Objective Catechol-O-methyltransferase (COMT), the product of the COMT gene, detoxifies the carcinogenic catechol estrogens. The aim of the present study was to examine the relationship between COMT Val158Met polymorphism and the risk of ovarian cancer. Material and methods The study groups consist of 94 individuals as a patients group with ovarian cancer (n=47) and control group (n=47). The allele and genotype frequencies were determined according to Hardy-Weinberg equilibrium (HWE). The allele and genotype frequencies. determined according to HWE. Genetic analysis were performed by real-time-polymerase chain reaction instrument, and the statistical analysis were performed by SPSS program. Results Although no significant relationship was obtained among groups (p=0.413) regarding COMT gene Val158Met polymorphism, the genotype frequencies for COMT Val158Met (rs4860) polymorphism in groups was homozygote wild type GG genotype 25.5%, heterozygote GA genotype 46.8%, homozygote mutant AA genotype 27.7%. Conclusion This study is the first to investigate the relationship between ovarian cancer and the Val158Met polymorphism in the COMT gene in a Turkish population. No statistically significant relationship was identified among genotypes belonging to the patient and control groups although sample sizes were relatively small and the analysis should be repeated in a larger cohort.

Objective: The aim of this research was to determine the association between the fok1 polymorphism and uterine leiomyomas.Material and Methods: For genotyping the fok1 polymorphism of the vitamin D receptor, real-time polymerase chain reaction was performedon blood samples of uterine leiomyoma (n27) and control (n33) groups. For statistical analyses, SPSS v.23 software (SPSS Inc., Chicago, IL,USA) was used.Results: A statistically significant difference was observed for the frequency of the CC genotype between the uterine leiomyoma and controlgroups, and the frequencies of the T allele in the uterine leiomyoma groups were significantly higher than in the control group.Conclusion: The presence of the fok1 CC genotype may be a risk-reducing factor and the T allele may be a potential risk factor for developinguterine leiomyoma. Objective: The aim of this study is investigate the role of the Twist homolog 1 (TWIST), serine peptidase inhibitor (SERPINB5), and plasminogen activator inhibitor 1 (SERPIN1) genes in uterine leiomyoma etiopathogenesis. Material and Methods: Twelve patients, aged between 39 and 58, and had a hysterectomy, were included in the study. The size of the leiomyomas was between 20 and 130 mm based on gross pathology after hysterectomy. Tissue samples were obtained from normal myometrium and leiomyoma (1 cm³) tissue of the uterus of the patients and stored at -86°C. Samples were divided to two groups after histopathological evaluation of the uterus: normal myometrial tissues as control group (Group 1) and leiomyoma tissue as the study group (Group 2). We aimed to determine the effects of melatonin treatment on endometrial implants in an oopherectomized rat endometriosis model.This study is a prospective, randomised, controlled experimental study. It was carried out at the Experimental Research Center of Yeditepe University (YUDETAM). Twenty-two, female, non-pregnant, nulligravid Spraque-Dawley albino rats were included in our study. Endometriosis was surgically induced in oopherectomized rats. Rats were randomised into two groups: control group and melatonin group. In the melatonin group, rats were treated with melatonin (20 mg/kg/day) for two weeks. After the operations were performed to assess the regression of the endometriotic lesions, melatonin treatment was stopped. At the end of the sixth week necropsies were performed to assess the rate of recurrence. The volume and histopathological scores of endometriotic foci were examined.Volumes of the endometriotic lesions significantly decreased in the melatonin group. Also, when the melatonin group was analysed within itself, endometriotic lesion volumes decreased and histopathological scores increased significantly.Melatonin causes regression of the endometriotic lesions in rats and improvement in their histopathological scores.Ooferektomize sıçan endometriozis modelinde melatoninin endometriotik odaklar üzerine etkisini incelemeyi amaçladık.Bu çalışma prospektif, randomize, kontrollü, deneysel bir çalışmadır. Çalışma Yeditepe Üniversitesi Deneysel Araştırma Merkezinde (YÜDETAM) yapıldı. Deneysel çalışmamıza 22 adet dişi, gebe olmayan, nulligravid Spraque-Dawley albino sıçan dahil edildi. Ooferektomize edilen sıçanlarda cerrahi olarak endometriozis oluşturuldu. Sıçanlar iki gruba ayrıldı: kontrol grubu ve melatonin grubu. Melatonin grubunda 2 hafta boyunca sıçanlara melatonin (20 mg/kg/gün) tedavisi verildi. Endometriotik regresyonu değerlendirmek için operasyonlar yapıldıktan sonra melatonin tedavisi sonlandırıldı. Rekürrens oranlarını değerlendirmek için altıncı haftanın sonunda nekropsiler yapıldı. Endometriotik odakların hacim ve histopatolojik skorları değerlendirildi.Endometriotik lezyonların hacimleri melatonin grubunda anlamlı derecede azaldı. Ayrıca, melatonin grubu kendi içerisinde incelendiğinde endometriotik lezyonların hacimlerinin anlamlı derecede azaldı ve histopatolojik skorları anlamlı derecede arttı.Melatonin sıçanlarda endometriotik odakların gerilemesine ve histopatolojik skorların iyileşmesine neden olur.

It can be assumed a high incidence of chromosomal abnormality among the patients who underwent ICSI cycle after searching the current literature. The huge amount of these abnormalities consist of low grade mosaicism. Besides adequate follicular response, the rate of fertilization failure is high in these group. If pregnancy achieved, spontan abortion usually occurs. Oocyte donation can be most appropriate treatment choise in some selected patients.

To determine the effects of the combination of letrozole and melatonin on surgically induced endometriosis.This prospective, randomized, controlled, experimental study was carried out at Yeditepe University Experimental Research Center (YUDETAM). Female non-pregnant, 17 nulligravid Wistar - Hannover albino rats with surgically induced endometriosis were used in this study. Endometriosis was induced by using homologous uterine horn transplantation in the rats. Four operations were performed on each rat. The induction of endometriosis was performed in the first operation. After two weeks of estradiol treatment the second operation was performed and endometriotic lesions were evaluated. Estrogen was then discontinued and in the study groups medications were started. During two weeks the rats were given medications and the third operation was performed for the assessment of the effects of the medications on the endometriotic foci. Then all the medications were stopped and estrogen was started again. Two weeks later all the rats were euthanized and recurrence of endometriosis was evaluated.The sum of the lesion volumes in the control group was 93.6±31.7 mm(3) at the end of the second week. After the cessation of estradiol it decreased to 85.0±23.8 mm(3) (P=0.31) and increased to 119.7±29.4 mm(3) at the sixth week (P=0.02). A significant reduction in histopathologic scores were seen after cessation of the estradiol (p=0.04). At the end of the sixth week, histopathological scores reached the pretreatment values. In the letrozole and melatonin group the sum of the lesion volumes decreased significantly after the treatment (82.8±21.0 mm(3) and 15.7±8.0 mm(3) respectively). At the end of the sixth week, the mean volume was calculated as 43.9±31.8 mm(3) (p=0.002). Histopathologic scores were 2.3±0.1, 2.0±0.2 and 2.2±0.3 at the end of the second, fourth and sixth weeks, respectively, in the letrozole and melatonin group.Letrozole and melatonin caused a significant regression in lesion volumes; however, histopathological scores of endometriotic lesions did not change significantly.Kombine letrozol ve melatonin tedavisinin cerrahi olarak indüklenmiş endometriosis üzerindeki etkisini belirlemek.Bu prospektif randomize kontrollü bir deneysel çalışma olarak dizayn edildi ve Yeditepe Üniversitesi Deneysel Hayvanlar Araştırma Merkezinde (YUDETAM) gerçekleştirildi. Cerrahi olarak endometriosis oluşturulmuş 17 tane nulligravida dişi Wistar – Hannover albino rat kullanıldı. Ratlara homolog uterin boynuz inokülasyonu yapılarak endometriosis oluşturuldu ve iki hafta aralar ile dört operasyon yapıldı. Endometriosis indüksiyonu ilk operasyonda yapıldı. İki haftalık östradiol tedavisini takiben ikinci operasyon yapıldı. Sonra östrojen kesildi ve çalışma grubuna ilaç başlandı. Takip eden iki haftanın sonunda üçüncü operasyon yapılarak endometriyal odaklar üzerinde ilaçların etkinliği araştırıldı. Üçüncü operasyondan sonra bütün ilaçlar kesildi ve östrojen tekrar başlandı. Iki haftalık östrojen tedavisinin ardından tüm ratlara ötanazi yapıldı ve rekürrens oranlarına bakıldı.Kontrol grubunda tedavinin başında lezyonların volüm ortalaması 93.6±31.7 mmÇalışmamızda Letrozol ve Melatonin endometriotik lezyonların boyutlarında gerileme oratya çıkarırken, histopatolojik skorlarda değişikliğe yol açmadı.
