open access article Insulating glass flat-plate collectors can save cost by being produced quickly and automatically in insulated glass production facilities, and they can be filled with argon to reduce heat loss. During its lifetime, the collector is likely to lose argon because of gradual material degradation of the sealing. However, information on the influence of the argon concentration on the collector efficiency is limited. Therefore, the objective of this research work was to analyse this effect. A theoretical material property calculation of argon-air mixtures was carried out to determine the convective losses with variable argon concentrations. Thermal collector performance was measured experimentally using an outdoor solar tracker test rig. The results strongly suggest, that the influence of argon concentration on both the convective losses and the thermal efficiency is non-linear. The measurements revealed that an argon concentration of 90 % can increase average thermal performance by percentage points. An increase in argon concentration from 0 % to 50 % has almost twice the effect on average thermal efficiency as an increase from 50 % to 90 %. Concluding from these results, an argon leakage threshold of 2.5 percentage points per year is proposed to avoid disproportionate loss of efficiency over time.

In this paper, we augment a model of endogenous credit cycles by Matsuyama et al. (2016) with human capital to study its impact on the stability of income evolution and its distribution. Human capital is modelled as pure external effect of production, following a learning-by-producing approach. Agents have access to two different investment projects, which differ substantially in their next generations spillover effects. Some generate pecuniary externalities and technological spillovers through human capital formation whereas others fail to do so. Moreover, the latter are subject to financial frictions. Together with the interaction between those projects, endogenous credit cycles occur and a pattern of boom and bust cycles can be observed. We explore the impact of human capital on the systems stability by providing analytical results and numerical simulations, which confirm a strong interaction between credit market frictions and the importance of human capital in the production process. In general, we found that an increasing importance of human capital has an ambiguous effect on the income evolution. Especially during transition periods, we observe a destabilising effect while with high human capital importance, the income path is stabilised. Moreover, an increase of the importance of human capital favours investment projects with positive intergenerational spillovers, thus driving up the labour income of the young generation. Thus, our study reveals that human capital is an essential factor for economic stability under the presence of financial frictions and has substantial positive effects on the income distribution.

This paper considers Bayesian estimation of the threshold vector error correction (TVECM) model in moderate to large dimensions. Using the lagged cointegrating error as a threshold variable gives rise to additional difficulties that typically are solved by utilizing large sample approximations. By relying on Markov chain Monte Carlo methods, we are enabled to circumvent these issues and avoid computationally-prohibitive estimation strategies like the grid search. Due to the proliferation of parameters, we use novel global-local shrinkage priors in the spirit of Griffin and Brown (2010). We illustrate the merits of our approach in an application to five exchange rates vis-a-vis the US dollar by means of a forecasting comparison. Our findings indicate that adopting a non-linear modeling approach improves the predictive accuracy for most currencies relative to a set of simpler benchmark models and the random walk.

44Sc is an increasingly investigated positron emitter for use in positron emission tomography (PET) imaging. However, 44Sc is a non-pure positron emitter, since prompt photons are co-emitted during the decay process. This study investigates coincidence energy spectra of 44Sc and its impact on PET quantification on a preclinical and clinical PET system in comparison with 18F. The raw data of the coincidence events revealed characteristic differences comparing the photon energy distribution of 44Sc and 18F. Due to prompt gamma emission of 44Sc, activity recovery is underestimated on PET systems. However, clinical PET imaging of 44Sc with acceptable quantitative accuracy appears feasible by using a single, constant correction factor.

We empirically investigate the role of central banks in the context of heterogeneous labor markets, jobless recoveries and job polarization. Specifically, we estimate the effect of monetary policy on the US labor market using disaggregated time series based on large scale survey data. The impact of interest rate changes on unemployment in 32 occupation groups is explored in a Bayesian factor-augmented vector autoregression framework. The results suggest largely heterogeneous impacts across various occupation groups. This heterogeneity can be explained by differential task profiles of the workers in their respective occupations. Workers with tasks that are easily automated or offshored as well as workers at the bottom of the skill distribution are disproportionately affected following a monetary policy shock. This implies that labor market participants that are highly vulnerable to structural developments such as skill-biased technological change and the globalization of labor markets are also most sensitive to conventional monetary policy measures. From a policy perspective, we conclude that central banks are unlikely to be able to take on a stabilizing role in the context of labor market polarization.

Gait dysfunction is common in patients with multiple sclerosis (PwMS). Treatment with prolonged-release fampridine (PR-fampridine) improves walking ability in some PwMS. Associated fampridine-induced changes in the walking pattern are still poorly understood but may provide a better understanding of the mechanisms underlying the beneficial drug effects. 61 PwMS were treated with PR-fampridine in a randomized, monocentric, double-blind and placebo-controlled clinical trial with crossover design (FAMPKIN). Drug-induced improvements in walking speed (Timed-25-Foot Walk; T25FW) and endurance (6-Minute Walk Test; 6MWT) were quantified. In this sub-study of the FAMPKIN trial, fampridine-induced changes in kinetic gait patterns were analyzed by pressure-based foot print analysis during treadmill walking. Vertical ground reaction forces were analyzed during different gait phases. Kinetic data of 44 PwMS was eligible for analysis. During double-blind treatment with PR-fampridine, patients performed significantly better in the T25FW and 6MWT than during placebo treatment (p < 0.0001 for both). At the group level (n = 44), there were no significant changes of gait kinetics under PR-fampridine vs. placebo. However, we found relevant changes of walking kinetics regarding forces during loading, single limb and pre-swing phase in a patient sub-group (n = 8). Interestingly, this sub-group demonstrated superior responsiveness to PR-fampridine in the clinical walking tests compared to those patients without any fampridine-induced changes in kinetics (n = 36). Our results demonstrate fampridine-induced changes in gait kinetics in a sub-group of PwMS. These gait pattern changes were accompanied by improved clinical walking performance under PR-fampridine. These results shed some light on the biomechanical changes in walking patterns underlying enhanced fampridine-induced gait performance.

Three unexpected products derived from the unsymmetrically substituted amidinate anion [MeC(NEt)(NtBu)]− are reported. Treatment of 1-tert-butyl-3-ethyl-carbodiimide, tBu-N C N–Et, with 1 equiv. of methyllithium in diethyl ether afforded colorless crystals of the unusual octanuclear lithium amidinate derivative {[LiN(Et)C(Me)N(tBu)]2⋅LiN(Et)C(CH2Li)N(tBu) Et2O}2 (1) in 57% yield instead of the expected lithium amidinate Li[MeC(NEt)(NtBu)]. In the molecule of 1, one central methyl group in a trimer of Li[MeC(NEt)(NtBu)] is deprotoned and two of these partially deprotonated trimers are linked through a central eight-membered [C–N–Li–CH2]2 ring to afford the octanuclear product 1. The same reaction carried out in 1,2-dimethoxyethane (DME) solution produced the oxygen-centered, heptanuclear lithium amidinate cluster {Li[MeC(NEt)(NtBu)]}5⋅Li2O (2) (21% yield). Treatment of anhydrous praseodymium trichloride with in situ-prepared Li[MeC(NEt)(NtBu)] in Et2O/THF afforded green (THF)Li[Pr{MeC(NEt)(NtBu)}4] (3) as the first anionic tetrakis(amidinato)lanthanide(III) complex in 55% isolated yield. The molecular and crystal structures of all three new compounds have been elucidated by X-ray diffraction.

Uncontrolled muscle spasms often develop after spinal cord injury. Structural and functional maladaptive changes in spinal neuronal circuits below the lesion site were postulated as an underlying mechanism but remain to be demonstrated in detail. To further explore the background of such secondary phenomena, rats received a complete sacral spinal cord transection at S(2) spinal level. Animals progressively developed signs of tail spasms starting 1 week after injury. Immunohistochemistry was performed on S(3/4) spinal cord sections from intact rats and animals were sacrificed 1, 4 and 12 weeks after injury. We found a progressive decrease of cholinergic input onto motoneuron somata starting 1 week post-lesion succeeded by shrinkage of the cholinergic interneuron cell bodies located around the central canal. The number of inhibitory GABAergic boutons in close contact with Ia afferent fibers was greatly reduced at 1 week after injury, potentially leading to a loss of inhibitory control of the Ia stretch reflex pathways. In addition, a gradual loss and shrinkage of GAD65 positive GABAergic cell bodies was detected in the medial portion of the spinal cord gray matter. These results show that major structural changes occur in the connectivity of the sacral spinal cord interneuronal circuits below the level of transection. They may contribute in an important way to the development of spastic symptoms after spinal cord injury, while reduced cholinergic input on motoneurons is assumed to result in the rapid exhaustion of the central drive required for the performance of locomotor movements in animals and humans.

The climate specific effectiveness of solar DEC-systems and relevant preferable adaptions of the system design have been so far predominantly analysed based on individual simulation for selected sites. In order to systematically deduce design-specific outline data for the application of the solar DEC-technology at climatically different sites a methodological zoning approach was further developed. A subsequent meteorological analysis for 17 sites mapping the world climate creates a transparent understanding on the activity of the specific system components. This results in a comprehension on the site-specific effectiveness of solar DEC-systems and provides insights on principally relevant and efficient system configurations.

The research project ‘Solar Heat in the Liquid Food Industry’ (part of the FORETA Research Network [1] ) focused on the development and optimisation of low-temperature heating systems for the liquid food industry. Its main objectives are in energy efficiency, waste heat recovery and the feasibility of a solar-thermal process heating system. Based on the particular simulation results the overall solar-thermal potentials for German breweries and dairies were determined. In this connection a literature review indicated that most of the potential studies on solar-thermal energy use in the industry or related to specific industrial sectors are based on the total use of low-temperature heat. In opposition to these results, the available and mainly limited roof area was found to be a more important aspect for the solar-thermal potential. Hence, the development of a methodology for a site- specific analysis was necessary. The interconnection of only a few defined evaluation criteria resulted in a more realistic estimation of the potential for a solar-thermal heat supply.

Restricted structural re-growth in the adult CNS is a major limitation to fully functional recovery following extensive CNS trauma. This limitation is partly due to the presence of growth inhibitory proteins, in particular, Nogo-A. Pre-clinical studies have demonstrated that intrathecally infused anti-Nogo-A antibodies are readily distributed via the cerebrospinal fluid penetrating throughout the spinal cord and brain, where they promote sprouting, axonal regeneration and improved functional recovery after CNS injury. Whether anti-Nogo-A treatments of intact animals might induce behavioral alterations has not been systematically tested. This is addressed here in an adult rat model of chronic intrathecal infusion of function-blocking anti-Nogo-A antibodies for 2 to 4weeks. We observed by proteomic and immunohistochemical techniques that chronic Nogo-A neutralization in the intact CNS increased expression of cytoskeletal, fiber-growth-related, and synaptic proteins in the hippocampus, a brain region which might be particularly sensitive to Nogo-A depletion due to the high expression level of Nogo-A. Despite such molecular and proteomic changes, Nogo-A blockade was not associated with any pronounced cognitive-behavioral changes indicative of hippocampal functional deficiency across several critical tests. Our results suggest that the plastic changes induced by Nogo-A blockade in the adult hippocampus are counter-balanced by homeostatic mechanisms in the intact and the injured CNS. The data indicate that anti-Nogo-A therapy appears safe in the adult CNS over 4weeks of continuous administration.

Pentafluorobenzyl (PFB) bromide (PFB-Br) is a versatile derivatization reagent for numerous classes of compounds. Under electron-capture negative-ion chemical ionization (ECNICI) conditions PFB derivatives of acidic compounds readily and abundantly ionize to produce intense anions due to [M-PFB](-). In the present article we investigated the PFB-Br derivatization of unlabelled acetaminophen (N-acetyl-p-aminophenol, NAPAP-d(0); paracetamol; MW 151) and tetradeuterated acetaminophen (NAPAP-d(4); MW 155) in anhydrous acetonitrile and their GC-ECNICI-MS behavior using methane as the buffer gas. In addition to the expected anions [M-PFB](-) at m/z 150 from NAPAP-d(0) and m/z 154 from NAPAP-d(4), we observed highly reproducibly almost equally intense anions at m/z 149 and m/z 153, respectively. Selected ion monitoring of these ions is suitable for specific and sensitive quantification of acetaminophen in human plasma and urine. Detailed investigations suggest in-source formation of N-acetyl-p-benzoquinone imine (NAPQI; MW 149), the putatively toxic acetaminophen metabolite, from the PFB ether derivative of NAPAP. GC-ECNICI-MS of non-derivatized NAPAP did not produce NAPQI. The peak area ratio of m/z 149 to m/z 150 and of m/z 153 to m/z 154 decreased with increasing ion-source temperature in the range 100-250°C. Most likely, NAPQI formed in the ion-source captures secondary electrons to become negatively charged (i.e., [NAPQI](-)) and thus detectable. Formation of NAPQI was not observed under electron ionization (EI) conditions, i.e., by GC-EI-MS, from derivatized and non-derivatized NAPAP. NAPQI was not detectable in flow injection analysis LC-MS of native NAPAP in positive electrospray ionization (ESI) mode, whereas in negative ESI mode low extent NAPQI formation was observed (5%). Our results suggest that oxidation of drug derivatives in the ion-sources of mass spectrometers may form intermediates that are produced from activated drugs in enzyme-catalyzed reactions.

A measurement setup combined with a Finite Element (FE) simulation is presented to determine the elasticity modulus of soft materials as a function of frequency. The longterm goal of this work is to measure in vitro the elasticity modulus of human vocal folds over a frequency range that coincides with the range of human phonation. The results will assist numerical simulations modeling the phonation process by providing correct material parameters. Furthermore, the measurements are locally applied, enabling to determine spatial differences along the surface of the material. In this work the method will be presented and validated by applying it to silicones with similar characteristics as human vocal folds. Three silicone samples with different consistency were tested over a frequency range of 20-250 Hz. The results of the pipette aspiration method revealed a strong frequency dependency of the elasticity modulus, especially below 100 Hz. In this frequency range the elasticity moduli of the samples varied between 5 and 27 kPa.

The mucin MUC1 is a candidate for use in specific immunotherapy against breast cancer, but this requires the large-scale production of a MUC1 antigen. In this study, a bioprocess for the expression of a recombinant MUC1 fusion protein with a cancer associated glycosylation in CHO-K1 cells has been developed. Cells permanently expressing parts of the extracellular portion of MUC1 fused to IgG Fc were directly transferred from adherent growth in serum-containing medium to suspension culture in the protein-free ProCHO4-CDM culture medium. Using the Cellferm-pro system, optimal culture parameter as pH and pO(2) were determined in parallel spinner flask batch cultures. A pH of 6.8-7.0 and a pO(2) of 40% of air saturation was found to give best cell growth and productivity of secreted recombinant protein. Specific productivity strongly depended the pO(2) and correlated with the online monitored oxygen uptake rate (OUR) of the cells, which indicates a positive influence of the rate of oxidative phosphorylation on productivity. The optimised conditions were applied to continuous perfusion culture which gave very high cell densities and space time yields of the recombinant MUC1 fusion protein, allowing production at gram scale. The product degradation was much lower in supernatants from continuous perfusion culture compared to batch mode. Antibodies reacting with cancer associated MUC1 glycoforms strongly bound to the fusion protein, indicating that the desired glycoforms were obtained and suggesting that the recombinant MUC1 protein could be tested for use in immunotherapy.

The "neurotrophin hypothesis" of depression predicts that depressive disorders in humans coincide with a decreased activity and/or expression of brain-derived neurotrophic factor (BDNF) in the brain. Therefore, we investigated whether mice with a reduced BDNF expression due to heterozygous gene disruption demonstrate depression-like neurochemical changes or behavioral symptoms. BNDF protein levels of adult BDNF(+/-) mice were reduced to about 60% in several brain areas investigated, including the hippocampus, frontal cortex, striatum, and hypothalamus. The content of monoamines (serotonin, norepinephrine, and dopamine) as well as of serotonin and dopamine degradation products was unchanged in these brain regions. By contrast, choline acetyltransferase activity was significantly reduced by 19% in the hippocampus of BDNF(+/-) mice, indicating that the cholinergic system of the basal forebrain is critically dependent on sufficient endogenous BDNF levels in adulthood. Moreover, BDNF(+/-) mice exhibited normal corticosterone and adrenocorticotropic hormone (ACTH) serum levels under baseline conditions and following immobilization stress. In a panel of behavioral tests investigating locomotor activity, exploration, anxiety, fear-associated learning, and behavioral despair, BDNF(+/-) mice were indistinguishable from wild-type littermates. Thus, a chronic reduction of BDNF protein content in adult mice is not sufficient to induce neurochemical or behavioral alterations that are reminiscent of depressive symptoms in humans.

In vitro pituitary adenylate cyclase activating polypeptide (PACAP) induces the expression of brain-derived neurotrophic factor (BDNF) via its specific receptor PAC1. Since BDNF has been implicated in learning paradigms and mice lacking functional PAC1 have deficits in hippocampus-dependent associative learning, we investigated whether PAC1 mutants show alterations in hippocampal expression of BDNF and its receptor TrkB. Semi-quantitative in situ-hybridization using exon-specific BDNF-probes revealed significantly reduced expression of the exon-III and exon-V-specific transcripts within the hippocampal CA3 region in PAC1-deficient mice. A similar trend was observed for the exon-I-specific transcript. The expression of the exon-III-specific transcript was also reduced within the dentate gyrus, while Trk B-expression did not differ between genotypes. Our data demonstrate that even in vivo PAC1-mediated signaling seems to play a pivotal role for the transcriptional regulation of BDNF.

Low temperature multilayer insulation systems are highly efficient in usual satellite applications. Thermal control in the field of Earth reentry systems as well as space power plants benefit from the application of high temperature multilayer insulations. The analytic prediction of high temperature multilayer insulation performance in a changing environment is enabled if the dependence of the heat transfer modes on the various design parameters is known. Systematic and tailored design of the insulation becomes feasible then. This is illustrated by three typical examples like a planetary probe for the exploration of the solar system, an Earth based space transportation system and a solar-dynamic space power system for low Earth orbit application.

This paper describes the optimization of a Cassegrainian collector/receiver system to be used in a solar-dynamic power module for a space station. After an introduction to solar-dynamic space power generation and its various alternatives of solar collectors, the Cassegrainian collector/receiver system is described in more detail, its advantages are outlined. Because a useful numeric tool for analysing the double reflector Cassegrainian collector/receiver system was not available, a computer code was developed. The code allows the variable modelling of the collector, the receiver, includes solar limb darkening effects, and offers the possibility to make realistic assumptions for collector/receiver system errors. Hence, this computer code sets the ground for real-life operation simulation. The exact knowledge of the intensity distribution in the receiver aperture plane and the receiver interior is indispensable for aperture optimization and receiver simulation and design. These intensity distributions are supplied by the computer program. Results of a sensitivity analysis are presented and discussed.
