We have previously shown that the myristoylated alanine-rich C kinase substrate, a primary protein kinase C substrate in brain that binds and cross-links filamentous actin, is enriched in neuronal growth cones and is developmentally regulated in brain. Here we examined myristoylated alanine-rich C kinase substrate expression in the facial motor nucleus during axonal regeneration following facial nerve axotomy or facial nerve resection lesions, which impede regeneration, or following motor neuron degeneration induced by the retrograde neurotoxin ricin. For comparative purposes, the protein kinase C substrates myristoylated alanine-rich C kinase substrate-like protein and growth-associated protein-43 were examined in parallel. Myristoylated alanine-rich C kinase substrate messenger RNA exhibited a robust increase in both neurons and non-neuronal cells in the facial motor nucleus beginning four days after axotomy, peaked at seven days (2.5-fold), and declined back to baseline levels by 40 days. Myristoylated alanine-rich C kinase substrate protein similarly exhibited a twofold elevation in the facial motor nucleus determined four and 14 days post-axotomy. Following nerve resection, myristoylated alanine-rich C kinase substrate messenger RNA levels increased at seven days and returned to baseline levels by 40 days. Unlike myristoylated alanine-rich C kinase substrate messenger RNA, myristoylated alanine-rich C kinase substrate-like messenger RNA levels did not increase in the facial motor nucleus at any time point following nerve axotomy or resection, whereas growth-associated protein-43 messenger RNA exhibited a rapid (one day) and prolonged (40 days) elevation in facial motor nucleus neurons following either nerve axotomy or resection. Ricin-induced degeneration of facial motor neurons elevated myristoylated alanine-rich C kinase substrate and myristoylated alanine-rich C kinase substrate-like messenger RNAs in both microglia (lectin-positive) and astrocytes (glial fibrillary acidic protein-positive).Collectively, these data demonstrate that myristoylated alanine-rich C kinase substrate exhibits a unique expression profile in the facial motor nucleus following facial nerve lesions, and it is proposed that myristoylated alanine-rich C kinase substrate may serve to mediate actin-membrane cytoskeletal plasticity in both neurons and glial cells in response to protein kinaseC-mediated signaling during nerve regeneration and degeneration.

Rats exposed to chronic intake of sodium barbital maintained high circulating levels of barbital in blood and brain and exhibited increased sensitivity ot audiogenic convulsions during the withdrawal period. Levels of gamma-aminobutyric acid (GABA), glutamate, guanosine 3',5'-monophosphate (cyclic GMP) and adenosine 3',5'-monophosphate (cyclic AMP) were measured in selected brain regions after sacrifice with high power microwave inactivation. Cyclic GMP during chronic barbital administration was significantly lower than controls in most brain regions, especially the hindbrain. During the withdrawal period cyclic GMP in the cerebellum was significantly increased, while returning at least to control levels in all other regions. GABA throughout the brain tended to be reduced during barbital dependence, while cyclic AMP and glutamate levels remained unchanged in all groups. These results indicate a possible role for cyclic GMP in the mediation of the central nervous system response during barbiturate dependence and withdrawal.

Long Evans rats were exposed continuously to lead (1,000 ppm of lead acetate in the drinking water of dam or weanling) throughout gestation, lactation and until sacrifice at 60 days of age. A matched group of control animals was exposed to distilled water. On the day of sacrifice, control and lead treated animals were administered methylatropine 40 min. and oxotremorine (a muscarinic agonist) 10 min. prior to sacrifice by microwave irradiation. Locomotor activity was monitored during the 10 min. immediately preceding sacrifice. Levels of cyclic AMP and cyclic GMP were determined in 21 brain regions by radioimmunoassay. Locomotor activity following oxotremorine was significantly reduced in both lead treated and control animals. Levels of cyclic GMP were increased in septal region and frontal cortex and reduced in cerebellum and inferior colliculus of both groups of animals. The level of cyclic AMP in the pituitary increased 20-25 fold with a greater response noted in lead treated rats. Elevations of cyclic AMP were also noted in the interpeduncular nucleus, corpus striatum and substantia nigra in both groups. These data suggest that the cellular response to a muscarinic agonist was significantly affected by chronic exposure to lead in the pituitary but not in the remainder of the central nervous system.

The release of endogenous dopamine and norepinephrine from six brain regions (cortex, hippocampus, hypothalamus, striatum, s. nigra, n. accumbens-o. tubercle) of chronically lead-exposed rats was compared to release from controls. Prenatal and postnatal chronic lead intoxication was induced by 1000 ppm lead acetate in the drinking water of dams 70 days prior to mating and throughout gestation and lactation. Offspring were also maintained on 1000 ppm lead acetate until sacrificed at approximately 60 days of age. Release was measured in vitro from tissue minces prepared from both lead-exposed and control rats. There were no significant differences in spontaneous or potassium-stimulated norepinephrine release between lead-exposed and control rats. Potassium and amphetamine both stimulated striatal dopamine release; however, no marked differences between lead-exposed and control tissue were seen. Potassium-stimulated release of dopamine from the nucleus accumbens-olfactory tubercle area was increased in the lead-exposed animals. This is interesting in light of the postulated role of the n. accumbens in the central regulation of motor activity and the reported alterations in activity in lead-exposed rodents.
