1. The Chemistry of P=N−P=X (X=S, O, NR) Linkages for the Synthesis of Dendritic Structures

Abstract: International audience; The Staudinger reaction between a phosphine and an azide, applied to phosphorus azides, has been used for the synthesis of a large variety of dendritic structures, incorporating P=N−P=X moieties (X = mainly S, but also O and N−R). Conjugation of the P=N bond with the P=X bond greatly stabilizes the P=N bond. Highly branched structures such as dendrons, dendrimers, Janus dendrimers, layered dendrimers, surface-block dendrimers, and diverse other dendritic structures incorporating such linkage have been elaborated. Accelerated methods of synthesis of dendrimers are also based on the Staudinger reaction. A versatile reactivity was observed exclusively on the sulfur atom of P=N−P=S linkages, such as alkylation or complexation. Alkylation on S induces a weakening of the strength of the P=S bond, which can be easily cleaved to generate phosphines able to react in Staudinger reactions inside the structure of dendrimers, thus affording highly sophisticated dendritic structures.

Abstract: International audience; Combined chemo/gene therapy of cancer through different action mechanisms has been emerging to enhance the therapeutic efficacy towards cancer, and still remains a challenging task due to the lack of highly effective and biocompatible nanocarriers. In this work, we report a new nanosystem based on amphiphilic phosphorus dendron (1-C12G1) micelles to co-deliver microRNA-21 inhibitor (miR-21i) and doxorubicin (DOX) for combination therapy of triple negative breast cancer. The amphiphilic phosphorus dendron bearing a long linear alkyl chain and ten protonated pyrrolidine surface groups was prepared and was demonstrated to form micelles in water solution and have a hydrodynamic size of 103.2 nm. The micelles are shown to be stable, enable encapsulation of an anticancer drug DOX with optimal loading content (80%) and encapsulation efficiency (98%), and can compress miR-21i to form polyplexes to render it with good stability against degradation. The co-delivery system of 1-C12G1@DOX/miR-21i polyplexes has a pH-dependent DOX release profile, and can be readily phagocytosed by cancer cells to inhibit them due to the different anticancer mechanisms, which was further validated after intravenous injection to treat an orthotopic triple-negative breast tumor model in vivo. With the proven biocompatibility under the studied doses, the developed amphiphilic phosphorus dendron micelles could be developed as an effective nanomedicine formulation for synergistic cancer therapy.

Abstract: International audience; This review presents the synthesis of dendrons (dendritic wedges) based on cyclotriphosphazene cores, in which one function is a fluorescent group, and the five remaining functions are used for growing the dendritic branches. The growing of the branches is carried out by a divergent method, using a two-step process which implies hydroxybenzaldehyde and the phosphorhydrazide H2NNMeP(S)Cl2. Four different fluorophores have been used, derived from maleimide, julolidine, pyrene, or from a near-infra-red fluorophore. Depending on the type of fluorophore and on the type of terminal functions of the dendrons, different applications have been targeted. One can cite experiments in catalysis or for the elaboration of materials, transfection experiments, the study of anti-cancer properties, and imaging in relation with the human immune system.

Abstract: International audience; Hydrogels are hydrophilic, three-dimensional networks able to imprison large amounts of water and are largely used in pharmaceutical formulations. Hydrogels are frequently obtained from hydrophilic polymers, either natural, biohybrid, or synthetic. Owing to their peculiar structure, dendrimers can be considered prospective building blocks for hydrogel networks. This review gathers the use of different types of amphiphilic dendritic structures able to generate physical hydrogels alone. Such dendritic structures comprise dendrimers, Janus dendrimers, and dendrons. The first part concerns different types of positively charged phosphorus dendrimers used to generate hydrogels, which are also suitable to form fibers, and for encapsulating diverse substances, or forming complexes with genetic materials for their slow delivery. The second part concerns PAMAM dendrimers functionalized with collagen mimetics. The third part concerns amphiphilic Janus dendrimers, whereas the fourth part displays different types of amphiphilic dendrons and their use, in particular in the fields of materials and drug delivery.

Abstract: Automated X-ray data recording and data reduction strategies are straightforward and powerful these days, as well as semi-automated routines to solve standard structures without, or at least with only little manual intervention and within a relatively short time. The structure determination of the amino-carborane closo-9-[4-(dibenzylamino)phenyl]-1,2-dicarbadodecaborane(12), C22H29B10N, is an example that automated routines still always have to be revised and that it may be necessary to be aware of possible problems of a structure determination.

Abstract: International audience; Dendrimers are highly branched macromolecules tailorable at will to fulfil precise requirements. They have generated a great many expectations and a huge number of publications and patents in relation to medicine, including in relation to personalized medicine, but have resulted in very poor clinical translation up to now. As clinical trials are the first steps in view of developing new compounds for (a personalized) medicine, this review focusses on the clinical trials carried out with dendrimers. Many of these clinical trials have been recently posted (2020–2022); thus, only very few concern phase 3. The safety and efficiency of essentially two main types of dendrimers, based on polylysine and polyamidoamide scaffolds, have been assessed up to now. These dendrimers were tested with the aim of treating mainly bacterial vaginosis, cancers, and COVID-19.

Abstract: International audience; Dendrimers are highly branched macromolecules possessing, in most cases, identical terminal functions. However, it is sometimes desirable to have two types of surface functions in order to fulfil specific properties. The stochastic functionalization is frequently used for such purposes, but the presence of an uncontrolled number of each type of terminal function, albeit acceptable for research purposes, has no practical use. Thus, it is highly desirable to find strategies suitable for the precise grafting of two different functional groups on the surface of dendrimers. The easiest way, and the most widely used, consists in using a bifunctional monomer to be grafted to all of the surface functions of the dendrimers. Two other strategies are known but are rarely used: the modification of an existing function, to generate two functions, and the sequential grafting of one function then of a second function. The three methods are illustrated in this review with polyphosphorhydrazone (PPH) dendrimers, together with their properties as catalysts, for materials, and as biological tools.

Abstract: International audience; Conventional small molecular chemical drugs always have challenging limitations in cancer therapy due to their high systemic toxicity and low therapeutic efficacy. Nanotechnology has been applied in drug delivery, bringing new promising potential to realize effective cancer treatment. In this context, we develop here a new nanomicellar drug delivery platform generated by amphiphilic phosphorus dendrons (1-C17G3.HCl), which could form micelles for effective encapsulation of a hydrophobic anticancer drug doxorubicin (DOX) with a high drug loading content (42.4%) and encapsulation efficiency (96.7%). Owing to the unique dendritic rigid structure and surface hydrophilic groups, large steady void space of micelles can be created for drug encapsulation. The created DOX-loaded micelles with a mean diameter of 26.3 nm have good colloidal stability. Strikingly, we show that the drug-free micelles possess good intrinsic anticancer activity and act collectively with DOX to take down breast cancer cells in vitro and the xenografted tumor model in vivo through upregulation of Bax, PTEN, and p53 proteins for enhanced cell apoptosis. Meanwhile, the resulting 1-C17G3.HCl@DOX micelles significantly abolish the toxicity relevant to the free drug. The findings of this study demonstrate a unique nanomicelle-based drug delivery system created with the self-assembling amphiphilic phosphorus dendrons that may be adapted for chemotherapy of different cancer types.

Abstract: International audience; Dendrimers are macromolecular hyperbranched nano-structures, which properties are mainly driven by the type of their terminal functions. This paper reports the synthesis of dendritic structures built from difunctional polyethylene glycols (PEGs) used as core. Three different PEG lengths have been used: PEG300 (ca 6 CH2CH2-O units), PEG1000 (ca 22 CH2CH2-O units), and PEG2000 (ca 45 CH2CH2-O units). The first level of branching units is based on a dialdehyde of a dipiperazine triazine derivative. The second and third level of branching units are constituted of phosphorhydrazone linkages. The growing of the dendritic branches was found extremely slow with both PEG1000 and PEG2000 used as core, and was stopped early, whereas this problem was not observed with PEG300 used as core. These PEG-cored dendritic structures were finally functionalized on the surface with two types of nitrogen ligands.

Abstract: International audience; The long-term treatment of tuberculosis (TB) sometimes leads to nonadherence to treatment, resulting in multidrug-resistant (MDR) and extensively drug-resistant (XDR) tuberculosis. Inadequate bioavailability of the drug is the main factor for therapeutic failure, which leads to the development of drug-resistant cases. Therefore, there is an urgent need to design and develop novel antimycobacterial agents minimizing the period of treatment and reducing the propagation of resistance at the same time. Here, we report the development of original and noncytotoxic polycationic phosphorus dendrimers essentially of generations 0 and 1, but also of generations 2–4, with pyrrolidinium, piperidinium, and related cyclic amino groups on the surface, as new antitubercular agents active per se, meaning with intrinsic activity. The strategy is based on the phenotypic screening of a newly designed phosphorus dendrimer library (generations 0–4) against three bacterial strains: attenuated Mycobacterium tuberculosis H37Ra, virulent M. tuberculosis H37Rv, and Mangora bovis BCG. The most potent polycationic phosphorus dendrimers 1G0,HCl and 2G0,HCl are active against all three strains with minimum inhibitory concentrations (MICs) between 3.12 and 25.0 μg/mL. Both are irregularly shaped nanoparticles with highly mobile branches presenting a radius of gyration of 7 Å, a diameter of maximal 25 Å, and a solvent-accessible surface area of dominantly positive potential energy with very localized negative patches arising from the central N3P3 core, which steadily interacts with water molecules. The most interesting is 2G0,HCl, showing relevant efficacy against single-drug-resistant (SDR) M. tuberculosis H37Rv, resistant to rifampicin, isoniaid, ethambutol, or streptomycin. Importantly, 2G0,HCl displayed significant in vivo efficacy based on bacterial counts in lungs of infected Balb/C mice at a dose of 50 mg/kg oral administration once a day for 2 weeks and superior efficacy in comparison to ethambutol and rifampicin. This series of polycationic phosphorus dendrimers represents first-in-class drugs to treat TB infection, could fulfill the clinical candidate pipe of this high burden of infectious disease, and play a part in addressing the continuous demand for new drugs.

Abstract: International audience; Various dendritic structures have been prepared during the last few decades and their properties have been emphasized in a number of reviews. Among these macromolecular structures, those incorporating phosphorus units – phosphorus dendrimers – or viologen linkages – viologen dendrimers – stand as the most promising in different fields spanning from chemistry, materials science to biology and nanomedicine. The association of the two complementary phosphorus and viologen structures resulted in a nascent family of structures referred to as “mixed” hybrid phosphorus–viologen dendrimers. In light of their specific properties, it is of interest to illustrate the conceptual preparation of not only a variety of sophisticated hybrid viologen–phosphorus dendrimers including classical dendrimers, but also other kinds of mixed macromolecules such as macromolecular asterisks, onion peel structures and covalent polydendrimer frameworks. Some preliminary results concerning the biological properties of these nano-objects as well as their use for the construction of organic–inorganic hybrid materials are presented.

Abstract: International audience; We designed and synthesized several families of novel amphiphilic fluorescent phosphorus dendron-based micelles showing relevant antiproliferative activities for use in the field of theranostic nanomedicine. Based on straightforward synthesis pathways, 12 amphiphilic phosphorus dendrons bearing 10 protonated cyclic amino groups (generation one), or 20 protonated amino groups (generation two), and 1 hydrophobic chain carrying 1 fluorophore moiety were created. The amphiphilic dendron micelles had the capacity to aggregate in solution using hydrophilic/hydrophobic interactions, which promoted the formation of polymeric micelles. These dendron-based micelles showed moderate to high antiproliferative activities against a panel of tumor cell lines. This paper presents for the first time the synthesis and our first investigations of new phosphorus dendron-based micelles for cancer therapy applications.

Abstract: International audience; Gene delivery, one important cancer-therapy mode, still remains to be challenging because of the shortage of highly efficient and safe nonviral vectors. Here, we revisit the development of cationic phosphorus dendrimers by synthesizing them with different generations (G1–3) and surface ligands (1-(2-aminoethyl) pyrrolidine, 1-(3-aminopropyl) piperidine, or 1-(2-aminoethyl) piperidine) for optimized gene delivery toward cancer-gene-therapy applications. First, the synthesized dendrimer derivatives were employed to condense plasmid DNA (pDNA) encoding enhanced green fluorescent protein (EGFP) to optimize their gene-delivery efficiency by varying the dendrimer generations and surface polycationic ligands. We show that all dendrimer/pDNA polyplexes display good cytocompatibility, and the 1-(2-aminoethyl) pyrrolidine-modified protonated G1 dendrimers (1-G1) display the best gene-delivery efficiency to HeLa cells under the same conditions through flow cytometry and fluorescence microscopic imaging analyses. Hence, 1-G1 dendrimers were then used as a vector to transfect pDNA encoding both EGFP and p53 protein for cancer-gene-therapy applications. Our results reveal that under the optimized conditions, the transfection of pDNA induces the significant p53 protein expression as verified through the resulted cell cycle arrest (regulation of p21 and Cdk4/Cyclin-D1 expression) and Western blotting. The cancer-gene-therapy potential of the polyplexes was finally validated through therapy of a xenografted tumor model after intratumoral injection without systemic toxicity. The developed cationic 1-G1 dendrimers may be adopted as a powerful vector system for gene therapy of cancer, as well as for highly effective gene therapy of other diseases.

Abstract: International audience; First-in-class CuII and AuIII metaled phosphorus dendrons were synthesized and showed significant antiproliferative activity against several aggressive breast cancer cell lines. The data suggest that the cytotoxicity increases with reducing length of the alkyl chains, whereas the replacement of CuII with AuIII considerably increases the antiproliferative activity of metaled phosphorus dendrons. Very interestingly, we found that the cell death pathway is related to the nature of the metal complexed by the plain dendrons. CuII metaled dendrons showed a potent caspase-independent cell death pathway, whereas AuIII metaled dendrons displayed a caspase-dependent apoptotic pathway. The complexation of plain dendrons with AuIII increased the cellular lethality versus dendrons with CuII and promoted the translocation of Bax into the mitochondria and the release of Cytochrome C (Cyto C).

Abstract: International audience; Attaining absolute control over a catalytic process and, therewith, exploiting its full potential pleases scientists in their ambitious and ongoing endeavor to perform catalysis like Nature does. In this regard, redox-switchable catalysis certainly holds great potential, constantly gaining importance in modern catalysis research. Herein, we report the application of P-stereogenic dendritic ferrocenyl phosphines in the ruthenium-catalyzed redox-switchable transfer hydrogenation of a ketone yielding an enantioenriched alcohol. By adding a chemical oxidant or reductant, the catalytic activity of the complexes was reversibly switched off and back on again over the course of the hydrogen transfer reaction. This has been rationalized mainly in terms of a distinct electronic communication between the redox-active group and the catalytic center. The highly functionalized dendritic catalysts presented here might impact the way prospective homogeneous catalysts will be designed.

Abstract: A new class of phosphorus dendritic compounds (PDCs) having a cyclotriphosphazene (P3N3) core and decorated with six &beta, cyclodextrin (&beta, CD) units, named P3N3-[O-C6H4-O-(CH2)n-&beta, CD]6, where n = 3 or 4 was designed, and the synthesis was performed using copper (I) catalyzed alkyne-azide cycloaddition (CuAAC). To obtain the complete substitution of the P3N3, two linkers consisting of an aromatic ring and an aliphatic chain of two different lengths were assessed. We found that, with both linkers, the total modification of the periphery was achieved. The two new obtained dendritic compounds presented a considerably high water solubility (&gt, 1 g/mL). The compounds comprised in this new class of PDCs are potential drug carrier candidates, since the conjugation of the &beta, CD units to the P3N3 core through the primary face will not only serve as surface cover but, also, provide them the faculty to encapsulate various drugs inside the &beta, CDs cavities.

Abstract: International audience; A small water-soluble phosphorus-containing dendrimer was engineered for the complexation of gold(I) and for its reduction under mild conditions. Gold nanoparticles were obtained as colloidal suspensions simply and only when the powdered form of this dendrimer was dissolved in water, as shown by transmission electron microscopy (TEM) and energy dispersive X-ray spectroscopy (EDX) analyses. The dendrimers acted simultaneously as mild reducers and as nanoreactors, favoring the self-assembly of gold atoms and promoting the growth and stabilization of isolated gold nanoparticles. Thus, an unprecedented method for the synthesis of colloidal suspensions of water-soluble gold nanoparticles was proposed in this work.

Abstract: Dendrimers are nanosized, arborescent polymers of which size and structure are perfectly controlled. This is one reason why they are widely used for biomedical purposes. Previously, we showed that a phosphorus-based dendrimer capped with anionic azabisphosphonate groups (so-called ABP dendrimer) has immuno-modulatory and anti-inflammatory properties towards human immune cells in vitro. Thereafter, we have shown that the ABP dendrimer has a promising therapeutic efficacy to treat models of chronic inflammatory disorders. On the way to clinical translation, the biodistribution and the safety of this drug-candidate has to be thoroughly assessed. In this article, we present preliminary non-clinical data regarding biodistribution, hematological safety, genotoxicity, maximal tolerated doses, and early cardiac safety of the ABP dendrimer. One of the genotoxicity assays reveals a potential mutagen effect of the item at a concentration above 200 &micro, M, i.e., up to 100 times the active dose in vitro on human immune cells. However, as the results obtained for all the other assays show that the ABP dendrimer has promising biodistribution and safety profiles, there is no red flag raised to hamper the regulatory pre-clinical development of the ABP dendrimer.

Abstract: International audience; Fluorescent derivatives of phosphorhydrazone dendrimers are reviewed. Diverse types of fluorophores have been used, such as pyrene, naphthol, anthracene, dansyl, diketone, phthalocyanine, maleimide, julolidine, rhodamine, fluorescein, or fluorene derivatives. The fluorescent groups can be located either as terminal groups on the surface, at the core, linked to the core (off-center), or to the branches of the dendritic structure. After fundamental research on their synthesis, these compounds have been used in the fields of catalysis, nanomaterials, OLEDs, sensors and biology/nanomedicine, in particular for monitoring transfection, or for their anti-inflammatory or anti-cancer properties.

Abstract: International audience; Phosphorus dendrimers (dendrimers having one phosphorus atom at each branching point) possess versatile properties, depending on the type of their terminal functions. This perspective article focusses on the synthesis and study of properties of phosphorus dendrimers functionalised on their surface by nitrogen ligands and their complexes. It is organized depending on the type of N-ligands (P,N ligands, N,N ligands, and N,N,N-ligands). Most properties concern catalysis, with palladium, copper, and scandium complexes. Recent advances concern anti-cancer properties, with dendrimers functionalised by copper and gold complexes of N,N-ligands.

Abstract: International audience; The synthesis and the photophysical properties of a new class of fully organic monodisperse nanoparticles for combined two-photon imaging and photodynamic therapy are described. The design of such nanoparticles is based on the covalent immobilization of a dedicated quadrupolar dye that combines excellent two-photon absorbing (2PA) properties, fluorescence and singlet oxygen generation ability, in a phosphorous-based dendrimeric architecture. First, a bifunctional quadrupolar dye bearing two different grafting moieties, a phenol function and an aldehyde function, was synthesized. It was then covalently grafted through its phenol function to a phosphorus-based dendrimer scaffold of generation 1. The remaining aldehyde functions were then used to continue the dendrimer synthesis up to generation 2, introducing finally 24 water-solubilizing triethyleneglycol chains at its periphery. A dendrimer confining 12 photoactive quadrupolar units in its inner scaffold and showing water solubility was thus obtained. Interestingly, the G1 and G2 dendrimers retain some fluorescence as well as significant singlet oxygen production efficiencies while they were found to show very high 2PA cross-sections in a broad range of the NIR biological spectral window. Hydrophilic dendrimer G2 was tested in vitro on breast cancer cells, first in one- and two-photon microscopy, which allowed for visualization of their cell internalization, then in two-photon photodynamic therapy. While being nontoxic in the dark and, more importantly, under exposure to daylight, dendrimer G2 proved to be a very efficient cell-death inducer only under two-photon irradiation in the NIR.

Despite the architectural similarity, organic and inorganic dendrimers differ in terms of methods of synthesis and characterization, and also by types of utilization. Inorganic dendrimers can offer completely new properties, as will be emphasized later in Chapter 10. The present chapter describes the synthesis of dendrimers comprising either silicon or phosphorus branching points with the emphasis on carbosilane and phosphorhydrazone dendrimers, both of which have obtained the largest attention of all inorganic dendrimers to date. Many other types of small dendrimers have been described in literature, but they will not be reported in this chapter, which focuses both on the above most widely used and also on the most original types of inorganic dendrimers. A few miscellaneous examples of dendrimers based on germanium, tin, or bismuth branching points will be given as well, although they are still rare among dendrimers.

Abstract: International audience; Different types of two-photon absorbing (TPA) fluorophores have been synthesized and specifically functionalized to be incorporated in the structure of phosphorus dendrimers (highly branched macromolecules). The TPA fluorophores were included in the periphery as terminal functions, in the core, or in the branches of the dendrimer structures, respectively. Also the functionalization in two compartments (core and surface, or branches and surface) was achieved. The consequences of the location of the fluorophores on the fluorescence and TPA properties have been studied. Several of these TPA fluorescent dendrimers have water-solubilizing functions as terminal groups, and fluorophores at the core or in the branches. They have been used as fluorescent tools in biology for different purposes, such as tracers for imaging blood vessels of living animals, for determining the phenotype of cells, for deciphering the mechanism of action of anticancer compounds, and for safer photodynamic therapy.

Abstract: The adsorption capacities of nano-sized organoclays composed of a stevensite-rich clay (R), phosphorus dendrimers (GC1 and GC2) and Na+-saturated clay were evaluated for their capacity to adsorb chromate and methylene blue (MB) in the range of 298–318 K. The adsorption kinetics and the isotherms were analysed based on kinetic equations and isotherm models and by adopting a non-linear regression procedure. In addition, the organoclays and the Na+-saturated clays were characterized principally by solid-state nuclear magnetic resonance spectroscopy. The pseudo-second-order rate equation described kinetics data well, and the adsorption rates were not limited by the intraparticle diffusion or by the liquid film diffusion. Both chemical species were adsorbed spontaneously (–31 <ΔG°T<–10 kJ/mol), but the adsorbents had a high affinity for MB species. The adsorption isotherms of chromate were fitted better by the Freundlich model, while those of MB followed the Langmuir model. Chromate adsorption took place at the edges and the free surfaces of stevensite, particularly at the protonated aluminols. MB was adsorbed as MBH2+and MB+. The MB protonation occurred at the clay surfaces, and MB+ions were located at the planar surfaces of stevensite as well as at the external surfaces of aggregates. Moreover, the tetrahedral sheet of stevensite involved in the formation of GC1-based organoclays was the subject of a partial chemical modification.

Abstract: A new phosphorus-containing dendrimer of the first-generation with potential pharmacological activity was synthesized and studied by spectral methods. The FTIR, FT Raman, 1 H and 31 P NMR spectra of the first generation dendrimer G 1 with a cyclotriphosphazene core, six branches O C 6 H 4 CH N N(CH 3 ) P(S) O C 6 H 4 (CH 2 ) 2 NH CO C 5 NH 4 were recorded. Amide groups of the dendrimer participate in the formation of an intermolecular hydrogen bond. Structure, geometric parameters, the frequency and intensity of the bands in the vibrational spectra were calculated using DFT with PBE functional and TZ2P basis set. Spectral characteristics, charge distribution and reactivity of the core, repeating units and terminal groups of the dendrimer were determined. The first-generation dendrimer molecule has the shape of a concave lens with a slightly non-planar cyclotriphosphazene core and flat repeating units. Repeating units are arranged symmetrically on three on each side of the core, there are no steric hindrances in it and the end groups are able to enter into subsequent reactions and dendrimer has a sufficiently large cavity for accommodating guest molecules. The HOMO covers the repeating units with a noticeable conjugation and the LUMO belongs to the terminal groups.

Abstract: International audience; Nanotechnology has the high capability to revolutionize many other technologies in many fields, due to the development of myriad nanodevices. Thus nanoparticles have been extensively studied over the last decade. Among all these, dendrimers pertain to the “nano-world” by virtue of their size (a few nanometers range). However, they are intrinsically different from classical ‘hard’ metallic nanoparticles because they are constituted exclusively of ‘soft’ organic matter (organic polymers). Their nanometric size effects can be observed and are reminiscent to those of the multivalent systems widely known in Nature. In this review we will report ways of incorporation of Au(I) or Au(III) selectively at the level of the core, within the cascade structure, regioselectively at a given generation, or on the outer shell of various phosphorus dendrimers. This can be achieved via the skeleton modification of these phosphorus dendrimers as well as with the selective incorporation of ligands as phosphine, thiol, iminophosphorane, thiophosphine, iminopyridine groups allowing complexation via formation of P–Au, S–Au, P=N–P=S–Au, or N–Au–N bonds. Original phosphorus dendritic structures as macromolecular asterisks, or onion peel dendritic structures allow to diversify the ways of obtaining either gold complexes or gold nanoparticles, some of them being used as chemical sensors. Finally, selected examples of applications of these gold nano-objects in nanomedicine for neuron activation or unprecedented anti-tumoral properties will be presented.

Abstract: International audience; Tuberculosis (TB) is a contagious infection that usually attacks not only the lungs, but also brain and spine. More than twenty drugs have been developed for the treatment of TB, but most of them were developed some years ago. They are used in different combinations. Isoniazid and Rifampicin are examples of the five first line TB drugs, whereas, for instance, Levofloxacin, Kanamycin and Linezolid belong to the second line drugs that are used for the treatment of drug resistant TB. Several new bicyclic nitroimidazoles (e.g., Delamanid) without mutagenic effects were developed. New TB drugs need to provide several main issues such as more effective, less toxic, and less expensive for drug resistant TB. Besides polymeric, metal-based nanoparticles, polymeric micelles and polymers, dendrimer nanostructures represent ideal delivery vehicles and offer high hopes for the future of nanomedicine. In this original review, we present and analyze the development of anti-TB drugs in combination with dendrimers. Few articles have highlighted the encapsulation of anti-TB drugs with dendrimers. Due to their unique structure, dendrimers represent attractive candidates for the encapsulation and conjugation of other anti-TB drugs presenting important drawbacks (e.g., solubility, toxicity, low bioavailability) that hinder their development, including clinic trials.

Abstract: International audience; Three generations of phosphorus dendrimers with pyrrolidine end groups were synthesized for use in biology and medicine. Neutral samples are soluble in organic solvents but after protonation these dendrimers become water-soluble and can be used for biological experiments. The FTIR and FT Raman spectra of three generations of dendrimers Gi constructed from the cyclotriphosphazene core, repeating units OC6H4CHNN(CH3)P(S)< and aminoethylpyrrolidine end groups NH(CH2)2C4NH9 were registered. Analysis of the IR spectra of Gi shows that the NH groups form a hydrogen bond with chlorine atoms. The calculation of the vibrational spectra was performed for the optimal structure of the first generation dendrimer in the framework of the DFT method. The first-generation dendrimer molecule has flat, repeating units and a plane of symmetry passing through the cyclotriphosphazene core. Calculation of the distribution of the potential energy made it possible to assign the bands in the experimental vibrational spectra. Amine groups show NH stretching bands at 3397 and 3200cm−1, NH+ stretching bands occur at 2645 and 2546cm−1 in the IR spectrum of G1. The stretching vibrations of the NH+ group are significantly shifted to low frequencies due to the formation of a hydrogen bond with the chlorine atom. The line at 1577cm−1 in the Raman spectrum is characteristic of repeating units.

Abstract: International audience; Difluoroboron complexes of monomeric and dimeric diketones and of generations 0-4 of phosphorus dendrimers ended by diketone ligands are synthesized and characterized. Their photoluminescence properties are measured. All compounds exhibit an intense absorption band in the near UV region. Both model dimers and dendrimers show a marked hypsochromic shift of this absorption band compared with the monomeric difluoroboron complex. The fluorescence of the dioxaborine complex subunit in the multichromophoric dendritic architectures is quenched compared with the emission of the isolated monomeric fluorophore, presumably due to interactions between the terminal groups of dendrimers.

Abstract: International audience; Phosphorus-containing dendrimers are attractive carriers for the delivery of bioactive molecules due to their very particular architecture, globular shape and chemical and physical properties. Herein we report a simple synthetic approach of ethacrynic acid derivatives loaded-phosphorus dendrimers by symmetrical grafting of ethacrynic acid analogues by nucleophilic substitution of each chlorine of P(S)Cl2 end groups. We report also for the first time an unsymmetrical grafting of two different phenol derivatives by a selective substitution of one chlorine of P(S)Cl2 end group followed by the grafting of the second phenol.

Abstract: International audience; A multivalent phosphorus dendrimer 1G3 and its corresponding Cu-complex, 1G3-Cu have been recently identified as agents retaining high antiproliferative potency. This antiproliferative capacity was preserved in cell lines overexpressing the efflux pump ABC B1, whereas cross-resistance was observed in ovarian cancer cell lines resistant to cisplatin. Theoretical 3D models were constructed: the dendrimers appear as irregularly shaped disk-like nano-objects of about 22 Å thickness and 49 Å diameter, which accumulated in cells after penetration by endocytosis. To get insight in their mode of action, cell death pathways have been examined in human cancer cell lines: early apoptosis was followed by secondary necrosis after multivalent phosphorus dendrimers exposure. The multivalent plain phosphorus dendrimer 1G3 moderately activated caspase-3 activity, in contrast with the multivalent Cu-conjugated phosphorus dendrimer 1G3-Cu which strikingly reduced the caspase-3 content and activity. This decrease of caspase activity is not related to the presence of copper, since inorganic copper has no or little effect on caspase-3. Conversely the potent apoptosis activation could be related to a noticeable translocation of Bax to the mitochondria, resulting in the release of AIF into the cytosol, its translocation to the nucleus and a severe DNA fragmentation, without alteration of the cell cycle. The multivalent Cu-conjugated phosphorus dendrimer is more efficient than its non-complexed analog to activate this pathway in close relationship with the higher antiproliferative potency. Therefore, this multivalent Cu-conjugated phosphorus dendrimer 1G3-Cu can be considered as a new and promising first-in-class antiproliferative agent with a distinctive mode of action, inducing apoptosis tumor cell death through Bax activation pathway.

Abstract: The FTIR and FT Raman spectra of the 11 generations of poly(phosphorhydrazone) dendrimers built from thiophosphoryl core with P−Cl ( G i ) and oxybenzaldehyde ( G' i ) terminal groups have been studied. Spectral pattern of dendrimers depends on the ratio of the number of end groups and repeating units. This ratio tends to r– 1 (r– functionality of the repeating unit) and becomes constant when the generation number exceeds 3. The experimental IR and Raman spectra of the different generations of dendrimers are very similar to each other in the agreement with the theory. Optimization of the structure and the analysis of normal vibrations was made for G' 0 and G 1 using the density functional theory (DFT). Calculations showed that the molecules of the first generation dendrimer have the form of a propeller with three blades and flat repeating units. Bond lengths and angles in the optimized geometry obtained by DFT show good agreement with experiment. Consideration of the molecular orbitals shows that in the repeating units and oxybenzaldehyde end groups of poly(phosphorhydrazone) dendrimers conjugation is realized. The oxybenzaldehyde terminal groups are characterized by a band 1702 cm −1 in the experimental IR spectra of G ’ i . The line in the Raman spectra of G i and G ’ i at 1576 cm −1 refers to the vibrations of the repeating units.

Analysis of the IR spectra of G 1 shows that the amide groups form an intermolecular hydrogen bond. Optimization of the structure and analysis of normal vibrations were carried out for dendrimer G 1 using the density functional theory (DFT). The six repeating units are arranged in a symmetric manner about the cyclophosphazene ring; each side of the ring contains three repeating units. It turned out that dendrimer G 1 is a double bowl with almost flat repeating units which represent the concave surface and a core as a bottom of the bowl. The assignment of bands in the vibrational spectra was carried out by analyzing the distribution of the potential energy. Amide groups show bands at 3393, 1675 cm −1 in the IR spectrum of G 1 . The line at 1577 cm −1 in the Raman spectrum is characteristic of repeating units.

Abstract: International audience; From biomaterials to imaging, and from drug delivery to drugs by themselves, phosphorus-containing dendrimers offer a large palette of biological properties, depending essentially on their types of terminal functions. The most salient examples of phosphorus dendrimers used for the elaboration of bio-chips and of supports for cell cultures, for imaging biological events, and for carrying and delivering drugs or biomacromolecules are presented in this feature article. Several phosphorus dendrimers can be considered also as drugs per se (by themselves) in particular to fight against cancers, neurodegenerative diseases, and inflammation, both in vitro and in vivo. Toxicity assays are also reported.

Abstract: International audience; Human natural killer (NK) cells play a key role in anti-cancer and anti-viral immunity, but their selective amplification in vitro is extremely tedious to achieve and remains one of the most challenging problems to solve for efficient NK cell-based immuno-therapeutic treatments against malignant diseases. Here we report that, when added to ex vivo culture of peripheral blood mononuclear cells from healthy volunteers or from cancer patients with multiple myeloma, poly (phosphorhydrazone) dendrimers capped with amino-bis(methylene phosphonate) end groups enable the efficient proliferation of NK cells with anti-cancer cytotoxicity in vivo. We also show that the amplification of the NK population relies on the preliminary activation of monocytes in the framework of a multistep cross-talk between monocytes and NK cells before the proliferation thereof. Thus poly(phosphorhydrazone) dendrimers represent a novel class of extremely promising drugs to develop NK-cell based anti-cancer therapies.

As remarkable properties, this dendritic PVDF displayed crystallinity (HRTEM highlighted crystalline disc-like zones of ca. 5 nm) and a much higher hydrophobicity than both its precursors with the water contact angle (WCA) reaching 108°.

Abstract: International audience; Dendrimers are hyperbranched macromolecules having a perfectly defined and multifunctionalized structure, constituted of branches emanating radially from a central core. The structure of dendrimers is particularly modular, and can incorporate in different parts coordination complexes. In this review, we will present the interplay between dendrimers and coordination chemistry in three main fields: catalysis, materials, and biology. Most of the examples will be taken from the work done with phosphorus-containing dendrimers, but the pioneering work carried out with other dendrimers will be also presented. One of the major improvements that metallodendrimers have afforded concerning catalysis is their easy recovery and reuse, bridging the gap between homogeneous and heterogeneous catalysis. Another major improvement concerns the “dendritic effect” which can afford impressive outcomes concerning the increase of yield and of enantioselectivity, together with a decrease of the leaching of metals, and consequently of waste. Dendrimers can be used also for the synthesis and stabilization of metallic nanoparticles, for the modification of metallic surfaces at the nanometric scale, and for the synthesis of mesoscopically ordered hybrid materials. Finally metallodendrimers have high potency against cancerous cell lines, and they appear to operate via a different mechanism of action compared with native metallodrugs, opening new avenues for the search of improved anti-cancer agents.

Abstract: International audience; The well-known diuretic Ethacrynic acid (EA, Edecrin), showing low anti-proliferative activities, was chemically modified at different positions. The new EA derivatives have been tested in vitro in anti-proliferative assays on both tumor KB (epidermal carcinoma) and leukemia HL60 (promyelocytic) cells suitable targets for anticancer activity. Reduction of the α-β double bond of EA completely abolished anti-cancer activities, whereas introduction of either 2-(4-substituted phenyl)ethanamine (series A) or 4-(4-substituted phenyl)piperazine (series B) moieties generated compounds showing moderate to strong anti-proliferative activities against human cancer cell lines. Several substitutions on the phenyl of these two moieties are tolerated. The mechanism of action of the EA derivatives prepared in this study is more complex than the inhibition of glutathione S-transferase π ascribed as unique effect to EA and might help to overcome tumor resistances.

Abstract: International audience; A new concept is presented, namely the synthesis of dendrimers intrinsically composed in alternation of building blocks pertaining to two known families of dendrimers: phosphorhydrazone dendrimers and triazine–piperazine dendrimers. These mixed dendrimers with layered controlled architecture inherit their easy 31P NMR characterization and their thermal stability from the phosphorhydrazone family, and their decreased solubility from the triazine–piperazine family. However, they have also their own and original characteristics. Both parent families are white powders, whereas the mixed dendrimers are yellow, orange, or red powders, depending on the generation. DFT calculations were carried out on model dendrons to understand these special color features. Remarkably, these dendrimers incorporating redox-active organic entities allow for the first time the monitoring of the growth of an organic dendrimer by electrochemistry while highlighting an even–odd generation behavior.

Abstract: International audience; Nanocomposites of phosphorus-based dendrimers and montmorillonite were prepared and examined using X-ray diffraction, thermal analysis, solid-state nuclear magnetic resonance, and high-resolution transmission electron microscope. The adsorption of chromate by the nanocomposites and the basic clay was studied in the temperature range 298-318 K. For the latter purpose, the kinetics was followed and adsorption isotherms were plotted. The results showed that intercalated nanocomposites (GC1-AT) were formed with the first generation of cationic dendrimers. The use of the second generation of cationic dendrimers resulted in the formation of a mixture of exfoliated and intercalated nanocomposites (GC2-AT). For both dendrimers, adsorption was by cation exchange. The kinetics of the adsorption of chromate on both nanocomposites and Na-saturated montmorillonite (Na-AT) followed the pseudo-second-order equation and the rate constants varied in 0.08-0.19 g/mmol s-1. The rate-limiting steps were discussed on the basis of the results of the external mass transfer and the internal diffusion models. Abstract: International audience; The influence of surface functions on the interactions between Poly(PhosphorHydrazone) PPH dendrimers and human monocytes is discussed on the basis of complementary biological and physicochemical studies on membrane models (monolayers and multi-lamellar vesicles). The studies were performed on both an active and non-toxic phosphonic acid capped dendrimer and a non-active but toxic carboxylic acid capped one. On the one hand, comparative studies of the behaviour of DPPC monolayers in the presence or absence of PPH dendrimers in the subphase showed differences in the phase transitions, highlighting interactions between both dendrimers and phospholipid monolayers, with a larger incidence for the carboxylic acid capped dendrimer (negative control), validating its cellular toxicity. On the other hand, comparative biological studies (activation of human monocytes and binding of fluorescent dendrimers on human monocytes) show the pre-eminence of phosphonic acid capped dendrimers towards specific binding and subsequent activation of human monocytes.

Abstract: International audience; Ruthenium nanoparticles (NPs) are prepared by milling under air ruthenium chloride (RuCl3), sodium borohydride (NaBH4) and a polyphosphorhydrazone (PPH) dendron (generation 0 to 2) having an alkyl chain at the focal point and triarylphosphines on the surface. The resulting NPs have a diameter in the 2 to 3 nm range and they are stable upon storage in solution or as powders. They can efficiently catalyze hydrogenation of styrene. The interaction between the dendrons and the NPs is studied, and the influence of the alkyl chain length and dendron generation is also discussed.
