Background: Injury risk in canine sprinting sports, such as greyhound racing and agility, have been previously documented through various surveys. Flyball, another sprinting canine sport with similar athletic requirements to agility, has yet to be assessed for factors associated with injury. The aim of this study was to determine injury prevalence and assess for risk factors for injury in flyball dogs. Methods: Survey data from 375 flyball participants was collected and analyzed. Data collected included patient-specific variables, equipment use, training/competition practices, and injury occurrence and localization. Univariate analysis was utilized for all variables of interest, followed by backwards nominal logistic regression to identify variables associated with increased risk of injury, with a p < 0.05 defined as significant. Results: Thirty-nine percent of dogs incurred at least one injury with 172 injuries reported. Injuries to the limbs were common (30.8% affecting forelimbs, 25.6% affecting hindlimbs), with the paw or nail the most frequently reported injured area (19.2%). Only protective wrap use and age were significantly associated with injury in the final nominal regression model ( p < 0.01). A biphasic injury rate with more injuries in younger dogs was observed, and injuries peaked by 6 years of age. Use of carpal wraps was positively associated with increased injury risk. Conclusions: These findings suggest an association between younger dogs and greater risk of injury, as well as identify a need to further investigate the utilization of wraps and potential association between injury risk among flyball participants., (Copyright © 2019 Montalbano, Gamble, Walden, Rouse, Mann, Sack, Wakshlag, Shmalberg and Wakshlag.)

Galectins are potent regulators of cell adhesion, growth and apoptosis in diverse cell types, including chondrocytes and synovial fibroblasts. Elevations in synovial fluid galectin-3 have been observed in rheumatoid arthritis, juvenile idiopathic arthritis and experimental inflammatory arthritis in animal models, whereas galectin-1 is thought to be protective. Less is known about galectins-1 and-3 in osteoarthritis (OA). Therefore, the purpose of this study was: (1) to determine whether galectin-1 and-3 synovial fluid concentrations and synovial membrane and cartilage histochemical staining were altered following osteochondral injury in an experimental equine osteoarthritis (OA) model and (2) to measure galectin-1 and-3 mRNA expression and synovial fluid concentrations in naturally occurring equine carpal OA. Synovial fluid galectin-1 and-3 concentrations were quantified using custom ELISAs in two research horse cohorts undergoing experimental OA induction ( n = 5 and 4) and in a cohort of horses with naturally occurring carpal OA ( n = 57). Galectin mRNA expression in synovial membrane and cartilage tissue obtained from carpal joints of horses with naturally occurring OA was measured using RT-qPCR, and galectin immunostaining was assessed in synovial membrane and osteochondral tissues in the experimental model ( n = 5). Synovial fluid galectin-1 and-3 concentrations increased following experimental carpal osteochondral fragmentation. Cartilage galectin-1 mRNA expression increased with OA severity in naturally occurring disease. The superficial zone of healthy articular cartilage stained intensely for galectin-3 in sham-operated joints, whereas galectin-1 staining was nearly absent. Chondrocyte galectin-1 and-3 immunoreactivity increased following cartilage injury, particularly in galectin-1 positive chondrones. Galectins-1 and-3 are present in healthy equine synovial fluid and increase following post-traumatic OA. Healthy superficial zone chondrocytes express galectin-3, whereas galectin-1 chondrocyte staining is limited predominantly to chondrones and injured cartilage. Further work is needed to clarify the functions of galectins-1 and-3 in healthy and OA joints.

Objectives: To examine the relative levels of heavy metals and arsenic content in commercial dog foods (arsenic, cadmium, lead and mercury) of 51 over-the-counter maintenance or all-life-stage dry dog foods. All products were chosen and segregated based on meat sources (fish, poultry, red-meat-17 products from each category) as animal protein sources being the primary contaminated ingredient due to bioaccumulation. Methods: Inductively coupled plasma mass spectrometry (ICP-MS) was performed on products that were classified as fish, red meat (beef, pork, venison, bison) or poultry (chicken, turkey, duck) based. A non-Gaussian data distribution for each heavy metal within category distribution led to non-parametric statistical testing and median (range) descriptive statistics. Comparison to average human consumption based on mg/megacalorie (Mcal)was also examined. Results: Based on caloric consumption, total arsenic and heavy metal consumption is higher in dogs than in humans; however chronic toxic exposure levels are highly unlikely. Fish-based diets had significantly higher arsenic, cadmium and mercury content than the poultry or red meat-based diets ( p < 0.01). Red meat-based diets (beef, venison and bison) had higher lead concentrations than poultry and fish-based diets ( p < 0.03). Clinical Significance: Based on the findings, commercial dog foods appear to be safe for chronic consumption and concentrations of the heavy metals were dependent on primary protein sources. Overall, poultry-based diets had relatively lower heavy metal and arsenic content than red meat and fish-based diets. Despite the safety of most pet foods occasional outliers for lead render some concern for chronic exposure based on other species toxicity data and a lack of data in dogs.

Objectives: The objectives of this study were to determine basic oral pharmacokinetics, and assess safety and analgesic efficacy of a cannabidiol (CBD) based oil in dogs with osteoarthritis (OA). Methods: Single-dose pharmacokinetics was performed using two different doses of CBD enriched (2 and 8 mg/kg) oil. Thereafter, a randomized placebo-controlled, veterinarian, and owner blinded, cross-over study was conducted. Dogs received each of two treatments: CBD oil (2 mg/kg) or placebo oil every 12 h. Each treatment lasted for 4 weeks with a 2-week washout period. Baseline veterinary assessment and owner questionnaires were completed before initiating each treatment and at weeks 2 and 4. Hematology, serum chemistry and physical examinations were performed at each visit. A mixed model analysis, analyzing the change from enrollment baseline for all other time points was utilized for all variables of interest, with a p ≤ 0.05 defined as significant. Results: Pharmacokinetics revealed an elimination half-life of 4.2 h at both doses and no observable side effects. Clinically, canine brief pain inventory and Hudson activity scores showed a significant decrease in pain and increase in activity ( p < 0.01) with CBD oil. Veterinary assessment showed decreased pain during CBD treatment ( p < 0.02). No side effects were reported by owners, however, serum chemistry showed an increase in alkaline phosphatase during CBD treatment ( p < 0.01). Clinical significance: This pharmacokinetic and clinical study suggests that 2 mg/kg of CBD twice daily can help increase comfort and activity in dogs with OA.

Serum muscle enzymes in endurance sled dogs peak within 2-4 days of racing. The object of this study was to compare mid-race serum chemistry profiles, select hormones, markers of inflammation, and the acute phase response in dogs that successfully completed half of the 2015 Yukon Quest sled dog race to their pre-racing samples ( n = 14), as well as mid-race samples of successful dogs to those who developed clinical exertional rhabdomyolysis ( ER ) ( n = 5). Concentrations of serum phosphorus in ER dogs were significantly elevated compared to healthy dogs (median 5.5 vs. 4.25 mg/dL, P < 0.01) at mid race. ALT, AST, and CK show a significant increase from pre-race baseline to mid-race chemistries ( P < 0.01), with more pronounced increases in dogs with ER compared to healthy racing dogs (CK- median 46,125 vs. 1,743 U/L; P < 0.01). Potassium concentrations were significantly decreased from pre-race baselines in all dogs (median 5.1 vs. 4.5 mEq/L; P < 0.01), and even lower in dogs with ER (median 3.5 mEq/L; P < 0.01) mid-race. No changes in serum pro-inflammatory cytokine concentrations were noted in any groups of dogs. C-reactive protein was elevated in both groups of dogs, but significantly higher in those with ER compared with healthy dogs mid-race (median 308 vs. 164 ug/mL; P < 0.01). Healthy dogs may have CK elevations over 10,000 U/L, and dogs with ER were over 30,000 U/L. Although potassium decreases in healthy endurance sled dogs during racing, it remains in the normal laboratory reference range; however ER dog potassium levels drop further to the point of hypokalemia. Lastly increases in CRP may be reflective of a physiological response to exercise over the course of a race; however high CRP in ER dogs may be capturing an early acute phase response.
