. mainland (European) populations (i) with four genes (nuclear and mitochondrial, 3781 bp) and (ii) in the chemical composition of male marking secretions (MMS), a key trait for mate attraction in bumblebees, by gas chromatography-mass spectrometry (GC-MS). Our results provide evidence for genetic differentiation in Corsican bumblebees and show that, contrary to theoretical expectations, island populations of bumblebees exhibit levels of genetic variation similar to the mainland populations. Likewise, our comparative chemical analyses of MMS indicate that Corsican populations of bumblebees are significantly differentiated from the mainland yet they hold comparative levels of within-population MMS variability compared to the mainland. Therefore, insularity has led Corsican populations to diverge both genetically and chemically from their mainland relatives, presumably through genetic drift, but without a decrease of genetic diversity in island populations. We hypothesize that MMS divergence in Corsican bumblebees was driven by a persistent lack of gene flow with mainland populations and reinforced by the preference of Corsican females for sympatric (Corsican) MMS. Many methods, based on morphological, molecular or chemical characters, have been used to address the question of species taxonomic status. Integrative taxonomy aims to define stronger supported taxonomic hypotheses by considering complementary datasets from different characters. By following an integrative approach, the present study includes molecular, chemical and morphological criteria to establish the taxonomic status of two rare and doubtful cuckoo bumblebee taxa: Bombus (Psithyrus) barbutellus and Bombus (Psithyrus) maxillosus. These two sympatric taxa are discriminated by few morphological criteria (mainly wing darkness and hair length). We used these morphological character diagnoses to establish an a priori status of our samples (23 specimens). We developed a combined molecular dataset from one nuclear gene, elongation factor 1α (EF-1α), and one mitochondrial gene, cytochrome c oxidase subunit I (COI), spanning 1623 bp, and a chemical dataset of sexual marking pheromones (73 compounds). The molecular data were subjected to maximum-likelihood and Bayesian phylogenetic inference under partitioned model and maximum parsimony. The chemical data were analysed by clustering and the two-group k-means method to test divergences between the two species. The resulting phylogenetic trees show no consistent divergence between the two taxa. Moreover, we found no divergence in the sexual marking pheromones in the clustering and two-group k-means analyses. These converging results support the conspecificity of both taxa. Nonetheless, our determinations using the traditional morphological criteria separated our samples into two taxa. We conclude that the morphological criteria seem to relate to intraspecific variations: B. maxillosus is regarded as a syn.n. of B. barbutellus. © 2011 The Authors. Il n'est pas possible d'identifier avec precision les especes de bourdons d'apres leurs caracteres morphologiques ou leurs couleurs. Par contre, les pheromones secretees par les glandes labiales des mâles sont fortement specifiques et bien adaptees a la definition d'une espece basee sur le concept de reconnaissance d'especes. Les taxons presentant les memes combinaisons de pheromones labiales peuvent etre consideres comme conspecifiques (= de la meme espece). Les auteurs ont re-examine le statut d'espece de bourdons de la zone palearctique occidentale appartenant au sous-genre Siricobombus Vogt : Bombus niveatus, B. sulfureus et B. vorticosus. Ces trois taxons vivent de preference dans les steppes de montagne : B. niveatus et B. sulfureus se limitent au Proche-Orient (Figs. 2, 3), B. vorticosus est egalement present dans les Balkans (Fig. 4). B. niveatus et B. vorticosus ne peuvent etre differencies morphologiquement que par la coloration et ne different de B. sulfureus que par la couleur du pelage et par des caracteres mineurs des genitalia (Fig. I). Parmi les 40 composes que nous avons identifies dans les secretions des glandes labiales, sept le sont pour la premiere fois. Les trois taxons du sous-genre Sibiricobombus different de tous les autres par une concentration en terpenes relativement plus grande que celle des autres composes et par le fait que les composes non terpeniques se limitent a des esters et a des acides gras simples. Alors que les secretions des glandes cephaliques des mâles de B. sulfureus sont tres differentes de celles de B. niveatus et B. vorticosus, nous n'avons pas trouve de difference significative entre ces dernieres (Tab. III). Nous en concluons que B. sulfureuse une espece valable, tandis que B. vorticosus n'est qu'une sous-espece de B. niveatus : Bombus niveatus ssp. vorticosus Gerstaecker nov. stalus.

Graphical Abstract Full-size image; International audience; Female alates of Nasutitermes ephratae termites from Guadeloupe and Nasutitermes sp. from Brazil produce a diterpene hydrocarbon of the molecular formula C20H30 as the main component of their tergal gland secretion. Analysis of NMR, IR, and mass spectra of the diterpene led to a structure of (11E)-trinervita-1(14),2,11-triene. Based on a comparison with the published oxygenated trinervitane skeleton from termites we prefer the enantiomer with absolute configurations (4R,7S,8R,15S,16S). The suggested structure is supported by ab initio quantum chemical calculation of 1H and 13C chemical shifts for the optimized geometry of the molecule.
