Current methods to farm tunas, especially Atlantic bluefin tunas (ABFTs; Thunnus thynnus), are unsustainable. This is driving a movement towards closed-cycle ABFT aquaculture, i.e., farming tuna from spawning and hatching to slaughter on land, to try and meet the demand for these fish. A common operational procedure in ballan wrasse production is transporting juveniles from one landbased farm to another for further on-growing. Episodes of increased mortality have been reported after such transportations. In this study, the relationship between transport stress and post-transport mortality at the on-growing facility was examined. It was also investigated if light sedation with AQUI-S® can mitigate stress during transport. Stress was quantified by measuring cortisol release rate to the tank water during transport. This was investigated in 10 commercial live carrier truck transports (6 without AQUI-S® sedation and 4 with sedation during loading and transport). The total time of transport varied between 12 and 21h. In general, mortality was significantly higher (1.0± 0.6% day-1) the first five days posttransport compared to 15-20days post transport (0.5% day-1). There was also a strong relationship between fish weight at transport and post-transport mortality, where higher mean weight at transport reduced mortality. In contrast to what was expected, AQUI-S® treatment during transport procedures increased cortisol excretion rate, suggesting a stimulating effect of AQUI-S® on the stress axis in ballan wrasse. Considering these results, the value of using AQUI-S® to reduce stress during transport of juvenile ballan wrasse might be questioned. However, there was no relationship between cortisol release rate during transport and post-transport mortality. Furthermore, this study emphasizes that water cortisol measurements can be used as a none-invasive tool for monitoring stress and can be integrated into the welfare evaluation during commercial fish transports. Assessing the potential impacts of wind farm noise on fish is a crucial aspect of Environmental Impact Assessment (EIA) studies. There is increasing evidence of disturbances and effects on hearing and behavior in animals. The black rockfish (Sebastes schlegelii) is a commercially valuable rocky reef fish native to East Asia. However, empirical studies that measure the actual consequences are lacking. In this study, we used auditory evoked potentials (AEP) to assess the effects of dominant frequency noise emitted by offshore wind farms on the auditory sensitivity, hearing threshold, swimming, and feeding behavior of juvenile black rockfish. The experimental findings revealed that the most sensitive sound frequency was 200 Hz, with the lowest hearing threshold recorded at 86.4 ± 3.4 dB re 1 mPa. Following 3 and 7 days of exposure to 200 Hz noise at 110 dB, threshold shifts in black rockfish reached 19.0 dB and 13.3 dB, respectively. During the subsequent recovery phase, these shifts decreased to approximately 9.8 dB after 3 days, respectively. The noise-exposed group exhibited higher swimming duration, moving distance, and caudal fin swing frequency compared to the control group without noise exposure. Furthermore, noise prolonged the feeding rate of black rockfish. Our findings provide the first evidence of noiseinduced temporary threshold shift and behavioral disturbances in juvenile black rockfish, implying potential fitness consequences associated with noise pollutant. The primary aim of this study was to comprehensively evaluate the welfare of Nile tilapia (Oreochromis niloticus) throughout their entire life cycle within aquaculture, spanning from reproduction to slaughter. The methodology was structured to identify welfare indicators closely aligned with the principles of animal freedoms defined by the Farm Animal Council, encompassing environmental, health, nutritional, behavioral, and psychological freedom. Notably, psychological freedom was inherently considered within the behavioral and physical analyses of the animals. To accomplish this, an integrative systematic literature review was conducted to define precise indicators and their corresponding reference values for each stage of tilapia cultivation. These reference values were subsequently categorized using a scoring system that assessed the deviation of each indicator from established ideal (score 1), tolerable (score 2), and critical (score 3) ranges for the welfare of the target species. Subsequently, a laboratory experiment was executed to validate the pre-selected health indicators, specifically tailored for the early life stages of tilapia. This test facilitated an assessment of the applicability of these indicators under operational conditions. Building on the insights gained from this experimentation, partial welfare indices (PWIs) were computed for each assessed freedom, culminating in the derivation of a general welfare index (GWI). Mathematical equations were employed to calculate these indices, offering a quantitative and standardized measure of welfare. This approach equips tilapia farmers and processors with the tools necessary for the continuous monitoring and enhancement of their production systems and stimulate the adoption of more sustainable and ethical practices within the tilapia farming.

The aim of this study was to investigate the physiological response of rainbow trout (Oncorhynchus mykiss) before slaughtering in the last phase of farming analyzing skin mucus and plasma. Two groups of rainbow trout were considered: Group UN (“unstressed”), represented by fish randomly captured from raceways, in the last phase of a standard fattening cycle; Group S (“stressed”), collected at the end of the pre-slaughtering tank, soon after slaughtering. The fish skin mucus was swabbed from head to tail using a sterile plastic spatula and the blood was collected through an endocardial puncture. qRT-PCR was used to study the gene expression in skin mucus. The mRNA expression levels of the IL-6 and IgD genes were higher in the S than in the Group UN. The plasma analysis showed an only a decrease in the glucose plasma levels in the Group S when compared to the Group UN. The present results indicated that the procedures adopted after slaughtering only affected changes in plasma glucose and skin mucus activity in rainbow trout suggesting that management protocol was compatible with non-stressful farming conditions.

Curiosity—the motivation to seek out information—has been studied widely across the animal kingdom. To investigate curiosity in zebrafish we presented 30 novel objects to groups of zebrafish housed in semi-naturalistic tanks (6 tanks; 10 fish/tank; 10-min presentations). During the first 100 s and final 100 s of each object's 10-min presentation period, we recorded each group's: (i) latency to approach the object, (ii) attraction to the object, (iii) social dynamics: agonistic behavior and group cohesion and coordination, and (iv) diving behavior, a stress response in zebrafish. Comparing these behaviors to a 100 s baseline period when no object was present, we tested for neophobia (avoidance of novelty), neophilia (overall attraction to novelty), sustained interest (prolonged attraction to at least some presentations), discriminant interest (certain objects eliciting more attention than others), habituation (loss of interest over time), and alterations to social and stress behaviors. Zebrafish groups readily approached all objects (1 s median latency), were neophilic throughout all object presentations, and showed systematic sustained interest only for some object presentations at the beginning of the study (object presentations 1–10). Over the course of the study, zebrafish also showed signs of habituation such that by the final ten object presentations (21-30), there were no signs of overall sustained interest. During the beginning of the study (object presentations 1–10), we also found evidence for specific object-driven interest, with object ID accounting for 11% of the variability in interest scores (p < 0.01), and object-driven interest corresponding to alterations in social behavior: decreased aggression (p < 0.02), increased group cohesion (p < 0.02), and increased group coordination (p < 0.05). By explicitly investigating curiosity in fish, this work reveals that under certain conditions, zebrafish voluntarily engage in cognitive stimulation opportunities. More work is needed to clarify what types of information zebrafish find most rewarding and how long-term exposure to such opportunities may affect fish welfare.

Environmental enrichment (EE) improves the growth rate and welfare of some cultured fishes. However, most cultured fish species are raised in non-enriched housing conditions. Clarias gariepinus is an important commercial fish species, but little is known about the effect of EE on their welfare. This study examined the effect of different EE on the survival rate (SR), growth [mean weight gain (MWG), specific growth rate (SGR) and feed conversion ratio (FCR)], behavioral (feed response, aggressive acts and shoaling time) and physiological responses (blood glucose) of C. gariepinus. One hundred and twenty juveniles of C. gariepinus (31.65 ± 0.69 g) were randomly allocated at 10 fish/tank and subjected to either Plant Enriched (PE), Substratum Enriched (SE), Plant and Substratum Enriched (PSE) and Non-Enriched (NE) tanks in triplicates for 56-days. Behavioral acts were observed for 10 min twice daily, and glucose level in blood samples was evaluated. Data were checked for normality using the Shapiro-Wilk test before being analyzed with the Kruskal-Wallis test. SR and MWG were significantly higher in Clarias gariepinus exposed to SE, with no significant differences among PE, PSE and NE treatments. There was no significant difference between the SGR of PSE and NE. FCR was similar between treatments. The highest condition factor (k) was recorded in SE tanks. Duration of feed response was shorter in SE, but there was no significant difference between the feed response of C. gariepinus exposed to PE and PSE. C. gariepinus exposed to PE, SE and PSE displayed a similar frequency of aggressive acts. African catfish reared in NE (barren) tanks had the least duration of shoaling period. The experiment consistently found the highest and least glucose values in PSE and SE. In conclusion, environmentally enriched housing tanks with SE resulted in the best MWG with a reduced level of aggression in C. gariepinus under laboratory conditions. Thus, EE might be applicable to boost fish productivity on a commercial scale.

The effects of stocking density on fish welfare are complex and involve many interacting parameters. This complex relationship between fish welfare, stocking density and influencing factors make it challenging to define a specific optimal (“golden”) stocking density. Indeed, previously published recommendations on stocking density for different species of aquaculture interest are incredibly variable even at the same life stage, and can also vary widely within a rearing unit. Production density can be estimated quite accurately if the farmer has good biomass control and a known water volume, but it is difficult to set minimum and maximum stocking density levels that will protect welfare. However, there is little doubt that stocking densities that are too low or too high can have negative impacts on welfare and/or production. Here, we propose how to select density on captive fish and monitor its potential effects integrating 1) solid welfare assessment based on operational welfare indicators and 2) good management practices. Regulation directly limiting stocking density is likely to be unworkable and ineffective, and a more rational option might be to prescribe acceptable levels of different welfare indicators (e.g., water quality, health, nutritional condition and behavioral indicators), which together with a positive economic balance of the company, allow to estimate the most suitable range of fish density for each particular species, life-stage and production systems.

Closed-containment (CCS) systems offer several advantages in controlling the production environment for Atlantic salmon (Salmo salar) aquaculture, especially at sea, where fish are more exposed to challenging environmental conditions. Here, we report the health and welfare of Atlantic salmon in FishGLOBE V5, a 3500 m3 semi-closed containment system. A group of 200,000 post-smolts was followed from a recirculating aquaculture system (RAS) facility, then for three and a half months in FishGLOBE V5, and three months after release in net pens. Fish samples were collected at four time points during the production period, while water quality was evaluated when the fish were in FishGLOBE V5. The water quality in FishGLOBE V5 was within the recommended range for salmon post-smolts. The mortality rate was 1.4% in FishGLOBE V5 and 4.9% in net pens. There was an increase in the prevalence of eye, snout and dorsal fin damages before the fish left the FishGLOBE V5 phase. The plasma level of magnesium was slightly elevated during this phase as well, indicating osmotic imbalance and stress. Histological evaluation of skin and gills showed sporadic cases of non-specific pathologies. In particular, the transitory stay in FishGLOBE V5 partially improved skin health, but not gill health. Gene expression profiling of some biomarkers showed that rearing fish in FishGLOBE V5 could influence the expression of genes involved in stress response, mucus production, and epithelial integrity. This study documented that rearing salmon for a certain period in FishGLOBE V5 during production affected different health and welfare indicators. These changes should be taken into consideration for the improvement of FishGLOBE V5 as a viable technology for post-smolt production at sea.

Still, the use of fish-based non-invasive welfare indicators, reflecting the physiological state of the fish, is limited in this type of system. Cortisol, the major stress-coping hormone in fish, diffuses through the gills. Consequently, waterborne cortisol is a potential fish-based non-invasive welfare indicator in RAS. However, its use in commercial rearing systems is sparse. In this study, we evaluated water cortisol levels and feeding behavior as welfare indicators of newly inserted smolt in a commercial RAS for harvest size Atlantic salmon. The RAS consisted of two parallel fish rearing raceways, raceways 1 and 2, sharing the same water treatment with common outlets and inlets. The smolts were inserted in raceway 1 while salmon that have been in the system for 6 months or more were kept in raceway 2. The smolt insertion period was 3 days. Samples for water cortisol levels were withdrawn the day before, 1 and 3 days after the smolt insertion period. Smolt insertion resulted in elevated water cortisol concentrations in the entire system, with the highest values in raceway 1, one day after smolt insertion. Estimated cortisol production in newly inserted smolt decreased over time, was similar to what has been reported in salmon adapting to experimental tanks. Feeding behavior indicated that the appetite was not fully resumed in the newly inserted smolts, while the appetite of fish in raceway 2 was unaffected by smolt insertion. These results, obtained in a highly intensive commercial RAS, suggest that waterborne cortisol together with feeding behavior can be used as indicators for adaptive processes associated with stress resilience in farmed fish. Aquaculture is a growing industry worldwide and Canadian finfish culture is dominated by marine salmonid farming. In part due to increasing public and stakeholder concerns around fish welfare protection, the first-ever Canadian Code of Practice for the Care and Handling of Farmed Salmonids was recently completed, following the National Farm Animal Care Council's (NFACC) rigorous Code development process. During this process, both the Scientific (responsible for reviewing existing literature and producing a peer-reviewed report that informs the Code) and Code Development (a diverse group of stakeholders including aquaculture producers, fish transporters, aquaculture veterinarians, animal welfare advocates, food retailers, government, and researchers) Committees identified research gaps in tandem, as they worked through the literature on salmonid physiology, health, husbandry, and welfare. When those lists are combined with the results of a public “top-of-mind” survey conducted by NFACC, they reveal several overlapping areas of scientific, stakeholder, and public concern where scientific evidence is currently lacking: (1) biodensity; (2) health monitoring and management, with a focus on sea lice infection prevention and management; (3) feed quality and management, particularly whether feed restriction or deprivation has consequences for welfare; (4) enclosure design, especially focused on environmental enrichment provision and lighting design; and (5) slaughter and euthanasia. For each of these five research areas, we provide a brief overview of current research on the topic and outline the specific research gaps present. The final section of this review identifies future research avenues that will help address these research gaps, including using existing paradigms developed by terrestrial animal welfare researchers, developing novel methods for assessing fish welfare, and the validation of new salmonid welfare indices. We conclude that there is no dearth of relevant research to be done in the realm of farmed salmonid welfare that can support crucial evidence-based fish welfare policy development.

Advances in tag technology now make it possible to monitor the behavior of small groups of individual fish as bioindicators of population wellbeing in commercial aquaculture settings. For example, tags may detect unusual patterns in fish heart rate, which could serve as an early indicator of whether fish health or welfare is becoming compromised. Here, we investigated the use of commercially available heart rate biologgers implanted into 24 Atlantic salmon weighing 3.6 ± 0.8 kg (mean ± SD) to monitor fish over 5 months in a standard 12 m × 12 m square sea cage containing ∼6,000 conspecifics. Post tagging, fish established a diurnal heart rate rhythm within 24 h, which stabilized after 4 days. Whilst the registered tagged fish mortality over the trial period was 0%, only 75% of tagged fish were recaptured at harvest, resulting in an unexplained tag loss rate of 25%. After 5 months, tagged fish were approximately 20% lighter and 8% shorter, but of the similar condition when compared to untagged fish. Distinct diurnal heart rate patterns were observed and changed with seasonal day length of natural illumination. Fish exhibited lower heart rates at night [winter 39 ± 0.2 beats per min (bpm), spring 37 ± 0.2 bpm, summer 43 ± 0.3 bpm, mean ± SE] than during the day (winter 50 ± 0.3 bpm, spring 48 ± 0.2 bpm, summer 49 ± 0.2 bpm) with the difference between night and day heart rates near half during the summer (6 bpm) compared to winter and spring (both 11 bpm). When fish experienced moderate and severe crowding events in early summer, the highest hourly heart rates reached 60 ± 2.5 bpm and 72 ± 2.4 bpm, respectively, on the day of crowding. Here, if the negative sublethal effects on fish that carry tags (e.g., growth rate) can be substantially reduced, the ability to monitor diurnal heart rate patterns across seasons and detect changes during crowding events, and using heart rate biologgers could be a useful warning mechanism for detecting sudden changes in fish behavior in sea cages. Hepatic metabolic adjustments are key adaptive mechanisms to stress in fish targeting at increasing energy availability for the animal to efficiently cope with a stressor. Teleosts exhibit a broad variety of these metabolic responses, depending on the species biology, individual experiences and the challenge’s characteristics. Nevertheless, the molecular response to a prolonged stress can be more heterogeneous and far more complex to interpret than that to an acute stress. A comparative proteomics analysis was employed to discover the set of liver proteins involved in the adaptive processes that tune the physiological response of Sparus aurata to different suboptimal rearing conditions and physical challenges. Three separated trials were established where fish were submitted to different conditions (overcrowding, net handling and hypoxia). The response at the transcript level of 13 genes was also assessed. Mass spectrometric analysis revealed 71 differential abundant proteins distributed among the trials. Prolonged exposure to stress seems to have induced widespread changes in amino acid, carbohydrate, and lipid metabolisms, antioxidant response and protein folding, sorting and degradation processes. Two genes corresponding to heat-shock proteins were found to be differently expressed in net handled fish. These results shed light on the dynamics and extent of this species’ metabolic reprogramming under different challenges, supporting future studies on stress markers’ discovery and fish welfare research.

Integration of technological solutions aims to improve accuracy, precision and repeatability in farming operations, and biosensor devices are increasingly used for understanding basic biology during livestock production. The aim of this study was to design and validate a miniaturized tri-axial accelerometer for non-invasive monitoring of farmed fish with re-programmable schedule protocols. The current device (AE-FishBIT v.1s) is a small (14 mm × 7 mm × 7 mm), stand-alone system with a total mass of 600 mg, which allows monitoring animals from 30 to 35 g onwards. The device was attached to the operculum of gilthead sea bream (Sparus aurata) and European sea bass (Dicentrarchus labrax) juveniles for monitoring their physical activity by measurements of movement accelerations in x- and y-axes, while records of operculum beats (z-axis) served as a measurement of respiratory frequency. Data post-processing of exercised fish in swimming test chambers revealed an exponential increase of fish accelerations with the increase of fish speed from 1 body-length to 4 body-lengths per second, while a close relationship between oxygen consumption (MO2) and opercular frequency was consistently found. Preliminary tests in free-swimming fish kept in rearing tanks also showed that device data recording was able to detect changes in daily fish activity. The usefulness of low computational load for data pre-processing with on-board algorithms was verified from low to submaximal exercise, increasing this procedure the autonomy of the system up to 6 h of data recording with different programmable schedules. Visual observations regarding tissue damage, feeding behavior and circulating levels of stress markers (cortisol, glucose, and lactate) did not reveal at short term a negative impact of device tagging. Reduced plasma levels of triglycerides revealed a transient inhibition of feed intake in small fish (sea bream 50–90 g, sea bass 100–200 g), but this disturbance was not detected in larger fish. All this considered together is the proof of concept that miniaturized devices are suitable for non-invasive and reliable metabolic phenotyping of farmed fish to improve their overall performance and welfare. Further work is underway for improving the attachment procedure and the full device packaging.

Fish have evolved a biological clock to cope with environmental cycles, so they display circadian rhythms in most physiological functions including stress response. Photoperiodic information is transduced by the pineal organ into a rhythmic secretion of melatonin, which is released into the blood circulation with high concentrations at night and low during the day. The melatonin rhythmic profile is under the control of circadian clocks in most fish (except salmonids), and it is considered as an important output of the circadian system, thus modulating most daily behavioral and physiological rhythms. Lighting conditions (intensity and spectrum) change in the underwater environment and affect fish embryo and larvae development: constant light/darkness or red lights can lead to increased malformations and mortality, whereas blue light usually results in best hatching rates and growth performance in marine fish. Many factors display daily rhythms along the hypothalamus-pituitary-interrenal (HPI) axis that controls stress response in fish, including corticotropin-releasing hormone (Crh) and its binding protein (Crhbp), proopiomelanocortin A and B (Pomca and Pomcb), and plasma cortisol, glucose, and lactate. Many of these circadian rhythms are under the control of endogenous molecular clocks, which consist of self-sustained transcriptional-translational feedback loops involving the cyclic expression of circadian clock genes (clock, bmal, per , and cry) which persists under constant light or darkness. Exposing fish to a stressor can result in altered rhythms of most stress indicators, such as cortisol, glucose, and lactate among others, as well as daily rhythms of most behavioral and physiological functions. In addition, crh and pomca expression profiles can be affected by other factors such as light spectrum, which strongly influence the expression profile of growth-related (igf1a, igf2a) genes. Additionally, the daily cycle of water temperature (warmer at day and cooler at night) is another factor that has to be considered. The response to any acute stressor is not only species dependent, but also depends on the time of the day when the stress occurs: nocturnal species show higher responses when stressed during day time, whereas diurnal fish respond stronger at night. Melatonin administration in fish has sedative effects with a reduction in locomotor activity and cortisol levels, as well as reduced liver glycogen and dopaminergic and serotonergic activities within the hypothalamus. Transport procedures usually cause fish stress. The purpose of this study was to investigate the effect of transport stress on blood biochemical profiles, oxidative stress biomarkers, and hepatic heat shock proteins (HSPs) of channel catfish (Ictalurus punctatus). Fish (body weight 55.57 ± 5.13 g) were randomly distributed to two groups, the control, and the treatment. The control group was kept under the normal culture conditions. The treatment group was exposed to the process of transport (3.5 h). Fish samples were collected before transport, after packing and at 0, 1, 6, 24, 72, and 168 h after transport, respectively. Transport caused a significant increase in the serum concentrations of cortisol, glucose, total cholesterol, and triglyceride, as well as, the activity of aspartate aminotransferase at 0 and 1 h after transport compared with non-transported fish and the basal level. Blood total protein content significantly declined in the transported fish. Total antioxidant capacity (T-AOC), malonaldehyde content, and the activities of both glutathione peroxidase and catalase significantly increased in fish within 6 h after transport. The transported fish exhibited a significant higher level in either the concentration of nitric oxide or the mRNA expressions of both hepatic HSP70 and HSP90. It is concluded that transport triggers stress response of I. punctatus, leading to the obvious change in antioxidant capacity. I. punctatus need to be more care after transport to recover from transport stress.

The present study aims at identifying the lowest number of fish (European seabass) that could be used for monitoring and/or experimental purposes in small-scale fish facilities by quantifying the effect that the number of individuals has on the Shannon entropy (SE) of the trajectory followed by the shoal’s centroid. Two different experiments were performed: (i) one starting with 50 fish and decreasing to 25, 13, and 1 fish, and (ii) a second experiment starting with one fish, adding one new fish per day during 5 days, ending up with five fish in the tank. The fish were recorded for 1h daily, during which time a stochastic event (a hit in the tank) was introduced. The SE values were calculated from the images corresponding to three arbitrary basal (shoaling) periods of 3.5 min prior to the event, and to the 3.5 min period immediately after the event (schooling response). Taking both experiments together, the coefficient of variation (CV) of the SE among measurements was largest for one fish systems (CV 37.12 and 17.94% for the daily average basal and response SE, respectively) and decreased concomitantly with the number of fish (CV 8.6–10% for the basal SE of 2 to 5 fish systems and 5.86, 2.69, and 2.31% for the basal SE of 13, 25, and 50 fish, respectively). The SE of the systems kept a power relationship with the number of fish (basal: R2= 0.93 and response: R2= 0.92).
