Perineuronal nets (PNs) are a unique and complex meshwork of specific extracellular matrix molecules that ensheath a subset of neurons in many regions of the central nervous system (CNS). PNs appear late in development and are supposed to restrict synaptic plasticity and to stabilize functional neuronal connections. PNs were further hypothesized to create a charged milieu around the neurons and thus, might directly modulate synaptic activity. Although PNs were first described more than 120 years ago, their exact functions still remain elusive. The purpose of the present review is to propose the nuclei of the auditory system, which are highly enriched in PN-wearing neurons, as particularly suitable structures to study the functional significance of PNs. We provide a detailed description of the distribution of PNs from the cochlear nucleus to the auditory cortex considering distinct markers for detection of PNs. We further point to the suitability of specific auditory neurons to serve as promising model systems to study in detail the contribution of PNs to synaptic physiology and also more generally to the functionality of the brain.

Adamsky et al. show that stimulation of astrocytes potentiates synaptic transmission and enhances behavioral performance in memory tasks. These results demonstrate that astrocytes are actively involved in synaptic physiology and brain function and lend further support to the idea that animal behavior results from the coordinated activity of neurons and astrocytes.

An increasing number of studies indicate that there exists greater diversity of cortical neurons than previously appreciated. In the present report, we use a combination of physiological and morphological methods to characterize cortical neurons in infragranular layers with apical dendrites pointing toward the white-matter compared to those neurons with apical dendrites pointing toward the pia in both mouse and rat neocortex. Several features of the dendritic morphology and intrinsic and synaptic physiology of these “inverted” neurons revealed numerous differences among this cell type between species. We also found differences between the different cell types within the same species. These data reveal that similar cell types in the rat and mouse may not always share similar physiological and morphological properties. These data are relevant to models of information processing through micro- and larger neocortical circuits and indicate that different cell types found within similar lamina can have different functional properties.

Synapses exhibit diverse functional properties, and it seems likely that these properties are specialized to perform specific computations. The Drosophila antennal lobe provides a useful experimental preparation for exploring the relationship between synaptic physiology and neural computations. This review summarizes recent progress in describing synaptic properties in the Drosophila antennal lobe. These studies reveal that several types of synapses in this circuit are highly specialized, and that these specializations are in some cases under tight regulatory control. These synaptic specializations can be understood in terms of the computational features they confer on the circuit. Specifically, many of these properties appear to promote odor detection when odor concentrations are low, while promoting adaptive gain control when odor concentrations are high.

Synapses are the fundamental units of information processing in the mammalian brain. Much of our understanding of their functional properties comes from voltage-clamp analysis, the predominant approach for investigating synaptic physiology. Here, we reveal that voltage clamp is completely ineffective for most excitatory synapses due to spine electrical compartmentalization. Under local dendritic voltage clamp, single-spine activation produced large spine head depolarizations that severely distorted measurements and recruited voltage-dependent channels. To overcome these voltage-clamp errors, we developed an approach to provide new, accurate measurements of synaptic conductance. Single-synapse AMPA conductance was much larger than previously appreciated, producing saturation effects on synaptic currents. We conclude that electrical compartmentalization profoundly shapes both synaptic function and how that function can be assessed with electrophysiological methods.

During the 1950s to 70s most of the mechanisms that control transmitter release from presynaptic nerve terminals were described at the neuromuscular junction. It was not, however, until the 1990s that the multiplicity of protein-protein interactions that govern this process began to be identified. The sheer numbers of proteins and the complexity of their interactions at first appears excessive, even redundant. However, studies of identified central synapses indicate that this molecular diversity may underlie a important functional diversity. The task of the neuromuscular junction is to relay faithfully the rate and pattern code generated by the motoneurone. To demonstrate phenomena such as facilitation and augmentation that are apparent only when the probability of release is low, experimental manipulation is required. In the cortex, however, low probability synapses displaying facilitation can be recorded in parallel with high probability synapses displaying depression. The mechanisms are largely the same as those displayed by the neuromuscular junction, but some are differentially expressed and controlled. Central synapses demonstrate exquisitely fine tuned information transfer, each of the many types displaying its own repertoire of pattern- and frequency-dependent properties. These appear tuned to match both the discharge pattern in the presynaptic neurone and the integrative requirements of the postsynaptic cell. The molecular identification of these differentially expressed frequency filters is now just coming into sight. This review attempts to correlate these two aspects of synaptic physiology and to identify the components of the release process that are responsible for the diversity of function.

A key question in synaptic physiology is what determines the release probability of a synaptic vesicle. In this issue of Neuron , Wadel et al. shed UV light on this problem, finding that hard-to-release vesicles are too far from Ca 2+ channels.

Do graded potential synapses work the same way as action potential synapses? Recent work emphasizes the differences and suggests that graded potential synapses are not all the same either.

Maitotoxin (MTX), applied in vitro to the mouse neuromuscular junction, briskly activates the spontaneous release of acetylcholine quanta, manifest as miniature end-plate potentials (MEPPs). This effect requires external Ca2+ and is accompanied by a steady post-junctional depolarization. After the peak activation of the spontaneous release process, the quantal discharge gradually declines with eventual abolishment of MEPPs. In contrast to the striking increase in MEPP frequency, the quantum content of the nerve-evoked end-plate potentials (EPPs) is increased only moderately by MTX. These effects are attributable to the ability of the toxin to elevate the level of free intracellular Ca2+ in the motor nerve terminal. With further characterization of its presynaptic site of action, maitotoxin may become a useful tool in studying synaptic physiology.

Slices of the field CA1 minus the stratum moleculare were prepared from the rat hippocampus and maintained in an in vitro recording chamber. The physiological properties of these "minislices" were similar to those reported for non-dissected hippocampal slices. Slices receiving various patterns of electrical stimulation through multiple electrodes were subsequently homogenized and crude membrane fractions prepared; the binding of [3H]glutamate was measured by a rapid filtration assay. Binding in membranes prepared from control slices exhibited kinetic properties and sensitivities to pharmacological and ionic manipulations which were comparable to those found in previous studies using conventional fractionation and assay techniques. Brief bursts of high-frequency stimulation increased [3H]glutamate binding compared to non-stimulated controls in 3 separate experiments. Stimulation at low frequency or at high frequency in low calcium medium did not produce this effect. In addition to suggesting that glutamate binding sites are regulated by patterns of afferent activity, these findings indicate that the minislice preparation should be of general utility in relating synaptic physiology to synaptic chemistry.

It is a sorry situation that despite considerable efforts we are totally presynaptic and postsynaptic spinal cord inhibitory systems, not to mention other areas. Sooner or later one hopes that the blunderbuss approach being pursued at present will lead to a breakthrough, but there is really little sing of this so far” (Burgen, 1964).
