Using sequential electron-beam evaporation of high-temperature dielectric (alumina ceramic) and magnetic (iron) targets in various gas atmospheres (helium, air, and oxygen) in medium vacuum (5–8 Pa), magneto-dielectric coatings with thickness of around 2 μm were deposited from a multicomponent beam plasma at a deposition rate of 0.2–0.3 μm/min. The coating magnetic properties were explored by the ferromagnetic resonance technique, revealing that their effective magnetization depends on the type of operating gas and varied from 4.2 to 6.8 kGs (for deposition in helium) to 0.3 kGs (in oxygen), which is characteristic of oxide ferromagnetic materials and is considerably lower than the corresponding value (∼22 kGs) for thin iron films formed by vacuum arc deposition in high vacuum. X-ray structural analysis of coatings deposited in medium vacuum in helium showed that the magnetic layer has a magnetite (Fe3O4) structure. The alumina ceramic layer provides the dielectric properties of the magneto-dielectric coating; a relative dielectric constant of 6.0 and a conductivity of 8.4 mS/m were achieved.

Objective Compression stockings and bandages are widely used after invasive treatment of varicose veins. The goals of compression after venous interventions are to reduce pain, bruising, and ecchymosis. Nevertheless, patients often report discomfort with the compression. To make postprocedural compression more tolerable, foot-sparing bandages were tested in a randomized clinical trial of noninferiority. Methods A total of 187 patients were randomized to use class II foot-sparing compression sleeves for the full leg or class II stockings after radiofrequency ablation with concomitant phlebectomy. The primary endpoint was the quality of life, measured using the Chronic Venous Disease Quality of Life Questionnaire 20-item scale 30 days after intervention. The secondary endpoints were pain in the leg and discomfort related to the compression garment, which were assessed using the visual analog scale (VAS) at 2, 7, 14, and 30 days. Results The global index score of the questionnaire was 66.1 and 70.6 and 83.8 and 87.7 for the sleeve and stocking groups before and 30 days after intervention, respectively (P = .542 and P = .150, respectively). The VAS for pain score in the operated leg was slightly higher in the sleeve group the day after the intervention (score, 2.1 vs 1.6; P = .03). At 7, 14, and 30 days, the VAS for pain scores did not differ significantly (score, 0.7 vs 0.5; 0.5 vs 0.3; and 0.1 vs 0.1, respectively; P = NS for all). The VAS for discomfort score was not significantly different statistically in the study group at 2 days (sleeve, 1.9; vs stocking, 1.4; P = .08) but was higher after 7 days (sleeve, 0.9; vs stocking, 0.6; P = .008). No difference in discomfort was found between the study and control groups at 14 or 30 days (sleeve, 0.6; vs stocking, 0.4; and sleeve, 0.4; vs stocking, 0.4, respectively; P = NS for both). Conclusions Quality of life after thermal ablation with phlebectomy improved equivalently in patients who had used class II compression sleeves for full legs and those who had used class II compression stockings. Pain and discomfort were slightly higher in the sleeve group.

Boron-containing films are promising materials for improving the hardness, wear-resistance and durability of mechanical parts used in industry. We describe a novel approach for deposition of such coatings – by using electron-beam evaporation of boron-containing target material (pure boron or boron nitride) in inert (helium) or chemically-active (oxygen, nitrogen, residual atmosphere) gases at medium vacuum. Boron-containing films deposited on titanium substrates were characterized using a nanohardness tester and a non-contact 3D profilometer. Elemental composition of the coatings was studied using raster electron microscope equipped with a device for EDX analysis. All boron-containing coatings provided significant improvement (by a factor of 4–16) in the microhardness of the substrate surface.

Adeno-associated virus (AAV) vectors are widely used in clinical gene therapy to correct genetic disease by in vivo gene transfer. Although the vectors are useful, in part because of their limited immunogenicity, immune responses directed at vector components have complicated applications in humans. These include, for instance, innate immune sensing of vector components by plasmacytoid dendritic cells (pDCs), which sense the vector DNA genome via Toll-like receptor 9. Adaptive immune responses employ antigen presentation by conventional dendritic cells (cDCs), which leads to cross-priming of capsid-specific CD8(+) T cells. In this study, we sought to determine the mechanisms that promote licensing of cDCs, which is requisite for CD8(+) T cell activation. Blockage of type 1 interferon (T1 IFN) signaling by monoclonal antibody therapy prevented cross-priming. Furthermore, experiments in cell-type-restricted knockout mice showed a specific requirement for the receptor for T1 IFN (IFNaR) in cDCs. In contrast, natural killer (NK) cells are not needed, indicating a direct rather than indirect effect of T1 IFN on cDCs. In addition, co-stimulation by CD4(+) T cells via CD40-CD40L was required for cross-priming, and blockage of co-stimulation but not of T1 IFN additionally reduced antibody formation against capsid. These mechanistic insights inform the development of targeted immune interventions.

Purpose: To investigate age-related differences in outcomes of critically ill patients with sepsis around the world., Methods: We performed a secondary analysis of data from the prospective ICON audit, in which all adult ( >16 years ) patients admitted to participating ICUs between May 8 and 18, 2012, were included, except admissions for routine postoperative observation. For this sub-analysis, the 10,012 patients with completed age data were included. They were divided into five age groups - 80 years. Sepsis was defined as infection plus at least one organ failure., Results: A total of 2963 patients had sepsis, with similar proportions across the age groups (80 = 30.9%). Hospital mortality increased with age and in patients >80 years was almost twice that of patients 70 years was independently associated with increased risk of dying., Conclusions: The odds for death in ICU patients with sepsis increased with age with the maximal rate of increase occurring between the ages of 71 and 77 years.
