Inbreeding and inbreeding depression are processes in small populations of particular interest for a range of human activities such as animal breeding, species conservation or pest management. In particular, biological control programs should benefit from a thorough understanding of the causes and consequences of inbreeding because natural enemies experience repetitive bottlenecks during importation, laboratory rearing, and introduction. Predicting the effect of inbreeding in Hymenopteran parasitoid wasps, frequently used in biological control programs, is nonetheless a difficult endeavor. In haplodiploid parasitoids, the purge of deleterious alleles via haploid males should reduce genetic load, but if these species also have complementary sex determination (CSD) abnormal diploid males will be produced, which may jeopardize the success of biological control introductions. Mastrus ridens is such a parasitoid wasp with CSD, introduced to control the codling moth, Cydia pomonella (L.). We studied its life history traits in the laboratory under two conditions: inbred (full sib) and outbred (non-sib) crosses, across five generations, in order to examine the consequences of inbreeding in this species. We found that in inbred lines non reproducing females live less, the number of daughters produced was lower, and that sex ratio (proportion of males) and proportion of diploid males were higher. Diploid males were able to produce fertile daughters, but fewer than haploid males. Lineage survival was similar for inbred and outbred lines across the five generations. The most significant decrease in fitness was thus a consequence of the production of diploid males, but this effect was not as extreme as in most other species with CSD, due to the fertility of diploid males. This study highlights the importance of determining the type of sex determination in parasitoid wasps used for biological control, and the importance of maintaining genetic diversity in species with CSD when importation or augmentation is the goal. International audience; The current bumblebee decline leads to inbreeding in populations that fosters a loss of allelic diversity and diploid male production. As diploid males are viable and their offspring are sterile, bumblebee populations can quickly fall in a vortex of extinction. In this article, we investigate for the first time a potential premating mechanism through a major chemical reproductive trait (male cephalic labial gland secretions) that could prevent monandrous virgin queens from mating with diploid males. We focus our study on the cephalic labial gland secretions of diploid and haploid males of Bombus terrestris (L.). Contrary to initial expectations, our results do not show any significant differentiation of cephalic labial gland secretions between diploid and haploid specimens. Queens seem therefore to be unable to avoid mating with diploid males based on their compositions of cephalic labial gland secretions. This suggests that the vortex of extinction of diploid males could not be stopped through premating avoidance based on the cephalic labial gland secretions but other mechanisms could avoid mating between diploid males and queens.

Sex determination is ruled by haplodiploidy in Hymenoptera, with haploid males arising from unfertilized eggs and diploid females from fertilized eggs. However, diploid males with null fitness are produced under Complementary Sex Determination (CSD), whenindividuals are homozygous for this locus. Diploid males are expected to be more frequent in genetically eroded populations (such as islands and captive populations), as genetic diversity at the csd locus should be low. However, only a few studies have focused on the relation between population size, genetic diversity and the proportion of diploid males in the field. Here, we developed new microsatellites markers in order to assess and compare genetic diversity and diploid male proportion in populations from three distinct habitat types (mainland, island or captive), in the parasitoid waspVenturia canescens. Eroded genetic diversity and higher diploid male proportion were found in island and captive populations, and habitat type had large effect on genetic diversity. Therefore, diploid male proportion reflects the decreasing genetic diversity in small and isolated populations. Thus, Hymenopteran populations can be at high extinction risk due to habitat destruction or fragmentation.

International audience; The parasitoid wasp Mastrus ridens (Hymenoptera: Ichneumonidae) is a particularly well-suited biological model to document the history and evolution of populations used in classical biological control, repeatedly moved from laboratory to laboratory worldwide and introduced in various environments. This specialist ectoparasitoid of the codling moth was first imported from Kazakhstan to the USA in the 1990's, and then sent to Argentina, Chile and New Zealand. More recently, it was sent to Australia and France from other laboratory colonies, and imported again from field collections in Kazakhstan to Chile. Here, we used DNA sequencing to confirm the taxonomic identity of several populations used for biological control worldwide, and developed microsatellite markers for population genetics studies. A multiplex PCR amplifying 11 polymorphic markers was designed. These markers were used to compare the genetic diversity of laboratory and field populations and evaluate genetic differentiation between them. Results showed that laboratory populations with the longest rearing history had lower genetic diversity. Moreover, the geno-typing of males with the markers revealed the occurrence of diploid males, which was further confirmed by flow cytometry, suggesting complementary sex determination (CSD) in this species. The percentage of diploid males in the populations ranged from 4% to 25% and were negatively correlated with diversity indices, which is consistent with a single-locus CSD and genetic bottlenecks in laboratory rearings. Molecular tools proved to be suitable and reliable for genetic diversity analyses in M. ridens, and should be implemented more frequently in classical biological control programs.

International audience; AbstractPresence of diploid males in wild bees reflects inbreeding and provides information about the health status of a colony or population. Detection of diploid males, and discrimination from haploid males and workers, has, however, been limited to molecular diagnostics. Here we present a novel method based on differences in wing shape, e.g., venation patterns in wings. The method is easy to apply and results, for Bombus terrestris, in very high discrimination success. Possible applications of the method are discussed.

Background Single-locus complementary sex determination (sl-CSD), which occurs in some insects of the order Hymenoptera, imposes a heavy genetic load that can drive small populations to extinction. The core process in these species is the development of individuals homozygous at the sex-determining locus into unfit diploid males. The risk of extinction of populations with sl-CSD is theoretically much higher if diploid males are viable and capable of mating but sterile, because diploid males then decrease the reproductive output of both their parents and the females with which they mate. Results In the parasitoid wasp Venturia canescens (Hymenoptera: Ichneumonidae), diploid males resembled their haploid counterparts in most respects, but their mating success was nevertheless lower than that of haploid males, especially when the two types of males were placed in competition. Furthermore, although diploid males transferred viable sperm during copulation, they sired no daughters: the females with which they mated produced only sons, like virgin females. A simulation model combining behavior, genetics and demography demonstrated that for two alternative hypotheses concerning the fertilization success of diploid sperm, the mating success of diploid males strongly affected population dynamics. Conclusion The performance of diploid males should be estimated in competitive situations. It is a crucial determinant of the probability of extinction. Under the hymenopteran single-locus complementary sex-determination system, production of diploid males results from homozygosity at the sex-determiner locus. This arises when both parents transmit identical alleles at the locus to the offspring. In species reproducing asexually through thelytokous parthenogenesis, production of diploid males may also occur when the sex locus undergoes recombination and becomes homozygous in the offspring. Diploid males represent a substantial genetic load in hymenopteran populations because they often produce unviable sperm or sire sterile triploid female offspring. In the Mediterranean ant Cataglyphis cursor, the queen and workers can produce female offspring through automictic thelytokous parthenogenesis with central fusion, a mode of parthenogenesis that increases homozygosity. We report, for the first time, the presence of about 39 % of colonies producing adult diploid males (seven colonies out of 18). Overall, 8 % of adult males were diploid (12 diploid males out of the 146 males genotyped). Genotyping workers from the seven colonies producing diploid males showed that three diploid males were sons of queens and produced by thelytoky, six were probably sons of workers also produced by thelytoky and three were non-natal. Furthermore, the mating of a diploid male with two virgin queens in the laboratory led to the production of sterile triploid workers, which shows that diploid males in C. cursor are fertile, mate successfully and produce viable and functional but probably sterile female offspring. Because diploid males originate from thelytokous reproduction, they are only produced during sexual production and hence do not impair colony growth, which could explain why they are not removed at early brood stages. © 2013 Springer-Verlag Berlin Heidelberg., SCOPUS: ar.j, info:eu-repo/semantics/published

International audience; The emerging threat of pollinator decline has motivated research on bee conservation biology in order to both understand the causes of declines and to develop appropriate conservation strategies. The application of genetics to the conservation of diploid animals has proven to be important for both overcoming genetic threats to population viability and for providing tools to guide conservation programs. However, the haplodiploid bees have several unusual genetic properties of relevance to their conservation, which warrant special attention. Here I review how haplodiploidy and complementary sex determination affect genetic parameters pertinent to the viability and future evolutionary potential of bee populations. I also review how genetic tools can improve the conservation management of bees. I find that bees are especially prone to extinction for genetic reasons, and that genetics can provide invaluable tools for managing bee populations to circumvent pollinator decline.
