ObjectivesThis study aimed to evaluate the agreement between explosive detection dog (EDD) handlers and a team of veterinarians in assessing body condition score (BCS) and muscle condition score (MCS), hypothesizing significant BCS differences between handlers and veterinarians, and no significant MCS differences in healthy active duty EDDs.MethodsThis prospective study analyzed variance and inter-rater intraclass correlation coefficients (ICC) of agreement within BCS and MCS assessments collected from the 43 EDDs by four blinded graders; the EDDs’ respective handler and three veterinarians with varying levels of veterinary expertise.ResultsThe results of the study showed that 74.4% of the EDD population was graded as ideal BCS (4 or 5 out of 9) by the handlers compared to 67.44% by the members of the veterinary team; however, the graders scored different subsets of individual EDDs as ideal. Normal MCS (3 out of 3) was assessed in 86.05% (n = 37) of EDDs by the handlers versus in 70.54% by the veterinary team.ConclusionThis study highlights the importance of standardized training and guidelines for BCS and MCS assessments in working dogs to improve agreement between all members of the healthcare team.

IntroductionCurrent high demand for effective odor detection dogs calls for the development of reliable methods for measuring performance-related behavioral phenotypes in these highly specialized working animals. The Canine Behavioral Assessment & Research Questionnaire (C-BARQ) is a widely used behavioral assessment tool among working dog organizations with a demonstrated ability to predict success/failure of dogs in training. However, this instrument was developed originally to study the prevalence of behavior problems in the pet dog population, and it therefore lacks the capacity to measure specific behavioral propensities that may also be important predictors of working dog success. The current paper examines the factor structure, internal reliability, and content validity of a modified version of the C-BARQ designed to evaluate four new domains of canine behavior in addition to those encompassed by the original C-BARQ. These domains, labeled Playfulness, Impulsivity, Distractibility, and Basophobia (fear of falling), respectively, describe aspects of canine behavior or temperament which are believed to contribute substantially to working dog performance.MethodsExploratory factor analysis (EFA) of owner/handler questionnaire responses based on a sample of 1,117 working odor detection dogs.ResultsA total of 15 factors were extracted by EFA, 10 of which correspond to original C-BARQ factors. The remaining 5 comprise the four new domains– Playfulness, Impulsivity, Distractibility, and Basophobia– as well as a fifth new factor labeled Food focus.DiscussionThe resulting Working Dog Canine Behavioral Assessment & Research Questionnaire (WDC-BARQ) successfully expands the measurement capacities of the original C-BARQ to include dimensions of behavior/temperament of particular relevance to many working dog populations.

IntroductionThe study investigated the utilization of odor detection dogs to identify the odor profile of Staphylococcus aureus (S. aureus) biofilms in pure in vitro samples and in in vivo biosamples from animals and humans with S. aureus periprosthetic joint infection (PJI). Biofilms form when bacterial communities aggregate on orthopedic implants leading to recalcitrant infections that are difficult to treat. Identifying PJI biofilm infections is challenging, and traditional microbiological cultures may yield negative results even in the presence of clinical signs.MethodsDogs were trained on pure in vitro S. aureus biofilms and tested on lacrimal fluid samples from an in vivo animal model (rabbits) and human patients with confirmed S. aureus PJI.ResultsThe results demonstrated that dogs achieved a high degree of sensitivity and specificity in detecting the odor profile associated with S. aureus biofilms in rabbit samples. Preliminary results suggest that dogs can recognize S. aureus volatile organic compounds (VOCs) in human lacrimal fluid samples.DiscussionTraining odor detection dogs on in vitro S. aureus, may provide an alternative to obtaining clinical samples for training and mitigates biosecurity hazards. The findings hold promise for culture-independent diagnostics, enabling early disease detection, and improved antimicrobial stewardship. In conclusion, this research demonstrates that dogs trained on in vitro S. aureus samples can identify the consistent VOC profile of PJI S. aureus biofilm infections. The study opens avenues for further investigations into a retained VOC profile of S. aureus biofilm infection. These advancements could revolutionize infectious disease diagnosis and treatment, leading to better patient outcomes and addressing the global challenge of antimicrobial resistance.

Multisite phosphorylation of guanylyl cyclase (GC)-A, also known as NPR-A or NPR1, is required for receptor activation by natriuretic peptides (NPs) because alanine substitutions for the first four GC-A phosphorylation sites produce an enzyme that cannot be stimulated by NPs. In contrast, single Glu substitutions for the first six chemically identified GC-A phosphorylation sites to mimic the negative charge of phosphate produced an enzyme that is activated by NPs but had an elevated Michaelis constant (Km), resulting in low activity. Here, we show that vicinal (double adjacent) Glu substitutions for the same sites to mimic the two negative charges of phosphate produced a near wild type (WT) enzyme with a low Km. Unlike the enzyme with single glutamate substitutions, the vicinally substituted enzyme did not require the functionally identified Ser-473-Glu substitution to achieve WT-like activity. Importantly, the negative charge associated with either phosphorylation or glutamate substitutions was required for allosteric activation of GC-A by ATP. We conclude that vicinal Glu substitutions are better phosphomimetics than single Glu substitutions and that phosphorylation is required for allosteric activation of GC-A in the absence and presence of NP. Finally, we suggest that the putative functionally identified phosphorylation sites, Ser-473 in GC-A and Ser-489 in GC-B, are not phosphorylation sites at all.

Similar to other organizations that encourage positive youth development, the American Kennel Club (AKC) created the Junior Showmanship program to develop skills and pave a path for the next generation of competitors in canine conformation. Although participants age out of the Junior Showmanship Program when they turn eighteen, the hope is that Juniors will continue to be active in dog sports into adulthood. Females are overly represented in all age groups in this survey and in current participation. Although both males and females most commonly stopped participating at age 17 or 18, males had a significantly higher drop out at age 15. Further study is warranted to investigate factors specifically pertaining to male participation. A strong Belief System Model which accounted for positive responses associated with camaraderie and mentoring during the showmanship program was significantly (p=0.01) associated with commitment, member interest and active participation as an adult. In addition, success in the show ring was associated with future involvement in conformation or dog sports. Thus, both perceived achievement and mentorship by adult “volunteers” were associated with continued participation.

The bacterium Bordetella bronchiseptica is responsible for serious respiratory disease in dogs, most often associated with ‘kennel cough’ (canine infectious tracheobronchitis). It is recommended that dogs are vaccinated against the bacterium every 6–12 months, either by oral or intranasal administration. Any impairment of dogs' olfactory capabilities due to medical treatments may impact their efficiency and accuracy in their jobs. This study examined (1) the effect of intranasal and oral vaccines on the olfactory capabilities of detection dogs; as well as (1) effects of the vaccines on canine behavior. Dogs that were vaccinated initially with the oral and 28 days later with intranasal B. bronchiseptica were generally slower to find the target odor than the dogs that were assigned intranasal then oral vaccine. This result prompted a second between-subjects study to further investigate any impact of intranasal administration of the B. bronchiseptica vaccine on the olfactory capabilities of dogs. The intranasal vaccine was of particular interest due to its prevalent use and potential for nasal inflammation leading to decreased olfactory capabilities. Neither odor threshold nor time spent searching for odor were affected by the intranasal vaccine. Behavioral analyses showed that behaviors associated with the dogs' positive and negative motivation affected their time spent finding the target odor; this suggests that behavior should be considered in future studies of olfactory performance.

ObjectiveSeptic peritonitis is associated with significant morbidity and mortality. As a potential therapeutic agent in the treatment of sepsis, 2-O, 3-O desulfated heparin (ODSH) reduces histones and platelet factor 4 (PF4) in mouse sepsis models. This pilot clinical trial evaluated the safety and effect of ODSH in client-owned dogs with septic peritonitis.InterventionsIn an IACUC-approved, open-label, prospective, dose-escalation clinical trial in 6 dogs with spontaneous septic peritonitis, ODSH administration was initiated following surgical explore to achieve source control. Acute patient physiology and laboratory evaluation (APPLEfast and APPLEfull) scores on admission, source of septic peritonitis, requirement for vasopressors, the administration of blood products, and survival to discharge were recorded. Platelet count, cell free DNA (cfDNA) concentration, and platelet factor 4 (PF4) concentrations were measured at the time of each ODSH dosage. A dose of ODSH was administered every 8 hs for a total of 4 doses (maximum total dosage 75 mg/kg) based on a pre-determined escalation protocol. Patients were monitored in the ICU following administration for evidence of clinical hemorrhage.Main ResultsThe mean APPLEfast and APPLEfull scores on admission were 22 +/- 6 and 32 +/-10, respectively. Four dogs received 4 total dosages of ODSH and 2 dogs received 3 total dosages of ODSH intravenously. The mean total dosage of ODSH administered during the study period was 48.3 +/- 21.6 mg/kg. No dog required dose de-escalation or had any evidence of bleeding. Four dogs survived to discharge.ConclusionsNo adverse effects of ODSH administration were documented in dogs with septic peritonitis. A randomized controlled trial is necessary to evaluate ODSH as a novel therapeutic in the treatment of septic peritonitis.

Dogs perform a variety of integral roles in our society, engaging in work ranging from assistance (e.g., service dogs, guide dogs) and therapy to detection (e.g., search-and-rescue dogs, explosive detection dogs) and protection (e.g., military and law enforcement dogs). However, success in these roles, which requires dogs to meet challenging behavioral criteria and to undergo extensive training, is far from guaranteed. Therefore, enhancing the selection process is critical for the effectiveness and efficiency of working dog programs and has the potential to optimize how resources are invested in these programs, increase the number of available working dogs, and improve working dog welfare. In this paper, we review two main approaches for achieving this goal: (1) developing selection tests and criteria that can efficiently and effectively identify ideal candidates from the overall pool of candidate dogs, and (2) developing approaches to enhance performance, both at the individual and population level, via improvements in rearing, training, and breeding. We summarize key findings from the empirical literature regarding best practices for assessing, selecting, and improving working dogs, and conclude with future steps and recommendations for working dog organizations, breeders, trainers, and researchers.

The objective of this study was to evaluate 4 pre-exercise hydration strategies (oral water, chicken-flavored water, chicken-flavored oral electrolyte solution, and subcutaneous electrolyte solution) in working dogs conducting rigorous tracking operations in hot and arid conditions. In a randomized cross-over field study, 7 Border Patrol Search, Trauma, and Rescue (BORSTAR) Unit dogs working/training out of Fort Bliss in El Paso, Texas were randomly assigned to one of 4 different hydration strategy treatments each day for 4 days of study participation. Dogs were provided hydration treatment prior to running 2 separate one-mile tracks and were offered water while tracking. Body weight, blood, and urine were collected at the beginning of the study day and at the completion of each track. Core body temperatures were recorded using internal temperature sensing capsules. The impact of hydration strategy on change in weight, peak temperature, and serum chemical, hematological, and urinary parameters were analyzed using the COIN procedure in Ra. Compared to the other 3 hydration strategies, dogs receiving chicken-flavored water had higher blood creatine kinase values at the end of the second track (p = 0.0361). Otherwise, hydration strategy had minimal effects on blood or urine parameters. Total fluid intake was lower with water only compared to the other three hydration strategies. Dogs developed elevated core body temperatures (median 41°C; 106°F) without signs of heat exhaustion or heat stroke. Alternate hydration strategies increased total fluid intake compared to water alone; however, chicken-flavored water resulted in increased markers of muscle injury suggesting electrolyte-enriched strategies may have an advantage as a hydration strategy. Additionally, electrolyte-enriched fluids before exercise may help these dogs maintain lower peak temperatures.

Recent literature has demonstrated that dogs have the potential to detect, and communicate the presence of, various human diseases. However, there is a lack of investigation into whether commonplace training differences within the field could influence a dog's behavior during a biomedical detection task. Here we report on the behavior of four dogs trained to alert to blood plasma samples taken from individuals with ovarian cancer. One hundred trials per dog were selected from routine video recordings collected over a period of 13 months. Videos were coded frame by frame to quantify sample checking, alerting behavior, and durations of alert. Dogs had previously been trained to elicit a final response behavior once they had located the target odor. Two dogs had a “sit” response while the other two had a “stand-stare” response. Alert behavior was categorized as true positive (a correct alert to a cancer sample) or false positive (an incorrect alert to biological and non-biological controls and distractors). Hesitations were also recorded, where the dog either checks the sample twice or, spends a longer duration of time sniffing the sample than a true pass without carrying out their final response. Results show individual variation in the total frequency of false alerts elicited. However, the rate of hesitations appears to be influenced by alert style, with stand-stare dogs carrying out 40 and 32, respectively (total = 72) and sit dogs carrying out 7 and 8, respectively (total = 15). The stand-stare dogs had a non-significant difference in the duration of their true and false positive alerts. In contrast, the sit dogs showed a significant difference (p < 0.001), maintaining their false alerts for, on average, two times the duration of their true alerts. Stand-stare dogs increased the duration of time spent in contact with the port when plasma samples were present, whereas sit dogs spent on average 0.3 s in contact with the port regardless of what sample type it contained. These findings suggest that the type of operant response a biomedical detection dog has been trained may influence their sample checking and response behavior.

Citizen-driven Renewable Energy (RE) projects of various kinds, known collectively as community energy (CE), have an important part to play in the worldwide transition to cleaner energy systems. On the basis of evidence from 8 European countries, we investigate CE, over approximately the last 50 years (c.1970–2018), through the lens of Social Innovation (SI). We carry out a detailed review of literature around the social dimension of renewable energy; we collect, describe and map CE initiatives from Belgium, France, Germany, Italy, Poland, Spain, Sweden, and the UK; and we unpack the SI concept into 4 operational criteria which we suggest are essential to recognizing SI in CE. These are: (1) Crises and opportunities; (2) the agency of civil society; (3) reconfiguration of social practices, institutions and networks; (4) new ways of working. We identify three main phases of SI in CE. The environmental movements of the 1960s and the “oil shocks” of the 1970s provided the catalyst for a series of innovative societal responses around energy and self-sufficiency. A second wave of SI relates to the mainstreaming of RE and associated government support mechanisms. In this phase, with some important exceptions, successful CE initiatives were mainly confined to those countries where they were already embedded as innovators in the previous phase. The third phase of CE innovation relates to the societal response to the Great Recession that began in 2008 and lasted most of the subsequent decade. CE initiatives formed around this time were also strongly focused around democratization of energy and citizen empowerment in the context of rising energy prices, a weak economy, and a production and supply system dominated by excessively powerful multinational energy firms. CE initiatives today are more diverse than at any time previously, and are likely to continue to act as incubators for pioneering initiatives addressing virtually all aspects of energy. However, large multinational energy firms remain the dominant vehicle for delivery of the energy transition, and the apparent excitement in European policy circles for “community energy” does not extend to democratization of energy or genuine empowerment of citizens.

Behavioral traits such as trainability, fearlessness, and energy are required for dogs to succeed as search-and-rescue (SAR) dogs. Certification by the Federal Emergency Management Agency (FEMA) ensures that dogs and handlers have extensive training and have demonstrated specific skills in the field. To determine whether behavioral differences exist between SAR and pet dogs, and between FEMA-certified USAR and non-FEMA-certified SAR dogs, the Canine Behavioral Assessment and Research Questionnaire (C-BARQ) was administered to 129 SAR dogs participating in the post-9/11 medical surveillance study and a breed-matched sample of 2,131 pet dogs. Non-parametric mixed models were fit for each C-BARQ subscale with explanatory variables SAR/non-SAR status, FEMA certification status, breed, sex, neuter status, and age. SAR dogs had higher scores for trainability (P < 0.001) and energy (P < 0.001), and lower scores for aggression toward strangers (P < 0.01), aggression and fear toward dogs (P < 0.01), fear of dogs (P < 0.001), chasing (P < 0.001), fear of strangers (P < 0.001), and non-social fear (P < 0.001) than pet dogs. FEMA-certification was associated with lower fear of dogs (P < 0.05) and separation-related issues (P < 0.01) than non-FEMA certified SAR dogs. The traits identified in this study could provide guidance for more efficient selection of candidate SAR dogs and breeding stock.

Disaster search dogs traverse diverse and unstable surfaces found in collapsed buildings. It is unknown if the physical conditioning on a treadmill involves the same muscle groups that are involved in rubble search. This 14-week prospective cohort study was conducted to investigate changes within the thermal gradients of specific dog muscles following treadmill compared to rubble search. Nine dogs, ranging in age from 6 months to 4 years, were randomly assigned to one of two groups. Each week the two groups would participate in either 20 min of treadmill or rubble searches. Prior to exercise, the dogs were weighed and then kenneled in a temperature-controlled study room for 20 min at 21°C. Pre-exercise thermal images were then captured of the standing dog from the dorsal, left and right lateral, and caudal perspectives, and of the sitting dog from the rostral perspective. Following a 10-min warm-up period of stretches, dogs proceeded to either treadmill or search. Upon completion, dogs were kenneled in the study room for 20 min prior to post-exercise thermal images. Images were sectioned into 22 muscle regions, the pre-exercise images were subtracted from the post-exercise images to determine the temperature difference (ΔT) for that dog, on that day, for that activity. Thermography measures radiant energy, temperature, and converts this information into an image. This study looked at ΔT within a region pre and post-exercise. The study failed to find a statistically significant difference in the ΔT within each muscle group between treadmill and search activities. There was a decrease in ΔT within all muscle regions over the of the study except for the right cranial shoulder, right caudal shoulder, and right hamstring for the treadmill activity only. The decrease was significant in the pelvis, left longissimus, right cranial shoulder for the search activity, left oblique, left caudal shoulder, and left quadricep muscular regions. These findings suggest that ΔT in muscle groups are similar between treadmill exercise and rubble search. Regardless of the exercise type, 14 weeks of structured Search and Rescue training and treadmill exercise resulted in less ΔT associated with a structured weekly exercise.

Dogs are trained for a variety of working roles including assistance, protection, and detection work. Many canine working roles, in their modern iterations, were developed at the turn of the 20th century and training practices have since largely been passed down from trainer to trainer. In parallel, research in psychology has advanced our understanding of animal behavior, and specifically canine learning and cognition, over the last 20 years; however, this field has had little focus or practical impact on working dog training. The aims of this narrative review are to (1) orient the reader to key advances in animal behavior that we view as having important implications for working dog training, (2) highlight where such information is already implemented, and (3) indicate areas for future collaborative research bridging the gap between research and practice. Through a selective review of research on canine learning and behavior and training of working dogs, we hope to combine advances from scientists and practitioners to lead to better, more targeted, and functional research for working dogs.

The metabolism of cancer cells is an issue of dealing with fluctuating and limiting levels of nutrients in a precarious microenvironment to ensure their vitality and propagation. Glucose and glutamine are central metabolites for catabolic and anabolic metabolism, which is in the limelight of numerous diagnostic methods and therapeutic targeting. Understanding tumor metabolism in conditions of nutrient depletion is important for such applications and for interpreting the readouts. To exemplify the metabolic network of tumor cells in a model system, the fate 13C6-glucose was tracked in a breast cancer cell line growing in variable low glucose/low glutamine conditions. 13C-glucose-derived metabolites allowed to deduce the engagement of metabolic pathways, namely glycolysis, the TCA-cycle including glutamine and pyruvate anaplerosis, amino acid synthesis (serine, glycine, aspartate, glutamate), gluconeogenesis, and pyruvate replenishment. While the metabolic program did not change, limiting glucose and glutamine supply reduced cellular metabolite levels and enhanced pyruvate recycling as well as pyruvate carboxylation for entry into the TCA-cycle. Otherwise, the same metabolic pathways, including gluconeogenesis, were similarly engaged with physiologically saturating as with limiting glucose and glutamine. Therefore, the metabolic plasticity in precarious nutritional microenvironment does not require metabolic reprogramming, but is based on dynamic changes in metabolite quantity, reaction rates, and directions of the existing metabolic network.

Fit to Work is a formalized working dog foundational physical fitness assessment and development program. The Penn Vet Working Dog Center developed this program to address the needs of working dog handlers, trainers, and programs for simple, effective, and efficient methods to develop and assess working dog physical fitness. Fit to Work focuses on the foundational fitness modalities of strength, stability, mobility, and proprioception. The Penn Vet Working Dog Center piloted and refined this program over 3 months in a closed population of 31 working dogs in training. Fit to Work consists of posture development and maintenance, warm-up and cool-down routines, training exercises, and assessment methods. To simplify implementation for dogs and personnel, the foundational training program incorporates a discrete number of exercises, standardized progression steps, defined criteria for progression, and a reduced emphasis on learned behaviors. Fit to Work also enables safe and progressive assessment of foundational fitness through a tiered and inexpensive process. Future research will focus on validation of training and assessment methods, development of assessment standards, and correlation of physical fitness with operational performance.

Currently, demand for US-bred and born detector dogs exceeds available supply, while reliance on foreign-bred sources introduces many unnecessary and unwanted risks. With proper management of a domestic supply line, U.S. breeders can improve both health and behavior by applying scientific principles to breeding and raising of detector dogs. A cooperative national detector dog breeding and development program will mitigate the current shortage of domestic-bred dogs that meet the health and behavior standards required by government, military, and law enforcement agencies. To coordinate such a cooperative, we propose a Detector Dog Center of Excellence (DDCoE) led by representatives of academic canine science programs guided by an advisory board of stakeholders. As a non-governmental organization, the DDCoE will oversee selective breeding of dogs owned by breeders, purchase the resulting puppies, and its members will supervise puppy raising until dogs are of a suitable age to be purchased by government agencies or other working dog organizations. The DDCoE will serve as an approved vendor to facilitate the procurement process. Breeding decisions will be based on proven quantitative genetic methods implemented by a specialized database. A national working dog semen bank will ensure conservation of diverse genetic material and enhance selection response by providing numerous potential sires. As a data collection and genetic evaluation center, the DDCoE will lead research to define quantitative traits involved in odor detection, to understand how these traits develop, and methods to optimize training of dogs endowed with enhanced odor detection ability.

Tumors are metabolic entities wherein cancer cells adapt their metabolism to their oncogenic agenda and microenvironmental influences. Metabolically different cancer cell subpopulations collaborate to optimize nutrient delivery with respect to immediate bioenergetic and biosynthetic needs. They can also metabolically exploit host cells. These metabolic networks are directly linked with cancer progression, treatment, resistance, and relapse. Conversely, metabolic alterations in cancer are exploited for anticancer therapy, imaging, and stratification for personalized treatments. These topics were addressed at the 4th annual meeting of the International Society of Cancer Metabolism (ISCaM) in Bertinoro, Italy, on 19–21 October 2017.

Exercise-related physiological changes were evaluated in hydrated, exercise-conditioned working dogs with free access to tap water (TW) with or without a nutrient-enriched water supplement (NW). Physiological samples and measures were collected before and after work-related field tasks in a warm and moderately humid ambient environment. In a cross-over design study, 12 dogs (age range 8–23 months) were evaluated on 3 separate occasions within each period with exercise bouts up to 30 min, on days −4, 3, and 11. Dogs were offered either ad libitum TW or portion-controlled NW daily plus ad libitum TW. Prior to and serially after exercise, pulse rate (PR), core (BTcore) and ear (BTear) temperature were recorded. Urine was collected first thing in the morning, whereas blood samples collected and body weight (BW) recorded pre- and immediately post exercise. Ambient temperature was above 21.7°C (71°F) and relative humidity ranged from 36 to 76%. Activity parameters, AM urine measures, post-exercise percent change of BW, resting PR and resting BTcore did not differ between treatment groups on any exercise day. At the completion of exercise, mean BTcore for all dogs ranged from 104.8 to 105.6°F. Immediate post-exercise BTear was always lower compared to BTcore and means ranged from 103.3 to 104.0°F. The effect of time was highly significant (P < 0.001) for both BT measures with both BTcore and BTear recovering to resting levels by 60 min post exercise. PR and several blood values showed a significant main effect of time. Over the recovery period, dogs in the NW group had lower mean BTear and PR by 0.6°F and 3.4 bpm, respectively. Daily ingestion of a NW in combination with free access to TW can reduce the post-exercise-related BTcore and BTear hyperthermia, and improve pulse rate recovery following exercise in this population of working dogs undergoing 30 min bout of exercise.
