Bacterial biofilm formation and attachment to hosts are mediated by carbohydrate-binding lectins, exopolysaccharides, and their interactions in the extracellular matrix (ECM). During tomato infection Ralstonia pseudosolanacearum (Rps) GMI1000 highly expresses three lectins: LecM, LecF, and LecX. The latter two are uncharacterized. We evaluated the roles in bacterial wilt disease of LecF, a fucose-binding lectin, LecX, a xylose-binding lectin, and the Rps exopolysaccharide EPS I. Interestingly, single and double lectin mutants attached to tomato roots better and formed more biofilm under static conditions in vitro. Consistent with this finding, static bacterial aggregation was suppressed by heterologous expression of lecFGMI1000 and lecXGMI1000 in other Ralstonia strains that naturally lack these lectins. Crude ECM from a ΔlecF/X double mutant was more adhesive than the wild-type ECM, and LecF and LecX increased Rps attachment to ECM. The enhanced adhesiveness of the ΔlecF/X ECM could explain the double mutants hyper-attachment in static conditions. Unexpectedly, mutating lectins decreased Rps attachment and biofilm viscosity under shear stress, which this pathogen experiences in plant xylem. LecF, LecX, and EPS I were all essential for biofilm development in xylem fluid flowing through cellulose-coated microfluidic channels. These results suggest that under shear stress, LecF and LecX increase Rps attachment by interacting with the ECM and plant cell wall components like cellulose. In static conditions such as on root surfaces and in clogged xylem vessels, the same lectins suppress attachment to facilitate pathogen dispersal. Thus, Rps lectins have a dual biological function that depends on the physical environment.

Large gas bubbles can reach the surface of pools of mud and lava where they burst, often through the formation and expansion of circular holes. Bursting bubbles release volatiles and generate spatter, and hence play a key role in volcanic degassing and volcanic edifice construction. Here, we study the ascent and rupture of bubbles using a combination of field observations at Pâclele Mici (Romania), laboratory experiments with mud from the Imperial Valley (California, USA), numerical simulations and theoretical models. Numerical simulations predict that bubbles ascend through the mud as elliptical caps that develop a dimple at the apex as they impinge on the free surface. We documented the rupture of bubbles in nature and under laboratory conditions using high-speed video. The bursting of mud bubbles starts with the nucleation of multiple holes, which form at a near-constant rate and in quick succession. The quasi-circular holes rapidly grow and coalesce, and the sheet evolves towards a filamentous structure that finally falls back into the mud pool, sometimes breaking up into droplets. The rate of expansion of holes in the sheet can be explained by a generalization of the Taylor-Culick theory, which is shown to hold independent of the fluid rheology.

We study hydrodynamic interactions and clustering mechanisms of active membrane inclusions within lipid bilayers. Pairs of inclusions display unique oscillatory dynamics that disappear when the 3D fluid adjacent to the membrane is confined. We reduce the governing equations to a coupled dynamical system whose phase behavior reveals the striking role of bulk confinement in enhancing cluster formation within the membrane. Using numerical simulations, we then extend this finding to demonstrate the role of confinement in controlling large-scale aggregation of membrane inclusions.

Surfactants - molecules and particles that preferentially adsorb to fluid interfaces - play a ubiquitous role in the fluids of industry, of nature, and of life. Since most surfactants cannot be seen directly, their behavior must be inferred from their impact on observed flows, like the buoyant rise of a bubble, or the thickness of a coating film. In so doing, however, a difficulty arises: physically distinct surfactant processes can affect measurable flows in qualitatively identical ways, raising the specter of confusion or even misinterpretation. This Perspective describes, in one coherent piece, both the equilibrium properties and dynamic processes of surfactants, to better enable the fluid mechanics community to understand, interpret, and design surfactant/fluid systems. Specifically, §2 treats the equilibrium thermodynamics of surfactants at interfaces, including surface pressure, isotherms of soluble and insoluble surfactants, and surface dilatational moduli (Gibbs and Marangoni). §3 describes surfactant dynamics in fluid systems, including surfactant transport and interfacial stress boundary conditions, the competition between surface diffusion, advection, and adsorption/desorption, Marangoni stresses and flows, and surface excess rheology. §4 discusses paradigmatic problems from fluid mechanics that are impacted by surfactants, including translating drops and bubbles, surfactant adsorption to clean and oscillating interfaces; capillary wave damping, thin film dynamics, foam drainage, and the dynamics of particles and probes at surfactant-laden interfaces. Finally, §5 discusses the additional richness and complexity that frequently arise in 'real' surfactants, including phase transitions, phase coexistence, and polycrystalline phases within surfactant monolayers, and their impact on non-Newtonian surface rheology.

The surface shear viscosity of an insoluble surfactant monolayer often depends strongly on its surface pressure. Here, we show that a particle moving within a bounded monolayer breaks the kinematic reversibility of low-Reynolds-number flows. The Lorentz reciprocal theorem allows such irreversibilities to be computed without solving the full nonlinear equations, giving the leading-order contribution of surface pressure-dependent surface viscosity. In particular, we show that a disc translating or rotating near an interfacial boundary experiences a force in the direction perpendicular to that boundary. In unbounded monolayers, coupled modes of motion can also lead to non-intuitive trajectories, which we illustrate using an interfacial analogue of the Magnus effect. This perturbative approach can be extended to more complex geometries, and to two-dimensional suspensions more generally.

The surface shear rheology of many insoluble surfactants depends strongly on the surface pressure (or concentration) of that surfactant. Here we highlight the dramatic consequences that surface-pressure-dependent surface viscosities have on interfacially dominant flows, by considering lubrication-style geometries within high Boussinesq (Bo) number flows. As with three-dimensional lubrication, high-Bo surfactant flows through thin gaps give high surface pressures, which in turn increase the local surface viscosity, further amplifying lubrication stresses and surface pressures. Despite their strong nonlinearity, the governing equations are separable, so that results from two-dimensional Newtonian lubrication analyses may be immediately adapted to treat surfactant monolayers with a general functional form of ηs(Π). Three paradigmatic systems are analyzed to reveal qualitatively new features: a maximum, self-limiting value for surfactant fluxes and particle migration velocities appears for Π-thickening surfactants, and kinematic reversibility is broken for the journal bearing and for suspensions more generally.

Three-dimensional numerical simulations are performed to study the stability of a sedimenting suspension of weakly flexible fibers. It is well known that a suspension of rigid rods sedimenting under gravity at low Reynolds number is unstable to concentration fluctuations owing to hydrodynamic interactions. Flexible fibers, however, reorient while settling and even weak flexibility can alter their collective dynamics. In our recent work [Manikantan et al., "The instability of a sedimenting suspension of weakly flexible fibres," J. Fluid Mech. 756, 935-964 (2014)], we developed a mean-field theory to predict the linear stability of such a system. Here, we verify these predictions using accurate and efficient particle simulations based on a slender-body model. We also demonstrate the mechanisms by which flexibility-induced reorientation alters suspension microstructure, and through it, its stability. Specifically, we first show that the anisotropy of the base state in the case of a suspension of flexible fibers has a destabilizing effect compared to a suspension of rigid rods. Second, a conflicting effect of flexibility is also shown to suppress particle clustering and slow down the growth of the instability. The relative magnitude of filament flexibility and rotational Brownian motion dictates which effect dominates, and our simulations qualitatively follow theoretically predicted trends. The mechanism for either effects is tied to the flexibility-induced reorientation of particles, which we illustrate using velocity and orientation statistics from our simulations. Finally, we also show that, in the case of an initially homogeneous and isotropic suspension, flexibility always acts to suppress the growth of the instability.

An analytical expression for the fluctuation-rounded stretch-coil transition of semiflexible polymers in extensional flows is derived. The competition between elasticity and tension is known to cause a buckling instability in filaments placed near hyperbolic stagnation points and the effect of thermal fluctuations on this transition has yet to receive full quantitative treatment. Motivated by the findings of recent experiments as well as our simulations, we solve for the amplitude of the first buckled mode near the onset of the instability. This reveals a stochastic supercritical bifurcation, which is in excellent agreement with full numerical simulations.

Suspensions of sedimenting slender fibres in a viscous fluid are known to be unstable to fluctuations of concentration. In this paper we develop a theory for the role of fibre flexibility in sedimenting suspensions in the asymptotic regime of weakly flexible bodies (large elasto-gravitation number). Unlike the behaviour of straight fibres, individual flexible filaments rotate as they sediment, leading to an anisotropic base state of fibre orientations in an otherwise homogeneous suspension. A mean-field theory is derived to describe the evolution of fibre concentration and orientation fields, and we explore the stability of the base state to perturbations of fibre concentration. We show that fibre flexibility affects suspension stability in two distinct and competing ways: the anisotropy of the base state renders the suspension more unstable to perturbations, while individual particle self-rotation acts to prevent clustering and stabilizes the suspension. In the presence of thermal noise, the dominant effect depends critically upon the relative scales of flexible fibre self-rotation compared to rotational Brownian motion.

The dynamics of a flexible filament sedimenting in a viscous fluid are explored analytically and numerically. Compared with the well-studied case of sedimenting rigid rods, the introduction of filament compliance is shown to cause a significant alteration in the long-time sedimentation orientation and filament geometry. A model is developed by balancing viscous, elastic and gravitational forces in a slender-body theory for zero-Reynolds-number flows, and the filament dynamics are characterized by a dimensionless elasto-gravitation number. Filaments of both non-uniform and uniform cross-sectional thickness are considered. In the weakly flexible regime, a multiple-scale asymptotic expansion is used to obtain expressions for filament translations, rotations and shapes. These are shown to match excellently with full numerical simulations. Furthermore, we show that trajectories of sedimenting flexible filaments, unlike their rigid counterparts, are restricted to a cloud whose envelope is determined by the elasto-gravitation number. In the highly flexible regime we show that a filament sedimenting along its long axis is susceptible to a buckling instability. A linear stability analysis provides a dispersion relation, illustrating clearly the competing effects of the compressive stress and the restoring elastic force in the buckling process. The instability travels as a wave along the filament opposite the direction of gravity as it grows and the predicted growth rates are shown to compare favourably with numerical simulations. The linear eigenmodes of the governing equation are also studied, which agree well with the finite-amplitude buckled shapes arising in simulations. © 2013 Cambridge University Press.

The dynamics and transport properties of Brownian semiflexible filaments suspended in a two-dimensional array of counter-rotating Taylor-Green vortices are investigated using numerical simulations based on slender-body theory for low-Reynolds-number hydrodynamics. Such a flow setup has been previously proposed to capture some of the dynamics of biological polymers in motility assays. A buckling instability permits elastic filaments to migrate across such a cellular lattice in a "Brownian-like" manner even in the athermal limit. However, thermal fluctuations alter these dynamics qualitatively by driving polymers across streamlines, leading to their frequent trapping within vortical cells. As a result, thermal fluctuations, characterized here by the persistence length, are shown to lead to subdiffusive transport at long times, and this qualitative shift in behavior is substantiated by the slow decay of waiting-time distributions as a consequence of trapping events during which the filaments remain in a particular cell for extended periods of time. Velocity and mass distributions of polymers reveal statistically preferred positions within a unit cell that further corroborate this systematic shift from transport to trapping with increasing fluctuations. Comparisons to results from a continuum model for the complementary case of rigid Brownian rods in such a flow also highlight the role of elastic flexibility in dictating the nature of polymer transport. © 2013 AIP Publishing LLC.
