Condition-dependence ejaculate traits is taxonomically widespread and entails important adaptive potential. A recent meta-analysis (Macartney et al. 2019)1 reports that numerous ejaculate traits will be reduced by nutrient limitation, especially sperm number. We predicted the number of sperm transferred to a female (vs. produced / stored) to be an informative predictor of a male's fitness and post-copulatory reproductive strategy. Empirical studies in internally fertilising species, however, rarely examine this trait, mainly because of practical difficulties, such as the visualisation of the sperm. We overcome this hurdle by using males with glow-in-the-dark-sperm and study how the sperm depletion rate (over successive matings, a realistic scenario) is affected by the dietary regime. We control for genetic background using males from multiple isogenic lines.
