The aim of this study was to investigate the effect of pre-slaughter handling on the occurrence of PSE (Pale, Soft, and Exudative) meat in swine slaughtered at a commercial slaughterhouse located in the metropolitan region of Dourados, Mato Grosso do Sul, Brazil. Based on the database (n=1,832 carcasses), it was possible to apply the integrated multivariate analysis for the purpose of identifying, among the selected variables, those of greatest relevance to this study. Results of the Principal Component Analysis showed that the first five components explained 89.28% of total variance. In the Factor Analysis, the first factor represented the thermal stress and fatiguing conditions for swine during pre-slaughter handling. In general, this study indicated the importance of the pre-slaughter handling stages, evidencing those of greatest stress and threat to animal welfare and pork quality, which are transport time, resting period, lairage time before unloading, unloading time, and ambience. Este trabalho objetivou investigar a influência do manejo pré-abate na incidência de carne PSE (Pale, Soft and Exudative), em suínos abatidos em um abatedouro comercial, localizado na região metropolitana de Dourados-MS. A partir do banco de dados (n=1.832 carcaças), aplicou-se análise multivariada integrada, com a finalidade de identificar, dentre as variáveis selecionadas, quais representavam maior relevância para o estudo. Os resultados da Análise de Componentes Principais indicaram que os cinco primeiros componentes explicaram 89,28% da variância total. Na Análise de Fatores, o primeiro fator representou as condições fatigantes e de estresse térmico para os suínos, durante o manejo pré-abate. De modo geral, o estudo indicou a importância das etapas do manejo pré-abate, evidenciando aquelas que representam maior estresse e ameaça ao bem-estar animal e à qualidade da carne suína, quais sejam: o tempo de transporte, de descanso médio nas baias, de espera no pátio, de descarregamento e ambiência.

Avaliou-se o efeito do período de descanso pré-abate e do sexo sobre a incidência de carne PSE (pale, soft, exudative) em suínos. Foram realizadas três visitas a abatedouro comercial, nas quais se mensurou o pH das carcaças (n=2128) aos 45 minutos após o abate, de lotes compostos por fêmeas, machos castrados cirurgicamente e machos imunocastrados e de lotes mistos (fêmeas e machos castrados cirurgicamente), submetidos a período de descanso que variou de duas a 16 horas. Carcaças com pH45≤5,8 foram classificadas como PSE, e com pH45>5,8 como normais. Em função do número de animais avaliados dentro de cada categoria, estimou-se a frequência de carne PSE. A incidência total de carne PSE foi de 10,1%, sendo maior nos lotes de animais imunocastrados (13,5%) quando comparados aos lotes de fêmeas (8,6%) ou de machos castrados cirurgicamente (8,5%). Períodos de descanso menores que seis horas e acima de 14 horas aumentaram a incidência de carne PSE. Período de descanso entre seis e oito horas minimizaram a ocorrência de carnes PSE. Were evaluated the effect of lairage pre-slaughter and sex on the incidence of PSE (pale, soft and exudative) meat in pigs. There were three visits to a commercial slaughterhouse, in which the pH of carcasses (n=2128) was measured at 45 minutes after slaughter of lots consisting exclusively of females, males castrated surgically, males immunologically castrated and mixed lots (females and castrated surgically) underwent a resting time, and time between the shipment and slaughter ranged from two to 16 hours. Carcasses with pH45≤5.8 were classified as PSE and with pH45>5.8 as normal. Depending on the number of animals evaluated in each category we estimated the frequency of PSE. The overall incidence of PSE meat was 10.1%, higher in lots of animals immunocastrated (13.5%) compared to lots of females (8.6%) or castrated males (8.5%). Resting time of less than six hours and up to 14 hours increased the incidence of PSE meat. Resting time between six and eight hours minimized the occurrence of PSE meat.

In this study, L* (lightness), a* (redness), and b* (yellowness) values of 500 chicken breast fillets were determined using a portable colorimeter (Minolta, model CR-400) in a commercial processing plant. Fillets were considered pale when their L* was >49. Out of those samples, 30 fillets with normal color and 30 pale fillets were evaluated as to pH, drip loss, cooking loss, water holding capacity, shear force, and submitted to sensorial analysis. An incidence of 10.20% PSE meat was determined. Pale and normal fillets presented significantly different (p0.05) between pale and normal fillets. Despite the significant differences in meat physical properties, these were not perceived by consumers in terms of tenderness, aspect, and flavor. The observed incidence of PSE may cause losses due to its low water retention capacity. Total RNA isolated from Pectoralis major muscle from PSE (L*24h>53.0, pH"53) meats of two phenotypically distinct chicken lines, broiler and layer, was used to investigate the α-ryr and β-ryr gene expression by real-time RT-PCR approach. Mean relative quantification (RQ) values were lower (p0.05) in α-ryr gene expression regardless of line studied. The β-ryr RQ results suggested that in PSE samples an alteration might occur in the regular ratio (1:1) of α-RyR/β-RyR normally found in avian muscles. These results provided the first evidence of PSE meat occurrence as a result of the differential expression of ryanodine receptor genes which might lead to an increased in Ca2+ availability at the cell milieu.As proteínas α-RyR e β-RyR apresentam papéis distintos no mecanismo de excitação-contração com diferenças em seus mecanismos de ativação e respostas a ligantes. O RNA total de filé de peito (Pectoralis major m) com PSE (L*24h>53,0; pH 5,8) e não-PSE (4453) de duas linhagens distintas, de corte e de postura, foram utilizadas para estudar a expressão gênica dos genes α-ryr β-ryr por PCR-em-tempo-real. Os valores médios de expressão gênicas relativas (RQ) foram inferiores (p0,05) na expressão do , independentemente da linhagem estudada. Os resultados de RQ para β-ryr indicaram nas amostras PSE, uma alteração na proporção (1:1) de α-RyR/β-RyR comumente encontrada em músculos de aves. Estes resultados originam a primeira evidência da ocorrência de carnes PSE como resultado de uma disponibilidade acentuada de Ca2+ no citosol pela expressão diferenciada de proteínas receptoras de rianodina.

The objective of this work was to investigate the use of a halothane to screen broiler chickens prone to develop PSE (Pale, Soft, Exudative) meat through a special gas chamber connected to a veterinarian anaesthetic apparatus developed in our laboratory. Anaesthesia was induced with approx. 3.0% halothane at a flow rate of 6.0 Lm-1 in pure oxygen for 5 min. Commercial male broilers (n=342) aged 46 days old were evaluated and classified as either halothane sensitive (HAL+) or insensitive (HAL-), depending on the leg rigidity response. Approximately 27.5% (n=94) of broilers were HAL+ and 72.5% (n=248) were HAL-. This is a simple and rapid technique to evaluate broiler sensitivity to halothane and identify broilers prone to develop PSE meat. The occurrence of PSE meat in HAL insensitive broiler chicken samples suggests that other factors are related to PSE occurrence.O objetivo deste trabalho foi investigar o uso do halotano para avaliar e identificar frangos propensos ao desenvolvimento de carnes PSE (Pálida, Flácida, Exsudativa) por meio de uma câmara desenvolvida em nossos laboratórios. Esta foi conectada a um equipamento de anestesia veterinária e a anestesia foi então induzida com aproximadamente 3,0% de halotano a um fluxo de 6,0 L/min em puro oxigênio. A sensibilidade de três frangos foi avaliada simultaneamente. Os frangos de linhagem comercial (n=352) com 46 dias de idade foram expostos à 3,0% de halotano por 5 min e classificados como halotano sensível (HAL +) e não-sensível (HAL -), baseado no enrijecimento dos seus membros inferiores. Aproximadamente 27,5% (n=94) frangos foram HAL+ e 72,5% (n=248) HAL-. Amostras do músculo Pectoralis major (n=75) foram coletadas após o abate dos animais para as medidas de pH e cor, determinadas nos filés mantidos a 4 C por 24 h postmortem. Frangos HAL+ apresentaram maior porcentagem de carnes PSE quando comparadas ao grupo HAL-. Este é um teste simples e rápido para a avaliação da sensibilidade das aves ao halotano e identificar frangos propensos ao desenvolvimento de carnes PSE embora amostras de HAL - também apresentaram carnes PSE sugerindo que outros fatores estão relacionados com a formação da anormalidade.
