Animal models are commonly used for drug screening before clinical trials. However, developing these models is time-consuming, and the results obtained from these models may differ from clinical outcomes due to the differences between animals and humans. To this end, 3D bioprinting offers several advantages for drug screening, such as high reproducibility and improved throughput, in addition to the human cells that can be used to generate these models. Here, we report the development of an animal patient-derived in vitro breast cancer model for drug screening using digital light processing (DLP) bioprinting. These bioprinted models demonstrated good cytocompatibility and preserved phenotypes of the cells. DLP enabled rapid fabrication with blood vessel-like channels to replicate, to a good extent, the tumor microenvironment. Our findings suggested that the improved microenvironment, provided by vascular structures within the bioprinted models, played a crucial role in reducing the chemoresistance of drugs. The fight against insect pests primarily relies on the utilization of synthetic insecticides. However, improper application of these chemicals can lead to detrimental effects on both the environment and human health, as well as foster the development of insect resistance. Consequently, novel strategies must be implemented to address the challenges stemming from the prolonged use of synthetic insecticides in agricultural and public health environments. Certain strategies involve the combination of crop protectants, which not only enhance insecticidal effectiveness but also reduce application rates. Plant-based natural products emerge as promising alternatives for insect management. Monoterpenes, which are abundant plant compounds produced through the activation of various enzymes, have attracted significant attention for their effectiveness in insect control. Notably, they are prolific in fragrance-producing plants. This review explores the plant defense, insecticidal, and antimicrobial characteristics of monoterpenes against insect pests, shedding light on their potential modes of action and possibilities for commercialization. Monoterpenoids are interesting hydrocarbons typically found in essential oils and have a significant role in medicinal and biological purposes. The goal of this study was to investigate the effects of two monoterpenoids, carvacrol (CAR) and menthol (MEN), supplemented with Moringa oleifera leaf meal (MOLM) based diets on growth parameters, digestibility and body composition of Nile tilapia ( Oreochromis niloticus ). Alongside the basal diet (control-T1), nine experimental diets supplemented with categorized levels of CAR and MEN at 200, 300 and 400 mg/kg individually and their mixtures (MIX) (1:1) (CAR-T2, 200; T3, 300; T4, 400 mg/kg, MEN-T5, 200; T6, 300; T7, 400 mg/kg and MIX- (1:1) T8, 200; T9, 300; T10, 400 mg/kg) were fed to the fingerlings (6.55 ± 0.03 g) for the period of 60 days. Monoterpenoids supplementation led to significantly ( p <0.05) better growth, feed utilization and nutrient digestibility in comparison to the control group. The highest growth, feed efficiency and nutrient digestibility were noticed in fishes fed with a diet supplemented with 200 mg/kg MIX. Interestingly, fishes fed with diets containing monoterpenoids had significantly higher levels of protein and ash, but with lower lipid in comparison to the control group. Conclusively, the dietary supplements like CAR and MEN improved the health status of Nile tilapia when given either individually or in a mixture,. Cancer and diabetes represent significant challenges in the field of biomedicine, with major and global impacts on public health. Acacia nilotica , commonly called 'gum arabic tree,' is recognized for its unique biomedical properties. The current study aimed to investigate the pharmacological potential of A. nilotica -based zinc-oxide nanoparticles (ZnO-NPs) in comparison to the ethanol and methanol-based extracts against cancer, diabetes, and oxidative stress. Green synthesis of ZnO-NPs was performed using barks of Acacia nilotica . Different techniques for the characterization of ZnO-NPs, including UV-Visible spectroscopy, Scanning Electron Microscopy, Fourier Transmission Infrared (FT-IR) spectroscopy, and X-ray Diffraction (XRD), were utilized. The morphological analysis of ZnO-NPs revealed that the fine NPs have mean particle sizes of 15 ± 1.5 nm. For the solvent based-extraction, leaves and barks were utilized and dissolved into ethanol and methanol for further processing. The MTT assay revealed that the optimum concentration of ZnO-NPs to inhibit the proliferation of liver cancer cell line HepG2 was 100 μg/mL where 67.0 % inhibition was observed; and both ethanol- and methanol-based extracts showed optimum inhibition at 100 μg/mL. The DPPH assay further demonstrated that 250 μg/mL of ZnO-NPs and 1000 μg/mL of both ethanol- and methanol-based extracts, as the optimum concentration for antioxidant activity (with 73.1 %, 68.9 % and 68.2 % inhibition respectively). The α-Glucosidase inhibition assay revealed that 250 μg/mL of ZnO-NPs and 10 μg/mL of both ethanol- and methanol-based extracts as the optimum concentration for antidiabetic activity (with 95 %, 93.7 % and 93.4 % inhibition respectively). The study provided interesting insights into the efficacy and reliability of ZnO-NPs for potential pharmacological application. Tamarindus indica , a beneficial herb, has many health benefits but there is limited research on its use in fish nutrition industry. The current study investigated the effects of incorporating extracts of T. indica into the canola meal-based diets of Cyprinus carpio (common carp); following which, the growth, digestibility, carcass and hematological markers were assessed. A total of six diets were formulated with varying concentrations of T. indica extracts (TIE) viz, 0 %, 0.5 %, 1 %, 1.5 %, 2 % and 2.5 %. The fish (N = 270, 15 fish/tank with triplicates) in each tank were fed experimental diets for 70 days. The study demonstrated that TIE supplementation significantly improved the growth of common carp when compared to 0 % TIE level (control). The best results were observed at 1 % TIE level for the specific growth rate (1.68 ± 0.03 %), weight gain (15.00 ± 0.57 g), and feed conversion ratio (1.36 ± 0.05). Conversely, the 2.5 % TIE level gave the least improvement in terms of growth performance. Specifically for nutrient digestibility, the maximum values of crude protein (CP, 67.60 ± 0.83 %), crude fat (CF, 67.49 ± 0.45 %) and gross energy (GE, 70.90 ± 0.56 %) were recorded at 1 % TIE level. In addition, the best results of body composition (protein: 63.92 ± 0.06 %, ash: 18.60 ± 0.03 %, fat: 7.12 ± 0.02 % and moisture: 10.36 ± 0.04 %) and hematological indices, were measured in carps fed with 1 % supplementation level. Endophytic bacteria, living inside plants, are competent plant colonizers, capable of enhancing immune responses in plants and establishing a symbiotic relationship with them. Endophytic bacteria are able to control phytopathogenic fungi while exhibiting plant growth-promoting activity. Here, we discussed the mechanisms of phytopathogenic fungi control and plant growth-promoting actions discovered in some major groups of beneficial endophytic bacteria such as Bacillus , Paenibacillus, and Pseudomonas . Most of the studied strains in these genera were isolated from the rhizosphere and soils, and a more extensive study of these endophytic bacteria is needed. It is essential to understand the underlying biocontrol and plant growth-promoting mechanisms and to develop an effective screening approach for selecting potential endophytic bacteria for various applications. We have suggested a screening strategy to identify potentially useful endophytic bacteria based on mechanistic phenomena. Exogenous application of iron (Fe) may alleviate salinity stress in plants growing in saline soils. This comparative study evaluated the comparative residual effects of iron nanoparticles (FNp) with two other Fe sources including iron-sulphate (FS) and iron-chelate (FC) on maize ( Zea mays L.) crop grown under salt stress. All three Fe sources were applied at the rate of 15 and 25 mg/kg of soil before the sowing of wheat (an earlier crop; following the sequence of crop rotation) and no further Fe amendments were added later for the maize crop. Results revealed that FNp application at 25 mg/kg (FNp-2) substantially increased maize height, root length, root dry weight, shoot dry weight, and grain weightby 80.7%, 111.1%, 45.7%, 59.5%, and 77.2% respectively, as compared to the normal controls; and 62.6%, 81.3%, 65.1%, 78%, and 61.2% as compared to salt-stressed controls, respectively. The FNp-2 treatment gave higher activities of antioxidant enzymes, such as superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase compared to salt stressed control (50.6%, 51%, 48.5%, and 49.2%, respectively). The FNp-2 treatment also produced more photosynthetic pigments and better physiological markers: higher chlorophyll a contents by 49.9%, chlorophyll b contents by 67.2%, carotenoids by 62.5%, total chlorophyll contents by 50.3%, membrane stability index by 59.1%, leaf water relative contents by 60.3% as compared to salt stressed control. The highest Fe and Zn concentrations in maize roots, shoots, and grains were observed in FNp treatment as compared to salts stressed control. Higher application rates of Fe from all the sources also delivered better outcomes in alleviating salinity stress in maize compared to their respective low application rates. Extreme hot conditions during summers, high poverty rate and continuous electricity load shedding affect commercial manufacturing and sale of ice in many countries. The vendors prepared ice using untreated piped water, tanker water and ground water. These waters may contain hazardous pollutants and ice made from them will pose a potential human health risk. Thus, it is important to regularly monitor the chemical composition of water sources and the quality of the manufactured ice. A contemporary examination was carried out to evaluate the physico-chemical properties and heavy metals and metalloids in the ice sold in all the districts of Karachi, Pakistan. This pioneering study was an innovative effort to assess the ice quality in relation to potential pollutant hazards to human health; with concomitant geospatial information. The geospatial distribution of ice quality and major constituents were among the measured parameters; carefully associated with further geospatial information, determined using GIS (Geographic Information Systems) and PCA (Principal Component Analysis) techniques. Interestingly, the physico-chemical analyses revealed that the ice quality was marginally adequate and the total mean metal-metalloid contents were in the sequence of Pb > Ni > Zn > Fe > Cr > As. The concentrations of these metals were above the upper allowable limits with reference to the recommended WHO guidelines. We observed that 57.1% and 35.7% ice samples had good physico-chemical properties assessed using the Ice Quality Index (IQI). Conversely, the IQI for metals showed that the ice was unsafe for human consumption. In terms of health risk assessment, the overall mean CDI (Chronic Daily Intake) and HQ (Hazard Quotient) values were in the order of Pb () > Ni (3.2) > Zn (2.3) > Fe (2.1) > Cr (1.6) > As (0.5) and Pb (7.4) > As (1.7) > Cr (0.5) > Ni (0.4 > Zn (0.008) > Fe (0.003), respectively. High protein content, excellent amino acid profile, absence of anti-nutritional factors (ANFs), high digestibility and good palatability of fishmeal (FM), make it a major source of protein in aquaculture. Naturally derived FM is at risk due to an increase in its demand, unsustainable practices, and price. Thus, there is an urgent need to find affordable and suitable protein sources to replace FM. Plant protein sources are suitable due to their widespread availability and low cost. However, they contained certain ANFs, deficiency of some amino acids, low nutrient bioavailability and poor digestibility due to presence of starch and fiber. These unfavourable characteristics make them less suitable for feed as compared to FM. Thus, these potential challenges and limitations associated with various plant proteins have to be overcome by using different methods, i.e. enzymatic pretreatments, solvent extraction, heat treatments and fermentation, that are discussed briefly in this review. This review assessed the impacts of plant products on growth performance, body composition, flesh quality, changes in metabolic activities and immune response of fishes. To minimize the negative effects and to enhance nutritional value of plant products, beneficial functional additives such as citric acid, phytase and probiotics could be incorporated into the plant-based FM. Interestingly, these additives improve growth of fishes by increasing digestibility and nutrient utilization of plant based feeds. The contamination of farm soils with heavy metals (HMs) has raised significant concerns due to the increased bioavailability and accumulation of HMs in agricultural food crops. To address this issue, a survey experiment was conducted in the suburbs of Multan and Faisalabad to investigate the spatial distribution, bioaccumulation, translocation, and health risks of cadmium (Cd) and lead (Pb) in agricultural crops. The results show a considerable concentration of Cd and Pb in soils irrigated with wastewater, even though these levels were below the permissible limits in water and soil matrices. The pollution index for Cd was mostly greater than 1 at the selected sites, indicating its accumulation in soil over time due to wastewater irrigation. Conversely, the pollution index for Pb was below 1 at all sites. Among the plants, Zea mays accumulated the highest concentration of Cd and Pb. The translocation factor from soil to root was highest for Brassica olearecea (7.037 for Cd) and Zea mays (6.383 for Pb). The target hazard quotient (THQ) value of Cd exceeded the non-carcinogenic limit for most vegetables. The highest value was found in Allium cepa (5.256) and the lowest in Allium sativum (0.040). In contrast, the THQ level of Pb was below the non-carcinogenic limit for most vegetables, except for Allium cepa (1.479), Solanum lycopersicum (1.367), and Solanum tuberosum (1.326). The study highlights that Allium cepa poses the highest health risk for humans, while Medicago sativa poses the highest risk for animals due to Cd and Pb contamination. A sustainable and resilient crop production system is essential to meet the global food demands. Traditional chemical-based farming practices have become ineffective due to increased population pressures and extreme climate variations. Recently, nanobiotechnology is considered to be a promising approach for sustainable crop production by improving the targeted nutrient delivery, pest management efficacy, genome editing efficiency, and smart plant sensor implications. This review provides deeper mechanistic insights into the potential applications of engineered nanomaterials for improved crop stress resilience and productivity. We also have discussed the technology readiness level of nano-based strategies to provide a clear picture of our current perspectives of the field. Current challenges and implications in the way of upscaling nanobiotechnology in the crop production are discussed along with the regulatory requirements to mitigate associated risks and facilitate public acceptability in order to develop research objectives that facilitate a sustainable nano-enabled Agri-tech revolution. The SARS COV-2 and its variants are spreading around the world at an alarming speed, due to its higher transmissibility and the conformational changes caused by mutations. The resulting COVID-19 pandemic has imposed severe health consequences on human health. Several countries of the world including Pakistan have studied its genome extensively and provided productive findings. In the current study, the mCSM, DynaMut2, and I-Mutant servers were used to analyze the effect of identified mutations on the structural stability of spike protein however, the molecular docking and simulations approaches were used to evaluate the dynamics of the bonding network between the wild-type and mutant spike proteins with furin. We addressed the mutational modifications that have occurred in the spike protein of SARS-COV-2 that were found in 215 Pakistani's isolates of COVID-19 patients to study the influence of mutations on the stability of the protein and its interaction with the host cell. We found 7 single amino acid substitute mutations in various domains that reside in spike protein. The H49Y, N74K, G181V, and G446V were found in the S1 domain while the D614A, V622F, and Q677H mutations were found in the central helices of the spike protein. Based on the observation, G181V, G446V, D614A, and V622F mutants were found highly destabilizing and responsible for structural perturbation. Protein-protein docking and molecular simulation analysis with that of furin have predicted that all the mutants enhanced the binding efficiency however, the V622F mutant has greatly altered the binding capacity which is further verified by the K D value (7.1 E -14 ) and therefore may enhance the spike protein cleavage by Furin and increase the rate of infectivity by SARS-CoV-2. On the other hand, the total binding energy for each complex was calculated which revealed -50.57 kcal/mol for the wild type, for G181V -52.69 kcal/mol, for G446V -56.44 kcal/mol, for D614A -59.78 kcal/mol while for V622F the TBE was calculated to be -85.84 kcal/mol. The purpose of this research was to conduct a scientometric evaluation of the literature pertaining to plastic sand in order to evaluate its many aspects. Conventional review studies have several limitations when it comes to their capacity to completely and properly link different sections of the published research. Some of the more complicated features of advanced research are co-occurrence analysis, science mapping and co-citation analysis. During the study, the most inventive authors/researchers renowned for citations, the sources with the largest number of publications, the actively involved domains, and co-occurrences of keywords in the research on plastic sand are investigated. This study is limited to scientometric analysis of the available literature data on plastic sand. The VOSviewer application (version 1.6.18) was used to perform the analysis after bibliometric data for 4512 publications were extracted from the Scopus database and utilised in the extraction process from the year 2021 to June 2022. COVID-19 caused online buying channels to flourish across the globe. However, the extent to which online channels in Pakistan assisted peoples in coping with the pandemic remains unknown. This study aims to examine peoples behavior and perceptions regarding online food purchasing and its impact on different aspects of food security. The data were collected through online surveys of 1067 respondents in Punjab and Sindh provinces during the COVID-19 pandemic. We find that peoples access to food was adversely affected by the pandemic. However, people are increasingly purchasing food online, which has improved their accessibility to food. According to the findings, 62.51% of respondents reported to have changed their perception and behavior regarding online food purchasing. In addition, almost 46.40% of peoples reported that online shopping increased their access to food during the pandemic. Using logit regression, economic analysis shows that education, monthly income, and access to basic necessities such as clean drinking water, better sanitation, and better employment are positively related to online buying behavior.
