Software Defined Networking (SDN) is a networking architecture within the control is centralized through a software-based controller. Like Cyber-Physical Systems manager, this centralization eases the support of advanced application. Being a single point of attack makes the controller a preferred target in case of attack. To enhance the control plane against cyberattacks, an observer is introduced and is in charge of the detection of cyber-attacks on the nominal controller. In this objective, a detection of anomalies method in the activity of the control is proposed. This activity is defined as the events at the interface of communication between the controller and the network plant. In this paper, a non-deterministic control is considered which means that the decisions are stochastic. Hence, a probabilistic approach is proposed which aims to evaluate the deviation of the likelihood of the sequence of decisions taken by the controller. The formalism used to determine the likelihood is the Hidden Markov Model which permits to infer over the internal states of the controller through the observations. This method is discussed on a network case study.

International audience; Software Defined Networking (SDN) is a networking paradigm which proposed to decouple the forwarding and the control planes. Security and safety threat challenges at the control level are divided into the reinforcement of the controller, whatever the reason. This work aims to consider both threats and pave the way for a multi-controller architecture without East-West interface. Considering one nominal controller in charge of the data plane computation, we designed a second one in order to control the consistency of the decisions made by the controller, i.e. only through observing the activity of the command (i.e. the management traffic). Compared to related works, no direct exchanges between the controllers are required. The detection logic is introduced theoretically and it mainly relies on two phases: the learning of the decisions and the verification that each decision taken fits with the data plane estimate. The algorithm, implemented on ONOS, is discussed in a case study.

International audience; Purpose: Both perceived and objectively-measured attributes of the built environment have been related to active transport. They do not refer, however, to the same construct: objective measurements provide an inventory of a neighborhood?s active-friendly attributes, while the perceptions of individuals refer to their appraisal of their neighborhood opportunities for active travel. Both aspects are complementary in explaining active transport. This study examines the independent associations of perceived and objective environmental attributes with active transport, and whether perceptions moderate the objectively-measured environment active transport relationship.Methods: Based on a sample of 2670 participants from the NutriNet-Sante? Study (France), we examined (i) the concordance between perceptions and objective measurements of residentialbased environmental attributes using kappa statistics, (ii) the associations between perceived and objectively-measured attributes and active transport by trip purpose (commuting and errand) using zero-inflated negative-binomial regressions, and (iii) the potential moderation effect of perceptions on the associations between objectively-measured environmental attributes and active transport. Information related to demographics, perceptions and transport modes was collected in 2013.Results: A poor to fair level of agreement was observed between perceived , objectively -measured environmental attributes; both were associated with active transport for commuting and errands. Environmental correlates differed between the likelihood to engage in active transport and the time spent in transport. Perceptions positively moderated the associations be-tween active transport and objectively-measured walkability and access to facilities. The differ-ences in predicted active transport time between a high versus low perception of the environmental attributes for a given neighborhood ranged from 20 to 55 min for active commuting , from 84 to 37 min for active transport for errands.Conclusion: Associations between objectively-measured environmental attributes and active transport are dependent on individuals & rsquo; perceptions of opportunities provided by their residential neighborhood. Increasing individuals & rsquo; awareness of active-friendly environmental attributes may be crucial for enhancing active transport.

Skeletal muscle regenerates ad integrum after a sterile acute injury thanks to satellite cells (muscle stem cells). Inflammation, and notably macrophages, plays important roles during this process. Just after injury, monocytes infiltrate the tissue from the blood and convert into pro-inflammatory damaged associated macrophages. These macrophages phagocyte muscle debris and promote the proliferation of muscle stem cells. Then, macrophages switch their phenotype toward an anti-inflammatory restorative profile and promote muscle stem differentiation, fusion and myofiber growth. This sequence of macrophage profile is essential for an efficient skeletal muscle regeneration. The lab has shown that this phenotype switch is dependent of AMP kinase (AMPK)a1, a major energetic sensor in the cell controlling cellular metabolism. Besides, glucocorticoids have been used for decades for their anti-inflammatory effects on inflammation. Their actions are mediated by the Glucocorticoid Receptor which induces or represses gene expression by direct or indirect DNA-binding. As AMPKa1 and glucocorticoids induce similar anti-inflammatory effects on macrophages, we hypothesized that these 2 pathways could be interconnected in macrophages to allow the resolution of inflammation and muscle repair. Data from an in vitro model of skeletal muscle injury using bone marrow derived macrophages showed that: i) glucocorticoids induce AMPK phosphorylation; ii) AMPKa1 is required for the functional acquisition of the anti-inflammatory phenotype induced by glucocorticoids. Indeed, AMPKa1-deficient macrophages did not switch their phenotype and did not sustain myogenesis. In vivo experiments using LysMCre/+;AMPKa1fl/fl mice in which AMPKa1 is depleted only in myeloid cells, showed that macrophagic AMPK drove the beneficial effects of glucocorticoids during skeletal muscle regeneration. Inversely, in absence of AMPK in macrophages, glucocorticoids induced a delayed muscle regeneration and a modification in myofiber maturation, assessed by the alteration of myosin heavy chain expression. Adult skeletal muscle is capable of complete regeneration after an acute injury. The main parameter studied to assess muscle regeneration efficacy is the cross-sectional area (CSA) of the myofibers as myofiber size correlates with muscle force. CSA analysis can be time-consuming and may trigger variability in the results when performed manually. This is why programs were developed to completely automate the analysis of the CSA, such as SMASH, MyoVision, or MuscleJ softwares. Although these softwares are efficient to measure CSA on normal or hypertrophic/atrophic muscle, they fail to efficiently measure CSA on regenerating muscles. We developed Open-CSAM, an ImageJ macro, to perform a high throughput semi-automated analysis of CSA on skeletal muscle from various experimental conditions. The macro allows the experimenter to adjust the analysis and correct the mistakes done by the automation, which is not possible with fully automated programs. We showed that Open-CSAM was more accurate to measure CSA in regenerating and dystrophic muscles as compared with SMASH, MyoVision, and MuscleJ softwares and that the inter-experimenter variability was negligible. We also showed that, to obtain a representative CSA measurement, it was necessary to analyze the whole muscle section and not randomly selected pictures, a process that was easily and accurately be performed using Open-CSAM. To conclude, we show here an easy and experimenter-controlled tool to measure CSA in muscles from any experimental condition, including regenerating muscle. Analysis of circulating cell-free fetal DNA (cffDNA) in maternal plasma is very promising for early diagnosis of monogenic diseases. However, this approach is not yet available for routine use and remains technically challenging because of the low concentration of cffDNA, which is swamped by the overwhelming maternal DNA.To make clinical applications more readily accessible, we propose a new approach based on mutant enrichment with 3′-modified oligonucleotides (MEMO) PCR along with real-time PCR to selectively amplify from the maternal blood the paternally inherited fetal allele that is not present in the maternal genome.The first proof of concept of this strategy was displayed for cystic fibrosis by the accuracy of our detection of the p.Gly542* mutation used as the initial developmental model. Subsequently, a retrospective study of plasmas originating from two pregnant women carrying a fetus with private mutation confirmed the effectiveness of our method. We confirmed the presence of cffDNA in the studied samples by the identification of a tri-allelic DNA profile using a miniSTR kit.This new non-invasive prenatal diagnosis test offers numerous advantages over current methods: it is simple, cost effective, time efficient and does not require complex equipment or bioinformatics settings. Moreover, our assays for different private mutations demonstrate the viability of this approach in clinical settings for monogenic disorders.

Times Cited: 0; International audience; In skeletal muscle, autophagy is activated in multiple physiological and pathological conditions, notably through the transcriptional regulation of autophagy-related genes by FoxO3. However, recent evidence suggests that autophagy could also be regulated by post-transcriptional mechanisms. The purpose of the study was therefore to determine the temporal regulation of transcriptional and post-transcriptional events involved in the control of autophagy during starvation (4 h) and nutrient restoration (4 h) in C2C12 myotubes. Starvation was associated with an activation of autophagy (decrease in mTOR activity, increase in AMPK activity and Ulk1 phosphorylation on Ser467), an increase in autophagy flux (increased LC3B-II/LC3B-I ratio, LC3B-II level and LC3B-positive punctate), and an increase in the content of autophagy-related proteins (Ulk1, Atg13, Vps34, and Atg5-Atg12 conjugate). Our data also indicated that the content of autophagy-related proteins was essentially maintained when nutrient sufficiency was restored. By contrast, mRNA level of Ulk1, Atg5, Bnip3, LC3B and Gabarapl1 did not increase in response to starvation. Accordingly, binding of FoxO3 transcription factor on LC3B promoter was only increased at the end of the starvation period, whereas mRNA levels of Atrogin1/MAFbx and MuRF1, two transcriptional targets of FoxO involved in ubiquitin-proteasome pathway, were markedly increased at this time. Together, these data provide evidence that target genes of FoxO3 are differentially regulated during starvation and that starvation of C2C12 myotubes is associated with a post-transcriptional regulation of autophagy.

Cachexia is a muscle-wasting syndrome that contributes significantly to morbidity and mortality of many patients with advanced cancers. However, little is understood about how the severe loss of skeletal muscle characterizing this condition occurs. In the current study, we tested the hypothesis that the muscle protein myostatin is involved in mediating the pathogenesis of cachexia-induced muscle wasting in tumor-bearing mice. Myostatin gene inactivation prevented the severe loss of skeletal muscle mass induced in mice engrafted with Lewis lung carcinoma (LLC) cells or in ApcMin/+ mice, an established model of colorectal cancer and cachexia. Mechanistically, myostatin loss attenuated the activation of muscle fiber proteolytic pathways by inhibiting the expression of atrophy-related genes, MuRF1 and MAFbx/Atrogin-1, along with autophagy-related genes. Notably, myostatin loss also impeded the growth of LLC tumors, the number and the size of intestinal polyps in ApcMin/+ mice, thus strongly increasing survival in both models. Gene expression analysis in the LLC model showed this phenotype to be associated with reduced expression of genes involved in tumor metabolism, activin signaling, and apoptosis. Taken together, our results reveal an essential role for myostatin in the pathogenesis of cancer cachexia and link this condition to tumor growth, with implications for furthering understanding of cancer as a systemic disease. Background Muscle atrophy associated with various pathophysiological conditions represents a major health problem, because of its contribution to the deterioration of patient status and its effect on mortality. Although the involvement of pro-inflammatory cytokines in this process is well recognized, the role of sphingolipid metabolism alterations induced by the cytokines has received little attention. Results We addressed this question both in vitro using differentiated myotubes treated with TNF-α, and in vivo in a murine model of tumor-induced cachexia. Myotube atrophy induced by TNF-α was accompanied by a substantial increase in cell ceramide levels, and could be mimicked by the addition of exogenous ceramides. It could be prevented by the addition of ceramide-synthesis inhibitors that targeted either the de novo pathway (myriocin), or the sphingomyelinases (GW4869 and 3-O-methylsphingomyelin). In the presence of TNF-α, ceramide-synthesis inhibitors significantly increased protein synthesis and decreased proteolysis. In parallel, they lowered the expression of both the Atrogin-1 and LC3b genes, involved in muscle protein degradation by proteasome and in autophagic proteolysis, respectively, and increased the proportion of inactive, phosphorylated Foxo3 transcription factor. Furthermore, these inhibitors increased the expression and/or phosphorylation levels of key factors regulating protein metabolism, including phospholipase D, an activator of mammalian target of rapamycin (mTOR), and the mTOR substrates S6K1 and Akt. In vivo, C26 carcinoma implantation induced a substantial increase in muscle ceramide, together with drastic muscle atrophy. Treatment of the animals with myriocin reduced the expression of the atrogenes Foxo3 and Atrogin-1, and partially protected muscle tissue from atrophy. Conclusions Ceramide accumulation induced by TNF-α or tumor development participates in the mechanism of muscle-cell atrophy, and sphingolipid metabolism is a logical target for pharmacological or nutritional interventions aiming at preserving muscle mass in pathological situations.

International audience; Growing evidences show that functionally relevant polymorphisms in various promoters alter both transcriptional activity and affinities of existing protein-DNA interactions, and thus influence disease progression in humans. We previously reported the -94G>T CFTR promoter variant in a female CF patient in whom any known disease-causing mutation has been detected. To investigate whether the -94G>T could be a regulatory variant, we have proceeded to in silico analyses and functional studies including EMSA and reporter gene assays. Our data indicate that the promoter variant decreases basal CFTR transcriptional activity in different epithelial cells and alters binding affinities of both Sp1 and USF nuclear proteins to the CFTR promoter. The present report provides evidence for the first functional polymorphism that negatively affects the CFTR transcriptional activity and demonstrates a cooperative role of Sp1 and USF transcription factors in transactivation of the CFTR gene promoter.

International audience; This chapter considers the notion of urban ambience and attempts to characterise its generative dimension separately from the growing social demand for conditioned atmospheres (supermarkets, subways, etc.). Following the work of Gilles Deleuze, this paper revises the idea of perception as an activity of knowledge and focuses on it as an aesthetic experience. This is a process that utilises the notions of measure and rhythm. Ordinary perception would consist of a process of adjusting our knowledge to our lived environment, a case in which the measure is major and the rhythm is minor – or a classic process of recognition. Perception is also a fruitful field for aesthetic experiences, lived in daily situations in which the rhythm would become major and the measure minor. Our assumption is that Venice’s ambiences, as Proust describes them, can be traced to this outburst of the synthesis of perception: Measure fails to build an object, rhythm falls into a proliferative perceptual experience, and imagination no longer serves the purpose of comprehension but instead produces a very expressive space-time without any concept. The chapter builds on these notions (measure, rhythm) to discuss the idea of ambience as an introductory experience to our ordinary environment.
