Mercury is an important source of environmental contamination affecting human beings throughout the world and especially in the Amazon. Riverside populations have been chronically exposed to relatively high levels of methylmercury for many years. Long-term effects of mercury exposure are not well known, but human genotoxicity was already showed in both in vitro and epidemiological studies. However, to date, only two studies were carried out in Amazonian populations with conflicting results and without comparing to a non-exposed population. Aiming to highlight this question and avoid interference factors, this work analyzed in vitro genotoxicity of mercury in blood lymphocytes of Amazonian individuals by two methods (micronucleus and chromosomal aberrations). Deleterious effects of low levels (1-500 μg/l or 0,004-2 μM) of methylmercury were only detected with the method to detect chromosomal aberrations. Mitotic index (proportion of cells in metaphase) was the parameter most sensible. Thus, this technique was applied for the analysis of an Amazonian non-exposed population (Panacauera) with similar social-economical characteristics of the exposed populations studied elsewhere. The mean of the mitotic index for Panacauera population was 0.0814 ± 0.0097. Inter-individual variability of this index had no relation with sex or age. This value was above those registered for some groups of exposed populations. This fact points to mercury as the main responsible for inhibiting the cell cycle and/or the loss of proliferative capacity of the cells. These results already support mitotic index as an essential parameter for the early diagnose of mercury genotoxicity in humans, and especially in Amazonian populations., (Copyright © 2010 Elsevier Ltd. This paper presents a review about mercury contamination and human exposure in the Tapajós River basin (Brazil), one of the major tributaries of the Amazon impacted by traditional gold mining from the mid 1980s. The most recent review in this region was published more than ten years ago and since then many articles about environment and especially human populations have revealed new aspects of mercury toxicology. Additionally, new biomarkers of mercury exposure and toxicity have been studied in these populations. However, there are still many open, about both mercury's biogeochemical cycle and mercury health risks. Methylmercury is an important source of environmental contamination and the central nervous system (CNS) is one of the main target organs. Methylmercury genotoxicity was already demonstrated in peripherical tissues but was never detected in the brain. Thus, the objective of this work was to verify its genotoxic effect using brain cell lines. Glioblastoma (U373) and neuroblastoma (B103) human cell lines were exposed to methylmercury (0-10 microM). By measuring cellular viability, concentrations inducing <20% of cellular death (P<0.05) were selected: 1 and 0.1 microM. To detect micronuclei, 200,000 cells were treated with methylmercury for 24 h, and then incubated with cytochalasin B (2 microg/ml) for 72 h (U373) or 48 h (B103). The binucleation index, frequency of micronucleated cells, micronucleation index, metaphasic index and index of nucleoplasmic bridges were determined. Statistical analysis showed indices and percentages significantly higher (P<0.05) in methylmercury-treated cells. Each cell line was shown to be differently sensitive to each biomarker of genotoxic damage, which seems to indicate the existence of different mechanisms of toxicity. This work demonstrates, for the first time, MeHg ability to provoke genotoxicity in cells of brain origin with relatively low levels of exposure.

Mercury is a hazardous metal responsible for environmental contamination and human intoxication. Methylmercury, a very toxic organic compound, bio-accumulates through food chain, and is responsible for chronic mercury exposure of riverside Amazonian communities with a diet rich in fish. Uncertainties about the reference exposure dose that could have damaging consequences for nervous system development makes necessary the biomonitoring of these Amazonian populations, especially children. In this work, a comparative study was performed in exposed and non-exposed children living in the Amazon. A total of 168 children were analyzed to find possible correlations between gender, age, location, and hair mercury content. For each location, no statistically significant differences (P<0.05) were detected for gender and age versus mercury content. However, mean mercury levels in hair samples may indicate a tendency of boys to average higher hair concentrations. Also, in the community with highest levels of mercury, the limit of 10 micro g/g of mercury was surpassed by 65% of 2-6 years and 50% of 7-12 years children but only by 27% of 0-1 year babies, pointing to a lower bioaccumulation and/or the existence of a protection mechanism in babies. Log normal distributions of mercury concentrations for each location showed that children from populations under influence of gold mining activity contain the highest mercury levels in hair samples, though this intoxication may have decreased when compared to previous studies. Knowledge originated by this monitoring will better assist in the development of prevention strategies and government actions targeting the mercury contamination of Amazonian environment.

Cultured retina cells released accumulated [3H]GABA (gamma-aminobutyric acid) when stimulated by L-glutamate, N-methyl-D-aspartate (NMDA) and kainate. In the absence of Mg2+, dopamine at 200 microM (IC50 60 microM), inhibited in more than 50% the release of [3H]GABA induced by L-glutamate and NMDA, but not by kainate. This effect was not blocked by the D1-like dopamine receptor antagonist, R-(+)-7-chloro-8-hydroxy-3-methyl- -phenyl-2,3,4,5-tetrahydro- H-3-benzazepine hydrochloride (SCH 23390), neither by haloperidol nor spiroperidol (dopamine D2-like receptor antagonists). The dopamine D1-like receptor agonist R(+)-1-phenyl-2,3,4,5-tetrahydro-(1H)-3-benzazepine-7,diol hydrochloride (SKF 38393) at 50 microM, but not its enantiomer, also inhibited the release of [3H]GABA induced by NMDA, but not by kainate; an effect that was not prevented by the antagonists mentioned above. (+/-)-6-Chloro-7,8-dihydroxy-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepin e hydrobromide (SKF 812497) had no effect. Neither 8BrcAMP (5 mM) nor forskolin (10 microM) inhibited the release of [3H]GABA. Our results suggest that dopamine and (+)-SKF 38393 inhibit the glutamate and NMDA-evoked [3H]GABA release through mechanisms that seem not to involve known dopaminergic receptor systems.
