Extracellular vesicles (EV), including exosomes and microvesicles, are nano-sized intercellular communication vehicles that participate in a multitude of physiological processes. Due to their biological properties, they are also promising candidates for the systemic delivery of therapeutic compounds, such as cytokines, chemotherapeutic drugs, siRNAs and viral vectors. However, low EV production yield and rapid clearance of administered EV by liver macrophages limit their potential use as therapeutic vehicles. We have used a hollow-fiber bioreactor for the efficient production of bioactive EV bearing the heterodimeric cytokine complex Interleukin-15:Interleukin-15 receptor alpha. Bioreactor culture yielded ∼40-fold more EV per mL conditioned medium, as compared to conventional cell culture. Biophysical analysis and comparative proteomics suggested a more diverse population of EV in the bioreactor preparations, while serum protein contaminants were detectable only in conventional culture EV preparations. We also identified the Scavenger Receptor Class A family (SR-A) as a novel monocyte/macrophage uptake receptor for EV. In vivo blockade of SR-A with dextran sulfate dramatically decreased EV liver clearance in mice, while enhancing tumor accumulation. Immunostimulatory properties of mushroom derived polysaccharides (PS) as stand-alone agents were tested. Next, PS were nanocomplexed with polyI:C (pIC) to yield stable nanoparticles around 200 nm in size evidenced by atomic force microscopy and dynamic light scattering analyses. PSs were selectively engaged by cells expressing TLR2 and initiated NFκB dependent signaling cascade leading to a Th1-biased cytokine/chemokine secretion in addition to bactericidal nitric oxide (NO) production from macrophages. Moreover, cells treated with nanoparticles led to synergistic IL6, production and upregulation of TNFα, MIP3α, IFNγ and IP10 transcript expression. In mice, PS-Ovalbumin-pIC formulation surpassed anti-OVA IgG responses when compared to either PS-OVA or pIC-OVA mediated immunity. Our results revealed that signal transduction initiated both by TLR2 and TLR3 via co-delivery of pIC by PS in nanoparticle depot delivery system is an effective immunization strategy. Naturally occurring, biocompatible, and biodegradable polyhydroxybutyrate-co-hydroxyvalerate (PHBV), and synthetic, non-degrading polyhydroxyethylmethacrylate (PHEMA) membranes were prepared and their mechanical properties were studied. Their performances were compared with the interpenetrating networks (IPN) prepared by photopolymerization of HEMA in the presence of PHBV. The modulus of elasticity, failure stress and failure strain indicated that the IPNs are viscoelastic with properties closer to PHEMA but much stronger than PHEMA homopolymers. Incorporation of PHBV (7, 14 and 22% HV) affected the mechanical properties positively. Increasing the PHBV content increased the modulus of elasticity and failure stress nearly in all samples tested. PHBV (7, 14, and 22% HV, 300 mg) samples showed an approximately 17-30 fold increase in terms of modulus of elasticity and 7-10 fold increase in terms of failure stress. The scanning electron micrographs of the membranes showed that the PHEMA membranes are more porous than the PHBV membranes but the IPN structure displayed channels on the membrane surface indicating that HEMA polymerization was achieved by using the PHBV as a scaffold. With the use of the present technique, it is possible to synthesize supramolecular structures from molecules that are not compatible and miscible with each other.
